Abstract Traditional fully-deterministic algorithms, which rely on physical equations and mathematical models, are the backbone of many scientific disciplines for decades. These algorithms are based on well-established principles and laws of physics, enabling a systematic and predictable approach to problem-solving. On the other hand, AI-based strategies emerge as a powerful tool for handling vast amounts of data and extracting patterns and relationships that might be challenging to identify through traditional algorithms. Here, we bridge these two realms by using AI to find an optimal mapping of meteorological features predicted two days ahead by the state-of-the-art numerical weather prediction model by the European Centre for Medium-range Weather Forecasts (ECMWF) into lightning flash occurrence. The prediction capability of the resulting AI-enhanced algorithm turns out to be significantly higher than that of the fully-deterministic algorithm employed in the ECMWF model. A remarkable Recall peak of about 95% within the 0-24 h forecast interval is obtained. This performance surpasses the 85% achieved by the ECMWF model at the same Precision of the AI algorithm.

Abstract Motivated by recent reports of a quantum-disordered ground state in the triangular lattice compound NaRuO2, we derive a j eff = 1/2 magnetic model for this system by means of first-principles calculations. The pseudospin Hamiltonian is dominated by bond-dependent off-diagonal Γ interactions, complemented by a ferromagnetic Heisenberg exchange and a notably antiferromagnetic Kitaev term. In addition to bilinear interactions, we find a sizable four-spin ring exchange contribution with a strongly anisotropic character, which has been so far overlooked when modeling Kitaev materials. The analysis of the magnetic model, based on the minimization of the classical energy and exact diagonalization of the quantum Hamiltonian, points toward the existence of a rather robust easy-plane ferromagnetic order, which cannot be easily destabilized by physically relevant perturbations.

The management of unowned cats is an emerging problem, with public institutions and citizens’ concerns regarding their care and arrangement. Little is known regarding the outcome of traumatic orthopedic injuries in these patients. Indeed, complete functional recovery (CFR) should be the goal of treatment for return to their original location or adoption. The aim was to identify clinical factors influencing CFR in traumatized unowned cats with orthopedic lesions. This category of cats referred by the veterinary public service over three years was enrolled. Various clinical variables were retrospectively collected from the medical records and evaluated by nominal logistic analysis. Forty-eight unowned cats were enrolled, with a median estimated age of 24 (1–180) months and a body weight of 3 (0.7–5) kg. Thirty-four (71%) patients reached CFR. Estimated age, body weight, time from trauma to therapeutic intervention, spine involvement, presence of comorbidities, hospitalization time, and the radiographic score results were significantly associated with CFR. A longer time to therapeutic intervention seemed to be associated with a better outcome. Probably, cats severely traumatized did not live long enough to be evaluated and treated. Lighter cats experienced more severe consequences following blunt trauma. Younger and lighter cats bore a higher risk of panleukopenia-related death.

The skill of ERA5 has been assessed in relation to the prediction of the wind energy associated with 28 SYNOP stations located in Italy for a time span of 20 years (2001–2020). For comparison, a WRF-based high-resolution downscaling (3 km horizontally) was also produced for the same period. We found that simple predictions based on materialized past wind measures outperform the wind energy predictions from ERA5. This result can be ascribed to the particularly complex characteristics of the Italian territory. Motivated by this expected behavior, we have implemented a Quantile Random Forest (QRF) calibration which greatly alleviates the problems encountered in the ERA5 reanalysis dataset. This technique provides a calibrated ensemble prediction system for the wind speed at the station. Surprisingly, the calibrated ERA5 outperforms wind energy estimations from the high-resolution 3-km downscaling. Once properly calibrated, the high-resolution downscaling provides predictions very similar to the calibrated ERA5. Limiting our conclusions to the estimation of wind energy over a long time span (here 20 years), having at disposal a high-resolution wind-field dataset does not necessarily mean greater accuracy. A careful calibration of the original coarser wind-field dataset produces better results than the raw high-resolution dataset.

