Ex situ threatened species management has both conservation and welfare objectives and these objectives often align, but can diverge. Areas of agreement can present win‐wins for achieving welfare and conservation objectives, while identifying areas of divergence is important to ensure management strategies achieve balance across objectives. We examined welfare and conservation objectives in the ex situ population of Extinct in the Wild sihek (Guam kingfisher, Todiramphus cinnamominus) by quantifying mortality rates, determining sex‐ and age‐specific causes of mortality and identifying associated welfare domains, as well as quantifying sex‐ and age‐specific differences in reproductive value and contributions to variation in population growth rate (λ). Females had significantly higher mortality rates than males, potentially impacting population viability and suggesting females may be more vulnerable to experiencing lower welfare than males. Mitigating causes of female mortality would therefore present a clear win‐win for both welfare and conservation objectives. Both causes of mortality and contributions to variation in λ were found to differ across sex‐ and age‐classes. In particular, nutritional and metabolic diseases tended to impact younger age‐classes and these age‐classes had large contributions to variation in λ. Mitigation of these diseases could therefore also present a win‐win for welfare and conservation objectives. However, we also identified a potential divergence between objectives: a major cause of female mortality was reproductive disease with older aged females primarily affected, but older aged females contributed little to variation in λ and had low reproductive value. Developing mitigation strategies for reproductive disease could therefore aid welfare objectives but have little benefit for conservation objectives, suggesting careful balancing across objectives is required. Our results highlight the need to explicitly consider conservation and welfare objectives in threatened species management, in particular in the context of an increasing conservation need for ex situ population management, coupled with increasing social concern for animal welfare. Receptor cells isolated rat fungiform taste buds exposed to free fatty acids (FFAs) were analyzed by patch-clamp recordings to determine the effects of FFAs on chemosensory cues. Patch-clamp analysis of rodent taste receptor cells indicated the role of FFAs generated by lingual lipase in attenuating delayed-rectifying K+ channels. Furthermore, G proteins were not required in FFA-induced inhibition of the delayed-rectifying K+ channel in rodent taste receptor cells.

Crop improvement programs need to be evaluated periodically to ensure current breeding and selection methods are operating optimally. Although such analyses have customarily been performed using data collected from designed experiments with preplanned hypotheses (a priori statistical approaches), data accumulated over several years during routine crop improvement activities represent a rich repository for exploratory research. Post-hoc analyses of data retrieved from a well maintained database are particularly adept at discovering patterns and/or relationships between subgroups of sampled populations that would otherwise remain undetected and undiscovered when relying solely upon a priori statistical approaches. Data from several selection cycles are available from the Louisiana State University Agricultural Center Sugarcane Improvement Program to permit an appraisal of its breeding and selection practices. This paper reviews the decision making processes and selection strategies that occur during parental selection and crossing. Exploratory analyses of the data were performed to detect patterns that might have resulted from crop improvement activities. Each stage and/or decision making process was analyzed retrospectively to assess the effectiveness and efficiency with which progeny progressed to advanced stages of the program. Also documented, were the rates at which, and, the characteristics for which previously desirable entries were eliminated in subsequent stages of the program. Research was conducted to evaluate response of sugarcane when grown in competition with bermudagrass. Two experiments were conducted, a density experiment where a single cultivar was planted with bermudagrass at four densities and a cultivar experiment where six sugarcane cultivars were planted with bermudagrass at two densities. In both experiments a Randomized Complete Block design with four replications was used and experiments were repeated twice. Single node stolon segments of bermudagrass and single node cuttings of sugarcane were pre-sprouted for 10 days and then transplanted into 26.5 L pots with a surface area of 0. 093m². Sugarcane and bermudagrass were allowed to compete in pots for 56 days. For the density experiment, 0, 1, 2, or 4 bermudagrass plants were grown with HoCP96-540 sugarcane. Sugarcane growth determined by longest leaf length was not affected when sugarcane was grown with 1 or 2 bermudagrass plants. With 4 bermudagrass plants, sugarcane leaf length was reduced 12% and dewlap height was not affected. Sugarcane shoot number was reduced at least 44% when in competition with bermudagrass. Sugarcane shoot weight was reduced by 51% when in competition with bermudagrass. Sugarcane root weight was not affected when grown with 1 bermudagrass plant. With 2 and 4 bermudagrass plants, sugarcane root weight was reduced 36 and 41%, respectively. In the cultivar experiment, 0 or 2 bermudagrass plants were grown with HoCP 96-540, L 97-128, L 99-226, HoCP 00-950, L 01-283, and L 03-371. Based on leaf length, dewlap height, and shoot number, bermudagrass was most competitive with HoCP 96-540 and L 01-283 and least competitive with L 99-226 and L 97-128. Number of shoots produced for L 99-226, L 97-128, and L 03-371 were twice that for HoCP 00-950 and L 01-283. Sugarcane shoot weight for L 99-226 and L 97-128 was equivalent and averaged 2.8 times that for L 01-283 and HoCP 96-540. Sugarcane root weight for L 99-226 and L 97-128 was equivalent and averaged 1.9 times that for HoCP 96-950, L 03-371, L 01-283 and HoCP 96-540. Averaged across cultivars, two bermudagrass plants per pot reduced sugarcane shoot weight 17% and sugarcane root weight was reduced 14%. [ABSTRACT FROM AUTHOR]

