BackgroundHepatic encephalopathy (HE) is a neurological disorder resulting from advanced liver injury. HE has a high mortality rate and poor prognosis. The pathogenesis of HE is still unclear, which has led to the lack of a satisfactory specific treatment method. There is increasing evidence that the intestinal flora affects the communication between the gut and the brain in the pathogenesis of HE. Adjusting the intestinal flora has had a beneficial effect on HE in recent studies, and the Qingchang Ligan formula (QCLG) has been shown in previous studies to regulate intestinal flora and metabolites. In this study, we established a thioacetamide-induced HE mouse model to evaluate the protective effect of QCLG on HE and explore its potential mechanism, which also demonstrated that intestinal flora dysbiosis is involved in the pathogenesis of HE.MethodsMice were intraperitoneally injected with thioacetamide (TAA, 150 mg/kg) to induce HE. Additionally, they were orally administered Qingchang Ligan Formula (QCLG) at a dose of 6.725 g/kg·d for seven days, while control mice received an equal volume of saline via gavage. Subsequently, samples were subjected to 16S ribosomal ribonucleic acid (rRNA) gene sequencing, high-performance liquid chromatography-mass spectrometry (LC-MS), and RNA-sequencing (RNA-seq) analysis.ResultQCLG improved weight loss, cognitive impairment, neurological function scores, blood ammonia, and brain gene expression of interleukin-6 (TNF-α), Interleukin-1β (IL-1β), and interleukin-6 (IL-6) induced by HE. Moreover, QCLG increased the levels of liver function indicators, including alanine aminotransferase (ALT), aspartate aminotransferase (AST), and serum TNF-α, IL-1β, and IL-6. 16S RNA sequencing revealed increased Oscillibacter, Colidextribacter, and Helicobacter in TAA-induced mouse fecal samples. Also, the abundance of Bifidobacterium decreases TAA-induced mouse fecal samples. In contrast, QCLG treatment significantly restored the gut microbial community. Metabolomics indicated significant differences in some metabolites among the normal control, treatment, and model groups, including 5-methoxytryptophan, Daidzein, Stercobilin, and Plumieride (PLU).ConclusionQCLG can alleviate neuroinflammation and prevent HE caused by liver injury by regulating intestinal flora in mouse models.

Abstract. Background:. Total human immunodeficiency virus (HIV) DNA and integrated HIV DNA are widely used markers of HIV persistence. Droplet digital polymerase chain reaction (ddPCR) can be used for absolute quantification without needing a standard curve. Here, we developed duplex ddPCR assays to detect and quantify total HIV DNA and integrated HIV DNA. Methods:. The limit of detection, dynamic ranges, sensitivity, and reproducibility were evaluated by plasmid constructs containing both the HIV long terminal repeat (LTR) and human CD3 gene (for total HIV DNA) and ACH-2 cells (for integrated HIV DNA). Forty-two cases on stable suppressive antiretroviral therapy (ART) were assayed in total HIV DNA and integrated HIV DNA. Correlation coefficient analysis was performed on the data related to DNA copies and cluster of differentiation 4 positive (CD4+) T-cell counts, CD8+ T-cell counts and CD4/CD8 T-cell ratio, respectively. The assay linear dynamic range and lower limit of detection (LLOD) were also assessed. Results:. The assay could detect the presence of HIV-1 copies 100% at concentrations of 6.3 copies/reaction, and the estimated LLOD of the ddPCR assay was 4.4 HIV DNA copies/reaction (95% confidence intervals [CI]: 3.6–6.5 copies/reaction) with linearity over a 5-log10-unit range in total HIV DNA assay. For the integrated HIV DNA assay, the LLOD was 8.0 copies/reaction (95% CI: 5.8–16.6 copies/reaction) with linearity over a 3-log10-unit range. BackgroundEngaging in anal sexual intercourse markedly increases the risk of developing HIV among men who have sex with men (MSM); oral sexual activities tend to uniquely introduce gut-derived microbes to salivary microbiota, which, combined with an individual’s positive HIV status, may greatly perturb oral microecology. However, till date, only a few published studies have addressed this aspect.MethodsBased on 16S rRNA sequencing data of bacterial taxa, MicroPITA picks representative samples for metagenomic analysis, effectively revealing how the development and progression of the HIV disease influences oral microbiota in MSM. Therefore, we collected samples from 11 HIV-negative and 44 HIV-positive MSM subjects (stage 0 was defined by HIV RNA positivity, but negative or indeterminate antibody status; stages 1, 2, and 3 were defined by CD4+ T lymphocyte counts ≥ 500, 200–499, and ≤ 200 or opportunistic infection) and selected 25 representative saliva samples (5 cases/stage) using MicroPITA. Metagenomic sequencing analysis were performed to explore whether positive HIV status changes salivary bacterial KEGG function and metabolic pathway in MSM.ResultsThe core functions of oral microbiota were maintained across each of the five groups, including metabolism, genetic and environmental information processing. All HIV-positive groups displayed KEGG functions of abnormal proliferation, most prominently at stage 0, and others related to metabolism. Clustering relationship analysis tentatively identified functional relationships between groups, with bacterial function being more similar between stage 0-control groups and stage 1-2 groups, whereas the stage 3 group exhibited large functional changes. Although we identified most metabolic pathways as being common to all five groups, several unique pathways formed clusters for certain groups; the stage 0 group had several, while the stage 2 and 3 groups had few, such clusters. The abundance of K03046 was positively correlated with CD4 counts.ConclusionAs HIV progresses, salivary bacterial function and metabolic pathways in MSM progressively changes, which may be related to HIV promoting abnormal energy metabolism and exacerbate pathogen virulence. Further, infection and drug resistance of acute stage and immune cell destruction of AIDS stage were abnormally increased, predicting an increased risk for MSM individuals to develop systemic and oral diseases.

