Abstract

The potential of a voltametric E-tongue coupled with a custom data pre-processing stage to improve the performance of machine learning techniques for rapid discrimination of tomato purées between cultivars of different economic value has been investigated. To this aim, a sensor array with screen-printed carbon electrodes modified with gold nanoparticles (GNP), copper nanoparticles (CNP) and bulk gold subsequently modified with poly(3,4-ethylenedioxythiophene) (PEDOT), was developed to acquire data to be transformed by a custom pre-processing pipeline and then processed by a set of commonly used classifiers. The GNP and CNP-modified electrodes, selected based on their sensitivity to soluble monosaccharides, demonstrated good ability in discriminating samples of different cultivars. Among the different data analysis methods tested, Linear Discriminant Analysis (LDA) proved to be particularly suitable, obtaining an average F1 score of 99.26%. The pre-processing stage was beneficial in reducing the number of input features, decreasing the computational cost, i.e., the number of computing operations to be performed, of the entire method and aiding future cost-efficient hardware implementation. These findings proved that coupling the multi-sensing platform featuring properly modified sensors with the custom pre-processing method developed and LDA provided an optimal tradeoff between analytical problem solving and reliable chemical information, as well as accuracy and computational complexity. 6. Magnetic Beads as Versatile Tools for Electrochemical Biosensing Platforms in Point‐of‐Care Testing.

Abstract

Point‐of‐care technologies need particularly stringent requirements, including portability, rapidity, affordability, and ease of use, enabling healthcare practitioners to perform reliable analyses in decentralized settings. Such diagnostic tests are usually based on bioassays that allow for the detection of clinically relevant biomarkers, namely proteins, antibodies and nucleic acids. In the context of electrochemical biosensors, the implementation of magnetic beads offers unique advantages stemming from their high surface‐to‐volume ratio, easy manipulation through magnetic fields and versatility in surface chemistries for bio‐conjugation. The present concept article highlights the advantages achieved through the implementation of magnetic beads in electrochemical binding assays, immunosensors and genosensors. These methods mostly exploit magnetic beads for immobilization purposes, allowing for the assembly of bio‐complexes in liquid suspensions under precise temperature and shaking control, as well as simplifying the manipulation steps to enable untrained personnel to perform analyses. In addition, magnetic beads can also take part in the detection mechanism, leading to innovative sensing strategies with great potential for point‐of‐care diagnostic applications. 16. A Stand-Alone Portable Potentiostat With Parallel Channels for Smart Electrochemical Analyses

Abstract

In recent years portable potentiostats have increased in popularity allowing to perform electrochemical analyses outside the laboratories, moving them to home or point of care (PoC) environments. In this context, the idea of performing multiple acquisitions at the same time is particularly attractive to deal with replicates or with simultaneous multiple quantitative assessments of different analytes, shortening the time required for the analyses and/or increasing data reliability. Multiple parallel channels on a compact and wireless device can maximize the overall system usability. In this article, a multichannel Wi-Fi-based portable potentiostat is described. The device features four independent channels, which can be conditioned with different voltages. The device was designed to minimize the component count and the power consumption, obtaining 23.5 mW per channel. The system was electrically characterized, obtaining an excellent linearity ( $R^{2} =0.9999$ ), a maximum channel-to-channel mismatch of 1% when the maximum current range is selected, and a negligible crosstalk among the channels. Moreover, the multichannel potentiostat was tested in the real case scenario of a semiquantitative evaluation of the anti-tissue transglutaminase target antibodies in celiac disease. Two replicates of IgG and IgA were simultaneously acquired, showing a good capability of separating the positive and the negative samples, with a reduction of the acquisition time of 76% with respect to a single channel solution.

Abstract

The present PhD thesis mainly focused on design, development and validation of amperometric genosensors and genoassays based on immobilization of PNA probes on advanced and highly performing micro- and nanomaterials. The application field of such devices concerned the rapid and reliable determination of non-amplified DNA from genetically modified organisms (i.e. transgenic soy). Other issues faced during the PhD program were i) the development of portable IoT devices for acquisition and storage/sharing of data from biosensors intended for determination of clinically relevant biomarkers and ii) the implementation of piezoelectric mass sensors based on the electropolymerization of cavitands on quartz crystals for monitoring of environmentally relevant aromatic compounds in wastewaters. Il presente lavoro di tesi di dottorato è focalizzato principalmente sulla progettazione, sviluppo e validazione di genosensori e genosaggi amperometrici basati sull'immobilizzazione di sonde PNA su micro e nanomateriali avanzati altamente performanti. Il campo di applicazione di tali dispositivi ha riguardato la determinazione rapida e affidabile di DNA non amplificato da organismi geneticamente modificati (soia transgenica). Altri progetti affrontati durante il programma di dottorato hanno riguardato i) lo sviluppo di dispositivi IoT portatili per l'acquisizione e l'archiviazione/condivisione di dati ottenuti da biosensori dedicati alla determinazione di biomarcatori di interesse clinico e ii) l'implementazione di sensori di massa piezoelettrici basati sull'elettropolimerizzazione di cavitandi su cristalli di quarzo per il monitoraggio di composti aromatici di interesse ambientale nelle acque reflue.
