Preventative malaria control in DRC is reliant on pyrethroid-treated nets, raising concerns over the potential impacts of insecticide resistance. Introduction In the Democratic Republic of the Congo, the malaria transmission landscape is influenced by diverse bioclimatic conditions, leading to varied transmission patterns across the country. Purpose The primary objective of this study was to investigate the bionomics of malaria vectors across 22 NMCP sentinel sites. By analyzing the different malaria transmission facies, we aimed to provide detailed insights into vector behavior and abundance to inform more effective malaria control strategies. Methods Mosquitoes were captured at 22 sentinel sites, with sampling conducted in 20 randomly selected households per site. These households were divided into two groups: ten utilized human bait traps, and the remaining ten employed pyrethrum spray catches. The captures were performed over five consecutive nights every six months. A total of 1,723 Anopheles mosquitoes were collected for analysis. Results The study identified five Anopheles species, with Anopheles gambiae s.l. being the most abundant and primary vector, representing 75% of the total captures. The other species included An. funestus group (15%), An. paludis (12%), An. coustani (0.3%), and An. nili (0.2%). Behavioral observations revealed that An. gambiae s.l. is active both indoors and outdoors, with peak biting occurring from 9 p.m. to 4 a.m., and an average biting density of 12 bites per person per night. 13209 mosquitoes (i.e., 88.5%) were captured using the HLC method, while, among the 1,723 mosquitoes captured using the PSC method, 51% were blood-fed, 24% were semi-gravid, 18% were gravid, and 7% were fasting. An. gambiae s.l. was found to be present year-round, contributing to continuous malaria transmission. Conclusion This study provides crucial data on the prevalence and behavior of malaria vectors, particularly emphasizing the role of Anopheles gambiae s.l. as the primary vector in malaria transmission. The findings highlight significant differences in bite rates among species and the year-round activity of An. gambiae s.l., underscoring the need for targeted vector control measures. The data will be used to refine and implement effective vector control strategies, such as long-lasting insecticide-treated nets (LLINs) and indoor residual spraying (IRS), to reduce malaria transmission and mortality rates at NMCP sentinel sites.

Arbovirus epidemics (chikungunya, dengue, West Nile fever, yellow fever and zika) are a growing threat in African areas where Aedes (Stegomyia) aegypti (Linnaeus, 1762) and Aedes albopictus (Skuse, 1895) are present. The lack of comprehensive sampling of these two vectors limits our understanding of their propagation dynamics in areas at risk of arboviruses. Here, we collected 6,943 observations (both larval and human capture) of Ae. aegypti and Ae. albopictus between 2020 and 2022. The study was carried out in the Vallee de la Funa, a post-epidemic zone in the city of Kinshasa, Democratic Republic of Congo. Our results provide important information for future basic and advanced studies on the ecology and phenology of these vectors, as well as on vector dynamics after a post-epidemic period. The data from this study are published in the public domain as the Darwin Core Archive in the Global Biodiversity Information Facility.

Abstract Background Resistance in malaria vectors to pyrethroids, the most widely used class of insecticides for malaria vector control, threatens the continued efficacy of vector control tools. Target-site resistance is an important genetic resistance mechanism caused by mutations in the voltage-gated sodium channel (Vgsc) gene that encodes the pyrethroid target-site. Understanding the geographic distribution of target-site resistance, and temporal trends across different vector species, can inform strategic deployment of vector control tools. Results We develop a Bayesian statistical spatiotemporal model to interpret species-specific trends in the frequency of the most common resistance mutations, Vgsc-995S and Vgsc-995F, in three major malaria vector species Anopheles gambiae, An. coluzzii, and An. arabiensis over the period 2005–2017. The models are informed by 2418 observations of the frequency of each mutation in field sampled mosquitoes collected from 27 countries spanning western and eastern regions of Africa. For nine selected countries, we develop annual predictive maps which reveal geographically structured patterns of spread of each mutation at regional and continental scales. The results show associations, as well as stark differences, in spread dynamics of the two mutations across the three vector species. The coverage of ITNs was an influential predictor of Vgsc allele frequencies, with modelled relationships between ITN coverage and allele frequencies varying across species and geographic regions. We found that our mapped Vgsc allele frequencies are a significant partial predictor of phenotypic resistance to the pyrethroid deltamethrin in An. gambiae complex populations. Conclusions Our predictive maps show how spatiotemporal trends in insecticide target-site resistance mechanisms in African An. gambiae vary across individual vector species and geographic regions. Molecular surveillance of resistance mechanisms will help to predict resistance phenotypes and track their spread.

