Understanding the composition of complex Escherichia coli populations from the environment is necessary for identifying strategies to reduce the impacts of fecal contamination and protect public health. Metabarcoding targeting the hypervariable gene gnd was used to reveal the complex population diversity of E. coli and phenotypically indistinct Escherichia species in water, soil, sediment, aquatic biofilm, and fecal samples from native forest and pastoral sites. The resulting amplicons were cross-referenced against a database containing over 700 different partial gnd sequences from E. coli/non-E. coli Escherichia species. Alpha and beta measures of diversity of Escherichia populations were lowest in feces, soil and sediment compared to water and aquatic biofilm samples. Escherichia populations recovered from extensive freshwater catchments dominated by sheep, beef and dairy farming were extremely diverse but well-separated from a wetland dairy site. In contrast, Escherichia populations from the low-impact native forest site with fewer fecal sources were less diverse. Metabarcoding distinguished E. coli populations important to fecal contamination monitoring from non-E. coli Escherichia environmental populations. These data represent in-depth analysis and geographic stability of Escherichia populations from environmental samples with extensive heterogeneity, and reveal links with diverse fecal sources, land-use and the overall burden of fecal contamination at sample sites. To evaluate the efficacy of laparoscopic-assisted Soave procedure of Hirschsprung's Disease (HSCR) in neonates and infants within 3 months of age. Patients with HSCR who were admitted to the Department of Fetal and Neonatal Surgery of Hunan Children's Hospital from June 2012 to July 2020 and underwent laparoscopic-assisted Soave procedure were selected. The clinical manifestations, surgical procedures, postoperative fecal contamination, constipation, enterocolitis, and defecation function were analyzed. A total of 267 children were included, 230 males and 37 females. Vaginal delivery occurred in 138 cases (51.7%) and cesarean delivery in 129 cases (48.3%). There were 217 cases (81.3%) of delayed excretion of meconium, preoperative HAEC (Hirschsprung's Disease Associated enterocolitis) 116 cases (43.4%), postoperative HAEC 54 cases (20.2%), 1 cases of postoperative death, 266 cases (99.6%) were cured. There were 37 cases (13.9%) of long-segment type, 226 cases (84.6%) of common type, and 4 cases (1.5%) of short-segment type. Of the 267 cases, 214 cases were followed up (80.1%), 60 cases with fecal contamination, the incidence rate was 28.0%, and 5 cases were tertiary fecal contamination. The overall incidence of constipation was 3.7% (8 cases), and 2 cases were third-degree constipation. Evaluation of defecation function: The excellent rate was 94.4%. Laparoscopic-assisted Soave procedure in neonates and infants within 3 months of age is generally safe and feasible, most infants have good postoperative defecation function, and the incidence of postoperative fecal contamination, constipation, and enterocolitis is relatively low. This work aimed to determine the level of some toxic elements (As, Cr, Cd, and Pb) in the water of the Rupsha River, Bangladesh, concerning their potential dangers to human exposure. The elemental concentrations (mg/L) were determined using Atomic Absorption Spectrometer and found to decrease in the order of Cr (0.041) > Pb (0.029) > As (0.004) > Cd (0.002). The level of elements in this river water surpasses various international limits, making it unfit for human consumption. Furthermore, the metal pollution index and contamination index indicated that the water was also unsuitable for this purpose. The elements chosen were persuasive to discern the hazard quotient of non-carcinogenic risk. Moreover, total targeted hazard quotient (TTHQ) values were found for adults and children within acceptable limits (TTHQ <1). The value of carcinogenic risk did not surpass the range (10−6 to 10−4) of the threshold limit. Due to their high-water consumption per unit of body-weight and physiological development, children were found to be more sensitive than adults. Multivariate analyses demonstrated that human activities were the primary origin of toxic elements in river water. According to the findings, urban and industrial effluents should be treated before being released into rivers. Development along the river bank must be carefully controlled to safeguard the river environment. Introduction: Integrated crop-livestock systems (ICLS) improve soil health and productivity but may lead to the transfer of foodborne pathogens to fresh produce from soil contaminated with the feces of grazing animals. Methods and results: Over 2 years (2021–2022), organic ICLS field trials were conducted in California (CA) and Minnesota (MN) to examine the presence of foodborne pathogens (Escherichia coli O157, non-O157 Shiga toxin producing E. coli (STEC), and Listeria monocytogenes) across three treatments (fallow, cover crop without grazing, and cover crop with grazing by small ruminants) in soils and produce. A Random Forest (RF) analytical approach was used to determine potential associations between meteorological and soil chemical factors, and the generic E. coli (gEc) presence in soil. The persistence of gEc in soil, as an indicator of fecal contamination, was evaluated using mixed effect zero-inflated negative binomial (ZINB) models, considering top-ranked meteorological factors identified from RF analyses. One produce sample (cucumber) tested positive for non-O157 STEC (0.6%, 1/157) from grazed soil in MN, with no other foodborne pathogens detected in produce. Soil contamination by non-O157 STEC increased in 2022 (2.7%, 15/552) compared to 2021 (0%, 0/504) from both states, aligning with the increased prevalence of non-O157 STEC in post-grazed fecal samples from sheep in CA (41.7%, 5/12) and goats (42.5%, 17/40) in MN in 2022. Concentrations (Most Probable Number/100 g) of gEc in grazed soil returned to levels comparable to non-grazed or fallow treatments within 87–147 days post-graze (DPG) in both years, with a significant decrease predicted after 32 DPG. Interestingly, non-O157 STEC was detected in soil even after concentrations of gEc declined 115–147 DPG. Discussion: Although the effects of meteorological factors and soil chemical characteristics were not as influential as treatment or sampling day effects, ZINB analyses with identified meteorological factors in grazed soil suggested that regional differences in gEc counts were likely influenced by maximum air/soil temperatures on the sampling day. Further investigation is needed to evaluate the adoption of the NOP 90-120-day interval rules between manure application by grazing and harvest, considering a wider range of environmental regions and the potential cumulative effect of continuous ICLS in the same field on pathogen loads. This study assessed O. viverrini egg contamination in underwater sediments from canals and aimed to reduce O. viverrini transmission in egg–positive and downstream communities by controlling human opisthorchiasis in endemic area of Northeast Thailand.Sediment samples from 14 sites in UdonThani municipality were determined for O. viverrini eggs. Human fecal samples and fish were examined for opisthorchiasis and metacercaria infection, respectively. Water samples were analysed for faecal coliforms bacteria. From 2017 to 2023, transmission control efforts were evaluated by monitoring changes in human and fish infection rates.In 2017 and 2018, O. viverrini eggs were found in canal sediments at 153 and 86 eggs/liter, respectively. Human opisthorchiasis prevalence was 5.3%, with cyprinoid fish showing a 25.0% infection rate (11.6 cysts/infected fish) and significant fecal contamination (>5,500 CFU/100 ml). Conversely, egg–negative sites showed a 0.5% human infection rate and no metacercariae were found. After health interventions (2019 to 2023), O. viverrini eggs were not detected in sediments from canals, resulting in a significant reduction in human opisthorchiasis prevalence to 0.9% at the previously egg–positive site and from 12.2% to 1.7% in downstream communities (p < 0.001). Additionally, the prevalence of metacercariae in cyprinoid fish decreased from 11.4% (7.6 cysts/infected fish) to 4.5% (2.4 cysts/infected fish), indicating reduced transmission.The presence of O. viverrini eggs in sediment was associated to human opisthorchiasis transmission and demonstrated the significant effectiveness of health interventions in controlling the disease. Water quality is a significant determinant of ecosystem health and human well-being particularly in cities undergoing rapid industrialization and population growth. This review highlights data from studies conducted between 2004 and 2024 to assess the water quality of surface and groundwater sources in Islamabad and Rawalpindi. The results indicate severe microbial contamination with high levels of total coliform, fecal coliform, and E. coli in both cities, with a potential risk of waterborne diseases. Heavy metal contamination exists, especially As and Pb, which is significantly higher in Rawalpindi due to the industries. Physicochemical parameters, such as turbidity and residual microbial contamination remain in treated water, implying inefficiency in the water treatment process. The contamination also shows seasonal fluctuation, increasing during monsoon due to sewage overflows and agricultural runoff. Both cities also have conventional water treatment infrastructure that causes widespread