Abstract Recent experimental findings have reported the presence of unconventional charge orders in the enlarged (2 × 2) unit-cell of kagome metals AV3Sb5 (A = K, Rb, Cs) and hinted towards specific topological signatures. Motivated by these discoveries, we investigate the types of topological phases that can be realized in such kagome superlattices. In this context, we employ a recently introduced statistical method capable of constructing topological models for any generic lattice. By analyzing large data sets generated from symmetry-guided distributions of randomized tight-binding parameters, and labeled with the corresponding topological index, we extract physically meaningful information. We illustrate the possible real-space manifestations of charge and bond modulations and associated flux patterns for different topological classes, and discuss their relation to present theoretical predictions and experimental signatures for the AV3Sb5 family. Simultaneously, we predict higher-order topological phases that may be realized by appropriately manipulating the currently known systems.

Pinch point singularities, associated with flat band magnetic excitations, are tell-tale signatures of Coulomb spin liquids. While their properties in the presence of quantum fluctuations have been widely studied, the fate of the complementary nonanalytic features—shaped as half moons and stars—arising from adjacent shallow dispersive bands has remained unexplored. Here, we address this question for the spin S=1/2 Heisenberg antiferromagnet on the kagome lattice with second and third neighbor couplings, which allows one to tune the classical ground state characterized by flat bands to one that is governed by shallow dispersive bands for intermediate coupling strengths. Employing the complementary strengths of variational Monte Carlo, pseudofermion functional renormalization group, and density-matrix renormalization group, we establish the quantum phase diagram of the model. The U(1) Dirac spin liquid ground state of the nearest-neighbor antiferromagnet remains remarkably robust till intermediate coupling strengths when it transitions into a pinwheel valence bond crystal displaying signatures of half moons in its structure factor. Our Letter thus identifies a microscopic setting that realizes one of the proximate orders of the Dirac spin liquid identified in a recent work [Song, Wang, Vishwanath, and He, Nat. Commun. 10, 4254 (2019)2041-172310.1038/s41467-019-11727-3]. For larger couplings, we obtain a collinear magnetically ordered ground state characterized by starlike patterns.

This study compared the postoperative analgesic efficacy of intraperitoneal and incisional lidocaine versus ropivacaine in dogs undergoing major abdominal surgeries. Dogs randomly received intraperitoneal lidocaine irrigation (4 mg kg-1, diluted to 5 ml kg-1, L group), ropivacaine (4 mg kg-1, diluted to 5 ml kg-1, R group) or 0.9% saline (5 ml kg-1, C group). Prior to skin closure, dogs received incisional lidocaine 2 mg kg-1 (group L), incisional ropivacaine 2 mg kg-1 (group R) or incisional saline 0.2 ml kg-1 (group C). Pain was assessed at different time points up to 24 hours after extubation, using the Short Form-Glasgow Composite Measure Pain Scale and VAS Scale. In group C, postoperative pain scores were significantly higher than in groups L and R from T0.5 to T6 (p < 0.05). In R group, postoperative pain scores were significantly lower than in groups L and C from T12 to T24 (p < 0.05). Rescue analgesia was administered to 5/11 dogs in L group, 1/10 dogs in R group and 8/10 dogs in C group. Groups L and R experienced a significantly lower postoperative pain during the first 6 hours after extubation, compared with group C. Ropivacaine provided lower postoperative pain scores than lidocaine and saline up to 24 hours after extubation. According to the obtained results, ropivacaine seemed to provide better and longer lasting postoperative analgesia compared with lidocaine. Therefore, intraperitoneal and incisional administration of ropivacaine in dogs undergoing major abdominal surgeries is recommended.

Abstract We investigate the magnetism of a previously unexplored distorted spin-1/2 kagome model consisting of three symmetry-inequivalent nearest-neighbor antiferromagnetic Heisenberg couplings J ⬡, J, and $$J^{\prime}$$ J ′ , and uncover a rich ground state phase diagram even at the classical level. Using analytical arguments and numerical techniques we identify a collinear $$\overrightarrow{Q}=0$$ Q → = 0 magnetic phase, two unusual non-collinear coplanar $$\overrightarrow{Q}=(1/3,1/3)$$ Q → = ( 1 / 3 , 1 / 3 ) phases and a classical spin liquid phase with a degenerate manifold of non-coplanar ground states, resembling the jammed spin liquid phase found in the context of a bond-disordered kagome antiferromagnet. We further show with density functional theory calculations that the recently synthesized Y-kapellasite Y3Cu9(OH)19Cl8 is a realization of this model and predict its ground state to lie in the region of $$\overrightarrow{Q}=(1/3,1/3)$$ Q → = ( 1 / 3 , 1 / 3 ) order, which remains stable even after the inclusion of quantum fluctuation effects within variational Monte Carlo and pseudofermion functional renormalization group. The presented model opens a new direction in the study of kagome antiferromagnets.