Background: Spontaneous hemorrhage from erosion of varicose veins through the skin is a serious and occasionally fatal complication of varicose vein disease. Various treatments, both acute and delayed, have been advocated. Our two-step clinical protocol was designed to prevent further hemorrhage without delay and to provide durable freedom from recurrent bleeding., Methods: All patients referred to our surgical vein practice with hemorrhage from varicose veins were entered into a prospective registry. On presentation, all patients underwent diagnostic duplex ultrasound for venous reflux. Immediate treatment consisted of ultrasound-guided polidocanol/CO 2 foam sclerotherapy of the bleeding varicosity and adjacent veins. Subsequent endovenous ablation of underlying incompetent axial veins, with concurrent microphlebectomy when indicated, was scheduled and performed within 8 weeks., Results: Fifty-nine patients were referred with recent hemorrhage from varicose veins over a period of 4 years. Fifty-six (95%) had an ultrasound-identified tributary varicosity underlying the point of bleeding, and three had a skin-surface erosion only and no ultrasound-identified underlying tributary. Of the 59 study patients, 52 underwent prompt polidocanol/CO 2 foam sclerotherapy, targeted to both the underlying tributary (when present) as well as the cutaneous bleeding varicosity. The remaining seven either declined sclerotherapy or were ineligible. Underlying incompetence of axial (great, small, or anterior saphenous) veins was found in 54 patients (92%). Of these patients with truncal vein incompetence, 48 underwent ablation-5 declined and 1 was too frail for any further intervention. There were no recurrent bleeds in the interval between immediate sclerotherapy and scheduled ablation. During a mean follow-up of 2.2 years, 55 of the 59 patients (93%) had no recurrence of bleeding. Four patients (7%) had late, recurrent hemorrhage: one had failed to return for their scheduled ablation, two were on chronic anticoagulation, and one had severe right heart failure. All four were retreated without further recurrence., Conclusions: A two-step protocol of immediate ultrasound-guided foam sclerotherapy, followed within 8 weeks by endovenous ablation of incompetent axial veins and concurrent microphlebectomy, provided rapid and efficient treatment with durable freedom from subsequent hemorrhage., Competing Interests: Disclosures None., (Copyright © 2024 The Author(s). Introduction: An arteriovenous fistulae for dialysis access is only functional if superficial enough for reliable cannulation. We describe a previously unreported technique to superficialize arteriovenous fistulae too deep for reliable cannulation., Method: The path of the fistula is marked using ultrasound, and three to four transverse incisions are made along this path. The subcutaneous tissue is separated from the dermis over this entire area. The fistula is then accessed through transverse incisions and by dividing the overlying adipose tissue. The mobilized vein is then "trapped" directly under the dermis by closing the superficial fascia and adipose tissue beneath it and using this tissue to elevate the fistula. No anastomotic revisions are necessary, and moderate straightening can be performed., Results: We have performed this procedure on 20 patients with 100% technical success. One fistula had thrombosis within 8-10 weeks of the superficialization procedure and one patient refused access due to pain, but all other fistulas were successfully cannulated. Two patients were lost to follow-up, and one patient developed skin breakdown in the area of the flap which delayed cannulation. Our 3-month patency was 94.4% with a functional patency, defined as a clinically patent fistula successfully being used for hemodialysis, of 87.5%., Conclusion: The technique described allows elevation of the vein to the level of the dermis without division and re-anastomosis or re-tunneling, through several small incisions maintaining virgin skin and normal contour for easier cannulation.

