Agriculture is one of the most essential activities for humanity. Systems capable of automatically harvesting a crop using robots or performing a reasonable production estimate can reduce costs and increase production efficiency. With the advancement of computer vision, image processing methods are becoming increasingly viable in solving agricultural problems. Thus, this work aims to count green oranges directly from trees through video footage filmed in line along a row of orange trees on a plantation. For the video image processing flow, a solution was proposed integrating the YOLOv4 network with object-tracking algorithms. In order to compare the performance of the counting algorithm using the YOLOv4 network, an optimal object detector was simulated in which frame-by-frame corrected detections were used in which all oranges in all video frames were detected, and there were no erroneous detections. Being the scientific and technological innovation the possibility of distinguishing the green color of the fruits from the green color of the leaves. The use of YOLOv4 together with object detectors managed to reduce the number of double counting errors and obtained a count close to the actual number of oranges visible in the video. The results were promising, with an mAP50 of 80.16%, mAP50:95 of 53.83%, precision of 0.92, recall of 0.93, F1-score of 0.93, and average IoU of 82.08%. Additionally, the counting algorithm successfully identified and counted 204 oranges, closely approaching the actual count of 208. The study also resulted in a database with an amount of 644 images containing 43,109 orange annotations that can be used in future works.

Coloquialmente nos referimos a los espejismos como ilusiones ópticas sobre un determinado objeto; apariencias de la realidad. El ejercicio del Derecho en Ecuador está compuesto por varias de estas distorsiones que en la práctica se ven reflejadas por las costumbres comunes de jueces y abogados. En tal virtud, este artículo se compone de 3 espejismos en los procesos de tutela constitucional: i) La ilusión de la verdad como objeto de la prueba; ii) la utopía de la corrección epistémica de los hechos a través de los poderes probatorios del juez; y, iii) los laberintos del estándar de prueba propuesto por la Corte Constitucional para los procesos constitucionales. Tres tópicos que jueces y abogados saben de su importancia en una democracia. Empero, parecería que cuando entramos a la sala de audiencias vale más un buen relato bien contado que la corrección epistémica en aras de la verdad y la justicia ¿Será que algún día tomamos a los hechos en serio?

La prueba ha sido uno de los tópicos que ha merecido un importante desarrollo estos últimos años, dado que los hechos no deben ser vistos de manera superficial. Requiere, más bien, tanta o más exigencia como cuando se interpreta y se argumenta sobre asuntos de Derecho. Esta idea está en el corazón de este trabajo que versará sobre el trazado que el legislador ha diseñado para la distribución del riesgo del error, a través de la inversión de la carga de prueba en la acción de protección. Como resultado, con aciertos y desaciertos, a través de un método dogmático jurídico, se ha identificado a las presunciones legales relativas como el eje de este sistema distributivo probatorio y al estándar de prueba como llave maestra que eleva el nivel de exigencia de la justificación de los hechos.

Recent studies have been evaluating the presence of patterns associated with the occurrence of cancer in different types of tissue present in the individual affected by the disease. In this article, we describe preliminary results for the automatic detection of cancer (Walker 256 tumor) in laboratory animals using preclinical microphotograph images of the subject’s liver tissue. In the proposed approach, two different types of descriptors were explored to capture texture properties from the images, and we also evaluated the complementarity between them. The first texture descriptor experimented is the widely known Local Phase Quantization (LPQ), which is a descriptor based on spectral information. The second one is built by the application of a granulometry given by a family of morphological filters. For classification, we have evaluated the algorithms Support Vector Machine (SVM), k-Nearest Neighbor (k-NN) and Logistic Regression. Experiments carried out on a carefully curated dataset developed by the Enteric Neural Plasticity Laboratory of the State University of Maringá showed that both texture descriptors provide good results in this scenario. The accuracy rates obtained using the SVM classifier were 96.67% for the texture operator based on granulometry and 91.16% for the LPQ operator. The dataset was made available also as a contribution of this work. In addition, it is important to remark that the best overall result was obtained by combining classifiers created using both descriptors in a late fusion strategy, achieving an accuracy of 99.16%. The results obtained show that it is possible to automatically perform the identification of cancer in laboratory animals by exploring texture properties found on the tissue taken from the liver. Moreover, we observed a high level of complementarity between the classifiers created using LPQ and granulometry properties in the application addressed here.

Este trabalho apresenta uma metodologia voltada para mensurar a velocidade de um corpo rígido em movimento, tendo como ponto de partida, uma sequência de imagens obtidas por meio de uma filmadora digital. No processo de mensuração da velocidade, na etapa de identificação dos corpos em movimento, é utilizada uma técnica de segmentação baseada na média temporal dos valores de cada pixel registrados em N quadros de imagens consecutivos, em que, após a identificação desses corpos, é realizada a transformação dos pontos no plano imagem para o espaço objeto, por meio do modelo matemático definido como funções racionais. Conhecendo-se as coordenadas dos pontos no espaço objeto, a velocidade de deslocamento do corpo pode ser calculada, através do intervalo de tempo obtido pela quantidade de quadros que a câmara captura por segundo. Para validar a metodologia proposta foi realizado um experimento que contou com a utilização de um aparelho denominado trilho de ar, cinco sensores fotoelétricos dispersos sobre o mesmo com distancias conhecidas entre si e um cronômetro ligado aos sensores. No momento em que um corpo em movimento passa por um sensor, o cronômetro é paralisado e com isso, se tem a distância percorrida pelo corpo entre cada sensor e, quanto tempo foi necessário para percorrer cada uma dessas distâncias. Com esses dados em mãos é realizada uma comparação dos resultados obtidos por meio da análise de sequência de imagens com os resultados obtidos através dos cronômetros e dos sensores fotoelétricos.

