Abstract Ginseng, from the roots of Panax ginseng C. A. Meyer, is a widely used herbal medicine in Asian countries, known for its excellent therapeutic properties. The growth of P. ginseng is depend on specific and strict environments, with a preference for wetness but intolerance for flooding. Under excessive soil moisture, some irregular rust-like substances are deposited on the root epidermis, causing ginseng rusty symptoms (GRS). This condition leads to a significant reduce in yield and quality, resulting in substantial economic loses. However, there is less knowledge on the cause of GRS and there are no effective treatments available for its treatment once it occurs. Unsuitable environments lead to the generation of large amounts of reactive oxygen species (ROS). We investigated the key indicators associated with the stress response during different physiological stages of GRS development. We observed a significant change in ROS level, MDA contents, antioxidant enzymes activities, and non-enzymatic antioxidants contents prior to the GRS. Through the analysis of soil features with an abundance of moisture, we further determined the source of ROS. The levels of nitrate reductase (NR) and nitric oxide synthase (NOS) activities in the inter-root soil of ginseng with GRS were significantly elevated compared to those of healthy ginseng. These enzymes boost nitric oxide (NO) levels, which in turn showed a favorable correlation with the GRS. The activities of nicotinamide adenine dinucleotide phosphate (NADPH) oxidase first rose and then decreased as GRS developed. Excess soil moisture causes a decrease in oxygen levels. This activated NR and NOS in the soil, resulting in a production of excess NO. The NO then diffused into the ginseng root and triggered a burst of ROS through NADPH located on the cell membrane. Additionally, Fe2+ in soil was oxidized to red Fe3+, and finally led to GRS. This conclusion was also verified by the Sodium Nitroprusside (SNP), a precursor compound producing NO. The presence of NO from NR and NOS in water-saturated soil is responsible for the generation of ROS. Among these, NO is the main component that contribute to the occurrence of GRS.

Introduction: Global warming is increasing interest in how aquatic animals can adjust their physiological performance and cope with temperature changes. Therefore, understanding the behavioral changes and molecular underpinnings in fish under warming is crucial for both the individual and groups survival. This could provide experimental evidence and resource for evaluating the impact of global warming. Objective: Three genetic families of common carp (Cyprinus carpio) were generated. These juveniles were constructed short-term (4 days) and long-term (30 days) warming groups to investigate the effects of warming on behavioral responses and to elucidate the potential underlying mechanisms of warming-driven behavior. Methods: Behavioral tests were used to explore the effects of short- and long-term exposure to warming on the swimming behavior of C. carpio. Brain transcriptome combined with measurement of nervous system activity was used to further investigated the comprehensive neuromolecular mechanisms under warming. Results: Long-term warming groups had a more significant impact on the decline of swimming behavior in juvenile C. carpio. Furthermore, brain comparative transcriptomic analysis combined with measurement of nervous system activity revealed that genes involved in cytoskeletal organization, mitochondrial regulation, and energy metabolism are major regulators of behavior in the juvenile under warming. Importantly, especially in the long-term warming groups, enrichment analysis of associated gene expression suggested functional alterations of synaptic transmission and signal transduction leading to swimming function impairment in the central nervous system, as revealed by behavioral tests. Conclusions: Our study provides evidence of the neurogenomic mechanism underlying the decreased swimming activity in juvenile C. carpio under warming. These findings have important implications for understanding the impacts of climate change on aquatic ecosystems and the organisms that inhabit them.

Background: The breeding of high-quality, high-yield, and disease-resistant varieties is closely related to food security. The investigation of breeding results relies on the evaluation of seed phenotype, which is a key step in the process of breeding. In the global digitalization trend, digital technology based on optical sensors can perform the digitization of seed phenotype in a non-contact, high throughput way, thus significantly improving breeding efficiency. Aim of review: This paper provides a comprehensive overview of the principles, characteristics, data processing methods, and bottlenecks associated with three digital technique types based on optical sensors: spectroscopy, digital imaging, and three-dimensional (3D) reconstruction techniques. In addition, the applicability and adaptability of digital techniques based on the optical sensors of maize seed phenotype traits, namely external visible phenotype (EVP) and internal invisible phenotype (IIP), are investigated. Furthermore, trends in future equipment, platform, phenotype data, and processing algorithms are discussed. This review offers conceptual and practical support for seed phenotype digitization based on optical sensors, which will provide reference and guidance for future research. Key scientific concepts of review: The digital techniques based on optical sensors can perform non-contact and high-throughput seed phenotype evaluation. Due to the distinct characteristics of optical sensors, matching suitable digital techniques according to seed phenotype traits can greatly reduce resource loss, and promote the efficiency of seed evaluation as well as breeding decision-making. Future research in phenotype equipment and platform, phenotype data, and processing algorithms will make digital techniques better meet the demands of seed phenotype evaluation, and promote automatic, integrated, and intelligent evaluation of seed phenotype, further helping to lessen the gap between digital techniques and seed phenotyping.

The keyhole phenomenon, frequently found in LPBF, is critically responsible for the formation of internal porosities which degrades the part quality. Owing to the ultra-high transience and complexity of the LPBF process, the underlying mechanisms during the keyholing process have not been well understood. In this work, a Multiphysics-based CFD model is developed to simulate the keyhole-penetration process in LPBF of CuCrZr alloy, and the ray-tracing laser heat source is implemented to visualize the multiple laser reflections. The simulation results are well validated in terms of the track size, porosity distribution and laser absorption by conducting the single-track experiment, X-CT scanning and calorimetric measurement, respectively. The flow patterns, driving forces and laser irradiation behaviors in the keyhole mode are collectively elucidated to understand the formation mechanisms of the keyhole-induced pores. The reduction of laser ray entries into the emerging cavity with a shrinking opening is noticed and contributes to the pore formation.

