1. There is a common agreement that, to maximise our understanding of trait-based community assembly, traits related to different organs should be considered as they relate to different ecological niche axes. Floral traits, in particular, have been suggested to play an important role in the plant community assembly along environmental gradients as they determine the reproductive success, one of the key functions in plants. However, the role of floral traits in community assembly research remains largely unverified empirically. 2. Using a large and novel dataset on plant traits and plant communities, we analysed the variability of six floral traits of 139 herbaceous species in 21 sites located along an elevation gradient (2,000–4,000 m) in the western Himalaya. The variability of floral traits along the gradient was analysed using community-weighted mean (CWM) trait values and functional diversities (FD) calculated for each study community. 3. The CWMs showed that with increasing elevation, flowering began later, and flowers tended to be larger with decreasing specific flower area (SFA). Along with the convergence in the onset of flowering and SFA (i.e., lower FD at high elevation sites), these patterns suggest that abiotic filtering and plant-pollinator interactions could affect the floral trait composition of the communities. Most likely, increasing low-temperature stress towards high-elevation sites selected for late-floweringspecies that produce flowers with larger display areas. The low abundance and activity of pollinators at high elevation could also explain why these traits were selected in the communities studied. Delayed flowering with increasing elevation might facilitate the phenological overlap of plants and their pollinators, as pollinator activity at higher elevation peaks in the second half of the vegetation period. The dominance of a species with low SFA and larger display area in high-altitude communities could be attributable to increased flower longevity and attraction of pollinators, respectively, to maximise pollination success under pollinator scarcity. 4.Synthesis. Our study provides empirical support for the recent argument that floral traits contribute considerably to the assembly of plant communities along environmental gradients. Thus, such traits should be included in community assembly research as they represent key ecological functions for reproduction.

Changes in phenology induced by climate change occur across the globe with important implications for ecosystem functioning and services, species performance and trophic interactions. Much of the work on phenology, especially leaf out and flowering, has been conducted on woody plant species. Less is known about the responses in phenology of herbaceous species induced by global change even though they represent a large and important part of biodiversity worldwide. A globally coordinated research effort is needed to understand the drivers and implications of such changes and to predict effects of global change on plant species phenology and related ecosystem processes. Here, we present the rationale of the PhenObs initiative-botanical gardens as a global phenological observation network. The initiative aims to collect data on plant phenology in botanical gardens which will be used alongside information on plant traits and site conditions to answer questions related to the consequences of global change: What is the variation in plant phenology in herbaceous species across the growing season and in response to changes in climate? How can plant phenology be predicted from species' trait composition, provenance, position and extent of the distribution range and species' phylogeny? What are the implications of this variation with respect to species performance and assembly, biotic interactions (e.g. plant-pollinator interactions) as well as ecosystem processes and services under changing land use and climate? Here, we lay out the development of a straightforward protocol that is appropriate for monitoring phenology across a vast diversity of growth forms of herbaceous species from various habitats and geographical regions. To focus on a key number of stages necessary to capture all aspects of plant species phenology, we analysed associations between 14 phenological stages. These data were derived from a 2-year study on 199 species in four German botanical gardens. Based on the relationships of the phenological stages, we propose to monitor three vegetative stages ('initial growth', 'leaves unfolding' and 'senescence') and two reproductive stages ('flowers open' and 'ripe fruits') to fully capture herbaceous species phenology. Purpose To evaluate morphological alterations of meibomian glands (MGs) in the dry anophthalmic socket syndrome (DASS). Methods Fifteen unilateral anophthalmic patients wearing cryolite glass prosthetic eyes were enrolled. All patients with clinical blepharitis or other significant eyelid abnormalities were excluded. In vivo laser scanning confocal microscopy (LSCM) of the MGs in the lower eyelids both on the anophthalmic side and the healthy fellow eye was performed to quantify acinar unit density, acinar unit diameter, acinar unit area, meibum secretion reflectivity, the inhomogeneous appearance of the glandular interstice, and inhomogeneous appearance of the acinar walls. Results The lower eyelids of the anophthalmic sockets revealed a significant reduction of the acinar unit density (p = 0.003) as well as a significantly more inhomogeneous appearance of the periglandular interstices (p = 0.018) and the acinar unit walls (p = 0.015) than the healthy fellow eyelid. However, there were no significant differences regarding the acinar unit diameter, acinar unit area, and meibum secretion reflectivity of the MGs on the anophthalmic side compared to the healthy fellow eyelid (p ≥ 0.05, respectively). Conclusions The eyelids of anophthalmic sockets without clinical blepharitis demonstrate a reduced density of MG acinar units and a more inhomogeneous appearance of the periglandular interstices and the acinar unit walls. This can cause meibomian gland dysfunction contributing to DASS and suggests early treatment of these symptomatic patients, even in the clinical absence of any blepharitis signs.