JUSTIFICATIVA E OBJETIVOS: O procedimento de correção de pé torto congênito (PTC) cursa com dor pós-operatória intensa. A técnica mais utilizada em crianças é a peridural caudal associada à anestesia geral. Tem como limitação a curta duração da analgesia pós-operatória. Os bloqueios de nervos periféricos têm sido apontados como procedimentos com baixa incidência de complicações e tempo prolongado de analgesia. O objetivo do estudo foi comparar o tempo de analgesia dos bloqueios nervosos periféricos e bloqueio caudal e o consumo de morfina nas primeiras 24 horas após a correção de PTC em crianças. MÉTODO: Estudo randômico, encoberto, em crianças submetidas à intervenção cirúrgica para liberação póstero-medial de PTC, alocadas em 4 grupos conforme a técnica anestésica: Caudal (ACa); Bloqueios isquiático e femoral (IF); Bloqueios isquiático e safeno (IS); Bloqueio isquiático e anestesia local (IL), associados à anestesia geral. Nas primeiras 24 horas os pacientes receberam dipirona e paracetamol via oral e foram avaliados por anestesiologista que desconhecia a técnica empregada. Conforme escores da escala CHIPPS (Children's and infants postoperative pain scale) era administrada morfina via oral (0,19 mg.kg-1 por dia). RESULTADOS: Foram estudadas 118 crianças distribuídas nos grupos ACa (30), IF (32), IS (28) IL (28). O tempo médio entre o bloqueio e a primeira dose de morfina foi 6,16 horas no grupo ACa, 7,05 horas no IF, 7,58 horas no IS e 8,18 horas no IL. O consumo de morfina foi 0,3 mg.kg-1 por dia nos quatro grupos. Não houve diferença significativa entre os grupos. CONCLUSÕES: Os bloqueios nervosos periféricos não promoveram maior tempo de analgesia, tampouco redução no consumo de morfina nas primeiras 24 horas em crianças submetidas à correção de PTC quando comparados ao bloqueio peridural caudal.JUSTIFICATIVA Y OBJETIVOS: El procedimiento de corrección de pie jorobado congénito (PJC), debuta con dolor postoperatorio intenso. La técnica más utilizada en niños es la epidural caudal asociada a la anestesia general. Posee la limitante de una corta duración de la analgesia postoperatoria. Los bloqueos de nervios periféricos han sido indicados como procedimientos con una baja incidencia de complicaciones y un tiempo prolongado de analgesia. El objetivo del estudio actual, fue comparar el tiempo de analgesia de los bloqueos nerviosos periféricos y del bloqueo caudal y el consumo de morfina, en las primeras 24 horas después de la corrección de PJC en niños. MÉTODO: Estudio randómico doble ciego, en niños sometidos a la intervención quirúrgica para liberación posteromedial de PJC, ubicadas en cuatro grupos conforme a la técnica anestésica: caudal (ACa); bloqueos isquiático y femoral (IF); bloqueos isquiático y safeno (IS); bloqueo isquiático y anestesia local (IL), asociados a la anestesia general. En las primeras 24 horas, los pacientes recibieron dipirona y paracetamol vía oral y fueron evaluados por un anestesiólogo que no conocía la técnica usada. Conforme a las puntuaciones de la escala CHIPPS (Children's and infants postoperative pain scale), se administraba morfina vía oral (0,19 mg.kg-1 por día). RESULTADOS: Fueron estudiados 118 niños distribuidos en los grupos ACa (30), IF (32), IS (28) IL (28). El tiempo promedio entre el bloqueo y la primera dosis de morfina fue de 6,16 horas en el grupo ACa, 7,05 horas en el IF, 7,58 horas en el IS y 8,18 horas en el IL. El consumo de morfina fue de 0,3 mg.kg-1 por día en los cuatro grupos. No hubo diferencia significativa entre los grupos. CONCLUSIONES: Los bloqueos nerviosos periféricos no promovieron un tiempo más elevado de analgesia, ni tampoco una reducción en el consumo de morfina en las primeras 24 horas, en niños sometidos a la corrección de PJC cuando se les comparó con el bloqueo epidural caudal.BACKGROUND AND OBJECTIVES: Correction of congenital clubfoot (CCF) is associated with severe postoperative pain. Caudal epidural block associated with general anesthesia is the anesthetic technique used more often in children, but it is limited by the short duration of the postoperative analgesia. Peripheral nerve blocks are associated with a low incidence of complications and prolonged analgesia. The objective of this study was to compare the duration of analgesia in peripheral nerve blocks and caudal block, as well as morphine consumption in the first 24 hours after correction of CCF in children. METHODS: This is a randomized, double-blind study with children undergoing surgeries for posteromedial release of CCF, who were divided in four groups according to the anesthetic technique: caudal (ACa), sciatic and femoral block (IF), sciatic and saphenous block (IS), and sciatic block and local anesthesia (IL), associated with general anesthesia. In the first 24 hours, patients received oral dypirone and acetaminophen, and they were evaluated by anesthesiologists who were unaware of the technique used. Oral morphine (0.19 mg.kg¹ per day) was administered according to the scores of the CHIPPS (Children's and infants' postoperative pain scale) scale. RESULTS: One hundred and eighteen children separated into four groups: ACa (30), IF (32), IS (28), and IL (28) participated in this study. The mean time between the blockade and the first dose of morphine was 6.16 hours in group ACa, 7.05 hours in group IF, 7.58 in IS, and 8.18 hours in IL. Morphine consumption was 0.3 mg.kg-1 per day in all four groups. Significant differences were not observed among the groups. CONCLUSIONS: peripheral nerve blocks did not promote longer lasting analgesia or a decrease in morphine consumption in the first 24 hours in children undergoing CCF correction when compared to caudal epidural block.