In this research, we report fabrication of AZ91 (Mg–Al–Mn–Zn) matrix composites reinforced with a hybrid of hexagonal boron nitride nanoplatelets (BNNP) and nanotubes (BNNT) using spark plasma sintering (SPS) and hot isostatic pressing (HIP). The resultant composites present dense microstructure featured by the refined α-Mg grains decorated with eutectic along the α-Mg grain boundaries, and trace amounts of in-situ interfacial-reaction products such as Mg3N2 and MgB2 are observed to serve as chemical anchors for stronger interface. More importantly, the AZ91 composites reinforced with a hybrid of 0.4 wt% BNNPs-0.1 wt% BNNTs displays ∼209 MPa of compressive yield strength, ∼329 MPa of ultimate compressive strength and ∼19.4% of elongation, achieving satisfactory balance of strength and ductility when compared to the AZ91 composite reinforced with sole BNNPs. This improvement can be attributed to the unique three-dimensional hybrid structure, where BNNPs maintain their high specific surface area, and the de-bundled BNNTs extend like tentacles, further contributing to their interfacial adhesion with the matrix. This research is expected to pave the way for the development of Mg-based composites with an optimal balance of strength and ductility.

The Yangtze River is the mother river of China. To promote the aquatic ecosystem protection of the great river, the Project of Yangtze Fisheries Resources and Environment Investigation (2017–2021) supported by the Ministry of Agriculture and Rural Affairs, P. R. China carried out by 24 institutes and universities that located in the Yangtze River basin surveys the status of (1) fish species composition and spatial distribution, (2) current fish abundance, (3) endangered fishes, (4) Yangtze finless porpoise, (5) aquatic eco-environments, (6) water-level fluctuation areas, (7) capture fisheries and recreational fisheries of the Yangtze River mainstream and 10 of its main tributaries, including Yalong River, Heng River, Min River (including Dadu River), Chishui River, Tuo River, Jialing River, Wu River, Han River, Dongting Lake and Poyang Lake. The results showed that there were 443 fish species (belonging to 163 genus, 37 families, and 18 orders) before 2017, but only 323 fish species (including 15 new recorded exotic species) were recorded in the project of 2017–2021. Among them, Cyprinus carpio, Carassius auratus, Hypophthalmichthys molitrix, Pelteobagrus fulvidraco, Coilia brachygnathus, Silurus asotus, Saurogobio dabryi, Ctenopharyngodon idellus, Pelteobagrus nitidus, Hemiculter leucisculus, Siniperca chuatsi, Coreius heterodon, Culter alburnus, Parabramis pekinensis, and Aristichthys nobilis were the dominant species across the whole Yangtze River system. It is estimated that there were 886 million individuals weighing 124.8 million kg, merely equivalent to 27.3% of the resources in 1950s, 30.9% of the resources in 1960s, or 58.7% of the resources in 1980s. In the new list of protected fishes that recorded in the Yangtze River system, only 15 of 29 were collected in this project of 2017–2021. Psephurus gladius has been affirmed to be Extinct by International Union for Conservation of Nature (IUCN). The wild individuals of Tenualosa reevesii and Luciobrama macrocephalus have disappeared for many years and maybe have been extinct already. Acipenser dabryanus has been affirmed to be Extinct in the Wild by IUCN. The natural propagations of A. sinensis, Myxocyprinus asiaticus and Trachidermus fasciatus have been interrupted for many years. The populations of Yangtze finless porpoise in the Yangtze River mainstream, Dongting Lake and Poyang Lake have steadily rising sizes and expanding distributions in 2017–2021. Parts of them migrate from one region to another with the seasons, which would result in the fluctuation of Yangtze finless porpoise population within some regions. The conventional indicators of water quality in the Yangtze River system were good and conformed to the water quality criteria of fishery in 2017–2021. In the last 40 years, the maximum surface water area in the Yangtze River basin extended to approximately 63,360 km2, the minimum surface water area covered approximately 26,396 km2, and the seasonal water-level fluctuation areas occupied approximately 36,964 km2. Compared with 1984–2000 period, the 2001–2020 period witnessed an overall decreasing trend in the frequency of surface water occurrence within about 25,869 km2 of aquatic areas. From 1984 to 2000 period to 2001–2020 period, permanent surface water has decreased by nearly 8,750 km2. In 2017, the fishermen were mainly 40–60 years old and their educational levels were mainly lower than junior high school. In 2017, most anglers were older than 40 and used hand rod and/or sea rod. Their average catch of each time was mainly less than 1 kg. Results suggested that the fishing ban in key waters of the Yangtze River basin is not only an ecological project of aquatic ecosystem conservation, but also a livelihood project for the fisherman to embrace industrial transformation and improve their living conditions. After the implementation of the fishing ban, the fisheries resources would gradually recover, and the Yangtze finless porpoise population size would also see a steady increase. However, the endangered species would remain threatened for a long time. The degraded waterbodies and water-level fluctuation areas would be the key restrictive factors for future aquatic ecosystem recovery in the Yangtze River basin. Since this survey was carried out before the fishing ban, the current results could provide a baseline for future evaluation of the effect of the Yangtze River fishing ban.

Abstract Background This study aimed to evaluate the causal relationship between Interleukin-27 (IL-27) and osteoporosis by bidirectional Mendelian randomization (MR) analysis. Methods Firstly, the genome-wide association study summary data of osteoporosis (finn-b-M13_OSTEOPOROSIS) and IL-27 levels (ebi-a-GCST90012017) were picked out from the Integrative Epidemiology Unit (IEU) OpenGWAS database. After filtrating instrumental variables (IVs), the bidirectional MR analysis between IL-27 levels and osteoporosis was performed by MR-Egger, Weighted median, Simple mode, Weighted mode, and Inverse variance weighted (IVW). Subsequently, the sensitivity analysis was adopted to evaluate the reliability of the MR results via the Heterogeneity, Horizontal pleiotropy test and Leave-One-Out (LOO) analysis. Finally, the enrichment analysis of genes corresponding to SNPs related to IL-27 levels derived from eQTLGen database was executed to explore in depth the biological function and regulatory mechanism of these genes on osteoporosis occurrence. Results The bidirectional MR results based on IVW method revealed that IL-27 level as a risk factor was causally related to osteoporosis (P = 0.004, odds ratio (OR) = 1.123, 95% confidence interval (CI) = 1.037–1.217), whereas osteoporosis was not in significant connection with IL-27 levels (P > 0.05). In regard to the sensitivity analysis for forward MR results, there was no heterogeneity and horizontal pleiotropy, and no SNPs relevant to IL-27 levels existed severe bias, suggesting the reliability of forward MR analysis. Furthermore, a total of 74 genes corresponding to 26 SNPs of IL-27 levels were obtained and were mainly involved in immune and inflammatory pathways including MyD88-dependent toll-like receptor signaling pathway, Toll-like receptor signaling pathway, cytosolic DNA-sensing pathway and so forth. Conclusions This study supported that IL-27 level as a risk factor was causally connected with osteoporosis and might regulate the disease occurrence and progression by means of immune and inflammatory mechanisms, which could provide important reference and evidence for further exploring the role of IL-27 in the development of osteoporosis.

