2. Characterization of N6-methyladenosine long non-coding RNAs in sporadic congenital cataract and age-related cataract

AIM: To characterize the N6-methyladenosine (m6A) modification patterns in long non-coding RNAs (lncRNAs) in sporadic congenital cataract (CC) and age-related cataract (ARC). METHODS: Anterior capsule of the lens were collected from patients with CC and ARC. Methylated RNA immunoprecipitation with next-generation sequencing and RNA sequencing were performed to identify m6A-tagged lncRNAs and lncRNAs expression. Kyoto Encyclopedia of Genes and Genomes pathway enrichment analyses and Gene Ontology annotation were used to predict potential functions of the m6A-lncRNAs. RESULTS: Large amount of m6A peaks within lncRNA were identified for both CC and ARC, while the level was much higher in ARC (49 870 peaks) than that in CC (18 688 peaks), yet those difference between ARC in younger age group (ARC-1) and ARC in elder age group (ARC-2) was quite slight. A total of 1305 hypermethylated and 1178 hypomethylated lncRNAs, as well as 182 differential expressed lncRNAs were exhibited in ARC compared with CC. On the other hand, 5893 hypermethylated and 5213 hypomethylated lncRNAs, as well as 155 significantly altered lncRNA were identified in ARC-2 compared with ARC-1. Altered lncRNAs in ARC were mainly associated with the organization and biogenesis of intracellular organelles, as well as nucleotide excision repair. CONCLUSION: Our results for the first time present an overview of the m6A methylomes of lncRNA in CC and ARC, providing a solid basis and uncovering a new insight to reveal the potential pathogenic mechanism of CC and ARC.

3. A modified deep learning method for Alzheimer’s disease detection based on the facial submicroscopic features in mice

Abstract Alzheimer’s disease (AD) is a chronic disease among people aged 65 and older. As the aging population continues to grow at a rapid pace, AD has emerged as a pressing public health issue globally. Early detection of the disease is important, because increasing evidence has illustrated that early diagnosis holds the key to effective treatment of AD. In this work, we developed and refined a multi-layer cyclic Residual convolutional neural network model, specifically tailored to identify AD-related submicroscopic characteristics in the facial images of mice. Our experiments involved classifying the mice into two distinct groups: a normal control group and an AD group. Compared with the other deep learning models, the proposed model achieved a better detection performance in the dataset of the mouse experiment. The accuracy, sensitivity, specificity and precision for AD identification with our proposed model were as high as 99.78%, 100%, 99.65% and 99.44%, respectively. Moreover, the heat maps of AD correlation in the facial images of the mice were acquired with the class activation mapping algorithm. It was proven that the facial images contained AD-related submicroscopic features. Consequently, through our mouse experiments, we validated the feasibility and accuracy of utilizing a facial image-based deep learning model for AD identification. Therefore, the present study suggests the potential of using facial images for AD detection in humans through deep learning-based methods.

Unique natural conditions and socio-cultural backgrounds influenced the evolution of island dwellings, a type of dwellings in China. Over time, the influence of socio-cultural factors, particularly on architectural functions, has gradually become prominent. This study takes Tongshan Ancient City in Dongshan County, Fujian Province, as an example to study the changes in function and socio-cultural factors, and clarify this mechanism. It investigates 52 dwellings, and summarizes the evolution of architectural functions. By analyzing two clues of changes, this study determined three main socio-cultural factors with the most influence: transformation of industrial structure, change of personnel composition, and continuity of religious beliefs.

Abstract The vacancy effect in quantum spin liquid (QSL) has been extensively studied. A finite density of random vacancies in the Kitaev model can lead to a pileup of low-energy density of states (DOS), which is generally experimentally determined by a scaling behavior of thermodynamic or magnetization quantities. Here, we report detailed muon spin relaxation (μSR) results of H3LiIr2O6, a Kitaev QSL candidate with vacancies. The absence of magnetic order is confirmed down to 80 mK, and the spin fluctuations are found to be persistent at low temperatures. Intriguingly, the time-field scaling law of longitudinal-field (LF)-μSR polarization is observed down to 0.1 K. This indicates a dynamical scaling, whose critical exponent of 0.46 is excellently consistent with the scaling behavior of specific heat and magnetization data. All the observations point to the finite DOS with the form $$N(E)\sim {E}^{-\nu }$$ N ( E ) ∼ E − ν , which is expected for the Kitaev QSL in the presence of vacancies. Our μSR study provides a dynamical fingerprint of the power-law low-energy DOS and introduces a crucial new insight into the vacancy effect in QSL.

