Abstract

Inflammatory bowel disease (IBD) is a clinical condition of the gastrointestinal tract that has significant incidence in childhood. Major symptoms include abdominal pain, dyspepsia, delayed gastric emptying, anorexia, diarrhea and weight loss. IBD etiopathogenesis is multifactorial, with a proven involvement of cytokines. In this regard, cytokines like resistin and adiponectin produced by adipose tissue play a crucial role in inflammation. Particularly, resistin seems related to IBD severity and is considered a promising marker of disease occurrence and progression. Unraveling its mechanism of action and downstream effectors is mandatory when designing novel therapies. This preclinical study aims to further elucidate the action of resistin in causing functional gastrointestinal alterations, comparing it with the well-defined effect of adiponectin. To this end, we carried out electrophysiological analysis on murine gastric fundus. We found that resistin, similarly to adiponectin, increases smooth muscle cell (SMC) capacitance, indicative of cell surface remodeling, which is consistent with relaxation. However, contrary to adiponectin, resistin unalters membrane potential and inward Ca2+ entry and scarcely affects outward current, suggesting its inefficacy in markedly modifying electrical phenomena on the SMC membrane. This outcome, supporting the role of resistin in gastrointestinal distention, as observed in IBD, rules out a strikingly direct effect on SMCs.

Abstract

Abstract Background Literature suggests colonic resection and primary anastomosis (RPA) instead of Hartmann’s procedure (HP) for the treatment of left-sided colonic emergencies. We aim to evaluate the surgical options globally used to treat patients with acute left-sided colonic emergencies and the factors that leading to the choice of treatment, comparing HP and RPA. Methods This is a prospective, international, multicenter, observational study registered on ClinicalTrials.gov. A total 1215 patients with left-sided colonic emergencies who required surgery were included from 204 centers during the period of March 1, 2020, to May 31, 2020. with a 1-year follow-up. Results 564 patients (43.1%) were females. The mean age was 65.9 ± 15.6 years. HP was performed in 697 (57.3%) patients and RPA in 384 (31.6%) cases. Complicated acute diverticulitis was the most common cause of left-sided colonic emergencies (40.2%), followed by colorectal malignancy (36.6%). Abstract

Biobased and biodegradable plastics have emerged as promising alternatives to conventional plastics offering the potential to reduce environmental impacts while promoting sustainability. This study focuses on the production of multilayer blown films with enhanced functional properties suitable for food packaging applications. Films were developed through co-extrusion in a three-layer film configuration, with Polybutylene Succinate (PBS) and Polybutylene Succinate Adipate (PBSA) as the external and internal layers, respectively. The functional layer consisted of Polyhydroxybutyrate (PHB) enhanced with nanoclays Cloisite® 30B at varying weight ratios. Films were also processed by manipulating the extruder screw speed of the functional layer to investigate its impact on the functional properties. Rheology, mechanical strength, and barrier performance were characterised to establish correlations between processing conditions and functional layer blends (Cloisite® 30B/PHB) on the properties of the resultant films. Rheological test results indicated that the system with 5% Cloisite® had the best polymer/nanofiller matrix dispersion. Mechanical and permeability tests showed that by varying the process conditions (the alteration of the thickness of the functionalized layer) resulted in an improvement in mechanical and barrier properties. Furthermore, the addition of the nanofiller resulted in a stiffening of the film with a subsequent decrease in permeability to oxygen and water vapour.

4. Development of Biodegradable PBS/PVOH-based Films and Evaluation of Performance for Food Packaging Applications

Abstract

The objective of this work deals with the realization of high O2-barrier blown films based on novel biodegradable blends of Polybutylene succinate (PBS) and Poly-vinyl alcohol (PVOH) for food packaging applications. Blends at different PBS/PVOH weight ratios (100/0, 80/20, 60/40, 0/100 wt%) were produced and their processability and miscibility was preliminary assessed by means of dynamic shear rheological tests. Afterwards, blown films were realized and then characterized in terms of chemical properties and oxygen barrier performance. The results revealed that, although immiscible, polymers in the blend show some degree of interaction. Moreover, as the concentration of PVOH in the films increased, a significant decrease in oxygen permeability was obtained with respect to the neat PBS film, up to 42% for the PBS/PVOH 60/40 sample. The findings of this study highlighted the promising perspectives of these films as sustainable packaging for sensitive foods with high O2-barrier requirements.

