International audience; Understanding how populations are inter-connected can help identify species that are vulnerable or those that are likely to persist over ecological and evolutionary time scales. Favia fragum and Favia gravida are sister species of brooding corals common in the Atlantic Ocean that are secondary reef-builders that contribute to habitat complexity in intertidal and shallow-water coral reefs. To better understand population connectivity in these two congeneric species with broad and presumably non-overlapping distributions, one single-copy nuclear marker (masc1) and one ribosomal marker (ITS) were used to estimate population genetic parameters for Favia in four biogeographic regions: the Caribbean, Brazilian, Ascension and Tropical Eastern Atlantic provinces. Strong and significant levels of population structure were found among all four biogeographic regions, with each province being characterized by nearly all private alleles. Phylogenetic analyses based on each gene and on a combined marker analysis showed strong support for three sister clades composed of individuals from either the Caribbean, South Atlantic (Brazilian + Tropical Eastern Atlantic), or Ascension provinces. However, low genetic diversity was observed at each location, and no differentiation was observed among populations along the entire coast of Brazil (~ 1600 km). Furthermore, the Tropical Eastern Atlantic was likely colonized by at least two independent founder events, one from the Caribbean and another from the Brazilian provinces. Life-history traits of the genus might explain the different patterns found. Favia fragum and F. gravida release zooxanthellae larvae monthly and may be capable of rafting, which could favor occasional long-distance dispersal. On the other hand, they are capable of self-fertilization and commonly disperse over a few meters which may explain low genetic variability locally. Our results show that while long-distance migration events are possible for Favia corals, they are insufficient to maintain connectivity across biogeographic regions. Lastly, the Ascension Island population was more divergent than the sister clades composed of Caribbean or South Atlantic Favia, indicating a possible cryptic lineage, but further work may clarify species boundaries within the Atlantic coral genus Favia

The coastal oceans can be highly variable, especially near ocean fronts. The Ushant Front is the dominant oceanographic feature in the Iroise Sea (NE Atlantic) during summer, separating warm stratified offshore waters from cool vertically-mixed nearshore waters. Mesozooplankton community structure was investigated over an annual cycle to examine relationships with oceanographic conditions. DNA metabarcoding of COI and 18S genes was used in communities from six sites along two cross-shelf transects. Taxonomic assignments of 380 and 296 OTUs (COI and 18S, respectively) identified 21 classes across 13 phyla. Meroplankton relative abundances peaked in spring and summer, particularly for polychaete and decapod larvae, respectively, corresponding to the reproductive periods of these taxa. Meroplankton was most affected by season, while holoplankton varied most by shelf position. Copepods with a mixed feeding strategy were associated with the most offshore sites, especially in the presence of the front, while filter-feeding or carnivorous copepods were associated with nearshore sites. In sum, mesozooplankton communities in well-mixed coastal waters were distinct from those found in the Ushant Front (high thermal stratification and chlorophyll-a). Furthermore, the benthic compartment, through its partial life cycle in the water column, contributed to high heterogeneity in planktonic communities over short temporal and spatial scales.

In contrast to most Caribbean corals, the mustard hill coral Porites astreoides Lamarck, 1816 has seen its relative coverage increasing across its distribution range in recent years. Interpreting the reasons of such success remains, however, challenging. On the one hand, genetic homogeneity has been observed over large distances, in line with a large effective population size and/or high migration rate. On the other hand, strong genetic differentiation has been recently observed across depths within regions, consistent with some of the life history traits of P. astreoides, such as brooding of larvae followed by a short larval pelagic duration. Based on seven informative microsatellite markers and extensive sampling covering a large part of the P. astreoides distribution in the Caribbean, including six depth transects, we assessed multi-scale genetic connectivity patterns to test whether populations were structured by bathymetry, microenvironments or geography. While genetic diversity was similar in P. astreoides populations, genetic differentiation was observed not only over broad spatial scales, but also at the island scale or within a site along a depth transect. Isolation by distance was observed across the Lesser Antilles populations. However, genetic homogeneity was also observed over large distances, which, in juxtaposition to significant genetic structure within various locations across the Caribbean, has led to a complex spatial pattern of genetic differentiation in P. astreoides for which several likely explanations, including mixed reproductive strategy, are discussed.

