Artificial laboratory evolution was used to produce mutant strains of Escherichia coli and methicillin-resistant Staphylococcus aureus (MRSA) able to survive on antimicrobial metallic copper surfaces. These mutants were 12- and 60-fold less susceptible to the copper-mediated contact-killing process than their respective parent strains. Growth of the mutant and its parent in complex growth medium was similar. Tolerance to copper ions of the mutants was unchanged. The mutant phenotype remained stable over about 250 generations under non-stress conditions. The mutants and their respective parental strains accumulated copper released from the metallic surfaces to a similar extent. Nevertheless, only the parental strains succumbed to copper stress when challenged on metallic copper surfaces, suffering complete destruction of the cell structure. Whole genome sequencing and global transcriptome analysis were used to decipher the genetic alterations in the mutant strains; however, these results did not explain the copper-tolerance phenotypes on the systemic level. Instead, the mutants shared features with those of stressed bacterial sub-populations entering the early or "shallow" persister state. In contrast to the canonical persister state, however, the ability to survive on solid copper surfaces was adopted by the majority of the mutant strain population. This indicated that application of solid copper surfaces in hospitals and elsewhere has to be accompanied by strict cleaning regimens to keep the copper surfaces active and prevent evolution of tolerant mutant strains.Significance Microbes are rapidly killed on solid copper surfaces by contact-killing. Copper surfaces thus have an important role to play in preventing the spread of nosocomial infections. Bacteria adapt to challenging natural and clinical environments through evolutionary processes, for instance by acquisition of beneficial spontaneous mutations. We wishes to address the question whether mutants can be selected that have evolved to survive contact-killing on solid copper surfaces. We isolated such mutants from E. coli and S. aureus MRSA by artificial laboratory evolution. The ability to survive on solid copper surfaces was a stable phenotype of the mutant population and not restricted to a small sub-population. As a consequence, standard operation procedures with strict hygienic measures are extremely important to prevent emergence and spread of copper-surface-tolerant persister-like bacterial strains if copper surfaces are to be sustainably used to limit the spread of pathogenic bacteria e.g., to curb nosocomial infections. Macarrão Trepador - Hortivale, no período de setembro/2000 a fevereiro/2001, no Centro de Ciências Agrárias da Universidade Federal da Paraíba, em Areia. O delineamento experimental utilizado foi em blocos casualizados, com os tratamentos distribuídos em esquema fatorial (3 x 5) + 1, correspondendo às fontes (nitrato de cálcio, sulfato de amônio e uréia) e doses (0, 25, 50, 75 e 100kg.ha-1) de nitrogênio, e um tratamento adicional sem adubação (testemunha), com quatro repetições. As fontes nitrato de cálcio e sulfato de amônio, na dose de 100kg.ha-1, e uréia, na dose de 55kg.ha-1 de N, proporcionaram as produções de sementes no feijão-vagem de 2.571, 3.219 e 2.221kg.ha-1, respectivamente. O nitrogênio, em todas as fontes, influenciou positivamente a germinação e o vigor (índice de velocidade de germinação e emergência em campo) da semente do feijão-vagem. As doses de 68,8 e 49kg.ha-1 de N, fontes nitrato de cálcio e uréia foram responsáveis pelos valores máximos para a porcentagem de germinação, 72 e 75%, respectivamente. Para a fonte sulfato de amônio ocorreu aumento linear da porcentagem de germinação, à medida que se elevaram as doses de nitrogênio, sendo que na dose de 100kg.ha-1 obteve-se um porcentual de 84%. O índice de velocidade de germinação apresentou valores mais elevados, 6,0; 7,7 e 6,9 nas doses de 49; 71 e 53kg.ha-1 de N, fontes nitrato de cálcio, uréia e sulfato de amônio, respectivamente. A emergência em campo aumentou linearmente com elevação das doses de N (fonte nitrato de cálcio), sendo a emergência máxima (70%) obtida na dose de 100 kg ha-1 de N em cada fonte. O sulfato de amônio deve ser recomendado como fonte de N, em programas de produção de sementes de feijão-vagem. The application of three nitrogen sources in five concentrations each were studied in the physiological quality and yield of snap-bean seeds, cv. Macarrão Trepador Hortivale, from September/2000 to February/2001, in the Centro de Ciências Agrárias of the Universidade Federal of Paraíba, in Areia. A randomized complete block design was used, with treatments distributed in factorial scheme of (3 x 5) + 1, corresponding to nitrogen sources (calcium nitrate, ammonium sulfate and urea) and the second the five N levels (0, 25, 50, 75 and 100 kg.ha-1), and an additional treatment (without fertilization), in four replications. At 100 kg.ha-1 of N, the sources nitrate of calcium and sulfate of ammonium and urea at 55 kg.ha-1 of N presented the highest snap-bean seed yields, 2.571, 3.219 and 2.221 kg.ha-1, respectively, which surpassed the national average by 1.517, 1.219 and 1.221 kg.ha-1. The N in all the sources influenced the germination and the vigor positively (germination speed index and field emergence) of snap-bean seeds. At 68,8 and 49kg.ha-1 of N, calcium nitrate and urea were responsible for the maximum values for germination percentage, 72% and 75%, respectively. For ammonium sulfate there was a linear increase in the germination percentage, as the levels of N increased and at 100 kg.ha-1 of 84% germination was obtained. The germination speed Index showed higher values, 6,0; 7,7 and 6,9 at 49; 71 and 53 kg.ha-1 N, calcium nitrate, urea and ammonium sulfate, respectively. The field emergence increased linearly with calcium nitrate concentrations reaching the maximum emergence (70%) obtained at the 100 kg ha-1 N concentration in each source. Hemangiomas are the most common benign tumors of infancy. Despite their self-limited course, infantile capillary hemangiomas can impair vital or sensory functions as vision and cause cosmetic deformity. The usual treatments include oral/intralesional steroids, alpha interferon, cytotoxins, pulsed dye laser and cosmetic surgery resection. These treatments are not free of multiple complications and toxic side effects. This report describes the case of a 3-month-old female baby with progressively increasing hemangioma of the left upper eyelid impinging over the visual field. The hemangioma promptly responded to low-dose oral propranolol. A clinical response was noticed few days after the beginning of the treatment, with regression to 1/4 of its original size in 45 days of treatment, and to less than 1/10 after 8 months, free of any major side effects. Hemangiomas são os tumores benignos mais comuns durante o primeiro ano de vida. Apesar do seu curso autolimitado, os hemangiomas capilares podem prejudicar funções vitais ou sensoriais como a visão e causar alteração estética. O tratamento usual inclui esteróides orais ou intralesionais, interferon alfa, citotoxinas, laser e ressecção cirúrgica. Entretanto estes tratamentos não estão livres de complicações e efeitos adversos. Este relato descreve o caso de um bebê feminino de 3 meses com um hemangioma rapidamente progressivo na pálpebra superior esquerda, causando obstrução no eixo visual. O hemangioma respondeu rapidamente a uma baixa dose oral de propranolol. A resposta clínica foi notada poucos dias após o início do tratamento, com regressão a 1/4 do seu tamanho original após 45 dias de tratamento, e a menos de 1/10 após 8 meses, sem ter apresentado nenhum efeito adverso.

A questão central deste trabalho é demonstrar a viabilidade técnica e econômica da estabilização de solos com os cimentos pozolânicos de cinza de casca de arroz e de resíduo cerâmico. Para alcançar este objetivo o trabalho foi desenvolvido em duas etapas: a primeira referente ao desenvolvimento dos cimentos pozolânicos e a segunda referente à estabilização do solo propriamente dita. Em ambas as etapas adotou-se o critério da simplicidade e da possibilidade de utilização de equipamentos leves e de absorção de mão-de-obra não especializada. Os resultados técnicos e econômicos referentes ao cimento pozolânico de cinza de casca de arroz foram animadores; porém o mesmo não ocorreu com o cimento pozolânico de resíduo cerâmico que apresentou um desempenho em um patamar inferior. Quanto à estabilização de solos, os experimentos e a avaliação econômica efetuada mostram que a eficácia dos cimentos pozolânicos utilizados é limitada sendo difícil a obtenção de resultados comparáveis com o atualmente obtido com o cimento Portland. The main scope of this work is to demonstrate the technical and economical viability of the soil stabilization with pozzolanic cements made from rice husk ash and waste bricks and tiles. In order to reach this objective the work has been developed in two stages: the first referes to the development of the pozzolanic cement and the second one referes to the soil stabilization. In both these stages the criteria that has been adopted was of the simplicity, of the use of light equipments and of non skilled labour. The technical and economical results referring to the rice husk ash pozzolanic cement were encouraging; but the same was not the case with the waste bricks and tiles pozzolanic cement that had an inferior performance. As regards the soil stabilization, the tests and the economical evaluation show that the efficiency of the pozzolanic cement is limited thus the results achieved are not as good as of the Portland cement.

