Abstract

The aim of this study was to investigate on the prevalence of L. infantum infection in foxes living in two distinct areas of southern and central Italy. Sixty eight carcasses of foxes were subjected to PCR analysis. At external examination no clinical signs of leishmaniasis were recorded. Ten foxes were found to be positive by Leishmania nested-PCR, of which 4 in spleen, 2 in lymph nodes and 4 in both organs. Our results show that in southern and central Italy foxes may me subclinically infected by Leishmania, but their importance as reservoirs for the maintenance of the infection remains to be determined.

Abstract

The Mt. Vesuvius area of the Campania region (southern Italy) is an endemic zone of canine Leishmaniasis (CanL). Phlebotomus perniciosus is the main vector involved in the transmission of Leishmania infantum to susceptible hosts. Noteworthy, the Mt. Vesuvius area has been considered a stable focus of intense L. infantum zoonotic transmission. Geographical information systems (GIS) are very useful tools for mapping and monitoring the distribution of CanL and its vectors in the area in relation to environmental features. A previous entomological study conducted in the area showed a different density of P. perniciosus among the two sides of the Mt. Vesuvius. Indeed, higher densities of P. perniciosus were found on the coastal side (density = 5.8) than on the Apennine side (density = 1.4).The predominance of green vegetated environments (forest, semi-­‐natural and agricultural areas) in the coastal side, in contrast with the predominance of artificial surfaces (namely urban environment) in the Apennine side, was considered responsible for the different P. perniciosus densities between the two surveyed areas. Therefore, a serological survey was conducted using GIS in order to map the distribution of CanL along the coastal and the Apennine sides of the Mt. Vesuvius area. Sera from 505 autochthonous owned dogs were examined by IFAT to detect antibodies to L. infantum. A titre of at least 1:80 was considered a positive result. Out of the 505 dogs examined, 213 (42.2%; 95%Confidence Interval = 37.9 -­‐ 46.6%) were found to have antibodies to L. infantum. The prevalence was not different between the two sides (42.6% in the Apennine side and 41.6% in the coastal side). The findings of the present study showed that even low densities of P. perniciosus, especially when associated with an urban environment, seem sufficient to ensure Leishmania transmission among susceptible hosts. In fact, no significant differences were found in CanL seroprevalence of the two areas. Comprehensive control programs, including the use of vaccine, are discussed to prevent CanL in such endemic areas as the Mt. Vesuvius.

Abstract

Lysozyme is an antibacterial enzyme that is found in all body fluids except cerebrospinal fluid, sweat and urine. The purpose of this study was to investigate enzyme activity in the colostrum and serum of Mediterranean buffaloes and their newborn calves in the first hours after calving, in order to gain information on the passive immunological status of neonatal buffalo calves. In addition, the correlation between levels of lysozyme and IgG in the serum of the newborn buffalo calves was investigated, in order to evaluate whether this enzyme could be useful as a marker of maternally derived γ-globulin intake. Eight multiparous healthy buffaloes, five to seven years of age, living with their calves in paddocks in the same herd in southern Italy, were studied. The activity of lysozyme was determined using an assay based on the classic lysoplate method of Osserman and Lawlor. The results demonstrate that the activity of lysozyme in the serum of newborn buffalo calves gradually increases during the first 60 hours after birth, and that the activity of lysozyme in colostrum is higher in buffaloes than in cows, suggesting that there may be a critical non-specific defence role for this enzyme in Mediterranean buffaloes, as has been reported for Indian buffaloes. Furthermore, the lysozyme activity in the serum of newborn buffalo calves may originate from gradual absorption following colostral intake and/or from early endogenous synthesis in the first hours of life. Finally, the results show that the serum lysozyme activity of newborn buffalo calves is not useful as a marker of passive IgG intake.

Abstract

A longitudinal study was carried out on kennelled stray dogs in a canine leishmaniasis (CanL) endemic area, to evaluate early and late diagnostic performance of a non-invasive conjunctival swab (CS) nested (n)-PCR analysis for Leishmania detection in 2 cohorts of dogs, respectively. (A) Sixty-five IFAT- and CS n-PCR-negative dogs exposed to, and followed up once or twice a month during a full sand fly season (July–November 2008). In parallel, a sand fly survey was performed on site using standard sticky traps set twice a month, for a cumulative surface of 63m2. (B) Seventeen IFAT- and CS n-PCR-negative dogs found positive in July 2008 at the peripheral blood buffy-coat (BC) n-PCR. These dogs were examined again by BC n-PCR in September and November 2008, and before the subsequent transmission season (May 2009) along with CS n-PCR and IFAT. None of the cohort (A) dogs converted to positive CS n-PCR during the transmission season. Although ∼2500 phlebotomine specimens were collected with peaks of 100–147 specimens/m2 sticky trap, the cumulative density of the only proven CanL vector in the area (Phlebotomus perniciosus) was found to be very low (0.5/m2). All cohort (B) dogs remained substantially seronegative; BC n-PCR showed an intermittent positive trend during the period surveyed, resulting in 82% conversions to negative by the end of the study, in contrast with 71% conversions to positive at the CS n-PCR analysis. In conclusion, while CS n-PCR was not found effective for the early detection of Leishmania contacts in dogs exposed to a low pressure of vectorial transmission, this assay showed to slowly convert to positive in a high rate of dogs, in the absence of seroconversion. CS n-PCR technique can be a suitable marker for assessing Leishmania exposure in dogs as a non-invasive alternative to current serological and molecular tools.
