Heritage buildings are crucial for any area's cultural and political aspects. Proper maintenance and monitoring are essential for the conservation of these buildings. However, manual inspections are time‐consuming and expensive. We propose a deep learning–based detection framework to identify the damages on the ancient architectural wall. The algorithm applied in this study is YOLOv5. Comparing its five different versions, it was decided to use YOLOv5m as the most accurate detection algorithm with a mAP of 0.801. The damage types identified are physical weathering and visitors' scratches. High‐resolution images were selected for the experiment and effectively identified image. In addition, the applied algorithm allows real‐time detection and the identification of seasonal sources of disruption, which is proved by the video test in this study. The findings contribute to the development of an intelligent tool for health monitoring with the goal of fast and remote damage detection in the routine maintenance of heritage buildings. This paper presents a rapid and precise deep learning-based approach for measuring cracks in concrete structures. The proposed methodology involves data acquisition, pre-processing, model construction and post-processing. The Deeplabv3+ model is used for crack detection, and the use of Coordinate Attention is introduced to enhance its performance. And the implications of attention locations in the model are discussed. Insertion position after atrous spatial pyramid pooling (ASPP) operation is the most effective and accurate. The crack widths are obtained through post-processing, and the actual width is determined using the width conversion equation. The proposed method achieves an effective crack detection MIoU of 85.95 and a calculated width error of 6.3%, with a reduction of 2.5% compared to traditional models. Numerical experiments and real-world building experiments have demonstrated the feasibility and effectiveness of the proposed method. Summary: Anuran amphibians are susceptible to infection by intestinal nematodes, but the damage and response mechanisms that occur in their intestines after infection are only partially understood. In this study, the intestinal disruption and response mechanisms in Amolops wuyiensis frogs infected with Cosmocercoides wuyiensis n. sp. were revealed through analysis of the intestinal histopathology, digestive enzyme activity, transcriptome and intestinal microbiota. Tissue section analysis showed histological damage and inflammation in the infected intestine, and the digestive enzyme activity indicated a decrease in digestion and absorption of some nutrients. We found that infection led to differences in the intestinal microbiota composition, including lower diversity and symbiotic relationships. The greater relative abundance of the genera Burkholderia and Rhodococcus may enhance intestinal immunity to resist pathogenic infections. A comparison of the transcriptomes of infected and uninfected intestines revealed 1055 differentially expressed genes. GO enrichment and KEGG pathways analyses suggested that the guts of infected C. wuyiensis n. sp. show enhanced complement activation, cell adhesion molecule function, NOD‐like receptor signalling pathway activity and other innate immunity responses. Among the adaptive immune responses, the intestinal immune network for IgA production was significantly enriched, and the expression of IL‐17D and transforming growth factor beta‐1 genes were upregulated in the infected intestine. These results imply that C. wuyiensis n. sp. infection of A. wuyiensis intestine may trigger innate and adaptive immune responses, which reduce the post‐infection burden. Furthermore, the intestine of A. wuyiensis may also respond to C. wuyiensis n. sp. infection by increasing metallocarboxypeptidase activity and accelerating smooth muscle contraction. BACKGROUND: The brown planthopper (BPH), Nilaparvata lugens, is a serious insect pest of rice. Autophagy and its related gene ATG3 play multiple roles in insects. However, information about the functions of ATG3 in BPH (NlATG3) is unavailable, and its potential as a target for pest control remains unclear. RESULTS: RT‐qPCR results showed a relatively low expression of NlATG3 in 1st–4th‐instar nymphs, which increased through 9‐day‐old adults. The expression of NlATG3 increased continuously in 1‐day‐old through 5‐day‐old eggs, whereas it decreased thereafter. The mRNA level of NlATG3 was markedly higher in the ovary (1.16) and head (1.00) compared to the rest body parts of BPH adults. Injecting nymphs with dsNlATG3 at doses from 62.5 to 250 ng per insect had strong lethal effect upon them. For the 5th‐instar nymphs, all individuals died within 5 days after receiving the dsNlATG3, and importantly, no individual successfully molted. Transmission electron microscopy revealed the new cuticle of nymphs injected with dsNlATG3 became loose and curved, which is clearly different from that of the control. Correspondingly, the obvious vesicles in epidermal cells disappeared after dsNlATG3‐treatment. RNAi of NlATG3 significantly reduced the total