A maior preocupação deste trabalho é com as moradias das populações de baixa renda que na maioria dos casos são precárias não atendendo aos requisitos mínimos de habitabilidade. Para encarar este problema utilizamos como instrumento o conceito de tecnologia apropriada tendo em vista que as tecnologias tradicionais e modernas não tem apresentado bons resultados neste campo. Isto não quer dizer que estamos defendendo apenas uma saída tecnológica para os problemas da habitação popular. Para exemplificar a viabilidade deste enfoque, estudamos duas aplicações, em tijolos e em paredes monolíticas de solo-cimento, onde se colocaram as questões de escolha do solo, dosagem e ensaios de avaliação tais como: resistência à compressão, durabilidade, absorção d\'água, impermeabilidade. Procuramos também, através das aplicações, disseminar a metodologia empregada, que acreditamos possa ser utilizada no desenvolvimento de outros materiais. The principal objective of this Project is housing for low income people, wich is in the most part poorly constructed without the minimum standards of habitability. To face this problem, the appropriate technology approach were applied bering in mind that traditional and modern ones have not been suited to best resolve this type of housing. But this does not mean that this housing problem is only a technological challenge. As an example two cases were studied, one in pressed earth and the other in rammed earth, both using soil-cement. Soil selection, the cement to soil mixture, compressive strength, durability, water absorption and impermeability tests were evaluated. Besides these examples it is considered that this methodology can be applied to other studies of other materials.

A material that presents both the appropriate set of bulk properties in conjunction with an optimal surface performance is hardly found. For this reason, there is the need of modifying its surface. This is a standard procedure in many application fields but particularly important in the medical and environmental research. In this chapter, we describe the use of sputtering, as the chosen technology for the deposition of thin films. The use of the modified surfaces in the medical and environmental fields will be highlighted by two case studies in each one. In biomedicine, the surface modification of medical invasive devices for orthopedic and neural applications will be presented. For the environmental aspect, the results of two bioremediation tools, for arsenic and uranium removal, based on the immobilization of bacterial cells will be discussed.

Trabalho de Projeto do Mestrado Integrado em Engenharia Biomédica apresentado à Faculdade de Ciências e Tecnologia Os biomateriais utilizados em engenharia de tecidos são, normalmente, poliméricos. Dada a procura, cada vez maior, de materiais biodegradáveis, sobretudo de origem natural, os biopolímeros produzidos pelas bactérias são, de acordo com a literatura, adequados para aplicações biomédicas. Para além de propriedades como a biocompatibilidade, os scaffolds requerem um desempenho termomecânico superior aos polímeros biodegradáveis. Em resposta a este desafio, são produzidos compósitos de matriz polimérica reforçados com fibras naturais.Neste estudo foi avaliado o biopolímero produzido pela bactéria Phyllobacterium myrsinacearum, nunca referido na literatura científica ou em patentes. A caracterização revelou que a celulose bacteriana (CB) é, contrariamente a toda a celulose produzida por bactérias referida na literatura, hidrófoba. O biopolímero é constituído por fibrilas de comprimento micrométrico e com diâmetro nanométrico que apresenta uma estrutura semicristalina. Apresenta também propriedades mecânicas superiores às reportadas na literatura, nomeadamente no que diz respeito ao seu módulo de elasticidade e dureza.Os nanocompósitos de policaprolactona reforçados com CB foram produzidos por modificação ex situ e impressos tridimensionalmente por processo aditivo (3D). Tanto os homopolímeros de PCL (PCL 6800 e PCL 8502A) como os compósitos (PCL 6800/CB e PCL 8502A/CB) foram caracterizados quanto à sua cristalinidade, estrutura química, comportamento térmico e molhabilidade. As propriedades mecânicas foram avaliadas por ensaios de tração e flexão. A partir das caracterizações efetuadas foi concluído que as fibras de CB se comportam, eficazmente, como reforço mecânico Usually biomaterials used in tissue engineering are polymers. Given the demand for biodegradable materiais, mainly those of natural source, biopolymers produced by bacteria are, according to the literature, suitable for biomedical applications. Besides biocompatibility, scaffolds require a superior thermomechanical performance when compared to biodegradable polymers. As an answer to this challenge, composite materials of polymeric matrix reinforced with natural fibers are produced. In the present study, the biopolymer produced by the bacteria Phyllobacterium myrsinacearum, which was never reported in cientific literature or patents, was evaluated. The characterization revelead that bacterial cellulose (BC) is, instead of all cellulose produced by bacteria in literature, hydrophobic. The biopolymer consists on fibrils of micrometric length and nanometric diameter, and presents semicrystaline structure. It also presents superior mechanical properties compared to those reported in the literature, namely those which concern elastic modulus and hardeness. The CB-reinforced polycaprolactone nanocomposites were produced by ex situ modification and printed by additive manufacturing (3D printing). The hompolymers (PCL 6800/CB e PCL 8502A/CB) and composites (PCL 6800/CB e PCL 8502A/CB) were characterized by their crystallinity, chemical structure, thermal behaviour and wettability. The mechanical properties were evaluated by tensile and flexural tests. After characterization it was concluded that BC fibers acted, effectively, as a mechanical reinforcement. FCT