Ecological restoration is becoming increasingly important in addressing the global crisis of ecosystem degradation. Ecosystem services, as a concept that connects people and natural ecosystems, can be incorporated into the design, practice and evaluation of restoration projects to promote both the socioeconomic and ecological effects of restoration. However, the full incorporation of ecosystem services in landscape-scale restoration projects is still greatly lacking. In this study, we used the Dashihe mine tailings restoration project in Hebei, China, as a case study and analyzed data from field surveys, remote sensing and literature reviews to study the following two questions: 1) How would multiple ecosystem service supply capabilities and their tradeoffs change after ecological restoration; 2) How can multiple ecosystem service supply capabilities be maximized while minimizing their tradeoffs? The supply capabilities of four important ecosystem services (i.e., soil retention, dust deposition, carbon sequestration and habitat maintenance) and their tradeoff intensities under six restoration scenarios were quantified and compared. The results showed that restored ecosystems outperformed unrestored and even the reference conditions in terms of ecosystem service supply enhancement. However, there is still much room for improvement compared to the recommended restoration scenario. The recommended scenario could result in both higher service supplies and lower tradeoff intensities among the services. On this basis, we call for more incorporation of ecosystem service supply and tradeoff analysis in future ecological restoration projects to reverse the declining trends of ecosystem services across the globe.

Changes in land surface properties during urbanization have a significant impact on variations in precipitation. Little research has been carried out on spatial heterogeneity and influence strength of the driving factors of precipitation changes at different urbanization scales. Using a trend analysis and multi-scale geographically weighted regression, this study analysed the spatial heterogeneity and impact scale of driving factors of precipitation changes in 156 urban units in the Beijing-Tianjin-Hebei urban agglomeration region (Jing-Jin-Ji). In summer, RAD (radiation), RHU (relative humidity), WIN (wind speed), and POP (urban population density) were found to act on a small regional scale, AOD (aerosol optical depth) on a medium regional scale, and NDVI (normalized difference vegetation index), NLI (night time light intensity), UHI (urban heat island intensity), and AREA (urban area size) on a global scale. In winter, AREA and WIN acted on a medium regional scale, UHI on a large regional scale, and AOD, NDVI and NLI on a global scale. Across the whole year, NDVI and AREA had a medium regional impact and NLI a large regional one. Variations in natural factors, such as RAD and RHU, had a great influence on the spatial heterogeneity of precipitation changes, whereas human factors, such as NLI and UHI, had a small influence. In summer, AOD mainly affected Tangshan and Qinhuangdao in the northeast and Cangzhou in the southeast of the Jing-Jin-Ji. RHU and AREA primarily affected the cities of Handan and Xingtai. In winter, NLI, AREA, WIN, and UHI had significant effects in the cities of Handan and Xingtai, with AREA being the most important factor. In the Shijiazhuang-Hengshui area, RAD and NLI played a significant role; in the Beijing-Zhangjiakou-Chengde area, the most important factor affecting precipitation changes was the variation in POP. These results provide a scientific basis for flood disaster risk management in the Jing-Jin-Ji and the establishment of differentiated climate policies in different cities.

