Abstract High-grade serous ovarian cancer (HGSOC) represents the most common and lethal subtype of ovarian cancer. Despite initial response to platinum-based standard therapy, patients commonly suffer from relapse that likely originates from drug-tolerant persister (DTP) cells. We generated isogenic clones of treatment-naïve and cisplatin-tolerant persister HGSOC cells. In addition, single-cell RNA sequencing of barcoded cells was performed in a xenograft model with HGSOC cell lines after platinum-based therapy. Published single-cell RNA-sequencing data from neo-adjuvant and non-treated HGSOC patients and patient data from TCGA were analyzed. DTP-derived cells exhibited morphological alterations and upregulation of epithelial-mesenchymal transition (EMT) markers. An aggressive subpopulation of DTP-derived cells showed high expression of the stress marker ATF3. Knockdown of ATF3 enhanced the sensitivity of aggressive DTP-derived cells to cisplatin-induced cell death, implying a role for ATF3 stress response in promoting a drug tolerant persister cell state. Furthermore, single cell lineage tracing to detect transcriptional changes in a HGSOC cell line-derived xenograft relapse model showed that cells derived from relapsed solid tumors express increased levels of EMT and multiple endoplasmic reticulum (ER) stress markers, including ATF3. Single cell RNA sequencing of epithelial cells from four HGSOC patients also identified a small cell population resembling DTP cells in all samples. Moreover, analysis of TCGA data from 259 HGSOC patients revealed a significant progression-free survival advantage for patients with low expression of the ATF3-associated partial EMT genes. These findings suggest that increased ATF3 expression together with partial EMT promote the development of aggressive DTP, and thereby relapse in HGSOC patients.

Das Handbuch versteht sich als Brücke zwischen aktueller Forschung und Praxis. Es enthält Beiträge aus unterschiedlichen Disziplinen und gibt länderübergreifende Einblicke in die vielfältigen Arbeitsfelder des politischen Journalismus – mit Schwerpunkten auf Deutschland, Österreich und der Schweiz. Diese Zusammenführung unterschiedlicher Perspektiven verdeutlicht den integrativen Anspruch des 900 Seiten starken Werks. Es hat sich zum Ziel gesetzt, «den politischen Journalismus in modernen […] europäischen […] Demokratien einmal gesamthaft darzustellen» (S. 17). Dies war, wie Prinzing und Blum in ihrer Einleitung einräumen, ein arbeitsintensives Unterfangen, das sich aber – wie der Erfolg zeigt – gelohnt hat. Das Handbuch gewinnt sein Publikum in der Medien- und Kommunikationswissenschaft, in der Medienpraxis und Politik, in der PR- und Öffentlichkeitsarbeit sowie in der Verwaltung und den Staatskanzleien. Der Erfolg gründet in verschiedenen Stärken des Buches.

Background Lack of routine follow-up for trauma patients after hospital discharge likely contributes to high rates of injury-related complications in Cameroon. Mobile phone contact may facilitate timely follow-up and reduce disability for high-risk patients. A previous single-center study showed promising feasibility of mobile health (mHealth) triage, but generalizability remains unknown. We evaluated the feasibility and acceptability of implementing a postdischarge mHealth triage tool at four hospitals in Cameroon.Methods Trauma patients from four Cameroonian hospitals were contacted at 2 weeks, 1, 3, and 6 months postdischarge. Program feasibility was assessed by calculating the proportion of successful contacts and overall cost. Odds of successful contact were compared using generalized estimating equations across patient socioeconomic status. Acceptability was assessed using a structured patient survey at 2 weeks and 6 months postdischarge.Results Of 3896 trauma patients, 59% were successfully contacted at 2 weeks postdischarge. Of these, 87% (1370/1587), 86% (1139/1330), and 90% (967/1069) were successfully reached at the 1-month, 3-month, and 6-month timepoints, respectively. The median cost per patient contact was US$3.17 (IQR 2.29–4.29). Higher socioeconomic status was independently associated with successful contact; rural poor patients were the least likely to be reached (adjusted OR 0.11; 95% CI 0.04 to 0.35). Almost all surveyed patients reported phone-based triage to be an acceptable follow-up method.Conclusion Telephone contact is a feasible and acceptable means to triage postdischarge trauma patients in Cameroon. While scaling an mHealth follow-up program has considerable potential to decrease injury morbidity in this setting, further research is needed to optimize inclusion of socioeconomically marginalized groups.Level of evidence Level III, prospective observational study.

IntroductionMalignant pleural mesothelioma (MPM) is a neoplasm with dismal prognosis and notorious resistance to the standard therapeutics cisplatin and pemetrexed. Chalcone derivatives are efficacious anti-cancer agents with minimal toxicity and have, therefore, gained pharmaceutical interest. Here, we investigated the efficacy of CIT-026 and CIT-223, two indolyl-chalcones (CITs), to inhibit growth and viability of MPM cells and defined the mechanism by which the compounds induce cell death.MethodsThe effects of CIT-026 and CIT-223 were analyzed in five MPM cell lines, using viability, immunofluorescence, real-time cell death monitoring, and tubulin polymerization assays, along with siRNA knockdown. Phospho-kinase arrays and immunoblotting were used to identify signaling molecules that contribute to cell death.ResultsCIT-026 and CIT-223 were toxic in all cell lines at sub-micromolar concentrations, in particular in MPM cells resistant to cisplatin and pemetrexed, while normal fibroblasts were only modestly affected. Both CITs targeted tubulin polymerization via (1) direct interaction with tubulin and (2) phosphorylation of microtubule regulators STMN1, CRMP2 and WNK1. Formation of aberrant tubulin fibers caused abnormal spindle morphology, mitotic arrest and apoptosis. CIT activity was not reduced in CRMP2-negative and STMN1-silenced MPM cells, indicating that direct tubulin targeting is sufficient for toxic effects of CITs.DiscussionCIT-026 and CIT-223 are highly effective inducers of tumor cell apoptosis by disrupting microtubule assembly, with only modest effects on non-malignant cells. CITs are potent anti-tumor agents against MPM cells, in particular cells resistant to standard therapeutics, and thus warrant further evaluation as potential small-molecule therapeutics in MPM.