JUSTIFICATIVA E OBJETIVOS: O bloqueio do plexo lombar pela via posterior promove analgesia pós-operatória efetiva na artroplastia total do quadril. Ropivacaína e bupivacaína não apresentaram qualquer diferença na eficácia analgésica em diferentes bloqueios de nervos periféricos. O objetivo deste estudo foi comparar a eficácia da analgesia pós-operatória resultante da administração em dose única da bupivacaína a 0,5% ou da ropivacaína a 0,5% no bloqueio do plexo lombar pela via posterior na artroplastia total do quadril. MÉTODO: Trinta e sete pacientes foram alocados aleatoriamente em dois grupos, segundo o anestésico local utilizado no bloqueio: Grupo B - bupivacaína a 0,5% com epinefrina 1:200.000 ou Grupo R - ropivacaína a 0,5%. Durante o período pós-operatório, os escores de dor e o consumo de morfina na analgesia controlada pelo paciente foram comparados entre os grupos. O sangramento durante a operação e a incidência de efeitos adversos e de complicações também foram comparados. RESULTADOS: Apesar dos escores de dor terem sido menores no Grupo R 8, 12 e 24 horas após o bloqueio, essas diferenças não foram clinicamente significativas. Regressão linear múltipla não identificou o anestésico local como variável independente. Não houve diferença no consumo de morfina, no sangramento intraoperatório e na incidência de complicações e efeitos adversos entre os dois grupos. CONCLUSÕES: A bupivacaína a 0,5% e a ropivacaína a 0,5% produziram alívio eficaz e prolongado da dor pós-operatória após artroplastia total do quadril, sem diferença clínica, quando doses equivalentes foram administradas no bloqueio do plexo lombar pela via posterior.JUSTIFICATIVA Y OBJETIVOS: El bloqueo del plexo lumbar por la vía posterior, genera una analgesia postoperatoria efectiva en la artroplastia total de la cadera. La ropivacaína y la bupivacaína no arrojaron ninguna diferencia en la eficacia analgésica en diferentes bloqueos de nervios periféricos. El objetivo de este estudio, fue comparar la eficacia de la analgesia postoperatoria, resultante de la administración en dosis única de la bupivacaína a 0,5% o de la ropivacaína a 0,5% en el bloqueo del plexo lumbar por la vía posterior en la artroplastia total de la cadera. MÉTODO: Treinta y siete pacientes fueron ubicados aleatoriamente en dos grupos según el anestésico local utilizado en el bloqueo: Grupo B - bupivacaína a 0,5% con epinefrina 1:200.000 o Grupo R - ropivacaína a 0,5%. Durante el período postoperatorio, los puntajes de dolor y el consumo de morfina en la analgesia controlada por el paciente, fueron comparados entre los grupos. El sangramiento durante la operación y la incidencia de efectos adversos y de complicaciones también fueron comparados. RESULTADOS: Pese a que los puntajes de dolor hayan sido menores en el Grupo R 8 horas, 12 horas y 24 horas después del bloqueo, esas diferencias no fueron clínicamente significativas. Ropivacaine and bupivacaine do not show differences in analgesic efficacy when used in different peripheral nerve blocks. The objective of this study was to compare the efficacy of postoperative analgesia resulting from the administration of a single dose of 0.5% bupivacaine or 0.5% ropivacaine in posterior lumbar plexus block for total hip arthroplasty. METHODS: Thirty-seven patients were randomly divided in two groups according to the local anesthetic used: Group B - 0.5% bupivacaine with 1:200,000 epinephrine; or group R - 0.5% ropivacaine. During the postoperative period, pain scores and morphine consumption in patient controlled analgesia were compared between groups. Bleeding during surgery and the incidence of side effects and complications were also compared. RESULTS: Although pain scores were lower in Group R 8 hours, 12 hours, and 24 hours after the blockade, these differences were not clinically significant. Multiple linear regression identified the local anesthetic as an independent variable. Differences in morphine consumption, intraoperative bleeding, and the incidence of complications and side effects were not observed between both groups. CONCLUSIONS: 0.5% Bupivacaine and 0.5% ropivacaine produced effective and prolonged postoperative pain relief after total hip arthroplasty, without clinical differences, when equivalent doses were administered for posterior lumbar plexus block.

This paper proposes an automatic technique for water bodies; shadow and cloud segmentation applied to imagery acquired by CBERS and Landsat satellites. The technique is based on spectral and color data. Experiments were accomplished using complex landscape images containing water bodies, clouds, shadows, exposed soil and varied vegetation. Straightforward operations are applied to enhance the features of interest, in order to improve the segmentation results with robustness. The radiometric response pattern of each feature in particular, as well its colors nature, are used to accomplish this enhance. To improve the discrimination process of those features, two features indexes was used: NDVI to indicate the vegetation presence and WWI to indicate the water presence. For CBERS images were necessary the application of an automatic radiometric adjustment process in order to achieve appropriate results. To accomplish that radiometric adjustment was proposed and evaluated a procedure that allows recovering color attributes information of the features that was degenerated by atmospheric effects action (absorption and scattering).

Este artigo apresenta uma técnica para segmentação de corpos d'água, nuvens e áreas afetadas por sombras presentes em imagens multiespectrais adquiridas pelos satélites CBERS e Landsat. Essa técnica é baseada somente nos dados espectrais e de cor das feições imageadas. Foram realizados vários experimentos usando imagens complexas contendo corpos d'água, nuvens, sombras, solo exposto e variados tipos de vegetação. Operações simples de imagens são aplica-das para realçar as feições de interesse. Esse realce é necessário para promover um nível de segurança maior nas opera-ções de segmentação das feições e é feito usando tanto os padrões da resposta radiométrica como a natureza da cor dessas feições. Para auxiliar o processo de discriminação dessas feições, dois índices indicativos são usados: NDVI para indicar a presença de vegetação e WWI para indicar a presença de água. Para imagens CBERS é necessário fazer um ajuste radiométrico nas imagens para se alcançar resultados mais adequados. Para realizar esse ajuste radiométrico foi proposto e avaliado um procedimento automático que permite recuperar informações sobre os atributos de cor das fei-ções que foram degenerados pela ação dos efeitos atmosféricos (absorção e espalhamento).