Rapid global changes are altering regional hydrothermal conditions, especially in ecologically vulnerable areas such as coastal regions, subsequently influencing the dynamics of vegetation growth. However, there is limited research investigating the response of vegetation in these regions to extreme climates and the associated time lag-accumulation relationships. This study utilized a combined approach of gradual and abrupt analysis to examine the spatiotemporal patterns of vegetation dynamics in the coastal provinces of China from 2000 to 2019. Additionally, we evaluated the time lag-accumulation response of vegetation to extreme climate events. The results showed that (1) extreme high temperatures and extreme precipitation had increased over the past two decades, with greater warming observed in high latitudes and concentrated precipitation increases in water-rich southern regions; (2) both gradual and abrupt analyses indicate significant vegetation improvement in coastal provinces; (3) significant lag-accumulation relationships were observed between vegetation and extreme climate in the coastal regions of China, and the time-accumulation effects were stronger than the time lag effects. The accumulation time of extreme temperatures was typically less than one month, and the accumulation time of extreme precipitation was 2–3 months. These findings are important for predicting the growth trend of coastal vegetation, understanding environmental changes, and anticipating ecosystem evolution.

BackgroundRhubarb is an important traditional Chinese medicine, and rhein is one of its most important active ingredients. Studies have found that rhein can improve ulcerative colitis by regulating gut microbes, but there are few reports on its effects on liver diseases. Therefore, this study aims to investigate these effects and underlying mechanisms.MethodsMice were given rhein (100 mg/kg), with both a normal control group and a model group receiving the same amount of normal saline for one week. Acute liver injury was induced in mice by intraperitoneal injection of D-GalN (800 mg/kg)/LPS (10 ug/kg). Samples (blood, liver, and stool) were then collected and assessed for histological lesions and used for 16S rRNA gene sequencing, high-performance liquid chromatography-mass spectrometry (LC-MS) and RNA-seq analysis.ResultsThe levels of ALT and AST in the Model group were abnormal higher compared to the normal control group, and the levels of ALT and AST were significantly relieved in the rhein group. Hepatic HE staining showed that the degree of liver injury in the rhein group was lighter than that in the model group, and microbiological results showed that norank_o:Clostridia_UCG-014, Lachnoclostridium, and Roseburia were more abundant in the model group compared to the normal control group. Notably, the rhein treatment group showed reshaped disturbance of intestinal microbial community by D-GalN/LPS and these mice also had higher levels of Verrucomicrobia, Akkermansiaceae and Bacteroidetes. Additionally, There were multiple metabolites that were significantly different between the normal control group and the model group, such as L-α-amino acid, ofloxacin-N-oxide, 1-hydroxy-1,3-diphenylpropan-2-one,and L-4-hydroxyglutamate semialdehyde, but that returned to normal levels after rhein treatment. The gene expression level in the model group also changed significantly, various genes such as Cxcl2, S100a9, Tnf, Ereg, and IL-10 were up-regulated, while Mfsd2a and Bhlhe41 were down-regulated, which were recovered after rhein treatment.ConclusionOverall, our results show that rhein alleviated D-GalN/LPS-induced acute liver injury in mice. It may help modulate gut microbiota in mice, thereby changing metabolism in the intestine. Meanwhile, rhein also may help regulate genes expression level to alleviate D-GalN/LPS-induced acute liver injury.

