9. Alzheimer disease macrophages shuttle amyloid-beta from neurons to vessels, contributing to amyloid angiopathy

Abstract

Neuronal accumulation of oligomeric amyloid-β (αβ) is considered the proximal cause of neuronal demise in Alzheimer disease (AD) patients. Blood-borne macrophages might reduce Aβ stress to neurons by immigration into the brain and phagocytosis of αβ. We tested migration and export across a blood-brain barrier model, and phagocytosis and clearance of αβ by AD and normal subjects' macrophages. Both AD and normal macrophages were inhibited in αβ export across the blood-brain barrier due to adherence of Aβ-engorged macrophages to the endothelial layer. In comparison to normal subjects' macrophages, AD macrophages ingested and cleared less αβ, and underwent apoptosis upon exposure to soluble, protofibrillar, or fibrillar αβ. Confocal microscopy of stained AD brain sections revealed oligomeric Aβ in neurons and apoptotic macrophages, which surrounded and infiltrated congophilic microvessels, and fibrillar Aβ in plaques and microvessel walls. After incubation with AD brain sections, normal subjects' monocytes intruded into neurons and uploaded oligomeric Aβ. In conclusion, in patients with AD, macrophages appear to shuttle Aβ from neurons to vessels where their apoptosis may release fibrillar Aβ, contributing to cerebral amyloid angiopathy. © Springer-Verlag 2008.

Abstract

The use of renewable vegetable oils derived from oilseed crops could have serious environmental impacts especially regarding competition for land and emissions in air and water. The environmental performances of Linum usitatissimum L. (Flax) and Camelina sativa L. (Camelina) oilseed crops for biodiesel production has been assessed by means of Life Cycle Assessment (LCA) approach, considering three steps: i) the cultivation, ii) oil seeds transport to pressing plant and seed pressing, iii) biodiesel production from vegetable oil by transesterification. For the biodiesel from flax and camelina, more than 90% of the environmental impact is related to seed production, followed by the transesterification of raw vegetable oil and seed pressing at lab scale. The environmental performances are worst for the camelina mainly due to a lower seed yield. Nevertheless, these differences are slightly reduced thanks to the higher HHV (Higher Heating Values) of the camelina biodiesel. For both the biodiesel, the main environmental hotspots are: the production of factors production, the nitrogen emissions associated with the application of fertiliser and the mechanisation of the field operations (in particular soil tillage and sowing) and the emission of N and P compounds related to fertilisers application. In comparison with rapeseed (from data of Ecoinvent 3), the biodiesel from flax and camelina shows a higher environmental impact due to the higher consumption of fertilizers in rapeseed crop management.

Abstract

Composting plants combine compost and energy production using the Anaerobic Digestion (AD). However, the AD process generates residue whose liquid fraction is polluting and therefore needs to be subjected to expensive waste treatments with the loss of the value of the mineral elements. Therefore, it is necessary to develop technologies allowing the treatment of liquid digestate fraction and the uptake of those minerals as plant nutrients. The purpose of the project DUCK-TECH has been to provide a solution through an integrated biological system, i.e., an aquatic plant treatment system (APTS). An APTS for the liquid digestate fraction treatment has been implemented using duckweed Lemnaceae aquatic plant species, taking advantage of their ability to recover nitrogen and phosphorous and to reduce COD. The aim of this study is to evaluate the phytoremediation and environmental performances of the APTS based on duckweed. The preliminary results highlight that the environmental impact of liquid fraction treatment with APTS deeply ranges, depending on the duckweed growing. A further increase of the performances could be achieved through the valorisation of the duckweed biomass as a starch source for new bio-products.