Abstract Enhanced expression of anti-apoptotic B-cell lymphoma 2 (BCL-2) protein is frequent in cancer. Targeting of BCL-2 with the specific inhibitor ABT-199 (Venetoclax) has significant clinical activity in malignant diseases such as chronic lymphocytic leukemia and multiple myeloma. The small molecule drug ABT-199 mimics the pro-apoptotic BCL-2 homology domain 3 of BH3-only proteins and blocks the hydrophobic BC-groove in BCL-2. We have previously shown that ABT-199 synergizes with the proteasome inhibitor (PI) bortezomib in soft tissue sarcoma derived cells and cell lines to induce apoptosis. Synergistic apoptosis induction relies on the pore-forming effector BAX and expression of the pro-apoptotic BH3-only protein NOXA. Bortezomib augments expression of NOXA by blocking its proteasomal degradation. Interestingly, shown here for the first time, expression of NOXA is strongly enhanced by ABT-199 induced integrated stress response (ISR). ISR transcription factors ATF3 & ATF4 mediate transactivation of the BH3-only protein NOXA which specifically inhibits the anti-apoptotic MCL-1. Thus, NOXA potentiates the efficacy of the BCL-2 inhibitor ABT-199 by simultaneous inhibition of MCL-1. Hence, ABT-199 has a double impact by directly blocking anti-apoptotic BCL-2 and inhibiting MCL-1 via transactivated NOXA. By preventing degradation of NOXA PIs synergize with ABT-199. Synergism of ABT-199 and PIs therefore occurs on several, previously unexpected levels. This finding should prompt clinical evaluation of combinatorial regimens in further malignancies.

Abstract Background Despite advances in treatment of patients with non-small cell lung cancer, carriers of certain genetic alterations are prone to failure. One such factor frequently mutated, is the tumor suppressor PTEN. These tumors are supposed to be more resistant to radiation, chemo- and immunotherapy. Results We demonstrate that loss of PTEN led to altered expression of transcriptional programs which directly regulate therapy resistance, resulting in establishment of radiation resistance. While PTEN-deficient tumor cells were not dependent on DNA-PK for IR resistance nor activated ATR during IR, they showed a significant dependence for the DNA damage kinase ATM. Pharmacologic inhibition of ATM, via KU-60019 and AZD1390 at non-toxic doses, restored and even synergized with IR in PTEN-deficient human and murine NSCLC cells as well in a multicellular organotypic ex vivo tumor model. Conclusion PTEN tumors are addicted to ATM to detect and repair radiation induced DNA damage. This creates an exploitable bottleneck. At least in cellulo and ex vivo we show that low concentration of ATM inhibitor is able to synergise with IR to treat PTEN-deficient tumors in genetically well-defined IR resistant lung cancer models.

Abstract This study evaluated the effect on physical and sensory properties of freezing Halved Avocados (HA) by Traditional Method (TM) in a continuous freezer and by electromagnetic field-assisted method (EM) with CAS (Cells Alive System) freezer, obtaining HATM and HAEM, respectively. Each HA from the same specimen (obtaining 2 HA: one for fresh and the other for frozen-thawed state) was coded and evaluated. The firmness of the upper part of the fresh HA was higher (p < 0.05) than the lower part. The freezing curve showed the crystallization phase in the HAEM (from 14.25 to 20.33 min), thus indicating the formation of ice crystals, as well as affecting the significant decrease (p < 0.05) in more than 60% of firmness in the thawed HA concerning the fresh state. Regarding the color, there were differences (p < 0.05) in saturation (C*) between fresh and thawed avocado (HATM and HAEM); however, there were no differences (p > 0.05) in the tonality (h*). Weight loss of thawed HA was less than 0.5%. The sensory evaluation indicated there were no significant differences (p < 0.05) in firmness between the HATM and HAEM using the triangular test.

Abstract Background Photodynamic therapy with a photosensitizer such as protoporphyrin-IX, a light sensitive metabolite of heme synthesis, is a highly selective treatment for various carcinomas. In previous studies, we found a significant down regulation of the relevant enzyme ferrochelatase in gastrointestinal carcinomas leading to an accumulation of protoporphyrin-IX within the tumor cells. Recent studies showed that a novel anti-cancer drug, Alectinib, an orally available, highly selective, potent second-generation inhibitor of anaplastic lymphoma tyrosinkinase binds to ferrochelatase. Therefore, we were interested to see whether Alectinib treatment might lead to an accumulation of protoporphyrin IX. Methods Tumor cells of different origin were cultured, treated with LED-light and Alectinib. Results were gained by flow cytometry, immunohistochemistry and western blotting. Apoptosis was determined by flow cytometric analysis of Annexin V-FITC stained cells. In addition, cells were counterstained with propidium iodide to distinguish early apoptotic cells and late apoptotic/necrotic cells. Results Here, we report that photodynamic treatment of tumor cell lines of different origin in combination with Alectinib increased protoporphyrin-IX specific fluorescence and concomitantly cell death. Conclusions The usage of Alectinib could be another step for enhancing the effectiveness of photodynamic therapy. Further experiments will show whether photodynamic therapy in combination with Alectinib could be a new strategy for the treatment of e.g. peritoneal disseminated carcinomas.

Abstract Hepatocellular carcinoma (HCC) represents a global health challenge with limited therapeutic options. Anti-angiogenic immune checkpoint inhibitor-based combination therapy has been introduced for progressed HCC, but improves survival only in a subset of HCC patients. Tyrosine-kinase inhibitors (TKI) such as sorafenib represent an alternative treatment option but have only modest efficacy. Using different HCC cell lines and HCC tissues from various patients reflecting HCC heterogeneity, we investigated whether the sorafenib response could be enhanced by combination with pro-apoptotic agents, such as TNF-related apoptosis-inducing ligand (TRAIL) or the BH3-mimetic ABT-737, which target the death receptor and mitochondrial pathway of apoptosis, respectively. We found that both agents could enhance sorafenib-induced cell death which was, however, dependent on specific BH3-only proteins. TRAIL augmented sorafenib-induced cell death only in NOXA-expressing HCC cells, whereas ABT-737 enhanced the sorafenib response also in NOXA-deficient cells. ABT-737, however, failed to augment sorafenib cytotoxicity in the absence of BIM, even when NOXA was strongly expressed. In the presence of NOXA, BIM-deficient HCC cells could be in turn strongly sensitized for cell death induction by the combination of sorafenib with TRAIL. Accordingly, HCC tissues sensitive to apoptosis induction by sorafenib and TRAIL revealed enhanced NOXA expression compared to HCC tissues resistant to this treatment combination. Thus, our results suggest that BH3-only protein expression determines the treatment response of HCC to different sorafenib-based drug combinations. Individual profiling of BH3-only protein expression might therefore assist patient stratification to certain TKI-based HCC therapies.