International audience; Species distributions have been profoundly affected by past climate change, and are expected to change considerably in response to future environmental change. To better apprehend how future climate change is likely to affect genetic diversity in marine populations, it is essential to first evaluate the processes that have shaped the current distribution of genetic diversity in the sea. The honeycomb worm is a reef-building polychaete that hosts high biodiversity. Here we show that the genetic diversity in populations of S. alveolata is highest towards the edges of the current species range and lowest at its center. Pleistocene glacial cycles likely led to extirpations of S. alveolata from central populations in the Bay of Biscay, with coalescent-based estimates of post-glacial colonization dating to the beginning of the Holocene interglacial, from 10,000 to 14,000 years ago. Meanwhile, populations in the Irish Sea and English Channel likely persisted in glacial refugia since the Eemian interglacial, 120,000 years ago. Northern populations host at least two sets of divergent haplotypes, indicating that two refugia possibly existed in the north, with Ireland being a likely second refugium. Within biogeographic regions, populations were overall well-connected, but strong genetic differentiation suggests that little exchange occurs between regions. These two unexpected reservoirs of genetic diversity at the range edges deserve greater attention as warming temperatures threaten trailing edge populations, while greater climatic variability threatens leading edge populations.

Aim To investigate some of the environmental variables underpinning the past and present distribution of an ecosystem engineer near its poleward range edge. Location >500 locations spanning >7,400 km around Ireland. Methods We collated past and present distribution records on a known climate change indicator, the reef‐forming worm Sabellaria alveolata (Linnaeus, 1767) in a biogeographic boundary region over 182 years (1836–2018). This included repeat sampling of 60 locations in the cooler 1950s and again in the warmer 2000s and 2010s. Using species distribution modelling, we identified some of the environmental drivers that likely underpin S. alveolata distribution towards the leading edge of its biogeographical range in Ireland. Results Through plotting 981 records of presence and absence, we revealed a discontinuous distribution with discretely bounded sub‐populations, and edges that coincide with the locations of tidal fronts. Repeat surveys of 60 locations across three time periods showed evidence of population increases, declines, local extirpation and recolonization events within the range, but no evidence of extensions beyond the previously identified distribution limits, despite decades of warming. At a regional scale, populations were relatively stable through time, but local populations in the cold Irish Sea appear highly dynamic and vulnerable to local extirpation risk. Contemporary distribution data (2013–2018) computed with modelled environmental data identified specific niche requirements which can explain the many distribution gaps, namely wave height, tidal amplitude, stratification index, then substrate type. Main conclusions In the face of climate warming, such specific niche requirements can create environmental barriers that may prevent species from extending beyond their leading edges. These boundaries may limit a species’ capacity to redistribute in response to global environmental change.

Background Under the threat of climate change populations can disperse, acclimatise or evolve in order to avoid fitness loss. In light of this, it is important to understand neutral gene flow patterns as a measure of dispersal potential, but also adaptive genetic variation as a measure of evolutionary potential. In order to assess genetic variation and how this relates to environment in the honeycomb worm (Sabellaria alveolata (L.)), a reef-building polychaete that supports high biodiversity, we carried out RAD sequencing using individuals from along its complete latitudinal range. Patterns of neutral population genetic structure were compared to larval dispersal as predicted by ocean circulation modelling, and outlier analyses and genotype-environment associations tests were used to attempt to identify loci under selection in relation to local temperature data. Results We genotyped 482 filtered SNPs, from 68 individuals across nine sites, 27 of which were identified as outliers using BAYESCAN and ARLEQUIN. All outlier loci were potentially under balancing selection, despite previous evidence of local adaptation in the system. Limited gene flow was observed among reef-sites (FST = 0.28 ± 0.10), in line with the low dispersal potential identified by the larval dispersal models. The North Atlantic reef emerged as an isolated population and this was linked to high local larval retention and the effect of the North Atlantic Current on dispersal. Conclusions As an isolated population, with limited potential for natural genetic or demographic augmentation from other reefs, the North Atlantic site warrants conservation attention in order to preserve not only this species, but above all the crucial functional ecological roles that are associated with their bioconstructions. Our study highlights the utility of using seascape genomics to identify populations of conservation concern.

Data that can be used to monitor biodiversity through time are essential for conservation and management. The reef-forming worm, Sabellaria alveolata (L. 1767) is currently classed as ‘Data Deficient’ due to an imbalance in the spread of data on its distribution. Little is known about the distribution of this species around Ireland. Using data archaeology, we collated past and present distribution records and discovered that S. alveolata has a discontinuous distribution with large gaps between populations. Many regions lack data and should be targeted for sampling. Biodiversity surveys revealed that S. alveolata supported diverse epibiotic algal communities. Retrograding (declining) reefs supported greater infaunal diversity than prograding (growing) reefs or sand, suggesting that S. alveolata is a dynamic ecosystem engineer that has a lasting legacy effect. Similar research should be carried out for other Data Deficient species, habitats and regions. Such data are invaluable resources for management and conservation.

