Esse artigo buscou descrever a utilização da odontologia legal na identificação de vítimas em desastres por meio de uma busca seletiva em banco de dados sobre o tema escolhido através de uma revisão.Para a coleta de dados, foi realizada uma busca extensa de artigos científicos nas seguintes bases de dados: PubMed, Scielo e Scopus, aonde os dados que foram para análise ​​serão discutidos na revisão de literatura, discussão e conclusão.Em resumo, que este estudo exponha a relevância da odontologia forense inserida no campo de identificação humana, trazendo todas as suas possibilidades de métodos de reconhecimento e suas utilizações.

Ximenia americana L. is popularly known as a plum or wild plum, which belongs to the Olacaceae family. Rhipicephalus (Boophilus) microplus represents one of the most important bovine ectoparasites that causes damage to the livestock sector. The objective of this study was to evaluate the acaricide effect of the X. americana stem bark hydroethanolic extract (EHXA) against R. (B.) microplus and its possible modulatory action when in association with cypermethrin, through the adult immersion test. The in vitro acaricide activity of the extract over R. (B.) microplus engorged females was tested using concentrations ranging from 5 to 80 mg mL− 1, which resulted in mortalities between 13.34 and 100 %, with a posture index (PI) between 0.038 and 0.226 and a posture inhibition index (PII) between 0.87 and 83.33 %. The modulatory effect of the EHXA (1.58 mg mL− 1) resulted in mortality between 0 and 46.6 %, with a posture index of 0.221 to 0.139 and a posture inhibition index between 3.01 and 38.49 %. The acaricide activity of the EHXA and its action on oviposition was demonstrated for the first time in this study, demonstrating the potential use of this extract as an auxiliary therapy in R. (B.) microplus control.

Undue exposure to antimicrobials has led to the acquisition and development of sophisticated bacterial resistance mechanisms, such as efflux pumps, which are able to expel or reduce the intracellular concentration of various antibiotics, making them ineffective. Therefore, inhibiting this mechanism is a promising way to minimize the phenomenon of resistance in bacteria. In this sense, the present study sought to evaluate the activity of the Carvacrol (CAR) and Thymol (THY) terpenes as possible Efflux Pump Inhibitors (EPIs), by determining the Minimum Inhibitory Concentration (MIC) and the association of these compounds in subinhibitory concentrations with the antibiotic Norfloxacin and with Ethidium Bromide (EtBr) against strains SA-1199 (wild-type) and SA-1199B (overexpresses NorA) of Staphylococcus aureus. In order to verify the interaction of the terpenes with the NorA efflux protein, an in silico molecular modeling study was carried out. The assays used to obtain the MIC of CAR and THY were performed by broth microdilution, while the Efflux Pump inhibitory test was performed by the MIC modification method of the antibiotic Norfloxacin and EtBr. docking was performed using the Molegro Virtual Docker (MVD) program. The results of the study revealed that CAR and THY have moderate bacterial activity and are capable of reducing the MIC of Norfloxacin antibiotic and EtBr in strains of S. aureus carrying the NorA efflux pump. The docking results showed that these terpenes act as possible competitive NorA inhibitors and can be investigated as adjuvants in combined therapies aimed at reducing antibiotic resistance.

The number of arbovirus cases has increased in recent years, demonstrating a need for investing in effective control actions. Among these actions, are strategies using biological control vectors, a field where Wolbachia pipientis has shown itself as useful. Wolbachia pipientis, an obligatory intracellular Gram-negative bacteria, which parasites arthropods naturally or through laboratory-induced infections, is capable of manipulating the reproduction of its host. A systematic literature review gathering studies on this bacteria over last 10 years (2007-2021) was performed given its important role in the reduction of insect disease vectors. 111 articles were found, from which 78 were used in this study. Information on the Wolbachia biology, mechanism of action and potential for the biological control of insect disease vectors was gathered. The present study may contribute to the knowledge surrounding the bacterium, as well as stimulate the production of other studies with the same theme.

The aim of this study was to evaluate the direct antibacterial activity, antibiotic modulation and toxicity of the essential oil of Ocimum gratissimum L. and its major compound Eugenol. The phytochemical analysis of the essential oil from leaves was investigated by Gas Chromatography coupled to Mass Spectrometry (CG-MS) and Eugenol was the major compound (63.25%). The Minimum Inhibitory Concentration and clinical antibiotics modulatory activities were performed using the 96-well plate microdilution methodology. Drosophila melanogaster was used as a model to assess the toxicity of the two substances. In conclusion, the eugenol and essential oil of Ocimum gratissimum L. showed direct antibacterial activity and associates with the tested antibiotics, which place them as promising agents against bacterial resistance. The essential oil showed less toxicity to fruit flies when compared to eugenol alone, although the results were numerically similar.

