The combination of straw return and nitrogen (N) fertilization is an effective strategy for increasing farmland productivity and improving soil structure while mitigating climate change. However, although straw return practices are widely recommended in agroecosystems targeting sustainable agriculture, few studies have explored changes in grain yield, resource use efficiency, water productivity, and potential interactions between these variables. A field experiment was conducted during 2022–2023 in Northeast China to study the effects of straw return modes and N fertilizer input on maize grain yield and the efficient use of radiation, water, and N. We investigated two straw return tillage methods (rotary tillage with straw returning, RTS; plow tillage with straw returning, PTS) and five N fertilizer levels (0, 112, 187, 262, and 337 kg N ha−1) based on a field location experiment that started in 2015. Compared with RTS treatment, the PTS treatment resulted in a higher photosynthetic capacity by regulating the leaf area index and photosynthetic metabolism, resulting in increased dry matter accumulation and transportation. Such physiological improvements are conducive to increasing the nutrient use efficiency, water productivity, and grain yield. Specifically, PTS combined with the N3 treatment was an efficient tillage mode that increased radiation use efficiency of grain by 123.48 % and 207.41 % and by 153.44 % and 167.37 % in 2022 and 2023, respectively, compared with the PTS and N0 treatment. Together with partial least squares path modeling, regression analysis, and random forest, the variations in the above parameters led to high productivity in terms of N uptake, water productivity, leaf area index, radiation use efficiency, N use efficiency, and dry matter accumulation. Our results highlight the key roles of straw return methods in affecting grain yield, and further suggest that PTS combined with optimized N fertilizer input can enhance resource utilization, leading to a productive, efficient, and sustainable maize population.

The Charge Coupled Device (CCD) scanner determines the concentration of the microarray by capturing the intensity of the fluorescent signal on the microarray in combination with the standard curve. Due to the characteristics of semiconductors, the CCD sensor in the scanner we designed suffers from saturation, the non-linear relationship between photoelectric response and the light intensity collected by CCD, which poses a challenge for fitting the standard curve of microarray scanner. The Least Squares Algorithm (LSA) still has a large relative error even in the case of high-order fitting, especially in the region of the fluorescence image with small gray level. However, the standard curve is critical to the highly accurate measuring of the instrument. In view of the poor curve fitting performance of LSA, Weighted Least Squares (WLS), and Penalized Least Squares (PLS), as well as the small dataset, this paper proposes the Multi-Layer Perceptron (MLP) neural network algorithm with the minimization of relative error as the constraint, which is applied to the standard curve fitting of the scanner. The gray-level of the fluorescent probe in detection image was obtained as the data set acquired by the microarray scanner at different exposure time. And the relative error and the standard deviation of the relative errors were used as evaluation indicators. In our experiments we compared the MLP neural network with relative error minimization as the constraint with the LSA and the MLP neural network with sum of square errors (SSE) minimization as the constraint. The experimental results show that the MLP neural network constrained by minimizing the relative error has good fitting performance for the standard curve of CCD scanner, with the maximum relative error of only 0.89% while the standard deviation of relative error of only 0.25%. It can be seen that this method provides a new approach for standard curve fitting of microarray scanner.

Lactate, an important metabolic product, provides energy to neural cells during energy depletion or high demand and acts as a signaling molecule in the central nervous system. Recent studies revealed that lactate-mediated protein lactylation regulates gene transcription and influences cell fate, metabolic processes, inflammation, and immune responses. This review comprehensively examines the regulatory roles and mechanisms of lactylation in neurodevelopment, neuropsychiatric disorders, brain tumors, and cerebrovascular diseases. This analysis indicates that lactylation has multifaceted effects on central nervous system function and pathology, particularly in hypoxia-induced brain damage. Highlighting its potential as a novel therapeutic target, lactylation may play a significant role in treating neurological diseases. By summarizing current findings, this review aims to provide insights and guide future research and clinical strategies for central nervous system disorders.