We evaluated the effectiveness of Anvirzel™, an aqueous extract of Nerium oleander on HIV infection of human peripheral blood mononuclear cells. Oleandrin, the principle cardiac glycoside (CG) in Anvirzel™ has been shown to exhibit anti-cancer properties but its efficacy against HIV is unknown. Treatment with Anvirzel™ significantly reduced the infectivity of virus produced from infected cells without any change in the total amount of virus produced. This is in contrast to treatment with AZT, a potent inhibitor of HIV replication that has been shown to significantly reduce virus production. Relative to untreated cultures, virus in cultures treated with oleandrin had significantly reduced expression of the envelope protein gp120, the sole determinant of virus infectivity, suggesting a novel mechanism underlying the impaired infectivity. These results support the potential utility of the Nerium oleander aqueous extract, containing the CG oleandrin as a novel candidate anti-HIV therapeutic., (Copyright © 2012 Elsevier B.V. Two adult female bontebok (Damaliscus pygarus dorcas) were euthanized because of signs of pneumonia and weakness (case 1), and a nonresponsive lameness with draining fistula (case 2). Necropsy findings were similar in both cases and consisted of disseminated granulomatous lesions in the liver, kidneys, spleen, lungs, pleural surfaces, and multiple lymph nodes. Mycobacterium kansasii was isolated from both cases after multiple attempts on a variety of samples by two laboratories. The remaining four animals in the herd were tested for antibody responses using the Chembio ElephantTB STAT-PAK, DPP VetTB kits, and multi-antigen print immunoassay (MAPIA), for immune reaction using the intradermal tuberculin test, and by tracheal wash cultures, and thoracic radiographs. Banked serum samples collected in 2005 and obtained from the original institution, revealed 1/9 (11.11%) seropositive animals using the three immunoassays. Retesting the current herd in 2008 showed 2/6 (33.33%) seropositive animals by the three tests, with MAPIA demonstrating antibody reactivity to MPB83 and MPB70 proteins. Inconsistent intradermal tuberculin test results, cross-reactivity in serologic assays designed for tuberculosis detection, difficulty in obtaining definitive identification by culture, and inability to identify a source of infection created challenges in distinguishing the atypical mycobacteriosis due to M. kansasii from the initially suspected tuberculous infection in this herd. Owing to regulatory considerations, differences in host-to-host transmission, and source of infection between Mycobacterium tuberculosis complex and nontuberculous mycobacteria, correct diagnosis is crucial for management of these diseases in wildlife species.

To reduce male-male aggression in collared lemurs (Eulemur collaris) at Disney's Animal Kingdom, a clinical trial of leuprolide acetate (Depo-Lupron) injections through one breeding season (24 wk), followed by surgical castration after the breeding season was implemented. Daily behavioral observations to record occurrences of aggression were conducted before and during the injection period, as well as after castration. Fecal sample collection began 2 wk before the first injection and continued throughout the clinical trial to determine testosterone metabolite concentration. Samples were collected 3 times per week. Fecal collection and behavioral observation continued for 5 mo after castration. Fecal samples from 3 intact males from other institutions were obtained to compare testosterone values. While the lemurs were treated with Depo-Lupron, testosterone concentrations spiked immediately after injection, and then returned to pretreatment, nonbreeding-season levels after varying lengths of time. Analysis of the behavioral data indicated that aggression was not significantly reduced with Depo-Lupron treatment. However, after castration, contact aggression and wounding decreased to zero. Although the sample is small, this study presents the first documented data on the effects that chemical treatment with Depo-Lupron and surgical castration have upon hormone levels and aggressive behavior in eulemurs.