Thus, the aim of the present study was to investigate the antibacterial activity and the O-eugenol potential inO-eugenol presented a MIC of 1024 While we can conclude that the tested product has an efflux pump inhibitory effect, further studies are needed to elucidate its mechanisms of action, in addition to assays using other strains to verify whether the substance has the same inhibitory effect.

Spondias mombin is used in the folk medicine for the treatment of diarrhea and dysentery, indicating that extracts obtained from this species may present pharmacological activities against pathogenic microorganisms. The purpose of this work was to investigate the chemical composition and evaluate the antimicrobial activity of extracts obtained from the leaves (aqueous) and bark (hydroethanolic) of S. mombin both as single treatments and in combination with conventional drugs. Following a qualitative chemical prospection, the extracts were analyzed by HPLC–DAD. The antimicrobial activities were evaluated by microdilution. The combined activity of drugs and extracts was verified by adding a subinhibitory concentration of the extract in the presence of variable drug concentrations. The Minimum Fungicidal Concentration (MFC) was determined by a subculture of the microdilution test, while the effect of the in vitro treatments on morphological transition was analyzed by subculture in moist chambers. While the qualitative analysis detected the presence of phenols and flavonoids, the HPLC analysis identified quercetin, caffeic acid, and catechin as major components in the leaf extract, whereas kaempferol and quercetin were found as major compounds in the bark extract. The extracts showed effective antibacterial activities only against the Gram-negative strains. With regard to the combined activity, the leaf extract potentiated the action of gentamicin and imipenem (against Staphylococcus aureus), while the bark extract potentiated the effect of norfloxacin (against S. aureus), imipenem (against Escherichia coli), and norfloxacin (against Pseudomonas aeruginosa). A more significant antifungal (fungistatic) effect was achieved with the bark extract (even though at high concentrations), which further enhanced the activity of fluconazole. The extracts also inhibited the emission of filaments by Candida albicans and Candida tropicalis. Together, these findings suggest that that the extract constituents may act by favoring the permeability of microbial cells to conventional drugs, as well as by affecting virulence mechanisms in Candida strains.

Thus, the aim of the present study was to investigate the antibacterial activity and the O-eugenol potential in Staphylococcus aureus resistance reversal by efflux pump inhibition, as well as to evaluate its toxicity in the Drosophila melanogaster arthropod model. The broth microdilution method was used to determine the minimum inhibitory concentration (MIC) and the O-eugenol efflux pump inhibition. For the D. melanogaster toxicity assays, mortality and locomotor system damage were performed using the fumigation method. Results. O-eugenol presented a MIC of 1024 μg/mL against S. aureus. The association of this compound with the antibiotic tetracycline demonstrated a synergistic effect ( p < 0.0001 ), this also being observed when the antibiotic was associated with ethidium bromide ( p < 0.0001 ); thus, these results may be attributable to an efflux pump inhibition. The D. melanogaster mortality and geotaxis assays revealed the compound is toxic, with an EC50 of 18 μg/mL within 48 hours of exposure. Conclusions. While we can conclude that the tested product has an efflux pump inhibitory effect, further studies are needed to elucidate its mechanisms of action, in addition to assays using other strains to verify whether the substance has the same inhibitory effect.

Amburana cearensis (Allemão) ACSm. belongs to the Fabaceae family and occurs in the Brazilian semiarid, Argentina, Paraguay, Bolivia, and Peru. Numerous studies that portray its ethnobotany, use in popular medicine, chemical composition, and biological activities exist in the literature. This review aimed to provide an overview of the chemical composition, ethnopharmacology, and biological activities associated with A. cearensis and its isolated constituents. Information was collected from internet searches in the Scopus, Medline, PubMed, Google Scholar, and ScienceDirect databases were performed covering publications from 1997–2020. An ethnopharmacological literature analysis revealed that A. cearensis is used to treat a wide range of respiratory disorders in addition to intestinal, circulatory, and inflammatory problems. Coumarins, flavonoids, phenolic glycosides, phenolic acids, phenylpropanoid derivatives, and triterpenoids, among others, have been reported as active compounds, with High-Performance Liquid Chromatography (HPLC) being the main analytical technique used. The A. cearensis extracts and compounds presented several biological activities, including antimicrobial, antinociceptive, anti-inflammatory, antioxidant, neuroprotective, and myorelaxant activities, among others. This review provides a useful bibliography for future investigations and A. cearensis applications; however, future studies should focus on its toxic effects and the mechanisms of action of its extracts and isolated constituents to guide clinical applications.