Atlantic salmon are one of the most important fish species in global aquaculture production. However, temperature increases attributed to climatic events impair the production of Atlantic salmon during summer. Additionally, the nutritional requirements for this species when reared under elevated temperatures require elucidation. To address this gap, a feeding trial was conducted to investigate the effect of glutamine supplementation-a functional amino acid (AA) important for energy production and gut health-on the growth, metabolism, gut morphology, antioxidant capacity and thermal tolerance of Atlantic salmon parr at elevated temperatures (22°C). Atlantic salmon were pair-fed three isoenergetic diets: a control diet (D1, no addition of glutamine), D2 (7% glutamine supplementation with other dietary AA levels reduced-isonitrogenous to D1) and D3 (6% glutamine and with the same AA profile as D1). Metabolic rate measurements and sampling commenced after 7.5 weeks on diets and 3 weeks of exposure to 22°C. Glutamine supplementation (D2 and D3) did not affect specific growth rate (SGR), condition factor, relative gut mass or carcass composition despite fish fed D3 having increased pyloric caeca fold height (hF). Resting, maximum and digestive metabolic rates were also unaffected by glutamine supplementation. Responses of organisms to climate warming are variable and complex. Effects on species distributions are already evident and mean global surface ocean temperatures are likely to warm by up to 4.1 °C by 2100, substantially impacting the physiology and distributions of ectotherms. The largest marine ectotherm, the whale shark Rhincodon typus, broadly prefers sea surface temperatures (SST) ranging from 23 to 30 °C. Whole-species distribution models have projected a poleward range shift under future scenarios of climate change, but these models do not consider intraspecific variation or phenotypic plasticity in thermal limits when modelling species responses, and the impact of climate warming on the energetic requirements of whale sharks is unknown. Using a dataset of 111 whale shark movement tracks from aggregation sites in five countries across the Indian Ocean and the latest Earth-system modelling produced from Coupled Model Intercomparison Project Phase 6 for the Intergovernmental Panel on Climate Change, we examined how SST and total zooplankton biomass, their main food source, may change in the future, and what this means for the energetic balance and extent of suitable habitat for whale sharks. Earth System Models, under three Shared Socioeconomic Pathways (SSPs; SSP1-2.6, SSP3-7.0 and SSP5-8.5), project that by 2100 mean SST in four regions where whale shark aggregations are found will increase by up to 4.9 °C relative to the present, while zooplankton biomass will decrease. This reduction in zooplankton is projected to be accompanied by an increase in the energetic requirements of whale sharks because warmer water temperatures will increase their metabolic rate. We found marked differences in projected changes in the extent of suitable habitat when comparing a whole-species distribution model to one including regional variation. This suggests that the conventional approach of combining data from different regions within a species' distribution could underestimate the amount of local adaptation in populations, although parameterising local models could also suffer from having insufficient data and lead to model mis-specification or highly uncertain estimates. Elevated ultraviolet radiation (UVR) is postulated as one of multiple, interrelated environmental stressors driving amphibian population declines globally. However, key knowledge gaps remain in elucidating the link between elevated UVR and amphibian declines in a changing climate, including whether timing and irradiance of UVR exposure in early life dictates the onset of detrimental carryover effects post-metamorphosis. In this study, striped marsh frog larvae (Limnodynastes peronii) were exposed to UVR at one of two different irradiances for up to 7 days, either as hatchlings (Gosner stage 23) or as older larvae (Gosner stage 25-28). These animals were then reared to metamorphosis in the absence of UVR to examine independent and interactive carryover effects throughout development. Older larvae were more sensitive to UVR than hatchlings, with 53.1% and 15.6% mortality in larvae exposed to high and low irradiance respectively, compared with no mortality of hatchlings in either irradiance treatment. Irradiance and timing of UVR exposure had interactive effects on larval body length, causing stunted growth patterns and a lack of compensatory growth following UVR exposure, particularly in animals exposed to high irradiance UVR later in development. Timing of UVR exposure also determined the severity of carryover effects into metamorphosis, including delayed metamorphosis and the first published account (to our knowledge) of latent UVR-induced depigmentation in an amphibian. These findings highlight how acute changes to the larval UVR exposure regime can impact on amphibian health later in life, with implications for our understanding of the effects of climate change on UVR-related amphibian declines., (© 2024 Wiley Periodicals LLC.)

An ectotherm's performance and physiological function are strongly tied to environmental temperature, and many ectotherms thermoregulate behaviourally to reach optimum body temperatures. Tropical ectotherms are already living in environments matching their thermal tolerance range and may be expected to conform to environmental temperatures. We tracked the body temperatures ( T b ) of 163 estuarine crocodiles across 13 years and compared T b of 39 crocodiles to water temperature gathered using fish-borne sensors ( T w ) across 3 years (2015-2018). While T b largely conformed closely to T w , we found inter- and intra-individual differences in relative body temperature ( T b - T w ) that depended on sex and body size as well as the time of day and year. Deviations from T w , especially during the warm parts of the year, suggest that thermoregulatory behaviour was taking place: we found patterns of warming and cooling events that seemed to mediate this variation in T b . Thermoregulatory behaviour was observed most frequently in larger individuals, with warming events common during winter and cooling events common during summer. By observing free-ranging animals across multiple years, we found that estuarine crocodiles show yearly patterns of active cooling and warming behaviours that modify their body temperature, highlighting their resilience in the face of recent climate warming. Our work also provides the first evidence for thermal type in large-bodied reptiles.

How animals move and associate with conspecifics is rarely random, with a population's spatial structure forming the foundation on which the social behaviours of individuals form. Studies examining the spatial-social interface typically measure averaged behavioural differences between individuals; however, this neglects the inherent variation present within individuals and how it may impact the spatial-social interface. Here, we investigated differences in among-individual (co)variance in sociability, activity and site fidelity in a population of wild estuarine crocodiles, Crocodylus porosus, across a 10-year period. By monitoring 118 crocodiles using coded acoustic transmitters and an array of fixed underwater receivers, we discovered that not only did individual crocodiles repeatably differ (among-individual variation) in each behaviour measured but also in how consistently they expressed these behaviours through time (within-individual variation). As expected, crocodile activity and sociability formed a behavioural syndrome, with more active individuals being less sociable. Interestingly, we also found that individuals that were either more sociable or displayed greater site fidelity were also more specialized (lower within-individual variation) in these behaviours. Together, our results provide important empirical evidence for the interplay between spatial, temporal and social individual-level behavioural variation and how these contribute to forming behavioural niches. Cardiac phenotypic plasticity, the remodelling of heart structure and function, is a response to any sustained (or repeated) stimulus or stressor that results in a change in heart performance. Cardiac plasticity can be either adaptive (beneficial) or maladaptive (pathological), depending on the nature and intensity of the stimulus. Here, we draw on articles