contamination in the distribution system. Public health impact is prevalent, and outbreaks of diseases from waterborne diseases including Hepatitis E reported both in urban and peri-urban areas quite frequently. A gap is critical in applying the Advanced Oxidation Process (AOP) and other emerging technologies for degrading persistent contaminants, most notably heavy metals and microbial pollutants in the AOPs. This review focuses on the urgency of upgrading infrastructure, strengthening the implementation of water quality standards, and using more advanced treatment technologies and offers recommendations to policymakers and stakeholders and technology solutions for tackling critical challenges in water quality management, and emphasizes the importance of continuous monitoring and the enforcement of regulations. Introduction: Seals, protected wild marine mammals, are widely found in waters around the world. However, rising concerns about their increasing numbers in some areas have led to potential worries regarding microbiological contamination of coastal areas by their feces, which could impact bathing and shellfish-harvesting activities. To the best of our knowledge, no study has been conducted on the bacterial and RNA viral communities present in the feces of both grey and harbor seals, which are the two main seal species observed in mainland France and overseas. Methods: Fecal bacterial (n = 132) and RNA viral (n = 40) communities of seals were analyzed using 16S rRNA gene amplicon high-throughput sequencing and viral RNA sequencing methods, respectively. In addition, to identify the specific characteristics of seal fecal microbial communities compared to other animal fecal microbial communities that may also contaminate coastal areas, the bacterial communities of seals were compared to those of wild waterbirds and breeding animals (i.e., cattle and pigs) which could be present in upstream catchments of coastal areas. Finally, ANCOM was used to identify unique and seal-associated Amplicon Sequence Variants (ASVs), aiming to develop a Microbial Source Tracking (MST) bacterial qPCR marker associated with seals. Results and discussion: The bacterial communities of grey and harbor seals were not found to be significantly different and were characterized by a predominance of Firmicutes, including the genera Clostridium sensu stricto 1 and Peptoclostridium , followed by Fusobacteriota with the genus Fusobacterium , and Bacteroidota with the genus Bacteroides. However, variations in bacterial communities between sites and individuals were observed. Similar observations were made for the RNA viral communities being characterized by a predominance of Picobirnaviridae (44% of total reads) and Astroviridae (15%). This study successfully developed a sensitive (89.8%) and specific (97.1%) MST qPCR marker targeting grey seal-associated bacteria belonging to the Bifidobacteriaceae family. This marker can be used to identify potential fecal contamination of coastal areas by seals and complements the MST toolboxes of markers already developed for humans, wild birds and livestock. This cross-sectional study aimed to assess the drinking water parameters in primary and secondary schools. A questionnaire encompassing the schools’ general characteristics and information about drinking water was administered to school administrators. Drinking water samples were taken from 60 schools to evaluate drinking water parameters. The data were analyzed using Chi-Square, Mann–Whitney U, and Kruskall–Wallis tests, T-test, and SPSS 22.0 software. Non-compliance with national legislation was observed in 16.7% of schools during drinking water analysis. The microbiological parameters exceeded the threshold values in 90% of the schools where the analysis results of the drinking water were deemed inadequate. The analysis yielded no discernible variations based on school district or type. Mains water was the primary drinking water source in 91.7% of schools. The rate was lower in rural schools compared to urban schools, with a difference of 85 and 95%, respectively. It was found that 41.7% of schools lacked canteens or the sale of packaged water. Consequently, the availability of tap water in schools falls short of the desired standard. It is imperative to investigate the factors leading to microbiological contamination in school drinking water and formulate short-, medium-, and long-term strategies to enhance its safety. Olive mill wastewater, a byproduct of olive oil production, poses a major environmental challenge for Mediterranean countries like Morocco. These residues are rich in polyphenols with notable antimicrobial properties. This study aimed to investigate the impact of anthropogenic factors on the quality of certain wells in the Oued Amlil region of the Taza province, Morocco. It was carried out during two distinct periods by sampling water from six wells: P1, P2, P3, P4, P5 and P6. The first period occurred during the winter season of December 2022, when olive crushing operations for oil production were at their peak. The second period took place in the summer (August 2022), when the activities of the oil mills were stopped. The objective is to assess microbiological quality by determining the microbial load in total germs at 37°C (TG), Fecal Coliforms (FC), Total Coliforms (TC), and Fecal Streptococci (FS), as well as performing a polyphenol assay. The analysis results showed high concentrations of TG (Maximum value = 3100/100ml), FC (Maximum value = 910/100ml), TC (Maximum value = 7120/100ml), and FS (Maximum value = 7000/100ml) in wells P1, P2, and P5, although they are not located on the flow path of groundwater contaminated by the Innaouene River and the local olive oil mills. However, despite their location along this axis, wells P3, P4, and P6 show lower contamination levels. In order to identify similarities between different water sources and/or different points of contamination acquisition, principal component analysis (PCA) was applied to the collected data and showed a significant negative correlation between polyphenols and bacteriological parameters, confirming the hypothesis of polyphenols' bactericidal effect on fecal bacteria. In addition, the degree of contamination varies from one well to another depending on their location with the direction of flow of water in the groundwater and the proximity of sources of pollution. The Silway River has historically failed to meet safe fecal coliform levels due to improper waste disposal. The river mouth is located in General Santos City, the tuna capital of the Philippines and a leading producer of hogs, cattle, and poultry. The buildup of contaminants due to direct discharge of waste from chicken farms and existing water quality conditions has led to higher fecal matter in the Silway River. While there were technical reports in the early 2000s about poultry farming, this is the first study where fecal coliform from poultry farming was detected in the Silway River using highly sensitive protocols like qPCR. This study characterized the effect of flow velocity and physicochemical water quality parameters on chicken fecal contamination. Gene markers such as Ckmito and ND5-CD were used to detect and quantify poultry manure contamination through microbial source tracking (MST) and environmental DNA (eDNA) profiling. The results of this study showed the presence of chicken fecal bacteria in all stations along the Silway River. The results revealed that normal levels of water quality parameters such as temperature, pH, and high TSS concentrations create favorable conditions for chicken fecal coliforms to thrive. Multiple regression analysis showed that flow velocity and DO significantly affect chicken fecal contamination. A lower cycle threshold (Ct) value indicated higher concentration of the marker ND5-CD, which means higher fecal contamination. It was found that there was an inverse relationship between the Ct value and both velocity (R2 = 0.55, p = 0.01) and DO (R2 = 0.98, p = 0.2), suggesting that low flow velocity and low DO can lead to higher fecal contamination. Findings of fecal contamination could negatively impact water resources, the health of nearby residents, and surrounding farms and industries, as well as the health and growth of fish. We recently have shown that the gut microbiota composition in female and male runners positively correlates with sports, and female runners show similar gut microbiome diversity to male runners. However, gut microbiota composition has not yet been fully investigated in other endurance athletes, such as cyclists. Therefore, in the current study, we investigated the gut microbiome profiles in competitive, non-professional female and male cyclists compared to what we have shown in runners. We aim to understand (1) whether the gut microbiome signature is sport-specific; (2) whether there is a microbiome difference between female and male cyclists and runners; and (3) whether the gut bacteria expressed in cyclists and runners correlates with exercise performance. Our study included 58 subjects: 18 cyclists (9 males), 22 runners (13 males), and 18 control subjects (9 males). Fecal samples were obtained and subjected to taxonomic analysis to assess the relative abundances of species across subjects based on 16S rRNA sequencing results. Both alpha and beta diversity of the bacterial communities were evaluated to identify compositional variations between the groups. Each participant completed a maximal oxygen consumption test and a time-to-exhaustion test at 85% of the measured VO2max. Cyclists performed the