Abstract

Despite growing concerns about privacy and an evolution in laws protecting users’ rights, there remains a gap between how industries manage data and how users can express their preferences. This imbalance often favors industries, forcing users to repeatedly define their privacy preferences each time they access a new website. This process contributes to the privacy paradox. We propose a user support tool named the User Privacy Preference Management System (UPPMS) that eliminates the need for users to handle intricate banners or deceptive patterns. We have set up a process to guide even a non-expert user in creating a standardized personal privacy policy, which is automatically applied to every visited website by interacting with cookie banners. The process of generating actions to apply the user’s policy leverages customized Large Language Models. Experiments demonstrate the feasibility of analyzing HTML code to understand and automatically interact with cookie banners, even implementing complex policies. Our proposal aims to address the privacy paradox related to cookie banners by reducing information overload and decision fatigue for users. It also simplifies user navigation by eliminating the need to repeatedly declare preferences in intricate cookie banners on every visited website, while protecting users from deceptive patterns.

6. HIF-1α/MMP-9 Axis Is Required in the Early Phases of Skeletal Myoblast Differentiation under Normoxia Condition In Vitro

Abstract

Hypoxia-inducible factor (HIF)-1α represents an oxygen-sensitive subunit of HIF transcriptional factor, which is usually degraded in normoxia and stabilized in hypoxia to regulate several target gene expressions. Nevertheless, in the skeletal muscle satellite stem cells (SCs), an oxygen level-independent regulation of HIF-1α has been observed. Although HIF-1α has been highlighted as a SC function regulator, its spatio-temporal expression and role during myogenic progression remain controversial. Herein, using biomolecular, biochemical, morphological and electrophysiological analyses, we analyzed HIF-1α expression, localization and role in differentiating murine C2C12 myoblasts and SCs under normoxia. In addition, we evaluated the role of matrix metalloproteinase (MMP)-9 as an HIF-1α effector, considering that MMP-9 is involved in myogenesis and is an HIF-1α target in different cell types. HIF-1α expression increased after 24/48 h of differentiating culture and tended to decline after 72 h/5 days. Committed and proliferating mononuclear myoblasts exhibited nuclear HIF-1α expression. Differently, the more differentiated elongated and parallel-aligned cells, which are likely ready to fuse with each other, show a mainly cytoplasmic localization of the factor. Multinucleated myotubes displayed both nuclear and cytoplasmic HIF-1α expression. The MMP-9 and MyoD (myogenic activation marker) expression synchronized with that of HIF-1α, increasing after 24 h of differentiation. By means of silencing HIF-1α and MMP-9 by short-interfering RNA and MMP-9 pharmacological inhibition, this study unraveled MMP-9’s role as an HIF-1α downstream effector and the fact that the HIF-1α/MMP-9 axis is essential in morpho-functional cell myogenic commitment.

Abstract

Adipokines are peptide hormones produced by the adipose tissue involved in several biological functions. Among adipokines, adiponectin (ADPN) has antidiabetic and anti-inflammatory properties. It can also modulate food intake at central and peripheral levels, acting on hypothalamus and facilitating gastric relaxation. ADPN exerts its action interacting with two distinct membrane receptors and triggering some well-defined signaling cascades. The ceramidase activity of ADPN receptor has been reported in many tissues: it converts ceramide into sphingosine. In turn, sphingosine kinase (SK) phosphorylates it into sphingosine-1 phosphate (S1P), a crucial mediator of many cellular processes including contractility. Using a multidisciplinary approach that combined biochemical, electrophysiological and morphological investigations, we explored for the first time the possible role of S1P metabolism in mediating ADPN effects on the murine gastric fundus muscle layer. By using a specific pharmacological inhibitor of SK2, we showed that ADPN affects smooth muscle cell membrane properties and contractile machinery via SK2 activation in gastric fundus, adding a piece of knowledge to the action mechanisms of this hormone. These findings help to identify ADPN and its receptors as new therapeutic targets or as possible prognostic markers for diseases with altered energy balance and for pathologies with fat mass content alterations.