Ecological theory postulates that plant trait research should consider multiple traits related to different organs and/or ontogenetic stages as such traits represent different ecological niche axes. We tested the seasonal variability of frost resistance and photosynthesis characteristics, namely net photosynthetic rate at saturating irradiance and ambient atmospheric CO2 concentrations, maximum carboxylation rate, and measurements based on the JIP-test, namely maximum quantum yield of PSII and the performance index. Additionally, we measured leaf functional traits, namely specific leaf area, leaf dry matter content, stomatal pore area index, and chlorophyll content. We assessed all traits weekly on two evergreen species Berberis vulgaris and Quercus × hispanica as well as the two summer green species Betula pendula and Quercus pubescens. We found that there was a high seasonal variability in all traits except stomatal pore area index. In a multivariate analysis, we could show trade-offs between growth rates and leaf thickness on the first axis, as well as resistance and performance on the second axis. The findings help understand ecophysiological processes and fine-tune biosphere models.

Patterns of flowering phenology, i.e. first and last flowering day (FFD and LFD) and flowering duration (FD) govern plant pollination and reproduction. Most research has focused on FFD whereas LFD, FD and second flowering events were neglected although they are crucial events. To assess whether flowering patterns change species-specifically with changing abiotic conditions and whether these are related to leaf persistence, reproduction and pollination syndrome, we monitored flowering phenology (including also second flowering events) of 29 herbaceous species along two elevational gradients ranging from 700 to 1800 m a.s.l. in two consecutive years. FFD was delayed with increasing elevation. LFD followed two alternative strategies: species which ceased flowering early in the season delayed LFD with increasing elevation, species which ceased flowering late did not change LFD. FD decreased with increasing elevation in most species. The timing of flowering phenology had strong influences on the intensity of change along the elevational gradient and other stages of flowering phenology. Many species showed a second flowering event which occurred irrespective of elevation, suggesting a link to unsuccessful pollination rather than temperature. Life history strategies helped explain patterns of flowering phenology and a species’ ability to track changes in abiotic conditions, e.g. evergreen species started to flower earlier than summer green species and insect-pollinated species were able to expand FD most. These findings give insight into species-specific changes in phenological patterns and thus plant performance with changing environment. They should be considered when assessing the consequences of environmental change at both the community and ecosystem level.

Ecosystem functions are heavily dependent on the functional composition of the plant community, i.e., the functional traits of plants forming the community. This, on the one hand, depends on plant occurrence, but on the other hand, depends on the intraspecific variability of functional traits of the species, which are influenced by climate and nutrient availability and affected by plant-plant interactions. To illustrate that, we studied the effects of drought and nitrogen addition (+ N), two important abiotic variables which are changing with ongoing global change, as well as their combined effect on the functional responses of grassland communities in semi-arid environments of Northern Africa comprising of natural and invasive species. We conducted an experiment where we planted three native species and one invasive plant species in artificial communities of five individuals per species per plot. We exposed these communities to four different treatments: a drought treatment, an N-addition treatment, the combination between drought and N-addition, as well as a control. Phenology has emerged as key indicator of the biological impacts of climate change, yet the role of functional traits constraining variation in herbaceous species’ phenology has received little attention. Botanical gardens are ideal places in which to investigate large numbers of species growing under common climate conditions. We ask whether interspecific variation in plant phenology is influenced by differences in functional traits. 2. We recorded onset, end, duration and intensity of initial growth, leafing out, leaf senescence, flowering and fruiting for 212 species across five botanical gardens in Germany. We measured functional traits, including plant height, absolute and specific leaf area, leaf dry matter content, leaf carbon and nitrogen content and seed mass and accounted for species’ relatedness. 3. Closely related species showed greater similarities in timing of phenological events than expected by chance, but species' traits had a high degree of explanatory power, pointing to paramount importance of species’ life-history strategies. Taller plants showed later timing of initial growth, and flowered, fruited and underwent leaf senescence later. Large-leaved species had shorter flowering and fruiting durations. 4. Taller, large-leaved species differ in their phenology and are more competitive than smaller, small-leaved species. We assume climate warming will change plant communities’ competitive hierarchies with consequences for biodiversity.