Motivated by recent experiments on Cs$_2$Cu$_3$SnF$_{12}$ and YCu$_{3}$(OH)$_{6}$Cl$_{3}$, we consider the ${S=1/2}$ Heisenberg model on the kagome lattice with nearest-neighbor super-exchange $J$ and (out-of-plane) Dzyaloshinskii-Moriya interaction $J_D$, which favors (in-plane) ${{\bf Q}=(0,0)}$ magnetic order. By using both variational Monte Carlo and tensor-network approaches, we show that the ground state develops a finite magnetization for $J_D/J \gtrsim 0.03 \mathrm{-} 0.04$; instead, for smaller values of the Dzyaloshinskii-Moriya interaction, the ground state has no magnetic order and, according to the fermionic wave function, develops a gap in the spinon spectrum, which vanishes for $J_D \to 0$. The small value of $J_D/J$ for the onset of magnetic order is particularly relevant for the interpretation of low-temperature behaviors of kagome antiferromagnets, including ZnCu$_{3}$(OH)$_{6}$Cl$_{2}$. For this reason, we assess the spin dynamical structure factor and the corresponding low-energy spectrum, by using the variational Monte Carlo technique. The existence of a continuum of excitations above the magnon modes is observed within the magnetically ordered phase, with a broad peak above the lowest-energy magnons, similarly to what has been detected by inelastic neutron scattering on Cs$_{2}$Cu$_{3}$SnF$_{12}$.

Here, the authors present the SAMMY-seq, a method for genome-wide characterization of heterochromatin dynamics and detect early stage alterations of heterochromatin structure in progeria primary fibroblasts, accompained by Polycomb dysfunctions.

Neuronal differentiation requires rearrangement of the transcriptional and chromatin landscapes of neural cells. Here, the authors study in-vitro neuronal differentiation of murine embryonic stem cells (ESCs) to show that this process is modulated by DOT1L activity, which regulates H3K79me2 accumulation, and preserves accessibility of SOX2-bound enhancers.

Abstract Genetic and epigenetic factors contribute to the development of the spinal cord. Failure in correct exertion of the developmental programs, including neurulation, neural tube closure and neurogenesis of the diverse spinal cord neuronal subtypes results in defects of variable severity. We here report on the histone methyltransferase Disruptor of Telomeric 1 Like (DOT1L), which mediates histone H3 lysine 79 (H3K79) methylation. Conditional inactivation of DOT1L using Wnt1-cre as driver (Dot1l-cKO) showed that DOT1L expression is essential for spinal cord neurogenesis and localization of diverse neuronal subtypes, similar to its function in the development of the cerebral cortex and cerebellum. Transcriptome analysis revealed that DOT1L deficiency favored differentiation over progenitor proliferation. Dot1l-cKO mainly decreased the numbers of dI1 interneurons expressing Lhx2. In contrast, Lhx9 expressing dI1 interneurons did not change in numbers but localized differently upon Dot1l-cKO. Similarly, loss of DOT1L affected localization but not generation of dI2, dI3, dI5, V0 and V1 interneurons. The resulting derailed interneuron patterns might be responsible for increased cell death, occurrence of which was restricted to the late developmental stage E18.5. Together our data indicate that DOT1L is essential for subtype-specific neurogenesis, migration and localization of dorsal and ventral interneurons in the developing spinal cord, in part by regulating transcriptional activation of Lhx2.

In Drosophila, dosage compensation involves a twofold transcriptional upregulation of the single male chromosome X. Here the authors show that global conformational differences are specifically present in the male X chromosome and detectable using Hi-C data, indicating that dosage compensation affects global chromosome structure.

