Using simultaneous, in situ optical spectroscopy and time-resolved, small-angle X-ray scattering (SAXS), we have directly monitored the seeded growth of nearly monodisperse gold nanorods using hydroquinone as the reductant. Growth of the rods is much slower than with the ascorbate ion, allowing the rate of growth along both the longitudinal and transverse directions to be independently determined. The thickness of the stabilizing CTAB layer (3.2 ± 0.3 nm) has also been extracted. We find that increasing the hydrogen tetrachloroaurate(III) concentration produces longer rods, while conversely, increasing the hydroquinone concentration reduces the final aspect ratio. The final number of gold rods is smaller than the initial number of seed particles and decreases in the presence of larger concentrations of HAuCl4. The SAXS data reveal an early transition from a spherical morphology to an ellipsoidal one and then to spherically capped cylinders. The growth curve exhibits at least three distinct regimes: an initial phase comprising spherical seed growth, followed by symmetry breaking and slow elongation. A third phase is marked by rapid rod growth and increases in the aspect ratio. This process is temporally well resolved from the initial symmetry breaking but typically occurs when the rods are around 6 nm in diameter using hydroquinone as the reductant. The results provide qualitative support for the “popcorn model” proposed by Edgar et al. [ Formation of Gold Nanorods by a Stochastic “Popcorn” Mechanism. ACS Nano 2012, 6, 1116 1125 ].

Neuropilin-2 (NRP2) is a prognostic indicator for reduced survival in bladder cancer (BCa) patients. Together with its major ligand, vascular endothelial growth factor (VEGF)-C, NRP2 expression is a predictive factor for treatment outcome in response to radiochemotherapy in BCa patients who underwent transurethral resection. Therefore, we investigated the benefit of combining cisplatin-based chemotherapy with irradiation treatment in the BCa cell line RT112 exhibiting or lacking endogenous NRP2 expression in order to evaluate NRP2 as potential therapeutic target. We have identified a high correlation of NRP2 and the Glioma-associated oncogene family zinc finger 2 (GLI2) transcripts in the cancer genome atlas (TCGA) cohort of BCa patients and a panel of 15 human BCa cell lines. Furthermore, we used in vitro BCa models to show the transforming growth factor-beta 1 (TGFb1)-dependent regulation of NRP2 and GLI2 expression levels. Since NRP2 was shown to bind TGFb1, associate with TGFb receptors and enhance TGFb1 signaling, we evaluated downstream signaling pathways using an epithelial to mesenchymal (EMT)-assay in combination with a PCR profiling array containing 84 genes related to EMT. Subsequent target validation in NRP2 knockout and knockdown models revealed secreted phosphoprotein 1 (SPP1/OPN/Osteopontin) as a downstream target positively regulated by NRP2.

In biological fluids, proteins bind to particles, forming so-called protein coronas. Such adsorbed protein layers significantly influence the biological interactions of particles, both in vitro and in vivo. The adsorbed protein layer is generally described as a two-component system comprising "hard" and "soft" protein coronas. However, a comprehensive picture regarding the protein corona structure is lacking. Herein, we introduce an experimental approach that allows for in situ monitoring of protein adsorption onto silica microparticles. The technique, which mimics flow in vascularized tumors, combines confocal laser scanning microscopy with microfluidics and allows the study of the time-evolution of protein corona formation. Our results show that protein corona formation is kinetically divided into three different phases: phase 1, proteins irreversibly and directly bound (under physiologically relevant conditions) to the particle surface; phase 2, irreversibly bound proteins interacting with preadsorbed proteins, and phase 3, reversibly bound "soft" protein corona proteins. Additionally, we investigate particle-protein interactions on low-fouling zwitterionic-coated particles where the adsorption of irreversibly bound proteins does not occur, and on such particles, only a "soft" protein corona is formed. The reported approach offers the potential to define new state-of-the art procedures for kinetics and protein fouling experiments.