The effects of maize/peanut intercropping on crop yields, peanut nodulation, biological nitrogen (N) fixation in peanuts, crop N uptake, and N use efficiency under different N application rates were studied. A long-term maize/peanut intercropping micro-plot experiment was started in 2015. The experiment included the following three planting patterns: maize sole crop (SM), peanut sole crop (SP), and maize and peanut intercropping (intercropping maize: IM; intercropping peanut: IP). Additionally, three N application rates were tested as follows: 0 kg·ha−1 (N0), 150 kg·ha−1 (N150), and 300 kg·ha−1 (N300). The results indicated that N fertilization significantly increased maize yield. Intercropping increased maize yield while decreasing peanut yield across different N application rates. Both N fertilization and intercropping significantly increased the maize harvest index (HI), whereas intercropping decreased the peanut HI under N300. The number and fresh weight of peanut nodules decreased with the increasing N application rate with reductions ranging from 31.15% to 45.23% and 39.60% to 46.67%, respectively. Intercropping increased the number of peanut nodules by an average of 62.56% under the N0 treatment. Intercropping significantly improved the N absorption capacity of the whole intercropping system, and the contribution of maize was higher than that of peanuts. Maize demonstrated a stronger competitive ability for N uptake compared with peanuts in the intercropping system. Intercropping significantly increased the N use efficiency for both maize and peanuts. However, the N use efficiency of maize increased with N application rates, while that of peanut decreased. Compared with sole crops, intercropping increased the partial factor productivity of maize by 55.2% but decreased that of peanuts by 56.3%. In conclusion, at an N application rate of 150 kg·ha−1, maize/peanut intercropping increased overall crop yield and improved the N absorption and use capacity of maize

Abstract Background The RNA m6A modification has been implicated in multiple neurological diseases as well as macrophage activation. However, whether it regulates microglial activation during hypoxic-ischemic brain damage (HIBD) in neonates remains unknown. Here, we aim to examine whether the m6A modification is involved in modulating microglial activation during HIBD. We employed an oxygen and glucose deprivation microglial model for in vitro studies and a neonatal mouse model of HIBD. The brain tissue was subjected to RNA-seq to screen for significant changes in the mRNA m6A regulator. Thereafter, we performed validation and bioinformatics analysis of the major m6A regulators. Results RNA-seq analysis revealed that, among 141 m6A regulators, 31 exhibited significant differential expression (FC (abs) ≥ 2) in HIBD mice. We then subjected the major m6A regulators Mettl3, Mettl14, Fto, Alkbh5, Ythdf1, and Ythdf2 to further validation, and the results showed that all were significantly downregulated in vitro and in vivo. GO analysis reveals that regulators are mainly involved in the regulation of cellular and metabolic processes. The KEGG results indicate the involvement of the signal transduction pathway. Conclusions Our findings demonstrate that m6A modification of mRNA plays a crucial role in the regulation of microglial activation in HIBD, with m6A-associated regulators acting as key modulators of microglial activation.

Maize/peanut intercropping may improve soil health through reducing nitrogen (N) fertilization. However, the effects of maize/peanut intercropping combined with reduced N fertilization on the soil fungal community structure have not been well reported. Using a long-term localized micro-zone experiment, we investigated the combined effects of intercropping and N fertilizer application on soil fungal community diversity and composition. Three cropping patterns (maize/peanut intercropping, maize monoculture, and peanut monoculture) and three N application levels (0 kg·hm−2, 150 kg·hm−2, and 300 kg·hm−2) were assessed. The results showed that the total numbers of fungal species and unique species (operational taxonomic units, OTUs) in both maize and peanut soils tended to first increase and then decrease with increasing N application. Compared with monoculture, the numbers of total OTUs and unique OTUs in intercropped maize soil decreased by 4.14% and 12.79%, respectively, but the total numbers of OTUs and unique OTUs in peanut soil increased by 1.08% and 3.78%, respectively. With increasing N application, the soil fungal Ace and Chao indices of maize soil first increased and then decreased, while the fungal Shannon, Ace, and Chao indices of peanut soil decreased. Compared with the monoculture system, intercropping significantly reduced the maize soil fungal Ace and Chao indices but increased the peanut soil fungal Shannon, Ace, and Chao indices. Nitrogen application and intercropping significantly altered the fungal community structure of maize soil, while N application had no significant effect on the fungal community structure of peanut soil, though intercropping significantly changed the fungal community structure of peanut soil. At the phylum level, Ascomycota, Basidiomycota, Mortierellomycota, unclassified_k_Fungi, and Chytridiomycota were the dominant taxa. Redundancy analysis (RDA) showed that soil nitrate (NO3−) content was the main environmental factor shaping the soil fungal community. In conclusion, excessive N fertilization (300 kg·hm−2) can reduce soil fungal community diversity; maize/peanut intercropping reversed the negative effect of N application on fungal community of peanut soil, but not that of maize soil. Soil NO3− content is the primary environmental driver of soil fungal communities.

