Purpose Brucellosis is a zoonotic disease of great public health importance. In wild animals, Brucella abortus is one of the most diagnosed species, mainly in enzootic environments where domestic animals share the same environment. B. abortus is common in environments shared by cattle, wild, and domestic animals. This study aimed to detect the presence of B. abortus DNA in free-ranging and captivity felids at Mato Grosso State, Brazil. Method Polymerase chain reaction, based on the genetic element IS711, was performed in blood samples collected from 23 free-ranging and captive felids. The species represented include Leopardus colocolo, Leopardus pardalis, Leopardus wiedii, Panthera onca, Puma concolor, and Puma yagouaroundi. Results DNA amplification of B. abortus was observed in only one captive P. concolor (4.34%). Conclusion The detection of this pathogen in captive animals using molecular tools demonstrates the importance of monitoring, as it raises concerns about the possibility of transmission between humans and wild and domestic animals, especially in regions of vast biodiversity, such as in the State of Mato Grosso, Brazil.

Bacteria of the genus Providencia are opportunistic pathogens in humans, widely distributed in the environment and associated with greater resistance to antibiotics, and this being uncommon the association with clinical diseases. This study reports the isolation of P. stuartii in two sheep that presented clinical signs of pneumonia. At necropsy there was severe and acute fibrinopurulent bronchopneumonia. Histologically, there were infiltrated neutrophils and fibrin in the alveolar lumen, and the alveolar septa presented multifocal thickening with moderate proliferation of pneumocytes and mononuclear interstitial infiltrate. Providencia sp. was isolated in the microbiological tests of the lung and tracheal secretions. The isolate was subjected to DNA extraction, polymerase chain reaction (PCR) for the 16SrRNA gene and sequencing of genomic DNA, which demonstrated 100% homology with P. stuartii. This is the first report of the presence of this microorganism as a cause of interstitial and fibrinopurulent bronchopneumonia in sheep. Therefore, it is suggested that epidemiological surveillance strategies should be carried out in animals to better understand their role in the dissemination of this pathogen. Las bacterias del género providencia son patógenos oportunistas en el ser humano, ampliamente distribuidos en el medio ambiente y asociados a una mayor resistencia a los antibióticos, siendo infrecuente la asociación con enfermedades clínicas. Este estudio reporta el aislamiento de P. stuartii en dos ovejas que presentaban signos clínicos de neumonía. En la necropsia había bronconeumonía fibrinopurulenta aguda y grave. Histológicamente había neutrófilos infiltrados y fibrina en la luz alveolar, y los septos alveolares presentaban engrosamiento multifocal con moderada proliferación de neumocitos e infiltrado intersticial mononuclear. Providencia sp. se aisló en las pruebas microbiológicas de las secreciones pulmonares y traqueales. El aislado se sometió a extracción de ADN, reacción en cadena de la polimerasa (PCR) para el gen 16SrRNA y secuenciación del ADN genómico, que demostró una homología del 100% con P. stuartii. Este es el primer reporte de la presencia de este microorganismo como causa de bronconeumonía intersticial y fibrinopurulenta en ovinos. Por tanto, se sugiere que se lleven a cabo estrategias de vigilancia epidemiológica en animales para comprender mejor su papel en la diseminación de este patógeno. Portanto, sugere-se que estratégias de vigilância epidemiológica devam ser realizadas em animais para melhor compreensão de seu papel na disseminação desse patógeno.

Swine respiratory diseases such as atrophic rhinitis and bronchopneumonia caused by Pasteurella (P.) multocida cause important economic losses to the modern swine industry. The purpose of this study was to characterize P. multocida strains isolated from swine lungs by RAPD (Randomly Amplified Polymorphic DNA) to demonstrate their genetic diversity. Ninety-four samples of fragments from lungs with pneumonia and sixty one samples without pneumonia were collected in slaughterhouses in Mato Grosso during the period from December 2009 to March 2010. Clinical cases in 2012 and 2013 were also included in this study. Among the lung fragments with macroscopic lesions, without macroscopic lesions and clinical samples, 40.42%, 4.49% and 100% were positive for P. multocida, respectively. Bacterial identification culturing was confirmed by PCR (polymerase chain reaction) by means of the amplification of the gene kmt1. RAPD technique was performed for 46 isolates, and in every isolate, a total of 7 to 11 amplification bands were detected, composed of 8 clusters based on genetic similarity. Thus, treatment, control and preventive measures should consider the genetic diversity of P. multocida populations in swine herds in order to improve the development of new protocols to produce antimicrobials and vaccines. RESUMO: As doenças respiratórias suínas como a rinite atrófica e broncopneumonia, associada a Pasteurella (P.) multocida causam importantes perdas econômicas na suinocultura moderna. O objetivo deste trabalho foi caracterizar isolados de P. multocida de pulmão suíno através do Randomly Amplified Polymorphic DNA (RAPD) para demonstrar a diversidade genética. Noventa e quatro amostras de fragmentos de pulmões com lesões de pneumonia e sessenta e uma amostras sem lesão foram coletadas em frigoríficos no Estado do Mato Grosso, durante o período de dezembro de 2009 a março de 2010. Amostra de casos clínicos ocorridos em 2012 e 2013 também foram inlcuídos. Amostras de pulmões com lesões macroscópicas, sem lesões macroscópicas e amostras clínicas apresentaram presença de 40,42%, 4,49% e 100% de isolamento para P. multocida, respectivamente. Os isolados foram todos confirmados através da PCR (Polymerase Chain Reaction) pela amplificação do gene kmt1. A técnica de RAPD foi realizada em 46 amostras e em cada isolado foi detectado 7 a 11 bandas, que foram agrupadas em 8 grupos baseados em suas similaridades genéticas. Dessa forma, tratamento, controle e medidas preventivas deveriam considerar a diversidade genética da população de P. multocida em rebanhos suínos para melhorar o desenvolvimento de novos protocolos para produção de antimicrobianos e vacinas.

