Abstract

Pygocentrus nattereri is a widely distributed species in the Neotropical region and a potential bio-indicator. Kidneys have functions in fish physiology, allowing them to live in different environments. We aimed to compare the histological characteristics of caudal kidneys between males and females, associating them with the renosomatic index (RSI). For this purpose, 15 males and 14 females were used for biometric and histological analyses. Structural volumetric density (SVD), renal corpuscle histometric measures, and hemosiderin and lipofuscin deposit frequency in macrophages melanogenic (MMs) were assessed. No biometric differences were observed between the sexes, but body weight and standard length were correlated with RSI. The SVD showed difference in hematopoietic tissue between female and males, whereas the density of the other structures was not different. The RSI was positively associated with hematopoietic tissue and proximal tubule density in contrast to distal tubules, blood vessels, collecting ducts and MMs. Females exhibited a higher renal corpuscle area, glomerulus area, distal tubule diameter, collecting tubule area, and collecting tubule lumen area. These differences may be due to metabolic differences between males and females. Sex effect in P. nattereri may define punctual differences in future studies on the metabolism and immunity of this species. Resumo Pygocentrus nattereri é uma espécie amplamente distribuída na região Neotropical e pode ser considerada como um potencial bioindicador ambiental. Os rins têm funções cruciais na fisiologia dos peixes, permitindo-lhes viver em diferentes ambientes. Nosso objetivo foi comparar as características histológicas dos rins excretores entre machos e fêmeas, associando-as ao índice renossomático (IRS). Para tanto, 15 machos e 14 fêmeas foram utilizados para análises biométricas e histológicas. Amostras do tecido renal foram processadas para densidade estrutural volumétrica, medidas histométricas do corpúsculo renal e frequência dos depósitos de hemossiderina e lipofuscina nos macrófagos melonogênicos (MMs). Não foram observadas diferenças biométricas entre os sexos, mas a massa corporal e o comprimento padrão foram correlacionados com IRS. Apenas a densidade estrutural volumétrica do tecido hematopoiético foi diferente entre machos e fêmeas. Não houve diferença nas demais estruturas. O IRS associou-se positivamente com o tecido hematopoiético e densidade dos túbulos proximais e negativamente com os túbulos distais, vasos sanguíneos, ductos coletores e MMs. As fêmeas apresentaram maior área do corpúsculo renal, área do glomérulo, diâmetro do túbulo distal, área do túbulo coletor e área do lúmen do túbulo coletor. Essas diferenças podem ser devida às diferenças metabólicas entre machos e fêmeas. Particularidade do efeito de sexo em P. nattereri pode definir diferenças pontuais em futuros estudos sobre o metabolismo e imunidade nesta espécie.

Abstract

The liver is a well‐defined organ regarding its functions; however, it may present morphological variations among species. Since anurans may be exposed to toxic substances in their environment, which can disrupt liver's morphology and function, descriptive studies of organs are important to be carried out under natural condition. Thus, this study aimed to describe morphological aspects of liver in three Neotropical anuran species commonly used as experimental models: Physalaemus cuvieri (Leptodactylidae), Leptodactylus fuscus (Leptodactylidae) and Rhinella diptycha (Bufonidae). Anatomical, histological and ultrastructural analyses of six male samples of each species were carried out. A restricted analysis of the three species showed that livers' anatomy and ultrastructure are quite similar—they present three lobes, rounded nucleus polyhedral hepatocytes and some glycogen contents; however, histologically, there was a difference in the structural arrangements. In R. diptycha the parenchyma was arranged in a single cord, whereas in P. cuvieri and L. fuscus, there were double ones. This parenchyma structuring is highly variable considering Amphibia class. Knowledge about hepatic morphology and ultrastructure in amphibians is a key measure to detect changes in experimental studies and to infer about possible physiological and / or metabolic changes, when these animals are exposed to adverse conditions. Abstract

Hematological parameters can provide key information to an animal health status. However, this information is usually hard to obtain. Here, we described the hematological parameters of Leptodactylus podicipinus in the Brazilian Pantanal. We measured red blood cell morphometrics, erythrogram, and leukogram. We also tested for phylogenetic signal in the erythrogram and leukogram of 48 frog species from 15 families, testing if body size explains their variation. Lymphocytes were the most abundant leukocytes (>60%) in L. podicipinus , followed by neutrophils (∼10%). Given that L. podicipinus is an abundant and widely distributed species in central Brazil, knowing its hematological pattern can help establish a baseline and improve its use as a bioindicator of environmental degradation. Mean corpuscular hemoglobin and value contributed more to the phylomorphospace of erythrogram, in which Leptodactylus spp. and Hypsiboas raniceps had lower values of these variables, whereas Bufotes viridis and Hyla arborea had high values. The phylogenetic signal was spread throughout the dimensions of the leukogram phylomorphospace. The variables that most contributed to it were total leukocytes counts, lymphocytes, and neutrophils. We also found a moderate phylogenetic signal for both the erythrogram and leukogram. Accordingly, body size accounted for a low proportion of variation in both the leukogram (4.7%) and erythrogram (0.57%). Abstract

Testicular melanization in anurans may be absent or present with different degrees of intensity. When present, the color is due to the concentration of melanocytes in external (surface) and interstitial tissue. The function of this coloration is not fully known and we ask if melanization during testicular development, in species with highly pigmented testes in adults, is correlated with germ cells maturation. To do so, we selected four anuran species with highly pigmented testes. Species differed in the amount of interstitial melanin. Pigmentation increased throughout ontogeny, but with interspecific variation in the onset and rate. The rate of melanization progressed with testicular differentiation. A threshold for interstitial pigmentation seems to be reached with the advancement of spermatogenesis. Overall, testicular pigmentation adds a new chapter to the study of sexual maturity in anurans.

Abstract

Here, we tested the dose dependent and short- and long-term effects of benzo[alpha]pyrene (BAP) on the thickness of small intestine and liver melanomacrophages (MMC) of Hypsiboas albopunctatus. We conducted an experiment with a fully-crossed 3x2 factorial design to answer these two questions. Male specimens were separated into groups injected with a subcutaneous solution containing either 3 or 7 mg/Kg of BAP. Specimens were killed either 72h or 168h after the first injection with BAP. Then, we measured the the thickness of small intestine and the area occupied by MMCs in the liver. Experimental groups had 6 animals each. Our sampling units were pictures of histological sections (25 per animal). For the intestinal epithelium we only analyzed 5 out of the 6 animals.

Abstract

Amphibians have pigmented cells in organs beyond just skin. Their functions involve free radical neutralisation, early innate response, and a relationship with environmental temperature and UV light. In gonads, pigment containing-cells seem to be restricted mainly to the testes and related to sperm production. However, we report for the first time ovarian melanisation in Pseudis minuta and its ontogenetic changes in larval and postmetamorphic stages. Melanin containing-cells on the ovarian surface initially appear at early premetamorphic stages whereas in the cortex they occur later. In consecutive stages, melanin containing-cells were more evident among oocytes but without a clear pattern, being located randomly within the germinal epithelium or in the stroma. Although their function is unclear, a relationship with the fast acquisition of sexual maturity must be further explored. [ABSTRACT FROM AUTHOR]