published in this Special Issue of Journal of Experimental Biology, and from the broader comparative physiology literature, to highlight the core components that enable cardiac plasticity, including structural remodelling, excitation-contraction coupling remodelling and metabolic rewiring. We discuss when and how these changes occur, with a focus on the underlying molecular mechanisms, from the regulation of gene transcription by epigenetic processes to post-translational modifications of cardiac proteins. Genome-wide association studies (GWASs) of major depressive disorder (MDD) have recently achieved extremely large sample sizes and yielded substantial numbers of genome-wide significant loci. Because of the approach to ascertainment and assessment in many of these studies, some of these loci appear to be associated with dysphoria rather than with MDD, potentially decreasing the clinical relevance of the findings. An alternative approach to MDD GWAS is to focus on the most severe forms of MDD, with the hope that this will enrich for loci of larger effect, rendering their identification plausible, and providing potentially more clinically actionable findings. Here we review the genetics of severe depression by using clinical markers of severity including: age of onset, recurrence, degree of impairment, and treatment with ECT. There is evidence for increased family-based and Single Nucleotide Polymorphism (SNP)-based estimates of heritability in recurrent and early-onset illness as well as severe functional impariment. GWAS have been performed looking at severe forms of MDD and a few genome-wide loci have been identified. Several whole exome sequencing studies have also been performed, identifying associated rare variants. Although these findings have not yet been rigorously replicated, the elevated heritability seen in severe MDD phenotypes suggests the value of pursuing additional genome-wide interrogation of samples from this population. The challenge now is generating a cohort of adequate size with consistent phenotyping that will allow for careful and robust classifications and distinctions to be made. Ultraviolet radiation (UVR) is a pervasive factor that has shaped the evolution of life on Earth. Ambient levels of UVR mediate key biological functions but can also cause severe lethal and sublethal effects in a wide range of organisms. Furthermore, UVR is a powerful modulator of the effects of other environmental factors on organismal physiology, such as temperature, disease, toxicology and pH, among others. This is critically important in the context of global change, where understanding the effects of multiple stressors is a key challenge for experimental biologists. Ecological physiologists rarely afford UVR discussion or include UVR in experimental design, even when it is directly relevant to their study system. During summer, farmed Atlantic salmon (Salmo salar) can experience prolonged periods of warming and low aquatic oxygen levels due to climate change. This often results in a drop in feed intake; however, the physiological mechanism behind this behaviour is unclear. Digestion is a metabolically expensive process that can demand a high proportion of an animal's energy budget and might not be sustainable under future warming scenarios. We investigated the effects of elevated temperature and acute hypoxia on specific dynamic action (SDA; the energetic cost of digestion), and how much of the energy budget (i.e. aerobic scope, AS) was occupied by SDA in juvenile Atlantic salmon. AS was 9% lower in 21°C-acclimated fish compared to fish reared at their optimum temperature (15°C) and was reduced by ~50% by acute hypoxia (50% air saturation) at both temperatures. Furthermore, we observed an increase in peak oxygen uptake rate during digestion which occupied ~13% of the AS at 15°C and ~20% of AS at 21°C, and increased the total cost of digestion at 21°C. The minimum oxygen tolerance threshold in digesting fish was ~42% and ~53% at 15 and 21°C, respectively, and when digesting fish were exposed to acute hypoxia, gut transit was delayed. Thus, these stressors result in a greater proportion of the available energy budget being directed away from digestion. In response to seasonal droughts, the green striped burrowing frog Cyclorana alboguttata enters a reversible hypometabolic state called aestivation where heart rate and oxygen consumption can be reduced despite warm (>25C°) ambient temperatures. With a view to understanding molecular mechanisms we profiled aestivating versus control gastrocnemius muscle using mRNA sequencing. This indicated an extensive metabolic reprogramming, with nearly a quarter of the entire transcriptome (3996 of 16,960 mRNA) exhibiting a nominal >2-fold change. Consistent with a physiological adaptation to spare carbohydrate reserves, carbohydrate catabolism was systemically downregulated. A 630-fold downregulation of ENO3 encoding the enolase enzyme was most striking. The 590 frog orthologs of mRNA encoding the mitoproteome were, viewed as a population, significantly downregulated during aestivation, although not to the same extent as mRNA encoding carbohydrate catabolism. Prominent examples include members of the TCA cycle (IDH2), electron transport chain (NDUFA6), the ATP synthase complex (ATP5F1B) and ADP/ATP intracellular transport (SLC25A4). Moreover, mRNA derived from the mt genome itself (e.g. mt-ND1) were also downregulated. Most prominent among the upregulated mRNA are those encoding aspects of regulated proteolysis including the proteosome (e.g. PSME4L), peptidases (USP25), atrogins (FBXO32) and ubiquitination (VCP). Finally, we note the ∼5-fold upregulation of the mRNA EIFG3 that encodes part of the EIF4F complex. This possesses global control of protein synthesis. Amphibian declines are sometimes correlated with increasing levels of ultraviolet radiation (UVR). While disease is often implicated in declines, environmental factors such as temperature and UVR play an important role in disease epidemiology. The mutagenic effects of UVR exposure on amphibians are worse at low temperatures. Amphibians from cold environments may be more susceptible to increasing UVR. However, larvae of some species demonstrate cold acclimation, reducing UV-induced DNA damage at low temperatures. Understanding of the mechanisms underpinning this response is lacking. We reared Limnodynastes peronii larvae in cool (15°C) or warm (25°C) waters before acutely exposing them to 1.5 h of high intensity (80 µW cm -2 ) UVBR. We measured the color of larvae and mRNA levels of a DNA repair enzyme. We reared larvae at 25°C in black or white containers to elicit a skin color response, and then measured DNA damage levels in the skin and remaining carcass following UVBR exposure. Cold-acclimated larvae were darker and displayed lower levels of DNA damage than warm-acclimated larvae. There was no difference in CPD-photolyase mRNA levels between cold- and warm-acclimated larvae. Skin darkening in larvae did not reduce their accumulation of DNA damage following UVR exposure. Our results showed that skin darkening does not explain cold-induced reductions in UV-associated DNA damage in L. peronii larvae. Statistical analyses that physiologists use to test hypotheses predominantly centre on means, but the tail ends of the response distribution can behave quite differently and underpin important scientific phenomena. We demonstrate that quantile regression (QR) offers a way to bypass some limitations of least squares regression (LSR) by building a picture of independent variable effects across the whole distribution of a dependent variable. We used LSR and QR with simulated and real datasets. With simulated data, LSR showed no change in the mean response but missed significant effects in the tails of the distribution found using QR. With real data, LSR showed a significant change in the mean response but missed a lack of response in the upper quantiles which was biologically revealing. Reef-building corals create one of the most biodiverse and economically important ecosystems on the planet. Unfortunately, global coral reef ecosystems experience threats from numerous natural stressors, which are amplified by human activities. One such threat is ultraviolet radiation (UVR) from the sun; a genotoxic stressor that is a double-edged sword for corals as they rely on sunlight for energy. More intense UVR has been shown to have greater direct impacts on animal physiology, and these may be exacerbated by co-occurring stressors. The aim of this systematic literature review was to examine if the