A combination of low serum calcium (Ca), high serum phosphorus (P), and low serum magnesium (Mg) has been observed in individual captive ruminants, primarily affecting kudu (Tragelaphus strepsiceros), eland (Taurotragus oryx), nyala (Tragelaphus angasii), bongo (Tragelaphus eurycerus), and giraffe (Giraffa camelopardalis). These mineral abnormalities have been associated with chronic laminitis, acute tetany, seizures, and death. Underlying rumen disease secondary to feeding highly fermentable carbohydrates was suspected to be contributing to the mineral deficiencies, and diet changes that decreased the amount of starch fed were implemented in 2003. Serum chemistry values from before and after the diet change were compared. The most notable improvement after the diet change was a decrease in mean serum P. Statistically significant decreases in mean serum P were observed for the kudu (102.1-66.4 ppm), eland (73.3-58.4 ppm), and bongo (92.1-64.2 ppm; P < 0.05). Although not statistically significant, mean serum P levels also decreased for nyala (99.3-86.8 ppm) and giraffe (82.6-68.7 ppm). Significant increases in mean serum Mg were also observed for kudu (15.9-17.9 ppm) and eland (17.1-19.7 ppm). A trend toward increased serum Mg was also observed in nyala, bongo, and giraffe after the diet change. No significant changes in mean serum Ca were observed in any of the five species evaluated, and Ca was within normal ranges for domestic ruminants. The mean Ca:P ratio increased to greater than one in every species after the diet change, with kudu, eland, and bongo showing a statistically significant change. The results of this study indicate that the diet change had a generally positive effect on serum P and Mg levels.

Mucosal dendritic cells have been implicated in the capture, storage, and transmission of HIV to CD4(+) T cells as well as in the promotion of HIV replication in activated CD4(+) T cells during the cognate T-cell and DC interaction. We report that HIV induces human genital mucosal epithelial cells to produce thymic stromal lymphopoietin (TSLP) via activation of the NFkappaB signaling pathway. The TSLP secreted by HIV exposed epithelial cells activated DC, which promoted proliferation and HIV-1 replication of co-cultured autologous CD4(+) T cells. In rhesus macaques, we observed dramatic increases in TSLP expression concurrent with an increase in viral replication in the vaginal tissues within the first 2 weeks after vaginal SIV exposure. These data suggest that HIV-mediated TSLP production by mucosal epithelial cells is a critical trigger for DC-mediated amplification of HIV-infection in activated CD4(+) T cells. The cross talk between mucosal epithelial cells and DC, mediated by HIV-induced TSLP, may be an important mechanism for the high rate of HIV infection in women through the vaginal mucosa.

Human immunodeficiency virus type 1 (HIV-1) infection is initiated by the binding of the viral envelope protein gp120 to the host cell CD4 receptor through a high-affinity interaction involving amino acids 39-60 within the CD4. We obtained evidence demonstrating functional importance of this region in CD4 for viral infectivity by showing that a synthetic peptide corresponding to this CD4 sequence exhibited competitive binding to gp120 and significantly reduced infection by diverse HIV-1 strains, including primary isolates. Treatment of HIV-1-infected cells with this CD4 peptide induced shedding of gp120 and exposure of the transmembrane protein gp41. Furthermore, we observed that deletion or substitution of arginine at position 59 (Arg(59)) within the CD4 peptide sequence abrogated its gp120-shedding property. These results indicate a critical role for Arg(59) in the CD4 for conformational changes in gp120 during the sequential process of entry and infection by HIV-1.