INTRODUCTION: White matter hyperintensity (WMH) volume on MRI is increased among presymptomatic individuals with autosomal dominant mutations for Alzheimer's disease (AD). One potential explanation is that WMH, conventionally considered a marker of cerebrovascular disease, are a reflection of cerebral amyloid angiopathy (CAA) and that increased WMH in this population is a manifestation of this vascular form of primary AD pathology. We examined whether the presence of cerebral microbleeds, a marker of CAA, mediates the relationship between WMH and estimated symptom onset in individuals with and without autosomal dominant mutations for AD. PARTICIPANTS AND METHODS: Participants (n = 175, mean age = 41.1 years) included 112 with an AD mutation and 63 first-degree non-carrier controls. We calculated the estimated years from expected symptom onset (EYO) and analyzed baseline MRI data for WMH volume and presence of cerebral microbleeds. Mixed effects regression and tests of mediation were used to examine microbleed and WMH differences between carriers and non-carriers and to test the whether the association between WMH and mutation status is dependent on the presence of microbleeds. RESULTS: Mutation carriers were more likely to have microbleeds than non-carriers (p<0.05) and individuals with microbleeds had higher WMH volume than those without (p<0.05). Total WMH volume was increased in mutation carriers compared with non-carriers, up to 20 years prior to EYO, after controlling for microbleed status, as we demonstrated previously. Formal testing of mediation demonstrated that 21% of the association between mutation status and WMH was mediated by presence of microbleeds (p = 0.03) but a significant direct effect of WMH remained (p = 0.02) after controlling for presence of microbleeds. DISCUSSION: Although there is some co-dependency between WMH and microbleeds, the observed increases in WMH among mutation carriers does not appear to be fully mediated by this marker of CAA

The formation of a biomolecular corona around engineered particles determines, in large part, their biological behavior in vitro and in vivo. To gain a fundamental understanding of how particle design and the biological milieu influence the formation of the "hard" biomolecular corona, we conduct a series of in vitro studies using microfluidics. This setup allows the generation of a dynamic incubation environment with precise control over the applied flow rate, stream orientation, and channel dimensions, thus allowing accurate control of the fluid flow and the shear applied to the proteins and particles. We used mesoporous silica particles, poly(2-methacryloyloxyethylphosphorylcholine) (PMPC)-coated silica hybrid particles, and PMPC replica particles (obtained by removal of the silica particle templates), representing high-, intermediate-, and low-fouling particle systems, respectively. The protein source used in the experiments was either human serum or human full blood. The effects of flow, particle surface properties, incubation medium, and incubation time on the formation of the biomolecular corona formation are examined. Our data show that protein adhesion on particles is enhanced after incubation in human blood compared to human serum and that dynamic incubation leads to a more complex corona. By varying the incubation time from 2 s to 15 min, we demonstrate that the "hard" biomolecular corona is kinetically subdivided into two phases comprising a tightly bound layer of proteins interacting directly with the particle surface and a loosely associated protein layer. Understanding the influence of particle design parameters and biological factors on the corona composition, as well as its dynamic assembly, may facilitate more accurate prediction of corona formation and therefore assist in the design of advanced drug delivery vehicles.

Imaging spectrometry of non-oceanic aquatic ecosystems has been in development since the late 1980s when the first airborne hyperspectral sensors were deployed over lakes. Most water quality management applications were, however, developed using multispectral mid-spatial resolution satellites or coarse spatial resolution ocean colour satellites till now. This situation is about to change with a suite of upcoming imaging spectrometers being deployed from experimental satellites or from the International Space Station. We review the science of developing applications for inland and coastal aquatic ecosystems that often are a mixture of optically shallow and optically deep waters, with gradients of clear to turbid and oligotrophic to hypertrophic productive waters and with varying bottom visibility with and without macrophytes, macro-algae, benthic micro-algae or corals. As the spaceborne, airborne and in situ optical sensors become increasingly available and appropriate for aquatic ecosystem detection, monitoring and assessment, the science-based applications will need to be further developed to an operational level. The Earth Observation-derived information products will range from more accurate estimates of turbidity and transparency measures, chlorophyll, suspended matter and coloured dissolved organic matter concentration, to more sophisticated products such as particle size distributions, phytoplankton functional types or distinguishing sources of suspended and coloured dissolved matter, estimating water depth and mapping types of heterogeneous substrates. We provide an overview of past science, current state of the art and future directions so that early career scientists as well as aquatic ecosystem managers and associated industry groups may be prepared for the imminent deluge of imaging spectrometry data.