The diagnostic process of inherited retinal dystrophies (IRD) is impeded by their low prevalence and the variability of the clinical presentations; however, for patients a valid diagnosis is vital for future planning and evaluating the potential of an appropriate early treatment to delay disease progression.Aim of the current study was to outline the patients' journeys until they receive the final diagnosis. This should help uncover diagnostic shortcomings and highlight potential for improvement with respect to the use of genetic diagnostic testing.Data were collected by questionnaires and an online survey conducted by the self-help association PRO RETINA Deutschland e. V. among patients with IRD. Data were analyzed by descriptive statistics.From 15 March to 22 April 2021, 183 questionnaires were completed and 42 online interviews conducted. The surveyed population consisted of 48% female patients, mean age was 55 years and first symptoms occurred at a mean age of 22 years. On average about 14 years passed from first symptoms until final diagnosis. Only 66% of the patients reported that they had received at least 1 diagnostic genetic testing; less than half of the patients (47%) received genetic counseling. The huge majority of patients (85%) would be interested in gene therapy.From the perspective of affected patients, a shortening of the time to diagnosis, the use of molecular genetic testing and the offer of genetic counseling are important to improve patient care for patients with IRD.HINTERGRUND: Die Diagnose erblicher Netzhautdystrophien ist aufgrund der niedrigen Prävalenz und der Variabilität der klinischen Präsentation schwierig. Dies sollte dazu dienen, mögliche Schwachstellen in der Diagnostik und Versorgung aufzudecken und Verbesserungspotenziale insbesondere im Hinblick auf die Nutzung genetischer Diagnostik aufzuzeigen.Die Daten wurden mittels Umfragebögen und Online-Interviews von der Selbsthilfevereinigung PRO RETINA Deutschland e. V. unter Patienten mit erblichen Netzhautdystrophien erhoben. Daten wurden mittels deskriptiver Statistik ausgewertet.Im Zeitraum vom 15.03. bis zum 22.04.2021 wurden 183 Umfragebögen ausgefüllt und 42 Online-Interviews durchgeführt. Die Umfragepopulation bestand aus 48 % weiblichen Teilnehmern, das Durchschnittsalter betrug 55 Jahre, erste Symptome traten im Schnitt im Alter von 22 Jahren auf. Von den ersten Symptomen bis zur finalen Diagnose vergingen etwa 14 Jahre. Lediglich 66 % der Befragten gaben an, dass mindestens 1 Gentest bei Ihnen durchgeführt wurde; weniger als die Hälfte (47 %) erhielt eine humangenetische Beratung. Die überwiegende Mehrheit (85 %) würde eine Gentherapie erwägen.Aus der Patientenperspektive sind eine Verkürzung des Diagnoseweges, das Angebot einer molekulargenetischen Diagnostik sowie einer humangenetischen Beratung wesentlich für die Verbesserung der Versorgung von Patienten mit erblichen Netzhautdystrophien.

Departamento de Medio Ambiente y Agronomía​ (INIA), Remote sensing capabilities to monitor evergreen broadleaved vegetation are limited by the low temporal variability in the greenness signal. With canopy greenness computed from digital repeat photography (PhenoCam), we investigated how canopy greenness related to seasonal changes in leaf age and traits as well as variation of trees’ water fluxes (characterized by sap flow and canopy conductance). The results showed that sprouting leaves are mainly responsible for the rapid increase in canopy green chromatic coordinate (GCC) in spring. We found statistically significantly differences in leaf traits and spectral properties among leaves of different leaf ages. Specifically, mean GCC of young leaves was 0.385 ± 0.010 (mean ± SD), while for mature and old leaves was 0.369 ± 0.003, and 0.376 ± 0.004, respectively. Thus, the temporal dynamics of canopy GCC can be explained by changes in leaf spectral properties and leaf age. Sap flow and canopy conductance are both well explained by a combination of environmental drivers and greenness (96% and 87% of the variance explained, respectively). In particular, air temperature and vapor pressure deficit (VPD) explained most of sap flow and canopy conductance variance, respectively. Besides, GCC is an important explanatory variable for variation of canopy conductance may because GCC can represent the leaf ontogeny information. We conclude that PhenoCam GCC can be used to identify the leaf flushing for evergreen broadleaved trees, which carries important information about leaf ontogeny and traits. Frost resistance (FR) is a highly adaptive trait and important for plant performance, survival and distribution. While overall seasonal changes in the frost resistance of herbaceous species are well documented, knowledge of the variability during the growth period is scarce. Responses could be expected due to differences in temperature yet investment in frost resistance might be at the expense of plant performance. To analyse temporal and spatial (i.e. same date but differing temperatures) variability, FR of leaves of six herbaceous species on five sampling dates were assessed along an elevational gradient in the northern limestone Alps. We used chlorophyll fluorescence techniques to calculate the lethal temperature of 90% of the population (LT90) thereof. To test the association with plant performance, we measured eco-morphological leaf traits (specific leaf area (SLA), leaf dry matter content, leaf phosphorous and magnesium content as well as stomatal pore area index (SPI)) in parallel. We found that FR as well as leaf traits exhibit a strong temporal variation whereas spatial variability was low. When analysing the relationship of FR to leaf traits we found that SLA as a proxy of growth rate was negatively associated with FR indicating a trade-off between growth and resistance, whereas SPI showed a positive relationship to FR. This finding gives further insight into the variability of traits and will help to improve predictions concerning plant performance and distribution under changing climate regimes.

