23. Luminescent Heterobimetallic PtII–AuIComplexes Bearing N-Heterocyclic Carbenes (NHCs) as Potent Anticancer Agents

Abstract

This study aims to probe into new series of heterobimetallic PtII–AuIcomplexes with a general formula of [Pt(p-MeC6H4)(dfppy)(μ-dppm)Au(NHC)]OTf, NHC = IPr, 2; IMes, 3; dfppy = 2-(2,4-difluorophenyl)pyridinate; dppm = 1,1-bis(diphenylphosphino)methane, which are the resultant of the reaction between [Pt(p-MeC6H4)(dfppy)(κ1-dppm)], 1, with [AuCl(NHC)], NHC = IPr, B; IMes, C, in the presence of [Ag(OTf)]. In the heterobimetallic complexes, the dppm ligand is settled between both metals as an unsymmetrical bridging ligand. Several techniques are employed to characterize the resulting compounds. Moreover, the photophysical properties of the complexes are investigated by means of UV–vis and photoluminescence spectroscopy. Furthermore, the experimental study is enriched by ab initio calculations (density functional theory (DFT) and time-dependent DFT (TD-DFT)) to assess the role of Pt and Au moieties in the observed optical properties. It is revealed that 1–3is luminescent in the solid state and solution at different temperatures. In addition, the achieved results indicate the emissive properties of 1–3are originated from a mixed 3IL/3MLCT excited state with major contribution of intraligand charge transfer (dfppy). A comparative study is conducted into the cytotoxic activities of starting materials and 1–3against different human cancer cell lines such as the pancreas (MIA-PaCa2), breast (MDA-MB-231), cervix (HeLa), and noncancerous breast epithelial cell line (MCF-10A). The achieved results suggest the heterobimetallic PtII–AuIspecies as optimal compounds that signify the existence of cooperative and synergistic effects in their structures. The complex 3is considered as the most cytotoxic compound with the maximum selectivity index in our screened complex series. Moreover, it is disclosed that 3effectively causes cell death by inducing apoptosis in MIA-PaCa2 cells. Furthermore, the finding results by fluorescent cell microscopy manifest cytoplasmic staining of 3rather than nucleus.

34. Half-Sandwich Cyclometalated RhIIIComplexes Bearing Thiolate Ligands: Biomolecular Interactions and In Vitroand In VivoEvaluations

Abstract

A class of cyclometalated RhIIIcomplexes [Cp*Rh(ppy)(SR)] bearing thiolate ligands, Cp* = pentamethylcyclopentadienyl, ppy = 2-phenylpyridinate, and R = pyridyl (Spy, 2), pyrimidyl (SpyN, 3), benzimidazolyl (Sbi, 4), and benzothiazolyl (Sbt, 5), were produced and identified by means of spectroscopic methods. The in vitrocytotoxicity of the RhIIIcompounds in three different human mortal cancerous cell lines (ovarian, SKOV3; breast, MCF-7; lung, A549) and a normal lung (MRC-5) cell line were evaluated, indicating the selectivity of these cyclometalated RhIIIcomplexes to cancer cells. Complex 5, selected for in vivoexperiment, has shown an effective inhibition of tumor growth in SKOV3 xenograft mouse model relative to control (p-values < 0.05 and < 0.01). Importantly, the outcomes of H&E (hematoxylin and eosin) staining and hematological analysis revealed negligible toxicity of 5compared to cisplatin on a functioning of the main organs of mouse. Molecular docking, UV–vis, and emission spectroscopies (fluorescence, 3D fluorescence, synchronous) techniques were carried out on 1–5to peruse the mechanism of the anticancer activities of these complexes. The obtained data help to manifest the binding affinity between the rhodium compounds and calf thymus DNA (CT-DNA) through the interaction by DNA minor groove and moderate binding affinity with bovine serum albumin (BSA), particularly with the cavity in the subdomain IIA. It can be concluded that the Rh-thiolate complexes are highly promising leads for the development of novel effective DNA-targeted anticancer drugs.

35. Half-Lantern Cyclometalated Platinum(II) Complexes as Anticancer Agents: MolecularDocking, Apoptosis, Cell Cycle Analysis, and Cytotoxic Activity Evaluations