Abstract

With an enormous range of applications, the Internet of Things (IoT) has magnetized industries and academicians from everywhere. IoT facilitates operations through ubiquitous connectivity by providing Internet access to all the devices with computing capabilities. With the evolution of wireless infrastructure, the focus from simple IoT has been shifted to smart, connected and mobile IoT (M-IoT) devices and platforms, which can enable low-complexity, low-cost and efficient computing through sensors, machines, and even crowdsourcing. All these devices can be grouped under a common term of M-IoT. Even though the positive impact on applications has been tremendous, security, privacy and trust are still the major concerns for such networks and insufficient enforcement of these requirements introduces non-negligible threats to M-IoT devices and platforms. Thus, it is important to understand the range of solutions which are available for providing a secure, privacy-compliant, and trustworthy mechanism for M-IoT. There is no direct survey available, which focuses on security, privacy, trust, secure protocols, physical layer security and handover protections in M-IoT. This paper covers such requisites and presents comparisons of state-the-art solutions for IoT which are applicable to security, privacy, and trust in smart and connected M-IoT networks. Apart from these, various challenges, applications, advantages, technologies, standards, open issues, and roadmap for security, privacy and trust are also discussed in this paper.

Abstract

Adiponectin (ADPN), a hormone produced by adipose tissue, facilitates gastric relaxation and can be a satiety signal in the network connecting peripheral organs and the central nervous system for feeding behavior control. Here, we performed preclinical research by morpho-functional analyses on murine gastric fundus smooth muscle to add insights into the molecular mechanisms underpinning ADPN action. Moreover, we conducted a clinical study to evaluate the potential use of ADPN as a biomarker for eating disorders (ED) based on the demonstrated gastric alterations and hormone level fluctuations that are often associated with ED. The clinical study recruited patients with ED and healthy controls who underwent blood draws for ADPN dosage and psychopathology evaluation tests. The findings of this basic research support the ADPN relaxant action, as indicated by the smooth muscle cell membrane pro-relaxant effects, with mild modifications of contractile apparatus and slight inhibitory effects on gap junctions. All of these actions engaged the ADPN/nitric oxide/guanylate cyclase pathway. The clinical data failed to unravel a correlation between ADPN levels and the considered ED, thus negating the potential use of ADPN as a valid biomarker for ED management for the moment. Nevertheless, this adipokine can modulate physiological eating behavior, and its effects deserve further investigation.

Abstract

The continuous and rapid spread of the COVID-19 pandemic has emphasized the need to seek new therapeutic and prophylactic treatments. Peptide inhibitors are a valid alternative approach for the treatment of emerging viral infections, mainly due to their low toxicity and high efficiency. Recently, two small nucleotide signatures were identified in the genome of some members of the Coronaviridae family and many other human pathogens. In this study, we investigated whether the corresponding amino acid sequences of such nucleotide sequences could have effects on the viral infection of two representative human coronaviruses: HCoV-OC43 and SARS-CoV-2. Our results showed that the synthetic peptides analyzed inhibit the infection of both coronaviruses in a dose-dependent manner by binding the RBD of the Spike protein, as suggested by molecular docking and validated by biochemical studies. The peptides tested do not provide toxicity on cultured cells or human erythrocytes and are resistant to human serum proteases, indicating that they may be very promising antiviral peptides.