test on an SRM ergometer, while runners used a motorized treadmill. Blood lactate levels were measured at 5 min intervals throughout the time-to-exhaustion trials. Alpha diversity demonstrated a significant difference (p-adj < 0.001) between cyclists and runners. Male cyclists showed significantly lower alpha diversity than runners (p-adj < 0.001). The taxonomic analysis of gut microbiota composition between cyclists, runners, and controls showed a lower or higher abundance of fifteen different bacteria. In cyclists, there was a significant positive correlation between six bacteria, and in runners, there was a significant positive correlation between eight bacteria, with weekly training volume, time-to-exhaustion, VO2max, and blood lactate levels. This study suggests potential sport-specific characteristics in long-distance cyclists' and runners' gut microbiome signatures. These findings emphasize the differences in gut microbiota between cyclists and runners, probably due to the difference in physiological and biomechanical conditions related to the activity mode during each sport. A conventional, group-specific PCR method was developed to identify each of the four previously defined major taxa (Myoviridae, Siphoviridae, Podoviridae and Microviridae) of somatic coliphages and used to classify isolates from sewage. Somatic coliphage infectivity detection, occurrence and survival in primary human sewage effluent was observed over time to further understand the presence and behavior of the groups of somatic coliphages at two environmental temperatures (4 and 25 °C). Over time, the taxonomic composition of the somatic coliphage population in sewage changed, with the Microviridae family becoming the most prevalent family in the sewage population after several weeks. Based on their persistence and prevalence in environmental waters, phages belonging to the Microviridae family provide supporting information on sewage contamination and possibly of human enteric viruses in sewage-contaminated water. Groundwater is an essential drinking water source for humans. However, improper groundwater management leads to fecal contamination and waterborne diseases caused by viral pathogens. Therefore, this study aimed to investigate norovirus (NoV) contamination by conducting nationwide monitoring over five years (2019–2023). Groundwater samples were analyzed for water quality parameters, indicator microorganisms, NoV prevalence, and viral genotypes. Water quality was assessed for temperature, turbidity, and residual chlorine, whereas microorganisms were analyzed for total coliforms, Escherichia coli, and NoV genotypes. Of the 600 sites, 11 (1.8%) were NoV-positive, irrespective of season or location. Low residual chlorine levels (0.02–0.75 mg/L) were observed, possibly limiting viral inactivation. Total coliforms were detected in only three NoV-positive samples, and E. coli was absent. NoV genotypes were identified as GI.1 and GII.4, with GII.4 being the most frequently detected genotype. Simple Summary: Wild bird species are known to be readily infected with and carry Salmonella enterica. This has two main consequences. Firstly, they may act as reservoirs in the transmission of infection to livestock species or zoonotic transmission to humans. Secondly, birds themselves may develop systemic infection, leading to high mortality disease. Recent studies have shown that specific genotypes adapted to and capable of causing disease in a range of bird species have recently evolved. Salmonella enterica is an important bacterial pathogen in humans and warm-blooded animals. Wild bird species represent both a potential reservoir for zoonotic infection and as a susceptible host to infection by host-adapted variants. Historically, wild birds were considered to be a major source of Salmonella infection in livestock, but in recent years, it has been more apparent that birds are more likely to act as a reservoir for recycling infection on farms rather than as the primary source of infection. Birds may also transmit infection to humans directly from feces or indirectly through fecal contamination of foods, including peanut butter. While many bird species can be infected with Salmonella, the rates of infection are variable, and most cases lead to intestinal carriage rather than disease. In this case, fecal shedding of Salmonella bacteria from birds can represent a risk for transmission to humans. As such, care is needed when in contact with fecal material such as that found on bird tables or feeders. In recent years, there have been emergences of Salmonella Typhimurium genotypes associated with high mortality in songbirds or passerine birds, resulting in 'die offs' in Europe, Israel, New Zealand and the US. Additionally, S. typhimurium DT2 and other variant Copenhagen genotypes are associated with high mortality disease in pigeons. These genotypes show evidence of evolution towards adaptation to specific hosts, with pseudogenes leading to loss of functional metabolic pathways and specific virulence factors. Escherichia coli harbors various virulence factors responsible for diarrhea and other diseases in neonatal calves. The antimicrobial resistance (AMR) among the diarrheagenic and non-diarrheagenic E. coli may impose a potential health hazard among the calves and make the antibacterial therapy ineffective. The virulence gene expression among AMR strains plays a crucial role in establishing diarrheal disease with therapeutic complications. This study investigates the diversity of E. coli isolates from neonatal calves. E. coli isolated from the fecal samples were subsequently subjected to pathotyping and virulotyping using the PCR technique. AMR profiling was done by phenotypic and genotypic methods. The ERIC pattern of E. coli pathotypes was compared and correlated with the pattern of AMR and virulence traits. 179 isolates were obtained from 158 fecal samples collected from neonatal calves. Forty-two isolates were pathotypable; predominated by shigatoxigenic E. coli. Seventy-four isolates were ESBL producers. Nineteen isolates were carbapenem resistant and 68 isolates exhibited quinolone resistance. blaAmpC, blaVIM and qnrS were the predominant genes detected among ESBL, carbapenem and quinolone resistance genes, respectively. The present study indicated diverse virulence profiles and the presence of significant ESBL, carbapenem and fluoroquinolone resistance among E. coli pathotypes from calves. The analysis of resistance profiling indicates the risks associated with the indiscriminate antibiotic use among farm animals, which may pose a potential health hazard to livestock owners. Based on a systematic review of convolutional neural networks (CNN), this study explores the efficacy of small imaging sensors in monitoring the real-time presence of cyanotoxins and hazardous contaminants in urban ecosystems. To develop a machine learning-based CNN, this study first investigated the relationships between the prevalence of hazardous algal blooms (HABs) and faecal indicator bacteria (FIB) in waterways and aquifers of certain semi-arid zones of Sri Lanka, Sweden and New York (United States). By incorporating a popularly known AbspectroscoPY framework to effectively process the spectrophotometric data of the obtained samples, the formulation subsequently reveals strong positive correlations between FIB coliforms and nutrient loads (particularly nitrate and phosphate). A corroborative association with the incidence of chronic kidney disease of uncertain aetiology (CKDu) among the residents of the studied regions further affirms the reliability of the methodology. These findings underline the need for policymakers to consider the geographical and land-use traits of urban habitats in strategies aimed at reducing water-borne health hazards. It is critical to attribute faecal contamination to its original source in order to assess public health risks and implement effective interventions to mitigate future contamination. This study aimed to identify the primary sources of faecal contamination in water using microbial source tracking markers in water-stressed rural communities. A total of 1128 water samples were collected sequentially from the main source (river/borehole) to the households. Six host-specific genetic markers were used to detect faecal contamination in the water samples (BacHum and HF183, BacCow, Pig-2-Bac, Cytb and BacCan). Of the 564 water samples tested during the wet season, 37.94% (n = 214) were positive for human and animal-specific Bacteroidales marker genes, while 31.73% (n = 179) of the 564 tested during the dry season were also positive. During the wet season, animal faecal contamination was more prevalent among the positive samples (Cytb: 20.56%, n = 44; BacCan: 19.16%, n = 41). By contrast, human-origin faecal contamination was dominant during the dry season (BacHum: 23.46%, n = 42; HF183: 21.23%, n = 38). Identifying the origin of faecal contamination will assist in implementing targeted intervention strategies for the effective prevention of pathogen transmission in water-stressed rural communities in order to protect public health. Diarrheal diseases remain a leading factor behind child mortality in Pakistan. Children residing in households with unimproved sanitation and water facilities suffer more from diarrheal illness. One of the important pathways that has not been well investigated in Pakistan is the household floor, which carries significant fecal contamination. For this purpose, the present study assesses the impact of household flooring on the incidence of diarrhea among children aged 