Abstract

International audience; The monitoring of the water cycle at the Earth surface which tightly interacts with the climate change processes as well as a number of practical applications (agriculture, soil and water quality assessment, irrigation and water resource management, etc...) requires surface temperature measurements at local scale. Such is the goal of the Indian-French high spatio-temporal TRISHNA mission (Thermal infraRed Imaging Satellite for High-resolution Natural resource Assessment). The scientific objectives of the mission and research work conducted to consolidate the mission specifications are presented. Progress in modelling of surface fluxes is then discussed. The main specifications of the mission such as the revisit, the spatial resolution, the overpass time, the spectral bands and the orbit are analyzed and justified. The resulting baseline of the mission is given.

Abstract

The aim of this report is to provide an overview of the final version of the PIPER framework and application. The software, along with its documentation, and not the report, constitutes the main part of the deliverable. The software and documentation were already distributed at the Final Workshop and online (under the Open Source license GPLv2 or later for the software, and the GNU FDL 1.3 license for the documentation). The documentation includes detailed descriptions of the framework principles, user interface, metadata, along with the modules and their parameters. It also includes application scenarios (called workflows). Information about the use of the modules is complemented by Tutorials that were developed as part of WP1 (online on the wiki) and explanatory videos were developed as part of WP4 (videos of the final workshop, now available on YouTube). The headers in the source code files (also available online) list the main contributors to the software. The report will therefore not provide details about information that is already available elsewhere but will only provide a very brief summary of the functionalities available. Some of the descriptions are excerpts of the manual.

Abstract

International audience; Finite Element Human Body Models (HBM) have become powerful tools for injury prediction. However, they are developed for a few limited size and age in a few standardized positions, limiting their usage. The aim of the FP7 European Commission co-funded project PIPER, initiated in November 2013, is to develop a modular open source (License GPL v3) framework with advanced numerical methods to personalize and position a HBM. Challenges are to design a user friendly software, usable for any Finite Element code (customizable parser) and any HBM through the definition of HBM metadata, which proposes numerical methods to transform HBMs that lead to an output model, the mesh quality of which is preserved. The current version is developed using standard tools and codes (C++, python, Qt, VTK...). Dual Kriging interpolation method [1] is implemented for non-linear scaling of the HBM. For positioning, a lightweight physics models is generated from HBM. It can be interactively manipulated by the user to define the targeted position. In addition, specific approaches are available, namely the Frame-based approach [1] and the Contour based approach [2]. The preliminary version of the software (windows and linux versions) was tested by 5 partners on 3 models in predefined scenarios (see figure). The performance evaluation of the transformation methods regarding the mesh quality demonstrates the critical need for mesh improvement tools. This issue however strongly depends on the design features of the HBMs such as the coupling between soft tissue deformation and the bones transformation. Finally, to help the definition of a complete set of personalizing or positioning targets, statistical a priori knowledge will be introduced.

Plant biomass is a large source of fermentable sugars for the synthesis of bioproducts using engineered microbes. These sugars are stored as cell wall polymers, mainly cellulose and hemicellulose, and are embedded with lignin, which makes their enzymatic hydrolysis challenging. One of the strategies to reduce cell wall recalcitrance is the modification of lignin content and composition. Lignin is a phenolic polymer of methylated aromatic alcohols and its synthesis in tissues developing secondary cell walls is a significant sink for the consumption of the methyl donor S-adenosylmethionine (AdoMet). In this study, we demonstrate in Arabidopsis stems that targeted expression of AdoMet hydrolase (AdoMetase, E.C. 3.3.1.2) in secondary cell wall synthesizing tissues reduces the AdoMet pool and impacts lignin content and composition. In particular, both NMR analysis and pyrolysis gas chromatography mass spectrometry of lignin in engineered biomass showed relative enrichment of non-methylated p-hydroxycinnamyl (H) units and a reduction of dimethylated syringyl (S) units. This indicates a lower degree of methylation compared to that in wild-type lignin. Quantification of cell wall-bound hydroxycinnamates revealed a reduction of ferulate in AdoMetase transgenic lines. Biomass from transgenic lines, in contrast to that in control plants, exhibits an enrichment of glucose content and a reduction in the degree of hemicellulose glucuronoxylan methylation. We also show that these modifications resulted in a reduction of cell wall recalcitrance, because sugar yield generated by enzymatic biomass saccharification was greater than that of wild-type plants. Considering that transgenic plants show no important diminution of biomass yields, and that heterologous expression of AdoMetase protein can be spatiotemporally optimized, this novel approach provides a valuable option for the improvement of lignocellulosic biomass feedstock.