6. Fasting hypertriglyceridemia in relation to mortality in an elderly male Chinese population

Abstract We investigated fasting hypertriglyceridemia as predictors of all‐cause, cardiovascular, and non‐cardiovascular mortality in an elderly male Chinese population, while accounting for various conventional cardiovascular risk factors. Our participants were elderly men recruited from residents living in a suburban town of Shanghai (≥60 years of age, n = 1583). Hypertriglyceridemia was defined as a fasting serum triglycerides concentration ≥1.70 mmol/L. Subgroup analyses were performed according to current smoking (yes vs. no), alcohol intake (yes vs. no), and the presence and absence of hypertension and hyperglycemia. During a median of 7.9 years follow‐up, all‐cause, cardiovascular, and non‐cardiovascular deaths occurred in 279, 112, and 167 participants, respectively. After adjustment for confounding factors, fasting hypertriglyceridemia was not significantly (p ≥ .33) associated with the risk of all‐cause, cardiovascular, and non‐cardiovascular mortality. However, there was significant (p = .03) interaction between hypertriglyceridemia and the presence and absence of hypertension in relation to all‐cause mortality. In normotensive, but not hypertensive individuals, hypertriglyceridemia was significantly associated with a higher risk of all‐cause mortality (hazard ratio 1.57, 95% confidence interval 1.06–2.31). In further non‐parametric analyses in normotensive individuals, the age‐standardized rate for all‐cause mortality increased from 18.9 in quartile 1 to 20.0, to 24.7, and to 39.9 per 1000 person‐years in quartiles 2, 3, and 4 of serum triglycerides concentration, respectively (ptrend = .0004). Similar results were observed for cardiovascular mortality. Our study in elderly male Chinese showed that fasting hypertriglyceridemia was associated with a higher risk of all‐cause and cardiovascular mortality in patients with normotension but not those with hypertension.

Background: MicroRNAs (miRNAs) show promise as blood-based tumor markers for germ cell tumors (GCTs), with miRNA-371-3p being the most studied. The marginal benefit of including other candidate miRNAs to aid with the management of testicular GCTs remains unclear. Objective: To assess the performance of our combined miRNA assay (371a-3p and 372-3p) in patients with clinically localized testicular masses. Design, setting, and participants: This was a retrospective review of patients prospectively enrolled in an ongoing protocol collecting serum miR-371a-3p and miR-372-3p levels (together, Memorial Sloan Kettering Cancer Center [MSK] miRNA assay [MMA]) in patients with a suspected or diagnosed testicular GCT. Outcome measurements and statistical analysis: The coprimary outcomes of interest were sensitivity and specificity of miR-371a-3p and 372-3p, individually and together, to detect nonteratomatous GCTs in the orchiectomy specimen. Secondary outcomes included additional assay diagnostic parameters, the relationship of patient and disease factors with variations in miRNA levels, and temporal patterns of miRNA normalization after orchiectomy. Results and limitations: Sixty-two patients were included, 52 had a viable GCT at orchiectomy, and ten had no cancer or a non-GCT. Forty-six patients with a GCT had positive preorchiectomy MMA (sensitivity 88.5% [95% confidence interval {CI}: 79.8, 97.2]), and one patient had positive preorchiectomy MMA but no GCT (specificity 90.0% [95% CI: 71.4, 100]). The diagnostic performance of miR-371a-3-p and miR-372-3p was similar. The time for miRNA to decrease to undetectable levels varied, with some patients having positive levels up to 3 wk after orchiectomy. Conclusions: The biomarkers miR-371a-3p and miR-372-3p demonstrated high sensitivity and specificity for localized testicular GCTs, but causes of variation in relative miRNA levels and time to normalization for individual patients remain unclear. Patient summary: We studied the ability of the blood-based biomarkers miR-371a-3p and miR-372-3p to detect testicular cancer (germ cell tumors) in patients with small testicular masses. We found that together and individually these were sensitive and specific for testicular cancer.

8. HIF-1α-HPRT1 axis promotes tumorigenesis and gefitinib resistance by enhancing purine metabolism in EGFR-mutant lung adenocarcinoma

Abstract Background The mutations of oncogenic epidermal growth factor receptor (EGFR) is an important cause of lung adenocarcinoma (LUAD) malignance. It has been knowm that metabolic reprogramming is an important hallmark of malignant tumors, and purine metabolism is a key metabolic pathway for tumor progression and drug resistance, but its relationship with the EGFR-mutant LUAD is unclear. Methods Metabolic reprogramming was studied through capillary electrophoresis-time of flight mass spectrometry (CE-TOF/MS)-based metabolic profiling analysis. Cell proliferation in vitro was evaluated by EdU staining and cell cycle assay. Tumorigenicity in vivo was tested by subcutaneous tumor formation experiment in nude mice. The binding of hypoxia-inducible factor-1 alpha (HIF-1α) and hypoxanthine phosphoribosyltransferase 1 (HPRT1) was detected by DNA pull‑down assay and Chromatin immunoprecipitation (ChIP) assays. HIF-1α, HPRT1, DNA damage and cell apoptosis related genes were examined by western blot. In addition, RNA sequencing, mass spectrometry and bioinformatics analysis were performed. Results We found that mutated EGFR (muEGFR) upregulates HPRT1 to promote purine metabolism and tumorigenesis of EGFR-mutant LUAD. Mechanistically, muEGFR increases HIF-1α expression through protein stability. Meanwhile, up-regulated HIF-1α bound to the promoter of HPRT1 and transcriptionally activates HPRT1 expression, enhancing purine metabolism to maintain rapid tumor cell proliferation in EGFR-mutant LUAD. Further, gefitinib inhibited the synthesis of purine nucleotides, and HPRT1 inhibition increased the sensitivity of gefitinib to EGFR-mutant LUAD. Conclusions Our study reveals that muEGFR-HIF-1α-HPRT1 axis plays a key role in EGFR-mutant LUAD and provides a new strategy-inhibiting purine metabolism for treating EGFR-mutant LUAD.

