Bovine respiratory disease (BRD) represents a significant challenge in cattle management due to its multifactorial nature and lack of a gold standard diagnostic method. Procalcitonin (PCT) has emerged as a potential biomarker for bacterial infections in various species, including cattle. This study aimed to investigate plasma PCT concentration variations in pre-weaned dairy calves categorized as BRD-positive using clinical scores (WRSC; BRD-positive ≥5), thoracic ultrasonography with two cut-off (TUS; BRD-positive ≥1 or ≥3), or a combination of both methods (WRSC/TUS1cm or WRSC/TUS3cm). Additionally, the accuracy of PCT in diagnosing BRD was evaluated. A cross-sectional study was conducted on a convenience sample of 226 pre-weaned Italian-Friesian female calves. Clinical scoring, TUS, and plasma PCT analysis were performed. Calves were categorized based on TUS findings, clinical scores, or a combination of both methods. Statistical analyses were conducted to assess the differences in PCT concentrations among different groups and to determine the diagnostic accuracy of PCT. Results showed a significant increase in PCT levels in calves with lung consolidation detected by TUS using a 1 cm cutoff. However, the diagnostic accuracy of PCT in discriminating BRD-positive cases was poor (area under the curve 0.62). The optimal cutoff value for PCT was determined to be 86.63 pg/mL, with sensitivity of 49.7%, specificity of 71.8%, positive predictive value of 79.4% and negative predictive value of 39.5%. In conclusion, while PCT showed potential as a biomarker for BRD, its diagnostic accuracy was limited in this study. Future research should focus on integrating PCT measurements with other diagnostic methods and conducting longitudinal cohort studies to better understand its role in BRD diagnosis and management.

Abstract Since the late twentieth century, the global fashion industry has been increasingly embracing the business model known as fast fashion. Characterised by rapid production cycles, fleeting trends, low-cost garments and large-scale production, fast fashion seems to meet consumer demand for affordable and trendy clothing. However, its environmental impact as a major polluter poses significant challenges to sustainability and circularity initiatives. This article presents the results of a systematic literature review, exploring the unsustainable consequences of fast fashion, focusing on both demand and supply side, from a geographical perspective. Using a Global North–Global South framework, it explores differences in socio-economic structures, consumption and production patterns, access to resources and environmental impacts. The analysis suggests that a fair and equitable transition towards a sustainable and circular fashion industry will require the links between business, society and nature to be reconsidered, to avoid perpetuating the inequalities associated with the global linear capitalist economy. The findings highlight the importance of both markets and institutions in sustainable growth. In the Global North, the most frequently discussed topics relate to investment and research and development with respect to new technologies or system innovations often with the support of well-structured political guidance. Conversely, in the Global sustainable initiatives tend to be scattered, country-specific and intricately tied to particular socio-economic and cultural contexts.

Procalcitonin (PCT) and protein carbonylated content (PCC) are promising biomarkers for bacterial infection and inflammation in veterinary medicine. This study examined plasma PCT and PCC levels in healthy cows (H) and cows with subclinical mastitis (SCM). A total of 130 cows (65 H and 65 SCM) were included in this study. Blood samples were collected, and plasma was frozen at −80 °C. PCT levels were determined using a bovine procalcitonin ELISA kit, while PCC was measured following the methodology of Levine et al. Statistical analysis revealed a significant difference in PCT levels between H (75.4 pg/mL) and SCM (107.3 pg/mL) cows (p < 0.001) and significantly lower concentrations of PCC in the SCM group (H: 0.102 nmol/mL/mg, SCM: 0.046 nmol/mL/mg; p < 0.001). The PCT cut-off value for distinguishing healthy and subclinical mastitis animals was >89.8 pg/mL (AUC 0.695), with a sensitivity of 66.2% and specificity of 69.2%. PCT showed potential value as a diagnostic tool to help in decision making for subclinical mastitis cases, while PCC requires further studies to investigate the trend of this biomarker during localized pathology.