Zooxanthellate corals live in symbiosis with phototrophic dinoflagellates of the family Symbiodiniaceae, enabling the host coral to dwell in shallow, nutrient-poor marine waters. The South Atlantic Ocean is characterized by low coral diversity with high levels of endemism. However, little is known about coral-dinoflagellate associations in the region. This study examined the diversity of Symbiodiniaceae associated with the scleractinian coral Favia gravida across its distributional range using the ITS-2 marker. This brooding coral endemic to the South Atlantic can be found across a wide range of latitudes and longitudes, including the Mid-Atlantic islands. Even though it occurs primarily in shallower environments, F. gravida is among the few coral species that live in habitats with extreme environmental conditions (high irradiance, temperature, and turbidity) such as very shallow tide pools. In the present study, we show that F. gravida exhibits some degree of flexibility in its symbiotic association with zooxanthellae across its range. F. gravida associates predominantly with Cladocopium C3 (ITS2 type Symbiodinium C3) but also with Symbiodinium A3, Symbiodinium linucheae (ITS2 type A4), Cladocopium C1, Cladocopium C130, and Fugacium F3. Symbiont diversity varied across biogeographic regions (Symbiodinium A3 and S. linucheae were found in the Tropical Eastern Atlantic, Cladocopium C1 in the Mid-Atlantic, and other subtypes in the Southwestern Atlantic) and was affected by local environmental conditions. In addition, Symbiodiniaceae diversity was highest in a southwestern Atlantic oceanic island (Rocas Atoll). Understanding the relationship between corals and their algal symbionts is critical in determining the factors that control the ecological niches of zooxanthellate corals and their symbionts, and identifying host-symbiont pairs that may be more resistant to environmental changes.

WOS:000464891100006; International audience; The honeycomb worm Sabellaria alveolata is capable of building extensive bioconstructions, including what are currently considered Europe's largest biogenic reefs. The size and volume of these bioconstructions, however, vary greatly, such that not all habitats engineered by S. alveolata may be easily identified as reefs. Given that European environmental legislation protects marine habitats that are classified as "reefs", it is important to identity a clear set of definition criteria. Furthermore, quantifiable and unequivocal criteria are also needed to evaluate the ecological (health) state of these reefs, in order to best monitor and protect them. Here we propose new terminology to describe the physical appearance of these bioconstructions and attempt to link these physical criteria to the physiological state of the tube-building polychaete. We tested whether a bioconstruction displaying outward signs of growth is built by "healthy" worms devoid of physiological stress by analysing three macromolecules (carbohydrates, proteins, lipids), four polar lipid fatty acids, six neutral lipid fatty acid markers and three metabolic enzymes (citrate synthase, catalase and superoxide dismutase). The worms were sampled in bioconstructions of different "Type" (veneer vs. hummock), "Phase" (progradation vs. retrogradation), and "Shore Level" (high shore vs. low shore) at Champeaux in Mont-Saint-Michel Bay, France. Our results show that worms sampled in retrograding reefs (i.e. displaying signs of erosion and colonisation by epibionts such as oysters or mussels), were less physiologically stressed than worms sampled in prograding bioconstructions, possibly due to lower intraspecific competition and hence greater food availability. We therefore suggest management measures should encompass the whole mosaic of biogenic construction Types and Phases. We propose the inclusion of the polar lipid fatty acid arachidonic acid, in combination with the activity of two metabolic enzymes, citrate synthase and superoxide dismutase, as the three key biochemical markers to consider for quantitative information on the physiological state of this particular ecosystem engineer. Our results also revealed the influence of both sex and size on fatty acid and enzyme levels, highlighting the importance of taking into account both these variables when sampling and subsequently pooling individuals by sex and size category for laboratory analyses. Once seasonal and site variation have been addressed, these biochemical indicators could be examined in parallel with S. alveolata bioconstruction physical criteria as part of a European-wide protocol for monitoring ecological status in this potential reef habitat.