Background: Pilonidal disease (PD) represents one of the most common proctological diseases in young adults. Although several approaches to treating PD have been described, there is still a lack of agreement on which is the best. The aim of this study was to evaluate the long-term efficacy of endoscopic pilonidal sinus treatment (EPSiT) at a tertiary care academic center.Methods: Between June 2017 and January 2021, a total of 32 patients [12 women (37.5%) and 20 men (62.5%)] with a mean age of 29.22 ± 12.98 years were treated with EPSiT. Pre- and post-operative symptoms were assessed with a score of 0–5. Success was defined as the absence of any subjective symptoms, as well as by complete post-operative wound healing.Results: Most of the patients had a midline external opening (17/32; 53.1%), with a mean number of external openings of 2.41 (1–4) ± 1.04. The median post-operative pain score was 0, and the mean follow-up period was 22 (4–42) ± 11.49 months. The time to wound healing was reduced in patients with one opening (28.14 ± 4.06 days) compared to patients with two or more openings (33.64 ± 7.3 days) (p = 0.067). The mean operative time was longer in patients who subsequently had a recurrence (41.75 ± 6.24 vs. 34.18 ± 6.24 min; p = 0.031). The overall success rate was 87.5% (28/32), and the mean time to recurrence was 3.25 (2–5) ± 1.26 months.Conclusions: EPSiT represents a viable option for the treatment of PD. More evidence and a longer follow-up period are needed to validate the results.

Sentinel lymph node (SLN) biopsy is a well-established staging tool in canine oncology. This study aims to explore the feasibility of SLN biopsy in dogs with scars from prior excised solid malignancies that were referred for further tumor staging and/or adjuvant treatment options. Mapping was either performed using radiopharmaceutical, methylene blue, and/or near-infrared fluorescent (NIRF) imaging. Thirty-three dogs with 34 scars from prior excision of the mast cell tumor (MCT) (n = 29), soft tissue sarcoma (n = 2), oral melanoma (n = 1), subungual melanoma (n = 1), and mammary adenocarcinoma (n = 1) were retrospectively enrolled. Primary treatment consisted of curative intent/wide tumor excisions in 50.0% of dogs and marginal excision in the remaining 50.0%. The median time between tumor excision and SLN biopsy was 50 days (range 17–110 days). The procedure was successful in 31/34 scars, translating to a detection rate of 91.2%. The SLN did not correspond to the regional lymph node in 19/31 scars (61.3%). SLN metastases were histologically identified in 13/31 (41.9%) dogs, all of them affected by MCT. Based on our results, SLN biopsy using lymphoscintigraphy/methylene blue and/or NIRF is feasible in dogs presenting with scars from the prior surgical excision of solid tumors, and should be suggested for accurate nodal staging.

Despite an increasing research conducted on ocean plastic pollution over the last decade, there are still large knowledge gaps in our current understanding of how floating plastic debris accumulating in subtropical oceanic gyres may harm the surface-associated pelagic community known as neuston. Removing floating plastic debris from the surface ocean can minimize potentially adverse effects of plastic pollution on the neuston, as well as prevent the formation of large quantities of secondary micro- and nanoplastics. However, due to the scarcity of observational data from remote and difficult to access offshore waters, neuston dynamics in subtropical oceanic gyres and thus the potential impacts of plastic pollution as well as of cleanup activities on the neuston remain uncertain. Here, we provide rare observational data of the relative distribution of floating plastic debris (0.05–5 cm in size) and members of the neuston in the eastern North Pacific Ocean. Our results reveal that the dominant neustonic species co-occurring with high concentrations of floating plastic debris in the North Pacific Garbage Patch (NPGP) such as Porpita porpita, Halobates spp., pteropods, isopods, heteropods, and crabs depict either a low atmospheric drag due to physical attributes or a potential plastic-associated fitness benefit such as increased surface area for oviposition and structure for habitat. We further observe relatively higher plastic to organism ratios inside the NPGP for most target species compared to waters outside the NPGP. The findings presented here provide a first observational baseline to develop ecological models that can help evaluate the long-term risks of plastic pollution and of offshore cleanup activities for neuston in the eastern North Pacific Ocean. We further suggest that offshore mitigation strategies aiming at removing floating plastic debris from the ocean surface need to evaluate both, the direct impact of neuston bycatch during plastic removal on neuston population dynamics, as well as the potential benefits of reducing the negative effects of plastic pollution on the neuston.