6–59 months. Data for this cross-sectional study has been taken from Pakistan Demographic and Health Survey (2017–18). Descriptive statistics and multilevel logistic regression were employed. The results revealed that improved household flooring leads to a substantial reduction in childhood diarrhea. Also, well-educated mothers through better hygiene and correct dietary practices reduce diarrheal illness among children even in an unhygienic environment. The significance of Escherichia albertii as a foodborne pathogen is increasingly acknowledged, but the assessment of its occurrence and transmission remains challenging due to the lack of validation of selective isolation, detection, and identification methods. The aim of the present study was to examine its presence on various meat samples at the retail level in order to assess a potential foodborne transmission and its occurrence in clinical stool samples. First, the evaluation and selection of a selective enrichment broth and isolation medium, combined with an optimized identification by MALDI-TOF MS, as well as a suitable DNA extraction method and a PCR-based detection strategy were developed. After the evaluation of existing isolation strategies and the formulation of an adapted enrichment and isolation medium, 100% isolation specificity was not achieved. An identity confirmation of suspected colonies remains necessary. A total of 292 samples, including 45 beef fillet, 51 minced beef, 50 pork fillet, 30 minced pork, 30 chicken carcass, 51 chicken fillet, and 35 minced chicken samples were examined. Samples were all collected at the retail level, including supermarkets and local butcheries. Escherichia albertii was isolated from two chicken fillets (3.9%) and additionally detected in one minced chicken (4.5%) and two other chicken fillet (4.5%) samples by a PCR assay. All beef and pork samples tested negative for its presence, but transmission through these meat types cannot be excluded, as it potentially correlates with the level of fecal contamination that was significantly higher on poultry products. With other hygienic conditions and processing steps applied, the presence of E. albertii on food can therefore differ in other parts of the world. Escherichia albertii was present in 0.4% of the 2419 clinical stool samples examined. The future development of a chromogenic isolation medium, as well as further extensive epidemiologic approaches and a genomic comparison of human, food, and animal isolates, could enhance the assessment of the emerging pathogen status and its potential as a foodborne hazard. Enteropathogenic (EPEC), necrotoxigenic (NTEC), and Shiga-toxin producing Escherichia coli (STEC) are pathotypes responsible for severe clinical forms in humans and animals. They can be shed in the feces of animals with consequent environmental contamination. This study evaluated the antibacterial activity of essential oils (EOs) from oregano (Origanum vulgare), savory (Satureja montana), thyme (Thymus vulgaris), and their blend against EPEC, NTEC, and STEC strains previously isolated from avian fecal samples. Minimum inhibitory concentration values between 0.039% and 0.156% were found with O. vulgare EO, between ≤0.0195% and 0.156% with both S. montana and T. vulgaris EOs, and between 0.039% and ≤0.0195% with the blend. The mixture with equal parts of EOs from oregano, savory and thyme seems a promising alternative product to combat pathogenic E. coli strains responsible for environmental contamination. The Banger River, bordered by settlements, industrial sites, and agricultural land, has faced critical pollution challenges due to waste generated by surrounding activities. This contamination has led to a significant decline in both the quality and quantity of available clean water, a situation further exacerbated by rapid population growth. For years, the presence and severity of fecal pollution in natural water bodies have been monitored through the measurement of coliform bacteria, serving as a vital indicator. In this study, numerical simulation models were utilized to assess the distribution of total coliforms in the waters of Pekalongan. The findings indicated that the distribution of total coliforms was significantly affected by tidal currents, which predominantly flowed westward along the coastline. The coliforms were sourced from various runoff pathways throughout the Banger River, with concentrations increasing up to 65,693 CFU/mL as one moves further west. In the coastal area of Pekalongan, coliform concentrations range between 15,252 and 62,332 CFU/mL. Hurricane Harvey caused widespread flooding along the Texas Gulf Coast in August 2017; some areas of Houston received >150 cm of rainfall within a few days. Due to concerns over fecal contamination of floodwaters, surface water samples were collected at six locations in the southeastern Houston area immediately before and after the hurricane and then every 1 to 2 weeks thereafter over a 2-month period. Total E. coli was enumerated using the IDEXX Quanti-Tray/2000 system. DNA extracted from water samples was analyzed via quantitative real-time PCR (qPCR) for general and source-specific total Bacteroidales and human Bacteroidales markers, and digital PCR (dPCR) for antibiotic resistance genes (ARG) and a plasmid (pBI143) associated with human waste. SourceTracker2 was used to determine human source contributions based on metagenomic analysis of PCR-amplified 16S rRNA gene fragments. Samples collected immediately after the hurricane had elevated levels of E. coli, ranging from 488 to 1,733 CFU 100 ml−1. After 1 week, E. coli levels decreased to <100 MPN 100 ml−1. Total Bacteroidales numbers were elevated immediately following the hurricane and remained high for 12 days. Human-source contributions, as assessed by PCR methods and metagenomic analysis, peaked within 12 days after the hurricane consistently across all sampling sites. Multiple regression analysis of environmental parameters, copies of ARG and pBI143, and metagenomic data confirmed that human waste caused the dramatic, short-term, high levels of fecal contamination of floodwaters generated by Hurricane Harvey. Background: Current methods for obtaining fecal egg counts in horses are often inaccurate and variable depending on the analyst's skill and experience. Automated digital scanning of fecal sample slides integrated with analysis by an artificial intelligence (AI) algorithm is a viable, emerging alternative that can mitigate operator variation compared to conventional methods in companion animal fecal parasite diagnostics. Vetscan Imagyst is a novel fecal parasite detection system that uploads the scanned image to the cloud where proprietary software analyzes captured images for diagnostic recognition by a deep learning, object detection AI algorithm. The study describes the use and validation of Vetscan Imagyst in equine parasitology. Methods: The primary objective of the study was to evaluate the performance of the Vetscan Imagyst system in terms of diagnostic sensitivity and specificity in testing equine fecal samples (n = 108) for ova from two parasites that commonly infect horses, strongyles and Parascaris spp., compared to reference assays performed by expert parasitologists using a Mini-FLOTAC technique. Two different fecal flotation solutions were used to prepare the sample slides, NaNO3 and Sheather's sugar solution. Results: Diagnostic sensitivity of the Vetscan Imagyst algorithm for strongyles versus the manual reference test was 99.2% for samples prepared with NaNO3 solution and 100.0% for samples prepared with Sheather's sugar solution. Sensitivity for Parascaris spp. was 88.9% and 99.9%, respectively, for samples prepared with NaNO3 and Sheather's sugar solutions. Diagnostic specificity for strongyles was 91.4% and 99.9%, respectively, for samples prepared with NaNO3 and Sheather's sugar solutions. Specificity for Parascaris spp. was 93.6% and 99.9%, respectively, for samples prepared with NaNO3 and Sheather's sugar solutions. Lin's concordance correlation coefficients for VETSCAN IMAGYST eggs per gram counts versus those determined by the expert parasitologist were 0.924–0.978 for strongyles and 0.944–0.955 for Parascaris spp., depending on the flotation solution. Conclusions: Sensitivity and specificity results for detecting strongyles and Parascaris spp. in equine fecal samples showed that Vetscan Imagyst can consistently provide diagnostic accuracy equivalent to manual evaluations by skilled parasitologists. As an automated method driven by a deep learning AI algorithm, VETSCAN IMAGYST has the potential to avoid variations in analyst characteristics, thus providing more consistent results in a timely manner, in either clinical or laboratory settings. This study analysed the water quality of 156 samples collected from 13 different points within the Chief Albert Luthuli Local Municipality between January and December 2021, focusing on compliance with the South African National Standard 241 (SANS 241). Microbiological assessments revealed widespread contamination, with heterotrophic plate counts (HPCs) exceeding the recommended range in nearly all samples. The median HPC levels were particularly concerning, with the untreated source (RE000) exhibiting a median of 2555 CFU/mL. Total coliforms and Escherichia coli were present at alarming levels, indicating significant faecal contamination; the highest total coliform count, 2420 CFU/100 mL, was recorded at RE000. Physically, while all samples complied with electrical conductivity standards (0–70 µS/cm), turbidity levels showed variability, with