Background: The emergence of fungal resistance to commercial drugs has been observed, and because of that, research with natural products have been performed with the aim of obtaining bioactive compounds. Objective: Evaluate the chemical composition and antifungal activity of the ethanolic extract of Costus cf. arabicus L leaves (EECAL) over strains of the genus Candida, as well as its inhibitory potential over yeast virulence. Methods: The composition of EECAL was analyzed through High Performance Liquid Chromatography (HPLC). The Minimum Inhibitory Concentration (MIC) was determined by broth microdilution using spectrophotometer readings and the Minimal Fungicidal Concentration (MFC) was investigated. The reading data of the MIC was used to trace a cellular growth curve and calculate the Inhibitory Concentration for 50% of the cells (IC50) of the extract and fluconazole. The effect over the yeast morphology was verified using wet-chamber microculture and visualized through optical microscopy (40x). Results: HPLC detected the presence of flavonoids and phenolic acids. The extract presented fungistatic effect (MIC of 8.192 µg/mL). The IC50 of the extract and fluconazole varied between 4,008.7 to 5,116.8 µg/mL and 44.0 to 83.1 µg/mL, respectively. The extract inhibited the formation of hyphae at MICx2 against CA LM 77. For the CA INCQS 40006, the inhibition was verified at MIC/2. In the CT LM 23 and CT INCQS 40042 strains the presence of hyphae was considered absent at the MIC. Conclusion: The extract presented antifungal action on cell growth at elevated concentrations and an inhibitory effect of dimorphism in the tested Candida species.

Purpose: Alternative methods for the use of animals in research have gained increasing importance, due to assessments evaluating the real need for their use and the development of legislation that regulates the subject. The principle of the 3R’s (replacement, reduction and refinement) has been an important reference, such that in vitro, ex vivo and cord replacement methods have achieved a prominent place in research. Methods: Therefore, due to successful results from studies developed with these methods, the present study aimed to evaluate the myorelaxant effect of the Dysphania ambrosioides essential oil (EODa) using a Sus scrofa domesticus coronary artery model, and the toxicity of both the Dysphania ambrosioides essential oil and its major constituent, α-terpinene, against Drosophila melanogaster in toxicity and negative geotaxis assays. Results: The EODa relaxed the smooth muscle of swine coronary arteries precontracted with K+ and 5-HT in assays using Sus scrofa domesticus coronary arteries. The toxicity results presented LC50 values of 1.546 mg/mL and 2.282 mg/mL for the EODa and α-terpinene, respectively, thus showing the EODa and α-terpinene presented toxicity to these dipterans, with the EODa being more toxic. Conclusions: Moreover, the results reveal the possibility of using the EODa in vascular disease studies since it promoted the relaxation of the Sus scrofa domesticus coronary smooth muscle.

Os fungos sao micro-organismos comuns no ambiente, sendo que alguns vivem de forma comensal, habitando a superficie da pele, mucosas, trato respiratorio, gastrointestinal e urogenital de humanos e animais, como e o caso das especies pertencentes ao genero Candida . O fluconazol e um dos farmacos mais usados para combater as enfermidades causadas por esses micro-organismos, atuando em um alvo especifico dos fungos, entretanto, muitas linhagens ja lhe sao resistentes. A resistencia a medicamentos tem despertado a necessidade de procurar novas alternativas de cunho natural. Libidibia ferrea popularmente conhecida como juca ou pau ferro e uma especie vegetal da familia Fabaceae, que possui em sua composicao substâncias avaliadas quanto a seu efeito antimicrobiano. Este estudo tem como objetivo verificar o efeito antifungico do extrato, sozinho e combinado com o Fluconazol na inibicao de crescimento de Candida albicans, Candida tropicalis e Candida kruzei, bem como na virulencia fungica. Os ensaios para determinacao da Concentracao Inibitoria de 50% dos micro-organismos (CI 50 ) , foram realizados por microdiluicao (8.192 a 8 μg/mL) e na combinacao extrato/farmaco, o extrato foi avaliado em concentracao subinibitoria (CFM/16). A Concentracao Fungicida Minima (CFM) foi verificada por subcultivo em meio solido. O efeito inibidor sobre a morfologia fungica foi verificado em câmaras umidas. O extrato apresentou efeito inibidor sobre os micro-organismos em concentracoes elevadas e sem relevância clinica, demonstrando um comportamento de resistencia ao extrato em questao. A CI 50 evidencia a potencializacao do farmaco pelo extrato, onde a combinacao dos dois reduziu a concentracao de efeito do fluconazol frente a todas as linhagens. A CFM foi ≥ 16.384 µg/mL. Em relacao a morfologia fungica, o extrato inibiu a transicao morfologica de C. albicans e C. tropicalis na concentracao mais elevada. A especie tem constituintes ativos para potencializacao de efeito de farmaco comercial e para interferir na expressao de um importante fator de virulencia fungica.