Background Pediatric high-grade gliomas (pHGGs) are among the most common and incurable malignant neoplasms of childhood. Despite aggressive, multimodal treatment, the outcome of children with high-grade gliomas has not significantly improved over the past decades, prompting the development of innovative approaches.Methods To develop an effective treatment, we aimed at improving the suboptimal antitumor efficacy of oncolytic adenoviruses (OAs) by testing the combination with a gene-therapy approach using a bispecific T-cell engager (BiTE) directed towards the erythropoietin-producing human hepatocellular carcinoma A2 receptor (EphA2), conveyed by a replication-incompetent adenoviral vector (EphA2 adenovirus (EAd)). The combinatorial approach was tested in vitro, in vivo and thoroughly characterized at a molecular level.Results After confirming the relevance of EphA2 as target in pHGGs, documenting a significant correlation with worse clinical outcome of the patients, we showed that the proposed strategy provides significant EphA2-BiTE amplification and enhanced tumor cell apoptosis, on coculture with T cells. Moreover, T-cell activation through an agonistic anti-CD28 antibody further increased the activation/proliferation profiles and functional response against infected tumor cells, inducing eradication of highly resistant, primary pHGG cells. The gene-expression analysis of tumor cells and T cells, after coculture, revealed the importance of both EphA2-BiTE and costimulation in the proposed system. These in vitro observations translated into significant tumor control in vivo, in both subcutaneous and a more challenging orthotopic model.Conclusions The combination of OA and EphA2-BiTE gene therapy strongly enhances the antitumor activity of OA, inducing the eradication of highly resistant tumor cells, thus supporting the clinical translation of the approach.

Millions of tons of buoyant plastic materials enter oceans annually, the majority originating from terrestrial sources and transported to oceans where oceanographic processes disperse or accumulate them. Some of these materials beach while others accumulate in convergent zones in coastal seas and the open ocean. Although accumulations associated with subtropical gyres, for example, the “Great Pacific Garbage Patch” (GPGP) are well-known, coastal accumulation zones have received less attention. Here we report quantities and characteristics of plastics accumulated in fronts encountered within the Ashmore Reef marine park (Pulau Pasir), northern Australia. These areas, as well as surrounding waters, were sampled using Manta trawls, drone, and snorkel surveys conducted in October 2018. With mean plastic concentrations of 523,146 pieces km−2 for plastics > 500 micron these hotpots contained plastic concentrations an order of magnitude higher than surrounding waters (16,561 pieces km−2) and comparable to the largest known accumulation zone: the GPGP. Furthermore, the mean mass within hotspots was 5,161 g km–2 vs. 9 g km–2 in surrounding waters. Therefore, we classify the features described in this study as types of “Coastal Garbage Patches” (CGPs). Importantly, the coastal fronts accumulating plastics in CGPs are key habitats for many marine species. Biomass outnumbered plastics by weight, with a ratio of 0.521 in CGPs and 0.016 in surrounding waters vs. 287.7 recorded in the GPGP. Polymer types found between the CGPs and GPGP were similar, but plastic films vastly dominated in the CGPs, whilst they were amongst the rarest types found in the GPGP. This study demonstrates the existence of CGPs coinciding with high priority conservation zones in coastal waters and highlights a need for further research into these environments.

Enhancers are non-coding regulatory elements that are distant from their target gene. Their characterization still remains elusive especially due to challenges in achieving a comprehensive pairing of enhancers and target genes. A number of computational biology solutions have been proposed to address this problem leveraging the increasing availability of functional genomics data and the improved mechanistic understanding of enhancer action.In this review we focus on computational methods for genome-wide definition of enhancer-target gene pairs. We outline the different classes of methods, as well as their main advantages and limitations. The types of information integrated by each method, along with details on their applicability are presented and discussed. We especially highlight the technical challenges that are still unresolved and hamper the effective achievement of a satisfactory and comprehensive solution.We expect this field will keep evolving in the coming years due to the ever-growing availability of data and increasing insights into enhancers crucial role in regulating genome functionality.

This article analyzes the development of the papal magisterium on human mobility during the period that went from the election of Leo XIII (1878) to the death of Paul VI (1978). What emerges from the research is a complex situation, characterized by a consistent thought in which each Pope chose to preserve specific elements while making his own specific, individual contribution to the problem. The article begins with Leo XIII and his concern for the faith of emigrants, it then reviews Pope Pius’ papacy where the tensions due to the Fascist regime characterized the period up to Pius XII. Paolo VI opened the papal magisterium to the new perspectives proposed by the Second Vatican Council as well as the great changes of human mobility that occurred after the second world war. This study, therefore, analyses an important Catholic line of thought in order to understand both the present day situation and the modernity and complexity of Pope Francis’ papacy.