Welfare assessment of dairy calves is commonly conducted through the observation of their behavior and the analysis of physiological parameters. Despite the large number of studies on this topic, there is a lack of research on the possible correlation between the physiological parameters and behavior at basal level in dairy calves. For this reason, the present study aimed to investigate the possible correlations between serum cortisol, neutrophil-to-lymphocyte ratio (N/L) and locomotor play behavior in 21 Friesian dairy calves within their first month of life. Every week, an arena test after blood collection was performed. A significant positive correlation between serum cortisol levels and N/L (rrm = 0.381; 95% CI = 0.083, 0.617; p = 0.012) and a negative correlation between time spent running and N/L (rrm = −0.351; 95% CI = −0.575, −0.077; p = 0.012) were found. On the contrary, no significant correlation between time spent running and cortisol levels was detected. Cortisol levels and N/L were higher in the newborn and had a constant decrease until reaching a stable value after the 24th day of life. These findings suggest a better predictivity of N/L, compared to serum cortisol, on locomotor play behavior.

Robust Animal-Based Measures (ABMs) are fundamental in order to assess animal welfare, however in semi-extensive sheep farming systems is not easy to collect ABMs without inducing additional stress in the animals. Heart rate variability (HRV) is a non-invasive technique of assessing stress levels related to animal welfare. It is considered a sensitive indicator of the functional regulatory characteristics of the autonomic nervous system. Several studies investigated the use of HRV for welfare assessment in dairy cows while research on sheep is scarce. Moreover, assessing HRV in small ruminants at pasture is critical because of the lack of a solution adoptable for field conditions. This study aimed to evaluate if a smart textiles technology is comparable to a Standard base-apex electrocardiogram (ECG) for measuring HRV in small ruminants. Eight healthy Massese dairy sheep were recruited. Standard base-apex ECG and smart textiles technology (Smartex ECG) were simultaneously acquired for 5 min in the standing, unsedated, unclipped sheep. The ECG tracings were recorded when animals were standing quietly. The Bland-Altman test and the linear regression analysis were applied after parameter extraction in time, frequency, and non-linear methods to compare Smartex against standard base-apex ECG systems. The Bland-Altman test was applied to all HRV extracted parameters (Mean RR, pNN50, RMSSD, LF/HF, SampEn, SD1, SD2, stdRR) to evaluate the agreement between the two different instruments, and a linear regression analysis was performed to evaluate the relationship between the two methods. The smart textiles biotechnology was simple to wear and clean. It can be worn without using glue and without shaving the sheep's wool, limiting animal handling and stress. Bland Altman test reported a robust agreement between the two systems. In fact, the regression analysis of HRV parameters showed that half of the parameters recorded had an R2 coefficient >0.75. Results also showed a very small reproducibility coefficient that indicated that the two methods were really close to each other. Smartex textiles technology can be used for HRV evaluation in sheep species as a potential ABM for animal welfare assessment.

Background: The approach to respiratory diseases in donkeys is similar to that for horses, nevertheless bronchoalveolar fluid (BALF) and tracheal wash (TW) cytology in this species have been described only a few times in the literature. Aim: To describe BALF cytological findings in a cohort of 24 healthy Amiata donkeys. Methods: Bronchoalveolar lavage (BAL) was performed under standing sedation with a large animal where the broncho-alveolar lavage catheter passed blindly through the nasal passage into the trachea. Results: The total nucleated cell count (TNCC) of the BALF was found similar to that already described in healthy horses and donkeys. No differences in the differential count were observed according to age and sex. A decreased macrophage percentage and an increased eosinophil percentage was observed in our donkey population when compared to the existing reference range for horses. Conclusions: The reference intervals for BAL cytology in donkeys may be significantly different for those referred for horses.

We evaluated the preventive effects of the oral administration of chestnut tannins (Castanea sativa) together with its potential metabolic effect on calf diarrhoea. Forty Italian Friesian female calves were included and divided into Group C (control group) and Group T (tannin-treated group). From the third day of life (T0) for the following 56 days (T56), calves from Group C received 2 L of warm water, while 10 g of chestnut tannin powder extract were added to Group T. Calves were weighed at birth and at T56. Daily faecal score evaluation was performed according to the literature. The age at diarrhoea onset (TDE) and the duration of the diarrhoeic episode were recorded. Blood methaemoglobin and liver enzymes were evaluated weekly starting from T0 to T56 by spectrophotometer and clinical chemistry analysis, respectively. The t-Student and chi-square tests were performed. The TDE was higher (p = .04) in Group T than in Group C (12.0 ± 8.2 and 7.7 ± 3.8 days, respectively). There were no differences for ADG between the groups. Group C spent 24.4% of the whole period with diarrhoea, whereas Group T experienced diarrhoea for 18.9% of the period (p = .001). All the blood and serum analytes were within physiological values. The administration of tannins in calves from the third day of life seemed to delay the onset of diarrhoea by almost four days, suggesting effective preventive action of chestnut tannins.HIGHLIGHTS The oral administration of chestnut tannins in calves seemed to delay the onset of diarrhoea by almost four days, thus showing a preventive effect. The oral administration of chestnut tannins may lead to a better and shorter recovery of the calf, thus decreasing the use of antibiotics.

