Abstract

The first scientific meeting of the newly established European SYSGENET network took place at the Helmholtz Centre for Infection Research (HZI) in Braunschweig, April 7-9, 2010. About 50 researchers working in the field of systems genetics using mouse genetic reference populations (GRP) participated in the meeting and exchanged their results, phenotyping approaches, and data analysis tools for studying systems genetics. In addition, the future of GRP resources and phenotyping in Europe was discussed.

Abstract

The European Mouse Mutagenesis Consortium is the European initiative contributing to the international effort on functional annotation of the mouse genome. Its objectives are to establish and integrate mutagenesis platforms, gene expression resources, phenotyping units, storage and distribution centers and bioinformatics resources. The combined efforts will accelerate our understanding of gene function and of human health and disease.

26. Differences in the organization and chromosomal allocation of satellite DNA between the European long tailed house mice Mus domesticus and Mus musculus.

Abstract

We compared the organization of satellite DNA (stDNA) and its chromosomal allocation in Mus domesticus and in Mus musculus. The two stDNAs show similar restriction fragment profiles after digestion (probed with M. domesticus stDNA) with some endonucleases of which restriction sequences are present in the 230-240 bp repetitive unit of the M. domesticus stDNA. In contrast, EcoRI digestion reveals that M. musculus stDNA lacks most of the GAATTC restriction sites, particularly at the level of the half-monomer. The chromosome distribution of stDNA (revealed by an M. domesticus stDNA probe) shows different patterns in the M. domesticus and M. musculus karyotypes, with about 60% of M. domesticus stDNA retained in the M. musculus genome. It is particularly noteworthy that the pericentromeric regions of M. musculus chromosomes 1 and X are totally devoid of M. domesticus stDNA sequences. In both groups, the differences in energy transfer between the stDNA-bound fluorochromes Hoechst 33258 and propidium iodide suggest that AT-rich repeated sequences have a much more clustered array in the M. domesticus stDNA, as if they are organized in tandem repeats longer than those of M. musculus. Considering the data as a whole, it seems likely that the evolutionary paths of the two stDNAs diverged after the generation of the ancestral 230-240 bp stDNA repetitive unit through the amplification, in the M. domesticus genome, of a family repeat which included the EcoRI GAATTC restriction sequence. 31. Nonrandom association between a specific autosome and the X chromosome in meiosis of the male mouse: possible consequence of the homologous centromeres’ separation

Abstract

A nonrandom association is described between chromosome No. 15, which is involved in the T(14;15)6Ca translocation, and the X chromosome in diakinesis/ metaphase I plates of the male mouse. The tight attachment of centromeric heterochromatin (CH) regions of chromosomes X and 15 is predominantly found in those primary spermatocytes in which the distance between the homologous CH regions of the No. 15s exceeds some critical value. The rule holds true for both T6 homozygotes (i.e., for those of their primary spermatocytes displaying two 15t univalents) and T6 heterozygotes (i.e., for translocation configurations “III + I” and certain types of “chain-of-four” quadrivalents). The tight X-15 CH association is highly specific, since out of 1540 primary spermatocytes of the T6 heterozygotes, only 3 showed the tight X-14 CH association. In contrast to the tight CH attachment, the frequency of loose X-15 chromosome association does not show dependence upon the centromere separation of the No. 15s. The nature of the described CH association remains to be clarified; for the present, it seems that the occurrence of a nucleolus organizer on the translocated chromosome 15t presumably plays a role in the causal mechanism of the loose X-15t CH association. Some circumstantial evidence indicates the adverse effect of the observed tight CH attachment on the course of spermatogenesis.

34. Allelic profile at 37 biochemical loci of two inbred strains of the house mouse derived from wild Mus musculus musculus

Abstract

Two newly established inbred strains derived from Mus musculus musculus, designated PWD/Ph (F29) and PWK/Ph (F33), were examined for their alleles at 37 biochemical loci located on 12 different chromosomes. The allelic pattern showed characteristic differences from those observed in common inbred strains. The genetic distance Dbetween PWK/Ph and PWD/Ph was 0·027, whereas the corresponding values for the genetic distances between PWK/Ph and C57BL/6J, DBA/2J, BALB/cJ and SWR/J were 0·777, 0·721, 0·721 and 0·838 respectively. New allozymes are described as being controlled by the loci Es-23, Pre-2and Tam-1. The genetic relationship to M.m. molossinusis indicated by identical alleles at six other loci (Es-2, Es-9, Es-10, Es-11, Es-18and Es-22).

Abstract

Ubiquitination is a universal protein degradation pathway in which the molecules of 8.5-kDa proteolytic peptide ubiquitin are covalently attached to the epsilon-amino group of the substrate's lysine residues. Little is known about the importance of this highly conserved mechanism for protein recycling in mammalian gametogenesis and fertilization. The data obtained by the students and faculty of the international training course Window to the Zygote 2000 demonstrate the accumulation of ubiquitin-cross-reactive structures in the trophoblast, but not in the inner cell mass of the expanding bovine and mouse blastocysts. This observation suggests that a major burst of ubiquitin-dependent proteolysis occurs in the trophoblast of mammalian peri-implantation embryos. This event may be important for the success of blastocyst hatching, differentiation of embryonic stem cells into soma and germ line, and/or implantation in both naturally conceived and reconstructed mammalian embryos.
