Background Cell disruption is a key unit operation to make valuable, intracellular target products accessible for further downstream unit operations. Independent of the applied cell disruption method, each cell disruption process must be evaluated with respect to disruption efficiency and potential product loss. Current state-of-the-art methods, like measuring the total amount of released protein and plating-out assays, are usually time-delayed and involve manual intervention making them error-prone. An automated method to monitor cell disruption efficiency at-line is not available to date. Results In the current study we implemented a methodology, which we had originally developed to monitor E. coli cell integrity during bioreactor cultivations, to automatically monitor and evaluate cell disruption of a recombinant E. coli strain by high-pressure homogenization. We compared our tool with a library of state-of-the-art methods, analyzed the effect of freezing the biomass before high-pressure homogenization and finally investigated this unit operation in more detail by a multivariate approach. Conclusion A combination of HPLC and automated data analysis describes a valuable, novel tool to monitor and evaluate cell disruption processes. Our methodology, which can be used both in upstream (USP) and downstream processing (DSP), describes a valuable tool to evaluate cell disruption processes as it can be implemented at-line, gives results within minutes after sampling and does not need manual intervention.

Wetter und Klima werden signifikant durch den turbulenten Austausch von Wärme, Impuls, Wasser und Spurenstoffen zwischen der Atmosphäre und dem Ozean beeinflusst. Diese Austauschprozesse finden innerhalb der atmosphärischen und der oberen ozeanischen Grenzschicht statt und hängen im Wesentlichen von der Temperaturdifferenz zwischen den beiden Systemen und den jeweiligen Strömungen ab. Eine typische meteorologische Situation mit großen Temperaturdifferenzen stellen polare Kaltluftausbrüche (KLA) dar, wobei eine kalte und trockene Luftmasse durch einen synoptisch-skaligen Wind von einer eisbedeckten Landmasse über den wärmeren Ozean advehiert wird. Aufgrund einer heterogenen Eiskante ist die Konvektion über dem Ozean typischerweise in Form von Rollen organisiert. Diese Rollen erstrecken sich vertikal über die gesamte Grenzschicht und horizontal über mehrere hundert Kilometer. Die turbulenten Austauschprozesse innerhalb der Grenzschichten können in globalen numerischen Wetter-, Klima- und Ozeanmodellen nicht aufgelöst, sondern müssen parametrisiert werden. Dafür ist ein tieferes Verständnis dieser Prozesse notwendig. Bisherige Studien konnten zwar zeigen, dass Konvektionsrollen einen signifikanten Anteil am vertikalen Transport haben, jedoch nicht, ob dadurch der gesamte turbulente Transport verändert wird. Diese Frage wird in dieser Arbeit mithilfe von Grobstruktursimulationen (engl. Large-Eddy Simulation - LES) untersucht. Im ersten Teil dieser Studie wurden 12 unterschiedliche polare KLA-Situationen simuliert, wobei der synoptisch-skalige Wind und die Ozeantemperatur variierte. Mittels unterschiedlicher Meereisverteilungen konnte für jede KLA Situation jeweils ein Fall mit und ohne Rollen unter denselben meteorologischen Bedingungen simuliert werden. Die Eigenschaften der simulierten Rollen stimmen gut mit beobachteten Konvektionsrollen in KLA überein. Für jeden KLA wurde sowohl die Grenzschichtentwicklung zwischen dem Rollen und Nicht-Rollen Fall verglichen, als auch der Rollenanteil am vertikalen turbulenten Transport analysiert. Die Ergebnisse zeigen, dass die Rollen einen wesentlichen Anteil am vertikalen Transport haben, aber weder den gesamten Transport noch die Entwicklung der konvektiven Grenzschicht verändern. Dieses konnte für unterschiedliche Wellenlängen der Rollen und auch eine trockene Atmosphäre bestätigt werden. Wie bereits durch Beobachtungen bekannt, modifizieren atmosphärische Konvektionsrollen die Struktur der Meeresoberfläche inklusive der oberflächennahen Flüsse. Darum wurde im zweiten Teil dieser Studie der Einfluss der Konvektionsrollen auf den Ozean untersucht, und ob es ggf. zu Rückkopplungseffekten mit der Atmosphäre kommen könnte. Die Simulationen der oberen ozeanischen Grenzschicht wurden angetrieben durch oberflächennahe Flüsse einer der KLA Simulationen. Dabei wurden Simulationen mit und ohne Rollensignale an der Meeresoberfläche durchgeführt. Die Simulationen