Integrating social cognitive perspective, social exchange perspective, and social identity perspective, we developed a research model examining how leader humility affects follower prosocial behaviours via three psychological mechanisms (i.e., self‐efficacy, affective trust, and perceived insider status). We used two field survey studies to examine our research model. Using a three‐wave lagged design, Study 1 (N = 221) found that leader humility had positive indirect effects on employee knowledge exchange, an information‐based indicator of prosocial behaviours, through all three mechanisms. To constructively replicate and extend our findings, Study 2 (N = 230) used a multi‐source and three‐wave lagged design and incorporated two additional indicators of prosocial behaviours: helping behaviour and voice behaviour. The results showed that affective trust mediated the positive effects of leader humility on all indicators of prosocial behaviours, indicating that social exchange is a powerful theoretical lens in explaining the implications of leader humility. Self‐efficacy mediated the positive effect of leader humility on knowledge exchange and helping behaviour. However, perceived insider status did not exhibit any significant mediating effect. There are no Food and Drug Administration‐approved drugs for treating noise‐induced hearing loss (NIHL), reflecting the absence of clear specific therapeutic targets and effective delivery strategies. Noise trauma is demonstrated results in nicotinamide adenine dinucleotide (NAD+) downregulation and mitochondrial dysfunction in cochlear hair cells (HCs) and spiral ganglion neurons (SGNs) in mice, and NAD+ boosted by nicotinamide (NAM) supplementation maintains cochlear mitochondrial homeostasis and prevents neuroexcitatory toxic injury in vitro and ex vivo, also significantly ameliorated NIHL in vivo. To tackle the limited drug delivery efficiency due to sophisticated anatomical barriers and unique clearance pathway in ear, personalized NAM‐encapsulated porous gelatin methacryloyl (PGMA@NAM) are developed based on anatomy topography of murine temporal bone by micro‐computed tomography and reconstruction of round window (RW) niche, realizing hydrogel in situ implantation completely, NAM sustained‐release and long‐term auditory preservation in mice. Objective·To investigate the changes of mitochondria and nicotinamide adenine dinucleotide (NAD+ ) levels in the cochleae of C57BL/6J mice at different ages, and explore potential mechanism of age-related hearing loss. Methods·Forty C57BL/ 6J male mice aged 1, 4, 8, 12 months, respectively, were chosen and classified into 4 groups in terms of age (n=10). Auditory brain response (ABR) and distortion product otoacoustic emission (DPOAE) were conducted to detect the auditory function of mice at different ages; real-time quantitative PCR (RT-qPCR) was applied to compare the mRNA expression levels of the genes associated with mitochondrial energy metabolism in the cochleae of mice at different ages, including Ndufb5 (NADH: ubiquinone oxidoreductase subunit B5), Sdha (succinate dehydrogenase complex flavoprotein subunit A), Sdhc (succinate dehydrogenase complex subunit C), and Atp5b (ATP synthase, H+ transporting mitochondrial F1 complex, beta subunit); the changes of mitochondrial quantity in the cochlear hair cells from the mice aged 1 month and 12 months were observed by whole-mount immunofluorescence; the mitochondrial ultrastructure in the cochlear sensory epithelia including outer and inner hair cells, myelinate nerve fibers and spiral ganglion neurons of 1- and 12-month-old mice was observed by transmission electron microscope (TEM); NAD+ levels in the cochleae of mice at different ages were detected by quantitative colorimetry. Results·The ABR thresholds of the 12-month-old mice were significantly elevated in comparison with those of the 1-month-old mice at the frequency range of 5.66-- 45.00 kHz (P<0.01); and the DPOAE thresholds of the 12-month-old mice were significantly elevated in comparison with those of the 1-month-old mice at the frequency range of 11.32 -- 32.00 kHz (P<0.01). The expression levels of the genes related to mitochondrial functions including Ndufb5, Sdha, Sdhc, and Atp5b showed a downward trend with the age, which in the 8-month-old and 12-month-old mice were significantly different from those in the 1-month-old mice (P<0.05). Immunostaining showed that the number of mitochondria in cochlear inner hair cells of the 12-month-old mice was significantly lower than that of 1-month-old mice. It was observed by TEM that vacuolar degenerated mitochondria and larger lipofuscin existed in the inner hair cells, myelinate nerve fibers and spiral ganglion neurons of the 12-month-old mice. The NAD+ level showed a decreasing trend with age, declining significantly from 8 months of age, compared with the 1-month-old mice (P<0.01). Conclusion·Mitochondrial dysfunction with abnormal structure and descending NAD+ levels in the cochleae is consistent with hearing function deterioration in C57BL/6J aging mice. Sensorineural hearing loss (SNHL) becomes an inevitable worldwide public health issue, and deafness treatment is urgently imperative; yet their current curative therapy is limited. Auditory neuropathies (AN) were proved to play a substantial role in SNHL recently, and spiral ganglion neuron (SGN) dysfunction is a dominant pathogenesis of AN. Auditory pathway is a high energy consumption