Abstract

The human papillomavirus (HPV) is a non-enveloped DNA virus transmitted through skin-to-skin contact that infects epithelial and mucosal tissue. It has over 200 known genotypes, classified by their pathogenicity as high-risk and low-risk categories. High-risk HPV genotypes are associated with the development of different types of cancers, including cervical cancer, which is a leading cause of mortality in women. In clinical practice and the market, the principal tests used to detect HPV are based on cytology, hybrid detection, and qPCR. However, these methodologies may not be ideal for the required timely diagnosis. Tests have been developed based on isothermal nucleic acid amplification tests (INAATs) as alternatives. These tests offer multiple advantages over the qPCR, such as not requiring specialized laboratories, highly trained personnel, or expensive equipment like thermocyclers. This review analyzes the different INAATs applied for the detection of HPV, considering the specific characteristics of each test, including the HPV genotypes, gene target, the limit of detection (LOD), detection methods, and detection time. Additionally, we discuss the tests available on the market that are approved by the Food and Drug Administration (FDA). Finally, we address the challenges and potential solutions for the large-scale implementation of INAATs, particularly in rural or underserved areas. [ABSTRACT FROM AUTHOR]

Abstract

The encapsulation of gallic acid (GA) through several methods has enhanced its shelf life and facilitated industrial applications. Polymeric matrices made of alginate and pectin were evaluated to encapsulate GA via spray drying. The pH-responsive release mechanism was monitored to validate the matrices' performances as wall materials and extend the bioactive compound stability. The microcapsules produced were characterized via scanning electron microscopy (SEM), dynamic light scattering (DLS), Fourier-transform infrared spectroscopy (FTIR), and cyclic voltammetry (CV). The retention and encapsulation efficiency ranges were 45–82% and 79–90%, respectively. The higher values were reached at 3 and 0.75% (w/v) pectin and sodium alginate, respectively. The scanning electron microscopy showed smooth spherical capsules and the average particle size ranged from 1327 to 1591 nm. Their performance and stability were evaluated with optimal results at a pH value of 7 throughout the investigation period. Therefore, this work demonstrated the suitability of gallic acid encapsulation via spray drying using pectin and alginate, which are biopolymers that can be obtained from circular economy processes starting from agro-industrial biomass. The developed formulations provide an alternative to protecting and controlling the release of GA, promoting its application in the food, pharmaceutical, and cosmetic industries and allowing for the release of compounds with high bioactive potential. Abstract

Sialidosis is a rare autosomal recessive disease that presents with progressive lysosomal storage of sialylated glycopeptides and oligosaccharides caused by homozygous or compound heterozygous sequence variants in the neuraminidase 1 (NEU1) gene. These sequence variants can lead to sialidosis type I and II; the latter is the most severe and presents prenatally or at early age. However, sialidosis diagnosis is challenging, especially in those health systems with limited resources of developing countries. Consequently, it is necessary to dip into high-throughput molecular diagnostic tools to allow for an accurate diagnosis with better cost-effectiveness and turnaround time. We report a 4-member pedigree segregating an ultrarare missense variant, c.1109A>G; p.Tyr370Cys, in NEU1 as detected by whole-exome sequencing. Two short-lived siblings, who presented with previously unreported clinical features from such a homozygous sequence variant, were diagnosed with sialidosis type II. Additionally, we present a novel molecular model exhibiting the consequences of the variant in the sialidase-1 tridimensional structure. This study allowed us to provide a definitive diagnosis for our patients, increase our understanding of this pathogenic variant, and improve genetic counseling.