Solid-state fermentation in mud pits is a typical brewing process for nongxiangxing baijiu and fuyuxiangxing baijiu. Pit mud is the functional carrier of mud pits, in which the abundance, diversity, metabolic activity and interaction of microorganisms could directly affect the quality of baijiu. This paper reviews the application of traditional culture and non-culture techniques in research on microorganisms in pit mud, the spatiotemporal differences in microbial community structure, the functional microorganisms and their interactions, the development and improvement of the methods for the quality evaluation of pit mud, and pit mud maintenance and aging focusing on the functional strains, and summarizes the development history and application status of different techniques in the field of research on microorganisms related to the production of Chinese baijiu, the growth and decline patterns of prokaryotic and eukaryotic microorganisms during pit mud aging, the spatial difference in the distribution of prokaryotic and eukaryotic microorganisms in the pit, and the functions and interdependence of the core functional microorganisms in pit mud such as caproic acid bacteria, butyric acid bacteria, lactic acid bacteria, acetic acid bacteria and methane bacteria, the reason for pit mud aging, the transformation of the criteria for quality evaluation pit mud from traditional sensory and physicochemical analysis to the comprehensive evaluation system, and studies on pit mud maintenance and aging based on the functional properties of core strains. The purpose of this review is to provide a reference for evaluating the microbial diversity, exploring the metabolic function of microbial community and improving the quality of pit mud.

11. Large-scale toppling slope under water level fluctuation of reservoir: A case of Yunnan Province, China

Landslides induced by reservoir inundation are common in Southwest China, negatively influencing hydropower stations. The Wunonglong hydropower station dam was constructed in the upper reaches of the Lancang River, accordingly causing the water level at the Lajinshengu slope to increase by 30 m. A tension crack with a visible depth of 8 m was observed in the upper sector of the Lajinshengu slope after reservoir impoundment for 170 d. In the following days, numerous cracks appeared on the surface of the slope, and the maximum displacement of the slope reached 3.22 m. Then, a large-scale active deformation body within the Lajinshengu slope formed with an area of 2.62 × 105 m2 and a volume of 1.65 × 107 m3. Detailed field investigations, on-site monitoring, and centrifugal model tests were carried out to analyze the surface features, deformation characteristics, and failure mechanism of the Lajinshengu slope. The results show that the slope is an ancient landslide, divided into two parts (i.e. zone A and zone B) by the gully. Zone B is a traction landslide caused by the displacement of zone A. The long-term inundation weakens the soft rock at the slope foot, intensifying the toppling of bedrock and consequently triggering the sliding of the overburden in zone A. The failure mode of the Lajinshengu slope is a typical case of toppling-sliding failure, and the underlying rock toppling drives the overlying sliding. In addition, early identification methods for toppling deformation covered by overburdened soil were proposed based on monitoring data and deformation signs.

12. Performance of zero-valent iron immobilized on activated carbon cloth for the removal of phenol from wastewater

Abstract Background Discharge of large amounts of untreated industrial effluent into water bodies pose significant environmental challenges worldwide. This is due to the limitations of traditional wastewater treatment methods in the treatment of recalcitrant organic pollutants. Fenton processes involves the generation of hydroxyl radicals that are well suited to degrade organics in effluent water. This study focuses on reducing slag generation during Fenton processes and enhancing the reuse of nano-zero-valent iron (NZVI) through the immobilization of NZVI on activated carbon cloth (ACC) through a chitosan (CH) linker with phenol as a model pollutant. Results Microstructural and spectroscopic techniques were employed to study the materials prepared and 37.5 wt% iron loading was achieved. Phenol degradation of 96.3% at 40 °C at pH of 3.0 with 50 mM H2O2 was achieved using ACC-CH-NZVI. Adsorption and degradation studies carried out using ACC-CH-NZVI catalyst revealed that phenol adsorption onto ACC-CH-NZVI fits the Langmuir isotherm model, following the pseudo-second-order kinetic model and first-order reaction kinetics. Thermodynamic studies indicate the non-spontaneous, endothermic and irreversible nature of the removal process. Comparing ACC-CH-NZVI with ACC and ACC-CH, phenol removal using ACC drops from 87.8 to 39%, while using ACC-CH, the removal efficiency drops from 73 to 20.9% and using ACC-CH-NZVI, phenol removal drops from 96.3 to about 70% and total organic carbon removal drops from 79 to about 60% with minimal iron leaching, highlighting the superior performance of ACC-CH-ZVI and the role of NZVI in enhancing phenol removal. Conclusions The catalyst demonstrated good stability for phenol degradation to about 70% phenol removal from simulated wastewater and 60% TOC removal from industrial wastewater after five treatment cycles with minimal Fe leaching. 14. Fast and efficient isolation of murine circulating tumor cells using screencell technology for pre-clinical analyzes