Abstract

Intro Thrombotic complications are quite frequent during SARS-CoV2 infection, especially during severe disease and in hospitalized patients. The incidence in mild disease is not clear and probably few cases have been identified. Case report We reported a case of a 60-year-old man with no previous history admitted in our unit for splenic infarct and a floating clot into the descending aorta without signs of severe disease. Discussion Several mechanisms to clarify prothrombotic state have been described and the exact prevalence of thromboembolic phenomenon is probably underestimated. There is no consensus about the treatment and the indication to prevent these complications in non-hospitalized patients. Conclusion Thrombotic events should be suspected also in fit patients with previous or actual symptoms suggestive for Covid 19 infection. Further analysis should be performed to stratify the risk in non hospitalized patients and the indication for prophylactic treatment.

Abstract

Industrial assistive systems result from a multidisciplinary effort that integrates IoT (and Industrial IoT), Cognetics, and Artificial Intelligence. This paper evaluates the Prediction by Partial Matching algorithm as a component of an assembly assistance system that supports factory workers, by providing choices for the next manufacturing step. The evaluation of the proposed method was performed on datasets collected within an experiment involving trainees and experienced workers. The goal is to find out which method best suits the datasets in order to be integrated afterwards into our context-aware assistance system. The obtained results show that the Prediction by Partial Matching method presents a significant improvement with respect to the existing Markov predictors.

Abstract

Mobile cellular communications are experiencing an exponential growth in traffic load on Long Term Evolution (LTE) eNode B (eNB) components. Such load can be significantly contained by directly sharing content among nearby users through device-to-device (D2D) communications, so that repeated downloads of the same data can be avoided as much as possible. Accordingly, for the purpose of improving the efficiency of content sharing and decreasing the load on the eNB, it is important to maximize the number of simultaneous D2D transmissions. Specially, maximizing the number of D2D links can not only improve spectrum and energy efficiency but can also reduce transmission delay. However, enabling maximum D2D links in a cellular network poses two major challenges. First, the interference between the D2D and cellular communications could critically affect their performance. Second, the minimum quality of service (QoS) requirement of cellular and D2D communication must be guaranteed. Therefore, a selection of active links is critical to gain the maximum number of D2D links. This can be formulated as a classical integer linear programming problem (link scheduling) that is known to be NP-hard. This paper proposes to obtain a set of network features via deep learning for solving this challenging problem. The idea is to optimize the D2D link schedule problem with a deep neural network (DNN). This makes a significant time reduction for delay-sensitive operations, since the computational overhead is mainly spent in the training process of the model. The simulation performed on a randomly generated link schedule problem showed that our algorithm is capable of finding satisfactory D2D link scheduling solutions by reducing computation time up to 90% without significantly affecting their accuracy.

Abstract

Shallow, low-temperature geothermal resources can significantly reduce the environmental impact of heating and cooling. Based on a replicable standard workflow for three-dimensional (3D) geothermal modeling, an approach to the assessment of geothermal energy potential is proposed and applied to the young sedimentary basin of Pisa (north Tuscany, Italy), starting from the development of a geothermal geodatabase, with collated geological, stratigraphic, hydrogeological, geophysical and thermal data. The contents of the spatial database are integrated and processed using software for geological and geothermal modeling. The models are calibrated using borehole data. Model outputs are visualized as three-dimensional reconstructions of the subsoil units, their volumes and depths, the hydrogeological framework, and the distribution of subsoil temperatures and geothermal properties. The resulting deep knowledge of subsoil geology would facilitate the deployment of geothermal heat pump technology, site selection for well doublets (for open-loop systems), or vertical heat exchangers (for closed-loop systems). The reconstructed geological–hydrogeological models and the geothermal numerical simulations performed help to define the limits of sustainable utilization of an area’s geothermal potential.