Abstract Quantitative structure–property relationships have been extensively studied in the field of predicting retention times in liquid chromatography (LC). However, making transferable predictions is inherently complex because retention times are influenced by both the structure of the molecule and the chromatographic method used. Despite decades of development and numerous published machine learning models, the practical application of predicting small molecule retention time remains limited. The resulting models are typically limited to specific chromatographic conditions and the molecules used in their training and evaluation. Here, we have developed a comprehensive dataset comprising over 10,000 experimental retention times. These times were derived from 30 different reversed-phase liquid chromatography methods and pertain to a collection of 343 small molecules representing a wide range of chemical structures. These chromatographic methods encompass common LC setups for studying the retention behavior of small molecules. They offer a wide range of examples for modeling retention time with different LC setups.

Abstract Numerous studies have shown that the Krüppel-like factors (KLFs) family of transcription factors regulate various eukaryotic physiological processes including the proliferation, differentiation, senescence, death, and carcinogenesis of animal cells. In addition, they are involved in the regulation of key biological processes such as cell cycle, DNA repair, and immune response. Current studies focus on investigating the role of KLFs in normal physiological conditions and the incidence and development of diseases. However002C the significance of KLFs family genes in clear cell renal cell carcinoma (ccRCC) remains partly understood; therefore, an in-depth investigation of their role and clinical value in this cancer is desired. The study aimed to investigate the role of KLF family genes in the incidence, development, and prognosis of ccRCC, and to identify the related potential biomarkers and therapeutic targets. The expression of KLFs in the RNA sequencing data of 613 ccRCCs from the TCGA database was analyzed using R software, and UALCAN and GEPIA assessed the expression of KLF genes in ccRCC. Real-time fluorescence quantitative PCR analysis was performed using 10 pairs of paired ccRCC sample tissues and renal cancer cell lines from the First Affiliated Hospital of Nanchang University. Overall survival (OS), progression-free interval (PFI), and disease-specific survival (DSS) of Kidney Clear Cell Carcinoma (KIRC) samples at differential expressions of KLFs in the TCGA database were analyzed using the R software, followed by generating a nomogram prediction model. GSCALite assessed the interactions of KLF genes with miRNAs and generated network maps. Protein interaction network maps of 50 neighboring genes associated with KLF mutations were analyzed using STRING with GO and KEGG functional enrichment analyses. The cBioPortal determined the probability of KLF gene mutations and their impact on OS and disease-free survival (DFS) in patients with ccRCC. Immune cell infiltration of KLFs was analyzed using TIMER. Finally, GSCALite was used to analyze the drug sensitivity and associated pathways of action of KLFs. Correlation validation using cellular experiments. KLF3/5/9/15 were significantly downregulated in ccRCC tissues, whereas KLF16/17 were upregulated compared with the adjacent tissues. Patients with high mRNA levels of KLF16/17 showed significantly lower OS, PFI, and DSS, whereas KLF3/5/9 showed a reverse trend. In patients with ccRCC, a significant correlation was observed between KLF mutations and OS and DSS. Furthermore, the correlation of KLF3/5/9 with immune cell infiltration was stronger than that of KLF15/16, while KLF17 was significantly associated with the Epithelial-Mesenchymal Transition (EMT) pathway. Overexpression of KLF5 inhibits the proliferative and migratory capacity of renal cancer cells (786-O and OS-RC-2), as well as their sensitivity to relevant small molecule drugs. Our research revealed the expression levels and biological significance of KLF genes in ccRCC, particularly highlighting the potential of KLF5 as a promising biomarker and therapeutic target for effective prognosis and diagnosis of ccRCC.

In order to enhance the driving ability of autonomous vehicles on structured roads and enable them to plan safe and comfortable paths, we propose an obstacle avoidance path strategy for autonomous vehicles based on genetic algorithm. The use of Frenet-Serret enhances the adaptability of the algorithm in complex environments. In order to improve the generation and optimisation of obstacle avoidance trajectory, we establish an anti-collision model. When the vehicle faces a potential collision with an obstacle, the genetic algorithm quickly iterates and selects the first nine genes to generate the rough solution and convex space of the path. Combined with convex space, the quadratic programming method will numerically optimise the generated rough solution to generate an accurate path that satisfies the constraints. In addition, in order to ensure the safety and comfort in the process of obstacle avoidance, based on the dynamic constraints of the vehicle, the speed planning is used to determine the speed curve. We simulate in various scenarios involving moving obstacles. The real-time simulation based on the HIL platform proves that the proposed path planning strategy is effective in various driving scenarios.

Abstract Automated fiber placement (AFP) enables the efficient and precise fabrication of complex-shaped aerospace composite structures with lightweight and high-performance properties. However, due to the excessive compression on the inner edge of the tow placed along the curved trajectory, the resulting defects represented by buckling and wrinkles in spatial tow steering can induce poor manufacturing accuracy and quality degradation of products. In this paper, a theoretical model of tow buckling based on the first-order shear deformation laminate theory, linear elastic adhesion interface and Hertz compaction contact theory is proposed to analyze the formation mechanism of the wrinkles and predict the formation of defects by solving the critical radius of the trajectory, and finite element analysis involving the cohesive zone modeling (CZM) is innovated to simulate the local buckling state of the steered tow in AFP. Additionally, numerical parametric studies and experimental results indicate that mechanical properties and geometric parameters of the prepreg, the curvature of the placement trajectory and critical process parameters have a significant impact on buckling formation, and optimization of process parameters can achieve effective suppression of placement defects. This research proposes a theoretical modeling method for tow buckling, and conducts in-depth research on defect formation and suppression methods based on finite element simulation and placement experiments.

