Abstract

This article discusses the development of an additively manufactured template for physician-modified endografts, which are used to treat complex aortic aneurysms. The template is designed to improve the precision of fenestrations, which are manually cut by surgeons into the endoprosthesis. The template is patient-specific and simplifies, standardizes, and accelerates the modification process. The article describes the methods used to create the template, including segmentation of the patient-specific centerline and additive manufacturing. The results show that the template can be produced within a few hours, is biocompatible and sterilizable, and has a high accuracy in fenestration. The developed method allows for the creation of a sterilized, precise, and patient-specific template within one day. [Extracted from the article]

Abstract

Perivascular space (PVS) burden is an emerging, poorly understood, magnetic resonance imaging marker of cerebral small vessel disease, a leading cause of stroke and dementia. Genome-wide association studies in up to 40,095 participants (18 population-based cohorts, 66.3 ± 8.6 yr, 96.9% European ancestry) revealed 24 genome-wide significant PVS risk loci, mainly in the white matter. These were associated with white matter PVS already in young adults (N = 1,748; 22.1 ± 2.3 yr) and were enriched in early-onset leukodystrophy genes and genes expressed in fetal brain endothelial cells, suggesting early-life mechanisms. In total, 53% of white matter PVS risk loci showed nominally significant associations (27% after multiple-testing correction) in a Japanese population-based cohort (N = 2,862; 68.3 ± 5.3 yr). Mendelian randomization supported causal associations of high blood pressure with basal ganglia and hippocampal PVS, and of basal ganglia PVS and hippocampal PVS with stroke, accounting for blood pressure. Our findings provide insight into the biology of PVS and cerebral small vessel disease, pointing to pathways involving extracellular matrix, membrane transport and developmental processes, and the potential for genetically informed prioritization of drug targets.

Item does not contain fulltext, Major depressive disorder (MDD) is associated with an increased risk of brain atrophy, aging-related diseases, and mortality. We examined potential advanced brain aging in adult MDD patients, and whether this process is associated with clinical characteristics in a large multicenter international dataset. We performed a mega-analysis by pooling brain measures derived from T1-weighted MRI scans from 19 samples worldwide. Healthy brain aging was estimated by predicting chronological age (18-75 years) from 7 subcortical volumes, 34 cortical thickness and 34 surface area, lateral ventricles and total intracranial volume measures separately in 952 male and 1236 female controls from the ENIGMA MDD working group. The learned model coefficients were applied to 927 male controls and 986 depressed males, and 1199 female controls and 1689 depressed females to obtain independent unbiased brain-based age predictions. The difference between predicted "brain age" and chronological age was calculated to indicate brain-predicted age difference (brain-PAD). On average, MDD patients showed a higher brain-PAD of +1.08 (SE 0.22) years (Cohen's d = 0.14, 95% CI: 0.08-0.20) compared with controls. However, this difference did not seem to be driven by specific clinical characteristics (recurrent status, remission status, antidepressant medication use, age of onset, or symptom severity). This highly powered collaborative effort showed subtle patterns of age-related structural brain abnormalities in MDD. Substantial within-group variance and overlap between groups were observed. Longitudinal studies of MDD and somatic health outcomes are needed to further assess the clinical value of these brain-PAD estimates.

Abstract

We have carried out meta-analyses of genome-wide association studies (GWAS) (n = 23 784) of the first two principal components (PCs) that group together cortical regions with shared variance in their surface area. PC1 (global) captured variations of most regions, whereas PC2 (visual) was specific to the primary and secondary visual cortices. We identified a total of 18 (PC1) and 17 (PC2) independent loci, which were replicated in another 25 746 individuals. The loci of the global PC1 included those associated previously with intracranial volume and/or general cognitive function, such as MAPT and IGF2BP1. The loci of the visual PC2 included DAAM1, a key player in the planar-cell-polarity pathway. We then tested associations with occupational aptitudes and, as predicted, found that the global PC1 was associated with General Learning Ability, and the visual PC2 was associated with the Form Perception aptitude. These results suggest that interindividual variations in global and regional development of the human cerebral cortex (and its molecular architecture) cascade—albeit in a very limited manner—to behaviors as complex as the choice of one’s occupation.