Unrepaired DNA damage during embryonic development can be potentially inherited by a large population of cells. However, the quality control mechanisms that minimize the contribution of damaged cells to developing embryos remain poorly understood. Here, we uncovered an ATR- and CHK1-mediated transcriptional response to replication stress (RS) in mouse embryonic stem cells (ESCs) that induces genes expressed in totipotent two-cell (2C) stage embryos and 2C-like cells. This response is mediated by Dux, a multicopy retrogene defining the cleavage-specific transcriptional program in placental mammals. In response to RS, DUX triggers the transcription of 2C-like markers such as murine endogenous retrovirus-like elements (MERVL) and Zscan4. This response can also be elicited by ETAA1-mediated ATR activation in the absence of RS. ATR-mediated activation of DUX requires GRSF1-dependent post-transcriptional regulation of Dux mRNA. Strikingly, activation of ATR expands ESCs fate potential by extending their contribution to both embryonic and extra-embryonic tissues. These findings define a novel ATR dependent pathway involved in maintaining genome stability in developing embryos by controlling ESCs fate in response to RS.

The aim of this paper is to promote the use of mine clay washing residues for the preparation of alkali activated materials (AAMs). In particular, the influence of the calcination temperature of the clayey by-product on the geopolymerization process was investigated in terms of chemical stability and durability in water. The halloysitic clay, a mining by-product, has been used after calcination and mixed with an alkaline solution to form alkali activated binders. Attention was focused on the influence of the clay’s calcination treatment (450–500–600 °C) on the geopolymers’ microstructure of samples, remaining in the lower limit indicated by the literature for kaolinite or illite calcination. The mixtures of clay and alkali activators (NaOH 8M and Na-silicate) were cured at room temperature for 28 days. The influence of solid to liquid ratio in the mix formulation was also tested in terms of chemical stability measuring the pH and the ionic conductivity of the eluate after 24-h immersion time in water. The results reported values of ionic conductivity higher for samples made with untreated clay or with low temperature of calcination (≥756 mS/m) compared with values of samples made with calcined clay (292 mS/m). This result suggests that without a proper calcination of the as-received clay it was not possible to obtain 25 °C-consolidated AAMs with good chemical stability and dense microstructure. The measures of integrity test, pH, and ionic conductivity in water confirmed that the best sample is made with calcined clay at 600 °C, being similar (53% higher ionic conductivity of the eluate) or equal (integrity test and pH) to values recorded for the metakaolin-based geopolymer considered the reference material. These results were reflected in term of reticulation and morphology of samples through the analysis with scanning electron microscope (SEM) and X-ray diffraction (XRD), which show a dense and homogeneous microstructure predominantly amorphous with minor amounts of quartz, halloysite, and illite crystalline phases. Special attention was dedicated to this by-product to promote its use, given that kaolinite (and metakaolin), as primary mineral product, has a strong impact on the environment. The results obtained led us to consider this halloysite clay very interesting as an aluminosilicate precursor, and extensively deepening its properties and reactivity for the alkaline activation. In fact, the heart of this work is to study the possibility of reusing this by-product of an industrial process to obtain more sustainable high-performance binders.

Acute myeloid leukemia (AML) is characterized by recurrent mutations that affect normal hematopoiesis. The analysis of human AMLs has mostly been performed using end-point materials, such as cell lines and patient derived AMLs that also carry additional contributing mutations. The molecular effects of a single oncogenic hit, such as expression of the AML associated oncoprotein AML1-ETO on hematopoietic development and transformation into a (pre-) leukemic state still needs further investigation. Here we describe the development and characterization of an induced pluripotent stem cell (iPSC) system that allows in vitro differentiation towards different mature myeloid cell types such as monocytes and granulocytes. During in vitro differentiation we expressed the AML1-ETO fusion protein and examined the effects of the oncoprotein on differentiation and the underlying alterations in the gene program at 8 different time points. Our analysis revealed that AML1-ETO as a single oncogenic hit in a non-mutated background blocks granulocytic differentiation, deregulates the gene program via altering the acetylome of the differentiating granulocytic cells, and induces t(8;21) AML associated leukemic characteristics. Together, these results reveal that inducible oncogene expression during in vitro differentiation of iPS cells provides a valuable platform for analysis of aberrant regulation in disease.