QUESTIONS: Are the vegetation structure and soil properties of fallows similar to adjacent species‐rich Mediterranean steppe communities 35 years after abandonment? Is there a spontaneous redevelopment towards the original steppe vegetation after reintroduction of the traditional grazing system? Can differences in functional trait composition be identified according to different land‐use types? Do traits of selected species affect the ability to recolonize fallow land? LOCATION: Mediterranean steppe of La Crau (Southeastern France). METHODS: We conducted 80 vegetation surveys and soil analyses in the steppe and adjacent fallow land in six locations in La Crau in 2015; for three locations, data from 2001 were included. To compare the functional composition of steppe vs fallow land, community‐weighted means (CMW) of specific leaf area (SLA), leaf dry matter content (LDMC) and canopy height were calculated. To test whether trait variability is associated with the recolonization success of 12 selected target species, we measured SLA, LDMC, canopy height, plant width and aboveground biomass on eight replicate individuals per site and species. RESULTS: Fallow land was characterized by lower species richness and elevated phosphorus and potassium content in the soil. Comparing vegetation releves between 2001 and 2015 revealed that the fallows were developing towards steppe vegetation. CWMs of SLA were significantly lower and CWM of LDMC higher in steppe than in fallow land. There was no similarity in trait composition between steppe species that successfully recolonized the fallow‐land community, and no evidence that intraspecific trait variability is associated with recolonization success of these species. CONCLUSIONS: Even after decades of abandonment and reintroduction of the grazing system, the vegetation structure, soil properties and plant functional trait composition of fallows differ from those of adjacent steppe communities. These results illustrate the need for active restoration to accelerate the recovery of former arable land to the target steppe communities.

We tested whether cheap and quick chlorophyll (Chl) fluorescence can be used in ecophysiological field studies as proxies for gas-exchange measurements. We measured net photosynthetic rate at saturating irradiance and ambient atmospheric CO2 concentrations (PNsat), maximum carboxylation rate (Vcmax), maximum quantum yield of PSII (Fv/Fm), the performance index (PIabs), leaf nitrogen (Narea), and carbon isotope discrimination (Δ13C) within four herbaceous species along two elevational gradients. We analysed the relationship between Chl fluorescence and gas-exchange parameters and their link to indirect assessment of plant performance via ecophysiological traits. Fv/Fm showed no relationship to PNsat and only weak relationships to Vcmax. PIabs was positively related to PNsat and Vcmax. PIabs, PNsat, and Vcmax were positively associated with Narea and negatively to Δ13C, whereas Fv/Fm showed no relationship to Narea and a positive to Δ13C. Thus, PIabs might be suitable to characterize the photosynthetic activity when aiming on large numbers of samples.

Data is central in almost all scientific disciplines nowadays. Furthermore, intelligent systems have developed rapidly in recent years, so that in many disciplines the expectation is emerging that with the help of intelligent systems, significant challenges can be overcome and science can be done in completely new ways. In order for this to succeed, however, first, fundamental research in computer science is still required, and, second, generic tools must be developed on which specialized solutions can be built. In this paper, we introduce a recently started collaborative project funded by the Carl Zeiss Foundation, a virtual manufactory for digitization in the sciences, the “Werkstatt”, which is being established at the Michael Stifel Center Jena (MSCJ) for data-driven and simulation science to address fundamental questions in computer science and applications. The Werkstatt focuses on three key areas, which include generic tools for machine learning, knowledge generation using machine learning processes, and semantic methods for the data life cycle, as well as the application of these topics in different disciplines. Core and pilot projects address the key aspects of the topics and form the basis for sustainable work in the Werkstatt.