mit Rollensignal zeigen, dass eine sehr schwache rollenartige Zirkulation angeregt wird, welche sich über die gesamte ozeanische Grenzschicht erstreckt. Ein Vergleich der Grenzschichtentwicklung zwischen den Simulationen mit und ohne Rollensignal weist keine signifikanten Unterschiede auf. Dieses gilt auch für die Meeresoberfläche, so dass es keinen veränderten Wärme- oder Feuchteeintrag in die Atmosphäre gibt. Dieses Ergebnis konnte auch unter Berücksichtigung der Langmuir-Zirkulation im Ozean bestätigt werden. Diese Studie zeigt, dass atmosphärische Rollen, angeregt durch stromaufwärts gelegene Heterogenitäten, einen wesentlichen Anteil am vertikalen turbulenten Transport haben ohne den Gesamttransport zu erhöhen. Darum müssen solche Rollen nicht in Parametrisierungen globaler Modelle, welche keine großskalige Turbulenz auflösen, gesondert berücksichtigt werden., Weather and climate are significantly influenced by the turbulent exchange of heat, momentum, water and trace substances between the atmosphere and the ocean. These exchange processes take place within the atmospheric and upper oceanic boundary layer and depend essentially on the temperature difference between the two systems and the respective flows. A typical meteorological situation with large temperature differences are polar cold air outbreaks (CAO), where cold and dry air is advected by synoptic-scale wind from an ice-covered landmass over the warmer ocean. With structured ice edges, the convection over the ocean is typically organized into rolls, which extend vertically across the entire boundary layer and horizontally up to several hundreds of kilometers. The turbulent exchange processes within the boundary layers cannot be resolved in large-scale models, such as global numerical weather, climate and ocean models, and thus must be parametrized. Therefore, a deeper understanding of these processes is necessary. Although previous studies have shown that convective rolls play a significant role for the vertical turbulent transport, it is not clear if the boundary layer rolls affect the total turbulent transport. This question will be investigated in the current study by means of large-eddy simulations (LES). In the first part of this study, 12 different polar CAO situations were simulated, by varying the synoptic-scale wind and the ocean temperature. By means of different sea ice distributions, a case with and without rolls could be simulated under the same meteorological conditions for each CAO situation. The properties of the simulated rolls agreed well with observed convective rolls in CAO. The boundary layer development was compared between the roll and the nonroll case, and the roll contribution to the vertical turbulent transport was analyzed for each investigated CAO situation. Results show that although the rolls make a substantial contribution to the vertical transport, they neither affect the total transport nor the development of the boundary layer. These results could also be confirmed for different roll wavelengths and additionally for a dry atmosphere. As known from observations, atmospheric rolls modify the structure of the sea surface, including the surface fluxes. Therefore, the influence of atmospheric convective rolls on the ocean was investigated in the second part of this study, and, if feedback effects with the atmosphere could occur. For the simulations of the upper oceanic boundary layer, the flow was driven by the near-surface fluxes of one of the CAO simulations, again with and without roll signals at the sea surface. The ocean simulations with a roll signal showed that a very weak roll-like circulation is induced, which extends over the whole oceanic boundary layer. A comparison between the simulations with and without roll signals at the sea surface showed no significant differences in the boundary layer development of the ocean. This also applies to the sea surface, so that there is no feedback on the atmosphere in the form of a changing heat or moisture input. This result was also confirmed by considering the Langmuir circulation. The results of this study showed that atmospheric rolls triggered by upstream heterogeneities contribute substantially to the vertical turbulent transport, without increasing the total transport. Therefore, such rolls do not have to be considered in parametrization schemes of large-scale models, as long as these models do not resolve such large-scale turbulence.