Abstract Heat shock protein (HSP) 90, an important component of the molecular chaperone network, is closely concerned with cellular signaling pathways and stress response by participating in the process of maturation and activation of client proteins, playing a crucial role both in the normal and abnormal operation of the organism. In functionally defective tissues, programmed cell death (PCD) is one of the regulable fundamental mechanisms mediated by HSP90, including apoptosis, autophagy, necroptosis, ferroptosis, and others. Here, we show the complex relationship between HSP90 and different types of PCD in various diseases, and discuss the possibility of HSP90 as the common regulatory nodal in multiple PCD, which would provide a new perspective for the therapeutic approaches in disease.

Legume/cereal intercropping has the potential to maximize the use of resources to raise yields due to enhanced nitrogen (N) fixation by legume root nodules, while high N fertilization may inhibit the nodulation of legume. However, whether legume/cereal intercropping can promote the accumulation of soil N storage with N fertilization and its underlying mechanism are less clear. Here, we evaluated the long-term (5 years) effects of maize/peanut intercropping and mineral N fertilization on peanut soil total N content and soil N cycling functional genes. The experiment includes two planting patterns (peanut maize intercropping and peanut monocropping) with three N fertilization rates (0, 150, and 300 kg N ha−1). Intercropping increased soil total N content (STN) by average 18.2%, and the positive effect of intercropping on STN decreased with N application rate. Highest N application decreased the nodule fresh weight (NFW) by 64.3% and 46.0% in intercropping and monocropping system, respectively. However, intercropping has no effect on NFW. Intercropping increased the nifH gene abundance by average 26.5%. SEM analysis indicated that NFW and nifH gene abundance combined can explain 46% of the variance of STN. Our results indicate that biological N fixation but not mineral N fertilization enhances the accumulation of N in soil planted with peanut in maize/peanut intercropping system.

Traditional Chinese medicine (TCM) has attracted a great deal of attention in the treatment of cerebral ischemia is credited with the remarkable neuroprotective effects. However, the imperfect functional mechanism of TCM is a major obstacle to their application. Many studies have been conducted to illustrate the pathophysiology of post-ischemic cerebral ischemia by elucidating the neuronal cell death pathway. Meanwhile, a new type of cell death, ferroptosis, is gradually being recognized in various diseases and is becoming a new pathway of therapeutic intervention strategy to solve many health problems. Especially since ferroptosis has been found to be closely involved into the pathogenesis of cerebral ischemia, it has been considered as a key target in the treatment of cerebral ischemia. Therefore, this paper reviews the latest research findings about the treatment of cerebral ischemia with TCM focused on ferroptosis as a target. Also, in order to explores the possibility of a new approach to treat cerebral ischemia with TCM, we discusses the correlation between ferroptosis and other cell death pathways such as apoptosis and autophagy, which would provide references for the following researches.