The aims of the present study were to evaluate the preventive and the therapeutic effect of Stodi® as phytogenic feed additive rich in phenolic substances on the calf diarrhea, during the first 24 days of life. A total of 40 calves were included and randomly divided into Group C (control group) and Group T (treated group) with placebo or treatment administration started from the third day of life (T0). Calves belonged to group C received 2 L of warm water, while the calves assigned to group T received 2L of warm water plus 30 g of Stodi®. Solutions administration was maintained until day 21 (T21) that was the end of the experimental period. Calves were weighed at T0 and T21 to assess the average daily gain (ADG). Physical examination and fecal score evaluation were performed daily. The duration of a diarrheic episode, the age of the first diarrhea outbreak (TDE) and the frequency of diarrheic episodes were recorded. Complete blood count, methemoglobin and liver enzymes were evaluated at T0 and at T21 in all the calves by spectrophotometer and clinical chemistry analysis, respectively. Data were analyzed using a mixed model. A Chi-square and a Mann-Whitney test were also performed. No difference was found for ADG between the groups. The difference of mean age at TDE was not statistically significant between C and T group. The number of calves with diarrhea in the C group tended to be higher than that of T group (p = 0.13). Calves in group C spent more days with clinical sign of diarrhea compared to group T (p = 0.016). Complete blood count, methemoglobin and liver enzymes were within the reference ranges. The feed additive Stodi® seemed to be effective in shortening neonatal diarrhea episodes in calves thanks to the administration of 30 g per day of product. The fixed dosage of Stodi® used in our study did not show a preventive effect to reduce the incidence of calf diarrhea.

The aim of this study was to compare forelimb claw dimensions in buffalo cows, and to establish a reference values for functional claw trimming. Medial and lateral healthy front feet claws were evaluated from 20 young (YB) and 20 mature (MB) Egyptian buffalo cows (Bubalus bubalis). The dorsal wall length (TL), dorsal wall angle (TA), heel length (HBL), height (HBH) and width (HBW), sole (SL) and claw length (CL) and sole width (SW) were measured. Differences between YB and MB were evaluated using ANOVA test, while those between the lateral and medial claws with a paired t test. Before and after trimming, the HBL, HBW, HBH, SL and CL were significantly higher for both claws in MB, while the TA was significantly lower. Before trimming the lateral HBW and HBH were significantly higher than the medial, while the SL and CL of the medial claw were significantly higher than the lateral claw. After trimming, the lateral claw HBL was significantly higher in MB. The SW of the lateral claw was significantly higher than the medial claw in both YB and MB, and it did not change after trimming. These results can be considered as a guidance during front feet trimming in buffaloes.

Veterinary facility admissions of wild animals are increasing alongside the interest in wildlife diseases. To improve animal welfare, it is therefore important to increase veterinarians’ knowledge of wild animal medicine and to improve the clinical and diagnostic procedures, especially in the case of patients affected by trauma or multiple traumas. Blood analysis can be a quick and minimally invasive way of gathering useful clinical information for adequate treatment and management, and, together with a good clinical examination, to help predict hospitalisation outcomes. Few papers have reported reference ranges for the haematological and biochemical parameters of roe deer. This study evaluates the haematological and biochemical parameters in traumatised roe deer in relation to animal hospitalisation outcomes. The study was carried out on a cohort of 98 roe deer divided into groups according to their age and hospitalisation outcome. For each animal, a panel of haematological and biochemical parameters was performed. Significant differences were found between unweaned (4 months old) groups for total bilirubin. Creatine kinase, creatinine, and bilirubin may be useful indicators to correlate with the severity of trauma and help predict prognosis.

