Abstract

Pediatric medulloblastoma (MB) is the most common solid malignant brain neoplasm, with Group 3 (G3) MB representing the most aggressive subgroup. MYC amplification is an independent poor prognostic factor in G3 MB, however, therapeutic targeting of the MYC pathway remains limited and alternative therapies for G3 MB are urgently needed. Here we show that the RNA-binding protein, Musashi-1 (MSI1) is an essential mediator of G3 MB in both MYC-overexpressing mouse models and patient-derived xenografts. MSI1 inhibition abrogates tumor initiation and significantly prolongs survival in both models. We identify binding targets of MSI1 in normal neural and G3 MB stem cells and then cross referenced these data with unbiased large-scale screens at the transcriptomic, translatomic and proteomic levels to systematically dissect its functional role. Comparative integrative multi-omic analyses of these large datasets reveal cancer-selective MSI1-bound targets sharing multiple MYC associated pathways, providing a valuable resource for context-specific therapeutic targeting of G3 MB.

7. Linear and nonlinear optical properties of silica aerogel

Abstract

Scattering media have traditionally been seen as a hindrance to the controlled transport of light through media, creating the familiar speckle pattern. However such matter does not cause the loss of information but instead performs a highly complex deterministic operation on the incoming flux. Through sculpting the properties of the incoming wavefront, we can unlock the hidden characteristics of these media, affording us far more degrees of freedom than that which is available to us in traditional ballistic optics. These additional degrees of freedom have allowed for the creation of compact sophisticated optical devices based only on the deterministic nature of light scattering. Such devices include diffraction-limit-beating lenses, polarimeters, spectrometers, and some which can transmit entire images through a scattering substance. Additional degrees of freedom would allow for the creation of even more powerful devices, in new working regimes. In particular, the application of related techniques where the scattering material is actively modified is limited. This thesis is concerned with the use of optothermal nonlinearity in random media as a way to provide an additional degree of control over light which scatters through it. Specifically, we are concerned with silica aerogel as a platform for this study. Silica aerogel is a lightweight skeletal structure of silica fibrils, which results in a material which is up to 99.98 % by volume. This material exhibits a unique cocktail of properties of use such as near unitary refractive index, an order of magnitude lower thermal conductivity, and high optothermal nonlinearity. The latter two of these properties allow for the creation of localised steep thermal gradients, proportionally affecting the low refractive index significantly. Additionally through differing fabrication steps, the opacity, and as a result, we can adjust the scattering strength. In line with the development of light deterministic light scattering techniques in linear media, we develop through the use of pump-probe setups, a framework for the development of a similar line of techniques in nonlinear scattering media. We show that we can reversibly control the far-field propagation of light in weakly scattering silica aerogel. Following this, we show that nonlinear perturbation can be used to extend and modify the optical memory effect, where slight adjustments in scattering direction maintain the overall correlation of the scattered profile. Finally, we measure the nonlinear transmission matrix, a complete description of how any wavefront would pass through at a particular point in a scattering media, and how that scattering can be modified through the application of an optothermal nonlinearity. Extending the tool of scattering media into the nonlinear regime helps pave the way toward the next set of advances in the field of light scattering control.

Abstract

We report on the characterization of silica aerogel thermal optical nonlinearity, obtained by z-scan technique. The results show that typical silica aerogels have nonlinear optical coefficient similar to that of glass $(\simeq 10^{-12} $m$^2/$W), with negligible optical nonlinear absorption. The non\-li\-near coefficient can be increased to values in the range of $10^{-10} $m$^2/$W by embedding an absorbing dye in the aerogel. Abstract

We recently reported that atypical teratoid rhabdoid tumors (ATRTs) comprise at least two transcriptional subtypes with different clinical outcomes; however, the mechanisms underlying therapeutic heterogeneity remained unclear. In this study, we analyzed 191 primary ATRTs and 10 ATRT cell lines to define the genomic and epigenomic landscape of ATRTs and identify subgroup-specific therapeutic targets. We found ATRTs segregated into three epigenetic subgroups with distinct genomic profiles, SMARCB1 genotypes, and chromatin landscape that correlated with differential cellular responses to a panel of signaling and epigenetic inhibitors. Significantly, we discovered that differential methylation of a PDGFRB-associated enhancer confers specific sensitivity of group 2 ATRT cells to dasatinib and nilotinib, and suggest that these are promising therapies for this highly lethal ATRT subtype.

Abstract

The monocyte–macrophage system plays an important role in phagocytosis of pathogens and cellular debris following infection or tissue injury in several pathophysiological conditions. We examined ENaC/ASIC subunit transcript expression and the importance of select subunits in migration of bone marrow derived monocytes (freshly isolated) and macrophages (monocytes differentiated in culture). We also examined the effect of select subunit deletion on macrophage phenotype. BM monocytes were harvested from the femurs of male and female WT and KO mice (6–12 weeks of age). Our results show that α, β, γENaC, and ASIC1‐5 transcripts are expressed in BM macrophages and monocytes to varying degrees. At least αENaC, βENaC, and ASIC2 subunits contribute to chemotactic migration responses in BM monocyte–macrophages. Polarization markers (CD86, soluble TNFα) in BM macrophages from mice lacking ASIC2a plus βENaC were shifted towards the M1 phenotype. Furthermore, select M1 phenotypic markers were recovered with rescue of βENaC or ASIC2. Taken together, these data suggest that βENaC and ASIC2 play an important role in BM macrophage migration and loss of βENaC and/or ASIC2 partially polarizes macrophages to the M1 phenotype. Abstract