Modelers can improve a model by addressing the causes for the model errors (data errors and structural errors). This leads to implementing model enhancements (MEs), for example, meteorological data based on more monitoring stations, improved calibration data, and/or modifications in process formulations. However, deciding on which MEs to implement remains a matter of expert knowledge. After implementing multiple MEs, any improvement in model performance is not easily attributed, especially when considering different objectives or aspects of this improvement (e.g., better dynamics vs. reduced bias). We present an approach for comparing the effect of multiple MEs based on real observations and considering multiple objectives (MMEMO). A stepwise selection approach and structured plots help to address the multidimensionality of the problem. Tailored analyses allow a differentiated view on the effect of MEs and their interactions. MMEMO is applied to a case study employing the mesoscale hydro‐sedimentological model WASA‐SED for the Mediterranean‐mountainous Isábena catchment, northeast Spain. The investigated seven MEs show diverse effects: some MEs (e.g., rainfall data) cause improvements for most objectives, while other MEs (e.g., land use data) only affect a few objectives or even decrease model performance. Interaction of MEs was observed for roughly half of the MEs, confirming the need to address them in the analysis. Calibration and increasing the temporal resolution showed by far stronger impact than any of the other MEs. The proposed framework can be adopted in other studies to analyze the effect of MEs and, thus, facilitate the identification and implementation of the most promising MEs for comparable cases.

Amyloid imaging plays an important role in the research and diagnosis of dementing disorders. Substantial variation in quantitative methods to measure brain amyloid burden exists in the field. The aim of this work is to investigate the impact of methodological variations to the quantification of amyloid burden using data from the Dominantly Inherited Alzheimer's Network (DIAN), an autosomal dominant Alzheimer's disease population. Cross-sectional and longitudinal [11C]-Pittsburgh Compound B (PiB) PET imaging data from the DIAN study were analyzed. Four candidate reference regions were investigated for estimation of brain amyloid burden. A regional spread function based technique was also investigated for the correction of partial volume effects. Cerebellar cortex, brain-stem, and white matter regions all had stable tracer retention during the course of disease. Partial volume correction consistently improves sensitivity to group differences and longitudinal changes over time. White matter referencing improved statistical power in the detecting longitudinal changes in relative tracer retention; however, the reason for this improvement is unclear and requires further investigation. Full dynamic acquisition and kinetic modeling improved statistical power although it may add cost and time. Several technical variations to amyloid burden quantification were examined in this study. Partial volume correction emerged as the strategy that most consistently improved statistical power for the detection of both longitudinal changes and across-group differences. For the autosomal dominant Alzheimer's disease population with PiB imaging, utilizing brainstem as a reference region with partial volume correction may be optimal for current interventional trials. Further investigation of technical issues in quantitative amyloid imaging in different study populations using different amyloid imaging tracers is warranted.

INTRODUCTION: Multiple gene expression based prognostic biomarkers have been repeatedly identified in gastric carcinoma. However, without confirmation in an independent validation study, their clinical utility is limited. Our goal was to establish a robust database enabling the swift validation of previous and future gastric cancer survival biomarker candidates. RESULTS: The entire database incorporates 1,065 gastric carcinoma samples, gene expression data. Out of 29 established markers, higher expression of BECN1 (HR = 0.68, p = 1.5E-05), CASP3 (HR = 0.5, p = 6E-14), COX2 (HR = 0.72, p = 0.0013), CTGF (HR = 0.72, p = 0.00051), CTNNB1 (HR = 0.47, p = 4.3E-15), MET (HR = 0.63, p = 1.3E-05), and SIRT1 (HR = 0.64, p = 2.2E-07) correlated to longer OS. Higher expression of BIRC5 (HR = 1.45, p = 1E-04), CNTN1 (HR = 1.44, p = 3.5E- 05), EGFR (HR = 1.86, p = 8.5E-11), ERCC1 (HR = 1.36, p = 0.0012), HER2 (HR = 1.41, p = 0.00011), MMP2 (HR = 1.78, p = 2.6E-09), PFKB4 (HR = 1.56, p = 3.2E-07), SPHK1 (HR = 1.61, p = 3.1E-06), SP1 (HR = 1.45, p = 1.6E-05), TIMP1 (HR = 1.92, p = 2.2E- 10) and VEGF (HR = 1.53, p = 5.7E-06) were predictive for poor OS. MATERIALS AND METHODS: We integrated samples of three major cancer research centers (Berlin, Bethesda and Melbourne datasets) and publicly available datasets with available follow-up data to form a single integrated database. Subsequently, we performed a literature search for prognostic markers in gastric carcinomas (PubMed, 2012-2015) and re-validated their findings predicting first progression (FP) and overall survival (OS) using uni- and multivariate Cox proportional hazards regression analysis. CONCLUSIONS: The major advantage of our analysis is that we evaluated all genes in the same set of patients thereby making direct comparison of the markers feasible. The best performing genes include BIRC5, CASP3, CTNNB1, TIMP-1, MMP-2, SIRT, and VEGF.