In this study, the role of two reactive fillers, specifically a sand from a clay washing process as an alternative to waste glass powder and a commercial metakaolin (MK), into the geopolymerization process of waste clay-based materials was assessed. Three kinds of clayey wastes from mining operations—halloysitic, kaolinitic and smectitic clays—were tested as potential precursor of geopolymeric materials in view of a potential valorisation of these by-products. A mix-design based on the addition of low percentages (20%) of these fillers or MK to improve the mechanical and chemico-physical properties of geopolymeric formulations was evaluated. All the clays were thermally treated at a temperature of 650 °C, while the geopolymeric pastes were cured at room temperature. In particular, the chemical stability in water (pH and ionic conductivity of leachate water, weight loss), the variations in the microstructure (XRD, SEM), and in the mechanical performance (compressive strength) were analysed. The most reactive additive was MK, followed by sand and waste glass at very similar levels—1:1 or 2:1—depending upon the type of the clay but not strictly related to the clay type. The increase of geopolymeric gel densification due to the presence of MK and sand was replaced by a crack deflection mechanism in the case of the WG grains. The worst performance (chemical stability and mechanical properties) was found for the halloysitic clay, while kaolinitic and smectitic clays developed strengths slightly below 30 MPa.

The aim of this study was to evaluate the efficacy of oral transmucosal (OTM) cannabidiol (CBD), in addition to a multimodal pharmacological treatment for chronic osteoarthritis-related pain in dogs. Twenty-one dogs were randomly divided into two groups: in group CBD (n = 9), OTM CBD (2 mg kg−1 every 12 h) was included in the therapeutic protocol (anti-inflammatory drug, gabapentin, amitriptyline), while in group C (n = 12), CBD was not administered. Dogs were evaluated by owners based on the Canine Brief Pain Inventory scoring system before treatment initiation (T0), and one (T1), two (T2), four (T3) and twelve (T4) weeks thereafter. Pain Severity Score was significantly lower in CBD than in C group at T1 (p = 0.0002), T2 (p = 0.0043) and T3 (p = 0.016). Pain Interference Score was significantly lower in CBD than in C group at T1 (p = 0.0002), T2 (p = 0.0007) and T4 (p = 0.004). Quality of Life Index was significantly higher in CBD group at T1 (p = 0.003). The addition of OTM CBD showed promising results. Further pharmacokinetics and long-term studies in larger populations are needed to encourage its inclusion into a multimodal pharmacological approach for canine osteoarthritis-related pain.

In late summer and autumn Mediterranean coastal regions are quite regularly affected by small-scale, flood-producing convective systems. The complexity of mesoscale triggering mechanisms, related to low-level temperature gradients, moisture convergence, and topographic effects contributes to limit the predictability of such phenomena. In the present work, a severe convection episode associated to a flash flood occurred in Cannes (southern France) in October 2015, is investigated by means of numerical simulations with a state-of-the-art nonhydrostatic mesoscale model. In the modelling configuration operational at the University of Genoa precipitation maxima were underestimated and misplaced. The impact of model resolution as well as initial and boundary conditions on the quantitative precipitation forecasts is analyzed and discussed. In particular, the effect of ingesting a high-resolution satellite-derived sea surface temperature field is proven to be beneficial in terms of precipitation intensity and localization, especially when also associated with the most accurate lateral boundary conditions.

Understanding the nature of the excitation spectrum in quantum spin liquids is of fundamental importance, in particular for the experimental detection of candidate materials. However, current theoretical and numerical techniques have limited capabilities, especially in obtaining the dynamical structure factor, which gives a crucial characterization of the ultimate nature of the quantum state and may be directly assessed by inelastic neutron scattering. In this work, we investigate the low-energy properties of the S=1/2 Heisenberg model on the triangular lattice, including both nearest-neighbor J_{1} and next-nearest-neighbor J_{2} superexchanges, by a dynamical variational Monte Carlo approach that allows accurate results on spin models. For J_{2}=0, our calculations are compatible with the existence of a well-defined magnon in the whole Brillouin zone, with gapless excitations at K points (i.e., at the corners of the Brillouin zone). The strong renormalization of the magnon branch (also including rotonlike minima around the M points, i.e., midpoints of the border zone) is described by our Gutzwiller-projected state, where Abrikosov fermions are subject to a nontrivial magnetic π flux threading half of the triangular plaquettes. When increasing the frustrating ratio J_{2}/J_{1}, we detect a progressive softening of the magnon branch at M, which eventually becomes gapless within the spin-liquid phase. This feature is captured by the band structure of the unprojected wave function (with two Dirac points for each spin component). In addition, we observe an intense signal at low energies around the K points, which cannot be understood within the unprojected picture and emerges only when the Gutzwiller projection is considered, suggesting the relevance of gauge fields for the low-energy physics of spin liquids.