Abstract Circulating tumor cells (CTCs) represent a rare and heterogeneous population of cancer cells that are detached from the tumor site and entered blood or lymphatic circulation. Once disseminated in distant tissues, CTCs could remain dormant or create a tumor mass causing serious danger for patients. Many technologies exist to isolate CTCs from patients’ blood samples, mostly based on microfluidic systems or by sorting them according to their surface antigens, notably EpCAM, and/or cytokeratins for carcinoma. ScreenCell has developed an easy-to-use, antigen-independent, rapid, cost-effective, and efficient technology that isolates CTCs according to their bigger size compared to the blood cells. This study provides the technical information necessary to isolate and characterize CTCs from mouse blood. By using blood samples from transgenic mice with breast cancer or from WT mice in which we spiked cancer cells, we showed that ScreenCell technology is compatible with standard EDTA blood collection tubes. Furthermore, the ScreenCell Cyto kit could treat up to 500 µl and the ScreenCell MB kit up to 200 µl of mouse blood. As the ScreenCell MB kit captures unaltered live CTCs, we have shown that their DNA could be efficiently extracted, and the isolated cells could be grown in culture. In conclusion, ScreenCell provides a rapid, easy, antigen-independent, cost-effective, and efficient technology to isolate and characterize CTCs from the blood samples of cancer patients and murine models. Thanks to this technology CTCs could be captured fixed or alive. Murine cancer models are extensively used in pre-clinical studies. Therefore, this study demonstrates the crucial technical points necessary while manipulating mouse blood samples using ScreenCell technology.

Abstract Purpose Hepatocellular carcinoma (HCC) is the most prevalent malignancies worldwide. Recently, oxidative phosphorylation (OXPHOS) has received extensive concern as an emerging target in antitumor therapy. However, the OXPHOS-involved underlying genes and clinical utilization in HCC remain worth exploring. The present research aimed to create an OXPHOS-relevant signature in HCC. Patients and methods In this study, the prognostic signature genes linked with OXPHOS were identified, and prognostic models were built using least absolute shrinkage and selection operator (LASSO) cox regression analysis. Furthermore, the combination study of immune microenvironment and signature genes looked into the involvement of immune cells in signature-based genes in HCC. Following that, chemotherapeutic drug sensitivity and immunotherapy analysis was implemented to predict clinical efficacy in HCC patients. Finally, clinical samples were collected to measure the expression of OXPHOS-related signature genes. Results Following a series of screens, six prognostic signature genes related with OXPHOS were identified: MRPS23, MPV17, MAPK3, IGF2BP2, CDK5, and IDH2, on which a risk model was built. The findings revealed a significant drop in the survival rate of HCC patients as their risk score increased. Meanwhile, independent prognostic study demonstrated that the risk score could accurately identify HCC patients. Immuno-microenvironmental correlation research suggested that the prognostic characteristics could serve as a reference index for both immunotherapy and chemotherapy. Finally, RT-qPCR exhibited a trend in signature gene expression that was consistent with the results. Conclusion In this study, a total of six prognostic genes associated with OXPHOS were selected and a prognostic model was constructed, providing an essential reference for the study of OXPHOS in HCC.

Accurate ultra-short-term prediction of the Earth rotation parameters (ERP) holds paramount importance for real-time applications, particularly in reference frame conversion. Among them, diurnal rotation (UT1-UTC) which cannot be directly estimated through Global Navigation Satellite System (GNSS) techniques, significantly affects the rapid and ultra-rapid orbit determination of GNSS satellites. Presently, the traditional LS (least squares) + AR (autoregressive) and LS + MAR (multivariate autoregressive) hybrid methods stand as primary approaches for UT1-UTC ultra-short-term predictions (1–10 days). The LS + MAR hybrid method relies on the UT1-UTC and LOD (length of day) series. However, the correlation between LOD and first-order-difference UT1-UTC is stronger than that between LOD and UT1-UTC. In light of this, and with the aid of the first-order-difference UT1-UTC, we propose an enhanced LS + MAR hybrid method to UT1-UTC ultra-short-term prediction. By using the UT1-UTC and LOD data series of the IERS (International Earth Rotation and Reference Systems Service) EOP 14 C04 product, we conducted a thorough analysis and evaluation of the improved method's prediction performance compared to the traditional LS + AR and LS + MAR hybrid methods. According to the numerical results over more than 210 days, they demonstrate that, when considering the correlation information between the LOD and the first-order-difference UT1-UTC, the mean absolute errors (MAEs) of the improved LS + MAR hybrid method range from 21 to 934 μs in 1–10 days predictions. In comparison to the traditional LS + AR hybrid method, the MAEs show a reduction of 7–53 μs in 1–10 days predictions, with corresponding improvement percentages ranging from 1 to 28%. Similarly, when compared to the traditional LS + MAR hybrid method, the MAEs have a reduction of 5–42 μs in 1–10 days predictions, with corresponding improvement percentages ranging from 4–20%. Additionally, when aided by GNSS-derived LOD data series, the MAEs of improved LS + MAR hybrid method experience further reduction.