Caesalpinia ferrea C. Mart., popularly known as “Jucá” or “Pau-ferro”, belongs to the Fabaceae family, and is classified as a native and endemic species in Brazil. Numerous studies that portray its ethnobotany, chemical composition, and biological activities exist in the literature. The present study aimed to systematically review publications addressing the botanical aspects, uses in popular medicine, phytochemical composition, and bioactivities of C. ferrea. The searches focused on publications from 2015 to March 2020 using the Scopus, Periódicos Capes, PubMed, Google Scholar, and ScienceDirect databases. The leaves, fruits, seeds, and bark from C. ferrea are used in popular medicine to treat disorders affecting several systems, including the circulatory, immune, cardiovascular, digestive, respiratory, genitourinary, musculoskeletal, and conjunctive systems. The most commonly found chemical classes in phytochemical studies are flavonoids, polyphenols, terpenoids, tannins, saponins, steroids, and other phenolic compounds. The biological properties of the extracts and isolated compounds of C. ferrea most cited in the literature were antibacterial, antifungal, antioxidant, antiproliferative, anti-inflammatory, and healing potential. However, further studies are still needed to clarify a link between its traditional uses, the active compounds, and the reported pharmacological activities, as well as detailed research to determine the toxicological profile of C. ferrea.

The indiscriminate use of antibiotics contributes significantly to the selection of bacteria resistant to several antibiotics. Among the resistance mechanisms are the Efflux Pumps which are responsible for extruding solutes from the cell cytoplasm through proteins in the cell membrane. Because of this, new strategies are needed to control multidrug-resistant pathogenic strains. In this way, the objective of this study was to evaluate the antibacterial activity of eugenol by inhibition of TetK Efflux Pump in strains of Staphylococcus aureus resistant to Tetracycline, in addition to evaluating its toxicity in Drosophila melanogaster. To determine the Minimum Inhibitory Concentration (MIC), the broth microdilution method was used. The modulated effect of antibiotic and Ethidium Bromide associated with eugenol in subinhibitory concentrations (MIC/8) was evaluated. To evaluate the toxic effect of eugenol on D. melanogaster, fumigation tests were used, in which the parameters of mortality and damage to the locomotor system were evaluated. The results showed that eugenol has no direct activity in S. aureus, with an MIC ≥1024 μg/mL. However, it demonstrated that the synergistic potential when associated with Tetracycline, reducing the MIC of the antibiotic, already associated with Ethidium Bromide, had an antagonistic effect. When the toxicity in D. melanogaster was evaluated, eugenol demonstrated a non-toxic profile, since it presented EC50: 2036 μL/mL in 48 h of exposure. In conclusion, eugenol had no relevant direct effect against S. aureus, however, it potentialized the action of the antibiotic by decreasing its MIC.

The Staphylococcus aureus bacteria is a pathogen considered opportunistic and that has been acquiring resistance to several classes of antibiotics, mainly due to the synthesis of efflux pumps, which are proteins that expel these drugs intracellularly, reducing their effectiveness. The objective of this study was to evaluate the ability of isoeugenol to inhibit S. aureus efflux pumps and to determine its toxicity against a eukaryotic model (Drosophila melanogaster). IS-58, K2068 and K4414 S. aureus strains were used in the study. Isoeugenol minimum inhibitory concentration (MIC) and antibiotic modulation were evaluated in efflux pump inhibitory tests as well as in ethidium bromide (EtBr) assays. Toxicity tests against D. melanogaster assessed mortality and negative geotaxis. Isoeugenol obtained a relevant MIC result and a synergism was observed when isoeugenol was associated with the antibiotics, mainly with ciprofloxacin. Isoeugenol was able to affect all three efflux pumps tested, especially in strain K4414. The mortality of D. melanogaster caused by isoeugenol administration started after 12 h of exposure, being volume dependent and having an LC50 of 81.69 μL/L. In the negative geotaxis test, a statistical difference was observed after 24h of exposure compared to the control, demonstrating that damage to the locomotor apparatus had occurred. Based on the results, isoeugenol is a putative efflux pump inhibitor, becoming an alternative in blocking these proteins, and demonstrated acute toxicity against D. melanogaster.