Equine Gastric Ulcer Syndrome (EGUS) is a worldwide disease of the stomach that can be found in different categories of horses. Different clinical signs may be present, but a large number of horses are asymptomatic. The aim of this study was to identify a possible correlation between body condition score (BCS) and EGUS in a population of horses. A total of 203 non-athlete horses were submitted for gastroscopies, and the presence and severity of lesions of the glandular and squamous mucosa were recorded. A board-certified veterinarian blinded to the gastroscopy results assessed the BCS of the horse. In the study population, no correlation was found between BCS and the presence of gastric lesions in either the glandular or the squamous mucosa. The disease of the squamous or glandular mucosa cannot be excluded based only on the presence of normal or increased BCS in non-athlete horses.

The body condition score (BCS) represents a practical but subjective method for assessing body fat reserves. Real time ultrasonography (RTU) has been proposed as an accurate method to objectively measure subcutaneous fat (SF) thickness and predict body fat reserves in cows, horses and donkeys. The aim of the present study was to describe RTU measures of SF thickness during periparturient period in jennies. The present prospective cohort study evaluated six dairy jennies. SF RTU were performed at 15 and 7 days before the presumptive delivery, and 2, 15 and 30 days after delivery. A portable ultrasound machine and multifrequency linear transducer (5–7.5 MHz) was used. RTU images were obtained in six sites (S1–S6). Results at each time point were reported as mean ± standard deviation and compared through time. A total of 180 images were evaluated. RTU technique was easy to perform and well tolerated. No statistically significant differences were found of each site during time, except for S2 and S6a: S2 at T2 and S6a at T1 were significatively different to values obtained at T5. The RTU mean values were above those reported by others, suggesting major physio-logical challenges related to energy balance and fat mobilization in pregnant jennies bred for milking production. BCS and sites through observational time have shown a good and reliable association. Our study could give preliminary indications on fat reserves in different body locations evaluated thanks to RTU and it show no significative variation of SF thickness, in pregnant and lactating jennies.

Literature on the protein carbonyl content (PCC) evaluation in horses is scarce, thus the aims were to evaluate the PCC in healthy and SIRS (Systemic Inflammatory Response Syndrome) horses and to investigate the performances of PCC in terms of sensitivity, specificity, and likelihood ratio in identifying SIRS positive and negative horses. A total of 72 adult horses were included. All the horses underwent to a complete physical examination, blood analysis, and were evaluated for the SIRS status. Blood samples were collected once in healthy horses and at admission time, then 24, 48, 72, and 96 h after admission in sick animals. PCC was evaluated using a method previously described. Data were statistically analyzed to verify differences in PCC between healthy vs. SIRS positive or SIRS negative horses at all sampling time. The receiver operating characteristic (ROC) curve was performed to verify sensitivity and specificity of PCC in the diagnosis of SIRS-positive and SIRS negative horses. The healthy horses were standardbred mares with a median age of 8.5 years. The sick horses were 31/54 females, 16/54 geldings, and 7/54 stallions of different breeds and with a median age of 12 years old. Eight out of 54 sick horses were SIRS negative, while 46/54 were SIRS positive. Statistically significant differences were obtained between healthy and SIRS positive horses, while no differences were observed between healthy and SIRS negative horses at any sampling time. The best cutoff value of PCC to discriminate between SIRS positive, SIRS negative, and healthy horses, the sensitivity and specificity of cutoff point, the area under receiver operating characteristic curve, the 95% confidence intervals, and the likelihood ratio were reported. We found higher PCC values in sick SIRS-positive horses vs. healthy ones with a decrement over time, while no differences at admission, nor during the observational period, were obtained in sick but SIRS-negative horses. The value of 0.049 nmol/ml/mg is reported as a potential cutoff for the diagnosis of SIRS positivity vs. healthy horses with a sensibility of 74.5% and a specificity of 72.2%. In conclusion, PCC seems to be a sensitive and specific marker for SIRS in horses.