In this study, a fluorescence detection method combining aptamer-modified up-conversion nanoparticles (UCNPs) and magnetic nanoparticles (MNPs) was developed for detection of Clenbuterol (CLB). The aptamer-modified magnetic NPs captured CLB, which reacted with the aptamer-modified UCNPs and generated a sandwich complex. The aptamer-modified UCNPs acted as a fluorescence source. The MNP-CLB-UCNP complex was retrieved from the solution using an magnetic field, and the fluorescence intensity was detected by fluorescence spectrophotometry with excitation and emission spectra at 980 nm and in the 400–800 nm region, respectively. The results showed that the fluorescence intensity gradually increased with increasing concentrations of CLB with a good specify. The method was highly sensitive for the quantification of CLB, with a limit of detection of 0.304 ng mL−1. The recovery rate of CLB from pork samples ranged from 84 % to 94.87 %. This fluorescence method enables the sensitive, precise, and accurate quantification of CLB residues in pork samples.

Few studies have examined the morphological alterations in the CT blend sign in ICH. Therefore, we assessed changes in the CT blend sign and their association with hematoma expansion (HE) and adverse outcomes in ICH patients.MethodsWe recorded the clinical and radiographic parameters of patients with ICH and blend sign on brain CT. The patients were categorized into two groups, with changes in the relatively hypoattenuating region of the blend sign (CHB group) and with no changes in the relatively hypoattenuating region of the blend sign (NHB groups). We performed univariate and multivariate logistic regression analyses to examine the correlations between CHB and HE and poor outcomes. Furthermore, receiver operating characteristic curve analysis was used to confirm the predictive power of CHB.ResultsIn total, 183 patients were included in the study, of whom 74 (40.4%) demonstrated changes in the hypoattenuating region of the blend sign, whereas 109 (59.6%) did not. Consequently, there has been a compelling exploration into the realm of nanoregulators to refine and enhance these procedures. This review embarks on a meticulous examination of the intricate anatomical structure of the ovary and its microenvironment, thereby establishing a robust groundwork for the development of nanomodulators. It systematically categorizes nanoregulators and delves deeply into their functions and mechanisms, meticulously tailored for optimizing ovarian tissue cryopreservation and transplantation. Furthermore, the review imparts valuable insights into the practical applications and obstacles encountered in clinical settings associated with OTC-T. Abstract To understand the mechanical response pattern of the existing structure and ground due to the construction of metro tunnels underneath, the finite difference method is adopted to study the torsional deformation and stress variation of the existing structure and the effect of underground carriageway structures on the surface subsidence. The curves of the maximum differential subsidence, torsion angle, and distortion of the cross‐section of the existing structure show two peaks in succession during traversing of two metro tunnels beneath it. The torsion angle of the existing structure changes when the two tunnels traverse beneath it in opposite directions. The first traversing of the shield tunnel mainly induces the magnitude variation in torsional deformation of the existing structure, but the second traversing of the subsurface tunnel may cause a dynamic change in the magnitude and form of torsional deformation in the existing structure. The shielding effect can reduce the surface subsidence caused by metro tunnel excavation to a certain extent, and the development trend of subsidence becomes slower as the excavation continues.

In light of increasingly prominent environmental issues, inspiring green supply chain (GSC) members to engage in collaborative innovation is crucial to improve environmental performance. In this paper, in relation to a two-level GSC consisting of manufacturers and suppliers, differential equations involving the greenness of intermediate and final products as state variables are constructed considering the effect of digital capability on green innovation. Subsequently, designs for three incentive mechanisms—the greenness reward, the R&D effort reward, and the digital construction reward—are presented, and their long-term dynamic effects on the economic, environmental, and social benefits are compared and analyzed. Finally, the impacts of consumer green preference and the contribution of digital capability to the advancement of green innovation are explored. The findings show that all these incentives can boost economic, environmental, and social benefits while motivating the supplier. To achieve the best incentive effect, the reward coefficient should fall within a specific range. The digital construction reward mechanism is the most favourable in the initial stage, while the R&D effort reward mechanism is the most appropriate in the long term. The promotion effects of digital capability on green innovation and consumer green preference have the potential to enhance economic, environmental, and social performance.