nine samples exceeding the acceptable threshold of 1 NTU. The highest turbidity, 7.55 NTU, was recorded at the untreated source (RE000). Chemically, pH levels fell within safe ranges (6.0–9.0), with all samples being compliant with standards. However, only one sample (E001) met the free chlorine standard of 0.3–0.6 mg/L, suggesting inadequate disinfection practices. The untreated source was classified as 'Unsuitable for drinking' (WQI = 207.27). Access to safe drinking water is a fundamental human need for health and well-being implemented globally by the United Nations under Sustainable Development Goal (SDG) 6. Storing drinking water is common in rural areas of Ethiopia due to off-premises water sources and intermittent piped water supply. However, this practice can lead to further contamination during collection, transport, and storage, posing a risk to public health. The objective of this study was to identify the determinant factors of drinking water quality at the point of use in the rural setting of northwestern Ethiopia, South Gondar zone. A questionnaire survey was conducted, and water samples from 720 households were collected during the wet and dry seasons. The determinant factors were identified using the multivariable logistic regression model. About 39.2% of the surveyed households had basic water supply services, 41.9% were using unimproved sources, and 8.3% were using surface water. Only 9.4% were using basic sanitation services, and 57.2% were practicing open defecation. Safe water storage was practiced by 84.3% of households, while only 2% engaged in household water treatment. About 14% of dry and 8% of wet season samples from the storage were free from fecal coliform bacteria. Furthermore, 52.9% of dry and 62.2% of wet season samples fell under the high microbial health risk category. The season of the year, the water source type, storage washing methods, and the socioeconomic status of the household were identified as key predictors of household drinking water fecal contamination using the multivariable logistic regression model. It was observed that the drinking water in households had a high load of fecal contamination, posing health risks to consumers. To tackle these problems, our study recommends that stakeholders should enhance access to improved water sources, implement source-level water treatment, increase access to improved sanitation facilities, advocate for safe household water management practices, and endorse household water treatment methods. Background: Pressure sores significantly affect elderly patients, with the sacrum being especially vulnerable, often due to its proximity to the anus and potential for fecal contamination. Despite preventive measures such as frequent repositioning and specialized cushions, some sores still necessitate surgical intervention. Postoperative care focuses on monitoring, hygiene, and pressure alleviation. This study assessed the efficacy of Closed-Incision Negative Pressure Wound Therapy (CI-NPWT) for postoperative wound management in patients with sacral pressure sores treated with local flaps. Methods: A retrospective analysis was performed on sacral sore patients who underwent reconstructive surgery from March 2019 through April 2023. Surgical procedures involved debridement and wound coverage using gluteal artery perforator-based fasciocutaneous flaps, followed by postoperative monitoring. Patients were grouped into conventional monitoring or NPWT management, with the latter utilizing the INFOV.A.C. The therapy unit was calibrated to 125 mmHg. Data on patient demographics, flap metrics, fluid drainage amounts, and six-month postoperative outcomes were collected, and then analyzed with SPSS Statistics. Results: In this study of 52 patients with sacral pressure sores, the NPWT group (n = 25) showed significantly fewer flap complications and a lower drainage volume on the seventh postoperative day (mean 17.2 cc) compared to the conventional dressing group (mean 27.8 cc, P < 0.05). No postoperative complications were observed in the NPWT group, whereas the conventional group (n = 27) experienced one infection and three cases of dehiscence. Both groups achieved a 100% flap survival rate. Conclusion: Closed-incision negative pressure wound therapy (CI-NPWT) shows promise in reducing wound dehiscence and infection rates in elderly patients undergoing sacral pressure sore reconstruction with local flaps. However, further research with larger, randomized studies is needed to confirm its effectiveness as an alternative to conventional postoperative care. Trial registration. Gut microbial dysbiosis, especially bacteriome, has been implicated in Alzheimer's disease (AD). However, nonbacterial members of the gut microbiome in AD, such as the mycobiome, archaeome, and virome, are unexplored. Here, we perform higher-resolution shotgun metagenomic sequencing on fecal samples collected longitudinally from a mouse model of AD to investigate longitudinal and multi-kingdom gut microbiome profiling. Shotgun metagenomic sequencing of fecal samples from AD mice and healthy mice returns 41,222 bacterial, 414 fungal, 1836 archaeal, and 1916 viral species across all time points. The ecological network pattern of the gut microbiome in AD mice is characterized by more complex bacterial–bacterial interactions and fungal–fungal interactions, as well as simpler archaeal–archaeal interactions and viral–viral interactions. The development of AD is accompanied by multi-kingdom shifts in the gut microbiome composition, as evidenced by the identification of 1177 differential bacterial, 84 differential fungal, 59 differential archaeal, and 10 differential viral species between healthy and AD mice across all time points. In addition, the functional potential of the gut microbiome is partially altered in the development of AD. Collectively, our findings uncover longitudinal and multi-kingdom gut microbiome alterations in AD and provide a motivation for considering microbiome-based therapeutics during the prevention and treatment of AD. For decades, the risk of exposure to infectious diseases in recreational beaches has been evaluated through the quantification of fecal indicator bacteria in water samples using culture methods. The analyses of sand samples have recently been developed as a complement to the monitoring of recreational waters in beach quality assessments. The growing use of molecular techniques for environmental monitoring allows for the rapid detection of pathogenic genes, thus providing more accurate information regarding the health risk of exposure to contaminated sand. The aim of this work was to determine the relationship between the fecal indicators abundance in water and sand and the presence of Shiga toxin-producer Escherichia coli (STEC) in sand by analyzing samples from touristic beaches using culture-dependent (fecal coliforms assay) and culture-independent (real-time PCR of stx1, stx2, and eae genes) techniques. We found a high concentration of coliform bacteria in water and sand in several beaches in eastern Uruguay, with different levels of sanitation networks and levels of urbanization. The presence of STEC virulence genes (mainly stx1) was confirmed in 8 out of 20 sand samples. The recreational use of sandy beaches may imply a risk to the health of its users, especially near streams and creek outflows, thus highlighting the need of monitoring sand bacteriological quality and pathogens using molecular tools. Surface waters within the basin of the Laurentian Great Lakes are impacted by microbial contamination from municipal wastewater and agricultural runoff, as well as from other sources. In particular, microbial contamination of drinking water is an ongoing problem within many Indigenous communities located in the basin. However, it is difficult to identify the sources of microbial contamination using the traditional monitoring approaches with fecal indicator bacteria, such as total coliforms and Escherichia coli (E. coli). In this study, we evaluated whether surface waters in the basin are contaminated with fecal bacteria of human origin using chemical indicators of domestic wastewater (i.e., caffeine and sucralose) and with Bacteroidales 16S rRNA markers. Study areas included the Grand River watershed within the Lake Erie basin and three nearshore locations within the Great Lakes basin. Two of these sites are sources of drinking water for Indigenous communities. We assessed whether there were relationships between the concentrations of fecal indicator microorganisms and chemical indicators of domestic wastewater at selected study locations. Analysis of genetic markers indicated that about 30% of the Bacteroidales bacteria present at a site in the Grand River were of human fecal origin and the balance were of bovine or general animal origin. The presence of caffeine and sucralose in surface waters indicated that there was upstream contamination by domestic wastewater. However, in the drinking water treatment plant operated by Six Nations of the Grand River, the levels of these chemical indicators and fecal bacteria were reduced by the advanced water treatment technologies. The concentrations of sucralose and caffeine collectively were strongly correlated with the levels of total coliforms in samples from the Grand River (R2 = 0.75) and with levels of E. coli in samples from the Great Lakes basin (R2 = 0.97), but there appeared to be an upper threshold for this relationship. These data indicate that analysis of caffeine and sucralose and genetic markers for strains of Bacteroidales fecal bacteria may be useful