Ocean acidification is a major global stressor that leads to substantial changes in seawater carbonate chemistry, with potentially significant consequences for calcifying organisms. Marine shelled mollusks are ecologically and economically important species providing essential ecosystem services and food sources for other species. Because they use calcium carbonate (CaCO3) to produce their shells, mollusks are among the most vulnerable invertebrates to ocean acidification, with early developmental stages being particularly sensitive to pH changes. This study investigated the effects of CO2-induced ocean acidification on larval development of the European abalone Haliotis tuberculata, a commercially important gastropod species. Abalone larvae were exposed to a range of reduced pHs (8.0, 7.7 and 7.6) over the course of their development cycle, from early-hatched trochophore to pre-metamorphic veliger. Biological responses were evaluated by measuring the survival rate, morphology and development, growth rate and shell calcification. Larval survival was significantly lower in acidified conditions than in control conditions. Similarly, larval size was consistently smaller under low pH conditions. Larval development was also affected, with evidence of a developmental delay and an increase in the proportion of malformed or unshelled larvae. In shelled larvae, the intensity of birefringence decreased under low pH conditions, suggesting a reduction in shell mineralization. The reef-building fire coralMillepora alcicornisis reported from Ascension Island (South Atlantic). This record, based on a recent expedition to Ascension, is the first in which the identification of aMilleporacoral is supported by photographic evidence from the field and by morphological and molecular analyses of collected specimens. This finding is discussed in relation to earlierMilleporarecords from Ascension and the biogeographic range of this particular species in the Atlantic.

The presence of five shallow-water scleractinian species at Ascension Island is confirmed, i.e. Favia gravida, Madracis sp., Siderastrea radians, Polycyathus atlanticus and Rhizosmilia gerdae. The three former are zooxanthellate, the two latter azooxanthellate. A sixth species, Cladocora debilis (azooxanthellate), dredged from moderately deep water is also expected to occur in shallow water. Madracis sp. and P. atlanticus are new records for the island. A previous record of Astrangia solitaria at Ascension is now referred to as P. atlanticus. Favia gravida, S. radians and C. debilis are amphi-Atlantic. Rhizosmilia gerdae is currently known only from Ascension Island and the Western Atlantic. None of the species are endemic to Ascension Island. No member of the family Dendrophylliidae has as yet been found at Ascension, whereas that family is represented at its nearest neighbour, St Helena Island.

Scleractinian corals (i.e. hard corals) play a fundamental role in building and maintaining coral reefs, one of the most diverse ecosystems on Earth. Nevertheless, their phylogenies remain largely unresolved and little is known about dispersal and survival of their planktonic larval phase. The small subunit ribosomal RNA (SSU rRNA) is a commonly used gene for DNA barcoding in several metazoans, and small variable regions of SSU rRNA are widely adopted as barcode marker to investigate marine plankton community structure worldwide. Here, we provide a large sequence data set of the complete SSU rRNA gene from 298 specimens, representing all known extant reef coral families and a total of 106 genera. The secondary structure was extremely conserved within the order with few exceptions due to insertions or deletions occurring in the variable regions. Remarkable differences in SSU rRNA length and base composition were detected between and within acroporids (Acropora, Montipora, Isopora and Alveopora) compared to other corals. The V4 and V9 regions seem to be promising barcode loci because variation at commonly used barcode primer binding sites was extremely low, while their levels of divergence allowed families and genera to be distinguished. A time-calibrated phylogeny of Scleractinia is provided, and mutation rate heterogeneity is demonstrated across main lineages. The use of this data set as a valuable reference for investigating aspects of ecology, biology, molecular taxonomy and evolution of scleractinian corals is discussed.

International audience; Aim : To verify the synonymy of the reef‐building polychaete Phragmatopoma caudata (described for the Caribbean) and Phragmatopoma lapidosa (described for Brazil) using molecular data. To evaluate the patterns of genetic diversity and connectivity among populations from Florida to South Brazil.Location : Intertidal zone in the western Atlantic biogeographical Region: Brazil, eastern Caribbean and Florida (USA).Methods : DNA sequence data from one mitochondrial (cox‐1) and one nuclear ribosomal (ITS‐1) loci were obtained from 11 populations of P. caudata spanning the coasts of Brazil, eastern Caribbean and Florida. Phylogenetic relationships among populations of P. caudata and other members of the genus were inferred by Bayesian methods. Population differentiation was evaluated by Bayesian analysis of population structure (baps), AMOVA and pairwise φst. Demographic history was inferred by Bayesian skyline plots.Results : Phylogenetic inference supported the interpretation of a single species of Phragmatopoma spanning the Brazilian and Caribbean Provinces of the western Atlantic Region. Little population structure was observed across the species distribution, with the exception of the Florida population. The baps analysis supported a 2‐population model, with population differentiation being strong and significant between Florida and all other Atlantic populations for cox‐1, and significant between Florida and most populations for ITS‐1. Differences in genetic diversity were not significant between Caribbean and Brazilian populations, although several populations in Brazil had low values for diversity indices. Bayesian skyline plots indicate population expansion starting at c. 200 ka.Main conclusions : Phragmatopoma caudata is able to maintain genetic connectivity across most of its geographical range, with population differentiation being observed only between Florida and all other localities, possibly due to ecological speciation in the transition zone between tropical and subtropical environments. Long‐distance connectivity across much of the species range is likely the result of long‐lived larvae that are tolerant to a wide range of environmental conditions.