Abstract Lung diseases globally impose a significant pathological burden and mortality rate, particularly the differential diagnosis between adenocarcinoma, squamous cell carcinoma, and small cell lung carcinoma, which is paramount in determining optimal treatment strategies and improving clinical prognoses. Faced with the challenge of improving diagnostic precision and stability, this study has developed an innovative deep learning-based model. This model employs a Feature Pyramid Network (FPN) and Squeeze-and-Excitation (SE) modules combined with a Residual Network (ResNet18), to enhance the processing capabilities for complex images and conduct multi-scale analysis of each channel's importance in classifying lung cancer. Moreover, the performance of the model is further enhanced by employing knowledge distillation from larger teacher models to more compact student models. Subjected to rigorous five-fold cross-validation, our model outperforms existing models on all performance metrics, exhibiting exceptional diagnostic accuracy. Ablation studies on various model components have verified that each addition effectively improves model performance, achieving an average accuracy of 98.84% and a Matthews Correlation Coefficient (MCC) of 98.83%. Collectively, the results indicate that our model significantly improves the accuracy of disease diagnosis, providing physicians with more precise clinical decision-making support.

31. Experimental study on mix proportion optimization of anti-calcium dissolution shotcrete for tunnels based on response surface methodology

Aiming at the issue of crystallization and blockage of drainage system due to the massive calcium loss from the tunnel shotcrete, a self-designed tunnel seepage crystallization modelling system was developed. This system was produced in conjunction with the initial tunnel support shotcrete construction and drainage pipe installation, and is capable of simulating both the seepage process of groundwater in the shotcrete and the process of crystallization in the drainage pipe. Based on three different mechanisms of anti-crystallization, which include absorbing free calcium, reducing the porosity and increasing hydrophobicity, antialkali agent, nano-calcium carbonate, and silane were selected to test, respectively. Firstly, the suitable dosing ranges of these three external admixtures for resisting calcium loss in shotcrete were determined by single factor tests, which were 7%–11%, 4%–8%, and 0.3%–0.5%, respectively. Thereafter, the response surface method was employed to evaluate the interaction of antialkali agent, nano-calcium carbonate and silane on calcium loss in shotcrete, and to establish the relationship between them, and thus to determine the admixture ratio that can effectively reduce calcium loss crystallization in shotcrete, with the optimal admixture amounts of antialkali agent being 9.242%, nano-calcium carbonate 4.889% and silane 0.366%. Lastly, the reliability of the model test results was verified by the microscopic analysis, and the results showed that the total amount of calcium dissolution in the optimized group could be reduced by 75% compared with the blank control group, and was basically consistent with that derived from the response surface regression model, validating the high accuracy of the buildup response surface regression model. The present study can provide some ideas and references for reducing the seepage crystallization behavior of groundwater in the initial tunnel support shotcrete.

Abstract E6AP dysfunction is associated with Angelman syndrome and Autism spectrum disorder. Additionally, the host E6AP is hijacked by the high-risk HPV E6 to aberrantly ubiquitinate the tumor suppressor p53, which is linked with development of multiple types of cancer, including most cervical cancers. Here we show that E6AP and the E6AP/E6 complex exist, respectively, as a monomer and a dimer of the E6AP/E6 protomer. The short α1-helix of E6AP transforms into a longer helical structure when in complex with E6. The extended α1-helices of the dimer intersect symmetrically and contribute to the dimerization. The two protomers sway around the crossed region of the two α1-helices to promote the attachment and detachment of substrates to the catalytic C-lobe of E6AP, thus facilitating ubiquitin transfer. These findings, complemented by mutagenesis analysis, suggest that the α1-helix, through conformational transformations, controls the transition between the inactive monomer and the active dimer of E6AP.

Abstract Background This study investigated the association between Anti-Müllerian Hormone (AMH) and relevant metabolic parameters and assessed its predictive value in the clinical diagnosis of polycystic ovarian syndrome (PCOS). Methods A total of 421 women aged 20–37 years were allocated to the PCOS (n = 168) and control (n = 253) groups, and their metabolic and hormonal parameters were compared. Spearman correlation analysis was conducted to investigate associations, binary logistic regression was used to determine PCOS risk factors, and receiver operating characteristic (ROC) curves were generated to evaluate the predictive value of AMH in diagnosing PCOS. Results The PCOS group demonstrated significantly higher blood lipid, luteinizing hormone (LH), and AMH levels than the control group. Glucose and lipid metabolism and hormonal disorders in the PCOS group were more significant than in the control group among individuals with and without obesity. LH, TSTO, and AMH were identified as independent risk factors for PCOS. AMH along with LH, and antral follicle count demonstrated a high predictive value for diagnosing PCOS. Conclusion AMH exhibited robust diagnostic use for identifying PCOS and could be considered a marker for screening PCOS to improve PCOS diagnostic accuracy. Attention should be paid to the effect of glucose and lipid metabolism on the hormonal and related parameters of PCOS populations.