Abstract The filamentous ‘Pf’ bacteriophages of Pseudomonas aeruginosa play roles in biofilm formation and virulence, but mechanisms governing Pf prophage activation in biofilms are unclear. Here, we identify a prophage regulatory module, KKP (kinase-kinase-phosphatase), that controls virion production of co-resident Pf prophages and mediates host defense against diverse lytic phages. KKP consists of Ser/Thr kinases PfkA and PfkB, and phosphatase PfpC. The kinases have multiple host targets, one of which is MvaU, a host nucleoid-binding protein and known prophage-silencing factor. Characterization of KKP deletion and overexpression strains with transcriptional, protein-level and prophage-based approaches indicates that shifts in the balance between kinase and phosphatase activities regulate phage production by controlling MvaU phosphorylation. In addition, KKP acts as a tripartite toxin-antitoxin system that provides defense against some lytic phages. A conserved lytic phage replication protein inhibits the KKP phosphatase PfpC, stimulating toxic kinase activity and blocking lytic phage production. Thus, KKP represents a phosphorylation-based mechanism for prophage regulation and antiphage defense. The conservation of KKP gene clusters in >1000 diverse temperate prophages suggests that integrated control of temperate and lytic phage infection by KKP-like regulatory modules may play a widespread role in shaping host cell physiology.

Abstract Subarachnoid hemorrhage (SAH) is one of the most severe type of cerebral strokes, which can cause multiple cellular changes in the brain leading to neuronal injury and neurological deficits. Specifically, SAH can impair adult neurogenesis in the hippocampal dentate gyrus, thus may affecting poststroke neurological and cognitive recovery. Here, we identified a non-canonical role of milk fat globule epidermal growth factor 8 (MFGE8) in rat brain after experimental SAH, involving a stimulation on adult hippocampal neurogenesis(AHN). Experimental SAH was induced in Sprague-Dawley rats via endovascular perforation, with the in vivo effect of MFGE8 evaluated via the application of recombinant human MFGE8 (rhMFGE8) along with pharmacological interventions, as determined by hemorrhagic grading, neurobehavioral test, and histological and biochemical analyses of neurogenesis related markers. Results: Levels of the endogenous hippocampal MFGE8 protein, integrin-β3 and protein kinase B (p-Akt) were elevated in the SAH relative to control groups, while that of hippocalcin (HPCA) and cyclin D1 showed the opposite change. Intraventricular rhMGFE8 infusion reversed the decrease in doublecortin (DCX) immature neurons in the DG after SAH, along with improved the short/long term neurobehavioral scores. rhMGFE8 treatment elevated the levels of phosphatidylinositol 3-kinase (PI3K), p-Akt, mammalian target of rapamycin (mTOR), CyclinD1, HPCA and DCX in hippocampal lysates, but not that of integrin β3 and Akt, at 24 hr after SAH. Treatment of integrin β3 siRNA, the PI3K selective inhibitor ly294002 or Akt selective inhibitor MK2206 abolished the effects of rhMGFE8 after SAH. In conclusion, MFGE8 is upregulated in the hippocampus in adult rats with reduced granule cell genesis. rhMFGE8 administration can rescue this impaired adult neurogenesis and improve neurobehavioral recovery. Mechanistically, the effect of MFGE8 on hippocampal adult neurogenesis is mediated by the activation of integrin β3/Akt pathway. These findings suggest that exogenous MFGE8 may be of potential therapeutic value in SAH management. Graphical abstract and proposed pathway of rhMFGE8 administration attenuate hippocampal injury by improving neurogenesis in SAH models. SAH caused hippocampal injury and neurogenesis interruption. Administered exogenous MFGE8, recombinant human MFGE8(rhMFGE8), could ameliorate hippocampal injury and improve neurological functions after SAH. Mechanistically, MFGE8 bind to the receptor integrin β3, which activated the PI3K/Akt pathway to increase the mTOR expression, and further promote the expression of cyclin D1, HPCA and DCX. rhMFGE8 could attenuated hippocampal injury by improving neurogenesis after SAH, however, know down integrin β3 or pharmacological inhibited PI3K/Akt by ly294002 or MK2206 reversed the neuro-protective effect of rhMFGE8.

Abstract College students face uncertainties during job searches due to a lack of career planning, unclear objectives, and ineffective search strategies, leading to poor employment outcomes. Fuzzy Control (FC) based Job Search Strategies (JS2) are proposed in this research as an optimized technique named FC-JS2-TSC. This technique combines Takagi-Sugeno (TS) fuzzy inference with Cuckoo (C) search optimization. The primary goals are improving individualized advice and creating an integrated system to deal with job search concerns. The FC uses fuzzy logic and sets to model uncertainties such as vague job desires and ever-changing market circumstances. Individual student profiles and preferences are used to fine-tune methods by cuckoo search. Through experimental validation, we can see that FC-JS2-TSC outperforms previous methods in terms of both job strategy selection and results. As a measure of system efficacy, the results demonstrate a high Cronbach's alpha reliability of 0.96, a low RMSEA of 0.04 and 96.6% regarding job offers. By adjusting tactics in response to uncertainty, the innovative FC-JS2-TSC algorithm facilitates data-driven, personalized decision-making, ultimately leading to more efficient job searches. It has an integrated design that combines optimization with fuzzy logic's uncertainty handling to ensure students have the best possible chance of success in their job searches.

Abstract Accumulated evidence suggested that gut microbial dysbiosis interplayed with progressive chronic kidney disease (CKD). However, no available therapy is effective in suppressing progressive CKD. Here, using microbiomics in 480 participants including healthy controls and patients with stage 1–5 CKD, we identified an elongation taxonomic chain Bacilli-Lactobacillales-Lactobacillaceae-Lactobacillus-Lactobacillus johnsonii correlated with patients with CKD progression, whose abundance strongly correlated with clinical kidney markers. L. johnsonii abundance reduced with progressive CKD in rats with adenine-induced CKD. L. johnsonii supplementation ameliorated kidney lesion. Serum indole-3-aldehyde (IAld), whose level strongly negatively correlated with creatinine level in CKD rats, decreased in serum of rats induced using unilateral ureteral obstruction (UUO) and 5/6 nephrectomy (NX) as well as late CKD patients. Treatment with IAld dampened kidney lesion through suppressing aryl hydrocarbon receptor (AHR) signal in rats with CKD or UUO, and in cultured 1-hydroxypyrene-induced HK-2 cells. Renoprotective effect of IAld was partially diminished in AHR deficiency mice and HK-2 cells. Our further data showed that treatment with L. johnsonii attenuated kidney lesion by suppressing AHR signal via increasing serum IAld level. Taken together, targeting L. johnsonii might reverse patients with CKD. This study provides a deeper understanding of how microbial-produced tryptophan metabolism affects host disease and discovers potential pathways for prophylactic and therapeutic treatments for CKD patients.