Today’s product requirements demand an ever increasing functionality for the same space and usually the same number of components. Thereby, the quality, reliability and robustness of these products should be preserved or even be increased. This target conflict cannot be solved without compromises. The research community between the Institute of Machine Components (IMA), University of Stuttgart, and the Knorr-Bremse Systeme für Nutzfahrzeuge GmbH is seeking for new solutions for these challenges. The new approaches for designing robust and reliable products are being implemented directly in a current development project of an innovative Air Disc Brake (ADB). With “Systematic Method for Axiomatic Robustness-Testing” (SMART), reliability methods and the basic concept of Robust Design methodology are related to the Taguchi Method. SMART is based on three phases: System, Parameter and Tolerance Design; accordingly, the sample phases of VDA (Association of German Automotives) are used as milestones. In the System Design, SMART focuses on the decreasing complexity according to the functional dependences of the DPs, thus precluding early random failures. In the Parameter Design phase, SMART gives the developer an approach for modeling an adaptive simulation model (SIM-SMART). This model also enables the simulation of random and possible fatigue failures in addition to the nominally robust DPs. In the early stage of product development, reliability predictions are possible. In the iterative Tolerance Design phase, the final tolerance limits for robust and reliable products are defined with consideration of compromises in terms of costs, quality and technical feasibility. With the application of SMART, a design concept of a new generation of an ADB with less complexity is created. The extensive functions for flexible function studies are modeled with the objective of SIM-SMART. Accordingly to this model, parameter studies for determination of the nominal adjustment levels can be performed and their random and fatigue failures modeled. In conclusion, more accurate reliability test strategies are recommended using the definition of tolerance limits. The cost aspect and technical feasibility are also taken into account. So far, SMART has not been added to the iterative Tolerance Design phase. With this paper, the method is not only extended to this phase, but also sufficiently validated. In addition, SMART can predict and analyze random failures. With its three coherent and iterative phases, it is an as yet unpublished and unimplemented approach for designing even more robust and reliable products. Robust Design Methodology and reliability methods are fundamental building blocks for products with high quality requirements. SMART presents an approach to support the designing of robust, reliable, highly functional and innovative ADB.

We analyze around 200 different financial time series, i.e. components of Dow Jones, Nasdaq, FTSE and Nikkei with seven different VaR approaches. We differentiate our analysis according to characteristics that can be observed. Our analysis shows that in high risk situations in which the time series show high volatility risk and high fat tail risk the current Basle II guidelines fail in the attempt to cushion against large losses by higher capital requirements. One of the factors causing this problem is that the builtin positive incentive of the penalty factor resulting from the Basle II backtesting is set too weak. Therefore, we propose adjustments regarding the Basle II penalty factor that take different risk situations into account and lead to higher capital buffers for forecast models with a systematic risk underestimation.