In this report, a comprehensive review of the mechanisms, efficiency, and emerging technologies associated with ZVM-based water treatment is provided. The underlying objectives for which the review aimed to address include (i) providing an understanding of the ZVMs used in water treatment applications and their properties, (ii) reviewing the mechanisms employed by ZVMs to sequestrate contaminants, (iii) evaluating the efficiency of ZVMs in the removal of contaminants and (iv) exploring the various emerging technologies used in ZVM-based water treatment and to provide some recommendations for future research.It was concluded from the work that ZVMs abate contaminants found in wastewater through an interplay and synergy of physical, chemical and catalytic mechanisms. ZVMs, when used with other treatment techniques, provide better benefits in the treatment of diverse contaminants. To help achieve the full scale utilization of ZVMs potential, sustained research efforts combined with innovative approaches are needed for sustainable and efficient water treatment solutions. The review offers insights into technologies needed to eliminate diverse contaminants from wastewater, addressing important considerations regarding sustainability and future directions of ZVM-based water treatment technologies.

Green hydrogen production from water is one attractive route to non-fossil fuel and a potential source of clean energy. Hydrogen is not only a zero-carbon energy source but can also be utilized as an efficient storage of electrical energy generated through various other sources, such as wind and solar. Cost-effective and environmentally benign direct hydrogen production through neutral water (∼pH 7) reduction is particularly challenging due to the low concentration of protons. There is currently a major need for easy-to-prepare, robust, as well as active electrode materials. Herein we report three new molecular electrodes that were prepared by anchoring commercially available, and environmentally benign cobalt-containing electrocatalysts with three different ligand frameworks (porphyrin, phthalocyanine, and corrin) on a structurally modified graphite foil surface. Under the studied reaction conditions (over 7 h at 22°C), the electrode with Co-porphyrin is the most efficient for the water reduction with starting ∼740 mV onset potential (OP) (vs. RHE, current density 2.5 mA/cm2) and a Tafel slope (TS) of 103 mV/dec. It is followed by the molecular electrodes having Co-phthalocyanine [825 mV (OP), 138 mV/dec (TS)] and Vitamin-B12 (Co-corrin moiety) [830 mV (OP), 194 mv/dec (TS)]. A clear time-dependent improvement (>200 mV over 3 h) in the H2 production overpotential with the Co-porphyrin-containing cathode was observed. This is attributed to the activation due to water coordination to the Co-center. A long-term chronopotentiometric stability test shows a steady production of hydrogen from all three cathode surfaces throughout seven hours, confirmed using an H2 needle sensor. At a current density of 10 mA/cm2, the Co-porphyrin-containing electrode showed a TOF value of 0.45 s−1 at 870 mV vs. RHE, whereas the Co-phthalocyanine and Vitamin-B12-containing electrodes showed 0.37 and 0.4 s−1 at 1.22 V and 1.15 V (vs. RHE), respectively.

Background: To evaluate the efficacy and safety of allogenic CD8 + natural killer T (CD8+ NKT) immunotherapy combined with gefitinib in the treatment of advanced or metastatic EGFR mutant non-small cell lung cancer (NSCLC). Methods: This study is prospective. The NSCLC patients with exon 19 (Ex19del) or exon 21 L858R point mutations, and response to gefitinib treatment were enrolled into the trial to be randomly assigned into the gefitinib arm and the gefitinib/NKT arm. Allogenic CD8+ NKT cells were cultured in vitro and adaptive transferred into the patients via vein in the gefitinib/NKT arm. The primary endpoint was progression-free survival (PFS). Secondary endpoint analysis included time to disease progression (TTP), overall survival (OS), levels of serum tumour markers for carcinoembryonic antigen (CEA) and alanine aminotransferase (ALT) in the blood, the response rate and safety. From July 2017 to June 2021, 19 patients were randomly assigned to the gefitinib arm (n = 8) and the gefitinib/NKT arm (n = 11). Results: The estimated median survival PFS in the gefitinib/NKT arm was significantly longer than that of the gefitinib arm (12 months vs 7 months). Similar results were also observed for the median TTP. Moreover, the gefitinib/NKT arm had better CEA control than the gefitinib arm. Clinical grade 3 adverse reactions occurred in 64% and 39% of patients in the gefitinib/NKT arm and the gefitinib arm, respectively. The most common grade 3 adverse events in the gefitinib/NKT arm included abnormal liver function in 8 cases (73%) and diarrhoea in 1 case (9%), both of which resolved after drug intervention. Conclusion: The PFS of EGFR-mutated advanced NSCLC treated with allogenic CD8+ NKT cells combined with gefitinib was longer than that of gefitinib alone. No obvious serious adverse reactions occurred, and the patients compliance and survival status were good.

Abstract Wearing face masks is the best way to stop the spread of respiratory infections. However, if masks are not sterilized, changing them too frequently can actually increase the risk of cross‐contamination. Herein, the construction of an antipathogen photocatalytic mask with carbon vacancy‐modified carbon nitride nanosheets (g‐C3N4‐VC Ns) coated on the non‐woven fabrics of the out layer of the mask, offering effective and long‐term protection against damaging pathogens when exposed to light is reported. The introduced carbon vacancies are found capable of creating energy‐disordered sites and inducing energetic electric force to overcome the Coulomb interactions between electron‐hole pairs, thus promoting the electron‐hole separation to achieve a high generation of reactive oxygen species (ROS). Thanks to its high activity in generating ROS upon exposure to light, the as‐prepared photocatalytic mask shows high pathogen sterilization performance. This, in turn, prolongs the mask's protective lifetime, decreases the need for regular replacement, and decreases medical waste production. The work demonstrated here opens new viewpoints in designing pathogens biocidal protective devices for health protection, offering significant promise in specific environment self‐protection.