Dilated cardiomyopathy is a relatively common pathology in captive flying foxes (Pteropus spp.). The goal of this study was to establish quantitative reference range measurements that could be used to support a diagnosis of cardiac disease in these animals. Lateral and ventrodorsal thoracic radiographs from apparently healthy flying foxes (n = 66) of three species (Rodriguez island flying fox, P. rodricensis, n = 18; small island flying fox, P. hypomelanus, n = 16; and Malaysian flying fox, P. vampyrus, n = 32) were evaluated objectively to describe the cardiac appearance. Absolute and relative cardiac dimensions also were measured. The same methods were used to evaluate radiographs from flying foxes (n = 9) with known dilated or acute cardiomyopathy. The following ratios were most appropriate for categorizing normal cardiac silhouette size. In the ventrodorsal projection, heart width to thoracic width and heart width to clavicle length were the preferred measurements. In the lateral projection, heart width compared with thoracic height was the preferred measurement. From radiographs of the bats with known dilated and acute cardiomyopathy, the apicobasilar heart length compared with thoracic height and heart width compared with thoracic height on lateral films were the most sensitive ratios for diagnosing cardiomegaly.

Six adult female tigers (Panthera tigris) were anesthetized repeatedly for elective medical procedures using 3 mg medetomidine and 200 mg ketamine i.m. Inductions were rapid and smooth, although supplemental ketamine was needed for safe transport after induction in 6 of 17 procedures. Reversal of the medetomidine-induced sedation with 15 mg atipamezole i.m. 59-232 min after induction resulted in smooth, rapid recoveries.

To determine whether mitral valve annulus displacement (MVAD) can be used to assess septal contractility in patients with paradoxical septal motion, we assessed four atrioventricular regions (septum, lateral wall, anterior wall, and inferior wall) by MVAD in 80 consecutive patients. The patients were divided into five groups: group 1 (control) (n = 20), normal left ventricular (LV) systolic function; group 2 (n = 15), paradoxical septal motion resulting from left bundle branch block (LBBB) and normal segmental and global LV systolic function; group 3 (n = 19), paradoxical septal motion as a result of cardiac surgery, and normal segmental and global LV systolic function; group 4 (n = 11), paradoxical septal motion resulting from LBBB, dilated cardiomyopathy, and severely depressed LV systolic function; group 5 (n = 15), septal hypokinesis with either normal or mildly depressed global LV systolic function. In groups 1, 2, and 3, 80% to 100% of patients had septal and other regional MVAD > or = 1.0 cm. The average MVAD in group 4 (dilated cardiomyopathy), was significantly decreased ( < or = 0.8 cm) in all four regions (p < 0.01 compared with groups 1, 2, and 3). In group 5 (septal hypokinesis), the septal MVAD was > or = 1.0 cm in only 13% of the patients (p < 0.025 compared with groups 1, 2, and 3). In conclusion, patients with paradoxical septal motion caused by LBBB or cardiac surgery have preserved septal contractility when evaluated by MVAD.

Living-related liver transplantation is becoming more commonplace worldwide in the treatment of end-stage liver disease in the pediatric age group. Our LRD experience has resulted in patient and graft survival rates comparable to our cadaveric donor recipients. The incidence and severity of acute rejection episodes were similar. This differs from the clear immunologic advantage of living-related donation in kidney transplantation. It may, however, reflect the relatively small numbers in our LRD group. Overall, however, the technical complications are manageable with early intervention, yielding acceptable results.

1. Brief planned and crisis admissions to an inpatient psychiatric unit are presented as a component supportive of outpatient care and case management for chronically mentally ill patients. 2. Typical patients admitted to this inpatient short-term program are those experiencing a life stress or temporary crisis; chronically mentally ill patients who cycle and experience exacerbations of their illness; and those who need to make the transition from an acute inpatient unit to community living. 3. The Brief Admission Program treats the individual's response to and the consequences of a lifelong illness and enables the patient to return to his previous level of functioning and continue necessary outpatient treatment.

Usher syndrome is an autosomal recessive disease characterized by dual sensory impairments; affected individuals are born with a sensorineural hearing loss and ultimately lose their sight as retinitis pigmentosa develops. Conventional protein markers previously tested in a Louisiana Acadian kindred suggested tentative linkage to vitamin D-binding protein on chromosome 4. DNA linkage studies do not confirm this linkage relationship and exclude much of chromosome 4 as the site of the Usher syndrome gene in these families.
