Background and Purpose: The relationship between osteopathic (COMLEX) and allopathic (USMLE) exam scores in not well described. We sought to describe the relationship between COMLEX and USMLE scores for osteopathic medical students who reported scores for both exams during application to internal medicine residency. Methods: Cross sectional study of 588 matched USMLE/COMLEX Step 1 scores and 241 matched Step 2 scores. Means, standard deviations, and pairwise correlations for matched scores were calculated. Results: USMLE and COMLEX paired scores resulted in a Pearson's correlation of 0.85 for Step 1 scores and 0.79 for Step 2 scores. COMLEX means were 560 and 561 for Level 1 and 2 and USMLE means were 209 and 215 for Step 1 and 2. Conclusions: USMLE and COMLEX scores are strongly related for internal medicine residency applicants who took both exams. Our sample performed approximately 0.8 standard deviation higher that the national COMLEX means on both Level 1 and 2 but at the national mean on USMLE Step 1 and 2. The variation in scoring methodology and potential differences between the study sample and individual applicant characteristics suggesteject caution should be taken in using a given COMLEX score as a predictor of performance on the USMLE. Background: Insufficient sleep and insomnia are associated with alcohol use as well as anxiety during adolescence and young adulthood. A negative reinforcement path to explain the association between sleep difficulties and alcohol misuse has been proposed. Within this pathway, it is speculated that while sober, insomnia and insufficient sleep lead to increased anxiety as well as anxiolytic responses to alcohol, thereby increasing the risk for both alcohol use and alcohol use problems. No work to date has examined the negative reinforcement path to alcohol use among adolescents who have consumed alcohol. Objectives: The current study aims to address this gap in the literature by examining if sleep quality is related to adolescent alcohol use problems and frequency through serial indirect effects of adolescent anxiety symptoms and coping motives for alcohol use. A total of 147 adolescents between the ages of 13 and 18 years ( M age = 16.31; SD = 0.96) from all geographic regions in the US were recruited using social media platforms (i.e., Facebook and Instagram). Participants who reported having tried alcohol at least once completed self-report measures to examine sleep quality, anxiety symptoms, alcohol use problems, alcohol use frequency, and coping motives for alcohol use. Results: Adolescent sleep quality was found to be associated with higher levels of both alcohol use problems and alcohol use frequency through the serial indirect effect of anxiety symptoms and coping motives for alcohol use. Conclusions: Overall, these findings represent a step towards understanding the complex relationship between sleep quality, alcohol, anxiety, and coping motives among adolescents.

Much research has demonstrated that psychopathology can be described in terms of broad dimensions, representing liability for multiple psychiatric disorders. Broad spectra of psychopathology (e.g., internalizing and externalizing) are increasingly used as targets for research investigating the development, etiology, and course of psychopathology because they account for patterns of relatedness among disorders that were once presumed distinct. Thus, these spectra represent alluring targets due to their comprehensive and parsimonious nature. Nevertheless, little research has established the role of individual disorders over and above broad dimensions in the study of psychopathology. In the current study, we investigate whether there are unique etiological associations between individual internalizing disorders and personality traits after accounting for their etiological associations with a broad internalizing dimension. We used a community sample of twins (Npairs = 448) ages 4 to 19 to examine the etiological associations between internalizing psychopathology and Big Five personality dimensions. In terms of genetic covariation, a broad internalizing dimension was positively associated with neuroticism and negatively associated with extraversion, agreeableness, and conscientiousness. Moreover, internalizing accounted for most of the genetic variance shared between individual internalizing disorders and personality traits. Nevertheless, there were unique genetic associations between the following pairs of personality traits and disorders: neuroticism and social anxiety, extraversion and social anxiety, agreeableness and depression, and conscientiousness and compulsions. There was little evidence of environmental influences shared between internalizing and personality. In sum, a broad internalizing dimension adequately accounted for almost all of the etiologic covariation between internalizing disorders and personality, with several interesting exceptions. Introduction: Adolescence is characterized by the onset of a relatively specific set of socioemotional disorders (i.e., depression, generalized anxiety disorder, social anxiety disorder, and eating disorders) as well as body dysmorphia symptoms. Appearance-related concerns are a central feature of these disorders. Emerging evidence in adults suggests that appearance-related safety behaviors may play an instrumental role in the onset and maintenance of a number of disorders. To date, no work has examined appearance-related safety behaviors during adolescence. The present study examined the extent to which appearance-related safety behaviors may be associated with socioemotional and body dysmorphia symptoms during adolescence., Methods: Adolescents between the ages of 13 and 17 years old (N = 387, M age  = 14.82 years, 31.3% identified as male, 47.0% identified as female, and 19.1% identified as nonbinary/third gender, 2.6% declined to report gender identity) completed measures assessing negative affect, anxiety-relevant safety behavior use, cognitive reappraisal, expressive suppression, appearance-related safety behaviors, body dysmorphia symptoms, and socioemotional symptoms. Structural Equation Modeling was used to test hypotheses., Results: The results of this study suggest that appearance-related safety behaviors evidenced associations with latent factors corresponding to affective (i.e., depression, generalized anxiety disorder, social anxiety), eating disorders, and body dysmorphia symptoms after controlling for previously established vulnerability factors., Conclusions: These findings demonstrate that appearance-related safety behaviors may evidence transdiagnostic associations with socioemotional symptoms and body dysmorphia symptoms during adolescence., (© 2022 The Foundation for Professionals in Services for Adolescents.)