WOS:000406954100003; Fire corals are the only branching corals in the South Atlantic and provide an important ecological role as habitat-builders in the region. With three endemic species (Millepora brazilensis, M. nitida and M. taboret and one amphi-Atlantic species (M. alcicornis), fire coral diversity in the Brazilian Province rivals that of the Caribbean Province. Phylogenetic relationships and patterns of population genetic structure and diversity were investigated in all four fire coral species occurring in the Brazilian Province to understand patterns of speciation and biogeography in the genus. A total of 273 colonies from the four species were collected from 17 locations spanning their geographic ranges. Sequences from the 16S ribosomal DNA (rDNA) were used to evaluate phylogenetic relationships. Patterns in genetic diversity and connectivity were inferred by measures of molecular diversity, analyses of molecular variance, pairwise differentiation, and by spatial analyses of molecular variance. Morphometrics of the endemic species M. braziliensis and M. nitida were evaluated by discriminant function analysis; macro-morphological characters were not sufficient to distinguish the two species. Genetic analyses showed that, although they are closely related, each species forms a well-supported Glade. Furthermore, the endemic species characterized a distinct biogeographic barrier: M. braziliensis is restricted to the north of the Sao Francisco River, whereas M. nitida occurs only to the south. Millepora laboreli is restricted to a single location and has low genetic diversity. In contrast, the amphi-Atlantic species M. alcicornis shows high genetic connectivity within the Brazilian Province, and within the Caribbean Province (including Bermuda), despite low levels of gene flow between these populations and across the tropical Atlantic. These patterns reflect the importance of the Amazon Orinoco Plume and the Mid Atlantic Barrier as biogeographic barriers, and suggest that, while M. alcicornis is capable of long-distance dispersal, the three endemics have restricted ranges and more limited dispersal capabilities.

International audience; Since 1997, populations of the European abalone Haliotis tuberculata suffer mass mortalities attributed to the bacterium Vibrio harveyi. These mortalities occur at the spawning season, when the abalone immune system is depressed, and when temperatures exceed 17 °C, leading to favorable conditions for V. harveyi proliferation. In order to identify mechanisms of disease resistance, experimental successive infections were carried out on two geographically distinct Brittany populations: one that has suffered recurrent mortalities (Saint-Malo) and one that has not been impacted by the disease (Molène). Furthermore, abalone surviving these two successive bacterial challenges and uninfected abalone were used for several post-infection analyses. The Saint-Malo population was found to be resistant to V. harveyi infection, with a survival rate of 95% compared to 51% for Molène. While in vitro quantification of phagocytosis by flow cytometry showed strong inhibition following the first infection, no inhibition of phagocytosis was observed following the second infection for Saint-Malo, suggesting an immune priming effect. Moreover, assays of phagocytosis of GFP-labelled V. harveyi performed two months post-infection show an inhibition of phagocytosis by extracellular products of V. harveyi for uninfected abalone, while no effect was observed for previously infected abalone from Saint-Malo, suggesting that the effects of immune priming may last upwards of two months. Detection of V. harveyi by qPCR showed that a significantly greater number of abalone from the susceptible population were positive for V. harveyi in the gills, indicating that portal of entry effectors may play a role in resistance to the disease. Collectively, these results suggest a potential synergistic effect of gills and hemolymph in the resistance of H. tuberculata against V. harveyi with an important involvement of the gills, the portal of entry of the bacteria.