Abstract

Background and Objective: In the design of modern metal-based anticancer drugs, platinum-based complexeshave gained growing interest. In this study, the anticancer activity of half-lantern cyclometalated Pt(II)‒Pt(II)complexes was evaluated using MTT, apoptosis, cell cycle analysis, and DNA binding studies. Materials and Methods: The cytotoxicity of Pt(II)‒Pt(II) complexes were evaluated against different cancer cell lines,such as human lung (A549), breast (MCF-7, and MDA-MB-231), ovarian (SKOV-3), and colon (HT-29) as well asnormal breast (MCF-10A), and human lung fibroblast (MRC-5) cells using MTT assay. BioLegend's PE Annexin, VApoptosis Detection Kit with 7AAD, was applied to assess the apoptotic effects of 1A and 1B compound againstMCF-7 and A549 cell lines. Cell cycle analysis was determined using the flow cytometry method. The interaction ofcompounds with four different DNA structures with PDB codes (1BNA, 1LU5, 3CO3, and 198D) has been investigatedby molecular docking. To achieve binding to DNA experimentally, the electrophoresis mobility shift assay andcomet assay were applied. Results: In the evaluation of cytotoxic effects, 1A showed the highest cytotoxicity among the studied compounds, andit showed higher potency with more selectivity against normal cell lines than cisplatin. This compound had IC50 of7.24, 2.21, 1.18, 2.71, 10.65, 18.32, and 49.21 μM against A549, SKOV3, HT29, MCF-7, MDA-MB-231, MRC-5, andMCF-10A, respectively, whereas cisplatin had IC50 of 9.75, 19.02, 107.23, 15.20, 18.09, 14.36, and 24.21 μm, respectively,on the same cell lines. In order to check the DNA binding activity of 1A, and 1B, electrophoretic mobility wasalso conducted, which indicated that the binding of these compounds led to a slight change in electrophoretic mobilityto DNA. The migration of chromosomal DNA from the nucleus in the form of a tail or comet was executed in thecomet assay of 1A on MCF-7. Examination of apoptosis of 1A and 1B on the MCF-7 cancer cell line showed that itcould increase induction of apoptosis in this cancerous cell in a concentration-dependent manner. Investigating theeffect of 1A using cell cycle analysis on MCF-7 cancer cell line showed that this complex affects stage G1 and S of thecell cycle. Conclusion: 1A has the potential to play a significant role in future biopharmaceutical studies.

41. Specific targeting of HER2-positive human breast carcinoma SK-BR-3 cells by amygdaline-ZHER2 affibody conjugate.

Abstract

Amygdalin induces apoptotic death in several carcinoma cells. Affibody is an engineered protein with a high affinity for human epidermal receptor 2 (HER2). We assessed the cytotoxic effects of the amygdalin-ZHER2 affibody conjugate on two breast carcinoma cell lines. The ZHER2 affibody gene was synthesized and transferred into E. coli BL21 as an expression host. After purification, the ZHER2 affibody was conjugated to amygdalin. The cytotoxic effects of amygdalin and its ZHER2 affibody conjugate on the SK-BR-3, with overexpression of HER2, and MCF-7 cells were evaluated by MTT assay. The effects of amygdalin and its conjugate on apoptotic death and expression of pro-apoptotic Bax and anti-apoptotic Bcl-2 proteins were measured. Amygdalin individually showed a potent cytotoxic effect against both MCF-7 (IC50 = 14.2 mg ml−1) and SK-BR-3 cells (IC50 = 13.7 mg ml−1). However, the amygdalin-ZHER2 affibody conjugate had a more cytotoxic effect on SK-BR-3 (IC50 = 8.27 mg ml−1) than MCF-7 cells (IC50 = 19.8 mg ml−1). Amygdalin had a significant apoptotic effect on both cell lines and the effect of its conjugate on SK-BR-3 cells was significantly more potent than MCF-7 cells. Amygdalin increased Bax and decreased Bcl-2 expression in both cell lines. However, the effect of its conjugate on the Bax and Bcl-2 expression in SK-BR-3 was more potent than MCF-7 cells. In conclusion, the amygdalin-ZHER2 affibody conjugate may be considered as a valuable candidate for specific treatment of breast cancer patients with overexpression of HER2. However, further in vivo studies are required to explain the antitumoral effects of constructed amygdalin-ZHER2 affibody conjugate. [ABSTRACT FROM AUTHOR]

Through a salt metathesis reaction, the chloride ligand in 1 was replaced by potassium benzothiazole-2-thiolate (Kbt) and potassium benzoimidazole-2-thiolate (Kbi) to afford complexes (PhCH2NC)Au(κ¹-S-bt)], 2a and (PhCH2NC)Au(κ¹-S-bi)], 2b, respectively, which were characterized by NMR spectroscopy. The cytotoxic activities of 2a and 2b were evaluated against three human cancer cell lines, including A549 (lung), SKOV3 (ovary), and MCF-7 (breast). Our results indicated that 2a exhibited comparable cytotoxicity on investigated cell lines with cisplatin. It showed a good antiproliferative activity with IC50 of 19.46, 11.76 and 13.27 μM against A549, SKOV3 and MCF-7 cell lines, respectively. The effects of these complexes on the proliferation of the non-tumorigenic epithelial breast cell line (MCF-10A) showed their good selectivity between the tumorigenic and non-tumorigenic cell lines. Molecular docking simulation studies were also conducted to determine the specific binding site and binding mode of the synthesized gold complexes to DNA and thioredoxinreductase (TrxR) as their proposed targets. Abstract

Dichloroacetate (DCA) as a small and active anticancer agent through inhibition of pyruvate dehydrogenase kinases (PDKs), prevents proliferation of tumor growth. In this research, a series of novel piperidine and piperazine derivatives of DCA have been designed and subjected to molecular docking studies. Based on the docking results, nine compounds with the lowest binding energy and better interaction with PDKs isoenzymes have been selected and synthesized. The cytotoxic activities of the synthesized compounds have been evaluated against HT-29 and MCF7 human cancer cell lines. These compounds show moderate potencies with much higher anticancer activity than DCA. The most active of the series, f1, showed IC50 value of 7.79 µM against HT-29 cell line. [ABSTRACT FROM AUTHOR]