39. A Novel RNA Methylation-Related Prognostic Signature and its Tumor Microenvironment Characterization in Hepatocellular Carcinoma

Introduction Hepatocellular carcinoma (HCC) is one of the most common malignant tumors of the digestive system. RNA methylation plays an important role in tumorigenesis and metastasis, which could alter gene expression and even function at multiple levels, such as RNA splicing, stability, translocation, and translation. In this study, we aimed to conduct a comprehensive analysis of RNA methylation-related genes (RMGs) in HCC and their relationship with survival and clinical features. Methods A retrospective analysis was performed using publicly available HCC-related datasets. The differentially expressed genes (DEGs) between HCC and controls were identified from TCGA-LlHC and intersected with RMGs to obtain differentially expressed RNA methylation-related genes (DERMGs). Regression analysis was used to screen for prognostic genes and construct risk models. Simultaneously, clinical, immune infiltration and therapeutic efficacy analyses were performed. Finally, multivariate cox regression was used to identify independent risk factors, and quantitative real-time polymerase chain reaction (qRT-PCR) was used to validate the expression levels of the core genes of the model. Results A 21-gene risk model for HCC was established with excellent performance based on ROC curves and survival analysis. Risk scores correlated with tumor grade, pathologic T, and TNM stage. Immune infiltration analysis showed correlations with immune scores, 11 immune cells, and 30 immune checkpoints. Low-risk patients showed a higher susceptibility to immunotherapy. The risk score and TNM stage were independent prognostic factors. qRT-PCR confirmed higher expression of PRDM9, ALPP, and GAD1 in HCC. Conclusions This study identified RNA methylation-related signature genes in HCC and constructed a risk model that predicts patient outcomes and reflects the immune microenvironment. Prognostic genes are involved in complex regulatory mechanisms, which may be useful for cancer diagnosis, prognosis, and therapy.

The closed-loop supply chain (CLSC) focusing on product recycling is getting more and more attention. The question of how to motivate CLSC participants to implement product recycling effectively has come into focus. We study the optimal strategy of a two-echelon CLSC under different incentive mechanisms, characterize the return rate in the form of a state equation for its dynamic behavior and construct a closed-loop supply chain dynamic model. Subsequently, we discuss two incentives—the cooperative promotion incentive and wholesale price discount incentive—based on the no-incentive game and investigate the optimal strategy, steady-state return rate, and revenue of each CLSC player in the three scenarios. Then, the choice of incentives by manufacturers and retailers under different scenarios is determined by comparative analysis. The results indicate that retailers always favor the cooperative promotion incentive. For manufacturers, the wholesale price discount incentive is more advantageous only when the recycling efforts outperform retailers’ promotion efforts in unit cost. In most other cases, the cooperative promotional incentive is more popular since it will generate more revenue for each participant and the entire supply chain. Moreover, there is a Profit-Pareto-improvement region when the CLSC implements the cooperative promotion incentive.

41. Experimental investigation on calcium dissolution performance of shotcrete with different waterproofing agents

The crystalline blockage of tunnel drainage pipes poses a common challenge to tunnel engineering, particularly in regions with abundant groundwater and severe ion erosion. Based on the design of concrete mix proportion, this paper improves the calcium ion erosion resistance of shotcrete by adding permeable crystalline sodium methyl silicate waterproofing agent and silane waterproofing agent. Concurrently, the mechanical properties and microscopic characterization of waterproofing agent concrete are carried to determine the optimal dosage for practical engineering applications. The experimental results reveal that the sodium methyl silicate waterproofing agent has minimal impact on the calcium ion dissolution resistance of shotcrete. In contrast, the silane waterproofing agent significantly enhances the calcium dissolution performance of shotcrete, and the optimum dosage is identified as 0.4%. During long-term maintenance, the silane waterproofing agent proves more effective in improving the compressive strength of shotcrete compared to the sodium methyl silicate waterproofing agent, showing an increase rate exceeding 20%. Both waterproofing agents contribute to the enhancement of the tensile strength of shotcrete, and the enhancement effect is consistent with the compressive strength. The permeable crystalline waterproofing agent expedites the cement hydration process, resulting in the formation of higher-density calcium silicate hydrates (C-S-H) gel and ettringite (AFt). These substances fill the pores of shotcrete leading to a more compact internal structure. The calcium ion dissolution and mechanical strength test results collectively validate the superior effectiveness crystallization prevention of aterproofing agents . This research provides valuable insights for addressing the tunnel crystallization phenomenon caused by the calcium dissolution.