Background : Commercially harvested wild medicinal plants are often subject to overharvesting. This study aims to examine the ecological requirements, collection status, harvest practices, sustainability of harvest and trade chain of five medicinal plant species and assesses the socio-economic importance of these medicinal plants for local households. Methods : We studied the abundance of five selected medicinal plant species in the surrounding areas of five selected villages. Harvest and trade related information was recorded by interviewing plant collectors and middlemen as well as pharmacies in Germany. For the ecological analyses concerning the occurrence of plants we performed ANOVA followed by LSD Post-Hoc tests. Correlations between profit from collection of medicinal plants and income per household in the five villages were calculated with the Pearson Correlation Coefficient. Results : The results showed that (1) harvest practices are destructive for individual plants, (2) economic contribution of plant harvest varies between villages and species and (3) the amount of cash earned is negatively related to average per capita income in the village. Conclusion : A management plan for sustainable harvest or cultivation is recommended to ensure the future existence of the plant species as well as the sustainability of market supply. Anthropogenic nitrogen (N) deposition and resulting differences in ecosystem N and phosphorus (P) ratios are expected to impact photosynthetic capacity, that is, maximum gross primary productivity (GPP). However, the interplay between N and P availability with other critical resources on seasonal dynamics of ecosystem productivity remains largely unknown. In a Mediterranean tree–grass ecosystem, we established three landscape-level (24 ha) nutrient addition treatments: N addition (NT), N and P addition (NPT), and a control site (CT). We analyzed the response of ecosystem to altered nutrient stoichiometry using eddy covariance fluxes measurements, satellite observations, and digital repeat photography. A set of metrics, including phenological transition dates (PTDs; timing of green-up and dry-down), slopes during green-up and dry-down period, and seasonal amplitude, were extracted from time series of GPP and used to represent the seasonality of vegetation activity. The seasonal amplitude of GPP was higher for NT and NPT than CT, which was attributed to changes in structure and physiology induced by fertilization. PTDs were mainly driven by rainfall and exhibited no significant differences among treatments during the green-up period. Yet, both fertilized sites senesced earlier during the dry-down period (17–19 days), which was more pronounced in the NT due to larger evapotranspiration and water usage. Fertilization also resulted in a faster increase in GPP during the green-up period and a sharper decline in GPP during the dry-down period, with less prominent decline response in NPT. Overall, we demonstrated seasonality of vegetation activity was altered after fertilization and the importance of nutrient–water interaction in such water-limited ecosystems. With the projected warming-drying trend, the positive effects of N fertilization induced by N deposition on GPP may be counteracted by an earlier and faster dry-down in particular in areas where the N:P ratio increases, with potential impact on the carbon cycle of water-limited ecosystems., The authors acknowledge the Alexander von Humboldt Foundation for supporting this research with the Max-Planck Prize to Markus Reichstein. Phenological responses to changing temperatures are known as “fingerprints of climate change,” yet these reactions are highly species specific. To assess whether different plant characteristics are related to these species‐specific responses in flowering phenology, we observed the first flowering day (FFD) of ten herbaceous species along two elevational gradients, representing temperature gradients. On the same populations, we measured traits being associated with (1) plant performance (specific leaf area), (2) leaf biochemistry (leaf C, N, P, K, and Mg content), and (3) water‐use efficiency (stomatal pore area index and stable carbon isotopes concentration). We found that as elevation increased, FFD was delayed for all species with a highly species‐specific rate. Populations at higher elevations needed less temperature accumulation to start flowering than populations of the same species at lower elevations. Surprisingly, traits explained a higher proportion of variance in the phenological data than elevation. Earlier flowering was associated with higher water‐use efficiency, higher leaf C, and lower leaf P content. In addition to that, the intensity of shifts in FFD was related to leaf N and K. These results propose that traits have a high potential in explaining phenological variations, which even surpassed the effect of temperature changes in our study. Therefore, they have a high potential to be included in future analyses studying the effects of climate change and will help to improve predictions of vegetation changes.

Lymphangiogenesis is essential for fluid homeostasis in vascularized tissues. In the normally avascular cornea, however, pathological lymphangiogenesis mediates diseases like corneal transplant rejection, dry eye disease, and allergy. So far, a physiological role for lymphangiogenesis in a primarily avascular site such as the cornea has not been described. Using a mouse model of perforating corneal injury that causes acute and severe fluid accumulation in the cornea, we show that lymphatics transiently and selectively invade the cornea and regulate the resolution of corneal edema. Pharmacological blockade of lymphangiogenesis via VEGFR-3 inhibition results in increased corneal thickness due to delayed drainage of corneal edema and a trend towards prolonged corneal opacification. Notably, lymphatics are also detectable in the cornea of a patient with acute edema due to spontaneous Descemet´s (basement) membrane rupture in keratoconus, mimicking this animal model and highlighting the clinical relevance of lymphangiogenesis in corneal fluid homeostasis. Together, our findings provide evidence that lymphangiogenesis plays an unexpectedly beneficial role in the regulation of corneal edema and transparency. This might open new treatment options in blinding diseases associated with corneal edema and transparency loss. Furthermore, we demonstrate for the first time that physiological lymphangiogenesis also occurs in primarily avascular sites.