Abstract Background Between 2011 and 2018, an estimated 134.8 million pyrethroid-treated long-lasting insecticidal nets (LLINs) were distributed nationwide in the Democratic Republic of Congo (DRC) for malaria control. Pyrethroid resistance has developed in DRC in recent years, but the intensity of resistance and impact on LLIN efficacy was not known. Therefore, the intensity of resistance of Anopheles gambiae sensu lato (s.l.) to permethrin and deltamethrin was monitored before and after a mass distribution of LLINs in Kinshasa in December 2016, and in 6 other sites across the country in 2017 and 11 sites in 2018. Methods In Kinshasa, CDC bottle bioassays using 1, 2, 5, and 10 times the diagnostic dose of permethrin and deltamethrin were conducted using An. gambiae s.l. collected as larvae and reared to adults. Bioassays were conducted in four sites in Kinshasa province 6 months before a mass distribution of deltamethrin-treated LLINs and then two, six, and 10 months after the distribution. One site in neighbouring Kongo Central province was used as a control (no mass campaign of LLIN distribution during the study). Nationwide intensity assays were conducted in six sites in 2017 using CDC bottle bioassays and in 11 sites in 2018 using WHO intensity assays. A sub-sample of An. gambiae s.l. was tested by PCR to determine species composition and frequency of kdr-1014F and 1014S alleles. Results In June 2016, before LLIN distribution, permethrin resistance intensity was high in Kinshasa; the mean mortality rate was 43% at the 5× concentration and 73% at the 10× concentration. Bioassays at 3 time points after LLIN distribution showed considerable variation by site and time and there was no consistent evidence for an increase in pyrethroid resistance intensity compared to the neighbouring control site. Tests of An. gambiae s.l. in 6 sites across the country in 2017 and 11 sites in 2018 showed all populations were resistant to the diagnostic doses of 3 pyrethroids. In 2018, the intensity of resistance varied by site, but was generally moderate for all three pyrethroids, with survivors at ×5 the diagnostic dose. Anopheles gambiae sensu stricto (s.s.) was the most common species identified across 11 sites in DRC, but in Kinshasa, An. gambiae s.s. (91%) and Anopheles coluzzii (8%) were sympatric. Conclusions Moderate or high intensity pyrethroid resistance was detected nationwide in DRC and is a serious threat to sustained malaria control with pyrethroid LLINs. Next generation nets (PBO nets or bi-treated nets) should be considered for mass distribution.

BackgroundProspective malaria public health interventions are initially tested for entomological impact using standardised experimental hut trials. In some cases, data are collated as aggregated counts of potential outcomes from mosquito feeding attempts given the presence of an insecticidal intervention. Comprehensive data i.e. full breakdowns of probable outcomes of mosquito feeding attempts, are more rarely available. Bayesian evidence synthesis is a framework that explicitly combines data sources to enable the joint estimation of parameters and their uncertainties. The aggregated and comprehensive data can be combined using an evidence synthesis approach to enhance our inference about the potential impact of vector control products across different settings over time.MethodsAggregated and comprehensive data from a meta-analysis of the impact of Pirimiphos-methyl, an indoor residual spray (IRS) product active ingredient, used on wall surfaces to kill mosquitoes and reduce malaria transmission, were analysed using a series of statistical models to understand the benefits and limitations of each.ResultsMany more data are available in aggregated format (N = 23 datasets, 4 studies) relative to comprehensive format (N = 2 datasets, 1 study). The evidence synthesis model had the smallest uncertainty at predicting the probability of mosquitoes dying or surviving and blood-feeding. Generating odds ratios from the correlated Bernoulli random sample indicates that when mortality and blood-feeding are positively correlated, as exhibited in our data, the number of successfully fed mosquitoes will be under-estimated. Analysis of either dataset alone is problematic because aggregated data require an assumption of independence and there are few and variable data in the comprehensive format.ConclusionsWe developed an approach to combine sources from trials to maximise the inference that can be made from such data and that is applicable to other systems. Bayesian evidence synthesis enables inference from multiple datasets simultaneously to give a more informative result and highlight conflicts between sources. Advantages and limitations of these models are discussed.