Abstract Purpose This current study attempted to investigate whether one-stitch method (OM) of temporary ileostomy influenced the stoma-related complications after laparoscopic low anterior resection (LLAR). Methods We searched for eligible studies in four databases including PubMed, Embase, Cochrane Library, and CNKI from inception to July 20, 2023. Both surgical outcomes and stoma-related complications were compared between the OM group and the traditional method (TM) group. The Newcastle–Ottawa Scale (NOS) was adopted for quality assessment. RevMan 5.4 was conducted for data analyzing. Results Totally 590 patients from six studies were enrolled in this study (272 patients in the OM group and 318 patients in the TM group). No significant difference was found in baseline information (P > 0.05). Cellulose macrofibers (MFs) are gaining increasing interest as natural and biodegradable alternatives to fossil-derived polymers for both structural and functional applications. However, simultaneously achieving their exceptional mechanical performance and desired functionality is challenging and requires complex processing. Here, we reported a one-step approach using a tension-assisted twisting (TAT) technique for MF fabrication from bacterial cellulose (BC). The TAT stretches and aligns BC nanofibers pre-arranged in hydrogel tubes to form MFs with compactly assembled structures and enhanced hydrogen bonding among neighboring nanofibers. The as-prepared BC MFs exhibited a very high tensile strength of 1 057 MPa and exceptional lifting capacity (over 340 000 when normalized by their own weight). Moreover, due to the volume expansion of BC nanofibers upon water exposure, BC MFs quickly harvested energy from environmental moisture to untwist the bundled networks, thus generating a torsional spinning with a peak rotation speed of 884 r/(min·m). The demonstrated rapid and intense actuation response makes the MFs ideal candidates for diverse humidity-response-based applications beyond advanced actuators, remote rain indicators, intelligent switches, and smart curtains.

Abstract The current study aimed to evaluate the effect of lymph node ratio (LNR) on the short-term and long-term outcomes of colorectal cancer (CRC) patients who underwent radical CRC surgery. We retrospectively collected CRC patients who underwent radical surgery from Jan 2011 to Jan 2020 in a single-center hospital. The patients were divided into the high LNR group and the low group according to the median. The baseline information and the short-term outcomes were compared between the high group and the low group. Univariate and multivariate logistic regression was performed to analyze the independent predictors for overall survival (OS) and disease-free survival (DFS). A 1:1 proportional propensity score matching (PSM) was used to reduce the selection bias between the two groups. Kaplan–Meier method was used to estimate the OS and DFS between the two groups in different T stages. A total of 1434 CRC patients undergoing radical surgery were enrolled in this study, and there were 730 (50.9%) patients in the low LNR group and 704 (49.1%) patients in the high LNR group. Abstract Background The relationship between obstructive sleep apnea (OSA) and the occurrence of arrhythmias and heart rate variability (HRV) in hypertensive patients is not elucidated. Our study investigates the association between OSA, arrhythmias, and HRV in hypertensive patients. Methods We conducted a cross-sectional analysis involving hypertensive patients divided based on their apnea-hypopnea index (AHI) into two groups: the AHI ≤ 15 and the AHI > 15. All participants underwent polysomnography (PSG), 24-hour dynamic electrocardiography (DCG), cardiac Doppler ultrasound, and other relevant evaluations. Results The AHI > 15 group showed a significantly higher prevalence of frequent atrial premature beats and atrial tachycardia (P = 0.030 and P = 0.035, respectively) than the AHI ≤ 15 group. Time-domain analysis indicated that the standard deviation of normal-to-normal R-R intervals (SDNN) and the standard deviation of every 5-minute normal-to-normal R-R intervals (SDANN) were significantly higher in the AHI > 15 group (P = 0.020 and P = 0.033, respectively). Frequency domain analysis revealed that the low-frequency (LF), high-frequency (HF) components, and the LF/HF ratio were also significantly elevated in the AHI > 15 group (P 15 group (P 15 are at an increased risk for atrial arrhythmias and left atrial dilation, with HRV significantly correlating with OSA severity.

Abstract Background Pruning is an important cultivation management option that has important effects on peach yield and quality. However, the effects of pruning on the overall genetic and metabolic changes in peach leaves and fruits are poorly understood. Results The transcriptomic and metabolomic profiles of leaves and fruits from trees subjected to pruning and unpruning treatments were measured. A total of 20,633 genes and 622 metabolites were detected. Compared with those in the control, 1,127 differentially expressed genes (DEGs) and 77 differentially expressed metabolites (DEMs) were identified in leaves from pruned and unpruned trees (pdLvsupdL), whereas 423 DEGs and 29 DEMs were identified in fruits from the pairwise comparison pdFvsupdF. The content of three auxin analogues was upregulated in the leaves of pruned trees, the content of all flavonoids detected in the leaves decreased, and the expression of almost all genes involved in the flavonoid biosynthesis pathway decreased. The phenolic acid and amino acid metabolites detected in fruits from pruned trees were downregulated, and all terpenoids were upregulated. The correlation analysis revealed that DEGs and DEMs in leaves were enriched in tryptophan metabolism, auxin signal transduction, and flavonoid biosynthesis. DEGs and DEMs in fruits were enriched in flavonoid and phenylpropanoid biosynthesis, as well as L-glutamic acid biosynthesis. Conclusions Pruning has different effects on the leaves and fruits of peach trees, affecting mainly the secondary metabolism and hormone signalling pathways in leaves and amino acid biosynthesis in fruits.

Bamboo nodes (BNs) is a rare disease detected in the Department of Otorhinolaryngology. Here, we reported two cases of BNs, highlighting a potential association of BNs with autoimmune diseases. Upon presentation, the primary concerns of both patients were hoarseness and foreign body sensation. In both cases a co-existence between BNs and an autoimmune disease was identified. In one case, BNs were initially diagnosed while undifferentiated connective tissue disease was later identified by serological testing. In contrast, the other patient did not show any BN-related symptom until two months of therapy for undifferentiated connective tissue disease. Both cases received oral administration of medicines for immunotherapy, with one taking voice training. Both recovered from the larynx symptoms after 10 to 12 months of therapy. This report reminded us of the potential association of BNs with autoimmune diseases. Voice training may be an effective adjuvant intervention for the treatment of BNs.