ObjectiveBacterial DNA (bactDNA) detection can be used to quickly identify pathogenic bacteria and has been studied on ascitic fluid. We aimed to retrospectively analyze the diagnostic value and applicational prospect of the bactDNA load in spontaneous bacterial peritonitis (SBP).MethodWe extracted viable bactDNA from ascitic samples of 250 patients with decompensated cirrhosis collected from October 2019 to April 2021 and detected the bactDNA by droplet digital polymerase chain reaction (ddPCR). We used ascitic samples of a baseline cohort of 191 patients to establish diagnostic thresholds for SBP and analyze the patients’ diagnostic performance based on ascites polymorphonuclear (PMN) and clinical manifestation. We performed bactDNA quantification analysis on 13 patients with a PMN less than 250 cells/mm3 but with clinical symptoms. The dynamic changes of the bactDNA load from eight patients (before, during, and after SBP) were analyzed.ResultsAfter the removal of free DNA, the bactDNA detected by ddPCR was generally decreased (1.75 vs. 1.5 log copies/µl, P < 0.001). Compared with the traditional culture and PMN count in the SBP diagnosis, the bactDNA showed that the ddPCR sensitivity was 80.5%, specificity was 95.3%, positive predictive value was 82.5%, and negative predictive value was 94.7%, based on clinical composite criteria. In patients with a PMN less than 250 cells/mm3, the bactDNA load of 13 patients with symptoms was significantly higher than those without symptoms (2.7 vs. 1.7 log copies/µl, P < 0.001). The bactDNA in eight patients had SBP that decreased by 1.6 log copies/µl after 48 h of antibiotic treatment and by 1.0 log copies/µl after 3 days of continued treatment.ConclusionBactDNA detection can be used to further enhance the diagnostic efficiency of SBP. Therefore, the application of ddPCR assay not only can be used to discriminate and quantify bacteria but also can be used in the clinical assessment for antibiotics treatment.

The Qingchangligan formula (QCLGF) is a traditional Chinese medicine that has significant clinical potential for patients with acute liver failure (ALF). However, the experimental evidence of the effect of QCLGF on ALF and the associated mechanisms remain elusive. We aimed to evaluate the function of QCLGF in ALF and the underlying mechanism. ALF was induced in rats by intraperitoneal injection of D-GalN (1100 mg/kg). The Qingchangligan formula was administered to the rats (6.725 g/kg · d) for 5 days, and the model group and the control group were given the same amount of physiological saline. Then 16S rRNA gene sequencing, high performance gas chromatography-mass spectrometry (GC-MS), and RNA-seq analysis were performed on the samples. The levels of ALT and AST in the ALF rats were abnormal (5322.08 ± 566.27 U/L and 7655.95 ± 1238.08 U/L, respectively) compared with the normal control (98.98 ± 6.90 U/L and 99.63 ± 10.94 U/L, respectively). The levels of ALT and AST in the QCLGF rats (2997.67 ± 469.24 U/L and 4158.40 ± 596.07 U/L, respectively) were closer the normal control group. Liver HE staining showed that the degree of liver damage in the QCLGF rats was lighter than that in the ALF rats. The overall structure of the gut microbiota after ALF was significantly altered, including Proteobacteria, Blautia, Romboutsia, Parabacteroides, UCG-008, Parasutterella, Ruminococcus, norank_f:Lachnospiraceae, the Eubacterium_xylanophilum_group, Oscillibacter, and Eisenbergiella. QCLGF balanced the structure and abundance of intestinal flora. The levels of D(+)galactose, isopropyl beta-D-1-thiogalactopyranoside and D-mannitol were lighter in the plasma of the ALF rats than in the normal control rats, but there were significantly elevated levels of those metabolites in the QCLGF rats. The gene expression changed significantly in the ALF rats. QCLGF regulated the expression of THBS1 and the KEGG pathways of carbohydrate metabolism, lipid metabolism, signal transduction, the immune system, and infectious disease: bacterial. QCLGF may alleviating intestinal flora disorder, regulating galactose metabolism and downregulating the expression of THBS1 to alleviate D-GalN induced acute liver failure.