Stomata enable plants to balance uptake of CO 2 with water loss via transpiration. Previous studies have shown that stomatal size and density trade-off with one another and are related to a suite of environmental factors (light, atmospheric CO 2 concentrations, moisture and temperature) which influence gas exchange. We examined the hypothesis that the habitat preferences of species are reflected in stomatal traits. Thus, we examined the relation of stomatal size and density of 36 herbaceous species along elevational gradients in relation to Ellenberg indicator values for light and moisture. The light value is a proxy for the transpiration demand due to exposure to radiation and air mass exchange, the moisture value reflects water supply. Additionally we measured carbon isotope discrimination (Δ 13 C), a proxy for intrinsic water use efficiency and internal CO 2 concentration. Stomatal size changed in parallel on both sides of a leaf, whereas changes in density differed between sides depending on species identity. There was an increase in absolute variation of sizes and densities with increasing mean size and density respectively, but not in relative variation. Species with few but large stomata tended to adjust stomatal size across environmental gradients, whereas species with small but many stomata mainly adjusted stomatal density. A higher Ellenberg indicator value for light and a lower value for moisture was associated with equal distribution of stomata between leaf sides. The carbon isotope discrimination data indicated that amphistomatic species, which illustrated a preference for high radiation and high air mass exchange as well as for dry habitats, had higher water use efficiency. We conclude that stomatal traits such as size, density or the distribution between the two sides of the leaf are indicators of how species optimize carbon uptake and balance water loss and radiation gain.

Parry-Romberg syndrome is a rare disease characterized by slowly progressive atrophy affecting facial subcutaneous tissues, including the underlying muscles and osteocartilaginous structures. Various periocular, ocular, and neuro-ophthalmological manifestations have been described in Parry-Romberg syndrome. The most common periocular disorders include enophthalmos, eyelid, and orbit alterations. The most frequent ocular disorders include corneal and retinal changes, and the most common neuro-ophthalmological disorders involve optic nerve, ocular motor and pupillary dysfunction. Besides the characteristic facial abnormalities, systemic manifestations may occur, including neurologic, dermatologic, cardiac, endocrine, infectious, orthodontic, and maxillofacial disorders. So far, mainly brief case reports describe these ophthalmological findings. Therefore, we summarize the ocular, periocular, and neuro-ophthalmological findings in detail, describe diagnostic modalities, and outline therapeutic options.

Vascular endothelial growth factor A (VEGF) induces angiogenesis and vascular hyperpermeability in ocular tissues and is therefore a key therapeutic target for eye conditions in which these processes are dysregulated. In contrast, the therapeutic potential of VEGF’s neurotrophic roles in the eye has remained unexploited. In particular, it is not known whether modulating levels of any of the 3 major alternatively spliced VEGF isoforms might provide a therapeutic approach to promote neural health in the eye without inducing vascular pathology. Here, we have used a variety of mouse models to demonstrate differences in overall VEGF levels and VEGF isoform ratios across tissues in the healthy eye. We further show that VEGF isoform expression was differentially regulated in retinal versus corneal disease models. Among the 3 major isoforms — termed VEGF120, VEGF164, and VEGF188 — VEGF188 was upregulated to the greatest extent in injured cornea, where it was both necessary and sufficient for corneal nerve regeneration. Moreover, topical VEGF188 application further promoted corneal nerve regeneration without inducing pathological neovascularization. VEGF isoform modulation should therefore be explored further for its potential in promoting neural health in the eye., The alternative VEGF splice form VEGF188 promotes nerve regeneration in corneal injury and may provide a therapeutic to treat corneal blindness.

Traits are widely used to detect and explain responses of ecosystem processes to environmental changes. Various studies use trait data from databases, often providing one value per trait and species, neglecting intraspecific trait variability along spatio-temporal gradients. Handbooks for standardized trait measurements claim that traits should be measured at an ‘optimal’ stage, which is typically defined to be reached when the plants are in full blossom. However, it is unclear whether this method is appropriate. The main aim of this study was to quantify the extent to which trait values vary with season and phenology, a type of variation that has been overlooked so far relative to other sources of intraspecific variation. Further, we aimed to investigate whether species rankings remain consistent throughout the year. From April to November 2012, we monitored seven leaf traits [specific leaf area SLA, leaf dry matter content, chlorophyll fluorescence parameters (Fv/Fm, performance-index PI], stomatal density, stomatal size and the stomatal pore area index SPI) of 15 summer green woody species weekly under controlled conditions. In parallel, we recorded phenological stages. The results showed that all traits varied significantly throughout the year in a species-specific manner. We detected trait relationships with vegetative but not with flowering phenology. Species rankings were inconsistent throughout the season in all traits. We concluded that the seasonal timing of trait measurements is crucial. Most notably SLA, Fv/Fm and stomatal size were the most robust traits in terms of small intraspecific and large interspecific variation and showed largely consistent species rankings across seasons.