A new inverse kinematic-optical-thermal (IKOT) model is introduced to control the process temperature in laser assisted tape winding and placement processes. The optimum time-dependent laser power distribution is obtained by employing a grid of independent laser cells while keeping the temperature of substrate and tape at the target temperature. Multi-layer cylindrical hoop winding with laser grids of 1 × 1, 28 × 1, and 28 × 11 and helical winding of a pressure vessel with laser grids of 22 × 1 and 22 × 11 are considered. It is found that the optimized laser power distribution pattern remains the same during the consecutive hoop winding process while the total power reduces to compensate the heat accumulation. A more non-uniform laser power distribution is obtained for the helical winding because the substrate curvature changes drastically at the dome section of the pressure vessel. The change in the optimum total laser power is found to be almost constant for the helical winding case. Finally, the IKOT model is evaluated by analyzing the effect of the computational parameters on the optimized process temperature.

Objectives In Cameroon, long-term outcomes after discharge from trauma are largely unknown, limiting our ability to identify opportunities to reduce the burden of injury. In this study, we evaluated injury-related death and disability in Cameroonian trauma patients over a 6-month period after hospital discharge.Design Prospective cohort study.Setting Four hospitals in the Littoral and Southwest regions of Cameroon.Participants A total of 1914 patients entered the study, 1304 were successfully contacted. Inclusion criteria were patients discharged after being treated for traumatic injury at each of four participating hospitals during a 20-month period. Those who did not possess a cellular phone or were unable to provide a phone number were excluded.Primary and secondary outcome measures The Glasgow Outcome Scale—Extended (GOSE) was administered to trauma patients at 2 weeks, 1 month, 3 months and 6 months post discharge. Median GOSE scores for each timepoint were compared and regression analyses were performed to determine associations with death and disability.Results Of 71 deaths recorded, 90% occurred by 2 weeks post discharge. Media fragmentation and polarization have contributed to blurring the lines between professional and non-professional journalism. Internationally, more fragmented-polarized media environments are often associated with the emergence of non-professional news providers, the weakening of journalistic standards, and the segmentation of audiences along ideological leanings. Furthermore, these environments are home to partisan and alternative news media outlets, some of which try to actively undermine the credibility of traditional mainstream media in their reporting. By following an audience-centric approach, this study investigates the consequences of more fragmented-polarized media environments and consumption habits on users’ perceptions of news media performance. We use online-survey data from five countries that differ in the extent of fragmentation and polarization in the media environment (CH = 1,859, DK = 2,667, IT = 2,121, PL = 2,536, US = 3,493). We find that perceptions of high news media performance are more likely to be expressed by citizens from less fragmented-polarized media environments. Positive perceptions of news media performance are also stronger among users of traditional media, and those who inform themselves in a more attitude-congruent manner. By contrast, citizens from more fragmented-polarized media environments and users of alternative news media tend to express less satisfaction with news media performance. Based on these results, we argue that perceptions of news media performance among news users are shaped by their individual media choices as well as by the composition of the news media environments that surrounds them.

Obesity increases the risk of developing or worsening many disorders particularly type 2 diabetes, a difficult-to-control chronic disease, treated with different drugs even with insulin, frequently without reaching therapeutic goals neither reducing cardiovascular risk. Therapy becomes more complex when type 2 diabetes is associated with severe obesity. The use of bariatric and metabolic surgery in patients with severe obesity and diabetes is assessed, and the different surgical techniques, assumptions about its mechanism of action, selection of patients and results are also reviewed. Bariatric surgery has proven to be effective in the treatment of severe obesity and type 2 diabetes in patients with BMI >35 kg/m2. This type of surgery has shown to be acceptable and well tolerated in selected and low risk patients capable of treatment Bariatric surgery has sufficient scientific evidence in these patients. However, there is no strong evidence of metabolic surgery in patients with BMI

Even though pesticides constitute a very essential component of improved and modern agriculture, the abuse of pesticides has brought substantial poisoning worldwide, especially in developing countries. This review investigates the possible sources by which people are exposed to pesticides worldwide and the impact on their livelihood. The group who often are exposed to these chemicals includes, farm workers, children, farmers, and family members who have direct link to the pesticide application sites as well as persons who are exposed to these pesticides through the domestic use of waterbodies and consumption of foodstuff or crops sprayed with these pesticides. Central to this review is the critical discussions of the different scientific research findings on health effects and risks related to pesticides usage. Again, organophosphates and organochlorines pesticides are found in most of the pesticide contaminated sites and can remain in soil and waterbodies for a longtime. This work has also provided cases of incidence of carcinogenicity in humans, as a result of pesticides use. We concluded that, breeding or developing insect tolerant or resistant crops may curb the profuse use of pesticides in agriculture.

Soybean is one of the promising food crops and feeds which contributes significantly to the development of agriculture in Burkina Faso. However, very few improved soybean varieties have been developed in the country. The introduction of new genotypes with high agronomic potential and adapted to the climatic conditions of the Sudanian zone of Burkina Faso will boost soybean production in the region. Twenty-four (24) newly introduced soybean genotypes were evaluated for their agromorphological and adaptation characteristics in the Sudanian zone of Burkina Faso. The variability parameters such as genotypic and phenotypic coefficient of variation, broad sense heritability, and expected genetic advance were estimated for 14 agromorphological characters. Objective: To evaluate the association between hyperglycemia treatment and mortality in patients with diabetes and COVID-19 in a Peruvian hospital. Methods: A retrospective cohort study was conducted between March and July 2020. Individual-level data were extracted from an implemented virtual platform. We included patients with type 2 diabetes hospitalized with COVID-19. The assessed outcome was in-hospital mortality. The Independent variable of interest was hyperglycemic treatment. We used Poisson regressions with robust variance to obtain crude and adjusted relative risks (RR) and their 95% confidence intervals (95% CI). Results: Out of 1635 patients hospitalized for COVID-19 during the study period, 248 patients with diabetes mellitus were included. The majority were men (66.9%), the median age was 62 years. Ninety-seven patients died in the hospital (39.1%). The median glycemia on admission was 222.5 mg/dL. At 48 h after hospital admission, 125 patients (50.4%) received sliding scale insulin alone (SSI), 46 (18.5%) received a fixed-dose insulin regimen. In the adjusted analysis, the group with SSI at 48 h of hospitalization had higher mortality than those with fixed-dose insulin (adjusted RR: 1.69; 95% CI: 1.01 – 2.83), and those and who continued with SSI in the following days had higher mortality compared to the group that switched to fixed-dose insulin (adjusted RR: 1.64; 95% CI: 1.17 – 2.32). Conclusion: Among assessed patients with diabetes and COVID-19, more than a third died during hospitalization. Early and continuous use of the sliding scale was associated with higher mortality compared to fixed-dose insulin regimens.