system, and SGNs required sufficient mitochondria. Mitochondria are known treatment target of SNHL, but mitochondrion mechanism and pathology in SGNs are not valued. Mitochondrial dysfunction and pharmacological therapy were studied in neurodegeneration, providing new insights in mitochondrion-targeted treatment of AN. In this review, we summarized mitochondrial biological functions related to SGNs and discussed interaction between mitochondrial dysfunction and AN, as well as existing mitochondrion treatment for SNHL. While several studies have investigated the effect of employees' need for achievement on their work‐related outcomes, how leader‐follower congruence in need for achievement is related to critical workplace outcomes remains underexplored. Integrating person‐environment theory and similarity‐attraction theory, this study investigated the influences of leader‐follower congruence in need for achievement on followers' job performance and job well‐being through the mediating effect of leader‐member exchange (LMX). Using cross‐level polynomial regression and response surface analysis to analyze the data from 391 leaders and 1,124 followers, we found that leader‐follower congruence in need for achievement had a positive effect on followers' job performance and job well‐being though the mediating role of LMX. • The fluorescent component ratio (I T : I C) P is effective for tracing anthropogenic POM. • Resuspended POM in the turbidity maximum zone was dominated by refractory POM. • The turbidity maximum zone in spring removed most of the riverine labile POM. • Labile POM export in summer contributes to deoxygenation processes in shelf area. • Refractory POM export predominantly occurs in the high runoff summer period. Base-extracted particulate organic matter (BEPOM) collected in low (spring) and high (summer) runoff seasons in the Changjiang Estuary and adjacent nearshore/offshore shelf areas was examined using absorption and fluorescence spectroscopy. Parallel factor analysis (PARAFAC) identified two humic-like (C1p and C2p) biologically-refractory components and two protein-like (C3p and C4p) bioreactive components. Absorption coefficient a 350p, C1p and C2p of BEPOM showed strong positive correlations with particulate organic carbon (POC), and were generally higher in spring than in summer. C4p correlated positively with Chl a , and was higher in summer than in spring. In the inner estuary, an anthropogenic POM signal from the highly polluted Huangpu River was effectively identified by the fluorescent component ratio (I T : I C) P. In the nearshore area during spring, resuspension within the well-developed turbidity maximum zone (TMZ) supplied large amounts of refractory POM into the water column. POM fractions from an autochthonous source (i.e., algal bloom) were dominant in the offshore area during the summer. A multivariate regression model inferred that POM export fluxes from the Changjiang River during spring had a higher percentage of labile POM fractions, which were subsequently retained by the TMZ. However, these labile fractions were discharged into the offshore shelf area contributing to hypoxia in the summer. The summer high runoff season also transported large amounts of refractory POM that was subsequently buried in the sediment. This study highlights the utilization of optical analyses to trace pollution sources and reveal POM fate and transport dynamics allowing better assessment of its ecological consequences in estuaries. CRISPR/Cas9 gene editing enables the treatment of hearing loss in congenitally deaf neonatal mice via both viral and non-viral delivery. While adeno-associated virus (AAV)-mediated gene delivery systems have been shown to be effective tools for gene replacement in the inner ear, application of the AAV-mediated CRISPR/Cas9 gene-editing approach for this purpose is yet to be documented. Based on our previous findings, we focused on the effects of several AAVs delivered via canalostomy injection in adult mice. Among the AAVs examined, AAV8 showed the greatest efficiency and specificity in transducing inner hair cells (IHC). The ability of Cre-expressing AAV8 to activate Cas9 in floxed-Cas9 knock-in (Cas9 KI) mice was further evaluated. We compared the effects of six different promoters (CMV, CAG, hSyn, CaMKIIa, GFAP, and ALB) of AAV8 delivered to the inner ear of adult Cas9 KI mice. Our findings showed that three AAV groups (CMV, CAG and hSyn promoters) infected the inner ear efficiently with different tropisms. Notably, AAVs with CMV, CAG, and hSyn promoters infected diverse cell types in mature murine cochleae, including IHCs. In particular, AAV8-hSyn showed high affinity to IHCs and spiral ganglion neurons (SGN). Neither the AAV8 virus itself (except AAV8-CAG) nor the surgical procedures used caused damage to HCs or impaired normal hearing. Our findings indicated that injection of AAV-Cre into mature inner ear efficiently induces Cas9 activation to achieve safe and efficient gene editing and different constituent promoters confer diverse infection patterns in cochlea, expanding the repertoire of gene-editing tools for regulating gene expression in target cells of the inner ear as part of the collective effort to rescue genetic hearing loss and develop effective gene therapy techniques. • AAV8 with different promoters activated Cas9 expression in different cell types specifically in mature Cas9 Knock-in mice cochleae safely.
