Within the herein presented research, we studied the applicability of flax fabrics for composite parts in personal watercrafts in order to enhance damping of vibrations from the engine and noise reduction (which is relatively high for contemporary carbon constructions). Since the composite parts are intended to be exposed to humid environments requiring high levels of mechanical properties, a carbon–flax composite was selected. Samples of carbon, fiberglass, flax, and hybrid carbon–flax twill and biax fabrics were subjected to tensile and three-point bending tests. The mechanical properties were also tested after exposure of the samples to a humid environment. Damping was assessed by vibration and noise measurements directly on the complete float for samples as well as real parts. The hybrid carbon–flax material exhibited lower values of tensile strength than the carbon material (760 MPa compared to 463 MPa), but, at the same time, significantly higher than the other tested materials, or flax itself (115 MPa for a twill fabric). A similar trend in the results was observed for the three-point bending tests. Vibration tests and noise measurements showed reductions in vibration amplitude and frequency when using the carbon–flax hybrid material; the frequency response function for the watercraft part assembled from the hybrid material was 50% lower than for that made of carbon. Testing of samples located in a humid environment showed the necessity of surface treatment to prevent moisture absorption (mechanical properties were reduced at minimum by 28%). The tests confirmed that the hybrid material is satisfactory in terms of strength and its contribution to noise and vibration damping.

Purpose: Titanium dioxide nanoparticles, 25 nm in size of crystallites (TiO2 P25), are among the most produced nanomaterials worldwide. The broad use of TiO2 P25 in material science has implied a request to evaluate their biological effects, especially in the lungs. Hence, the pulmonary A549 cell line has been used to estimate the effects of TiO2 P25. However, the reports have provided dissimilar results on caused toxicity. Surprisingly, the physicochemical factors influencing TiO2 P25 action in biological models have not been evaluated in most reports. Thus, the objective of the present study is to characterize the preparation of TiO2 P25 for biological testing in A549 cells and to evaluate their biological effects.Methods: We determined the size and crystallinity of TiO2 P25. We used four techniques for TiO2 P25 dispersion. We estimated the colloid stability of TiO2 P25 in distilled water, isotonic NaCl solution, and cell culture medium. We applied the optimal dispersion conditions for testing the biological effects of TiO2 P25 (0-100 mu g.mL-1) in A549 cells using biochemical assays (dehydrogenase activity, glutathione levels) and microscopy.Results: We found that the use of fetal bovine serum in culture medium is essential to maintain sufficient colloid stability of dispersed TiO2 P25. Under these conditions, TiO2 P25 were unable to induce a significant impairment of A549 cells according to the results of biochemical and microscopy evaluations. When the defined parameters for the use of TiO2 P25 in A549 cells were met, similar results on the biological effects of TiO2 P25 were obtained in two independent cell laboratories.Conclusion: We optimized the experimental conditions of TiO2 P25 preparation for toxicity testing in A549 cells. The results presented here on TiO2 P25-induced cellular effects are reproducible. Therefore, our results can be helpful for other researchers using TiO2 P25 as a reference material.

Wire electrical discharge machining (WEDM) is an unconventional and very efficient technology for precision machining of the Nimonic C 263 super alloy, which is very widespread, especially in the energy, aerospace and automotive industries. Due to electrical discharge, defects in the form of cracks or burned cavities often occur on the machined surfaces, which negatively affect the correct functionality and service life of the manufactured components. To increase the efficiency of the machining of Nimonic C 263 using WEDM, in this study, extensive design of experiments was carried out, monitoring input factors in the form of machine parameters like Pulse off time, Gap voltage, Discharge current, Pulse on time and Wire feed, the output of which was comprehensive information about the behaviour of such machined surfaces, which allowed the optimization of the entire machining process. Thus, the optimization of the Cutting speed was performed in relation to the quality of the machined surface and the machining accuracy, as well as an analysis of the chemical composition of the machined surfaces and a detailed analysis of the lamella using a transmission electron microscope. A detailed study of the occurrence of surface or subsurface defects was also included. It was found that with the help of complex optimization tools, it is possible to significantly increase the efficiency of the machining of the Nimonic C 263 super alloy and achieve both financial savings in the form of shortened machine time and increasing the quality of machined surfaces., Elektroerozivní drátové řezání (WEDM) je nekonvenční a velmi efektivní technologie pro precizní obrábění super slitiny Nimonic C 263, která je velmi rozšířená především v energetickém, leteckém a automobilním průmyslu. Vlivem elektroeroze často vznikají na obrobených površích defekty v podobě trhlin či vypálených kavit, které negativně ovlivňují správnou funkčnost a očekávanou životnost vyrobených součástí. Za účelem zefektivnění obrábění Nimonic C 263 pomocí WEDM byl v rámci této studie proveden rozsáhlý plánovaný experiment, při kterém byly sledovány vstupní faktory v podobě parametrů nastavení stroje Pulse off time, Gap voltage, Discharge current, Pulse on time a Wire feed, jehož výstupem bylo získání komplexních informací o chování takto obrobených povrchů, což umožnilo optimalizaci celého obráběcího procesu. Byla tedy provedena optimalizace rychlosti řezání v návaznosti na jakost obrobeného povrchu a přesnost obrábění, dále byla provedena analýza chemického složení obrobených povrchů a také detailní analýza lamely pomocí transmisního elektronového mikroskopu. Součástí byla i detailní studie výskytu povrchových či podpovrchových defektů. Bylo zjištěno, že pomocí komplexních optimalizačních nástrojů je možné obrábění super slitiny Nimonic C 263 výrazně zefektivnit a dosáhnout tak jak finanční úspory v podobě zkrácení strojního času tak zvýšení jakosti obráběných povrchů.

