Hybrid breeding has a great potential to significantly boost wheat yields. Ideal male pollinators would be taller in stature, contain many spikelets well-spaced along the spike and exhibit high extrusion of large anthers. Most importantly, flowering time would match with that of the female parent. Available genetic resources for developing an elite wheat pollinator are limited, and the genetic basis for many of these traits is largely unknown. Here, we report on the genetic analysis of pollinator traits using biparental mapping populations. We identified two anther extrusion QTLs of medium effect, one on chromosome 1BL and the other on 4BS coinciding with the semi-dwarfing Rht-B1 locus. The effect of Rht-B1 alleles on anther extrusion is genotype dependent, while tall plant Rht-B1a allele is consistently associated with large anthers. Multiple QTLs were identified at the Ppd-D1 locus for anther length, spikelet number and spike length, with the photoperiod-sensitive Ppd-D1b allele associated with favourable pollinator traits in the populations studied. We also demonstrated that homeoloci, Rht-D1 and Ppd-B1, influence anther length among other traits. These results suggest that combinations of Rht-B1 and Ppd-D1 alleles control multiple pollinator traits and should be major targets of hybrid wheat breeding programs.

Soil salinity is a major limitation to cereal production. Breeding new salt-tolerant cultivars has the potential to improve cereal crop yields. In this study, a doubled haploid bread wheat mapping population, derived from the bi-parental cross of Excalibur × Kukri, was grown in a glasshouse under control and salinity treatments and evaluated using high-throughput non-destructive imaging technology. Quantitative trait locus (QTL) analysis of this population detected multiple QTL under salt and control treatments. Of these, six QTL were detected in the salt treatment including one for maintenance of shoot growth under salinity (QG(1–5).asl-7A), one for leaf Na+ exclusion (QNa.asl-7A) and four for leaf K+ accumulation (QK.asl-2B.1, QK.asl-2B.2, QK.asl-5A and QK:Na.asl-6A). The beneficial allele for QG(1–5).asl-7A (the maintenance of shoot growth under salinity) was present in six out of 44 mainly Australian bread and durum wheat cultivars. The effect of each QTL allele on grain yield was tested in a range of salinity concentrations at three field sites across 2 years. In six out of nine field trials with different levels of salinity stress, lines with alleles for Na+ exclusion and/or K+maintenance at three QTL (QNa.asl-7A, QK.asl-2B.2 and QK:Na.asl-6A) excluded more Na+ or accumulated more K+ compared to lines without these alleles. Importantly, the QK.asl-2B.2allele for higher K+ accumulation was found to be associated with higher grain yield at all field sites. Several alleles at other QTL were associated with higher grain yields at selected field sites.

The combined assembly results in 18 super-scaffolds across the chromosome. The value of finished genome regions is demonstrated for two approximately 2.5 Mb regions associated with yield and the grain quality phenotype of fructan carbohydrate grain levels. In addition, the 50 Mb centromere region analysis incorporates cytological data highlighting the importance of non-sequence data in the assembly of this complex genome region. Conclusions: Sufficient genome sequence information is shown to now be available for the wheat community to produce sequence-finished releases of each chromosome of the reference genome. The high-level completion identified that an array of seven fructosyl transferase genes underpins grain quality and that yield attributes are affected by five F-box-only-protein-ubiquitin ligase domain and four root-specific lipid transfer domain genes. The completed sequence also includes the centromere.

Historically, wheat yields in drought-prone Australian environments have been consistently increasing for over a century. There is currently an agreement that approximately half of that increase is attributable to breeding programmes, but their physiological basis remains poorly documented. In this investigation, we hypothesized that limited whole-plant transpiration rate (TR) under high atmospheric vapour pressure deficit (VPD) could result in advantageous water conservation and crop yield increase under south Australian conditions. Therefore, TR response to VPD was measured in the 0.9-3.2 kPa range for a group of 23 wheat cultivars that were released from 1890 to 2008. Consistent with a water-conservation hypothesis, all genotypes displayed a VPD break point (BP) in TR with increasing VPD such that TR was limited at VPD above a BP of about 2 kPa. The BP and slope of TR with increasing VPD above the break point were correlated with the year of release, although the changes were in different directions. Such changes in these transpiration parameters were independent of plant leaf area and only marginally correlated with Zadok's stages. These results indicated that selection over 120 years by breeders for yield increase unconsciously resulted in genotype selection for the expression of the limited-TR trait. © 2016 Blackwell Verlag GmbH.

The research identified rye chromosome 4R arms associated with good pollinator traits, and demonstrated possible use of rye genetic resources to develop elite pollinators for hybrid wheat breeding. Bread wheat (Triticum aestivum) is a predominantly self-pollinating plant which has relatively small-sized anthers and produces a low number of pollen grains. These features limit the suitability of most wheat lines as pollinators for hybrid seed production. One strategy for improving the pollination ability of wheat is to introgress cross-pollination traits from related species. One such species is rye (Secale cereale L.), which has suitable traits such as high anther extrusion, long anthers containing large amounts of pollen and long pollen viability. Therefore, introducing these traits into wheat is of great interest in hybrid wheat breeding. Here, we investigated wheat-rye chromosome addition lines for the effects of rye chromosomes on anther and pollen development in wheat. Using a single nucleotide polymorphism genotyping array, we detected 984 polymorphic markers that showed expected syntenic relationships between wheat and rye. Our results revealed that the addition of rye chromosomes 1R or 2R reduced pollen fertility, while addition of rye chromosome 4R increased anther size by 16% and pollen grain number by 33%. The effect on anther length was associated with increases in both cell size and the number of endothecium cells and was attributed to the long arm of chromosome 4R. In contrast, the effect on pollen grain number was attributed to the short arm of chromosome 4R. These results indicate that rye chromosome 4R contains at least two genetic factors associated with increased anther size and pollen grain number that can favourably affect pollination traits in wheat.

MicroRNAs (miRNAs) are a class of important biological elements that regulates gene expression by translational repression or transcript degradation both in plants and animals. The origin, distribution and evolution of the miRNA genes have been systematically studied in many model plants, but they are not well understood in wheat. In this study, we have systematically identified the miRNA precursors and mature miRNAs in wheat group 7 chromosomes by computational analysis in the isolated chromosome sequences. A total of 716 precursors belonging to 43 miRNA families were identified, of which 12 miRNAs families (27.91 %) were not reported in wheat. Further analysis found that there were less mature miRNAs but more miRNA precursors distributing on 7D compared to 7A and 7B, suggesting that wheat group 7 homoeologous chromosome eliminated some miRNA precursors with the potential to yield the same miRNA and formed a more precise regulatory network via the two hybridization events. The target genes of all the newly identified miRNAs were also predicted. The results of this study are beneficial to improve our understanding of the distribution of wheat miRNAs and its evolutionary role in polyploidisation.

We propose in this paper a fully automatic approach for image-based plant stomata phenotyping. Given a microscopic image of a plant leaf surface, our goal is to automatically detect stomata cells and measure their morphological and structural features, such as stomata opening length and width, and size of the guard cells. The main challenge in developing such tool is the lack of contrast between the stomata cell region and its surrounding background. Our approach uses template matching to detect individual stomata cells and local analysis to measure stomata features within the detected stomata regions. It is fully automatic and computationally efficient. Thus, it will enable plant biologists to perform large scale analysis of stomata morphology, which in turn will help in developing understanding and controlling plant's response to various environmental stresses (e.g. drought and soil salinity).