Salivary virome is important for oral ecosystem, but there are few reports on people living with HIV. We performed metagenomic sequencing to compare composition and functional genes of salivary virobiota between one HIV-negative and four HIV-positive groups in which participants were all men who have sex with men (MSM) with different immunosuppression statuses (five samples per group) to find the evidence that salivary virobiota plays a role in the pathogenesis of oral disease. Acute-stage subjects achieved a positive result of HIV RNA, but HIV antibody negative or indeterminate, whereas individuals with mild, moderate, and severe immunosuppression exhibited CD4+ T-lymphocyte counts of at least 500, 200–499, and less than 200 cells/μL or opportunistic infection, respectively. The results showed the composition of salivary virus genera in subjects with mild immunosuppression was the most similar to that in healthy people, followed by that in the acute stage; under severe immunosuppression, virus genera were suppressed and more similar to that under moderate immunosuppression. Furthermore, abnormally high abundance of Lymphocryptovirus was particularly obvious in MSM with HIV infection. Analysis of KEGG Pathway revealed that Caulobacter cell cycle, which affects cell duplication, became shorter in HIV-positive subjects. It is worth noting that in acute-stage participants, protein digestion and absorption related to the anti-HIV-1 activity of secretory leukocyte protease inhibitor was increased. Moreover, in the severely immunosuppressed subjects, glutathione metabolism, which is associated with the activation of lymphocytes, was enhanced. Nevertheless, the ecological dysbiosis in HIV-positive salivary virobiota possibly depended on the changes in blood viral load, and salivary dysfunction of MSM infected with HIV may be related to CD4 counts. Ribonucleoside diphosphate reductase subunit M1 in purine metabolism was negatively correlated, though weakly, to CD4 counts, which may be related to the promotion of HIV-1 DNA synthesis in peripheral blood lymphocytes. 7-Cyano-7-deazaguanine synthase in folate biosynthesis was weakly positively correlated with HIV viral load, suggesting that this compound was produced excessively to correct oral dysfunction for maintaining normal cell development. Despite the limited number of samples, the present study provided insight into the potential role of salivary virome in the oral function of HIV infected MSM.

Analysis focused on sub-regional differentiation of vegetation greenness and their dominant drivers are needed to properly develop targeted strategies for sustainable management. In this study, we took China as a case study area, and analyzed the spatiotemporal heterogeneity of vegetation greenness and its strength of association with both environmental (topographical factors and hydrothermal conditions) and anthropogenic factors (land use type and population density) across six eco-geographic regions during 1982–2015. The whole period was divided into two periods by the turning point of 1998, after which China has implemented numerous forest protection projects. The attribution results based on the Geodetector method show the followings: (1) In China, precipitation is the dominant factor in landscape variation of Normalized Difference Vegetation Index (NDVI) with a strength of association of 85%. Additionally, precipitation is also the dominant factor in arid and semi-arid regions, including Northern semiarid (NS) region, Northwestern arid (NWA) region and Qinghai-Tibet Plateau (QTP) region. The dominant factors differ across diverse eco-geographic regions; for example, slope dominates in sub-tropical/tropical humid (STH) and middle temperate humid/sub-humid (MTH) regions. (2) Generally, the strength of association between vegetation and temperature decreases across China over the past 34 years, meaning that the limiting effect of temperature on the NDVI is weakened, similarly, the controlling effect of water conditions is also weakened. In contrast, the spatial association between anthropogenic factors and NDVI is enhanced. (3) The temporal dynamics of strength of association between factors and the NDVI differ in diverse periods and regions; for example, the strength of association between wind speed and NDVI decreased during 1982–1998, but increased during 1999–2015 in temperate humid/sub-humid (WTH) region; however, decreasing trends were revealed in the QTP region in both periods. Our study highlights that variation of NDVI is mainly attributed to climate change and land cover change. Generally, the limiting impact of hydrothermal conditions on NDVI weakens, and the controlling effect of human activity increases over time.

