Bipolar disorder (BD) is a heritable disorder characterized by shifts in mood that manifest in manic or depressive episodes. Clinical studies have identified abnormalities of the circadian system in BD patients as a hallmark of underlying pathophysiology. Fibroblasts are a well-established in vitro model for measuring circadian patterns. We set out to examine the underlying genetic architecture of circadian rhythm in fibroblasts, with the goal to assess its contribution to the polygenic nature of BD disease risk. We collected, from primary cell lines of 6 healthy individuals, temporal genomic features over a 48 h period from transcriptomic data (RNA-seq) and open chromatin data (ATAC-seq). The RNA-seq data showed that only a limited number of genes, primarily the known core clock genes such as ARNTL, CRY1, PER3, NR1D2 and TEF display circadian patterns of expression consistently across cell cultures. The ATAC-seq data identified that distinct transcription factor families, like those with the basic helix-loop-helix motif, were associated with regions that were increasing in accessibility over time. Whereas known glucocorticoid receptor target motifs were identified in those regions that were decreasing in accessibility. Further evaluation of these regions using stratified linkage disequilibrium score regression analysis failed to identify a significant presence of them in the known genetic architecture of BD, and other psychiatric disorders or neurobehavioral traits in which the circadian rhythm is affected. In this study, we characterize the biological pathways that are activated in this in vitro circadian model, evaluating the relevance of these processes in the context of the genetic architecture of BD and other disorders, highlighting its limitations and future applications for circadian genomic studies.

Latina women living in the USA experience disproportionately higher rates of psychological distress compared to their non-Latina White counterparts. Poor maternal mental health during pregnancy can contribute to intergenerational mental health disparities. Through this pathway, mothers experiences, environments, and exposures (henceforth exposures) during pregnancy become biologically embodied and can negatively affect the fetus and life-long developmental trajectories of her child. One of the exposures that can affect mother-offspring dyads is the neighborhood. With the goal of integrating anthropological and sociological theories to explain mental health disparities among pregnant Latina women, we explored how perceptions of neighbor attitudes may influence mental health during pregnancy. We analyzed self-reported responses from 239 pregnant Latina women in Southern California (131 foreign-born, 108 US-born) on their mental health and perceived attitudes of their neighbors using multiple linear regression models. Among foreign-born Latina women, living in neighborhoods with more favorable views of Latinos was associated with lower depression scores (pooled β =  - .70, SE = .29, p = .019) and lower pregnancy-related anxiety scores (pooled β =  - .11, SE = .05, p = .021), but greater state anxiety scores (pooled β = .09, SE = .04, p = .021). Among US-born women, there were no associations between neighbor attitudes and mental health. Overall, results suggest that social environments are correlated with mental health and that foreign-born and US-born Latinas have varied mental health experiences in the USA. Our findings highlight the importance of improving aspects of neighborhood cohesion as part of maternal-fetal care management.

Tardigrades are microscopic animals that survive desiccation by inducing biostasis. To survive drying tardigrades rely on intrinsically disordered CAHS proteins, which also function to prevent perturbations induced by drying in vitro and in heterologous systems. CAHS proteins have been shown to form gels both in vitro and in vivo, which has been speculated to be linked to their protective capacity. However, the sequence features and mechanisms underlying gel formation and the necessity of gelation for protection have not been demonstrated. Here we report a mechanism of fibrillization and gelation for CAHS D similar to that of intermediate filament assembly. We show that in vitro, gelation restricts molecular motion, immobilizing and protecting labile material from the harmful effects of drying. In vivo, we observe that CAHS D forms fibrillar networks during osmotic stress. Fibrillar networking of CAHS D improves survival of osmotically shocked cells. We observe two emergent properties associated with fibrillization; (i) prevention of cell volume change and (ii) reduction of metabolic activity during osmotic shock. We find that there is no significant correlation between maintenance of cell volume and survival, while there is a significant correlation between reduced metabolism and survival. Importantly, CAHS Ds fibrillar network formation is reversible and metabolic rates return to control levels after CAHS fibers are resolved. This work provides insights into how tardigrades induce reversible biostasis through the self-assembly of labile CAHS gels.