Abstract

Breast cancer (BC) is one of the leading causes of death in women worldwide. A major challenge in BC is chemoresistance, which is often modulated by epigenetic regulators such as long non-coding RNAs (lncRNAs). Because these regulator lncRNAs may play diverse roles, determining their specific pathways and/or functions is crucial to identify possible biomarkers and/or therapeutic targets for BC. In this study, we used gene expression microarrays in order to identify lncRNAs related to the BC biology. We found, among six differentially expressed (DE) lncRNAs, that the expression of lnc-ERP44-3:6 was consistently down-regulated in all breast tumor tissues compared to normal adjacent tissues of BC patients from northeastern Mexico. Since the role of lnc-ERP44-3:6 remains unknown in BC, we induced its transient overexpression in three different BC cell lines (i.e., MCF10A, MCF-7, and HCC1395) by transfection in order to elucidate its potential downstream effects. Remarkably, our results showed a significant increase in cell death and chemosensitivity to cisplatin at 48 h post-transfection (p < 0.01). In addition, we observed that GAPDH and FAS were up-regulated following the overexpression of lnc-ERP44-3:6. As GAPDH and FAS promote apoptosis in cancer, our findings suggest that the lnc-ERP44-3:6 overexpression induces cell death possibly via up-regulation of one or both genes in BC cell lines. To the best of our knowledge, this is the first study evaluating the overexpression of lnc-ERP44-3:6 and providing insights about its role in BC cells, which suggests that this lncRNA may be an interesting candidate as a potential therapeutic target for BC in our population. Abstract

Introducción: El desarrollo del SNC es un proceso compuesto de una serie de modificaciones y una compleja expresión de genes espacio-temporal. Aunado a esto, la complejidad anatómica en mamíferos hace que se presenten problemas para el análisis de la función y expresión de los genes. El reto más difícil es el hecho de que hay cientos de tipos de linajes celulares funcionalmente y morfológicamente definidos. Lo que da como resultado que la interpretación de los fenotipos causados por anomalías en el SNC, sea en ocasiones muy difícil de asociar a patologías neurológicas provocadas por la alteración de un gen o grupo de genes. En la actualidad, se tiene un limitado conocimiento sobre los genes y la función de genes involucrados en procesos biológicos en SNC. Por lo tanto, es de vital importancia el descubrimiento de nuevos genes y su función en el SNC. Esto nos permitirá relacionar el origen de diferentes patologías neurológicas descubiertas y aún por descubrir. Tal es el caso de ZMIZ1, del cual no se tiene claro su papel en el SNC. Este gen se encontró truncado y fusionado con otro gen debido a una translocación equilibrada de novo entre los cromosomas 10 y 19, en una niña con DI y múltiples alteraciones neuropsiquiátricas. Objetivo: Analizar la expresión relativa de Zmiz1 en las diferentes etapas de desarrollo de cerebro de rata, y en el transcriptoma de la línea celular de gioblastoma C6 de rata antes y tras la inhibición de ZMIZ1 por un ARNi. A partir de los genes diferencialmente expresados en la línea celular, sugerir las posibles vías reguladas por dicho gen en células nerviosas. Material y Métodos: Se analizó la expresión relativa de ZMIZ1 en las diferentes etapas de desarrollo de cerebro de rata (embrión, recién nacido y adulto) y en la línea celular de glioblastoma C6, mediante PCR tiempo real. Abstract

Sargassumbiomass is a potential ingredient for aquaculture formulations due to its high nutritional value and availability, as it contains a variety of essential nutrients including proteins, carbohydrates, lipids, vitamins, antioxidants, fiber, and minerals, which are essential for aquatic growing, development and health parameters including digestibility and immune response against diseases. Therefore, in this work, five experimental diets were formulated considering 2.5 % and 5 % levels of inclusion of Sargassumspp. from the Mexican Caribbean coasts (SC), and from the Gulf of California (SG), and one reference diet as a control. The characterization of the raw material and the proximal analysis of the five diets were evaluated according to standard reference methods. A 35-day feeding trial was conducted using juvenile Litopenaeus vannameishowed a shrimp survival rate of 97 ± 6 % (p > 0.05) on SC. Excellent weight gain (WG) performance of SC and SG diets at 2.5 % of inclusion resulted in >10 % WG compared to the control. The specific activity of amylase, lipase, trypsin, and chymotrypsin showed a significant increase by including Sargassumin diets, compared with the reference diet. Incorporating Sargassumat 2.5 % in feed formulations for shrimps showed a great opportunity to use it as an ingredient and improved production yields. The digestion and absorption parameters were represented by the enzymatic activity in which the diets based on Sargassumspp. from the Mexican Caribbean coast (SC) have resulted in the best performance.