The urban thermal environment is affected by multiple urban form and natural environment factors; research on the accurate prediction of the urban thermal environment, considering the interaction among different urban environmental factors, is still lacking. The development of a machine learning model provides a good means of solving complex problems. This study aims to clarify the relationship between urban environmental variables and the urban thermal environment through high-precision machine learning models as well as provide scenarios of future urban thermal environment developments. We defined an urban thermal environment index (UTEI), considering twelve urban form and natural indicators sourced from the remote sensing data of 150 cities in the Jing-Jin-Ji region from 2000 to 2015. We achieved accurate predictions of UTEI through training a gradient-boosted regression trees model. By unpacking the model, we found that the contribution rate of elevation (ELEV) was the highest. Among all the urban form indicators, the elongation index (ELONG), urban population (POP), nighttime light intensity (NLI), urban area size (AREA), and urban shape index (SHAPE) also had high contributions. We set up five scenarios to simulate the possible impact of different urban form factors on the overall urban thermal environment quality in the region. Under extremely deteriorated patterns that do not control urban expansion and vegetation reduction, the average UTEI could be as high as 0.55–0.76 °C in summer and 0.24–0.29 °C in winter, yet in the extremely optimized situation, UTEI decreased by 0.69 °C in summer and 0.56 °C in winter. Results showed that better urban form improves the quality of urban environments and can provide important insights for urban planners to mitigate urban heat island problems.

Urbanization has a significant influence on precipitation, but existing studies lack the spatial and temporal heterogeneity analysis of its impact on precipitation in urban areas at different levels. This study investigates the spatial heterogeneity of precipitation and the influencing factors on six dimensions in 156 urban areas in the Beijing–Tianjin–Hebei urban agglomeration from 2000 to 2018, utilizing a mixed-methods analytical approach. The results show that the change in the natural factor layer caused by urbanization was the most important factor, affecting urban precipitation variation in summer and over the whole year, accounting for 34.5% and 10.7%, respectively. However, the contribution of the urban thermal environment in summer cannot be ignored, and the change in the urban thermal environment caused by human activities in winter is an important influencing factor. When considering the optimal combination of factors, relative humidity was shown to be significant in the spatial variations in precipitation during summer, which contributed 26.2%, followed by human activity as indicated by night-time light intensity. Over the whole year, aerosol optical depth makes the substantial contribution of 21.8% to urban precipitation change. These results provide benchmarks for improving the adaptability of urban-environment change and urban planning in the context of urbanization.

The increasing degree of urbanization has continuously aggravated the surface urban heat island (sUHI) effect in China. To investigate the correlation between spatiotemporal changes of sUHI and urbanization in Beijing, land surface temperature in summer from 2000 to 2017 and the distribution of local climate zones (LCZs) in 2003, 2005, 2010, and 2017 was retrieved using remote sensing data and used to analyze the sUHI area and intensity change. The statistical method GeoDetector was utilized to investigate the explanatory ability of LCZs and population as the driving factors. The year of 2006 was identified as the main turning year for sUHI evolution. The variation the sUHI from 2000 showed first an increasing trend, and then a decreasing one. The sUHI pattern changed before and after 2009. Before 2009, the sUHI mainly increased in the suburbs, and then, the enhancement area moved to the central area. The sUHI intensity change under different LCZ conversion conditions showed that the LCZ conversion influences the sUHI intensity significantly. Based on population distribution data, we found that the relationship between population density and sUHI gets weaker with increasing population density. The result of GeoDetector indicated that the LCZ is the main factor influencing the sUHI, but population density is an important auxiliary factor. This research reveals the sUHI variation pattern in Beijing from 2000 and could help city managers plan thermally comfortable urban environments with a better understanding of the effect of urban spatial form and population density on sUHIs.