same applies to corals; that is, if the co-exposure of a constant stressor (UVR) with other stressors has a greater impact on coral physiology than if these stressors occurred separately. We reviewed the biochemical and cellular processes impacted by UVR and the defenses corals have against UVR. The main stressors investigated with UVR were temperature, nitrate, nutrient, oil, water motion, and photosynthetically active radiation (PAR). UVR generally worsened the physiological impacts of other stressors (e.g., by decreasing zooxanthellae and chlorophyll densities). There were species-specific differences in their tolerance to UVR (differences in zooxanthellae populations, sunscreen production and depth) and environmental stress (e.g., resilience to some oils), and that ambient levels of UVR were often beneficial (i.e., nullifying impacts of nitrates). We highlight areas of future investigation including examining and assessing other interacting stressors and their impacts (e.g., ocean acidification, ocean deoxygenation, toxins and pollutants), investigating impacts of multiple stressors with UVR on the coral microbiome, and elucidating the effects of multi-stressors with UVR across early-life history stages (especially larvae). UVR is a pervasive stressor to corals and can interact with other environmental conditions to compromise the resilience of corals. To celebrate its centenary year, Journal of Experimental Biology (JEB) commissioned a collection of articles examining the past, present and future of experimental biology. This Commentary closes the collection by considering the important research opportunities and challenges that await us in the future. We expect that researchers will harness the power of technological advances, such as '-omics' and gene editing, to probe resistance and resilience to environmental change as well as other organismal responses. The capacity to handle large data sets will allow high-resolution data to be collected for individual animals and to understand population, species and community responses. The availability of large data sets will also place greater emphasis on approaches such as modeling and simulations. Finally, the increasing sophistication of biologgers will allow more comprehensive data to be collected for individual animals in the wild. Ultraviolet radiation (UVR) from the sun is a natural daytime stressor for vertebrates in both terrestrial and aquatic ecosystems. UVR effects on the physiology of vertebrates manifest at the cellular level, but have bottom-up effects at the tissue level and on whole-animal performance and behaviours. Climate change and habitat loss (i.e. loss of shelter from UVR) could interact with and exacerbate the genotoxic and cytotoxic impacts of UVR on vertebrates. Therefore, it is important to understand the range and magnitude of effects that UVR can have on a diversity of physiological metrics, and how these may be shaped by taxa, life stage or geographical range in the major vertebrate groups. Using a meta-analytical approach, we used 895 observations from 47 different vertebrate species (fish, amphibian, reptile and bird), and 51 physiological metrics (i.e. cellular, tissue and whole-animal metrics), across 73 independent studies, to elucidate the general patterns of UVR effects on vertebrate physiology. We found that while UVR's impacts on vertebrates are generally negative, fish and amphibians were the most susceptible taxa, adult and larvae were the most susceptible life stages, and animals inhabiting temperate and tropical latitudes were the most susceptible to UVR stress. This information is critical to further our understanding of the adaptive capacity of vulnerable taxon to UVR stress, and the wide-spread sublethal physiological effects of UVR on vertebrates, such as DNA damage and cellular stress, which may translate up to impaired growth and locomotor performance. These impairments to individual fitness highlighted by our study may potentially cause disruptions at the ecosystem scale, especially if the effects of this pervasive diurnal stressor are exacerbated by climate change and reduced refuge due to habitat loss and degradation. Increases in ultraviolet radiation (UVR) correlate spatially and temporally with global amphibian population declines and interact with other stressors such as disease and temperature. Declines have largely occurred in high-altitude areas associated with greater UVR and cooler temperatures. UVR is a powerful mutagenic harming organisms largely by damaging DNA. When acutely exposed to UVR at cool temperatures, amphibian larvae have increased levels of DNA damage. Amphibians may compensate for the depressive effects of temperature on DNA damage through acclimatisation, but it is unknown whether they have this capacity. We reared striped marsh frog larvae (Limnodynastes peronii) in warm (25 °C) and cool (15 °C) temperatures under a low or moderate daily dose of UVR (10 and 40 μW cm -2 UV-B for 1 h at midday, respectively) for 18-20 days and then measured DNA damage resulting from an acute high UVR dose (80 μW cm -2 UV-B for 1.5 h) at a range of temperatures (10, 15, 20, 25, and 30 °C). Larvae acclimated to 15 °C and exposed to UVR at 15 °C completely compensated UVR-induced DNA damage compared with 25 °C acclimated larvae exposed to UVR at 25 °C. Additionally, warm-acclimated larvae had higher DNA damage than cold-acclimated larvae across test temperatures, which indicated a cost of living in warmer temperatures. Larvae reared under elevated UVR levels showed no evidence of UVR acclimation resulting in lower DNA damage following high UVR exposure. Our finding that thermal acclimation in L. peronii larvae compensated UVR-induced DNA damage at low temperatures suggested that aquatic ectotherms living in cool temperatures may be more resilient to high UVR than previously realised. Ecological carryover effects, or delayed effects of the environment on an organism's phenotype, are central predictors of individual fitness and a key issue in conservation biology. Climate change imposes increasingly variable environmental conditions that may be challenging to early life-history stages in animals with complex life histories, leading to detrimental physiological and fitness effects in later life. Yet, the latent nature of carryover effects, combined with the long temporal scales over which they can manifest, means that this phenomenon remains understudied and is often overlooked in short-term studies limited to single life-history stages. Herein, we review evidence for the physiological carryover effects induced by elevated ultraviolet radiation (UVR; 280-400 nm) as a potential contributor to recent amphibian population declines. UVR exposure causes a suite of molecular, cellular and physiological consequences known to underpin carryover effects in other taxa, but there is a lack of research linking embryonic and larval UVR exposures to fitness consequences post-metamorphosis in amphibians. We propose that the key impacts of UVR on disease-related amphibian declines are facilitated through carryover effects that bridge embryonic and larval UVR exposure with potential increased disease susceptibility post-metamorphosis. We conclude by identifying a practical direction for the study of ecological carryover effects in amphibians that could guide future ecological research in the broader field of conservation physiology. Given limited resources for wildlife conservation paired with an urgency to halt declines and rebuild populations, it is imperative that management actions are tactical and effective. Mechanisms are about how a system works and can inform threat identification and mitigation such that conservation actions that work can be identified. Here, we call for a more mechanistic approach to wildlife conservation and management where behavioral and physiological tools and knowledge are used to characterize drivers of decline, identify environmental thresholds, reveal strategies that would restore populations, and prioritize conservation actions. Increasing drought frequency and duration pose a significant threat to fish species in dryland river systems. As ectotherms, fish thermal and hypoxia tolerances directly determine the capacity of species to persist in these environments during low flow periods when water temperatures are high and waterbodies become highly stratified. Chronic thermal stress can compound the impacts of acute hypoxic events on fish resulting in significant fish mortality; however, it is not known if all size classes are equally susceptible, or if the