00000; International audience; The European abalone Haliotis tuberculata is a delicacy and consequently a commercially valuable gastropod species. Aquaculture production and wild populations are subjected to multiple climate-associated stressors and anthropogenic pressures, including rising sea-surface temperatures, ocean acidification and an emerging pathogenic Vibrio infection. Transcript expression data provides a valuable resource for understanding abalone responses to variation in the biotic and abiotic environment. To generate an extensive transcriptome, we performed next-generation sequencing of RNA on larvae exposed to temperature and pH variation and on haemolymph of adults from two wild populations after experimental infection with Vibrio harveyi. We obtained more than 1.5 billion raw paired-end reads, which were assembled into 328,519 contigs. Filtration and clustering produced a transcriptome of 41,099 transcripts, of which 10,626 (25.85%) were annotated with Blast hits, and 7380 of these were annotated with Gene Ontology (GO) terms in Blast2Go. A differential expression analysis comparing all samples from the two life stages identified 5690 and 10,759 transcripts with significantly higher expression in larvae and adult haemolymph respectively. This is the greatest sequencing effort yet in the Haliotis genus, and provides the first high-throughput transcriptomic resource for H. tuberculata.

Acclimation and adaptation, which are key to species survival in a changing climate, can be observed in terms of membrane lipid composition. Remodelling membrane lipids, via homeoviscous adaptation (HVA), counteracts membrane dysfunction due to temperature in poikilotherms. In order to assess the potential for acclimation and adaptation in the honeycomb worm, Sabellaria alveolata, a reef-building polychaete that supports high biodiversity, we carried out common-garden experiments using individuals from along its latitudinal range. Individuals were exposed to a stepwise temperature increase from 15 °C to 25 °C and membrane lipid composition assessed. Our results suggest that S. alveolata was able to acclimate to higher temperatures, as observed by a decrease in unsaturation index and 20:5n-3. However, over the long-term at 25 °C, lipid composition patterns are not consistent with HVA expectations and suggest a stress response. Furthermore, unsaturation index of individuals from the two coldest sites were higher than those from the two warmest sites, with individuals from the thermally intermediate site being in-between, likely reflecting local adaptation to temperature. Therefore, lipid remodelling appears limited at the highest temperatures in S. alveolata, suggesting that individuals inhabiting warm environments may be close to their upper thermal tolerance limits and at risk in a changing climate.

Since 1997, populations of the European abalone Haliotis tuberculata suffer mass mortalities attributed to the bacterium Vibrio harveyi. These mortalities typically coincide with the spawning season and when temperatures exceed 17 ° C, a temperature below which the bacterial proliferation is insufficient to cause damage to the abalone. To study abalone resistance mechanisms, experimental infections were carried out on geographically distinct populations thought to have differential response to disease in the field. A first set of successive infections identified one resistant (11% mortality) and one susceptible (58% mortality) population. Two sets of successive infections were used to highlight the resistance mechanisms of this population: (1) to evaluate the immune state of the resistant population compared to a susceptible, and (2) to compare differences in global gene expression (RNA-Seq) of hemocytes in the resistant and susceptible populations. While in vivo quantification of phagocytosis by flow cytometry showed strong inhibition following the first infection, inhibition of phagocytosis was less pronounced following the second infection, suggesting an immune priming effect. Fluorescent 3D microscopy (Vivatome) validated the capability of abalone hemocytes to phagocytose V. harveyi. In vitro analyses showed a significant negative impact of extracellular products of V. harveyi on phagocytosis. Furthermore, the RNA-Seq analysis showed 2956 transcripts (24, 9% annotated) with altered expression between the resistant and susceptible abalone populations following experimental infection. In the total annotated transcriptome, 288 transcripts were related to immune functions, including several F-type lectins, toll-like receptor, tumor necrosis factor and other immune genes of interest.

Recent advances in morphometrics and genetics have led to the discovery of numerous cryptic species in coral reef ecosystems. A prime example is the Montastraea annularis scleractinian coral species complex, in which morphological, genetic, and reproductive data concur on species boundaries, allowing evaluation of long-term patterns of speciation and evolutionary innovation. Here we test for cryptic species in the Atlantic species, Montastraea cavernosa, long recognized as polymorphic. Our modern samples consist of 94 colonies collected at four locations (Belize, Panama, Puerto Rico in the Caribbean; Sao Tome in the Eastern Atlantic). Our fossil samples consist of 78 colonies from the Plio-Pleistocene of Costa Rica and Panama. Landmark morphometric data were collected on thin sections of 46 modern and 78 fossil colonies. Mahalanobis distances between colonies were calculated using Bookstein coordinates, revealing two modern and four fossil morphotypes. The remaining 48 of the 94 modern colonies were assigned to morphotype using discriminant analysis of calical measurements. Cross-tabulation and multiple comparisons tests show no significant morphological differences among geographic locations or water depths. Patterns of variation within and among fossil morphotypes are similar to modern morphotypes. DNA sequence data were collected for two polymorphic nuclear loci (β-tub1 and β-tub2) on all 94 modern colonies. Haplotype networks show that both genes consist of two clades, but morphotypes are not associated with genetic clades. Genotype frequencies and two-locus genotype assignments indicate genetic exchange across clades, and ϕst values show no genetic differentiation between morphotypes at different locations. Taken together, our morphological and genetic results do not provide evidence for cryptic species in M. cavernosa, but indicate instead that this species has an unusually high degree of polymorphism over a wide geographic area and persisting for >25 million years (myr).