Compounds capable of inhibiting the efflux pump mechanism are a promising alternative against bacterial resistance because, when combined with antibiotics, they can increase the effectiveness of these drugs by inhibiting active efflux. Elaiophylin, derived from Streptomyces hygroscopicus, is a natural antibiotic that exhibits a variety of biological activities, including antibacterial activity. However, its potential as an inhibitor of the bacterial efflux mechanism has not been investigated. This study evaluated the ability of Elaiophylin to inhibit the NorA efflux pump in Staphylococcus aureus strains. Therefore, tests were performed to obtain the Minimum Inhibitory Concentration (MIC) and to verify the ability of Elaiophylin to potentiate the MIC of the antibiotic Norfloxacin and Ethidium Bromide (EtBr), known substrates of NorA efflux. Real-time PCR and molecular docking assays were also performed to assess the potential of Elaiophylin against NorA. The strains SA-1199 (wild type) and SA-1199B (NorA over-expressed) of S. aureus were used for this study. The results showed that Elaiophylin significantly decreased the MIC of Norfloxacin and EtBr, increasing the activity of these substrates against S. aureus, which carries the efflux protein NorA. However, Elaiophylin provided a non-significant reduction in norA gene expression, however, molecular docking demonstrated a high binding affinity between Elaiophylin and NorA efflux protein, indicating that Elaiophylin can act as a potential NorA in S. aureus.

The increase in microorganisms with resistance to medications has caused a strong preoccupation within the medical and scientific community. Animal toxins studies, such as parotoid glandular secretions from amphibians, possesses a great potential in the development of drugs, such as antimicrobials, as these possess bioactive compounds. It was evaluated Rhinella jimi (Stevaux, 2002) glandular secretions against standard and multi-resistant bacterial strains; the effect of secretions combined with drugs; and determined the toxicity using two biologic in vivo models, and a in vitro model with mice livers. Standard strains were used for the determination of the Minimum Inhibitory Concentration (MIC), while for the modulatory activity of antibiotics, the clinical isolates Escherichia coli 06, Pseudomonas aeruginosa 03 and Staphylococcus aureus 10 were used. Modulatory activity was evaluated by the broth microdilution method with aminoglycosides and β-lactams as target antibiotics. The secretions in association with the antibiotics have a significant reduction in MIC, both the aminoglycosides and β-lactams. The toxicity and cytotoxicity results were lower than the values used in the modulation. R. jimi glandular secretions demonstrated clinically relevant results regarding the modulation of the tested antimicrobials.

Bioactive phytocompounds are studied by several bioactivities demonstrated, as their cytotoxic effects. The aim of this work was to evaluate the phytochemical profile, the toxic effect using the Drosophila melanogaster animal model and the anti-inflammatory and antimicrobial effect of the Alternanthera brasiliana (EEAB) ethanol extract. The phytochemical profile was performed using HPLC. The cytotoxic effect was evaluated in vivo using D. melanogaster. The anti-inflammatory effect was determined by neurogenic and antiedematogenic assays, and the antimicrobial activity was assayed using a microdilution method to determine the minimum inhibitory concentration (MIC) of the EEAB alone and in association with antibiotics. The main compound identified on the EEAB was luteolin (1.93%). Its cytotoxic effect was demonstrated after 24 h in the concentrations of 10, 20 and 40 mg/mL. The extract demonstrated an antiedematogenic effect, with a reduction of the edema between 35.57 and 64.17%. The MIC of the extract was ≥1.024 μg/mL, thus being considered clinically irrelevant. However, when the EEAB was associated with gentamicin, a synergism against all bacterial strains assayed was observed: Staphylococcus aureus (SA10), Escherichia coli (EC06) and Pseudomonas aeruginosa (PA24). Due to these results, the EEAB demonstrated a low toxicity in vivo and anti-inflammatory and synergistic activities. These are promising results, mainly against microbial pathogens, and the compounds identified can be a source of carbon backbones for the discovery and creation of new drugs.

Presently, results from a study carried out in this area using the essential oil from the Calyptranthes concinna species, a representative from the Myrtaceae family, are reported. The essential oil was obtained by hydrodistillation and gas chromatography coupled to mass spectrometry was used to identify its chemical constituents. Antibacterial activity was determined using the broth microdilution method, thus obtaining the Minimal Inhibitory Concentration (MIC) value, from which the subinhibitory concentration (MIC/8) was derived. The C. concinna essential oil presented antibacterial activity against both standard and multiresistant bacteria. In addition, the oil demonstrated an antibiotic activity potentiation against Staphylococcus aureus and Escherichia coli when in combination with the antibiotic gentamicin, reducing the MIC from 141.38 &mu, g/mL and 208.63 &mu, g/mL to 64 &mu, g/mL and 128 &mu, g/mL, respectively. Conclusions: Findings from the present study suggest this oil is promising in terms of its antimicrobial activity.