allometric scaling of physiological processes means some size classes are disproportionately affected. We investigated the physiological responses of Murray cod ( Maccullochella peelii ) over a four-fold body size range (0.2-3000 g) to acute changes in water temperature and oxygen concentration following 4 weeks of acclimation to representative spring (20°C) and summer (28°C) water temperatures. We recorded maximum thermal tolerance ( CT max ), oxygen limited thermal tolerance (P CT max ), lowest tolerable oxygen level (as the oxygen level at which lose equilibrium; O 2,LOE ) , gill ventilation rates and aerial surface respiration threshold, blood oxygen transport capacity and lactate accumulation. Acclimation to elevated water temperatures improved thermal and hypoxia tolerance metrics across all size classes. However, body size significantly affected thermal and hypoxia responses. Small M. peelii were significantly less hypoxia tolerant than larger individuals, while larger fish were significantly less thermal tolerant than smaller fish. Hypoxia constrained thermal tolerance in M. peelii , with both small and large fish disproportionally compromised relative to mid-sized fish. Our findings indicate that both very small/young (larvae, fry, fingerlings) and very large/older M. peelii in dryland rivers are at significant risk from the combined impacts of a warming and drying climate and water extraction. Anthropogenic ozone depletion has led to a 2-5% increase in ultraviolet B radiation (UVBR) levels reaching the earth's surface. Exposure to UVBR causes harmful DNA damage in amphibians, but this is minimized by DNA repair enzymes such as thermally sensitive cyclobutane pyrimidine dimer (CPD)-photolyase, with cool temperatures slowing repair rates. It is unknown whether amphibian species differ in the repair response to a given dose of UVBR across temperatures. We reared larvae of three species ( Limnodynastes peronii , Limnodynastes tasmaniensis and Platyplectrum ornatum ) at 25°C and acutely exposed them to 80 µW cm -2 UVBR for 2 h at either 20°C or 30°C. UVBR-mediated DNA damage was measured as larvae repaired damage in photoreactive light at their exposure temperatures. Cool temperatures increased DNA damage in two species and slowed DNA repair rate in P. ornatum . The magnitude of DNA damage incurred from UVBR was species-specific. Platyplectrum ornatum had the lowest CPDs and DNA repair rates, and the depressive effects of low temperature on photorepair were greater in L. tasmaniensis . Considering the susceptibility of most aquatic organisms to UVBR, this research highlighted a need to consider the complexity of species-specific physiology when forecasting the influence of changing UVBR and temperature in aquatic ecosystems.

Determining the contribution of elevated ultraviolet-B radiation (UVBR; 280-315 nm) to amphibian population declines is being hindered by a lack of knowledge about how different acute UVBR exposure regimes during early life-history stages might affect post-metamorphic stages via long-term carryover effects. We acutely exposed tadpoles of the Australian green tree frog (Litoria caerulea) to a combination of different UVBR irradiances and doses in a multi-factorial laboratory experiment, and then reared them to metamorphosis in the absence of UVBR to assess carryover effects in subsequent juvenile frogs. Dose and irradiance of acute UVBR exposure influenced carryover effects into metamorphosis in somewhat opposing manners. Higher doses of UVBR exposure in larvae yielded improved rates of metamorphosis. However, exposure at a high irradiance resulted in frogs metamorphosing smaller in size and in poorer condition than frogs exposed to low and medium irradiance UVBR as larvae. We also demonstrate some of the first empirical evidence of UVBR-induced telomere shortening in vivo, which is one possible mechanism for life-history trade-offs impacting condition post-metamorphosis. Levels of ultraviolet (UV) radiation have increased in many parts of the world due to the anthropogenic destruction of the ozone layer. UV radiation is a potent immunosuppressant and can increase the susceptibility of animal hosts to pathogens. UV radiation can directly alter immune function via immunosuppression and photoimmunotolerance; however, UV may also influence pathogen defences by affecting the distribution of energy resources among competing physiological processes. Both defence against UV damage and repair of incurred damage, as well as the maintenance of immune defences and responding to an immune challenge, are energetically expensive. These competing demands for finite energy resources could trade off against one another, resulting in sub-optimal performance in one or both processes. We examined the potential for a disease-related energy trade-off in green tree frog ( Litoria caerulea ) larvae. Larvae were reared under high- or low-UV conditions for 12 weeks during which time we measured growth rates, metabolic rate and susceptibility to the amphibian fungal pathogen, Batrachochytrium dendrobatidis ( Bd ). We found that larvae exposed to high levels of UV radiation had higher rates of energy expenditure than those exposed to low UV levels; however, UV exposure did not affect growth rates or developmental timings. Larvae exposed to high UV radiation also experienced greater Bd infection rates and carried a higher infection burden than those not exposed to elevated UV radiation. We propose that the increased energetic costs of responding to UV radiation were traded off against immune defences to protect larval growth rates. Many aquatically respiring animals acutely exposed to low pH waters suffer inhibition of ion uptake and loss of branchial (gill) epithelial integrity, culminating in a fatal loss of body Na+. Environmental calcium levels ([Ca2+]e) are pivotal in maintaining branchial junction integrity, with supplemental Ca2+ reversing the negative effects of low pH in some animals. Tolerance of some naturally acidic environments by aquatic animals is further complicated by low [Ca2+]e, yet many of these environments are surprisingly biodiverse. How animals overcome the damaging actions of low pH and low environmental Ca2+ remains unknown. We examined the effects of [Ca2+]e on the response to low pH in larvae of the highly acid-tolerant frog Limnodynastes terraereginae. Acute exposure to low pH water in the presence of low (5 μmol l-1) [Ca2+]e increased net Na+ efflux. Provision of additional [Ca2+]e reduced net Na+ efflux, but the effect was saturable. Acclimation to both low and high (250 μmol l-1) [Ca2+]e improved the resistance of larvae to Na+ efflux at low pH. Exposure to the Ca2+ channel inhibitor ruthenium red resulted in an abrupt loss of tolerance in low pH-acclimated larvae. Acclimation to acidic water increased branchial gene expression of the intracellular Ca2+ transport protein calbindin, consistent with a role for increased transcellular Ca2+ trafficking in the tolerance of acidic water. Climate and land-use changes are expected to increase the future occurrence of wildfires, with potentially devastating consequences for freshwater species and ecosystems. Wildfires that burn in close proximity to freshwater systems can significantly alter the physicochemical properties of water. Following wildfires and heavy rain, freshwater species must contend with complex combinations of wildfire ash components (nutrients, polycyclic aromatic hydrocarbons, and metals), altered light and thermal regimes, and periods of low oxygen that together can lead to mass mortality events. However, the responses of aquatic fauna to wildfire disturbances are poorly understood. Here we provide a systematic review of available evidence on how aquatic animals respond to and recover from wildfire disturbance. Two databases (Web of Science and Scopus) were used to identify key literature. A total of 83 studies from across 11 countries were identified to have assessed the risk of wildfires on aquatic animals. We provide a summary of the main ecosystem-level changes associated with wildfires and the main responses of aquatic fauna to such disturbances. We pay special focus to physiological tools and biomarkers used to assess how wildfires impact aquatic animals. We conclude by providing an overview of how physiological biomarkers can further our understanding of wildfire-related impacts on aquatic fauna, and how different physiological tools can be incorporated into management and conservation plans and serve as early warning signs of wildfire disturbances., (© 2022 John Wiley & Sons Ltd.)