Background: We analyzed the clinical data of wildlife ungulates admitted for emergency care to the Veterinary Teaching Hospital (VTH), Department of Veterinary Medicine, University of Pisa over a 9-years period.Methods: Clinical data of all the wildlife ungulates admitted to the VTH were recorded. Blood samples were also taken from the animals for hematological and biochemical analysis. An assessment of ecto- and endoparasites was carried out, diagnostic imaging assessment was performed, and the outcomes were recorded.Results: Data concerning clinical parameters, blood work, parasitological analysis, and diagnostic imaging diagnosis were expressed as prevalence.Conclusion: The rescue and emergency treatments were related mostly to traumas caused by car accidents, followed by other causes. The traumatic injuries were mostly severe, characterized by multiple lesions involving hard and soft tissues. In this study, traffic accidents were the main cause of wildlife rescue and emergency management. This is probably due to the increased population of ungulates over the years, along with the considerable anthropization of the Pisa area.

Immunoglobulin type G (IgG) concentration both in jennies' colostrum and in serum of donkey foals are mostly unknown in the first 24 h after delivery. The aims of the present study were to evaluate the IgG concentrations of colostrum during the first 24 h of lactation of Amiata jennies, the absorption of colostrum and the weekly body weight gain of the donkey foals. IgG concentrations were assessed in the jennies' colostrum and in the serum of donkey foals. Colostrum was collected in 9 jennies ready after delivery, and at 6, 12, 24 h after foaling from both halves. Serum was collected at the same sampling times from 9 donkey foals. Donkey foals were weighted at birth and then weekly until the 28th days of life. Temporal changes of IgG concentrations in dam's colostrum and in donkey foal serum were analyzed by a linear regression model and a general linear model, respectively. Results showed that colostrum IgG concentration were similar between the left and the right half. Colostrum IgG concentrations decreased continuously throughout the time in all jennies by 0.0244 Log10 mg/mL per hour. Serum IgG concentrations in donkey foals at birth was significantly lower compared to other times. No correlation was found between the colostrum IgG concentrations and the average weekly body weight gain of the donkey foal. The pattern of colostrum IgG levels in jennies and serum IgG concentration in donkey foals seem to be similar to what reported for equine. However, the donkey foals seem to be less agammaglobulinemic at birth compared to the horse foal. The pattern and both serum and colostrum concentrations evaluated in the Amiata donkeys were slightly different from results reported in other donkey breeds, underlying the importance of setting references specific to breed.

The aim was to evaluate the use of a bovine procalcitonin (PCT) ELISA kit (Cusabio, China) for assessing PCT in bovine milk samples. Validation was performed by using 10 plasma and corresponding milk samples from mastitic cows. The limit of detection (LOD) was calculated. The coefficient of variation (CV%) of the readings of five plasma samples measured five times in the same plate (intra-assay) and the CV% of the same five samples read five times in three separate plates was evaluated. Parallelism was determined by serial twofold dilutions of five plasma and corresponding milk samples. Milk samples were analyzed with and without centrifugation. Regarding plasma PCT, the method presented an inter- and intra-CV < 23.7% and parallelism had very good recovery values. The ELISA kit studied can measure bovine plasma PCT concentrations. The kit antibodies fail in binding PCT in milk samples because all centrifuged milk samples showed a lower LOD than blank samples. Only three uncentrifuged milk samples showed measurable PCT concentrations. Due to these results, the commercial ELISA kit investigated could not be employed for the detection of PCT in milk samples.

The period from late gestation to early lactation is characterized by changes in energy metabolism. The aim of the current study was to analyze the plasma total lipid fraction using gas chromatography (GC) analysis, in order to highlight metabolic changes from the pre-foaling to post-foaling periods. Eleven pluriparous dairy jennies (mean age of 11.88 ± 3.79 years) belonging to the Amiata donkey breed were enrolled. Blood sampling was performed at 15 days before foaling (T0), and 15 (T1), 30 (T2), 60 (T3), and 90 (T4) days after foaling, for biochemical and GC analysis. A total of 37 fatty acids were identified in plasma samples: 4 medium chain (MCFA), 24 long chain (LCFA), and 9 very-long chain (VLCFA) fatty acids. Among them, 20 fatty acids changed significantly, and two fatty acid showed a trend toward significance. Furthermore, the LCFA, saturated, unsaturated, monounsaturated, and polyunsaturated ω-3 fatty acids changed significantly during the study period. The main alterations were between T0 and the other time points and appeared to be related to lipid metabolism, cellular structure and function, and inflammatory and immune responses. Our findings reveal greater energy requirements at the end of gestation compared to early lactation in donkeys.