Both maternal symptoms and adolescent offspring characteristics are associated with maladaptive parenting among families at risk for anxiety. One disorder that may be particularly associated with maladaptive parenting behaviors is generalized anxiety disorder (GAD). Previous work suggests that offspring negative affect (NA) is associated with different levels of maladaptive parenting behaviors among mothers with GAD. No work to date, however, has examined the association between offspring reported NA, maternal GAD, and maternal worry about offspring or maternal perceptions of psychological control (PC) among mothers of adolescents. Sixty-five mothers who were elevated in anxious arousal and their adolescent offspring between the ages of 12 and 16 years old (n = 65, 55% male, M age = 13.89) reported on parenting (mother report), NA (offspring report), and GAD symptoms (assessed via structured clinical interview), and maternal anxiety sensitivity (AS) symptoms. Study results indicated that maternal GAD status interacted with offspring NA in relation to maternal reported use of PC and worry about offspring. Specifically, offspring NA was positively related to PC for mothers without GAD, but not for mothers with GAD. Further, for mothers with GAD, offspring NA was negatively related to worry about offspring, but this relation did not persist for mothers without GAD. Maternal AS was related to overall higher levels of worry about offspring and PC. Mothers with GAD report using higher levels of maladaptive parenting when offspring report lower levels of NA, and lower levels when offspring report high NA. Background: Cigarette use during adolescence has been linked to increased risk for insomnia symptoms, but limited work has examined factors that may account for this association. Adolescent cigarette use and anxiety symptoms characterized by physiological hyperarousal evidence bidirectional associations, as do anxiety symptoms and insomnia symptoms. This suggests that adolescent cigarette use, anxiety symptoms characterized by physiological hyperarousal, and insomnia symptoms may increase and maintain one another. The current study tests physiological hyperarousal anxiety symptoms as a potential indirect effect in the cigarette-insomnia symptoms link across adolescence and young adulthood., Methods: We examined data from adolescents and young adults from Waves 1, 2, 3 and 4 of the National Longitudinal Study of Adolescent to Adult Health (N = 2,432 with full data). Insomnia symptoms were assessed at baseline (ages 12-16 years), 1 year later (13-17 years), and 14 years after baseline (26 - 30 years) among a nationally representative sample of adolescents. Cigarette use was assessed at baseline, 1 year later, 6 years after baseline, and 14 years after baseline. Anxiety symptoms were assessed at baseline and 1 year later., Results: Structural equation models indicated that anxiety symptoms exerted an indirect effect on the longitudinal associations between adolescent cigarette use and adult insomnia symptoms. Anxiety symptoms and cigarette use evidenced bidirectional associations during adolescence., Conclusions: These results suggest that increases in anxiety symptoms characterized by physiological hyperarousal may be one mechanism whereby cigarette use during adolescence is associated with increased insomnia symptoms during early adulthood. Introduction: Sleep disturbances are common among adolescents and are associated with elevated anxiety, and difficulties managing affect. Familial conflict is associated with both anxiety sensitivity and adolescent sleep disturbances. No work to date has examined how adolescent sleep disturbances may interact with anxiety sensitivity in relation to adolescent affective responding to parent-adolescent conflict. The current study was designed to address this gap in the literature by examining how adolescent sleep disturbances, anxiety sensitivity, conflict elicited anger, and conflict avoidance are associated., Method: Seventy-two American adolescents (n = 39 males) between the ages of 12 and 16 years (M age  = 13.84, SD = 1.38) completed a baseline assessment as well as a well-validated mother-adolescent laboratory-based conflict task., Results: For youth low in anxiety sensitivity, greater sleep disturbance related positively to conflict-elicited anger, which in turn predicted higher conflict avoidance. In contrast, this indirect effect was not significant for adolescents relatively higher in anxiety sensitivity. Instead, for these adolescents, increased sleep disturbances were associated with lower levels of conflict elicited anger., Conclusions: Results suggest that the effects of sleep disturbances on conflict elicited anger may vary as a function of adolescent anxiety vulnerability. The Arabidopsis thaliana wall-associated kinases (WAKs) bind to pectin with an extracellular domain and also contain a cytoplasmic protein kinase domain. WAKs are required for cell elongation and modulate sugar metabolism. This work shows that in leaf protoplasts a WAK1-GFP fusion protein accumulates in a cytoplasmic compartment that contains pectin. The WAK compartment contains markers for the Golgi, the site of pectin synthesis. The migration of WAK1-GFP to the cell surface is far slower than that of a cell surface receptor not associated with the cell wall, is influenced by the presence of fucose side chains on one or more unidentified molecules that might include pectin, and is dependent upon cellulose synthesis on the plasma membrane. WAK is crosslinked into a detergent-insoluble complex within the cytoplasmic compartment before it appears on the cell surface, and this is independent of fucose modification or cellulose synthesis. Thus, the assembly and crosslinking of WAKs may begin at an early stage within a cytoplasmic compartment rather than in the cell wall itself, and is coordinated with synthesis of surface cellulose.