Abstract Background Isolated sulfite oxidase deficiency (ISOD) is an autosomal recessive disorder caused by a deficiency of sulfite oxidase, which is encoded by the sulfite oxidase gene (SUOX). Clinically, the disorder is classified as one of two forms: the late-onset mild form or the classic early-onset form. The latter is life-threatening and always leads to death during early childhood. Mild ISOD cases are rare and may benefit from dietary therapy. To date, no cases of ISOD have been reported to recover spontaneously. Here, we present three mild ISOD cases in one family, each with a stable clinical course and spontaneous recovery. Case presentation All three siblings had two novel compound heterozygous mutations in the SUOX gene (NM_000456; c.1096C > T [p.R366C] and c.1376G > A [p.R459Q]). The siblings included two males and one female with late ages of onset (12–16 months) and presented with specific neuroimaging abnormalities limited to the bilateral globus pallidus and substantia nigra. The three patients had decreased plasma homocysteine levels. They exhibited a monophasic clinical course continuing up to 8.5 years even without dietary therapy. Conclusion This is the first report of mild ISOD cases with a stable clinical course and spontaneous recovery without dietary therapy. Our study provides an expansion for the clinical spectrum of ISOD. Furthermore, we highlight the importance of including ISOD in the differential diagnosis for patients presenting with late-onset symptoms, bilaterally symmetric regions of abnormal intensities in the basal ganglia, and decreased plasma homocysteine levels.

Polycaprolactone (PCL) is one of the promising linear aliphatic polyesters which can be used as mulching film. Although it has suitable glass transition temperature and good biodegradability, further practical applications are restricted by the limited temperature-increasing and moisturizing properties. The rational design of the PCL structure is a good strategy to enhance the related properties. In this study, thermally-induced phase separation (TIPS) was introduced to fabricate a PCL nanoporous thin film. The introduction of a nanoporous structure on the PCL surface (np-PCL) exhibited enhanced temperature-increasing and moisturizing properties when used as mulch film. In detail, the average soil temperature of np-PCL was increased to 17.81 °C, when compared with common PCL of 17.42 °C and PBAT of 17.50 °C, and approaches to PE of 18.02 °C. In terms of water vapor transmission rate, the value for np-PCL is 637 gm−2day−1, which was much less than the common PCL of 786 and PBAT of 890 gm−2day−1. As a result, the weed biomass under the np-PCL was suppressed to be 0.35 kg m−2, almost half of the common PCL and PBAT. In addition, the np-PCL shows good thermal stability with an onset decomposition temperature of 295 °C. The degradation mechanism and rate of the np-PCL in different pH environments were also studied to explore the influence of nanoporous structure. This work highlights the importance of the nanoporous structure in PCL to enhance the temperature-increasing and moisturizing properties of PCL-based biodegradable mulching film.

Ischemic brain injuries are common diseases with high morbidity, disability, and mortality rates, which have significant impacts on human health and life. Microglia are resident cells of the central nervous system (CNS). The inflammatory responses mediated by microglia play an important role in the occurrence and development of ischemic brain injuries. This article summarizes the activation, polarization, depletion, and repopulation of microglia after ischemic brain injuries, proposing new treatment strategies for such injuries through the modulation of microglial function.

Abstract Background To provide a synopsis of the current understanding of the association between variants of HNF1B and cancer susceptibility, we conducted a comprehensive research synopsis and meta-analysis to evaluate associations between HNF1B variants and prostate and endometrial cancers. Results Eighteen studies totaling 34,937 patients and 55,969 controls were eligible for this meta-analysis. Four variants showed a significant association with the risk of individual cancer. Strong significant associations were found between rs4430796 A and the risk of both prostate cancer (OR = 1.247, p = 2.21 × 10− 77) and endometrial cancer (OR = 1.217, p = 8.98 × 10− 16); the AA, AG genotypes also showed strong significant associations with the risk of prostate cancer (OR1 = 1.517, p = 4.46 × 10− 22; OR2 = 1.180, p = 0.002). There was a strong significant association between rs7501939 G and the risk of prostate cancer (OR = 1.201, p = 9.31 × 10− 31). Strong significant association was found between rs11649743 G (OR = 1.138, p = 1.08 × 10− 12), rs3760511 C (OR = 1.214, p = 1.57 × 10− 19) and the prostate cancer risk;the GG, AG genotypes of rs11649743 also showed strong significant associations with the risk of prostate cancer (OR1 = 1.496, p = 3.32 × 10− 6; OR2 = 1.276, p = 7.82 × 10− 6). All the cumulative epidemiological evidence of associations was graded as strong. Conclusions Our study summarizes the evidence and helps to reveal that common variants of HNF1B are associated with risk of prostate and endometrial cancer.