The aim of this study was to evaluate three commercially available rapid immunochromatographic tests for detection of Cryptosporidium parvum antigens in faeces of naturally infected neonatal diarrhoeic calves. FASTest® CRYPTO strip, FASTest® CRYPTO-GIARDIA Strip and TETRASTRIPS® were compared for their sensitivity, specificity, positive predictive value and negative predictive value using a cumulative positivity as gold standard. In addition, the agreement between each test and the gold standard was evaluated by Cohen’s Kappa (k) value. The highest infection rate was observed by FASTest® CRYPTO-GIARDIA Strip (65.15%), followed by FASTest® CRYPTO strip (63.64%) and TETRASTRIPS® (56.06%,). A very good diagnostic performance of all the three tests was observed. FASTest® CRYPTO strip (k = 0.935) and FASTest® CRYPTO-GIARDIA Strip (k = 0.968) had the highest sensitivity (100%) while TETRASTRIPS® (k = 0.875) had the highest specificity (100%). Eimeria spp oocysts were present in six samples but cross-reaction with this protozoan was not observed. These assays were not time-consuming and very easy to perform and to read. Based on our results, we recommend the use of FASTest® CRYPTO strip, FASTest® CRYPTO-GIARDIA Strip or/and TETRASTRIPS® for detection of C. parvum antigens in faeces of neonatal diarrhoeic calves.

Our aim was to measure the concentrations of derived reactive oxygen metabolite (d-ROMs) and biological antioxidant potential (BAP) of donkey mares and foals at delivery and to verify the protective role of the placenta against fetal oxidative stress. A total of 15 Amiata jennies with a physiological gestation length and delivery were included together with 17 foals (two twin foalings). After delivery, maternal and foal venous blood samples were collected along with blood from the artery. Circulating lactate and plasma d-ROMs and BAP were evaluated. The Wilcoxon test for paired data was applied to verify differences in d-ROMs and BAP values, while the Spearman test was used to evaluate correlations. A significantly higher d-ROMs concentration was found in jennies compared to their foals, and to the umbilical artery blood. The BAP was higher in jennies than in their foals, but no differences were observed in the umbilical artery blood. No difference was found between foals and their umbilical cord. Blood lactate was higher in foals than in their dams. Positive correlations were found between mares and umbilical cord for BAP and d-ROMs, and between mares and foals and umbilical cord for BAP. In conclusion, the placenta may be a protective factor for the fetus. As with equine foals, the antioxidant system of donkey foals does not seems to be effective at birth.

Colic horses show systemic inflammatory response syndrome (SIRS) clinical signs. Procalcitonin (PCT) showed increased circulating levels in sick horses. This study compares plasma PCT concentrations in healthy vs. SIRS negative/positive colic horses over time, and evaluates PCT and SIRS score potential correlation, to verify the usefulness of PCT for the evaluation of SIRS severity. Ninety-one horses were included; 43/91 were healthy, on basis of physical examination, blood work and SIRS score (score = 0), while 48/91 were sick colic horses, classified as SIRS-negative (score < 2) and positive (score ≥ 2). Moreover, a 0–6 point-scale SIRS score was calculated (assessing mucous membrane color and blood lactate concentration). PCT was evaluated at admission, and at 24, 48, 72 and 96 h, using a commercial kit for equine species. We verified by the ANOVA test PCT differences between healthy vs. colic horses, healthy vs. SIRS-negative or SIRS-positive colic horses, at all sampling times, and the correlation between the SIRS score at admission with the SIRS score. Statistically significant differences were detected between healthy vs. all colic horses and between healthy vs. SIRS-positive or negative horses at all sampling times. No correlation was observed between the SIRS score at admission and PCT values. PCT was statistically higher in colic horses compared to the healthy ones, suggesting a role as a biomarker for colic.