Limited dispersal and connectivity in marine organisms can have negative fitness effects in populations that are small and isolated, but reduced genetic exchange may also promote the potential for local adaptation. Here, we compare the levels of genetic diversity and connectivity in the coral Montastraea cavernosa among both central and peripheral populations throughout its range in the Atlantic. Genetic data from one mitochondrial and two nuclear loci in 191 individuals show that M. cavernosa is subdivided into three genetically distinct regions in the Atlantic: Caribbean-North Atlantic, Western South Atlantic (Brazil) and Eastern Tropical Atlantic (West Africa). Within each region, populations have similar allele frequencies and levels of genetic diversity; indeed, no significant differentiation was found between populations separated by as much as 3000 km, suggesting that this coral species has the ability to disperse over large distances. Gene flow within regions does not, however, translate into connectivity across the entire Atlantic. Instead, substantial differences in allele frequencies across regions suggest that genetic exchange is infrequent between the Caribbean, Brazil and West Africa. Furthermore, markedly lower levels of genetic diversity are observed in the Brazilian and West African populations. Genetic diversity and connectivity may contribute to the resilience of a coral population to disturbance. Isolated peripheral populations may be more vulnerable to human impacts, disease or climate change relative to those in the genetically diverse Caribbean-North Atlantic region.

Recent genetic work on various coral genera has shown that morphological convergence between Atlantic and Pacific corals obscures evolutionary relationships and inferred levels of endemicity between the regions. Based on DNA sequences from nuclear and mitochondrial loci that provide higher resolution than those previously presented, this study shows that relationships within parts of the Atlantic coral fauna are also in need of substantial revision. The data presented here indicate that (1) the endemic Brazilian genus Mussismilia is a monophyletic clade, (2) Mussismilia is more closely related to the Caribbean Faviidae than Mussidae, the family in which it is currently placed, (3) the Brazilian endemic coral Favia leptophylla is much more closely related to Mussismilia than other species of Favia and has most likely been incorrectly placed in the genus Favia and (4) the other endemic Favia species found in Brazil, Favia gravida, is genetically distinct from Favia fragum, a Caribbean congener with which it is frequently synonymized. The nuclear data also suggest the possible presence of a cryptic species within Mussismilia, but additional sampling and morphological information is required to confirm this finding.

We investigated how ocean acidification in combination with warming affected D-veliger larvae of the Pacific oyster Crassostrea gigas. Larvae were reared for 40 h under either control (pH 8.1, 20 degrees C), acidified (pH 7.9, 20 degrees C), warm (pH 8.1, 22 degrees C) or warm acidified (pH 7.9, 22 degrees C) conditions. Larvae in acidified conditions were significantly smaller than in the control, but warm acidified conditions mitigated negative effects on size, and increased calcification. A proteomic approach employing two-dimensional electrophoresis (2-DE) was used to quantify proteins and relate their abundance to phenotypic traits. In total 12 differentially abundant spots were identified by nano-liquid chromatography-tandem mass spectrometry. These proteins had roles in metabolism, intra- and extra-cellular matrix formations, stress response, and as molecular chaperones. Seven spots responded to reduced pH, four to increased temperature, and six to acidification and warming. Reduced abundance of proteins such as ATP synthase and GAPDH, and increased abundance of superoxide dismutase, occurred when both pH and temperature changes were imposed, suggesting altered metabolism and enhanced oxidative stress. These results identify key proteins that may be involved in the acclimation of C gigas larvae to ocean acidification and warming.Significance: Increasing atmospheric CO2 raises sea surface temperatures and results in ocean acidification, two climatic variables known to impact marine organisms. Larvae of calcifying species may be particularly at risk to such changing environmental conditions. The Pacific oyster Crassostrea gigas is ecologically and commercially important, and understanding its ability to acclimate to climate change will help to predict how aquaculture of this species is likely to be impacted. Modest, yet realistic changes in pH and/or temperature may be more informative of how populations will respond to contemporary climate change. We showed that concurrent acidification and warming mitigates the negative effects of pH alone on size of larvae, but proteomic analysis reveals altered patterns of metabolism and an increase in oxidative stress suggesting non-additive effects of the interaction between pH and temperature on protein abundance. Thus, even small changes in climate may influence development, with potential consequences later in life.