Background: Hypoxic-ischemic encephalopathy (HIE) is a major contributor to child mortality and morbidity. Reliable prognostication for HIE is of key importance. Proton magnetic resonance spectroscopy (1H-MRS) is a quantitative, non-invasive method that has been demonstrated to be a suitable complementary tool for prediction. The aim of this study was to investigate the prognostic capability of 1H-MRS in the era of therapeutic hypothermia (TH).Methods: Databases, namely MEDLINE, Embase, Web of Science, and the Cochrane library (Cochrane Center Register of Controlled Trials), were searched for studies published before July 17, 2017. Study selection and data extraction were performed by two independent reviewers. The mean difference (MD) or standardized MD (SMD) and 95% confidence interval (CI) were calculated using random-effects models. Subgroup analyses were conducted based on the use of TH.Results: Among the 1,150 relevant studies, seven were included for meta-analysis, but only two small studies were conducted under TH. For 1H-MRS measurement, three peak area ratios revealed predictive values for adverse outcomes in TH subgroup and the combined results (with and without TH): N-acetylaspartate (NAA)/creatine in basal ganglia/thalamus (BG/T) in TH (MD −0.31, 95%CI −0.55 to −0.07) and combined results (MD −0.37, 95% CI −0.49 to −0.25); NAA/choline in BG/T in TH (MD −0.89, 95%CI −1.43 to −0.35) and combined results (MD −0.25, 95%CI −0.42 to −0.07); and myo-inositol/choline in cerebral cortex in TH (MD −1.94, 95%CI −3.69 to −0.19) and combined results (MD −1.64, 95%CI −2.64 to −0.64). Moreover, NAA relative concentration is associated with adverse outcomes: in TH (MD −0.04, 95%CI −0.06 to −0.02) and combined results (MD −0.06, 95%CI −0.11 to −0.01) in white matter; in TH (MD −0.04, 95%CI −0.07 to −0.01) and combined results (MD −0.05, 95%CI −0.07 to −0.02) in gray matter.Conclusions: NAA may be a potential marker in outcome prediction for all HIE subjects. It seems that MDs for the ratios including NAA are larger than for its relative concentration, and therefore are more likely to be measurable in a clinical context. Larger prospective multicenter studies with a standardized protocol for both measurement protocols and analysis methods are required in future studies.

Multiple independent cancer susceptibility loci at chromosome 8q24 have been identified by GWAS (Genome-wide association studies). Forty six articles including 60,293 cases and 62,971 controls were collected to conduct a meta-analysis to evaluate the associations between 21 variants in 8q24 and prostate cancer risk. Of the 21 variants located in 8q2\5 were significantly associated with the risk of prostate cancer. In particular, both homozygous AA and heterozygous CA genotypes of rs16901979, as well as the AA and CA genotypes of rs1447295, were associated with the risk of prostate cancer. Our study showed that variants in the 8q24 region are associated with prostate cancer risk in this large-scale research synopsis and meta-analysis. Further studies are needed to explore the role of the 8q24 variants involved in the etiology of prostate cancer.

A greenhouse pot test was conducted to study the impacts of replacing mineral fertilizer with organic fertilizers for one full growing period on soil fertility, tomato yield and quality using soils with different tomato planting history. Four types of fertilization regimes were compared: (1) conventional fertilizer with urea, (2) chicken manure compost, (3) vermicompost, and (4) no fertilizer. The effects on plant growth, yield and fruit quality and soil properties (including microbial biomass carbon and nitrogen, NH4+-N, NO3--N, soil water-soluble organic carbon, soil pH and electrical conductivity) were investigated in samples collected from the experimental soils at different tomato growth stages. The main results showed that: (1) vermicompost and chicken manure compost more effectively promoted plant growth, including stem diameter and plant height compared with other fertilizer treatments, in all three types of soil; (2) vermicompost improved fruit quality in each type of soil, and increased the sugar/acid ratio, and decreased nitrate concentration in fresh fruit compared with the CK treatment; (3) vermicompost led to greater improvements in fruit yield (74%), vitamin C (47%), and soluble sugar (71%) in soils with no tomato planting history compared with those in soils with long tomato planting history; and (4) vermicompost led to greater improvements in soil quality than chicken manure compost, including higher pH (averaged 7.37 vs. averaged 7.23) and lower soil electrical conductivity (averaged 204.1 vs. averaged 234.6 μS/cm) at the end of experiment in each type of soil. We conclude that vermicompost can be recommended as a fertilizer to improve tomato fruit quality and yield and soil quality, particularly for soils with no tomato planting history.