Background: The treatment of BRAF V600E gliomas with BRAF inhibitors (BRAFis) and MEK inhibitors (MEKis) has been increasingly integrated into clinical practice for pediatric low-grade gliomas (PLGGs) and pediatric high-grade gliomas (HGGs). However, some questions remain unanswered, such as the best time to start targeted therapy, duration of treatment, and discontinuation of therapy. Given that no clinical trial has been able to address these critical questions, we developed a Canadian Consensus statement for the treatment of BRAF V600E mutated pediatric as well as adolescent and young adult (AYA) gliomas. Methods: Canadian neuro-oncologists were invited to participate in the development of this consensus. The consensus was discussed during monthly web-based national meetings, and the algorithms were revised until a consensus was achieved. Results: A total of 26 participants were involved in the development of the algorithms. Two treatment algorithms are proposed, one for the initiation of treatment and one for the discontinuation of treatment. We suggest that most patients with BRAF V600E gliomas should be treated with BRAFis ± MEKis upfront. Discontinuation of treatment can be considered in certain circumstances, and we suggest a slow wean. Conclusions: Based on expert consensus in Canada, we developed algorithms for treatment initiation of children and AYA with BRAF V600E gliomas as well as a discontinuation algorithm. Abstract

Physical unclonable functions (PUFs) are complex physical objects that aim at overcoming the vulnerabilities of traditional cryptographic keys, promising a robust class of security primitives for different applications. Optical PUFs present advantages over traditional electronic realizations, namely, a stronger unclonability, but suffer from problems of reliability and weak unpredictability of the key. We here develop a two-step PUF generation strategy based on deep learning, which associates reliable keys verified against the National Institute of Standards and Technology (NIST) certification standards of true random generators for cryptography. The idea explored in this work is to decouple the design of the PUFs from the key generation and train a neural architecture to learn the mapping algorithm between the key and the PUF. We report experimental results with all-optical PUFs realized in silica aerogels and analyzed a population of 100 generated keys, each of 10,000 bit length. The key generated passed all tests required by the NIST standard, with proportion outcomes well beyond the NIST’s recommended threshold. The two-step key generation strategy studied in this work can be generalized to any PUF based on either optical or electronic implementations. It can help the design of robust PUFs for both secure authentications and encrypted communications.

Abstract

Background Cancer predisposition syndromes (CPSs) are responsible for at least 10% of cancer diagnoses in children and adolescents, most of which are not clinically recognised prior to cancer diagnosis. A variety of clinical screening guidelines are used in healthcare settings to help clinicians detect patients who have a higher likelihood of having a CPS. The McGill Interactive Pediatric OncoGenetic Guidelines (MIPOGG) is an electronic health decision support tool that uses algorithms to help clinicians determine if a child/adolescent diagnosed with cancer should be referred to genetics for a CPS evaluation. Methods This study assessed MIPOGG’s performance in identifying Li-Fraumeni, DICER1, Constitutional mismatch repair deficiency and Gorlin (nevoid basal cell carcinoma) syndromes in a retrospective series of 84 children diagnosed with cancer and one of these four CPSs in Canadian hospitals over an 18-year period. Results MIPOGG detected 82 of 83 (98.8%) evaluable patients with any one of these four genetic conditions and demonstrated an appropriate rationale for suggesting CPS evaluation. When compared with syndrome-specific clinical screening criteria, MIPOGG’s ability to correctly identify children with any of the four CPSs was equivalent to, or outperformed, existing clinical criteria respective to each CPS. Conclusion This study adds evidence that MIPOGG is an appropriate tool for CPS screening in clinical practice. MIPOGG’s strength is that it starts with a specific cancer diagnosis and incorporates criteria relevant for associated CPSs, making MIPOGG a more universally accessible diagnostic adjunct that does not require in-depth knowledge of each CPS. Abstract

The present study investigated the post-activation performance enhancement (PAPE) of isokinetic quadriceps and hamstrings torque after flywheel (FW)-squat vs. FW-deadlift in comparison to a control condition. Fifteen male athletes were enrolled in this randomised, crossover study. Each protocol consisted of 3 sets of 6 repetitions, with an inertial load of 0.029 kg.m2. Isokinetic quadriceps (knee extension) and hamstrings (knee flexion) concentric peak torque (60º/s) and hamstring eccentric peak torque (−60º/s) were measured 5 min after experimental or control conditions. A significant condition (PAPE) effect was reported (f = 4.067, p = 0.008) for isokinetic hamstrings eccentric peak torque following FW-squat and FW-deadlift, but no significant differences were found for quadriceps and hamstrings concentric peak torques. The significant difference averaged 14 Nm between FW-squat vs. control (95% CI: 2, 28; d = 0.75, moderate; p = 0.033), and 13 Nm between FW-deadlift vs. control (95% CI: 1, 25; d = 0.68, moderate; p = 0.038). This study reported that both FW-squat and FW-deadlift exercises are equivalently capable of generating PAPE of isokinetic hamstrings eccentric torque. Practitioners may use these findings to inform strength and power development during complex training sessions consisting of flywheel-based exercises prior to a sport-specific task.