number of eggs laid per female as well as the eggs' hatchability, especially in the dsNlATG3♀ × dsGFP♂ group, whose total number of eggs laid per female largely decreased by 80.4%, and whose eggs' hatchability was significantly reduced from 95.7% to zero, when compared with the control (dsGFP♀ × dsGFP♂). AbstractPurposeMethodsResultsConclusionDiabetic retinopathy (DR) is the most common complication of diabetes mellitus. Stimulator of interferon genes (STING) plays an important regulatory role in the transcription of several genes. This study aimed to mine and identify hub genes relevant to STING in DR.The STING-related genes (STING-RGs) were extracted from MSigDB database. Differentially expressed STING-RGs (DE-STING-RGs) were filtered by overlapping differentially expressed genes (DEGs) between DR and NC specimens and STING-RGs. A PPI network was established to mine hub genes. The ability of the hub genes to differentiate between DR and NC specimens was evaluated. Additionally, a ceRNA network was established to investigate the regulatory mechanisms of hub genes. Subsequently, the discrepancies in immune infiltration between DR and NC specimens were further explored. Additionally, we performed drug predictions. Finally, RT-qPCR of peripheral blood samples was used to validate the bioinformatics results.A grand total of four genes (IKBKG, STAT6, NFKBIA, and FCGR2A) related to STING were identified for DR. The AUC values of all four hub genes were greater than 0.7, which indicated that the diagnostic value was acceptable. The ceRNA network contained four hub genes, 170 miRNAs, and 135 lncRNAs. In addition, immunoinfiltration analysis demonstrated that the abundance of activated B cells was notably different between the DR and NC specimens. Moreover, 32 drugs were included in the drug-gene network, with twelve drugs targeting STAT6, nine drugs targeting NFKBIA, four drugs targeted IKBKG, and seven drugs targeted FCGR2A. The expression of the four hub genes in blood samples determined by RT-qPCR was consistent with our analysis.In conclusion, four hub genes (IKBKG, STAT6, NFKBIA, and FCGR2A) related to STING with a diagnostic value for DR were identified by bioinformatics analysis, which might provide new insights into the evaluation and treatment of DR. Background: Inadequate levels of vitamin D (VitD) have been linked to increased rates of various health conditions and mortality. However, little is known about the relationship between mortality outcomes and 25-hydroxyvitamin D [25(OH)D] levels in individuals with rheumatoid arthritis (RA). This study aimed to examine this association using data from the National Health and Nutrition Examination Survey., Methods: A cohort of 2,290 individuals aged 20 to 85 years with RA was analyzed. Lower 25(OH)D levels were inversely associated with all-cause mortality, with a hazard ratio (HR) of 0.91 (0.87 to 0.96) per 10 nmol/L increase. Comparatively, the HR for the VitD insufficiency group was 0.64 (0.50 to 0.83), and for the VitD sufficiency group, it was 0.60 (0.44 to 0.80), both compared to the VitD deficiency group. Cause-specific analysis showed that higher 25(OH)D levels were associated with reduced mortality from heart disease (HR: 0.88, 0.82 to 0.95) and malignant neoplasms (HR: 0.86, 0.79 to 0.94). No significant correlation was found between 25(OH)D levels and cause-specific mortalities for other conditions., Results: Stratified by gender, the HR for males was 0.92 (0.85 to 0.99) and for females was 0.91 (0.86 to 0.98) per 10 nmol/L increase in 25(OH)D levels. Among individuals aged 20-59 years, no significant correlation was observed, while for those aged 60 years and older, the HR was 0.86 (0.82 to 0.90) per 10 nmol/L increase. Nonlinear analysis identified a sharp increase in HR below 59.95 nmol/L, while HR remained below 1 for 25(OH)D levels above 59.95 nmol/L., Conclusion: This study reveals a strong negative correlation between 25(OH)D levels and overall mortality in individuals with RA. Notably, this association is particularly significant for mortality related to heart disease and malignant neoplasms. Targeted VitD supplementation should be emphasized, especially in individuals aged 60 years and older with RA. Transition metal (Cu, Ni, Co) doped MnAl mixed oxide catalysts were prepared through a novel method involving the calcination of hydrotalcite precursors for the selective catalytic reduction of NO x with NH 3 (NH 3 -SCR). The effects of transition metal modification were confirmed by means of XRD, BET, TEM, XPS, NH 3 -TPD, and H 2 -TPR measurements. Experimental results evidenced that CoMnAl-LDO presented the highest NO x removal efficiency of over 80% and a relatively high N 2 selectivity of over 88% in a broad working temperature range (150-300 °C) among all the samples studied. Moreover, the CoMnAl-LDO sample possessed better stability and excellent resistance to H 2 O and SO 2 . The reasons for such results could be associated with the good dispersion of Co 3 O 4 and MnO x , which could consequently provide optimum redox behavior, plentiful acid sites, and strong NO x adsorption ability.