Abstract Background: There is a continued need for strategies to prevent influenza. While cetylpyridinium chloride (CPC), a broad-spectrum antimicrobial agent, has an extensive antimicrobial spectrum, its ability to affect respiratory viruses has not been studied in detail. Objectives: Here, we evaluate the ability of CPC to disrupt influenza viruses in vitro and in vivo. Methods: The virucidal activity of CPC was evaluated against susceptible and oseltamivir- resistant strains of influenza viruses. The effective virucidal concentration (EC) of CPC was determined using a hemagglutination assay and tissue culture infective dose assay. The effect of CPC on viral envelope morphology and ultrastructure was evaluated using transmission electron microscopy (TEM). The ability of influenza virus to develop resistance was evaluated after multiple passaging in sub-inhibitory concentrations of CPC. Finally, the efficacy of CPC in formulation to prevent and treat influenza infection was evaluated using the PR8 murine influenza model. Results: The virucidal effect of CPC occurred within 10 minutes, with mean EC50 and EC2log ranging between 5 to 20 µg/mL, for most strains of influenza tested regardless of type and resistance to oseltamivir. Examinations using TEM showed that CPC disrupted the integrity of the viral envelope and its morphology. Influenza viruses demonstrated no resistance to CPC despite prolonged exposure. Treated mice exhibited significantly increased survival and maintained body weight compared to untreated mice. Conclusions: The antimicrobial agent CPC possesses virucidal activity against susceptible and resistant strains of influenza virus by targeting and disrupting the viral envelope. Substantial virucidal activity is seen even at very low concentrations of CPC without development of resistance. Moreover, CPC in formulation reduces influenza-associated mortality and morbidity in vivo. ABSTRACT Objectives Cardiovascular risk estimated by several scores in patients with diabetes mellitus without a cardiovascular disease history and the association with carotid atherosclerotic plaque (CAP) were the aims of this study. Materials and methods Cardiovascular risk was calculate using United Kingdom Prospective Diabetes Study (UKPDS) risk engine, Framingham risk score for cardiovascular (FSCV) and coronary disease (FSCD), and the new score (NS) proposed by the 2013 ACC/AHA Guideline on the Treatment of Blood Cholesterol. Ultrasound was used to assess CAP occurrence. A receiver operating characteristic (ROC) analysis was performed. Results One hundred seventy patients (mean age 61.4 ± 11 years, 58.8% men) were included. Average FSCV, FSCD and NS values were 33.6% ± 21%, 20.6% ± 12% and 24.8% ± 18%, respectively. According to the UKPDS score, average risk of coronary disease and stroke were 22.1% ± 16% and 14.3% ± 19% respectively. Comparing the risks estimated by the different scores a significant correlation was found. The prevalence of CAP was 51%, in patients with the higher scores this prevalence was increased. ROC analysis showed a good discrimination power between subjects with or without CAP. Conclusion The cardiovascular risk estimated was high but heterogenic. The prevalence of CAP increased according to the strata of risk. Understanding the relationship between CAP and scores could improve the risk estimation in subjects with diabetes.

BackgroundRespiratory syncytial virus (RSV) is capable of transient viremia and extrapulmonary dissemination. Recently, this virus has been identified in fetal cord blood, suggesting the possibility of in utero acquisition in humans. Here, we assess permissivity and kinetics of RSV replication in primary human placental cells, examine their potential to transfer this infection to neighboring cells, and measure the inflammatory response evoked by the virus.Methods and findingsHuman placental villus tissue was collected immediately upon delivery and processed for isolation of placental cytotrophoblast, fibroblast, and macrophage (Hofbauer) cells. Isolated cells were infected with a recombinant RSV-A2 strain (rrRSV) expressing red fluorescent protein (RFP) and analyzed by fluorescence microscopy, Western blot, and quantitative PCR (qPCR). Based on RFP expression, rrRSV exhibited differential tropism for the three major placental cell types. Placental fibroblasts and Hofbauer cells were permissive and supported productive rrRSV replication. While infected cytotrophoblast cells expressed viral glycoprotein (G protein), only limited RSV replication was detected. Importantly, qPCR and fluorescence-focused unit assay revealed that the viral progeny remains trapped within infected Hofbauer cells for up to 30 days, with no release into surrounding media. Yet, Hofbauer cells passed the infection onto overlaid naïve 16HBE cells, suggesting contact-dependent trans-infection. Lastly, a significant increase in proinflammatory cytokines, particularly IL-6, TNF-alpha, and IFN-gamma was measured in the supernatant of infected Hofbauer cells by multiplex cytokine assay and conventional ELISA.ConclusionsThis study demonstrates that RSV can replicate in human placenta, exhibits differential tropism for distinct placental cell types, can be stored and transferred to neighboring cells by Hofbauer cells, and elicits an inflammatory response. It also supports the hypothesis that this respiratory virus can be vertically transferred to the fetus and potentially affect its development and the outcome of pregnancies.

For studying populism in a hybrid and high-choice media environment, the comparison of various media channels is especially instructive. We argue that populism-related communication is a combination of key messages (content) and certain stylistic devices (form), and we compare their utilization by a broad range of political actors on Facebook, Twitter, and televised talk shows across six countries (CH, DE, FR, IT, UK, and US). We conducted a content analysis of social media and talk show statements ( N = 2067) from 31 parties during a nonelection period of 3 months in 2015. We place special emphasis on stylistic devices and find that they can be grouped into three dimensions—equivalent to three dimensions used for populist key messages. We further find that political parties are generally more inclined to use populism-related communication on Facebook and Twitter than in political talk shows and that both new challenger parties and extreme parties use higher amounts of populist key messages and style elements.

In municipal governance around the world, the use of collective intelligence methods with dedicated tools and platforms is becoming the norm as a way to involve citizens, users and stakeholders in the design and implementation of policies. This new “open policy making” approach stands to benefit from the rise of artificial intelligence which can act as a cognitive agent to organize and summarize content, as well as a social agent interacting directly with participants. AI can also help fact check information and help generate automatic summaries and map concepts.

Objective: to describe the frequency of patients with diabetes mellitus type 2 hospitalized in Piura, Methods: we conducted a cross-sectional study in two hospitals of greater complexity care of EsSalud in Piura: Hospital III Jose Cayetano Heredia and Hospital II Jorge Reategui Delgado. Results: We obtained the clinical-demographic characteristics of the participants during the International Diabetes Day 2014. Of the 183 inpatients in both centers, 39 patients (21%) had diabetes mellitus type 2; the mean age was 67.8 ± 11.8. The medical records do not report hospital admission blood glucose and blood glucose during hospitalization in 17 and 24 participants, respectively. The medical records do not report weight of 31 participants. Diabetic foot was the primary cause of hospitalization. Conclusions: We conclude that diabetes mellitus type 2 is common in hospitalized patients of Essalud Piura, there is no proper record in the medical records of parameters such as weight and glycemic control.