Abstract The CMS detector, including its muon system, has been operating at the CERN LHC in increasingly challenging conditions for about 15 years. The muon detector was designed to provide excellent triggering and track reconstruction for muons produced in proton–proton collisons at an instantaneous luminosity ( $$\mathcal {L}$$ L ) of $$1 \times 10^{34}$$ 1 × 10 34 cm $$^{-2}$$ - 2 s $$^{-1}$$ - 1 . During the Run 2 data-taking period (2015–2018), the LHC achieved an instantaneous luminosity of twice its design value, resulting in larger background rates and making the efficient detection of muons more difficult. While some backgrounds result from natural radioactivity, cosmic rays, and interactions of the circulating protons with residual gas in the beam pipe, the dominant source of background hits in the muon system arises from proton–proton interactions themselves. Charged hadrons leaving the calorimeters produce energy deposits in the muon chambers. In addition, high-energy particles interacting in the hadron calorimeter and forward shielding elements generate thermal neutrons, which leak out of the calorimeter and shielding structures, filling the CMS cavern. We describe the method used to measure the background rates in the various muon subsystems. These rates, in conjunction with simulations, can be used to estimate the expected backgrounds in the High-Luminosity LHC. This machine will run for at least 10 years starting in 2029 reaching an instantaneous luminosity of $$\mathcal {L} = 5 \times \text {10}^\text {34}\,\text {cm}^\text {-2}\,\text {s}^\text {-1}$$ L = 5 × 10 34 cm -2 s -1 and increasing ultimately to $$\mathcal {L} = 7.5 \times \text {10}^\text {34}\,\text {cm}^\text {-2}\,\text {s}^\text {-1}$$ L = 7.5 × 10 34 cm -2 s -1 . These background estimates have been a key ingredient for the planning and design of the muon detector upgrade.

Abstract Background and objectives There is an urgent need to discover blood-based biomarkers of multiple sclerosis (MS) to better define the underlying biology of relapses and monitor disease progression. The main goal of this study is to search for candidate biomarkers of MS relapses associated with circulating extracellular vesicles (EVs), an emerging tool for biomarker discovery. Methods EVs, purified from unpaired plasma and CSF samples of RRMS patients by size-exclusion chromatography (SEC), underwent proteomic analysis to discover novel biomarkers associated with MS relapses. The candidate biomarkers of disease activity were detected by comparison approach between plasma- and CSF-EV proteomes associated with relapses. Among them, a selected potential biomarker was evaluated in a cohort of MS patients, using a novel and highly reproducible flow cytometry-based approach in order to detect low abundant EV subsets in a complex body fluid such as plasma. Results The proteomic profiles of both SEC-purified plasma EVs (from 6 patients in relapse and 5 patients in remission) and SEC-purified CSF EVs (from 4 patients in relapse and 3 patients in remission) revealed a set of proteins associated with MS relapses significant enriched in the synaptic transmission pathway. Among common proteins, excitatory amino-acid transporter 2, EAAT2, responsible for the majority of the glutamate uptake in CNS, was worthy of further investigation. By screening plasma samples from 110 MS patients, we found a significant association of plasma EV-carried EAAT2 protein (EV-EAAT2) with MS relapses, regardless of disease-modifying therapies. This finding was confirmed by investigating the presence of EV-EAAT2 in plasma samples collected longitudinally from 10 RRMS patients, during relapse and remission. Moreover, plasma EV-EAAT2 levels correlated positively with Expanded Disability Status Scale (EDSS) score in remitting MS patients but showed a negative correlation with age in patients with secondary progressive (SPMS). Conclusion Our results emphaticize the usefulness of plasma EVs as a source of accessible biomarkers to remotely analyse the CNS status. Plasma EV-EAAT2 showed to be a promising biomarker for MS relapses. Further studies are required to assess the clinical relevance of this biomarker also for disability progression independent of relapse activity and transition from RRMS towards SPMS.

The aim of the current study was to examine whether students' perceptions of teacher support at an individual-level, teacher support and well-functioning class climate at classroom-level, and teacher support and well-functioning class climate at school-level were associated with peer victimization. Data were obtained from a Student School Survey administered by the selected Swedish municipality. Multilevel analyses were based on 5,646 students in 277 classes and 27 schools. At the individual-level, girls and students who perceived greater teacher support than their classmates were victimized less often by their peers. In addition, students in schools with classes characterized by greater cooperation, cohesion, working atmosphere and respect toward their teachers tended to score lower on peer victimization. Within schools, students belonging to classes with a more well-functioning class climate than what was average in the school, and students belonging to classes that scored their teacher as more caring, fair and respectful compared to other classes in the school, were less likely to be targets of peer victimization.