tools for identifying the sources of microbiological contamination of surface waters and drinking water. Canada is a leading producer of wild lowbush blueberries, most of which are mechanically harvested, washed, individually quick frozen (IQF), and bulk packaged. Still, some berries are harvested by more gentle methods and sold as fresh-packed products. These berries do not undergo a wash step, nor are subjected to antimicrobial treatments. The purpose of this study was to conduct a microbiological survey of berries harvested in the province of Nova Scotia to assess their potential for harborage of bacterial foodborne pathogens. A combination of standardized plate count methods and 3M-Petrifilm protocols were used to enumerate total aerobic mesophilic bacteria (APC), yeasts and molds (YMC), coliforms, and generic E. coli, the latter being an indicator of fecal contamination. Overall, APC and YMC levels were 1.2 and 0.5 log greater, respectively, for berries collected early in the harvest season versus those acquired late season and varied significantly (p < 0.05) between farm (location) and harvest practices used. Berries harvested by our team using sanitized hand rakes (SH) had consistently lower APC and YMC levels than those harvested by farm crews. Yet, when gentle harvesting (GH) methods (hand-raking, walk-behind or modified mechanical harvesters) were employed on farms, lower numbers were generally observed compared to berries harvested by traditional tractor-mounted mechanized harvesters (MH). The presence of coliforms (and their levels) was also impacted by the harvest method, with detection rates of ~29%, 73%, and 92% in SH, GH, and MH samples, respectively. Mean counts were < 2.5 log10 CFU/g for both SH and GH berries, but significantly higher (p < 0.05) on MH berries (3.6 log10 CFU/g). Although ~56% of all berry samples collected (n = 350) contained coliforms, only 12 were positive for E. coli, 9 of which were MH samples. Only the latter had numbers > 2 log10 CFU/g, but none tested positive for Shiga toxin-producing serotype O157 (STEC O157) or Salmonella spp. when using internationally recognized selective enrichment and plating methods. ATP luminescence was used to assess the general hygiene of processing lines, whereby "hot spots" for microbial activity were identified, even after cleaning., Standard selective enrichment and plating methods were used for the detection of Listeria monocytogenes on 61 swab samples taken from berry totes or conveyor belts at different times during processing; 4 swabs tested positive for L. monocytogenes. However, the pathogen could not be detected by direct plating on selective agar without prior enrichment; this indicated its numbers were low. The results from this work demonstrated that alternative gentle harvest methods can reduce microbial numbers on wild blueberries. Although the frequency of fecal contamination in berry samples appeared to be low and targeted human pathogens were not detected; this represents a single study conducted over one harvest season. Therefore, it would be prudent for processors to seek effective antimicrobial technologies prior to packaging, while consumers should use caution and thoroughly wash produce before consumption. Where sporadic detection of L. monocytogenes was observed on environmental samples from the processing line, processors must ensure that effective sanitation programs are implemented to avoid potential food safety risks. Background: Untargeted lipidomics using collision-induced dissociation-based tandem mass spectrometry (CID-MS/MS) is essential for biological and clinical applications. However, annotation confidence still relies on manual curation by analytical chemists, despite the development of various software tools for automatic spectral processing based on rule-based fragment annotations. Methods: In this study, we present a novel machine learning model, MS2Lipid, for the prediction of known lipid subclasses from MS/MS queries, providing an orthogonal approach to existing lipidomics software programs in determining the lipid subclass of ion features. We designed a new descriptor, MCH (mode of carbon and hydrogen), to increase the specificity of lipid subclass prediction in nominal mass resolution MS data. Results: The model, trained with 6760 and 6862 manually curated MS/MS spectra for the positive and negative ion modes, respectively, classified queries into one or several of 97 lipid subclasses, achieving an accuracy of 97.4% in the test set. The program was further validated using various datasets from different instruments and curators, with the average accuracy exceeding 87.2%. Using an integrated approach with molecular spectral networking, we demonstrated the utility of MS2Lipid by annotating microbiota-derived esterified bile acids, whose abundance was significantly increased in fecal samples of obese patients in a human cohort study. This suggests that the machine learning model provides an independent criterion for lipid subclass classification, enhancing the annotation of lipid metabolites within known lipid classes. Surface water contamination by fecal matter threatens human health due to human and biological processes within a watershed, making socioeconomic development crucial for predicting and improving microbiological water quality. Consequently, climate change alters climatic parameters that affect flow regimes and the movement and fate of microorganisms. This study assessed the fate and transport of microbial Escherichia coli (E. coli) concentrations and their sources in the Tano River Basin in Ghana. Additionally, the study predicted future E. coli concentrations using climate change scenarios from the Intergovernmental Panel on Climate Change (IPCC)'s most recent representative concentration pathways (RCPs) and shared socioeconomic pathways (SSPs). Scenario_1 featured planned urbanization, enhanced manure and wastewater treatment, moderate population, livestock density growth, and climate change. Scenario_2 involved higher population growth, minimal improvements in wastewater management, zero manure treatment, higher livestock population, urbanization, and substantial climate change. Calibration and validation using E. coli data from June 2022 to April 2023 showed good agreement with observed concentrations (R2, 0.75 and 0.89; NSE, 0.69 and 0.68; PBIAS, 3.4 and 1.9, respectively). The measured and modeled E. coli concentrations were high, with the highest recording at 2.39 log cfu/100 ml during the rainy season. The study finds that the main causes of E. coli concentrations (44%) are point sources, primarily from human feces and livestock manure, followed by upstream pollution (34%) and non-point sources (22%). Non-point sources became the predominant contributors during periods of maximum discharge due to runoff from land and the dilution of point sources. Again Scenario_1 E. coli dropped to 68% and 97% of reference point levels by the 2050s and 2100s, respectively. E. coli concentrations decrease even more with subsequent treatment, such as tertiary treatment, manure treatment, or both. This study assessed the water quality for drinking and irrigation purposes on the southern slopes of Mt. Kilimanjaro during the dry season under low flow conditions. Fifty-one samples covering eight different water sources (i.e., stream water from natural and anthropogenic impacted streams, domestic water, spring water, rainfall, groundwater, lake water and water from irrigation canals) were collected in a snapshot sampling campaign over 10 days in February 2023. First, physical, chemical and biological parameters were analysed and compared with Tanzanian and international drinking and irrigation water quality requirements. The samples were then ranked according to their suitability for drinking and/or irrigation using water quality indices (WQI). All drinking water quality parameters except for E. coli and turbidity were within the permissible limits. A generalised problem of faecal contamination was found in the study area, including in domestic water, highlighting the need to identify sources of contamination and remediate them before distribution. The drinking water quality index (DWQI) classified 89% of the samples as unsuitable and 11% as excellent for drinking. Irrigation water quality parameters were within the guidelines of restriction of use except for pH in 5 samples. In contrast to the DWQI, the vast majority of the water samples (88%) can be used for irrigation without restrictions according to the irrigation water quality index (IWQI). The suitability of water for irrigation was also assessed using three indices, i.e., Kelley's Index, Soluble Sodium Percentage and Magnesium Ratio, which indicated potential problems with excess of sodium (about 30% of the samples) and magnesium (about 20%). Further studies combining suitability indices, soil characteristics and crop types are recommended to assess water quality for irrigation use. Article Highlights: The southern slopes of Mt. Access to safe drinking water sources and appropriate sanitation facilities remains a dream in low and middle-income countries including South Africa. This study identified the origin of faecal pollution by using quantitative polymerase chain reaction (qPCR) targeting host-specific Bacteroidales genetic markers to track the distribution of human-specific (BacHum) and animal-specific (cattle—BacCow, chicken—Cytb, pig—Pig-2-Bac, dog—BacCan) markers in water sources used by rural communities of the Vhembe District Municipality (VDM). Results revealed the prevalence of BacHum, BacCow, and