Inflammatorische Prozesse des Auges gehören zu den häufigsten Entitäten in der Augenheilkunde. Die Anwendung antientzündlicher Substanzen ist daher für den Behandler eine zentrale therapeutische Option. Hierbei kommen zumeist aufgrund der guten Zugänglichkeit des Auges topische Applikationen zur Anwendung. Substanzgruppen wie Antiallergika, nichtsteroidale Antirheumatika, Glukokortikoide, Calcineurininhibitoren, Serumaugentropfen sowie neuere, noch in der Entwicklung befindliche Substanzen werden spezifisch sowohl in schmalen als auch in breiten Indikationsspektren eingesetzt, weshalb die Kenntnis der Wirkmechanismen und der potenziellen Nebenwirkungen von großer Bedeutung ist. Dieser Artikel soll einen Überblick über die derzeitig verfügbaren antiinflammatorisch wirksamen Substanzen geben, die topisch verabreicht werden können. Hierbei wird auf eine detaillierte Anwendungsbeschreibung (Dosis, Applikationshäufigkeit, Dauer usw.) in Bezug auf die einzelnen Krankheitsbilder bewusst verzichtet. Ziel ist es, dem Leser pharmakologisches Basiswissen mit direktem ophthalmologischen Bezug zu vermitteln.

Stomata are mediators of gas exchange and thus important for photosynthesis and plant performance. The aim of this study was to analyze the ecological explanatory power of the stomatal pore area index (SPI) calculated via stomatal size and density. We studied the SPI on sun leaves of 22 herbaceous species on 22 study sites being distributed along two elevational gradients in the northern Alps ranging from 700 to 1800 m a.s.l.. We analyzed its correlation with other functional traits related to plant performance namely specific leaf area (SLA), area-based leaf nitrogen and carbon (N area and C area, respectively) as well as carbon discrimination Δ13C within as well as between species. On a subset of four species we also measured light-saturated net photosynthetic rate at ambient CO2 concentration (A sat) and stomatal conductance on all sites. We found that SPI was positively correlated with A sat, yet the relation was weaker than expected. The reaction of SPI along the elevational gradients was highly species-specific and related to variations in other investigated leaf traits. The relationship with functional traits, however, differed between the inter- and intraspecific level in strength and direction. SPI was positively related to N area and C area and negatively with SLA and Δ13C for most species. However, we found no significant relation considering species mean values for Δ13C and N area. The relation of SPI to SLA was the most consistent displaying no difference when comparing the relation between and within species. This research shows that different processes may act on different organizational levels leading to the detected differences in trait–trait correlations on the inter- and intraspecific levels. It may have important consequences also for macroecological and modelling studies.

The role of IL-10, a primarily anti-inflammatory cytokine, in the regulation of inflammatory lymphangiogenesis is undetermined. Herein, we show that IL-10 modulates corneal lymphangiogenesis and resolution of inflammation. IL-10 was not expressed in healthy corneas but was up-regulated in inflamed corneas by infiltrating macrophages. Macrophages up-regulated the expression of prolymphangiogenic vascular endothelial growth factor-C upon stimulation with IL-10. Consistently, corneal inflammation resulted in reduced expression of vascular endothelial growth factor-C and decreased corneal lymphangiogenesis in IL-10-deficient mice (IL-10(-/-)). The effect of IL-10 on lymphangiogenesis was indirect via macrophages, because IL-10 did not directly affect lymphatic endothelial cells. The expression of proinflammatory cytokines and the numbers of infiltrating macrophages increased and remained elevated in inflamed corneas of IL-10(-/-) mice, indicating that IL-10 deficiency led to more severe and prolonged inflammation. The corneal phenotype of IL-10 deficient mice was mimicked in mice with conditional deletion of Stat3 in myeloid cells (lysozyme M Cre mice Stat3(fl/fl) mice), corroborating the critical role of macrophages in the regulation of lymphangiogenesis. Furthermore, local treatment with IL-10 promoted lymphangiogenesis and faster egress of macrophages from inflamed corneas. Taken together, we demonstrate that IL-10 indirectly regulates inflammatory corneal lymphangiogenesis via macrophages. Reduced lymphangiogenesis in IL-10(-/-) and lysozyme M Cre Stat3(fl/fl) mice is associated with more severe inflammatory responses, whereas IL-10 treatment results in faster resolution of inflammation. IL-10 might be used therapeutically to terminate pathological inflammation.