In this study, headspace solid-phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS) and liquid chromatography-tandem mass spectrometry (LC-MS/MS) was employed to analyze the composition of volatile organic compounds (VOCs) and lipids in pot-stewed duck from three breeds, Cherry Valley, Jingjiang and Muscovy. A total of 53 VOCs and 3 538 lipids were identified. In addition, 16 characteristic VOCs were identified, and 1 387 potential lipid markers were selected to differentiate the three stewed ducks. Network analysis and Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment analysis revealed that phospholipids were closely linked to lipids and unsaturated fatty acids with more than 18 carbon atoms may be the major degradation products of lipids. The results of correlation analysis showed that VOCs were closely related to lipids, and phospholipid markers, mainly composed of phosphatidylcholine (PC) and phosphatidylethanolamine (PE), may contributed to the formation of characteristic VOCs. Cluster analysis showed that the flavor and lipid compositions of stewed Jingjiang and Muscovy ducks were relatively similar. This study may provide a theoretical basis for the flavor regulation of pot-stewed duck and the selection of duck breeds for the production of pot-stewed duck.

The Portland cement concrete pavement (PCCP) often suffers from different environmental distresses and vehicle load failure, resulting in slab corner fractures, potholes, and other diseases. Rapid repair has become one of the effective ways to open traffic rapidly. In this study, a novel type of rapid repair material, basalt fiber reinforced polymer modified magnesium phosphate cement (BFPMPC), is used to rapidly repair PCCP. Notably, the mechanical properties and characteristics of the repair interfaces which are named interfacial transition zones (ITZs) formed by BFPMPC and cement concrete are focused on as a decisive factor for the performance of the rapid repair. The changing trend of the elastic moduli was studied by nanoindentation experiments in the ITZs with the deconvolution analysis that the elastic moduli of certain kinds of substances can be determined. The experimental results show that the elastic modulus of ITZ-1 with a width of about 20 μm can be regarded as 0.098 times of the aggregate, and 0.51 times of the ordinary Portland cement (OPC) mortar. The BFPMPC-OPC mortar ITZ has roughly the same mechanical properties as the ITZ between aggregate and BFPMPC. A multi-scale representative two-dimensional model was established by random aggregate and a two-dimensional extended finite element method (XFEM) to study the mechanical properties of the repair interface. The simulation results show that the ITZ formed by the interface of BFPMPC and OPC mortar and basalt aggregate is the most vulnerable to failure, which is consistent with the nano-indentation experimental results.

Abstract In order to improve the driving ability of the explosion wave simulation equipment, reduce the erosion effect of condensed explosives on the explosion wave simulation equipment, improve the safety of the test process, and make better use of the meteorological detonation driving method, it is necessary to optimize the source of the shock wave load in the driving section. Based on the finite volume method of FLACS, a methane detonation driving model corresponding to the test is established to explore the feasibility of using methane as an explosion source to test the structure against explosion shock wave. A methane detonation drive test was carried out to verify the accuracy of the numerical model. Finally, an engineering model for attenuation of shock wave overpressure peak value in pipeline is established by dimensional analysis, and the model coefficient is determined by numerical simulation and test data. The results show that the blast pressure is the highest when the methane volume ratio reaches 9.5 vol% in the methane-air mixture. Simply increasing oxygen content has little effect on the peak overpressure and positive pressure duration of shock wave. In the pure oxygen environment, the detonation effect can be achieved when the volume ratio of methane to oxygen is 1:2, and the incident pressure of the shock wave is proportional to the volume of the gas cloud. When the gas cloud volume is constant, a reasonable selection of methane-oxygen mixture ratio can achieve a better detonation effect, which can effectively increase the peak overpressure of the shock wave in the test section. The research results can provide technical reference for the development of new explosion wave simulation equipment.

Abstract To address the measurement accuracy challenges posed by the internal flow complexity in atypical circular bend pipes with short turning sections and without extended straight pipe segments, this study designed an experimental circular “S”-shaped bent pipe with a diameter of 0.4 m and a bending angle of 135°. Numerical analysis was used to determine the stable region for velocity distribution within the experimental segment. Furthermore, a novel evaluation method based on the coefficient of variation was proposed to accurately locate the optimal position for installing thermal mass flow meters on the test cross section. Additionally, a formula for calculating the pipeline flow rate based on velocity differences was derived. This formula considers pipeline flow as the dependent variable and uses the velocity at two points in the test cross section as the independent variable. Experimental validation on a primary standard test bench demonstrated that the flow rate calculated by this method had an error controlled within 0.625% compared to the standard flow rate, thus effectively verifying the method’s high accuracy and engineering applicability. This research provides a new testing methodology and practical basis for flow measurement in complex pipeline systems, offering significant guidance for research and applications in related fields.

Abstract The extreme cooling of the Tibetan Plateau (TP) during the boreal winter typically poses threats to the local environment and people’s safety, and it is usually attributed to internal climate variability. Here we demonstrate that the five recent large tropical volcanic eruptions since 1880 have caused an average extreme cooling of up to −0.80 K on the TP in observations during the first boreal winter following the eruptions. This cooling effect is much larger than the global average terrestrial cooling of −0.30 K after the eruptions. The multi-model ensemble mean (MME) of the Atmospheric Model Intercomparison Project (AMIP) runs from Phase 6 of the Coupled Model Intercomparison Project (CMIP6), in which realistic sea surface temperatures (SST) were specified, can simulate an extreme TP cooling response of up to −0.79 K, which is much larger than the direct aerosol cooling of −0.36 K simulated by the historical runs. The positive North Atlantic Oscillation (NAO) anomaly during the post-eruption winter after the eruptions plays a key role in amplifying the TP cooling through atmospheric teleconnection, which overwhelms the warming response associated with the frequently occurring El Niños. The results from this study provide a perspective on the potential contribution of volcanic activity or stratospheric sulfur injection scenarios to specific TP cooling.