Eugenia uniflora is used in the Brazilian folk medicine to treat intestinal disorders and hypertension. However, scanty information exist on its potential toxicity to human, and little is known on its antioxidant activity in biological system. Hence, we investigated for the first time the potential toxic effects of ethanolic extract (EtOH) of E. uniflora (EEEU) in human leukocytes and erythrocytes, as well as its influence on membrane erythrocytes osmotic fragility. In addition, EEEU was chemically characterized and its antioxidant capacity was evaluated. We found that EEEU (1-480μg/mL) caused neither cytotoxicity nor DNA damage evaluated by Trypan blue and Comet assay, respectively. EEEU (1-480μg/mL) did not have any effect on membrane erythrocytes fragility. In addition, EEEU inhibited Fe2+-induced lipid peroxidation in rat brain and liver homogenates, and scavenged the DPPH radical. EEEU presented some polyphenolic compounds with high content such as quercetin, quercitrin, isoquercitrin, luteolin and ellagic acid, which may be at least in part responsible for its beneficial effects. Our results suggest that consumption of EEEU at relatively higher concentrations may not result in toxicity. However, further in vitro and in vivo studies should be conducted to ascertain its safety.

Among the bacterial resistance mechanisms, efflux pumps are responsible for expelling xenobiotics, including bacterial cell antibiotics. Given this problem, studies are investigating new alternatives for inhibiting bacterial growth or enhancing the antibiotic activity of drugs already on the market. With this in mind, this study aimed to evaluate the antibacterial activity of Estragole against the RN4220 Staphylococcus aureus strain, which carries the MsrA efflux pump, as well as Estragole's toxicity in the Drosophila melanogaster arthropod model. The broth microdilution method was used to perform the Minimum Inhibitory Concentration (MIC) tests. Estragole was used at a Sub-Inhibitory Concentration (MIC/8) in association with erythromycin and ethidium bromide to assess its combined effect. As for Estragole's toxicity evaluation over D. melanogaster, the fumigation bioassay and negative geotaxis methods were used. The results were expressed as an average of sextuplicate replicates. A Two-way ANOVA followed by Bonferroni's post hoc test was used. The present study demonstrated that Estragole did not show a direct antibacterial activity over the RN4220 S. aureus strain, since it obtained a MIC ≥1024 μg/mL. The association of estragole with erythromycin demonstrated a potentiation of the antibiotic effect, reducing the MIC from 512 to 256 μg/mL. On the other hand, when estragole was associated with ethidium bromide (EtBr), an antagonism was observed, increasing the MIC of EtBr from 32 to 50.7968 μg/mL, demonstrating that estragole did not inhibited directly the MsrA efflux pump mechanism. We conclude that estragole has no relevant direct effect over bacterial growth, however, when associated with erythromycin, this reduced its MIC, potentiating the effect of the antibiotic.

The use of plants with repellent properties has been evidenced as an important tool in the biocontrol of Culicidae that threaten public health. Studies on the main genera used for the biological control of these vectors were systematically reviewed including articles published from 2000 to 2019. Specialized databases such as Web of Science, Pubmed, Science Direct, Scopus BioMed, Bioline, Doaj and Scielo and the descriptors volatile oils, culicidae, arthropod vectors and medicinal plants were used. In the present study, 121 species distributed across 31 families were identified, with Lamiaceae, Myrtaceae and Verbenaceae possessing the largest number of species. For EO extraction virtually all plant parts were found to be used, including seeds, roots, rhizomes, stems, leaves, bracts, fruits, bark and inflorescences, these being mainly extract by hydrodistillation and steam distillation extraction. Gas chromatography coupled to mass spectrometry (GC-MS) was the main analytical technique used to identify the EO constituents. It is necessary to take into account the importance of developing alternative products which do not harm nature or the human species, nor any of the properties and uses these vegetables possess, which are essential for biological control.