The impact of obesity prevalence at early stages of life has been affected women at reproductive ages. Fertility rates, risk pregnancies, low weight at birth and cesarean section have been modified considerably. Bariatric surgery is a validated tool to improved fertility but malabsortion of nutrients as a consequence of surgery could affect mother and fetus nutrition and development. Gestacional diabetes diagnosis turns difficult in this group of obese patients after bariatric surgery. The aim of the recommendation was to offer tools for a good control and follow up of women at reproductive age.

The CYP2D6 gene encodes an enzyme important in the metabolism of many commonly used medications. Variation in CYP2D6 is associated with inter-individual differences in medication response, and genetic testing is used to optimize medication therapy. This report describes a retrospective study of CYP2D6 allele frequencies in a large population of 104,509 de-identified patient samples across all regions of the United States (US). Thirty-seven unique CYP2D6 alleles including structural variants were identified. A majority of these alleles had frequencies which matched published frequency data from smaller studies, while eight had no previously published frequencies. Importantly, CYP2D6 structural variants were observed in 13.1% of individuals and accounted for 7% of the total variants observed. The majority of structural variants detected (73%) were decreased-function or no-function alleles. As such, structural variants were found in approximately one-third (30%) of CYP2D6 poor metabolizers in this study. This is the first CYP2D6 study to evaluate, with a consistent methodology, both structural variants and single copy alleles in a large US population, and the results suggest that structural variants have a substantial impact on CYP2D6 function.

Pluripotent stem cells must strictly maintain genomic integrity to prevent transmission of mutations. In human induced pluripotent stem cells (iPSCs), we found that genome surveillance is achieved via two ways, namely, a hypersensitivity to apoptosis and a very low accumulation of DNA lesions. The low apoptosis threshold was mediated by constitutive p53 expression and a marked upregulation of proapoptotic p53 target genes of the BCL-2 family, ensuring the efficient iPSC removal upon genotoxic insults. Intriguingly, despite the elevated apoptosis sensitivity, both mitochondrial and nuclear DNA lesions induced by genotoxins were less frequent in iPSCs compared to fibroblasts. Gene profiling identified that mRNA expression of several antioxidant proteins was considerably upregulated in iPSCs. Knockdown of glutathione peroxidase-2 and depletion of glutathione impaired protection against DNA lesions. Thus, iPSCs ensure genomic integrity through enhanced apoptosis induction and increased antioxidant defense, contributing to protection against DNA damage.

Mehr als dreißig Jahre nach den Untersuchungen zur „optischen Kommentierung“ von Hans Mathias Kepplinger sind seither nur wenige Studien zur Darstellung von politischen Akteuren durch Kameraperspektiven und Einstellungsgrößen publiziert worden. Dabei haben kameratechnische Mittel neben verbalen Informationen einen nachgewiesenen Einfluss auf die Meinungsbildung der Rezipienten. Die vorliegende Studie untersucht inhaltsanalytisch 158 Fernsehnachrichtenbeiträge von zwei US-amerikanischen sowie deutschen Sendern in den Wahlkämpfen 2008 respektive 2009. Es zeigt sich, dass in den US-amerikanischen Sendungen Abweichungen von der Normalsicht und distanzierte Einstellungsgrößen angewendet werden. In den deutschen Beiträgen lassen sich ebenso Anwendungen von extremen Perspektiven feststellen, dafür wird stärker Nähe zu den Kandidaten hergestellt. Dabei ist länderübergreifend auf unterschiedliche Bildsprachen der Sender zu verweisen. Insgesamt zeigt sich transnational eine Zunahme abweichender Perspektiven und Einstellungsgrößen in den sogenannten Image Bites. Nicht zuletzt werden Unterschiede in den Darstellungen deutscher Spitzenkandidaten bei den ARD Tagesthemen festgestellt und deuten auf einen Visual Bias hin. Der Ländervergleich ermöglicht die Interpretation der Befunde vor dem Hintergrund unterschiedlicher Kommerzialisierungs- und Medialisierungsgrade.

Negativität ist ein zentrales Konzept in der Forschung zur politischen Kommunikation, doch hinsichtlich Konzeptualisierung und Operationalisierung besteht weitgehend Uneinigkeit. Auch beruhen viele Befunde auf der Analyse von Spezialzeiten wie dem Wahlkampf. Der vorliegende Beitrag zerlegt Negativität in die vier zentralen Dimensionen Tonalität, Konflikt, Untauglichkeit und Unvorteilhaftigkeit und untersucht die Politikberichterstattung von Leitmedien gezielt in den Routineperioden zwischen Wahlkämpfen. Er zieht Vergleiche zwischen drei deutschsprachigen Ländern (Deutschland, Österreich und der Schweiz), Online- und Offline-Angeboten, vier Mediengattungen (öffentlicher und privatwirtschaftlicher Rundfunk, Qualitäts- und Boulevardzeitungen) sowie verschiedenen Darstellungsformen und Themen. Es zeigt sich, dass Medien in Österreich, online und in meinungsbetonten Darstellungsformen negativer über Politik berichten als in ihren Vergleichsgruppen. Schweizerische Medien bringen mehr positiv gehaltene Beiträge und stellen politische Akteure seltener unvorteilhaft dar. Außerdem offenbart sich entgegen den gängigen Erwartungen, dass die Boulevardpresse unter allen Mediengattungen am wenigsten negativ schreibt.

Background: goliath grouper (Epinephelus itajara) is an economically valuable marine species and an excellent candidate for domestication for aquaculture purposes. If this grouper can osmoregulate in lowsalinity water, its cultivation can provide socio-economic benefits, for both coastal communities and the mainland agricultural sector. Objective: to evaluate the osmoregulatory capacity of juvenile goliath grouper when exposed to low-salinity water. Methods: juvenile goliath grouper (Epinephelus itajara) were either directly or gradually transferred from seawater to freshwater to test osmoregulatory ability. Body weight was assessed during acclimation and blood samples were taken to measure total osmolality and electrolytes. Results: all fish survived the transfer to freshwater and were maintained for up to 12 days after termination of the acclimation trials which lasted 72 hours. Juvenile goliath grouper were hyposmotic (342-462 mosmol/kg) to seawater and hyperosmotic (272-292 mosmol/kg) to freshwater. The gills and kidneys were found to have principal roles in the osmoregulatory processes. Numerous chloride cells were found on superficial regions of the gill filament epithelium, most likely serving to eliminate the excess of electrolytes while in seawater. The kidneys had numerous nephrons to make urine and retain electrolytes while in freshwater. Conclusions: these observations lead to the conclusions that juvenile goliath grouper have the ability to osmoregulate in freshwater and should be considered a marine euryhaline species. Such adaptability opens for consideration the possibility that goliath grouper could be successfully farmed in brackish water or even in freshwater.