Objectives: To evaluate and compare the effectiveness of Endoflas and Zinc Oxide Eugenol (ZOE) as root canal filling materials (RCFMs) for the pulpectomy of deciduous teeth by analyzing multiple clinical and radiographic success and failure follow-ups in previously published studies. Data: All clinical studies that investigated the pulpectomy of the deciduous teeth of children aged 3–9 years. Sources: The databases used for source identification included MEDLINE (via PubMed), Scopus, Web of Science, and the Cochrane Library. No limitations were imposed on the publication year or language. The selection of studies and extraction of relevant study characteristics were conducted from December 26, 2021, to September 7, 2023. Additionally, the risk of bias (RoB) in the included studies was evaluated by using a RoB instrument (RoB 2). Eligible studies were then combined, and a random-effects model was applied by using the maximum likelihood estimations of log risk ratios and their corresponding 95% confidence intervals. Study selection: Of the 3913 records found in the abovementioned databases, nine were eligible for systematic review and eight were eligible for meta-analysis. The studies included 628 pulpectomies of deciduous molar teeth in children. The overall results showed that compared with Endoflas, ZOE was associated with a higher risk ratio for clinical evaluation (LOG[RR] = 0.06, CI 0.03–0.09, p-value 0.001) and radiographic evaluation (LOG[RR] = 0.68, CI 0.35–1.00, p-value 0.001). This association was highly significant at 6- and 9-month follow-ups. Conclusion: Compared with ZOE, Endoflas was associated with a lower risk of the clinical and radiographic failure of deciduous teeth pulpectomy and a 6%–6.8% higher risk ratio, especially at 6- and 9-month follow-ups. Clinical significance: This study suggests the superiority of Endoflas over ZOE as an RCFM for deciduous teeth.

Abstract Bedload studies at the particle scale may help grasp the essence of the problem. Existing studies suffer from short filming durations, limited data volume, and a narrow range of sediment transport intensity variations. This paper employs the high‐speed photography technology and conducts experimental studies on bedload particle motion under 8 different sediment transport intensities. Using the latest image processing technology, over 6 million sediment particle coordinate points and nearly 400,000 particle motion trajectory curves were automatically obtained and used to compare the motion characteristics of bedload particles under different sediment transport intensities. The results show that under low sediment transport intensity, both the number of moving particles and particle motion velocity contribute to the bedload sediment transport rate, while under high‐intensity conditions, the transport rate mainly depends on the number of moving particles. The probability density distribution of sediment transport rate is concentrated and varies within a small range under low‐intensity conditions, exhibiting a tailing phenomenon. In contrast, under high‐intensity conditions, the range of sediment transport rate values increases, and the probability density curve tends to be symmetric, more closely approximating a normal distribution. Additionally, the paper compares the longitudinal and transverse motion velocities of particles and the coefficient of variation of the bedload sediment transport rate.

Background and purpose: Second-line chemotherapy is not effective in the treatment of gastric cancer. Bevacizumab is a molecularly targeted anticancer drug, and sindilizumab is a domestic programmed death-1 (PD-1) inhibitor. The combination of the two is a new direction for clinical treatment of gastric cancer. This study aimed to analyze the effects of bevacizumab assisted PD-1 inhibitor in the treatment of gastric cancer on serum miR-20a-5p and miR-515-3p. Methods: A retrospective study was conducted on 84 patients with gastric cancer treated in the Fourth Affiliated Hospital of Nanjing Medical University from January 2019 to July 2021, and they were divided into the observation group and the control group with 42 cases each according to different treatment plans. The control group was given second-line chemotherapy, and the observation group was given bevacizumab combined with PD-1 inhibitor (sindilizumab) on the basis of the control group. objective response rate (ORR), disease control rate (DCR), serum tumor markers, immune function indexes, serum miR-20a-5p and miR-515-3p levels and total incidence of toxic and side effects were compared between the two groups. This study has been approved by the Ethics Committee of the Fourth Affiliated Hospital of Nanjing Medical University (approval number: 20230531--K061). Results: The ORR (69.05%) and DCR (85.71%) of the observation group were higher than those of the control group (40.48% and 64.29%), and the difference was statistically significant (P0.05). Conclusion: Bevacizumab assisted sindilizumab can effectively improve the treatment efficiency of gastric cancer, improve immune function, and reduce the expression levels of serum miR-20a-5p, miR-515-3p and tumor markers, without increasing toxic and side effects.

Constructing structure-function relationships is critical for the rational design and development of efficient catalysts for CO2 electroreduction reaction (CO2RR). In2O3 is well-known for its specific ability to produce formic acid. However, how the crystal phase and surface affect the CO2RR activity is still unclear, making it difficult to further improve the intrinsic activity and screen for the most active structure. In this work, cubic and hexagonal In2O3 with different stable surfaces ((111) and (110) for cubic, (120) and (104) for hexagonal) are investigated for CO2RR. Theoretical results demonstrate that the adsorption of reactants on cubic In2O3 is stronger than that on hexagonal In2O3, with the cubic (111) surface being the most active for CO2RR. In experiments, synthesized cubic In2O3 nanosheets with predominantly exposed (111) surfaces exhibited a high HCOO– Faradaic efficiency (87.5%) and HCOO– current density (–16.7 mA cm–2) at –0.9 V vs RHE. In addition, an aqueous Zn-CO2 battery based on a cubic In2O3 cathode was assembled. Our work correlates the phases and surfaces with the CO2RR activity, and provides a fundamental understanding of the structure-function relationship of In2O3, thereby contributing to further improvements in its CO2RR activity. Moreover, the results provide a principle for the directional preparation of materials with optimal phases and surfaces for efficient electrocatalysis.

Abstract Macrophages (Mφ) autophagy is a pivotal contributor to inflammation-related diseases. However, the mechanistic details of its direct role in acute kidney injury (AKI) were unclear. Here, we show that Mφ promote AKI progression via crosstalk with tubular epithelial cells (TECs), and autophagy of Mφ was activated and then inhibited in cisplatin-induced AKI mice. Mφ-specific depletion of ATG7 (Atg7Δmye) aggravated kidney injury in AKI mice, which was associated with tubulointerstitial inflammation. Moreover, Mφ-derived exosomes from Atg7Δmye mice impaired TEC mitochondria in vitro, which may be attributable to miR-195a-5p enrichment in exosomes and its interaction with SIRT3 in TECs. Consistently, either miR-195a-5p inhibition or SIRT3 overexpression improved mitochondrial bioenergetics and renal function in vivo. Finally, adoptive transfer of Mφ from AKI mice to Mφ-depleted mice promotes the kidney injury response to cisplatin, which is alleviated when Mφ autophagy is activated with trehalose. We conclude that exosomal miR-195a-5p mediate the communication between autophagy-deficient Mφ and TECs, leading to impaired mitochondrial biogenetic in TECs and subsequent exacerbation of kidney injury in AKI mice via miR-195a-5p-SIRT3 axis.