BVDV ist dafür bekannt, dass zythopathogene Virusvarianten durch Integration zellulärer Sequenzen in das virale RNA-Genom entstehen. Die zellulären Insertionen induzieren nach Translation eine zusätzliche Spaltung des viralen Polyproteins. In der vorliegenden Arbeit wurden die Genome von verschiedenen BVDV Isolaten charakterisiert, die alle aus einem erkrankten Tier stammen. Analysen von cDNA-Klonen und RT-PCR Fragmenten erlaubten es, die Genomstrukturen einer Reihe von Viren zu rekonstruieren. Alle enthielten die gleiche zelluläre Sequenz, flankiert von unterschiedlich rearrangierten viralen Sequenzen. Die gefundenen Genomstrukturen lassen sich auf eine Grundstruktur zurückführen. Da die Entstehung solcher Virusvarianten mit weitgehend übereinstimmenden Genomstrukturen in unabhängigen Rekombinationsereignissen unwahrscheinlich ist, deuten diese Ergebnisse auf einen evolutionären Prozess hin, in dem eine Primärrekombinante gebildet und anschliessend durch weitere Rekombinationen verändert wurde. Bei der zellulären Insertion handelt es sich um ein Fragment des LC3 („light chain 3“), einem Bestandteil von Mikrotubuli assoziierten Proteinen. Aufgrund früherer Befunde war zu erwarten, dass das LC3 im viralen Polyprotein als Signal für eine proteolytische Prozessierung dient. Dies konnte in proteinbiochemischen Analysen bestätigt werden. Mutageneseexperimente erlaubten die Charakterisierung der Spaltstelle, die direkt am caroxyterminalen Ende der zellulären Insertion lokalisiert ist. In in vitro-Translationsstudien mit verschiedenen Proteaseinhibitoren konnte zudem gezeigt werden, dass es sich bei der prozessierenden Protease vermutlich um eine Cysteinprotease handelt. Expressionen in Zellen verschiedener Spezies zeigten, dass das prozessierende System außergewöhnlich gut konserviert ist, denn in Zellen von Hamster, Huhn, Fisch und Moskito wurde das bovine Substrat offensichtlich authentisch gespalten. LC3 bzw. das homologe Protein Aut7p/Apg8p der Hefe besitzen wesentliche Funktionen bei der Autophagie von hungernden Zellen. LC3 und Aut7p/Apg8p werden dazu durch carboxyterminale Proteasespaltung aktiviert. Nach Klonierung verschiedener potentieller Proteasegene aus Rinderzellen konnte durch in vitro-Kotranslationen und Expressions-Experimente in Hefezellen gezeigt werden, dass eine der potentiellen Proteasen tatsächlich LC3 prozessiert. Die Expression dieser und einer weiteren potentiellen Rinderprotease in Hefezellmutanten konnte eine Mutation, die zum Verlust der spezifischen Hefeprotease Aut2p/Apg4p führte, komplementieren. Die vorliegende Arbeit hat damit wichtige Erkenntnisse sowohl über die Grundlagen der Zytopathogenität von BVDV und die Entwicklung der MD als auch über ein hochkonserviertes zelluläres Protein-Prozessierungssystem erbracht. The bovine viral diarrhea virus (BVDV) is known for the ability to integrate cellular sequences into its viral RNA genome, resulting in cytopathogenic (cp) variants of the virus. After translation, the cellular insertions induce an additional cleavage of the viral polyprotein. In this thesis the genomes of different BVDV isolates obtained from a single infected animal were characterized. The sequence analysis of cDNA clones and of RT-PCR fragments allowed the reconstruction of the genome structures of a set of viruses. All of them contained the identical cellular sequence, flanked by variable rearranged viral sequences. The genomes of the different virus variants had closely related structures, making it likely that in the animal a primary recombinant virus arose, that was subsequently changed by further recombination events. The cellular insertion is a fragment of light chain 3 (LC3), a module of microtubule-associated proteins. As expected, LC3 was shown to serve as a signal for proteolytic processing. The cleavage site was identified directly at the carboxyterminus of the cellular insertion by mutagenesis experiments. In vitro-translation studies with different protease inhibitors indicated that the processing likely occurs by a cystein protease of the animal host. The expression of viral polyprotein in cells of hamster, chicken, fish, and mosquito revealed that the processing system is unusually well conserved LC3 and the homologous protein Aut7p/Apg8p from yeast have essential functions in autophagy of starving cells. For this, LC3 and Aut7p/Apg8p are activated by carboxyterminal protease cleavage. One protease from bovine cells bAut2B2 was identified that was capable of processing LC3 upon in vitro cotranslation and expression in yeast cells. The expression of bAut2B2 and bAut2A another potential bovine protease could complement a yeast mutant strain defective in the specific protease Aut2p/Apg4p. In summary, these results (1) give important insights into mechanism of cytopathogenity of BVDV and the subsequent development of mucosal disease (MD) and (2) lead to the identification of a highly conserved cellular protein processing system.