Efflux pumps and β-lactamases are mechanisms of bacterial resistance that exist in Staphylococcus aureus, where both mechanisms are expressed simultaneously in the SA K4100 strain, with its efflux pump being characterized as QacC (Quaternary Ammonium Compounds C). The search for inhibitors of these mechanisms has grown gradually, with research on isolated compounds, including terpenes, which have innumerable biological activities, being common. This study sought to evaluate the antibacterial activity of Terpinolene against the S. aureus K4100 strain, carrying a QacC efflux pump and β-lactamase, as well as to evaluate its toxicity in the Drosophila melanogaster arthropod model. Determination of the Minimum Inhibitory Concentration (MIC) was performed by broth microdilution. Efflux pump inhibition was evaluated by the MIC reduction of Oxacillin and Ethidium Bromide (EtBr). β-Lactamase inhibition was analyzed by the MIC reduction of Ampicillin with Sulbactam. Toxicity was verified by mortality parameters and locomotor assays in D. melanogaster. The results demonstrated that Terpinolene did not present a direct antibacterial activity (MIC ≥ 1024 μg/mL). However, a reduction in MIC was observed when Terpinolene was associated with Oxacillin (161.26–71.83 μg/mL) and EtBr (45.25–32 μg/mL), possibly by a β-lactamase and efflux pump inhibition, thus evidencing a modulatory activity. Terpinolene presented D. melanogaster mortality with an EC50 of 34.6 μL/L within 12 h of exposure. Additionally, Terpinolene presented damage to the locomotor system after the second hour of exposure, with the effect increasing in a concentration-dependent manner. In conclusion, new tests should be carried out to investigate the Terpinolene reinforcement of antibiotic activity and toxic activity mechanisms of action.

This study aimed to investigate the myorelaxant action of the Dysphania ambrosioides essential oil (EODa) and its major constituent α-terpinene on tracheal smooth muscle isolated from rats. In tracheal smooth muscle ex vivo, in organ baths, isometric contractions recordings were done in order to evaluated the effect of EODa (1–1000 μg/mL) and α-terpinene (1–3000 μg/mL) on the following parameters: basal tone, contractions evoked by potassium (KCl 60 mM), acetylcholine (ACh 10 μM) or serotonin (5-HT 10 μM). The EODa and its major constituent α-terpinene, did not statistically alter basal tone; however, they induced myorelaxant effects on top of contractions induced by KCl, ACh and 5-HT. EODa and α-terpinene also inhibited the contractions induced by barium in presence of High [K+] (80 mM). The data suggest that the relaxation induced by these agents is caused by the inhibition of L-type VGCC, inhibiting the inward Ca2+ current through these channels, but does not exclude the possibility of participation of other mechanisms. Results from this study also suggest the EODa, due to their efficacy on relaxation of the respiratory tract, posses a therapeutic potential as a antispasmodic agent for respiratory tract.

Paraquat (PQ) is a widely used herbicide with no antidote which is implicated in the pathogenesis of the Parkinson's disease. The present study then investigated the potential of caffeic acid (CA), a known antioxidant, cardioprotective and neuroprotective molecule to counteract oxidative stress mediated by PQ. In addition, molecular docking was performed to understand the mechanism underlying the inhibitory effect of CA against PQ poisoning. The fruit fly, Drosophila melanogaster, was exposed to PQ (0.44 mg/g of diet) in the absence or presence of CA (0.25, 0.5, 1 and 2 mg/g of died) for 7 days. Data showed that PQ-fed flies had higher incidence of mortality which was associated with mitochondrial dysfunction, increased free Fe(II) content and lipid peroxidation when compared to the control. Co-exposure with CA reduced mortality and markedly attenuated biochemical changes induced by PQ. The mechanism investigated using molecular docking revealed a strong interaction (−6.2 Kcal/mol) of CA with D. melanogaster transcriptional activation of nuclear factor erythroid 2-related factor 2 (Nrf2). This was characterized by the binding of CA to keap-1 domain of Nrf2. Taking together these results indicate the protective effect of CA against PQ-induced oxidative damage in D. melanogaster was likely through its coordination which hinders Nrf2-keap-1 binding leading to an increase of the antioxidant defense system.

In recent decades, aquatic environment pollution has become a global challenge due to a rapid industrial growth. In this context, adsorption has become one of the most used methods, gaining importance especially in the last decades, given the growing concern over environmental damages caused by these pollutants. Additionally, its low cost and bioavailability are attractive. Therefore, the present study proposed to investigate the adsorption potential of A. indica seeds for Cu(II) and Pb(II) removal in synthetic solutions. Equilibrium studies were carried out via isothermal adsorption in batch systems. Kinetic studies were used to evaluate the lowest equilibrium time. The two evaluated ions were better suited to the Langmuir model and Freundlich model. When applying Langmuir's model on Neem seed powder adsorption, maximum adsorption capacities of 11.54 mg g−1 and 17.96 mg g−1, in the crude form and 11.41 mg g−1 and 19.80 mg g−1 in the alkaline form were obtained for Cu(II) and Pb(II) ions, respectively. The equilibrium time was approximately 80 and 100 min for both ions. The kinetic model best adjusted to the phenomena was the pseudo-second order (Ho Model), suggesting adsorption mainly has a chemical nature for both the studied metals. The results showed Neem seed powder may be a sustainable, efficient and low cost alternative for the removal of Cu(II) and Pb(II) metal cations present in effluents.