Introduction: Staphylococcus pseudintermedius is coagulase-positive species of the Staphylococcus intermedius group. It is an opportunistic pathogen that can cause infection in various parts of the body and has a zoonotic potential. Although studies on the pathogenicity and epidemiology of S. pseudintermedius are limited, it is known that this bacterium has several virulence factors, including toxins. These toxins can be classified into three main groups: pyrogenic toxins with superantigenic properties such as toxic shock syndrome toxin and staphylococcal enterotoxins, exfoliative toxins, and cytotoxins such as hemolysins and leukocidins. Methodology: In this study, the occurrence of eight toxin genes (sea, sec, tst, SIET, EXI, LuK F-I, Luk S-I, and hlg ƴ) was examined by PCR in 58 isolates of S. pseudintermedius from four domestic animal species. Results: All S. pseudintermedius isolates had at least one of the eight toxin genes. The predominant toxin genes were Luk S-I (95%), Luk F-I (91%), and EXI (91%), and the least prevalent gene was hlg ƴ (5%). Significant association (p = 0.0175) was found between the occurrence patterns of genes hlg ƴ and Luk F-I. Conclusions: The frequent occurrence of these genes in S. pseudintermedius obtained from diseased animals indicates that these toxins may play an important role in the pathogenesis of infection among domestic animals.

The aim of this study was to determine the prevalence and diversity of veterinary clinical isolates of Staphylococcus and analyze their antimicrobial susceptibility. One hundred Staphylococcus spp. clinical isolates from domestic and wild animals were subjected to partial sequencing of the 16S rRNA gene to species determination. Antimicrobial susceptibility was obtained by a disk diffusion test against six antibiotics: amoxicillin (AMX), cephalexin (LEX), ciprofloxacin (CIP), erythromycin (ERY), gentamicin (GEN) and trimethoprim-sulfamethoxazole (SXT). The most common specie was S. pseudintermedius (61%, 61/100) and resistance to ERY (57%, 57/100), SXT (50%, 50/100) and AMX (46%, 46/100) was detected most frequently. In total, 40% (40/100) of Staphylococcus spp. exhibited a multidrug-resistant (MDR) phenotype. Results of this study emphasize that animals are reservoir of MDR Staphylococcus spp.

RESUMO: Objetivou-se identificar microrganismos isolados de diferentes tipos de ceratite ulcerativa em cães, juntamente com a sua susceptibilidade a antimicrobianos. O resultado do tratamento médico e cirúrgico também foi correlacionado com o tipo de isolado. Amostras para microbiologia foram obtidas com auxílio de swab estéril em 104 olhos de 72 pacientes sem histórico prévio de tratamento com antibióticos tópicos, atendidos no período de maio de 2012 a março de 2015. Os antibióticos testados foram: neomicina, gentamicina, tobramicina, cloranfenicol, polimixina B, ciprofloxacino, ofloxacino e moxifloxacina. No total, 131 bactérias foram isoladas de 96/104 olhos estudados, sendo o gênero Staphylococcus (48,09%) predominante, seguido por Pseudomonas aeruginosa (16,01%). O Shih Tzu foi a raça mais prevalente (33,33%) e o número de isolados gram-negativos foi significativamente maior nessa raça, comparativamente aos Pinschers (p=0,003), aos Filas, aos Poodles e aos sem raça definida (p=0,046). As bactérias isoladas neste estudo apresentaram maior susceptibilidade ao ofloxacino (84,55%), que foi significativamente mais eficaz em relação a neomicina e a polimixina B (p

Toxoplasmosis is caused by Toxoplasma gondii, an obligatory intracellular protozoan, which establishes acute and chronic infections in birds and mammals, including humans. This note reports, for the first time, the detection and sequencing of DNA from T. gondii in the peripheral blood of a young free range giant anteater (Myrmecophaga tridactyla). For the diagnosis, the following methods were used: polymerase chain reaction (PCR) and positive serology (1:800) by means of the modified agglutination test (MAT). Since this species may be consumed by humans and predated by wild felids, its importance is emphasized as a probable source of zoonotic infection, in addition to its possible participation in the infection enzootic cycle. Although, parasitemia has been confirmed in this specimen, it presented no clinical sign of infection. RESUMO: A toxoplasmose é causada pelo Toxoplasma gondii, um protozoário intracelular obrigatório, que estabelece infecções agudas e crônicas em aves e mamíferos, incluindo humanos. Esta nota relata, pela primeira vez, a detecção e o sequenciamento do DNA de T. gondii em sangue periférico de um filhote de tamanduá-bandeira (Myrmecophaga tridactyla) de vida livre. Para o diagnóstico, os seguintes métodos foram utilizados: reação em cadeia pela polimerase (PCR) e sorologia positiva (1:800) por meio de teste de aglutinação modificado (MAT). Por se tratar de uma espécie que pode ser consumida por humanos e predada por felídeos silvestres, sua importância é ressaltada como uma provável fonte de infecção zoonótica, além da sua possível participação no ciclo enzoótico de infecção. Embora a parasitemia tenha sido comprovada neste espécime, ele não apresentava sinais clínicos de infecção.
