Light use efficiency characterizes the ability of a crop to convert radiation into biomass. Determining optimum cultivar-specific photosynthetic photon flux density (PPFD) values from sole-source lighting can be used to optimize leaf expansion, maximize biomass, and shorten the production period. This study evaluated the growth of hydroponic lettuce (Lactuca sativa) 'Rex' cultivated under different PPFD levels using sole-source lighting. At lower PPFD levels of 201 to 292 µmol·m−2·s−1, the plant projected canopy size (PCS) and specific leaf area increased to enhance light capture by 36.2% as compared to higher PPFD levels (333 and 413 µmol·m−2·s−1), while plants exhibited 10.3% lower canopy overlap ratio and 27.8% lower shoot dry weights. Both low and high PPFD conditions lead to a similar trend in PCS among plants. Light use efficiency was not a major factor in influencing lettuce growth. Instead, the critical factor was the total incident light the plants received. In a production environment, delayed stress recognition can impact yield. Imaging can rapidly and effectively quantify stress symptoms using indexes such as normalized difference vegetation index (NDVI). Commercial systems are effective but cannot be easily customized for specific applications, particularly post-processing. We developed a low-cost customizable imaging system and validated the code to analyze images. Our objective was to verify the image analysis code and custom system could successfully quantify the changes in plant canopy reflectance. 'Supercascade Red', 'Wave© Purple', and 'Carpet Blue' Petunias (Petunia × hybridia) were transplanted individually and subjected to increasing fertilizer treatments and increasing substrate pH in a greenhouse. Treatments for the first trial were the addition of a controlled release fertilizer at six different rates (0, 0.5, 1, 2, 4, and 8 g/pot), and for the second trial, fertilizer solution with four pHs (4, 5.5, 7, and 8.5), with eight replications with one plant each. Plants were imaged twice a week using a commercial imaging system for fertilizer and thrice a week with the custom system for pH. The collected images were analyzed using an in-house program that calculated the indices for each pixel of the plant area. All cultivars showed a significant effect of fertilizer on the projected canopy size and dry weight of the above-substrate biomass and the fertilizer rate treatments (p < 0.01). Plant tissue nitrogen concentration as a function of the applied fertilizer rate showed a significant positive response for all three cultivars (p < 0.001). We verified that the image analysis code successfully quantified the changes in plant canopy reflectance as induced by increasing fertilizer application rate. There was no relationship between the pH and NDVI values for the cultivars tested (p > 0.05). Manganese and phosphorus had no significance with chlorophyll fluorescence for 'Carpet Blue' and 'Wave© Purple' (p > 0.05), though 'Supercascade Red' was found to have significance (p < 0.01). pH did not affect plant canopy size. Chlorophyll fluorescence pixel intensity against the projected canopy size had no significance except in 'Wave© Purple' (p = 0.005). NDVI as a function of the projected canopy size had no statistical significance. Canopy imaging offers a non-destructive, efficient way to objectively measure canopy size, detect stress symptoms, and assess pigment concentrations. While it is faster and easier than traditional destructive methods, manual image analysis, including segmentation and evaluation, can be time-consuming. To make imaging more widely accessible, it's essential to reduce the cost of imaging systems and automate the analysis process. We developed a low-cost imaging system with automated analysis using an embedded microcomputer equipped with a monochrome camera and a filter for a total hardware cost of ~USD 500. Our imaging system takes images under blue, green, red, and infrared light, as well as chlorophyll fluorescence. The system uses a Python-based program to collect and analyze images automatically. The multi-spectral imaging system separates plants from the background using a chlorophyll fluorescence image, which is also used to quantify canopy size. The system then generates normalized difference vegetation index (NDVI, "greenness") images and histograms, providing quantitative, spatially resolved information. We verified that these indices correlate with leaf chlorophyll content and can easily add other indices by installing light sources with the desired spectrums. Agriculture in controlled environments has gained popularity over time. Compared to traditional agriculture, controlled environments emerge as an alternative to mitigate the negative impacts of conventional farming methods. However, controlled environment agriculture, particularly plant factories with artificial lighting, incurs higher electricity costs, primarily for supplemental lighting and dehumidification of the cultivation area. Given these high costs, it is crucial to understand how efficiently plants utilize available light to convert it into biomass. This understanding can be used to design lighting strategies to reduce electricity usage. In this study, we cultivated 'Rex' lettuce (Lactuca sativa) plants on a soilless substrate and used an ebb-and-flow