This study designed a localized approach to working and writing action research suited for basic education. The researchers used multiphase mixed method research with three analysis stages focused on the identified problem. These stages include identifying the preferred approach in writing action research for basic education, designing a localized approach, and evaluating the developed action research approach. Five key informants participated in the preference assessment, while thirty respondents assessed the acceptability and significance levels of the designed localized approach. The key data analysis treatments employed qualitative data analysis, mean, and independent t-test. Preferred contextualized design for action research includes multiphase mixed-method research approaches. Grounded on the analyzed preference, the researchers designed the "Multi-Analysis Layer Nexus (MALN)" approach for basic education action research, featured with its focus approach matrix and a question-procedure-analysis (QPA) design alignment. The designed localized process received highly acceptable and highly significant ratings. No significant difference is derived from the two assessments. Recommendations included the conduct of MALN approach capacity building, skills and abilities profile, and data analysis treatments in the QPA matrix. The study is limited to the SALIKSIK (Strategic Action for Learning, Innovation, Knowledge Systems, and Instructional Keystones) Research Program of Lopez East and Lopez West Districts. Though, the DepEd Division of Quezon can use this for its localized action research approach. The developed MALN approach is an innovative, novel action research process specifically designed for basic education settings.

An on-farm observational study evaluated the use of individual automatic milk feeders (AMF) to wean 53 buffalo calves. Analysis of data retrieved from the AMF system over a 4-month period revealed positive indicators for animal adaptation, including rapid self-feeding behaviour, low incidence of feeding refusals, high milk intake, consistent weight gain, and increased frequency of daily feedings. The study identified potential areas for future research, such as determining the optimal number and frequency of feedings and the appropriate milk allowance for different stages of calf development. These findings suggest potential benefits for labour cost savings and emphasise the need to optimise AMF parameters to meet the specific needs of buffalo calves.

Abstract Background Thoracotomy is one of the surgical procedures most burdened by chronic post-operative pain. There is poor evidence regarding the possibility that even in pediatric patients, thoracotomy can be followed by post-operative pain. The primary objective of this analysis is to identify associations with home pain therapy, pain intensity, and possible protective factors acting on chronic pain in this population. Methods A retrospective cross-sectional study was conducted at Ospedale Pediatrico Bambino Gesù IRCCS. The study included pediatric patients undergoing thoracotomy. For statistical analyses, a logistic model and a zero-inflated strategy were implemented to explore associations and predict factors related to home-based analgesic therapy and pain intensity. Results Gender and age were identified as significant factors in the assignment of home therapy, with males having over seven times the risk compared to females (OR = 7.06, 95% CI = [2.11, 29.7]). At the last measurement, pain intensity was positively associated with age and the number of pain events during the week. Conclusions The study highlights significant factors influencing post-thoracotomy pain management in pediatric patients. These findings underscore the importance of tailored pain management strategies that consider gender and age to improve post-operative care and outcomes in pediatric thoracotomy patients.

This research use quantitative research methods that examines the impact of board independence, gender diversity, managerial ownership, institutional ownership, and ownership concentration on tax aggressiveness. It also investigates how audit quality moderates the relationship between these variables and tax aggressiveness. The research, based on a purposive sample of 566 companies from audited financial reports on the Indonesia Stock Exchange (BEI) spanning 2018 to 2022, uses panel regression with EViews version 12. Findings indicate that board independence, gender diversity, and ownership concentration do not significantly affect tax aggressiveness, while managerial and institutional ownership have a noteworthy impact. Ineffective implementation of independent commissioners' roles, male dominance in management, and the lack of expected theoretical impact on overseeing management do not influence tax aggressiveness. The research also shows that audit quality as moderating variable does not significantly moderate the relationship between these variables and tax aggressiveness.

This study aimed to describe the development of the Perceived Therapist’s Knowledge about Gender Identity Diversity Scale and to preliminarily validate this scale by describing its psychometric properties. This research instrument was constructed based on the existing literature and recommendations for instrument development. Initially, a 36-item scale was devised to assess perceived openness and knowledge about gender identity diversity in therapy. The content validation process involved 12 expert judges, leading to a refined 25-item scale. Participants consisting of 57 trans and non-binary Puerto Rican individuals completed the scale. Exploratory factor analysis revealed a unidimensional structure, supporting a single factor named “perceived knowledge about gender identity diversity in therapy.” The final scale demonstrated excellent reliability (α = 0.978; Sα = 0.980; ω = 0.979), indicating strong internal consistency. This validated scale contributes to assessing primarily Hispanic trans and non-binary individuals’ perceptions of their therapists’ knowledge about gender identity diversity.