The outbreak of Coronavirus Disease (COVID-19) in Wuhan have affected more than 250 countries and regions worldwide. However, most of the clinical studies have been focused on Wuhan, and little is known about the disease outside of Wuhan in China. In this retrospective cohort study, we report the early clinical features of 80 patients with COVID-19 admitted to the hospital in Beijing. The results show that 27 (33.8%) patients had severe illness. Six (7.5%) patients were admitted to the ICU, and 3 (3.8%) patients died. Forty-eight percent (39/80) of the patients had a history of living/traveling in Wuhan. Patients with severe- illness were significantly older (average age, 71 years old vs 44 years old) and had a high incidence of expectoration (59.3% vs 34.0%), shortness of breath (92.6% vs 9.4%), anorexia (51.9% vs 18.9%) and confusion(18.5% vs 0%) compared with nonsevere patients. The systolic blood pressure (median, 130 mmHg vs 120 mmHg) was higher and the oxygen saturation (median, 98.3% vs 92.0%) was significantly lower in severe patients than nonsevere patients. In addition, myoglobin (median, 56.0 ng/mL vs 35.0 ng/mL), troponin I (median, 0.02 pg/mL vs 0.01 pg/mL), C-reactive protein (median, 69.7 mg/L vs 12.9 mg/L) and neutrophils (median, 3.3×109/L vs 2.2×109/L) were significantly increased, while lymphocytes (median, 0.8×109/L vs 1.2×109/L), albumin (mean, 32.8 g/L vs 36.8 g/L) and the creatinine clearance rate (median, 91.2 vs 108.2 ml/min/1.73m2) were significantly decreased among severe patients. Our study revealed that older patients with high levels of C-reactive protein, myoglobin, troponin I, and neutrophil and high systolic blood pressure as well as low levels of lymphocytes, and albumin and a low creatinine clearance rate and oxygen saturation were more likely to have severe disease.

Deteriorating air quality is one of the most important environmental factors posing significant health risks to urban dwellers. Therefore, an exploration of the factors influencing air pollution and the formulation of targeted policies to address this issue are critically needed. Although many studies have used semi-parametric geographically weighted regression and geographically weighted regression to study the spatial heterogeneity characteristics of influencing factors of PM2.5 concentration change, due to the fixed bandwidth of these methods and other reasons, those studies still lack the ability to describe and explain cross-scale dynamics. The multi-scale geographically weighted regression (MGWR) method allows different variables to have different bandwidths, which can produce more realistic and useful spatial process models. By applying the MGWR method, this study investigated the spatial heterogeneity and spatial scales of impact of factors influencing PM2.5 concentrations in major Chinese cities during the period 2005–2015. This study showed the following: (1) Factors influencing changes in PM2.5 concentrations, such as technology, foreign investment levels, wind speed, precipitation, and Normalized Difference Vegetation Index (NDVI), evidenced significant spatial heterogeneity. Of these factors, precipitation, NDVI, and wind speed had small-scale regional effects, whose bandwidth ratios are all less than 20%, while foreign investment levels and technologies had medium-scale regional effects, whose bandwidth levels are 23% and 32%, respectively. Population, urbanization rates, and industrial structure demonstrated weak spatial heterogeneity, and the scale of their influence was predominantly global. (2) Overall, the change of NDVI was the most influential factor, which can explain 15.3% of the PM2.5 concentration change. Therefore, an enhanced protection of urban surface vegetation would be of universal significance. In some typical areas, dominant factors influencing pollution were evidently heterogeneous. Change in wind speed is a major factor that can explain 51.6% of the change in PM2.5 concentration in cities in the Central Plains, and change in foreign investment levels is the dominant influencing factor in cities in the Yunnan-Guizhou Plateau and the Sichuan Basin, explaining 30.6% and 44.2% of the PM2.5 concentration change, respectively. In cities located within the lower reaches of the Yangtze River, NDVI is a key factor, reducing PM2.5 concentrations by 9.7%. Those results can facilitate the development of region-specific measures and tailored urban policies to reduce PM2.5 pollution levels in different regions such as Northeast China and the Sichuan Basin.

At the city scale, the diurnal and seasonal variations in the relationship between urban form and the urban heat island effect remains poorly understood. To address this deficiency, we conducted an empirical study based on data from 150 cities in the Jing-Jin-Ji region of China from 2000 to 2015. The results derived from multiple regression models show that the effects of urban geometric complexity, elongation, and vegetation on urban heat island effect differ among different seasons and between day and night. The impacts of urban geometric factors and population density in summer, particularly those during the daytime, are significantly larger than those in winter. The influence of urban area and night light intensity is greater in winter than in summer and is greater during the day than at night. The effect of NDVI is greater in summer during the daytime. Urban vegetation is the factor with the greatest relative contribution during the daytime, and urban size is the dominant factor at night. Urban geometry is the secondary dominant factor in summer, although its contribution in winter is small. The relative contribution of urban geometry shows an upward trend at a decadal time scale, while that of vegetation decreases correspondingly. The results provide a valuable reference for top-level sustainable urban planning.