Abstract We apply the generalised Komar method proposed in [ arXiv:2208.05494 ] to Taub-NUT-AdS spacetime in the theory of Einstein gravity plus a cosmological constant. Based on a generalised closed 2-form, we derive the total mass and NUT charge of the Taub-NUT-AdS spacetime. Together with other thermodynamic quantities calculated through standard method, we conform the first law and Smarr relation. Then, we consider charged AdS NUT spacetimes in Einstein–Maxwell theory with a cosmological constant, and show that the generalised Komar method works, too. We obtain all the thermodynamic quantities, and the first law and Smarr relation are checked to be satisfied automatically.

Abstract Spectrum feature extraction plays a crucial role in identifying seismic events and calculating structural response parameters. However, the criteria for identifying effective modal components in Variational Mode Decomposition (VMD) are not well-defined, resulting in inaccurate spectrum feature extraction. To address this issue, we propose a novel spectrum feature extraction method that combines Allan variance, VMD, and power spectral density (PSD). Firstly, VMD is applied to filter noise components from triaxial accelerometer observations and add effective signals. Secondly, PSD is utilized to extract three groups of seismic frequencies (tri-axial accelerometers). Finally, the Allan method is introduced to identify the group of accelerometer observations with the highest reliability as the vibration frequency caused by the seismic excitation. We validate the effectiveness of our method by analyzing a Mw 2.6 micro-seismic event that occurred in Huairou, Beijing in 2022. The result shows that our proposed method accurately extracts spectrum features of the Great Wall. Specifically, the seismic excitation vibration frequencies at four monitoring stations were found to be 26.95 Hz, 12.89 Hz, 12.89 Hz, and 12.5 Hz. These findings underscore our method's utility in evaluating the Great Wall's structural response to seismic loading, which has significant implications for the conservation and protection of heritage structures.

Abstract Background Infectious bovine rhinotracheitis (IBR), caused by Bovine alphaherpesvirus-1 (BoAHV-1), is an acute, highly contagious disease primarily characterized by respiratory tract lesions in infected cattle. Due to its severe pathological damage and extensive transmission, it results in significant economic losses in the cattle industry. Accurate detection of BoAHV-1 is of paramount importance. In this study, we developed a real-time fluorescent quantitative PCR detection method for detecting BoAHV-1 infections. Utilizing this method, we tested clinical samples and successfully identified and isolated a strain of BoAHV-1.1 from positive samples. Subsequently, we conducted a genetic evolution analysis on the isolate strain’s gC, TK, gG, gD, and gE genes. Results The study developed a real-time quantitative PCR detection method using SYBR Green II, achieving a detection limit of 7.8 × 101 DNA copies/μL. Specificity and repeatability analyses demonstrated no cross-reactivity with other related pathogens, highlighting excellent repeatability. Using this method, 15 out of 86 clinical nasal swab samples from cattle were found to be positive (17.44%), which was higher than the results obtained from conventional PCR detection (13.95%, 12/86). The homology analysis and phylogenetic tree analysis of the gC, TK, gG, gD, and gE genes of the isolated strain indicate that the JL5 strain shares high homology with the BoAHV-1.1 reference strains. Amino acid sequence analysis revealed that gC, gE, and gG each had two amino acid mutations, while the TK gene had one synonymous mutation and one H to Y mutation, with no amino acid mutations observed in the gD gene. Phylogenetic tree analysis indicated that the JL5 strain belongs to the BoAHV-1.1 genotype and is closely related to American strains such as C33, C14, and C28. Conclusions The established real-time fluorescent quantitative PCR detection method exhibits good repeatability, specificity, and sensitivity. Furthermore, genetic evolution analysis of the isolated BoAHV-1 JL-5 strain indicates that it belongs to the BoAHV-1.1 subtype. These findings provide a foundation and data for the detection, prevention, and control Infectious Bovine Rhinotracheitis.

Abstract Hydrolyzed egg yolk peptide (YPEP) was shown to increase bone mineral density in ovariectomized rats. However, the underlying mechanism of YPEP on osteoporosis has not been explored. Recent studies have shown that Wnt/β-catenin signaling pathway and gut microbiota may be involved in the regulation of bone metabolism and the progression of osteoporosis. The present study aimed to explore the preventive effect of the YPEP supplementation on osteoporosis in ovariectomized (OVX) rats and to verify whether YPEP can improve osteoporosis by regulating Wnt/β-catenin signaling pathway and gut microbiota. The experiment included five groups: sham surgery group (SHAM), ovariectomy group (OVX), 17-β estradiol group (E2: 25 µg /kg/d 17β-estradiol), OVX with low-dose YPEP group (LYPEP: 10 mg /kg/d YPEP) and OVX with high-dose YPEP group (HYPEP: 40 mg /kg/d YPEP). In this study, all the bone samples used were femurs. Micro-CT analysis revealed improvements in both bone mineral density (BMD) and microstructure by YPEP treatment. The three-point mechanical bending test indicated an enhancement in the biomechanical properties of the YPEP groups. The serum levels of bone alkaline phosphatase (BALP), bone gla protein (BGP), calcium (Ca), and phosphorus (P) were markedly higher in the YPEP groups than in the OVX group. The LYPEP group had markedly lower levels of alkaline phosphatase (ALP), tartrate-resistant acid phosphatase (TRAP) and C-terminal telopeptide of type I collagen (CTX-I) than the OVX group. The YPEP groups had significantly higher protein levels of the Wnt3a, β-catenin, LRP5, RUNX2 and OPG of the Wnt/β-catenin signaling pathway compared with the OVX group. Compared to the OVX group, the ratio of OPG/RANKL was markedly higher in the LYPEP group. At the genus level, there was a significantly increase in relative abundance of Lachnospiraceae_NK4A136_group and a decrease in Escherichia_Shigella in YPEP groups, compared with the OVX group. However, in the correlation analysis, there was no correlation between these two bacteria and bone metabolism and microstructure indexes. These findings demonstrate that YPEP has the potential to improve osteoporosis, and the mechanism may be associated with its modulating effect on Wnt/β-catenin signaling pathway.