Purpose The shared resource theory between motor and cognitive control maintains that the central nervous system shares the same resources to respond to cognitive and motor demands; that is, there are competing demands in cognitive-motor interaction. Methods Through correlations between motor control and working memory in individuals with neurological injuries, this study aims to provide empirical evidence to support the above theory. Motor control was assessed in postural control and Dominant Upper Extremity (DUE) function and activity. Working memory (WM) was assessed via neuropsychological tests and Cognitive Event-related Potentials (ERPs). Fifty-six individuals with neurological injuries between the ages of 19 and 55 years participated. Results The neuropsychological working memory tests applied (Working Memory Index of the Wechsler Adult Intelligence Scale IV, Trail Making Test – part B, and Corsi Block-Tapping Test – backward span sequence) showed significant correlations between DUE functions and activities (ability to grasp, transport and release, and daily life tasks) (rho = [0.27]–[0.47]). Global postural control and WM did not show significant correlations, and nor did dominant upper extremity motor control with P300 wave latency and N200-P300 amplitudes of ERPs, except for grip strength. Conclusions The present results do not provide conclusive empirical evidence of the cognitive-motor interaction, based on the study of relationships between WM and motor control of DUE. However, there are striking correlations between WM and DUE function and activity, especially referring to instrumental activities of daily living. This finding could apply to the rehabilitation of people with neurological injuries and cognitive impairments.

Introduction Breast cancer survivors often experience pre and post-treatment physical and psychological symptoms, negatively affecting their quality of life. Regular physical exercise is associated with better quality of life and lower recurrence of cancer, and therefore all oncological patients are recommended to practice it in a regular basis. Despite this, breast cancer survivors have low adherence to physical exercise. The purpose of this study is to identify barriers, facilitators and preferences of Chilean breast cancer survivors to practice physical exercise. Methods Phenomenological qualitative study of 12 in-depth interviews with adjuvant radiation therapy concluded at least three months ago. Results Breast cancer survivors ignored the benefits of physical exercise during and after treatment. The barriers were physical symptoms, psychological barriers, sociocultural barriers, health system barriers, disinformation and sedentary lifestyle. Facilitators were coping with physical symoptoms, psychological issues, having information and active lifestyle. The preferences were painless and familiar exercises. Preferred exercise was walking. Conclusions Breast cancer survivors may adhere to physical exercise despite barriers when certain facilitators are present, which may be promoted by the health team when reporting the benefits of the physical exercise, prescribing personalized, safe and painless physical exercise and educating both patient and her family about the role of the physical exercise in cancer recovering process.

Este estudio aborda la evolución del marketing digital en el sector bancario latinoamericano durante el periodo 2020-2024, en un contexto marcado por la rápida digitalización impulsada por la pandemia de COVID-19. Su propósito es analizar las evidencias e investigaciones previas sobre el tema en el contexto latinoamericano. La metodología empleada fue una revisión sistemática de la literatura utilizando el método PRISMA, examinando artículos académicos en cinco buscadores: Google Académico, ProQuest, Scielo, Redalyc y Ebsco, en tres idiomas (español, inglés y portugués). Los resultados indican que la mayoría de las investigaciones tienen un enfoque cualitativo y exploratorio, evidenciando la novedad del tema tanto en la industria como en la región estudiada. La utilización de redes sociales para crear nuevos puntos de contacto con clientes y la creación de relaciones personalizadas a largo plazo con ellos, son algunas de las estrategias de marketing digital más destacadas. En conclusión, el marketing digital en el sector bancario ha demostrado ser una herramienta fundamental para generar valor y establecer conexiones duraderas con los clientes, en un nuevo contexto que incorpora las exigencias del mercado digital. Sin embargo, la investigación académica sobre el tema en la región estudiada aún se encuentra en una etapa preliminar.

Abstract Colombia is a tropical country with a rich fern flora. The Quindío Department is in an area which is undergoing significant environmental changes due to anthropogenic activities, such as coffee cultivation. Gleicheniaceae, a family of pioneer ferns, is known for preferentially occupying open, sunny, and disturbed habitats, such as roadsides. Our study aimed to conduct a taxonomic treatment of the Gleicheniaceae species in the Quindío Department in Colombia. Our results revealed the presence of 10 species distributed in four genera, with the Sticherus genus accounting for most of the species (seven in total). Notably, a new record of Sticherus (S. brevitomentosus) was found for Colombia, indicating the potential for the discovery of additional species in future studies. Furthermore, our results demonstrated that 42% of the 24 species reported in the Andean region were present in the Quindío Department, and Calarcá and Salento were the municipalities with the highest diversity. We have provided taxonomic descriptions for the genera and species, as well as an identification key, maps, images, and comments on the taxonomy and distribution of the species.