Body measurements could be used to estimate body weight (BW) with no need for a scale. The aim was to estimate heifers weight based on their body dimension characteristics. Twenty-five Holstein heifers represent the study group (SG); another 13 animals were evaluated as a validation group (VG). All the heifers were weighed (BW) and their wither height (WH), shin circumference (SC), heart girth circumference (HG), body length (BL), hip width (HW) and body condition score (BCS) were measured immediately after birth, and then weekly until 2 months and monthly until 15 months old. Equations were built with a stepwise regression in order to estimate the BW at each time using body measures for the SG. A linear regression was applied to evaluate the relationship between the estimated BW and the real BW. Equations found were to be statistically significant (r2 = 0.688 to 0.894; p < 0.0001). Three variables or fewer were needed for BW estimation a total of 11/23 times. Regression analysis indicated that the use of HG was promising in all the equations built for BW estimation. These models were feasible in the field; further studies will evaluate possible modifications to our equations based on different growing rate targets.

The aims of the present study were to evaluate the correlation between IgG Serum Radial Immunodiffusion (SRID), Electrophoresis Gamma Globulins (EGG), Electrophoresis Total Protein (ETP) and the serum total protein (TP) analyzed by refractometry and by a dry chemistry analyzer (Biuret) and to estimate serum IgG concentrations using serum TP. A total of 36 samples collected at four different times (birth, 6, 12, 24 h after birth) from nine Amiata donkey foals were evaluated with SRID, EGG, ETP, serum TP Biuret and refractometry. SRID IgG concentration increased significantly over time until T12. Serum TP analyzed with refractometry, electrophoresis and Biuret showed a statistically significant difference between T0 and T6 vs. T12 and T24. A good or strong correlation was found between different tests performed. Equations to quantify serum IgG were created and can be used for estimating the donkey foals’ serum IgG in the first day of life. Serum TP refractometry showed a high correlation with SRID IgG (0.91) which may be a particularly useful and economic instrument to estimate the transfer of immunity in donkey foals during the first day of life. Further studies evaluating a high number of animals are needed in order to set specific cut-off values.

We evaluated the udder cistern (UC) size during the dry period using ultrasound. Forty healthy quarters were evaluated in both the longitudinal and cross-section of the UC. Quarters were evaluated at the drying-off (T0) and 24 h later (T1), then regularly until the end of the dry period (T7–T58), during the colostrum production phase (TCPP) and at 7 days in milking (T7PP). The Spearman test was applied to find the correlation between the ultrasonographic UC size (UUCS) assessment and time. The Friedman test and Dunn’s test for multiple comparisons as a post-hoc test were performed to compare the forequarter and hindquarter cross-sections (FQCSs and HQCSs, respectively) and the forequarter and hindquarter longitudinal sections (FQLSs and HQLSs, respectively) at T0 vs. T58 vs. TCPP vs. T7PP. A total of 440 images were evaluated. A negative linear correlation between time and FQCS and FQLS (r = −0.95; p < 0.0004) and between time and HQCS and HQLS (r = −0.90; p < 0.002) was found. The UUCS decreased throughout the dry period, starting to increase at the beginning of the next lactation. Measuring the UUCS provides useful information for monitoring the dry period.

In human medicine, procalcitonin (PCT), the precursor of calcitonin, is used for the rapid identification of the origin and severity of sepsis. In veterinary medicine, PCT has been studied in horses, cattle, and dogs, but the use of PCT in diagnostic and/or prognostic settings is not possible because of the lack of validated assays to obtain reference ranges. The aim of the present study was the investigation of commercially available ELISA kits for the detection of canine and equine PCT in plasma samples. Validation of the ELISA kits was performed by using species-specific recombinant proteins spiked both in plasma and buffer samples; linearity, limit of detection (LOD), recovery, and intra-assay and inter-assay variability were calculated. Moreover, clinical samples obtained from sick and healthy animals were also analyzed with the tested kits. Canine PCT was measured with a recombinant canine and a canine PCT ELISA kit. Equine PCT was measured with an equine and a human ELISA PCT kit. Our data demonstrate that the canine recombinant PCT ELISA kit can be used to measure canine PCT in plasma samples, showing an intra-assay and inter-assay coefficient of variation less than 20% and a LOD of 11 pg/mL, whereas the present results do not support the use of the canine PCT ELISA kit. The human PCT ELISA kit is suitable to detect equine PCT with a LOD of 56 ng/mL, whereas the equine PCT ELISA kit did not detect recombinant equine PCT.