system for irrigation and fertilization. Plants grew in varying photosynthetic photon flux density (PPFD) levels ranging from 125 to 375 µmol·m−2·s−1 and were assessed for various physiological responses. Our findings revealed that plants exposed to higher light levels exhibited greater final dry weight, increased photosynthetic activity, higher water use efficiency, and accelerated growth compared to those under lower light conditions. Notably, plants subjected to higher light intensities did not show a significant increase in transpiration, suggesting a potential trade-off between energy expenditure on supplemental lighting and dehumidification. This finding opens the possibility of reducing energy consumption for dehumidification and achieving economic savings by subjecting plants to optimal growing conditions for shorter durations. In light of projected climate changes that are expected to disrupt crop yields in previously stable regions, it is essential to understand how these changes will affect food supplies. This study investigated the combined effect of light and temperature on the yield of wheat (Triticum aestivum L.) and rice (Oryza sativa L.) in a controlled environment. Three treatment combinations were tested: 30.2 mol.m−2.d−1 with a 17.5 °C average ("low"), 38.9 mol.m−2.d−1 and 22.5 °C ("medium"), and 47.5 mol.m−2.d−1 and 27.5 °C ("high"). The low treatment resulted in the highest wheat yield, with total dry biomasses of 233 g, 140 g, and 115 g for the low, medium, and high treatments. Rice showed the highest growth in the medium and high treatments, with total dry biomasses of 145 and 144 g. The medium and high treatment biomasses were not statistically significant for wheat or rice at 95% confidence. However, the wheat's edible biomass did significantly decrease between every treatment. These results demonstrate that wheat thrives in colder climates, even with less light available, while an increase in temperature will have either a positive or insignificant effect on rice within this study's temperature range. Global fertilizer production and mismanagement significantly contribute to many harmful environmental impacts, revealing the need for a greater understanding of crop growth and nutrient uptake, which can be used to optimize fertilizer management. This study experimentally adapts first-principles microbial modeling techniques to the hydroponic cultivation of Bibb lettuce (Lactuca sativa) under nitrogen-limited conditions. Monod and Michaelis–Menten's approaches are applied to predict biomass productivity and nutrient uptake and to evaluate the feasibility of reclaimed wastewater as a nutrient source of nitrogen. Experimental and modeling results reveal significantly different kinetic saturation constants ( K s = 1.331 and K m = 17.887 mg L−1) and a corresponding cell yield strongly dependent on nutrient concentration, producing visually and compositionally distinct tissue between treatments receiving ≤ 26.2 and ≥ 41.7 m g N L−1. The resulting Monod model overestimates dry mass predictions during low nutrient conditions, and the collective results support the development of a dynamic Monod curve that is temporally dependent during the plants' lifecycle. Despite this shortcoming, these results support the feasibility of reclaiming nitrogen from wastewater in hydroponic agriculture, expecting to produce lesser biomass lettuce exhibiting healthy tissue. Salinity stress is a common challenge in plant growth, impacting seed quality, germination, and general plant health. Sodium chloride (NaCl) ions disrupt membranes, causing ion leakage and reducing seed viability. Gibberellic acid (GA3) treatments have been found to promote germination and mitigate salinity stress on germination and plant growth. 'Bauer' and 'Muir' lettuce (Lactuca sativa) seeds were soaked in distilled water (control), 100 mM NaCl, 100 mM NaCl + 50 mg/L GA3, and 100 mM NaCl + 150 mg/L GA3 in Petri dishes and kept in a dark growth chamber at 25 °C for 24 h. After germination, seedlings were monitored using embedded cameras, capturing red, green, and blue (RGB) images from seeding to final harvest. Despite consistent germination rates, 'Bauer' seeds treated with NaCl showed reduced germination. Surprisingly, the 'Muir' cultivar's final dry weight differed across treatments, with the NaCl and high GA3 concentration combination yielding the poorest results (p < 0.05). This study highlights the efficacy of GA3 applications in improving germination rates. However, at elevated concentrations, it induced excessive hypocotyl elongation and pale seedlings, posing challenges for two-dimensional imaging. Nonetheless, a sigmoidal regression model using projected canopy size accurately predicted dry weight across growth stages and cultivars, emphasizing its reliability despite treatment variations (R2 = 0.96, RMSE = 0.11, p < 0.001). The decline in seed quality over time due to natural aging or mishandling requires assessing seed vigor for resilience in adverse conditions. Accelerated aging (AA) methods simulate seed deterioration by subjecting seeds to high temperatures and humidity. Saturated salt accelerated aging (SSAA) is an AA method adopted for small seeds like lettuce (Lactuca sativa). In this study, we subjected seeds of two lettuce cultivars ('Muir' and 'Bauer') to SSAA by sealing