Dosage compensation in Drosophila is mediated by the MSL complex, which increases male X-linked gene expression approximately 2-fold. The MSL complex preferentially binds the bodies of active genes on the male X, depositing H4K16ac with a 3′ bias. Two models have been proposed for the influence of the MSL complex on transcription: one based on promoter recruitment of RNA polymerase II (Pol II), and a second featuring enhanced transcriptional elongation. Here, we utilize nascent RNA sequencing to document dosage compensation during transcriptional elongation. We also compare X and autosomes from published data on paused and elongating polymerase in order to assess the role of Pol II recruitment. Our results support a model for differentially regulated elongation, starting with release from 5′ pausing and increasing through X-linked gene bodies. Our results highlight facilitated transcriptional elongation as a key mechanism for the coordinated regulation of a diverse set of genes.

Determine global gene dysregulation affecting 4q-linked (FSHD-1) and non 4q-linked (FSHD-2) cells during early stages of myogenic differentiation. This approach has been never applied to FSHD pathogenesis.By in vitro differentiation of FSHD-1 and FSHD-2 myoblasts and gene chip analysis we derived that gene expression profile is altered only in FSHD-1 myoblasts and FSHD-2 myotubes. The changes seen in FSHD-1 regarded a general defect in cell cycle progression, probably due to the upregulation of myogenic markers PAX3 and MYOD1, and a deficit of factors (SUV39H1 and HMGB2) involved in D4Z4 chromatin conformation. On the other hand, FSHD-2 mytubes were characterized by a general defect in RNA metabolism, protein synthesis and degradation and, to a lesser extent, in cell cycle. Common dysregulations regarded genes involved in response to oxidative stress and in sterol biosynthetic process. Interestingly, our results also suggest that miRNAs might be implied in both FSHD-1 and FSHD-2 gene dysregulation. Finally, in both cell differentiation systems, we did not observe a gradient of altered gene expression throughout the 4q35 chromosome.FSHD-1 and FSHD-2 cells showed, in different steps of myogenic differentiation, a global deregulation of gene expression rather than an alteration of expression of 4q35 specific genes. In general, FSHD-1 and FSHD-2 global gene deregulation interested common and distinctive biological processes. In this regard, defects of cell cycle progression (FSHD-1 and to a lesser extent FSHD-2), protein synthesis and degradation (FSHD-2), response to oxidative stress (FSHD-1 and FSHD-2), and cholesterol homeostasis (FSHD-1 and FSHD-2) may in general impair a correct myogenesis. Taken together our results recapitulate previously reported defects of FSHD-1, and add new insights into the gene deregulation characterizing both FSHD-1 and FSHD-2, in which miRNAs may play a role.

Objective. To perform a preliminary test of a new rehabilitation treatment (FIT-SAT), based on mirror mechanisms, for gracile muscles after smile surgery. Method. A pre- and postsurgery longitudinal design was adopted to study the efficacy of FIT-SAT. Four patients with bilateral facial nerve paralysis (Moebius syndrome) were included. They underwent two surgeries with free muscle transfers, one year apart from each other. The side of the face first operated on was rehabilitated with the traditional treatment, while the second side was rehabilitated with FIT-SAT. The FIT-SAT treatment includes video clips of an actor performing a unilateral or a bilateral smile to be imitated (FIT condition). In addition to this, while smiling, the participants close their hand in order to exploit the overlapped cortical motor representation of the hand and the mouth, which may facilitate the synergistic activity of the two effectors during the early phases of recruitment of the transplanted muscles (SAT). The treatment was also aimed at avoiding undesired movements such as teeth grinding. Discussion. Results support FIT-SAT as a viable alternative for smile rehabilitation after free muscle transfer. We propose that the treatment potentiates the effect of smile observation by activating the same neural structures responsible for the execution of the smile and therefore by facilitating its production. Closing of the hand induces cortical recruitment of hand motor neurons, recruiting the transplanted muscles, and reducing the risk of associating other unwanted movements such as teeth clenching to the smile movements.