The fibrillation and subsequent gelation of low molecular hydrogelators is usually triggered by external stimuli. Generally, strong acids are employed to trigger the self-assembly mechanism in pH-responsive supramolecular systems. However, the generation and design of novel stable gels with performing mechanical properties is a challenging task as a result of the uneven self-assembled networks formed. Here, we report the study of the self-assembly process of a low molecular weight hydrogelator (LMWG) in the proximity of its minimum gelation concentration (MGC = 0.3 mg/ml). At such high dilution, the generation of homogeneous gels with good mechanical properties by turning pH by strong acids is a demanding task as a result of the lack of monodisperse 1D self-assembled rod-like aggregates. A microfluidic device is employed here to gradually and homogeneously change the pH. We show that self-assembly of LMWG in well-defined structures can be enhanced by using the diffusive mixing occurring in the microfluidic reactor channel. For very short mixing times, aggregates with 2 nm cross-section are found in the region adjacent to the focused LMWG solution in contact with the low pH buffer solution streams, where the pH reaches values below the pKa of the LMWG and triggers the supramolecular self-assembly. For longer mixing times, aggregates grow in size and occupy homogeneously the micro channel. The results presented here show that better controlled self-assembly can be achieved using microfluidic mixing devices and early stages of self-assembly can be efficiently studied by coupling synchrotron SAXS with microfluidics.

Assessment of the network of toxicity pathways by Omics technologies and bioinformatic data processing paves the road toward a new toxicology for the twenty-first century. Especially, the upstream network of responses, taking place in toxicant-treated cells before a point of no return is reached, is still little explored. We studied the effects of the model neurotoxicant 1-methyl-4-phenylpyridinium (MPP+) by a combined metabolomics (mass spectrometry) and transcriptomics (microarrays and deep sequencing) approach to provide unbiased data on earliest cellular adaptations to stress. Neural precursor cells (LUHMES) were differentiated to homogeneous cultures of fully postmitotic human dopaminergic neurons, and then exposed to the mitochondrial respiratory chain inhibitor MPP+ (5 mu M). At 18-24 h after treatment, intracellular ATP and mitochondrial integrity were still close to control levels, but pronounced transcriptome and metabolome changes were seen. Data on altered glucose flux, depletion of phosphocreatine and oxidative stress (e.g., methionine sulfoxide formation) confirmed the validity of the approach. New findings were related to nuclear paraspeckle depletion, as well as an early activation of branches of the transsulfuration pathway to increase glutathione. Bioinformatic analysis of our data identified the transcription factor ATF-4 as an upstream regulator of early responses. Findings on this signaling pathway and on adaptive increases of glutathione production were confirmed biochemically. Metabolic and transcriptional profiling contributed complementary information on multiple primary and secondary changes that contribute to the cellular response to MPP+. Thus, combined 'Omics' analysis is a new unbiased approach to unravel earliest metabolic changes, whose balance decides on the final cell fate.

We present a versatile approach to produce macroscopic, substrate-supported arrays of plasmonic nanoparticles with well-defined interparticle spacing and a continuous particle size gradient. The arrays thus present a "plasmonic library" of locally noncoupling plasmonic particles of different sizes, which can serve as a platform for future combinatorial screening of size effects. The structures were prepared by substrate assembly of gold-core/poly(N-isopropylacrylamide)-shell particles and subsequent post-modification. Coupling of the localized surface plasmon resonance (LSPR) could be avoided since the polymer shell separates the encapsulated gold cores. To produce a particle array with a broad range of well-defined but laterally distinguishable particle sizes, the substrate was dip-coated in a growth solution, which resulted in an overgrowth of the gold cores controlled by the local exposure time. The kinetics was quantitatively analyzed and found to be diffusion rate controlled, allowing for precise tuning of particle size by adjusting the withdrawal speed. We determined the kinetics of the overgrowth process, investigated the LSPRs along the gradient by UV-vis extinction spectroscopy, and compared the spectroscopic results to the predictions from Mie theory, indicating the absence of local interparticle coupling. We finally discuss potential applications of these substrate-supported plasmonic particle libraries and perspectives toward extending the concept from size to composition variation and screening of plasmonic coupling effects.