Eugenia uniflora L. (Myrtaceae family), also known as “pitanga”, is a tree species widely used in popular medicine. Despite the well documented beneficial effects of the extracts and essential oils from this plant, little is known about its toxicity. We performed a phytochemical fingerprinting and evaluated the toxicity induced by the Eugenia uniflora leaves essential oil in a Drosophila melanogaster model. In order to understand the biochemical mechanisms involved in E. uniflora essential oil toxicity, changes in the Nrf2 signaling as well as the hallmarks of oxidative stress were measured. The exposure of adult flies to the essential oil via a fumigant method resulted in increased mortality and locomotor deficits. In parallel, an oxidative stress response signaling, evidenced by changes in ROS production, lipid peroxidation, alterations in the activity of antioxidant enzymes and expression of Nrf2 protein targets occurred. In the light of our findings, attention is drawn to the indiscriminate use of this plant for medicinal purposes. In addition, a potential bio-insecticidal activity of Eugenia uniflora volatile compounds is suggested, a fact that needs to be further explored.

Context Melissa officinalis subsp. inodora Bornm. (Lamiaceae) has been used since ancient times in folk medicine against various diseases, but it has not been investigated against protozoa. Objective To evaluate the activities of M. officinalis against Leishmania braziliensis, Leishmania infantum and Trypanosoma cruzi as well as its cytotoxicity in fibroblast cell line. Materials and methods The fresh leaves were chopped into 1 cm(2) pieces, washed and macerated with 99.9% of ethanol for 72 h at room temperature. Antiparasitic activity of M. officinalis was accessed by direct counting of cells after serial dilution, while the cytotoxicity of M. officinalis was evaluated in fibroblast cell line (NCTC929) by measuring the reduction of resazurin. The test duration was 24 h. High-performance liquid chromatography (HPLC) was used to characterise the extract. Results The extract at concentrations of 250 and 125 μg/mL inhibited 80.39 and 54.27% of promastigote (LC50 value = 105.78 μg/mL) form of L. infantum, 80.59 and 68.61% of L. brasiliensis (LC50 value = 110.69 μg/mL) and against epimastigote (LC50 value = 245.23 μg/mL) forms of T. cruzi with an inhibition of 54.45 and 22.26%, respectively, was observed. The maximum toxicity was noted at 500 μg/mL with 95.41% (LC50 value = 141.01 μg/mL). The HPLC analysis identified caffeic acid and rutin as the major compounds. Raphiodon echinus (R. echinus) is used in Brazilian folk medicine for the treatment of inflammation, coughs, and infectious diseases. However, no information is available on the potential antioxidant, cytotoxicity and genotoxicity of this plant. In this study, the polyphenolic constituents, antioxidant capacity and potential toxic effects of aqueous and ethanolic extracts of R. echinus on human erythrocytes and leukocytes were investigated for the first time. R. echinus extracts showed the presence of Gallic, chlorogenic, caffeic and ellagic acids, rutin, quercitrin and quercetin. Aqueous and ethanolic extracts of R. echinus exhibited antioxidant activity in DPPH radical scavenging with IC50 = 111.9 μg/mL (EtOH extract) and IC50 = 227.9 μg/mL (aqueous extract). The extracts inhibited Fe(2+) (10 μM) induced thiobarbituric acid reactive substances (TBARS) formation in rat brain and liver homogenates. The extracts (30-480 μg/mL) did not induce genotoxicity, cytotoxicity or osmotic fragility in human blood cells. The findings of this present study therefore suggest that the therapeutic effect of R. echinus may be, in part, related to its antioxidant potential. Nevertheless, further in vitro and in vivo studies are required to ascertain the safety margin of its use in folk medicine.

Lectins from the Diocleinae subtribe (Leguminosae) are highly similar proteins that promote various biological activities with distinctly differing potencies. The structural basis for this experimental data is not yet fully understood. Dioclea rostrata lectin was purified and crystallized by hanging-drop vapour diffusion at 293 K. The crystal belongs to the orthorhombic space group I222, with unit-cell parameters a = 61.51, b = 88.22, c = 87.76 A. Assuming the presence of one monomer per asymmetric unit, the solvent content was estimated to be about 47.9%. A complete data set was collected at 1.87 A resolution.