Phloem osmolality and its components are involved in basic cell metabolism, cell growth, and in various physiological processes including the ability of living cells to withstand drought and frost. Osmolality and sugar composition responses to environmental stresses have been extensively studied for leaves, but less for the secondary phloem of plant stems and branches. Leaf osmotic concentration and the share of pinitol and raffinose among soluble sugars increase with increasing drought or cold stress, and osmotic concentration is adjusted with osmoregulation. We hypothesize that similar responses occur in the secondary phloem of branches. We collected living bark samples from branches of adult Pinus sylvestris, Picea abies, Betula pendula and Populus tremula trees across Europe, from boreal Northern Finland to Mediterranean Portugal. In all studied species, the observed variation in phloem osmolality was mainly driven by variation in phloem water content, while tissue solute content was rather constant across regions. Osmoregulation, in which osmolality is controlled by variable tissue solute content, was stronger for Betula and Populus in comparison to the evergreen conifers. Osmolality was lowest in mid-latitude region, and from there increased by 37% towards northern Europe and 38% towards southern Europe due to low phloem water content in these regions. The ratio of raffinose to all soluble sugars was negligible at mid-latitudes and increased towards north and south, reflecting its role in cold and drought tolerance. For pinitol, another sugar known for contributing to stress tolerance, no such latitudinal pattern was observed. The proportion of sucrose was remarkably low and that of hexoses (i.e. glucose and fructose) high at mid-latitudes. The ratio of starch to all non-structural carbohydrates increased towards the northern latitudes in agreement with the build-up of osmotically inactive C reservoir that can be converted into soluble sugars during winter acclimation in these cold regions. Present results for the secondary phloem of trees suggest that adjustment with tissue water content plays an important role in osmolality dynamics. Furthermore, trees acclimated to dry and cold climate showed high phloem osmolality and raffinose proportion.

Understanding synaptic connectivity and plasticity within brain circuits and their relationship to learning and behavior is a fundamental quest in neuroscience. Visualizing the fine details of synapses using optical microscopy remains however a major technical challenge. Super resolution microscopy opens the possibility to reveal molecular features of synapses beyond the diffraction limit. With direct stochastic optical reconstruction microscopy, dSTORM, we image synaptic proteins in the brain tissue of the fruit fly, Drosophila melanogaster. Super resolution imaging of brain tissue harbors difficulties due to light scattering and the density of signals. In order to reduce out of focus signal, we take advantage of the genetic tools available in the Drosophila and have fluorescently tagged synaptic proteins expressed in only a small number of neurons. These neurons form synapses within the calyx of the mushroom body, a distinct brain region involved in associative memory formation. Our results show that super resolution microscopy, in combination with genetically labelled synaptic proteins, is a powerful tool to investigate synapses in a quantitative fashion providing an entry point for studies on synaptic plasticity during learning and memory formation

There is a heated debate about the effect of global change on tropical forests. Many scientists predict large-scale tree mortality while others point to mitigating roles of CO2 fertilization and – the notoriously unknown – physiological trait acclimation of trees. In this opinion article we provided a first quantification of the potential of trait acclimation to mitigate the negative effects of warming on tropical canopy tree growth and survival. We applied a physiological tree growth model that incorporates trait acclimation through an optimization approach. Our model estimated the maximum effect of acclimation when trees optimize traits that are strongly plastic on a week to annual time scale (leaf photosynthetic capacity, total leaf area, stem sapwood area) to maximize carbon gain. We simulated tree carbon gain for temperatures (25-35ºC) and ambient CO2 concentrations (390-800 ppm) predicted for the 21st century. Full trait acclimation increased simulated carbon gain by up to 10-20% and the maximum tolerated temperature by up to 2ºC, thus reducing risks of tree death under predicted warming. Functional trait acclimation may thus increase the resilience of tropical trees to warming, but cannot prevent tree death during extremely hot and dry years at current CO2 levels. We call for incorporating trait acclimation in field and experimental studies of plant functional traits, and in models that predict responses of tropical forests to climate change.

The use of plants as expression hosts for recombinant proteins is an increasingly attractive option for the production of complex and challenging biopharmaceuticals. Tools are needed at present to marry recent developments in high-yielding gene vectors for heterologous expression with routine protein purification techniques. In this study we designed the Cysta-tag, a new purification tag for immobilized metal affinity chromatography (IMAC) of plant-made proteins based on the protein-stabilizing fusion partner SlCYS8. We show that the Cysta-tag may be used to rapidly purify proteins under native conditions, and then be removed enzymatically to isolate the protein of interest. We also show that commonly used protease recognition sites for linking purification tags are differentially stable in leaves of the commonly used expression host Nicotiana benthamiana, with those linkers susceptible to cysteine proteases being less stable then serine protease-cleavable linkers. As an example we describe a Cysta-tag experimental scheme for the one-step purification of a clinically useful protein, human α1-antitrypsin, transiently expressed in N. benthamiana. With potential applicability to the variety of chromatography formats commercially available for IMAC-based protein purification, the Cysta-tag provides a convenient means for the efficient and cost-effective purification of recombinant proteins from plant tissues.

Myeloid cell leukemia sequence 1 (Mcl-1), an antiapoptotic member of the Bcl-2 family, is often overexpressed in tumor cells limiting the therapeutic success. Mcl-1 differs from other Bcl-2 members by its high turnover rate. Its expression level is tightly regulated by ubiquitylating and deubiquitylating enzymes. Interaction of Mcl-1 with certain Bcl-2 homology domain 3 (BH3)-only members of the Bcl-2 family can limit the access to Mcl-1 ubiquitin ligase E3 and stabilizes the antiapoptotic protein. In addition, the overexpression of the deubiquitinase ubiquitin-specific protease 9x (USP9x) can result in the accumulation of Mcl-1 by removing poly-ubiquitin chains from Mcl-1 preventing its proteasomal degradation. Analyzing radiation-induced apoptosis in Jurkat cells, we found that Mcl-1 was downregulated more efficiently in sensitive parental cells than in a resistant subclone. The decline of Mcl-1 correlated with cell death induction and clonogenic survival. Knockdown of BH3-only proteins Bim, Puma, and Noxa did not affect Mcl-1 level or radiation-induced apoptosis. However, ionizing radiation resulted in activation of USP9x and enhanced deubiquitination of Mcl-1 in the radioresistant cells preventing fast Mcl-1 degradation. USP9x knockdown enhanced radiation-induced decrease of Mcl-1 and sensitized the radioresistant cells to apoptosis induction, whereas USP9x knockdown alone did not change Mcl-1 level in unirradiated cells. Together, our results indicate that radiation-induced activation of USP9x inhibits Mcl-1 degradation and apoptosis resulting in increased radioresistance.