Abstract

A utilização do manejo convencional em solos sob cerrado tem acarretado modificações nas suas propriedades, bem como no comportamento e qualidade da sua matéria orgânica. O objetivo desta pesquisa foi investigar o impacto do manejo convencional nas propriedades físicas e no conteúdo e qualidade de substâncias húmicas de um Latossolo Vermelho-Amarelo, originalmente sob cerrado. O estudo foi realizado em uma propriedade agrícola no município de São Desidério-BA. Foram avaliadas quatro áreas sob diferentes períodos de uso e uma sob cerrado nativo. Os tempos de uso foram de um, dois, três e seis anos, sendo que a área estudada após seis anos passou por um período de três anos de pousio. O sistema de preparo do solo utilizado foi o tradicional da região, que consiste no uso de duas passadas de grade pesada aradora e duas de grade niveladora. Também foram realizadas, na ocasião de cada plantio, uma adubação com N-P-K + micronutrientes e calagens com base em resultados de análise de solo. As áreas foram cultivadas com milho e soja em sistema de rotação. Verificou-se que o manejo adotado não foi sustentável, pois já nos primeiros três anos de uso agrícola, ocorreu grande deterioração das propriedades físicas do solo, como densidade, porosidade e conteúdo de água disponível. O processo de humificação e a qualidade da matéria orgânica foram influenciados pela aplicação de calcário e pela rotação de culturas (milho/soja) e, ainda, que o sistema de manejo utilizado mostrou-se mais benéfico para o processo de estabilização das substâncias húmicas do que para as propriedades físicas dos solos estudados.The utilization of intensive conventional management system on soils under cerrado biome has caused modification in their properties, as well as, in the behavior and quality of the organic matter. The objective of this research was to evaluate the impact of conventional soil management practices upon physical properties, contents and qualities of humic substances of a Red-Yellow Oxisol, formerly, under cerrado (savana-like) natural vegetation. The study was accomplished on an private agricultural property in the municipal district of São Desidério-BA. They were appraised four areas under different length of times of agricultural use and one area under native cerrado (savana-like). The length of time under agriculture use was one, two, three and six years, and after six years the studied area was left uncultivated for a period of three years. The soil has been prepared under traditional systems which consists of plowing the land twice with heavy disks followed by twice grading. It was also applied, during the seedling time, N-P-K fertilizer plus micronutrients and lime according to soil analysis. The areas have been cultivated with corn and soybeans in a rotation system. It was observed that the adopted management system was not sustainable, because, at the first three years under cultivation occurred a great deterioration of physical properties such as soil density, porosity and available water contents. The humification process and the quality of the organic matter were influenced by both utilization of lime and by the rotation system (corn/soybeans). The used system was more benefic for the process of stabilization of the humic substances than for the physical properties of the studied soils.

Abstract

The identification of high-risk patients in the early stages of infected pancreatic necrosis (IPN) is critical, because it could help the clinicians to adopt more effective management strategies. We conducted a post hoc analysis of the MANCTRA-1 international study to assess the association between clinical risk factors and mortality among adult patients with IPN. Univariable and multivariable logistic regression models were used to identify prognostic factors of mortality. We identified 247 consecutive patients with IPN hospitalised between January 2019 and December 2020. History of uncontrolled arterial hypertension (p = 0.032; 95% CI 1.135–15.882; aOR 4.245), qSOFA (p = 0.005; 95% CI 1.359–5.879; aOR 2.828), renal failure (p = 0.022; 95% CI 1.138–5.442; aOR 2.489), and haemodynamic failure (p = 0.018; 95% CI 1.184–5.978; aOR 2.661), were identified as independent predictors of mortality in IPN patients. Cholangitis (p = 0.003; 95% CI 1.598–9.930; aOR 3.983), abdominal compartment syndrome (p = 0.032; 95% CI 1.090–6.967; aOR 2.735), and gastrointestinal/intra-abdominal bleeding (p = 0.009; 95% CI 1.286–5.712; aOR 2.710) were independently associated with the risk of mortality. Upfront open surgical necrosectomy was strongly associated with the risk of mortality (p p = 0.018; 95% CI 0.138–0.834; aOR 0.339) and enteral nutrition (p = 0.003; 95% CI 0.143–0.716; aOR 0.320) were found as protective factors. Organ failure, acute cholangitis, and upfront open surgical necrosectomy were the most significant predictors of mortality. Our study confirmed that, even in a subgroup of particularly ill patients such as those with IPN, upfront open surgery should be avoided as much as possible.