Abstract

We have carried out meta-analyses of genome-wide association studies (GWAS) (n = 23 784) of the first two principal components (PCs) that group together cortical regions with shared variance in their surface area. PC1 (global) captured variations of most regions, whereas PC2 (visual) was specific to the primary and secondary visual cortices. We identified a total of 18 (PC1) and 17 (PC2) independent loci, which were replicated in another 25 746 individuals. The loci of the global PC1 included those associated previously with intracranial volume and/or general cognitive function, such as MAPT and IGF2BP1. The loci of the visual PC2 included DAAM1, a key player in the planar-cell-polarity pathway. We then tested associations with occupational aptitudes and, as predicted, found that the global PC1 was associated with General Learning Ability, and the visual PC2 was associated with the Form Perception aptitude. These results suggest that interindividual variations in global and regional development of the human cerebral cortex (and its molecular architecture) cascade—albeit in a very limited manner—to behaviors as complex as the choice of one’s occupation.

Abstract

G protein-coupled receptors (GPCRs) are critical for cardiovascular physiology. Cardiac cells express >100 nonchemosensory GPCRs, indicating that important physiological and potential therapeutic targets remain to be discovered. Moreover, there is a growing appreciation that members of the large, distinct taste and odorant GPCR families have specific functions in tissues beyond the oronasal cavity, including in the brain, gastrointestinal tract and respiratory system. To date, these chemosensory GPCRs have not been systematically studied in the heart. We performed RT-qPCR taste receptor screens in rodent and human heart tissues that revealed discrete subsets of type 2 taste receptors (TAS2/Tas2) as well as Tas1r1 and Tas1r3 (comprising the umami receptor) are expressed. These taste GPCRs are present in cultured cardiac myocytes and fibroblasts, and by in situ hybridization can be visualized across the myocardium in isolated cardiac cells. Tas1r1 gene-targeted mice (Tas1r1(Cre)/Rosa26(tdRFP)) strikingly recapitulated these data. In vivo taste receptor expression levels were developmentally regulated in the postnatal period. Intriguingly, several Tas2rs were upregulated in cultured rat myocytes and in mouse heart in vivo following starvation. The discovery of taste GPCRs in the heart opens an exciting new field of cardiac research. We predict that these taste receptors may function as nutrient sensors in the heart.

Abstract

This report is intended to summarize major findings that emerged between 1992 and 1995 from the water-quality assessment of the Central Nebraska Basins Study Unit and to relate these findings to water-quality issues of regional and national concern. The information is primarily intended for those who are involved in waterresource management. Indeed, this report addresses many of the concerns raised by regulators, water-utility managers, industry representatives, and other scientists, engineers, public officials, and members of stakeholder groups who provided advice and input to the USGS during this NAWQA Study-Unit investigation. Yet, the information contained here may also interest those who simply wish to know more about the quality of water in the rivers and aquifers in the area where they live. Land use in central Nebraska appears to affect water quality significantly; streams in rangelands generally had fewer occurrences and smaller concentrations of pesticides than did streams in croplands where corn and soybeans were planted extensively. Subbasins with greater proportions of rangeland, such as the Dismal River, had negligible herbicide concentrations. The largest pesticide concentrations were in storm runoff following pesticide applications. Because some pesticide concentrations may exceed the U.S. Environmental Protection Agency’s (USEPA) drinking-water Maximum Contaminant Levels (MCLs) in storm runoff, the timing and intensity of rainfall has implications for drinking-water supplies. Pesticides in streams from storm runoff may enter alluvial aquifers as a consequence of ground-water withdrawals. Sites with degraded water chemistry commonly had degraded physical habitats as well. Streamflow regulation of the Platte River has affected water quality through habitat alterations that are deleterious to native species. Abstract