Abstract Background The current study shows the results of three years of IRS entomological monitoring (2016, before intervention; 2017 and 2018, after intervention) performed in Alibori and Donga, northern Benin. Methods Mosquito collections were performed on a monthly basis using human landing catches and pyrethrum spray catches in six districts including four treated with Actellic 300 CS (Kandi, Gogounou, Djougou and Copargo) and two untreated (Bembèrèkè and Kouandé) which served as control sites. Key transmission indicators of Anopheles gambiae (s.l.) as well as the residual activity of Actellic 300 CS assessed through WHO cone tests, were determined. Results The residual efficacy duration of Actellic 300 CS after the two IRS campaigns (2017 and 2018) was 4–5 months (May–September). Abstract Background This study aims to provide baseline data on the resistance status to insecticides, the frequency of mechanisms involved and the impact of the association with the synergist piperonyl butoxide (PBO) on resistant Anopheles gambiae (s.l.) populations in two regions of northern Benin, prior to an indoor residual spraying campaign and introduction of next generation long-lasting insecticidal nets (LLINs) incorporating PBO. Methods Adult Anopheles gambiae (s.l.) originating from larvae collected in two study regions (Alibori within the Kandi-Gogounou-Segbana districts and Donga within the Djougou-Copargo-Ouake districts) were tested with impregnated papers (bendiocarb 0.1%, pirimiphos-methyl 0.25%, permethrin 0.75% and deltamethrin 0.05%). The synergist PBO was used to check for the involvement of detoxification enzymes in pyrethroid resistant populations. Molecular analyses were performed for the identification of species within the Anopheles gambiae (s.l.) complex and kdr L1014F and G119S Ace-1 mutations. Biochemical assays assessed the activity of detoxification enzymes. Results Anopheles gambiae (s.l.) was resistant to pyrethroids, with a mortality range of 25–83% with deltamethrin and 6–55% with permethrin. A significant increase in mortality was observed after pre-exposure to PBO for both deltamethrin (63–99%) and permethrin (56–99%). With bendiocarb, An. gambiae (s.l.) were susceptible in Kandi (99% mortality), with possible resistance (92–95%) recorded in Djougou, Copargo, Gogounou, Ouake and Segbana. All study populations were fully susceptible to pirimiphos-methyl. The frequencies of resistant mutations varied according to species and sites: 0.67–0.88 for L1014F kdr and 0–0.06 for G119S Ace-1. Three study locations (Djougou, Gogounou and Kandi) showed high oxidase activity and four sites (Djougou, Ouake, Copargo and Kandi) showed elevated esterase activity. Conclusions This study confirms resistance to pyrethroids and suggests emerging bendiocarb resistance in An. gambiae (s.l.) populations in northern Benin. However, recovery of susceptibility to pyrethroids after PBO exposure, and susceptibility to organophosphates in the An. gambiae (s.l.) populations indicate that next generation LLINs incorporating PBO synergist combined with an indoor residual spraying (IRS) campaign with organophosphate insecticides may be regarded as alternative control tools.

Indoor residual spraying is a commonly used method for mosquito, and malaria, control and there are a number of available insecticides that are available for this. Here, the authors evaluate the efficacy of widely-used and novel insecticides against pyrethroid-resistant mosquitoes.

Abstract Background Polovodova method based on counting follicular dilatations estimates the number of egg-laying in mosquitoes. However, some researchers doubt the reliability of this method because of the absence of multiple dilatations in vectors after many gonotrophic cycles. It is in this context of controversy that our study was carried out to evaluate the importance of follicular dilatations in the determination of parity levels in An. gambiae s.s. Moreover, the application of this method allowed us to clarify the evolution of vectors’ infectivity to P. falciparum according to their parity level. Methods We used two techniques to determine the parity level in An. gambiae s.s. We used two batches of wild strain mosquitoes reproduced after a known number of egg-laying in laboratory. The first batch was submitted to oil injection in the ovaries using a micropipette. In the same way, the classic technique of ovaries dilaceration (a technique based on the Polovodova method) was applied to the second batch. In order to assess relationship between parity level and mosquitoes’ infectivity, Polovodova method was applied on vectors collected on humans. Finally, Heads and thoraces of these vectors were individually analyzed for P. falciparum antigen detection using an ELISA assay. Results In the first batch including 50 female mosquitoes “never laid”, 50 “laid once”, 50 “laid twice” and 48 “three times”, oil injection technique revealed 42 nulliparous, 44 uniparous, 46 biparous and 44 triparous respectively. Overall, Polovodova method was effective using oil injection technique (p > 0.05). However, in rural Kandi (Sonsoro) in northeastern Benin, several malaria cases are often diagnosed during dry seasons. The underlying factors accounting for this phenomenon remain unknown. Methods The entomological profile of Sonsoro has been studied compared to a location in urban Kandi (Gansosso) for a period of one year. During this period, Anopheles larval habitats were investigated and populations of Anopheles gambiae s.l. were sampled by human landing catches in both areas. Enzyme-linked immunosorbent assays (ELISA) for Plasmodium falciparum circumsporozoite protein (CSP) were conducted on vector specimens and the entomological inoculation rates (EIR) were determined per season (wet versus dry) in each area. In addition, during the severe drought period, Rapid Diagnostic Tests (RDTs) were conducted on school children under the age 10 years in these areas to provide a global view of drought-malaria prevalence and to perform a crossing with entomological data in Kandi. Results Overall, An. gambiae s.l. was particularly abundant in rural Kandi compared to the urban area with a significant decrease of vector density in both sites during the dry season. In this period, larval sampling data identified household water sources as potential breeding sites in urban and rural Kandi. We also observed a significant seasonal variation of the infectivity rate in both areas but for each period (season), the EIR was higher in the rural site than in the urban. Data of P. falciparum detection was the reflection of entomological findings. The droughtmalaria prevalence was 5.5 times higher in rural Kandi as compared to urban Kandi. The presence of a permanent water site and the low level of urbanization in rural Kandi were identified as a risk factor. Conclusion Our data showed a high level of malaria transmission in the municipality of Kandi. Household water source plays an important role in maintaining the breeding of anopheles larvae and the malaria transmission in Kandi.