them in a box containing 40 g/100 mL of a sodium chloride (NaCl) solution in a dark growth chamber at 41 °C for 24, 48, and 72 h with a control. We monitored their vigor using embedded computer cameras, tracking the projected canopy size (PCS) daily from sowing to harvest. The cultivar 'Muir' exhibited consistent PCS values across the treatments, while 'Bauer' showed PCS variations, with notable declines after prolonged aging. The germination rates dropped significantly after 48 and 72 h of SSAA. A nonlinear regression model revealed a strong relationship between PCS and shoot dry weight across harvests and cultivars (R2 = 0.93, RMSE = 0.15, p < 0.001). The research found that the projected canopy size and shoot dry weight increased over time with significant differences in treatments for the cultivar 'Bauer' but not for 'Muir,' with the canopy size being a strong predictor of dry weight and no significant impact from the SSAA treatments. This study highlights cultivar-specific responses to aging and demonstrates the efficacy of our imaging tool in predicting lettuce dry weight despite treatment variations. The survival and growth of young plants hinge on various factors, such as seed quality and environmental conditions. Assessing seedling potential/vigor for a robust crop yield is crucial but often resource-intensive. This study explores cost-effective imaging techniques for rapid evaluation of seedling vigor, offering a practical solution to a common problem in agricultural research. In the first phase, nine lettuce (Lactuca sativa) cultivars were sown in trays and monitored using chlorophyll fluorescence imaging thrice weekly for two weeks. The second phase involved integrating embedded computers equipped with cameras for phenotyping. These systems captured and analyzed images four times daily, covering the entire growth cycle from seeding to harvest for four specific cultivars. All resulting data were promptly uploaded to the cloud, allowing for remote access and providing real-time information on plant performance. Results consistently showed the 'Muir' cultivar to have a larger canopy size and better germination, though 'Sparx' and 'Crispino' surpassed it in final dry weight. A non-linear model accurately predicted lettuce plant weight using seedling canopy size in the first study. The second study improved prediction accuracy with a sigmoidal growth curve from multiple harvests (R2 = 0.88, RMSE = 0.27, p < 0.001). Rockwool and peatmoss are commonly used substrates in the greenhouse industry due to their quality, stable pH, exceptional water retention properties and air porosity that is important for plant root development. Although rockwool is commonly used in deep water culture (DWC) hydroponic systems as the base support, there is a lack of studies comparing different types of substrates that could be used in DWC systems, especially considering the increasing market value and awareness of sustainable production in controlled environment agriculture. We identified 13 commercial substrate mixes with different compositions and conducted a series of studies in a DWC system in a greenhouse for three seasons to evaluate their effects on arugula 'Slow Bolt' (Eruca sativa L.) and lettuce 'Summer Crisp' (Lactuca sativa L.) growth, yield, and quality. The substrates tested significantly influenced the growth, yield, and quality of both arugula and lettuce. The average leaf fresh weight per plant could range from 44 to 190 g for arugula and 89 to 265 g for lettuce. The peat-based products outperformed the coir and other inorganic substrates (phenolic foam, rockwool). The substrate with 75% peat + 25% fine coir produced the greatest plant height, width, and biomass for arugula and lettuce over three growing seasons. Examining arugula and lettuce growth, the fall season produced plants with higher water and nutrient use efficiency, while plants grown during the winter had lower resource use efficiency. Summary: The mechanisms underlying plant tolerance to boron (B) excess are far from fully understood. Here we characterized the role of the miR397‐CsiLAC4/CsiLAC17 (from Citrus sinensis) module in regulation of B flow.Live‐cell imaging techniques were used in localization studies. A tobacco transient expression system tested modulations of CsiLAC4 and CsiLAC17 by miR397. Transgenic Arabidopsis were generated to analyze the biological functions of CsiLAC4 and CsiLAC17. CsiLAC4's role in xylem lignification was determined by mRNA hybridization and cytochemistry. In situ B distribution was analyzed by laser ablation inductively coupled plasma mass spectrometry.CsiLAC4 and CsiLAC17 are predominantly localized in the apoplast of tobacco epidermal cells. Overexpression of CsiLAC4 in Arabidopsis improves the plants' tolerance to boric acid excess by triggering high‐B‐dependent lignification of the vascular system's cell wall and reducing free B content in roots and shoots. In Citrus, CsiLAC4 is expressed explicitly in the xylem parenchyma and is modulated by B‐responsive miR397. Upregulation of CsiLAC4 in Citrus results in lignification of the xylem cell walls, restricting B flow from xylem vessels to the phloem.CsiLAC4 contributes to plant tolerance to boric acid excess via high‐B‐dependent lignification of cell walls, which set up a 'physical barrier' preventing B flow.