International audience; : The deep reef refugia hypothesis proposes that deep reefs can act as local recruitment sources for shallow reefs following disturbance. To test this hypothesis, nine polymorphic DNA microsatellite loci were developed and used to assess vertical connectivity in 583 coral colonies of the Caribbean depth-generalist coral Montastraea cavernosa. Samples were collected from three depth zones (≤10, 15-20 and ≥25 m) at sites in Florida (within the Upper Keys, Lower Keys and Dry Tortugas), Bermuda, and the U.S. Virgin Islands. Migration rates were estimated to determine the probability of coral larval migration from shallow to deep and from deep to shallow. Finally, algal symbiont (Symbiodinium spp.) diversity and distribution were assessed in a subset of corals to test whether symbiont depth zonation might indicate limited vertical connectivity. Overall, analyses revealed significant genetic differentiation by depth in Florida, but not in Bermuda or the U.S. Virgin Islands, despite high levels of horizontal connectivity between these geographic locations at shallow depths. Within Florida, greater vertical connectivity was observed in the Dry Tortugas compared to the Lower or Upper Keys. However, at all sites, and regardless of the extent of vertical connectivity, migration occurred asymmetrically, with greater likelihood of migration from shallow to intermediate/deep habitats. Finally, most colonies hosted a single Symbiodinium type (C3), ruling out symbiont depth zonation of the dominant symbiont type as a structuring factor. Together, these findings suggest that the potential for shallow reefs to recover from deep-water refugia in M. cavernosa is location-specific, varying among and within geographic locations likely as a consequence of local hydrology.

Among Atlantic scleractinian corals, species diversity is highest in the Caribbean, but low diversity and high endemism are observed in various peripheral populations in central and eastern Atlantic islands and along the coasts of Brazil and West Africa. The degree of connectivity between these distantly separated populations is of interest because it provides insight into processes at both evolutionary and ecological time scales, such as speciation, recruitment dynamics and the persistence of coral populations. To assess connectivity in broadly distributed coral species of the Atlantic, DNA sequence data from two nuclear markers were obtained for six coral species spanning their distributional ranges. At basin-wide scales, significant differentiation was generally observed among populations in the Caribbean, Brazil and West Africa. Concordance of patterns in connectivity among co-distributed taxa indicates that extrinsic barriers, such as the Amazon freshwater plume or long stretches of open ocean, restrict dispersal of coral larvae from region to region. Within regions, dispersal ability appears to be influenced by aspects of reproduction and life history. Two broadcasting species, Siderastrea siderea and Montastraea cavernosa, were able to maintain gene flow among populations separated by as much as 1,200 km along the coast of Brazil. In contrast, brooding species, such as Favia gravida and Siderastrea radians, had more restricted gene flow along the Brazilian coast.

A global warming event that took place 55 million years ago at the end of the Palaeocene epoch is providing a picture of how Earth responds to climate change. The rapid rise in temperature was accompanied by turnovers in marine and terrestrial biota and changes in ocean chemistry and circulation. A study of carbon isotope records reveals a switch in the location of deep-water formation from the Southern Hemisphere to the Northern Hemisphere that was established within a few thousand years, but may have lasted for at least 40,000 years. This shows how greenhouse conditions can trigger quite rapid changes in deep ocean circulation that take much longer to be reversed. An exceptional analogue for the study of the causes and consequences of global warming occurs at the Palaeocene/Eocene Thermal Maximum, 55 million years ago. A rapid rise of global temperatures during this event accompanied turnovers in both marine1,2,3 and terrestrial biota4, as well as significant changes in ocean chemistry5,6 and circulation7,8. Here we present evidence for an abrupt shift in deep-ocean circulation using carbon isotope records from fourteen sites. These records indicate that deep-ocean circulation patterns changed from Southern Hemisphere overturning to Northern Hemisphere overturning at the start of the Palaeocene/Eocene Thermal Maximum. This shift in the location of deep-water formation persisted for at least 40,000 years, but eventually recovered to original circulation patterns. These results corroborate climate model inferences that a shift in deep-ocean circulation would deliver relatively warmer waters to the deep sea, thus producing further warming9. Greenhouse conditions can thus initiate abrupt deep-ocean circulation changes in less than a few thousand years, but may have lasting effects; in this case taking 100,000 years to revert to background conditions.