Although investigators have implicated hypoxic-ischemia (HI) as a potential cause of periventricular leukomalacia (PVL), the role of clinical risk factors or markers for HI in the development of PVL remains controversial. The aim of this study was to identify perinatal HI-related factors associated with PVL.The PubMed, EMBASE, and Cochrane Library databases were searched. The last search was performed on January 2017. Summary effect estimates (pooled odds ratios [ORs]) were calculated for each risk factor using fixed or random effects models with tests for heterogeneity and publication bias.Fifteen studies with a total of 12,851 participants were included in this meta-analysis, and 14 potential risk factors were analyzed. The pooled results showed that mothers with oligohydramnios (OR, 1.55; 95% confidence interval [CI], 1.05 to 2.30), preterm infants with acidemia (OR, 1.87; 95% CI, 1.18 to 2.97), 1-minute Apgar score

BACKGROUD:The role of maternal hypertensive disorders in pregnancy (HDP) in the development of retinopathy of prematurity (ROP) is unclear. METHODS:Studies were retrieved through literature searches in PubMed, EMBASE, Web of Science and the Cochrane Library up to May 5, 2016 without language restrictions. Cohort or case-control studies that reported the association of maternal hypertensive disorders and retinopathy of prematurity were eligible. Either a fixed- or a random-effects model was used to calculate the overall combined risk estimates. RESULTS:Thirteen cohort studies involving a total of 45082 individuals were included in the review. The pooled odds ratios of maternal hypertensive disorders in pregnancy for any stage and severe stages of ROP was 1.12 (95%CI: 0.90-1.40) and 0.80 (95%CI: 0.47-1.35), respectively. Sensitivity analyses confirmed that no single study qualitatively influenced the pooled OR. However, substantial heterogeneity and publication bias were observed in the meta-analysis. CONCLUSIONS:Additional larger, prospective and well-adjusted studies are needed to determine the association between HDP and ROP, especially regarding the effects of different types of maternal hypertensive disorders in pregnancy on retinopathy of prematurity.

Human growth transformation dependent protein (HGTD-P) is a newly identified protein that promotes neuronal apoptosis in hypoxia–ischemia brain damage (HIBD) in neonatal rats. However, the mechanisms regulating HGTD-P expression are not clear. Here we describe microRNAs targeted to HGTD-P and examine their effects on regulating neuronal apoptosis in HIBD. We use samples from cultured neurons after oxygen–glucose deprivation (OGD) and postnatal day 10 rat brains after hypoxia–ischemia (HI). RT-PCR, Western blotting, and immunostaining are used to detect the expression of HGTD-P and cleaved caspase 3, as well as real-time PCR detects microRNA expression. MicroRNA agomir is used to inhibit the expression of HGTD-P, and DAPI, TUNEL, and TTC staining are employed to detect cell apoptosis and brain damage. Moreover, in vitro processing assay is used to examine the mechanism by which HI down-regulates miR-139-5p expression. We found that miR-139-5p is down-regulated in neurons and rat brains after HI treatment. The expression pattern of miR-139-5p correlates inversely with that of HGTD-P. Furthermore, miR-139-5p agomir inhibits neuronal apoptosis and attenuates HIBD, which is concurrent with down-regulation of HGTD-P. Moreover, pre-miR-139 processing activity decreases in extracts from OGD neurons, and OGD neuronal extracts attenuates the processing of pre-miR-139 by Dicer. In conclusion, HI induces inhibitors which block the processing step of pre-miR-139, resulting in the down-regulation of mature miR-139-5p. The down-regulation of miR-139-5p plays a critical role in the up-regulation of HGTD-P expression. MiR-139-5p agomir attenuates brain damage when used 12 h after HI, providing a longer therapeutic window than anti-apoptosis compounds currently available.