There have been debates and a lack of understanding about the complex effects of urban-scale urban form on air pollution. Based on the remotely sensed data of 150 cities in the Beijing-Tianjin-Hebei agglomeration in China from 2000 to 2015, we studied the effects of urban form on fine particulate matter (PM2.5) and nitrogen dioxide (NO2) concentrations from multiple perspectives. The panel models show that the elastic coefficients of aggregation index and fractal dimension are the highest among all factors for the whole region. Population density, aggregation index, and fractal dimension have stronger influences on air pollution in small cities, while area size demonstrates the opposite effect. Population density has a stronger impact on medium/high-elevation cities, while night light intensity (NLI), fractal dimension, and area size show the opposite effect. Low road network density can enlarge the influence magnitude of NLI and population density. The results of the linear regression model with multiplicative interactions provide evidence of interactions between population density and NLI or aggregation index. The slope of the line that captures the relationship between NLI on PM2.5 is positive at low levels of population density, flat at medium levels of population density, and negative at high levels of population density. The study results also show that when increasing the population density, the air pollution in a city with low economic and low morphological aggregation degrees will be impacted more greatly.

More than 3000 cities in China were used to study the effect of urbanization and local climate variability on urban vegetation across different geographical and urbanization conditions. The national scale estimation shows that China’s urban vegetation depicts a trend of degradation from 2000 to 2015, especially in developed areas such as the Yangtze River Delta. According to the panel models, the increase of precipitation (PREC), solar radiation (SRAD), air temperature (TEMP), and specific humidity (SHUM) all enhance urban vegetation, while nighttime light intensity (NLI), population density (POPDEN), and fractal dimension (FRAC) do the opposite. The effects change along the East–West gradient; the influences of PREC and SHUM become greater, while those of TEMP, SRAD, NLI, AREA, and FRAC become smaller. PREC, SHUM, and SRAD play the most important roles in Northeast, Central, and North China, respectively. The role of FRAC and NLI in East China is much greater than in other regions. POPDEN remains influential across all altitudes, while FRAC affects only low-altitude cities. NLI plays a greater role in larger cities, while FRAC and POPDEN are the opposite. In cities outside of the five major urban agglomerations, PREC has a great influence while the key factors are more diversified inside.

BACKGROUND: Despite prolonged treatment with highly active antiretroviral therapy (HAART), the infectious HIV-1 continues to replicate and resides latently in the resting memory CD4+ T lymphocytes, which blocks the eradication of HIV-1. The viral persistence of HIV-1 is mainly caused by its proviral DNA being either linear nonintegrated, circular nonintegrated, or integrated. Previous reports have largely focused on the dynamics of HIV-1 DNA from the samples collected with relatively long time intervals during the process of disease and HAART treatment, which may have missed the intricate changes during the intervals in early treatment. METHODOLOGY/PRINCIPAL FINDINGS: In this study, we investigated the dynamics of HIV-1 DNA in patients during the early phase of HARRT treatment. Using optimized real time PCR, we observed significant changes in 2-LTR during the first 12-week of treatment, while total and integrated HIV-1 DNA remained stable. The doubling time and half-life of 2-LTR were not correlated with the baseline and the rate of changes in plasma viral load and various CD4+ T-cell populations. Longitudinal analyses on 2-LTR sequences and plasma lipopolysaccharide (LPS) levels did not reveal any significant changes in the same treatment period. CONCLUSIONS/SIGNIFICANCE: Our study revealed the rapid changes in 2-LTR concentration in a relatively large number of patients during the early HAART treatment. The rapid changes indicate the rapid infusion and clearance of cells bearing 2-LTR in the peripheral blood. Those changes are not expected to be caused by the blocking of viral integration, as our study did not include the integrase inhibitor raltegravir. Our study helps better understand the dynamics of HIV-DNA and its potential role as a biomarker for the diseases and for the treatment efficacy of HAART.