Cold water pollution (CWP) is caused by releases of unseasonably cold water from large, thermally stratified dams. Rapid and prolonged decreases in water temperature can have depressive effects on the metabolism, growth and swimming performance of fish. However, it is unknown if reducing the rate of temperature decrease could mitigate these negative effects by allowing thermal acclimation/acclimatization to occur. This study investigated the rate of temperature decrease as a potential CWP mitigation strategy in juvenile Murray cod Maccullochella peelii. M. peelii were exposed to a gradual, intermediate or rapid temperature decrease from 24 to 14°C. Energetic costs, locomotor performance, growth and survival were measured to determine if the initial thermal regime affected the thermal acclimation capacity of M. peelii. Cold exposure had significant acute and lasting depressive effects regardless of the rate of temperature decrease, although M. peelii showed varying degrees of thermal compensation in swimming performance and metabolism after 8 weeks of exposure to low temperatures. The short-term effects of CWP-like reductions in temperature are significant, but over time M. peelii can offset some of the depressive effects of CWP through thermal plasticity. This study highlights the importance of understanding physiological responses of fish to inform management and conservation. We conclude that rate of water temperature decline cannot be used to mitigate the sublethal effects of CWP on juvenile M. peelii but may still be useful for managing the negative effects in other native Australian fish species., (© 2022 The Authors. The global lockdown to mitigate COVID-19 pandemic health risks has altered human interactions with nature. Here, we report immediate impacts of changes in human activities on wildlife and environmental threats during the early lockdown months of 2020, based on 877 qualitative reports and 332 quantitative assessments from 89 different studies. Hundreds of reports of unusual species observations from around the world suggest that animals quickly responded to the reductions in human presence. However, negative effects of lockdown on conservation also emerged, as confinement resulted in some park officials being unable to perform conservation, restoration and enforcement tasks, resulting in local increases in illegal activities such as hunting. Overall, there is a complex mixture of positive and negative effects of the pandemic lockdown on nature, all of which have the potential to lead to cascading responses which in turn impact wildlife and nature conservation. While the net effect of the lockdown will need to be assessed over years as data becomes available and persistent effects emerge, immediate responses were detected across the world. Thus, initial qualitative and quantitative data arising from this serendipitous global quasi-experimental perturbation highlights the dual role that humans play in threatening and protecting species and ecosystems. Ecoimmunology is a rapidly developing field that explores how the environment shapes immune function, which in turn influences host-parasite relationships and disease outcomes. Host immune defence is a key fitness determinant because it underlies the capacity of animals to resist or tolerate potential infections. Importantly, immune function can be suppressed, depressed, reconfigured or stimulated by exposure to rapidly changing environmental drivers like temperature, pollutants and food availability. Thus, hosts may experience trade-offs resulting from altered investment in immune function under environmental stressors. As such, approaches in ecoimmunology can provide powerful tools to assist in the conservation of wildlife. Here, we provide case studies that explore the diverse ways that ecoimmunology can inform and advance conservation efforts, from understanding how Galapagos finches will fare with introduced parasites, to using methods from human oncology to design vaccines against a transmissible cancer in Tasmanian devils. In addition, we discuss the future of ecoimmunology and present 10 questions that can help guide this emerging field to better inform conservation decisions and biodiversity protection. From better linking changes in immune function to disease outcomes under different environmental conditions, to understanding how individual variation contributes to disease dynamics in wild populations, there is immense potential for ecoimmunology to inform the conservation of imperilled hosts in the face of new and re-emerging pathogens, in addition to improving the detection and management of emerging potential zoonoses., (© The Author(s) 2021. Despite decades of research, the role of elevated solar ultraviolet-B radiation (UVBR; 280-315 nm) in shaping amphibian populations remains ambiguous. These difficulties stem partly from a poor understanding of which parameters of UVBR exposure - dose, irradiance, and time interval - determine UVBR exposure health risk, and the potentially erroneous assumption that effects are proportional to the dose of exposure, irrespective of the administered regime (Bunsen-Roscoe Law of Reciprocity; BRL). We tested if the BRL holds with respect to UVBR-induced physiological effects in amphibians by acutely exposing tadpoles of the Australian green tree frog (Litoria caerulea) to a combination of different UVBR irradiances and doses in a fully factorial experiment. The BRL was invalid across all metrics assessed, with UVBR irradiance influencing the effects of a given dose on growth, coloration and burst swimming performance of larvae. We demonstrated some of the first empirical evidence for beneficial physiological effects of UVBR exposure in a larval amphibian, with improvements in growth, burst swimming performance and survival at the highest UVBR doses, contrary to hypotheses. Our findings demonstrate the species-specific nature of amphibian responses to UVBR, and the importance of UVBR irradiance in influencing the long-term physiological effects of a given dose of radiation.