In der Rekonstruktion groser durchgreifender Oberliddefekte hat sich die Bruckenlappenplastik nach Cutler-Beard etabliert. Ziel dieser Studie ist es, unsere Erfahrungen mit Technik, Komplikationen und Ergebnissen dieser Kutaneomuskulokonjunktivallappenplastik nach Tumorexzision darzustellen. Bei 18 Patienten wurde ein groser durchgreifender Oberliddefekt nach Tumorresektion mittels Cutler-Beard-Bruckenlappenplastik des Unterlides versorgt. Vier Patienten erhielten zusatzlich Spendersklera, 1 Patient kontralateralen Tarsus als Lidstabilisator. Chirurgisch erfolgreich konnte der Eingriff bei allen Patienten durchgefuhrt werden ohne Verletzung der Unterlidbrucke oder Infektion. Bei 3 der 13 Plastiken ohne zusatzlichen Lidstabilisator (23 %) kam es zu einem Oberlidentropium, das einen Revisionseingriff mit sekundarer Skleraimplantation erforderte. Die 5 primar mit Sklera- oder Tarsustransplantaten rekonstruierten Oberlider entwickelten keine Entropiumstellung. Die Cutler-Beard-Bruckenlappenplastik, die im Falle besonders ausgedehnter Defekte (> 75 % der Lidlange) mit der Transplantation von Stabilisationsersatzgewebe kombiniert werden kann, ist eine verlassliche Methode zur Rekonstruktion groser durchgreifender Oberliddefekte nach Tumorexzision.

Chronic inflammatory demyelinating polyneuropathy (CIDP) is an autoimmune-mediated peripheral neuropathy with multifocal involvement. Reliable biomarkers for diagnosis, disease progression, and treatment response remain to be developed. We assessed the utility of corneal confocal microscopy (CCM) as a diagnostic marker for CIDP in 16 patients. CCM parameters including corneal nerve fiber density (NFD), nerve fiber length, number of main nerve trunks, number of nerve branches, nerve tortuosity, and dendritic cell density (DCD) were compared to those from 15 healthy controls and correlated with clinical and electrophysiological findings. CIDP patients had a significantly lower corneal NFD compared to healthy controls. The total nerve fiber length and the number of nerve branches were significantly decreased, whereas nerve tortuosity was increased in patients with CIDP. There was no positive correlation between corneal NFD and clinical or electrophysiological assessments. The average DCD was not significantly different in CIDP patients and controls. CCM measures suggest damage to small sensory afferents in the cornea in CIDP patients. Further studies are needed to compare different neuropathic conditions and to explore longitudinal changes of CCM parameters.

To analyze potential alterations in corneal nerve morphology and function in different stages of Fuchs’ endothelial corneal dystrophy (FECD). Thirty eyes with FECD underwent in vivo confocal microscopy using the Heidelberg Retina Tomograph II (HRT II; Heidelberg Engineering, Heidelberg, Germany) and the Rostock Cornea Module (RCM) to quantify the morphology of the central subbasal corneal nerve plexus (total nerve length, total nerve number, number of main nerve trunks, number of nerve branches) as well as esthesiometry (using the Cochet-Bonnet esthesiometer) of the central cornea to determine central corneal sensation as a measure of nerve function. Findings were correlated with an age-matched control group of 30 healthy individuals. Comparisons to biomicroscopical stage of FECD, visual acuity and central corneal thickness were performed using Spearman correlation. The VEGF-A family plays a crucial role in the induction of pathological corneal neovascularization. The role of the different VEGF-A isoforms during lymphangiogenesis is only little-known. Current anti-angiogenic therapies in the eye and other organs inhibit all VEGF-A isoforms, and have effects on both blood and lymphatic vessels. Here we investigate whether selective targeting of the isoform VEGF 165 is able to inhibit corneal lymphangiogenesis under inflammatory conditions.The mouse model of suture-induced corneal neovascularization was used to assess the antihem- and antilymphangiogenic effect of topically applied pegaptanib. Corneal blood and lymph vascularized areas were analyzed morphometrically. Furthermore, we analyzed the proliferative effects of VEGF A 121, 165, and 189 on blood and lymphatic endothelial cells (BEC/LEC) via a cell-proliferation assay.Pegaptanib significantly inhibited inflammatory corneal hemangiogenesis (p 0.01), but not lymphangiogenesis in vivo (p 0.05), both topically as well as systemically, in the inflamed cornea. In vitro, BECs were more susceptible to pegaptanib than LECs.Targeting VEGF-A 165 significantly inhibits hem- but not lymphangiogenesis, suggesting VEGF-A 165 to be critical for hem-, but dispensable for lymphangiogenesis, at least in the inflamed cornea.
