Abstract

Mitochondrial DNA analysis demonstrates that out of 48 Candida albicans isolates, six exhibited clonal proliferation of the DNA. Data indicate that the nuclear genotypes of the strains carrying identical mitochondrial DNA haplotypes are similar to one another than those that possess randomly derived nuclear genotypes.

Abstract

A crt homolog was isolated, characterized and gene disrupted using homologous recombination to determine its role in the photochromogenicity of Mycobacterium marinum. Results of the study revealed that the expression of a gene replacement is significant for photochromogenicity. The gene disruption in mycobacterium gives way for the dissection of the molecular basis of mycobacterial pathogenesis.

Abstract

In vivo footprinting of Fremyella diplosiphon cultured in either green or red light demonstrated the presence of PepB, a DNA binding protein, upstream of thephycoerythrin operon gene. Results reveal that two guanosine residues in this segment are protected from methylation. Gel electrophoresis reveal that PepB from cultures grown in green light is identical to that of cultures grown underred light indicating that phototransduction is mediated via another regulatory element. In vitro foot printing studies were undertaken to determine the DNA binding site of PepB.

Abstract

Gene order evolution in two eukaryotes was studied by comparing the Saccharomyces cerevisiae genome sequence to extensive new data from whole-genome shotgun and cosmid sequencing of Candida albicans. Gene order is substantially different between these two yeasts, with only 9% of gene pairs that are adjacent in one species being conserved as adjacent in the other. Inversion of small segments of DNA, less than 10 genes long, has been a major cause of rearrangement, which means that even where a pair of genes has been conserved as adjacent, the transcriptional orientations of the two genes relative to one another are often different. We estimate that about 1,100 single-gene inversions have occurred since the divergence between these species. Other genes that are adjacent in one species are in the same neighborhood in the other, but their precise arrangement has been disrupted, probably by multiple successive multigene inversions. We estimate that gene adjacencies have been broken as frequently by local rearrangements as by chromosomal translocations or long-distance transpositions. A bias toward small inversions has been suggested by other studies on animals and plants and may be general among eukaryotes.

Abstract

Representational difference analysis of cDNAs (cDNA-RDA) is a sensitive subtractive hybridization technique capable of isolating rare mRNAs differentially expressed in two cell populations. cDNA-RDA can detect sequences represented at 0.0001% in the starting mRNA. By using reverse transcriptase polymerase chain reaction (PCR), cDNA-RDA also lends itself to studies in which samples are derived from limited numbers of cells. Standard cDNA-RDA protocols depend upon the presence of specific restriction enzyme sites in each cDNA, typically enzymes with four base recognition sequences. These sites are used to reduce the cDNA size range and provide primer sites for subsequent PCR amplification. Consequently, transcripts containing fewer than two of the chosen restriction sites are undetectable by cDNA-RDA. We have developed a restriction enzyme site-independent cDNA-RDA protocol called modified RDA (MRDA). We constructed MRDA test sequences from random hexamer-primed cDNA, thereby increasing the representation of mRNAs which are excluded by cDNA-RDA protocols. MRDA is also more efficient than cDNA-RDA at removing highly expressed housekeeping genes during the subtractive hybridization process, thereby allowing more efficient isolation of preferentially expressed mRNAs. Using MRDA, we isolated cDNAs differentially expressed between limited numbers of human CD4(+) naive and memory T lymphocyte subsets and skin- and gut-homing memory T cell subsets.
