Abstract

The introduction of non-native freshwater fish is a primary cause of aquatic biodiversity loss at global scale. Such introductions have a severe impact on freshwater ecosystems in terms of competition, predation, habitat alteration, genetic pollution and transmission of diseases and parasites. A systematic review was conducted on the helminths parasites of freshwater fish in the context of species introduction and a total of 199 publications were retrieved between 1969 and November 2022. Several scenarios may arise when a new fish species arrive in a recipient area. Non-native fish hosts can co-introduce their parasites without transmitting them to native fish (e.g., the case of North American Centrarchidae and their Monogenea parasites). Another possible outcome is the transfer of these parasites to the native fish fauna (spillover, e.g., the cases of the Nematoda Anguillicola crassus Kuwahara, Niimi & Itagaki, 1974 and the Monogenea Gyrodactylus salaris Malmberg, 1957). Reciprocally, non-native fish hosts may acquire parasites in their new distribution range whether these parasites are native or were previously introduced (e.g., the cases of Ponto-Caspian Gobiidae and the Chinese sleeper Perccottus glenii Dybowski, 1877). Acquired parasites can then be spilled back to the native fauna. This phenomenon is of particular interest when non-native fish hosts influence the dynamics of zoonotic parasites. Abstract

To assess the effect of anthropogenic activities on ecosystems, it is of prime importance to develop new tools enabling a rapid characterization of ecological communities. Freshwater ecosystems are particularly impacted and threatened by human activities and need thorough attention to preserve their biodiversity and the ecological services they provide. Studying such ecosystems is generally difficult because the associated organisms are hard to sample and to monitor. We present a ready-to-use environmental metabarcoding protocol to characterize and monitor the freshwater gastropods communities from water samples. The efficiency of this new tool was compared to a classical malacological survey at 19 sampled sites from 10 distinct rivers distributed over Corsica Island (France). From a single water sample, our eDNA monitoring tool provided a faithful characterization of the local malacofauna compared to the results obtained from the classical malacological survey, with 97.1% of species detection confirmed by both methods. The present tool successfully detected the 11 freshwater snail species previously reported in Corsica by malacological survey but was limited at the genus level for some species. Moreover, our malacological survey allowed an update of the local distribution of a wide diversity of freshwater snails including invasive species (i.e. Potamopyrgus antipodarum and Physa acuta) as well as snail hosts of pathogens of medical and veterinary importance (i.e. Bulinus truncatus and Galba truncatula). These results strengthened a previous hypothesis of an eventual competitive interaction between B. truncatus and P. antipodarum that could limit the endemization of the uro-genital bilharziasis in Corsica.

Abstract

Under ongoing climate changes, the development of large‐scale monitoring tools for assessing the risk of disease emergence constitutes an urging challenge. This is particularly the case for snail‐borne diseases such as the urogenital bilharziasis that emerged in Corsica and threat European countries. The expansion of this tropical disease mainly relies on the local presence of competent snail hosts such as Bulinus truncatus. Unfortunately, very little is known about the actual repartition of freshwater snails worldwide which makes new emergences difficult to predict. In this study, we developed two ready‐to‐use environmental DNA‐based methods for assessing the distribution of B. truncatus from water samples collected in the field. We used two approaches, a quantitative PCR (qPCR) and a droplet digital PCR (ddPCR) approach. We successfully detected B. truncatus in natural environments where the snail was previously visually reported. Our environmental DNA diagnostic methods showed a high sensitivity (≈60 DNA copy per mL of filtered water) and a high specificity to B. truncatus. Results obtained in qPCR and ddPCR were very similar. This study demonstrates that environmental DNA diagnostics tools enable a sensitive large‐scale monitoring of snail‐borne diseases hence allowing the delimitation of areas potentially threatened by urogenital schistosomiasis.

Abstract

Phoxinus minnows are small freshwater fish found throughout Eurasia. The taxonomy of this genus is still under investigation, and new species are regularly described. They are frequently introduced outside their native range due to their use as live bait for angling, and such fish introductions can have diverse impacts on the recipient areas. Before the rise in interest regarding the diversity of this genus in the past 15 years, studies carried out on their biology and ecology reported their findings for the Eurasian minnow Phoxinus phoxinus Linnaeus, 1758, which was then considered ubiquitous. A parasites checklist for Phoxinus minnows has yet to be published, and the recent progress on the taxonomy of this genus has enabled us to propose a reassignment of parasite records to their proper host. The most thoroughly studied potential species were the North European species Phoxinus sp. 7 sensu Palandačić et al. 2017, Phoxinus isetensis, and P. phoxinus. We were able to highlight a gap in knowledge for numerous species that have received little‐to‐no attention in terms of parasitology. Systematic molecular identification of Phoxinus minnows should be conducted when studying their parasites, as a reliable identification of the host is vital in parasitology research. Future research will certainly reveal a species‐specific composition of the parasitofauna occurring in Phoxinus, especially among widespread parasites such as Gyrodactylus and Diplostomum. Abstract

Abstract: Electron microscopy of the testes of the free-living flatworm Mesocastrada fuhrmanni collected from temporary freshwater ponds shows stages of spermiogenesis that are like other species of the Typhloplanidae. Spermiogenesis in Mesocastrada fuhrmanni is characterized by the presence, in the spermatid, of a differentiation zone underlain by peripheral microtubules and centered on two centrioles with an intercentriolar body. Two flagella of the 9+“1” pattern of the Trepaxonemata grow out in opposite directions from the centrioles. The flagella undergo a latero-ventral rotation, and a subsequent disto-proximal rotation of centrioles occurs in the spermatid. The former rotation involves the compression and the detachment of a row of cortical microtubules, and allows us to recognize a ventral from a dorsal side. Two features are of special interest at the end of differentiation: peripheral cortical microtubules lie parallel to the sperm axis near the anterior tip, but microtubules become twisted (about 40° with reference to the gamete axis) near the posterior extremity; in the same way, the posterior tip of the nucleus is spiralled. As far as we know, these features are observed for the first time in the Typhloplanidae. The pattern of spermiogenesis and the ultrastructural organization of the spermatozoon are compared with the available data on Typhloplanoida and in particular, species of the Typhloplanidae family. Abstract

The ultrastructure of the spermatozoon of Calliobothrium verticillatum(Cestoda, Tetraphyllidea, Oncobothriidae) parasite of the smoothhound shark, Mustelus mustelusL. (Pisces, Carcharhiniformes), was studied by transmission electron microscopy. This spermatozoon presents five regions characterized by several ultrastructural elements: an apical cone, a crested body, two axonemes of 9 + “1” pattern, electron-dense granules, a nucleus and cortical microtubules. In the present study, three of these features were the subject of a detailed attention. The first is the presence of two axonemes, which confirms that the Tetraphyllidea, Oncobothriidae possess two axonemes whereas the Tetraphyllidea, Phyllobothriidae possess only one axoneme. The second is the presence of one crested body, a criterion homogeneous in the Tetraphyllidea but heterogeneous among the different orders of Cestoda. The third is the number and the disposition of cortical microtubules. These three criteria seem to be interesting for phylogeny.