Fibrin glue is a two-component adhesive used to stop bleeding, seal wound edges and for scaffolds in tissue engineering. Autologous products result in reduced risk of contamination and immunological responses compared to commercially available fibrin glue. However, reproducibility due to patient dependent sealant properties of autologous fibrin glue preparation is low. In this study a fully automated production process for both the fibrinogen and thrombin component from small blood volumes was developed. The resulting fibrinogen concentration, thrombin activity and sealant properties of the fibrin glue were determined. The fully automated and closed production system proved to be a promising tool for fast and easy production of autologous fibrin glue with an effective adhesive quality.

Eighteen of the 86 variables examined, including riparian and instream habitat, macroinvertebrate communities, and water/sediment chemistry, were significantly correlated with percent impervious area. Variables related to channel condition, instream substrate, water chemistry, and residential and transportation right-of-way land uses were identified by principal components analysis as significant factors separating site groups. Detrended canonical correspondence analysis indicated that the macroinvertebrate communities responded to an urbanization gradient closely paralleling the percent of impervious area within the subbasin. A sliding regression analysis of variables significantly correlated with percent impervious area revealed 8 variables exhibiting threshold responses that correspond to a mean of 4.4 – 5.8% impervious area, much lower than mean values reported in other, similar investigations. As contributing factors to a subbasin's impervious area, storm drains and roads appeared to be important elements influencing the degradation of water quality with respect to the biota. Abstract

Unlabelled: Phaeochromocytomas usually occur sporadically but may be associated with dominant inherited cancer syndromes such as multiple endocrine neoplasia type 2 (MEN 2), von Hippel-Lindau disease (VHL) and type 1 neurofibromatosis. We report on a boy presenting at age 8 years with an isolated benign phaeochromocytoma of the left adrenal. Three years later a second adrenal phaeochromocytoma was diagnosed on the right side and removed. His family history was negative. Genetic analysis did not show a mutation in the MEN 2 susceptible proto-oncogene rearranged during transfection; however, we found a germline missense mutation in the VHL gene (nucleotide 695 G to A transversion) which has been described only twice before in the literature. Both parents had normal (wild type) VHL copies indicating that our patient had a de novo germline VHL mutation. Careful clinical evaluation of the patient at 18 years did not reveal any other manifestations of VHL disease.Conclusion: Carriers of von Hippel-Lindau germline mutations can present with a form fruste of von Hippel-Lindau disease presenting initially with unilateral phaeochromocytoma and therefore mutation analysis should be carried out. Abstract

After the nuclear reactor accident of Chernobyl, the “Integrated Measurement and Information System (IMIS) for Monitoring the Environmental Radioactivity and Detecting Emissions from Nuclear Plants was implemented in Germany. IMIS is a nationwide comprehensive measuring system which permanently monitors the radioactivity in all important environment media in the whole federal territory. At approximately 40 sites, the activity concentration of radioactive substances is measured in air and precipitations. At least 14 of them are responsible for trace monitoring of radionuclides in the air. The legal bases of IMIS prescribe regular inter-laboratory comparison analyses in cooperation with the Physikalisch-Technische Bundesanstalt (PTB), with the use of reference materials prepared by the Federal Coordinating Laboratories. In order to fulfil this requirement in the field of trace survey measurements in ground-level air, the Federal Office for Radiation Protection (“Bundesamt für Strahlenschutz”, BfS) and the PTB have conducted a comparison with real, dust-loaded reference filters in 2005. The comparison was organized within the framework of a cooperation of trace survey stations from Austria, Germany and Switzerland. The paper describes the preparation of the real, dust-loaded reference filters, the procedure for spiking the filters with the activity standard solution containing ²²Na, ⁸⁸Y, ⁸⁹Sr, ⁹⁰Sr, ¹²⁵Sb, ¹³³Ba, ¹³⁴Cs, and ²⁴¹Am. Some results are discussed and conclusions are given.