Background: People with color vision deficiencies report numerous limitations in daily life. However, they use basic color terms systematically and in a similar manner as people with people with normal color vision. We hypothesize that a possible explanation for this discrepancy between color perception and behavioral consequences might be found in the gaze behavior of people with color vision deficiency.Methods: A group of participants with color vision deficiencies and a control group performed several search tasks in a naturalistic setting on a lawn.Results: Search performance was similar in both groups in a color-unrelated search task as well as in a search for yellow targets. While searching for red targets, color vision deficient participants exhibited a strongly degraded performance. This was closely matched by the number of fixations on red objects shown by the two groups. Importantly, once they fixated a target, participants with color vision deficiencies exhibited only few identification errors. Conclusions: Participants with color vision deficiencies are not able to enhance their search for red targets on a (green) lawn by an efficient guiding mechanism. The data indicate that the impaired guiding is the main influence on search performance, while foveal identification (verification) largely unaffected.

Over the last 10-20 years, comparative research in the feld of communication has almost become fashionable. Many factors are responsible for this, for example: an increased awareness of globalisation as a communication-driven process; an awareness of increased transnational conglomerization of media organizations; and the increasing use of the Internet which facilitates easier access to information around the world. But the big question is how to organize collaborative international communication research efectively? Which models of cooperation are available to us, and what are their advantages and disadvantages? In this article, I analyze fve ways of doing collaborative researches and their respective challenges.

In functional magnetic resonance imaging (fMRI) studies that apply a ‘subsequent memory’ approach, successful encoding is indicated by increased fMRI activity during the encoding phase for hits versus misses, in areas underlying memory encoding such as the hippocampal formation. Signal-detection theory (SDT) can be used to analyze memory-related fMRI activity as a function of the participant’s memory trace strength (d’). The goal of the present study was to use SDT to examine the relationship between fMRI activity during incidental encoding and participants’ recognition performance. To implement a new approach, post-experimental group assignment into High- or Low Performers (HP or LP) was based on 29 healthy participants' recognition performance, assessed with SDT. The analyses focused on the interaction between the factors group (HP vs. LP) and recognition performance (hits vs. misses). A whole-brain analysis revealed increased activation for HP versus LP during incidental encoding for remembered versus forgotten items (hits > misses) in the insula / temporo-parietal junction (TPJ) and the FFG. Parameter estimates in these regions exhibited a significant positive correlation with (d’). As these brain regions are highly relevant for salience detection (insula), stimulus-driven attention (TPJ), and content-specific processing of mnemonic stimuli (FFG), we suggest that HPs’ elevated memory performance was associated with enhanced attentional and content-specific sensory processing during the encoding phase. We provide first correlative evidence that encoding-related activity in content-specific sensory areas and content-independent attention and salience detection areas influences memory performance in a task with incidental encoding of facial stimuli. Based on our findings, we discuss whether the aforementioned group differences in brain activity during incidental encoding might constitute the basis of general differences in memory performance between HP and LP.

Viral marketing seeks to maximize the spread of a campaignthrough an online social network, often targeting influential nodes with highcentrality. In this article, we analyze behavioral aspects of influentialusers in trust-based product reviews communities, quantifying emotionalexpression, helpfulness, and user activity level. We focus on two independentproduct review communities, Dooyoo and Epinions, in whichusers can write product reviews and define trust links to filter productrecommendations. Following the patterns of social contagion processes, wemeasure user social influence by means of the k-shell decomposition of trustnetworks. For each of these users, we apply sentiment analysis to extracttheir extent of positive, negative, and neutral emotional expression. Inaddition, we quantify the level of feedback they received in their reviews,the length of their contributions, and their level of activity over theirlifetime in the community. We find that users of both communities exhibit alarge heterogeneity of social influence, and that helpfulness votes and ageare significantly better predictors of the influence of an individual thansentiment. The most active of the analyzed communities shows a particularstructure, in which the inner core of users is qualitatively different fromits periphery in terms of a stronger positive and negative emotionalexpression. These results suggest that both objective and subjective aspectsof reviews are relevant to the communication of subjective experience.

Recent evidence suggests that the experience of stress can be communicated between individuals via chemosensory cues. Little is known, however, about the impact of these cues on neurophysiological responses during a socially threatening situation. In the current investigation we implemented a widely used paradigm to study social exclusion—Cyberball—to examine whether chemosensory cues signalling anxiety modulate the neuronal effects of ostracism. In a double-blind, within-subjects design, 24 healthy, normosmic participants were presented with chemosensory cues of anxiety (or control samples) and completed the Cyberball task while in a 3T fMRI scanner. Axillary sweat collected from male students awaiting an oral examination served as the anxiety cues while the chemosensory control stimuli consisted of sweat collected from the same individuals participating in an ergometer training session. The neuroimaging data revealed that under the control chemosensory condition, exclusion from Cyberball was associated with significantly higher orbitofrontal cortex and anterior cingulate cortex activity, which is consistent with previous studies in the field. However, when participants were primed with the anxiety sweat, the activity in these regions was not observed. Further, under exposure to anxiety cues during ostracism the participants showed deactivations in brain regions involved in memory (hippocampus), social cognition (middle temporal gyrus, superior temporal gyrus) and processing of salience (inferior frontal gyrus). These results suggest that successful communication of anxiety via the chemosensory domain may moderate the experience of social exclusion. It is possible that the anxiety signals make it easier for the individuals to detach from the group, pointing to the communicative role of chemosensory anxiety cues in enhancing adjustment mechanisms in light of a distressing situation.