BacCan in all surface water sources in Thulamela Local Municipality (TLM) and Collins Chabane Local Municipality (CLM) during wet (100%) and dry seasons (50–75%). Cytb was not detected in untreated spring water in TLM and CLM, and Pig-2-Bac was not detected in untreated hand-dug well water in TLM during both seasons. Accurate taxonomic species identification is essential to the study of mammals. Despite this necessity, rapid and accurate identification of cryptic, understudied, and elusive mammals remains challenging. Traditional barcoding of mitochondrial genes is standard for molecular identification but requires time-consuming wet-lab methodologies. Recent bioinformatic advancements for nanopore sequencing data offer exciting opportunities for noninvasive and field-based identification of mammals. Nanopore adaptive sampling (NAS), a polymerase chain reaction (PCR)-free method, selectively sequences regions of DNA according to user-specified reference databases. Here, we utilized NAS to enrich mammalian mitochondrial genome sequencing to identify species. Fecal DNA extractions were sequenced from 9 mammals, several collected in collaboration with Minnesota Tribal Nations, to demonstrate utility for NAS barcoding of noninvasive samples. By mapping to the entire National Center for Biotechnology Information mammalian mitochondrial reference genome database and bioinformatically analyzing highly similar matches, we successfully produced species identifications for all fecal samples. Eight of 9 species identifications matched previous PCR or animal/fecal appearance-based identifications. For the ninth species, our genetic data indicate a misidentification stemming from the original study. Our approach has a range of applications—particularly in field-based wildlife research, conservation, disease surveillance, and monitoring of wildlife trade. Of importance to Minnesota tribes is invasive species monitoring, detections, and confirmation as climate impacts cause changes in biodiversity and shifts in species distributions. The rapid assessment techniques described here will be useful as new introductions and range expansions of native and invasive species may first be detected by the presence of signs such as scat rather than direct observations and will be helpful for chronically understaffed tribal natural resources agencies. Background: Ensuring the availability of safe drinking water remains a critical challenge in developing countries, including Ethiopia. Therefore, this paper aimed to investigate the prevalence of fecal coliform and E. coli bacteria and, geographical, children availability, and seasonal exposure assessment through a meta-analysis. Methods: Two independent review groups extensively searched internet databases for English-language research articles published between 2013 and 2023. This systematic review and meta-analysis followed PRISMA guidelines. The methodological quality of each included study was evaluated using the STROBE guidelines. Publication bias was assessed by visual inspection of a funnel plot and then tested by the Egger regression test, and meta-analysis was performed using DerSimonian and Laird random-effects models with inverse variance weighting. Subgroup analyses were also conducted to explore heterogeneity. Results: Out of 48 potentially relevant studies, only 21 fulfilled the inclusion criteria and were considered for meta-analysis. The pooled prevalence of fecal coliform and E. coli was 64% (95% CI: 56.0–71.0%, I2 = 95.8%) and 54% (95% CI: 45.7–62.3%, I2 = 94.2%), respectively. Subgroup analysis revealed that the prevalence of fecal coliform bacteria increased during the wet season (70%) compared to the dry season (60%), particularly in households with under-five children (74%) compared to all households (61%), in rural (68%) versus urban (66%) areas, and in regions with high prevalence such as Amhara (71%), Gambela (71%), and Oromia (70%). Similarly, the prevalence of E. coli was higher in households with under-five children (66%) than in all households (46%). Conclusions: The analysis highlights the higher prevalence of fecal coliform and E. coli within households drinking water, indicating that these bacteria are a significant public health concern. Moreover, these findings emphasize the critical need for targeted interventions aimed at improving drinking water quality to reduce the risk of fecal contamination and enhance public health outcomes for susceptible groups, including households with under-five children, in particular geographical areas such as the Amhara, Gambela, and Oromia regions, as well as rural areas, at point-of-use, and during the rainy season. Escherichia coli is commonly found in the gastrointestinal tract of warm-blooded animals and is routinely used as an indicator of fecal contamination in recreational waters. While most E. coli are commensal, some can harbor pathogenic potential, posing a major public health risk. This study investigated the sources of fecal contamination in recreational waters through the characterization of E. coli isolates. Presumptive E. coli detection was performed using selective and differential media, confirmed by PCR, and followed by characterization. E. coli were detected in all studied aquatic environments, with most isolates belonging to phylogenetic groups D1 (24%, non-human mammals) and B1 (20%, birds), suggesting animals as primary contaminants sources. Among E. coli isolates, 35% were identified as diarrheagenic E. coli (DEC), with enterotoxigenic (34%) and enterohemorrhagic (26%) being the most prevalent pathotypes. Furthermore, 85% of all isolates and 86.5% of DEC isolates exhibited multi-drug resistance, with highest rates displayed in phylogenetic subgroups A1 (human and non-human mammals) and B1. This study highlights the importance of the One Health approach encompassing a human, animal, and environmental holistic health perspective to effectively manage public health strategies and ensure the safety of beachgoers. Background and purpose: Early detection, diagnosis, and treatment of colorectal cancer and its precancerous lesions can significantly improve patients' survival rates. The purpose of this research is to identify methylation markers specific to colorectal cancer tissues and validate their diagnostic capability in colorectal cancer and precancerous changes by measuring the level of DNA methylation in stool samples. Method: We analyzed samples from six cancer tissues and adjacent normal tissues and fecal samples from 758 participants, including 62 patients with interfering diseases. Bioinformatics databases were used to screen for candidate biomarkers for CRC, and quantitative methylation-specific PCR methods were applied for identification. The methylation levels of the candidate biomarkers in fecal and tissue samples were measured. Logistic regression and random forest models were built and validated using fecal sample data from one of the centers, and the independent or combined diagnostic value of the candidate biomarkers in fecal samples for CRC and precancerous lesions was analyzed. Finally, the diagnostic capability and stability of the model were validated at another medical center. Results: This study identified two colorectal cancer CpG sites with tissue specificity. These two biomarkers have certain diagnostic power when used individually, but their diagnostic value for colorectal cancer and colorectal adenoma is more significant when they are used in combination. Conclusion: The results indicate that a DNA methylation biomarker combined diagnostic model based on two CpG sites, cg13096260 and cg12587766, has the potential for screening and diagnosing precancerous lesions and colorectal cancer. Additionally, compared to traditional diagnostic models, machine learning algorithms perform better but may yield more false-positive results, necessitating further investigation. Porcine Teschoviruses (PTVs) are ubiquitous enteric viral pathogens that infect pigs and wild boars worldwide. PTVs have been responsible for causing the severe clinical disease (Teschen disease) to asymptomatic infections. However, to date, limited information is available on large-scale epidemiological data and molecular characterization of PTVs in several countries. In this study, we report epidemiological data on PTVs based on screening of 534 porcine fecal samples from different states of India and a RT-PCR based detection of PTVs shows a percent positivity of 8.24% (44/534). The PTV prevalence varied among different regions of the country with the highest detection rates observed in the state of Karnataka (38.1%). Phylogenetic analysis based on VP1 gene reveals the presence of PTV genotype 6 and 13 along with some unassigned novel genotypes which did not cluster with any of the established PTV genotypes (PTV 1–PTV 13). Indian PTV 6 strains are genetically closest to the Spanish strains (85.7–94.4%) whereas PTV 13 and novel genotype strains were found to be more similar to the Chinese strains (88.1–99.1%). Using recombination detection software, no Indian PTVs found to be recombinant on VP1 gene and selection pressure analysis revealed the purifying selection in the several sites of the VP1 gene of PTVs. The Bayesian analysis of Indian PTVs shows 1.16 × 10–4 substitution/site/year as the mean evolutionary rate. Further, isolation of the novel PTV strains from India and more detailed investigation much needed to know the evolutionary history of PTV strains circulating in porcine populations in India. Prevalence of water borne diseases is a public health concern. The study assessed the