Objective: To develop a quantitative severity stratification within the framework of a Physiologic State Classification (PSSC) system that can be applied to critically ill post-trauma patients with "sepsis/SIRS" and to relate PSSC to the nature of the plasma cytokine response., Materials and Methods: At each study time period, a patient was classified into one of seven physiologic States previously derived from clustering 17 cardiopulmonary and metabolic variables from 338 critically ill patients: R = reference, A = normal stress response, B = metabolic insufficiency, C1 (early) and C2 (late) = respiratory insufficiency, D = cardiogenic insufficiency, H = nonshock hypovolemia., Main Results: The PSSC used State data from a developmental set of 159 trauma patients in a logistic model (L2PDEATH) to provide a quantitative index of severity. This severity index was tested on 80 new trauma patients (mean injury Severity Score (ISS) = 27.6, 64% survivors). Using PSSC State distributions for evaluation of enzyme-linked immunosorbent assay (ELISA) measured cytokines interleukin (IL)-1, IL-6, IL-8, tumor necrosis factor (TNF) showed the multicytokine score to be greatest in those C2- and B-State regions associated with a higher severity as measured by L2PDEATH. Compared with ARDEATH of the Acute Physiology and Chronic Health Evaluation (APACHE) II scoring system, L2PDEATH provided a better indicator of severity of sepsis/systemic inflammatory response syndrome (SIRS) for posttrauma patients., Conclusions: PSSC allows classification of the physiologic and cytokine mediator response to trauma and permits stratification of severity in posttrauma critical illness.

We describe a simple procedure for isolation of Vicia villosa lectin with binding specificity for N-acetyl-D-galactosamine greater than D-galactose. Also, a rosette assay using V. villosa lectin coupled-SRBC is described which enables us to both visualize lectin binding cells and to fractionate lymphocytes from vitro primary mixed lymphocyte cultures on the basis of ability to bind V. villosa lectin.

Anti-human T-lymphocyte serum (anti-HTLA) was used to investigate the relationship between differentiation antigens of human (HTLA) and Macaca lymphocytes. The percentage of lymphocytes sensitive to anti-HTLA is the same in both species, with thymocytes having a higher concentration of HTLA than more mature T-lymphocytes. Macaca bone marrow cells acquire phenotypic properties of T-lymphocytes in vitro in the presence of calf thymic factors. Our data suggest that phylogenetic distinctions between various T-lymphocyte differentiation antigens may be utilized to provide antisera of narrow specificity.