In a hypothesis-and-theory paper, a functional approach to movement analysis in sports is introduced. In this approach, contrary to classical concepts, it is not anymore the ideal movement of elite athletes that is taken as a template for the movements produced by learners. Instead, movements are understood as the means to solve given tasks that in turn, are defined by to-be-achieved task goals. A functional analysis comprises the steps of (1) recognising constraints that define the functional structure, (2) identifying sub-actions that subserve the achievement of structure-dependent goals, (3) explicating modalities as specifics of the movement execution, and (4) assigning functions to actions, sub-actions and modalities. Regarding motor-control theory, a functional approach can be linked to a dynamical-system framework of behavioural shaping, to cognitive models of modular effect-related motor control as well as to explicit concepts of goal setting and goal achievement. Finally, it is shown that a functional approach is of particular help for sports practice in the context of structuring part practice, recognising functionally equivalent task solutions, finding innovative technique alternatives, distinguishing errors from style, and identifying root causes of movement errors.

In the wild, animals have developed survival strategies relying on their senses. The individual ability to identify threatening situations is crucial and leads to increase in the overall fitness of the species. Rodents, for example have developed in their nasal cavities specialized olfactory neurons implicated in the detection of volatile cues encoding for impending danger such as predator scents or alarm pheromones. In particular, the neurons of the Grueneberg ganglion (GG), an olfactory subsystem, are implicated in the detection of danger cues sharing a similar chemical signature, a heterocyclic sulfur- or nitrogen-containing motif. Here we used a from the wild to the lab approach to identify new molecules that are involuntarily emitted by predators and that initiate fear-related responses in the recipient animal, the putative prey. We collected urines from carnivores as sources of predator scents and first verified their impact on the blood pressure of the mice. With this approach, the urine of the mountain lion emerged as the most potent source of chemical stress. We then identified in this biological fluid, new volatile cues with characteristic GG-related fingerprints, in particular the methylated pyridine structures, 2,4-lutidine and its analogues. We finally verified their encoded danger quality and demonstrated their ability to mimic the effects of the predator urine on GG neurons, on mice blood pressure and in behavioral experiments. In summary, we were able to identify here, with the use of an integrative approach, new relevant molecules, the pyridine analogues, implicated in interspecies danger communication.

Fractionation of biomass by filtration is a standard method for sampling planktonic microbes. It is unclear how the taxonomic composition of filtered biomass changes depending on sample volume. Using seawater from a marine oxygen minimum zone, we quantified the 16S rRNA gene composition of biomass on a prefilter (1.6 μm pore-size) and a downstream 0.2 μm filter over sample volumes from 0.05 to 5 L. Significant community shifts occurred in both filter fractions, and were most dramatic in the prefilter community. Sequences matching Vibrionales decreased from ~40-60% of prefilter datasets at low volumes (0.05-0.5 L) to less than 5% at higher volumes, while groups such at the Chromatiales and Thiohalorhabdales followed opposite trends, increasing from minor representation to become the dominant taxa at higher volumes. Groups often associated with marine particles, including members of the Deltaproteobacteria, Planctomycetes and Bacteroidetes, were among those showing the greatest increase with volume (4 to 27-fold). Taxon richness (97% similarity clusters) also varied significantly with volume, and in opposing directions depending on filter fraction, highlighting potential biases in community complexity estimates. These data raise concerns for studies using filter fractionation for quantitative comparisons of aquatic microbial diversity, for example between free-living and particle-associated communities.

Bodily movement has long been employed as a foundation for cultivating mental skills such as attention, self-control or mindfulness, with recent studies documenting the positive impacts of mindful movement training, such as yoga and tai chi. A parallel mind-body connection has also been observed in many developmental disorders. We elaborate a spectrum of mindfulness by considering ADHD, in which deficient motor control correlates with impaired (disinhibited) behavioral control contributing to defining features of excessive distractibility and impulsivity. These data provide evidence for an important axis of variation for wellbeing, in which skillful cognitive control covaries with a capacity for skillful movement. We review empirical and theoretical literature on attention, cognitive control, mind wandering, mindfulness and skill learning, endorsing a model of skilled attention in which motor plans, attention, and executive goals are seen as mutually co-defining aspects of skilled behavior that are linked by reciprocal inhibitory and excitatory connections. Thus, any movement training should engage higher-order inhibition and selection and develop a repertoire of rehearsed procedures that coordinate goals, attention and motor plans. However, we propose that mindful movement practice may improve the functional quality of rehearsed procedures, cultivating a transferrable skill of attention. We adopt Langer’s spectrum of mindful learning that spans from mindlessness to engagement with the details of the present task and contrast this with the mental attitudes cultivated in standard mindfulness meditation. We particularly follow Feldenkrais’ suggestion that mindful learning of skills for organizing the body in movement might transfer to other forms of mental activity. The results of mindful movement training should be observed in multiple complementary measures, and may have tremendous potential benefit for individuals with ADHD and other populations.

Im Rahmen des Kongresses „Literaturwissenschaften in Frankfurt, 1914-1945“, der von Bernd Zegowitz und Frank Estelmann am 20. und 21. Juni 2014 an der Universität Frankfurt am Main organisiert wurde, gaben 13 Vortragende an zwei Tagen Einblicke sowohl in die Geschichte als auch in exemplarische Werke von Literaturwissenschaftlern, die in der Zeit zwischen der Universitätsgründung im Jahr 1914 und dem Ende des Nationalsozialismus 1945 an der Universität Frankfurt lehrten und forschten.

We use functional magnetic resonance imaging to investigate short-term neural effects of a brief sensorimotor intervention adapted from the Feldenkrais method, a movement-based learning method. Twenty-one participants (10 men, 19-30 years) took part in the study. Participants were in a supine position in the scanner with extended legs while an experienced Feldenkrais practitioner used a planar board to touch and apply minimal force to different parts of the sole and toes of their left foot under two experimental conditions. In the local condition, the practitioner explored movement within foot and ankle. In the global condition, the practitioner focused on the connection and support from the foot to the rest of the body. Before (baseline) and after each intervention (post-local, post-global), we measured brain activity during intermittent pushing/releasing with the left leg and during resting state. Independent localizer tasks were used to identify regions of interest (ROI).Brain activity during left-foot pushing did not significantly differ between conditions in sensorimotor areas. Resting state activity (regional homogeneity, ReHo) increased from baseline to post-local in medial right motor cortex, and from baseline to post-global in the left supplementary/cingulate motor area. Contrasting post-global to post-local showed higher ReHo in right lateral motor cortex. ROI analyses showed significant increases in ReHo in pushing-related areas from baseline to both post-local and post-global, and this increase tended to be more pronounced post-local. The results of this exploratory study show that a short, non-intrusive sensorimotor intervention can have short-term effects on spontaneous cortical activity in functionally related brain regions. Increased resting state activity in higher-order motor areas supports the hypothesis that the global intervention engages action-related neural processes.