Dois aspectos importantes são levantados quanto ao tema o universo de problemas de segurança vinculados aos incêndios em edifícios, e o uso de estruturas de aço, diante deste universo. Apresenta-se uma abordagem global dos problemas de segurança contra incêndio nos edifícios, onde o fenômeno do incêndio é analisado visando compreender os parâmetros de seu desenvolvimento, e o papel da estrutura situado no contexto geral, sendo destacadas particularidades relativas ao emprego do aço. Dentro das medidas de segurança no projeto de edifícios de estrutura de aço, são enfocados o desempenho da estrutura na situação de incêndio, e os \"aspectos arquitetônicos da segurança contra incêndio\" que envolvem medidas importantes na concepção dos edifícios, para o controle do crescimento e propagação do fogo. As exigências de resistência ao fogo de códigos e normas de construção para as estruturas são colocadas em pauta, traçando-se um confronto entre as exigências brasileiras e as de alguns outros países do mundo. Discorre-se acerca dos sistemas de proteção mais usados em vários países para incrementar a resistência ao fogo da estrutura de aço, quando se fazem necessários, e são apresentados aqueles em uso entre nós. O trabalho conclui com uma afirmação quanto à confiabilidade e a segurança das estruturas de aço, quando adequadamente projetadas, mostrando-as como alternativa válida e importante para a construção de edifícios no Brasil. Two approaches may be adopted the problems related to fire in buildings, and the problem of use of steel in building structures due to fire hazard. The general view of a fire in building is presented, with emphasis of the structure role in the building during the fire. The fire process itself is analyzed in order to understand the parameters related to fire development in a building, and the steel behavior is specially discussed. Fire safety measurements during a steel-framed building design is considered with two focus: the performance of structure during a fire and the architectural measures to the prevention or control of fire spread. The fire resistance requirements in the standards and codes are presented trying to compare the Brazilian requirements with same of the other countries. The most useful fire protection systems adopted in several countries are discussed as to improve the fire safety of the steel structure, and the systems in use in this country are presented. In conclusion, the steel structure for buildings is showed as a reliable and safe structure when properly designed, and it is a valuable alternative to other structures in use in Brazil.

O trabalho propõe uma metodologia para a condução de pesquisas de desenvolvimento de métodos, processos e sistemas construtivos voltadas para a criação de inovações tecnológicas que possam efetivamente contribuir para a evolução da atividade de construção de edifícios. São analisados os principais conceitos que identificam-se com o desenvolvimento tecnológico dos meios de produção de edifícios e estabelecidas definições para os mesmos. São também analisados os condicionantes que delimitam o contexto deste desenvolvimento no país. A metodologia proposta, entendida como um conjunto de preceitos destinados a orientar a condução de uma investigação e que se fundamenta na busca da máxima racionalização construtiva, é apresentada em dois níveis, o da doutrina e o da estratégia. A metodologia foi aplicada no desenvolvimento de um inovador processo construtivo de alvenaria estrutural de blocos cerâmicos. Este processo é apresentado e descreve-se como se deu o seu desenvolvimento, dos estudos iniciais à construção de edifícios protótipos. Conclue-se pela importância que a adoção de uma metodologia específica representa para a busca de soluções válidas para o conjunto da sociedade e como pode contribuir para a mudança da caótica situação atual por que passa a construção civil. This thesis proposes a methodology to carry on the development research on building methods, processes and systems aiming at technological innovations which may effectivelly contribute for buildings construction activities evolution. The main concepts related to the technological development of the ways for the production process are analysed and their definitions established. It is also analysed the facts that restrain such development in Brazil. The proposed methodology is defined as a set of principles which objective is to point out the guidance for a scientific investigation and supported by the maximum building rationalization. This methodology is presented in two distinctive levels: the doctrine and the strategy. The methodology is applied specifically in the development of an innovative structural masonry process with ceramic blocks. The process is described, as well as the development performed stages: from the first studies to the prototype buildings construction. It is concluded that by adopting a specific methodology important solutions for the society as a whole are obtained and it gives essential support to change the disordered situation currently faced by the civil construction.