Polycyklické aromatické uhlovodíky (PAU) představují skupinu významných kontaminantů pracovního i životního prostředí. Při expozici PAU v pracovním prostředí se vedle inhalační expoziční cesty může významně uplatňovat i expozice dermální. Stávající experimentální údaje o intenzitě a rychlosti penetrace látek do systémové cirkulace jsou zatím omezené. Předkládaný článek se zabývá metodickou a interpretační problematikou transepidermální absorpce PAU in vitro. Byla sledována intenzita (Flux) a rychlost (Lag time) penetrace naftalenu, fenanthrenu, pyrenu a benzo[a]pyrenu přes epidermální membránu z prasečího boltce. Experiment byl prováděn ve vertikálních statických difuzních komůrkách dle Franze (n = 32) a jako rozpouštědlo byl použit isopropyl-myristát. Parametr Flux (nmol/cm2/hod) dosáhl hodnot 95,7 ± 45,5 u naftalenu, 19,5 ± 8,7 u fenanthrenu, 4,38 ± 1,98 u pyrenu a 0,21 ± 0,08 u benzo[a]pyrenu. Parametr Lag time (hod) hodnot 0,26 ± 0,17 u naftalenu, 2,12 ± 0,41 u fenanthrenu, 3,25 ± 0,50 u pyrenu a 11,2 ± 4,08 u benzo[a]pyrenu. Hodnota parametru Flux klesala s molární hmotností PAU, zatímco hodnota parametru Lag time s molární hmotností PAU stoupala. Množství PAU, které za daný časový úsek penetrovalo přes epidermální membránu, se pohybovalo mezi 0,24 % (benzo[a]pyren) až 0,84 % (fenanthren) aplikované dávky. Penetrace PAU přes epidermální membránu vykazovala, v porovnání s experimenty na plné kůži, nižší stupeň variability dat. Z výsledků vyplývá, že použití epidermální membrány by mohlo zpřesňovat jak odhad vnitřní dávky PAU po dermální expozici, tak i odhad souvisejícího zdravotního rizika v rámci konzervativního expozičního scénáře. Experimenty s epidermální membránou jsou však časově i experimentálně náročné, bez možnosti objektivní kontroly integrity epidermální membrány, což může vést k finanční náročnosti testování, ztrátám vzorků a v neposlední řadě i k nárůstu rozdílů hodnot dat, získaných v různých laboratořích., Polycyclic aromatic hydrocarbons (PAHs) are a group of significant contaminants in the occupational and living environment. In addition to the inhalation route of exposure in the occupational environment, there is also significant the dermal route of exposure. Existing experimental data on the intensity and rate of penetration of these substances into systemic circulation are still limited. The presented paper is focused on methodological and interpretative issues of trans-epidermal absorption of PAHs in vitro. In this study, we assessed the intensity (Flux) and rate (Lag time) of penetration of naphthalene, phenanthrene, pyrene and benzo[a]pyrene through the epidermal membrane derived from a pig ear. The experiment was performed using static vertical Franz diffusion cells (n = 32) and isopropyl myristate was used as a solvent. Flux (nmol/cm2/hour) reached 95.7 ± 45.5 in the case of naphthalene, 19.5 ± 8.7 of phenanthrene, 4.38 ± 1.98 of pyrene and 0.21 ± 0.08 of benzo[a]pyrene. Lag time (hour) was 0.26 ± 0.17 in the case of naphthalene, 2.12 ± 0.41 of phenanthrene, 3.25 ± 0.50 of pyrene and 11.2 ± 4.08 of benzo[a]pyrene. The Flux value decreased with the molecular weight of PAHs, while the Lag time increased with the molecular weight of PAHs. The amount of PAHs that penetrated through the epidermal membrane in a given time period ranged between 0.24% (benzo[a]pyrene) and 0.84% (phenanthrene) of the applied dose. The penetration of PAHs through the epidermal membrane showed a lower degree of data variability compared to full thickness skin experiments. The results suggest that the use of the epidermal membrane could define more accurately both the estimation of the internal dose of PAHs after dermal exposure and the estimation of the associated health risk within a conservative exposure scenario.