Seeds of six soybean lines (Glycine max (L.) Merr. cv. Columbia, D68-127, Norredo, Sooty, T-102, Wilson 5) have been reported to lack the 120 000 dalton soybean lectin. Immunodiffusion and radioimmunoassay using anti-soybean lectin immunoglobulin failed to detect the lectin in seeds of five lines, but D68-127 seeds contained as much soybean lectin as the control line, Harosoy 63. The D68-127 seed lectin could be purified by affinity chromatography on Sepharose-N-caproylgalactosamine, and was indistinguishable from the conventional soybean lectin by the following criteria: electrophoretic migration in acidic and alkaline buffers, subunit molecular weight and composition, analytical isoelectric focusing, gel filtration chromatography. Phosphate-buffered saline extracts of roots, hypocotyls, stems, and leaves of 3--66-day-old Norredo and Harosoy 63 plants lacked soybean lectin, as determined by hemagglutination and radioimmunoassay (detection limit: 1.4 micrograms soybean lectin/g dry weight tissue). Cotyledons of Harosoy 63 (but not Norredo) contained large quantities of the lectin, which diminished as the plants aged. 5-day-old roots and hypocotyls of 20 soybean lines did not contain soybean lectin. Roots of Columbia, Norredo, Sooty, T-102, Wilson 5, and Harosoy 63 (control) were nodulated by a variety of strains of Rhizobium japonicum and Rhizobium sp.

A cross-sectional study of 60 multiple sclerosis outpatients assessed neuropsychological variables in relation to age, duration of disease, and degree of disability. The test battery included subtests of the WAIS (Vocabulary, Similarities, and Digit Span);the Raven matrices; and memory tests (Wechsler Paired Associates, Benton Visual Retention). Results showed that cognitive functioning of MS patients varies considerably. There is a general mild decline that affects abstract reasoning and memory as the disease progresses. These deficits are usually not sufficient to impair occupational or social functioning. In addition, there is spotty deterioration in certain cognitive functions for some patients (17%). Only a small subsample (6.7%), however, showed global deterioration significant enough to be highlighted by an Index of Dementia that was developed for this investigation.

Using a horse anti-human thymocyte serum made specific for T lymphocytes by absorption (anti-HTLA serum), a material reacting with it has been extracted from detergent-lysed peripheral blood lymphocytes. Acid elution and gel-filtration analysis allowed recovery of two peaks containing HTLA activity: one of 30,000-40,000 Daltons, the other being excluded by Sephadex G-200 and likely to be more than 300,000 Daltons. HTLA appeared to be released by lymphocytes incubated at 37 degrees, but not at 4 degrees, and significantly detectable after 10-24-h incubation. Finally co-capping experiments with anti-beta2m antibodies and absence of inhibition of cytolysis by anti-HTLA serum of celld beta2m.

Four of 14 strains of Rhizobium japonicum from soybean nodulated peanut (Arachis hypogaea L. cultivar Jumbo Virginia), and 3 of 8 Rhizobium sp. strains from peanut nodulate soybean (Glycine max (L.) Merr. cultivar Harosoy 63). Cells of three peanut rhizobia bound fluorescent- and radioisotope-labeled soybean lectin. Two of these strains failed to nodulate soybean, and conversely, two peanut strains that nodulated soybean did not bind to soybean lectin. Both culture medium and age had pronounced effects of the number of peanut rhizobia cells that bound fluorescent-labeled soybean lectin. Harosoy 63 soybean root exudates stimulated the growth of peanut rhizobia, but had no consistent influence on the number of cells that bound soybean lectin. Although extracellular soybean lectin receptors were present in culture fluids from each of the peanut rhizobia whose cells bound the lectin, the titer of receptors was greatest for strains 3G4b5. The affinity constants for the adherence of soybean lectin to Rhizobium sp. 3G4b5 cells from cultures of various ages ranged from 4.2 X 10(6) to 4.9 X 10(6) M-1, and the number of lectin binding sites per cell decreased as cells aged. Cells of the soybean and peanut rhizobia did not bind fluorescent- or radioisotope-labeled peanut lectin. The results indicate that there is no relationship between the ability of peanut and soybean rhizobia to nodulate the reciprocal host plant and their ability to bind to the lectin of that plant.

Cells from rat peritoneal exudate incubated in vitro with phage T2 yield RNA which induces cultures of rat lymph nodes to form antibodies. Complement fixation reveals T2 head, tail, and internal protein antigens in the RNA. Inoculating the RNA into mice that had previously been stimulated primarily with whole phage T2 results in a secondary response of neutralizing antibody.
