7. Toward Structural Correctness: Aquatolide and the Importance of 1D Proton NMR FID Archiving

Abstract

The revision of the structure of the sesquiterpene aquatolide from a bicyclo[2.2.0]hexane to a bicyclo[2.1.1]hexane structure using compelling NMR data, X-ray crystallography, and the recent confirmation via full synthesis exemplify that the achievement of "structural correctness" depends on the completeness of the experimental evidence. Archived FIDs and newly acquired aquatolide spectra demonstrate that archiving and rigorous interpretation of 1D (1)H NMR data may enhance the reproducibility of (bio)chemical research and curb the growing trend of structural misassignments. Despite being the most accessible NMR experiment, 1D (1)H spectra encode a wealth of information about bonds and molecular geometry that may be fully mined by (1)H iterative full spin analysis (HiFSA). Fully characterized 1D (1)H spectra are unideterminant for a given structure. The corresponding FIDs may be readily submitted with publications and collected in databases. Proton NMR spectra are indispensable for structural characterization even in conjunction with 2D data. Quantum interaction and linkage tables (QuILTs) are introduced for a more intuitive visualization of 1D J-coupling relationships, NOESY correlations, and heteronuclear experiments. Overall, this study represents a significant contribution to best practices in NMR-based structural analysis and dereplication.

17. Forensic Chemistry: The Revelation of Latent Fingerprints

Abstract

The visualization of latent fingerprints often involves the use of a chemical substance that creates a contrast between the fingerprint residues and the surface on which the print was deposited. The chemical-aided visualization techniques can be divided into two main categories: those that chemically react with the fingerprint residue and those that adhere to the fingerprint residue by intermolecular forces. A plethora of empirically effective fingerprint revelation methods have been developed but the chemistry is often incompletely understood. This article briefly describes the chemical rationale of most fingerprint visualization techniques practiced today. This material is suitable for a forensic chemistry course or for an introductory chemistry course to introduce the relevance of chemistry in law enforcement. This material may also apply to an organic or biochemistry course when discussing noncovalent intermolecular interactions.

Abstract

Nielsen et al. (1) argue that Van Doesum et al. (2) need to consider three points for their interpretation of a positive association between individual-level social mindfulness (SoMi) and environmental performance (EPI) at the country level (3). The association is weaker when 1) it is controlled for GDP and 2) when the data of three countries are removed; also, 3) the data do not address the association between SoMi and individual-level environmental concern. We discuss these points in turn.

Abstract

Humans are social animals, but not everyone will be mindful of others to the same extent. Individual differences have been found, but would social mindfulness also be shaped by one's location in the world? Expecting cross-national differences to exist, we examined if and how social mindfulness differs across countries. At little to no material cost, social mindfulness typically entails small acts of attention or kindness. Even though fairly common, such low-cost cooperation has received little empirical attention. Measuring social mindfulness across 31 samples from industrialized countries and regions (n = 8,354), we found considerable variation. Among selected country-level variables, greater social mindfulness was most strongly associated with countries' better general performance on environmental protection. Together, our findings contribute to the literature on prosociality by targeting the kind of everyday cooperation that is more focused on communicating benevolence than on providing material benefits.

25. Dehydration of 2-Methyl-1-Cyclohexanol: New Findings from a Popular Undergraduate Laboratory Experiment

Abstract

The mineral acid-catalyzed dehydration of 2-methyl-1-cyclohexanol has been a popular laboratory exercise in second-year organic chemistry for several decades. The dehydration experiment is often performed by organic chemistry students to illustrate Zaitsev's rule. However, sensitive analytical techniques reveal that the results do not entirely corroborate with Zaitsev's rule. Previous reports pertaining to this experiment have been limited by two factors: (i) it is difficult to separate 3- and 4-methyl-1-cyclohexene on a standard gas chromatography (GC) column and (ii) methods of accurate detection, identification, and quantification of trace minor products have been lacking. The current study uses gas chromatography--electron impact mass spectrometry (GC-MS) and quantitative nuclear magnetic resonance (qNMR) to identify and quantify not only the putative 1-, 3-, and 4-methyl-1-cyclohexene products but also several minor products such as methylenecyclohexane and ethylidenecyclopentane. Performing the dehydration with a single diastereomer of 2-methyl-1-cyclohexanol reveals a striking difference between the product distributions associated with cis and trans isomers. Some evidence continues to point towards an E2-like mechanism whereas other evidence, such as the presence of methylcyclohexene dimers in the still pot, points towards E1 carbocation intermediates. 26. Saying What You Mean: Teaching Mechanisms in Organic Chemistry

Abstract

Organic reactions in introductory organic chemistry courses are most commonly taught with a mechanism-based approach to the understanding of molecular reactivity. However, the effectiveness of the popular curved arrow representation to describe reaction mechanisms is often compromised by the overuse of shortcuts and obscure notation. The consistent use of balanced reaction equations, a concept that most students have learned and practiced in previous chemistry courses, is an effective technique in communicating reaction mechanisms. Other ways to introduce clarity and consistency to reaction mechanisms are: (i) avoiding the use of shortcut notations, (ii) including key electrons and bonds in structural representations, and (iii) distinguishing between covalent and ionic bonds. Abstract

Komatsu et al. (1) argue that Van Doesum et al. (2) may have overlooked the role of GDP in reporting a positive association between social mindfulness (SoMi) and the Environmental Performance Index (EPI) at country level. Although the relationship between EPI and SoMi is relatively weaker for countries with higher GDP, that does not imply that the overall observed relationship is a statistical artifact. Rather, it implies that GDP may be a moderator of the relationship between EPI and SoMi. The observed correlation is a valid result on average across countries, and the actual effect size would, at least to some degree, depend on GDP.

Abstract

Despite many decades of research to develop a malaria vaccine, only one vaccine candidate has been explored in pivotal phase III clinical trials. This candidate subunit vaccine consists of a portion of a single Plasmodium antigen, circumsporozoite protein (CSP). This antigen was initially identified in the murine malaria model and shown to contain an immunodominant and protective CD8+ T cell epitope specific to the H-2Kd (BALB/c)-restricted genetic background. A high-content screen for CD8+ epitopes in the H2Kb/Db (C57BL/6)-restricted genetic background, identified two distinct dominant epitopes. In this study, we present a characterization of one corresponding antigen, the Plasmodium sporozoite-specific protein S20. Plasmodium berghei S20 knockout sporozoites and liver stages developed normally in vitro and in vivo. This potent infectivity of s20(-) sporozoites permitted comparative analysis of knockout and wild-type parasites in cell-based vaccination. Protective immunity of irradiation-arrested s20(-) sporozoites in single, double and triple immunizations was similar to irradiated unaltered sporozoites in homologous challenge experiments. These findings demonstrate the presence of an immunogenic Plasmodium pre-erythrocytic determinant, which is not essential for eliciting protection. Although S20 is not needed for colonization of the mammalian host and for initiation of a blood infection, it is conserved amongst Plasmodium species. Malarial parasites express conserved, immunogenic proteins that are not required to establish infection but might play potential roles in diverting cellular immune responses.