quality of selected borehole waters in Jos North for their potability. The physicochemical parameters were assessed in addition to chlorides, phosphates, nitrates, and sulphates, total solids, total dissolved solids, total suspended solids, total hardness, using standard methods. Atomic absorption spectrophotometry used for elemental analysis of samples for manganese, lead, cadmium, copper and zinc. microbial analysis was carried out to test for presence of anaerobic bacteria, coliforms, Escherichia coli, Salmonella typhi, yeast and mould, Staphylococcus aureus using the plate count method. Water Quality Index by Weighted Arithmetic Index Method. Results ranged from 22.19-56.22 mg/L (chlorides), 0.00-0.07 mg/L (phosphates), 0.11-0.19 mS/cm (conductivity), 141.40-169.40 mg/L (alkalinity) and 86.70-160 mg/L (TSS). Kyan Primary school had Mn 0.632 mg/L against 0.500mg/L recommended by WHO. Microbial analysis revealed that Kyan primary school had 1 cfu per 100 mL water for E. coli, Salmonella typhi, yeast and mould. This indicated faecal contamination. The WQI revealed Village hostel, Kyan Village, Gadabiu had values of 6.54, 19.41, 10.97 are potable, Government College, Angwan Jarawa, Kyan Primary school had 28.90, 61.50, 93.60 are potable, poor, very poor, not potable. (Objective) This study aimed to develop the application of Polygonum lapathifolium in Luzhou-flavor Fuqu. (Methods) In the process of making Luzhou-flavor Fuqu, P. lapathifolium was added. The culture conditions such as the addition amount of P. lapathifolium, the inoculation amount of Luzhou-flavor Daqu, the moisture content and the culture time were optimized. The number of bacteria, the number of yeasts and the activity of glucoamylase were used as indicators to determine the optimal culture conditions. Under the optimal process conditions, GC-MS was used to detect the volatile flavor components in Luzhou-flavor Fuqu Baijiu. (Results) When the addition amount of P. lapathifolium was 8%, the inoculation amount of Luzhou-flavor Daqu was 4%, the moisture content was 55%, and the culture time was 48 h, the number of bacteria in Fuqu was 6.22X 107 CFU/g, the number of yeasts was 6.14X 108 CFU/g, and the glucoamylase activity was 671 U/g. 57 and 51 volatile flavor components were detected in Luzhou-flavor Fuqu Baijiu with and without P. lapathifolium. This research aims to understand the extent of microplastic contamination in Pekalongan waters, Central Java, and its potential impact on fishing grounds, aligning with Indonesia's National Action Plan for Handling Marine Debris 2018–2025. The study employs a 2D hydrodynamics modelling approach with Mike 21 Software to map the spatial distribution of microplastic movement concerning fishing areas during the west and east monsoon seasons. The results showed that microplastic particles follow tidal currents in Pekalongan waters, with their movement influenced by factors such as current, wind, and tidal conditions. The trajectory of microplastics entering fishing ground areas poses potential contamination risk for fish caught by fishermen, threatening the health of marine ecosystems and the stability of their structure and function. Monitoring of stream water quality is a key element of water resource management worldwide, but methods that are commonly used in temperate habitats may not be appropriate in humid tropical systems. We assessed the influence of four land uses on microbial water quality in 21 streams in the Panama Canal Watershed over a one-year period, using a common culture-based fecal indicator test and 16S rDNA metabarcoding. Each stream was located within one of four land uses: mature forest, secondary forest, silvopasture, and traditional cattle pasture. Culturing detected total coliforms and Escherichia coli across all sites but found no significant differences in concentrations between land uses. However, 16S rDNA metabarcoding revealed variability in the abundance of coliforms across land uses and several genera that can cause false positives in culture-based tests. Our results indicate that culture-based fecal indicator bacteria tests targeting coliforms may be poor indicators of fecal contamination in Neotropical oligotrophic streams and suggest that tests targeting members of the Bacteroidales would provide a more reliable indication of fecal contamination. Fecal material is one of the principal components in domestic wastewater (DWW). In this sense, Escherichia coli is a recognized indicator of fecal pollution due to its low-cost monitoring in DWW. Moreover, DWW are a pathogen reservoir that become a health hazard when its discharges are poured to water bodies without treatment. In this study, a simple and low-cost method was developed to functionalize the leaves (LC), peel (PC), and seeds (SC) of Moringa oleifera (MO), with an saline extract (NaCl 0.35 M), to obtain coagulants capable to reduce the presence of bacteria indicating fecal contamination and other pollution indicators in Guerrero, Mexico. The optimal doses were determined with a surface response using Sigma Plot® 8.10 software through a factor design of mixed levels. The optimal conditions for the reduction of the microbiological and physicochemical parameters were PC and SC, with concentrations of 70 g/l of the grinded material used to prepare the MO-coagulants, with doses of 1 and 6 mL, respectively, evaluated for the pollutant's removal in the wastewater samples. The results showed the removal of fecal coliforms (99.3%), E. coli (92%), nitrates (47.9%), total dissolved solids (63%), turbidity (90%) and a dissolved oxygen concentration (DO) of 7.1 g/l in the treated wastewater. The developed method, using MO, is a simple and efficient tool for the reduction of bacteria indicating fecal contamination and other pollution indicators present in DWW. Microbes present in the vermicompost plays a major role in exhibiting the beneficiary effect which results in significant plant growth and yield. In this study, we aim to analyze the microbial load present in the vermicompost which is prepared from different medicinal plants including Phyllanthus emblica, Senna auriculata, Justicia adhatoda, Annona squamosa at two different ratios of preparation with cow dung. To perform this, microbial load was evaluated before and after vermicompost formation. After completion, it was observed that fungal and actinomycetes loads were significantly higher in vermicompost samples prepared from Phyllanthus emblica plant samples. Concerning bacterial load, it was observed that vermicompost samples prepared from Senna auriculata showed the maximum load. These data provide us the information about the importance of the vermicompost with different medicinal plants in increasing the microbial load concentration which further enables plant growth and yield. It is also noted that the ratio of 1:2 gave better microbial load results than the 1:1 ratio sample. Micropollutants also known as "emerging micropollutants" (EMPs) have gained significant attention in research, with a particular emphasis on the ocean, marine water, and river pollution. The interest arises from the impact of EMPs on aquatic ecosystems, often originating from sources such as wastewater treatment plant (WWTP) and septic tanks leakage. However, EMPs pollution in surface water can also be caused by leakage of septic tanks. In Indonesia, where septic tanks maintenance is generally neglected, there is a potential risk of EMPs contamination. The outcome and transformation of EMPs, as well as their potential transport pathways, are influenced by sanitary conditions and the effectiveness of existing septic tanks. This concern has escalated to the point of causing fecal contamination in drinking water sources. Therefore, this research focuses on EMPs, specifically pharmaceutical EMPs, leakage from septic tanks, and their possible fate and transport. Access to safe drinking water sources and appropriate sanitation facilities remains a dream in low and middle-income countries including South Africa. This study identified the origin of faecal pollution by using quantitative polymerase chain reaction (qPCR) targeting host-specific Bacteroidales genetic markers to track the distribution of human-specific (BacHum) and animal-specific (cattle—BacCow, chicken—Cytb, pig—Pig-2-Bac, dog—BacCan) markers in water sources used by rural communities of the Vhembe District Municipality (VDM). Results revealed the prevalence of BacHum, BacCow, and BacCan in all surface water sources in Thulamela Local Municipality (TLM) and Collins Chabane Local Municipality (CLM) during wet (100%) and dry seasons (50–75%). In Fiji, 90% of the population has access to basic sanitation; however, there are still persistent health risks from endemic faecal-oral diseases such as typhoid fever. There is a need to assess the contribution of existing sanitation facilities in the faecal pathogen transmission pathway. This study was conducted as part of a larger planetary health study across 29 rural communities within five river catchments. This specific research aimed to characterise latrine front-ends, both infrastructure and usage behaviour, and to assess the faecal contamination levels on various frequently contacted latrine surfaces in rural Fiji. A sanitation survey, along with observation and latrine swab sampling, was conducted in households over three phases: baseline (n = 311) (Aug–Dec 2019), endline (n = 262) (Jun–Sep 2022) and an in-depth front-end study (n = 12) (Oct–Nov 2022). Of 311 households, almost all had pedestal-type latrines, predominately cistern-flush (83%), followed by pour-flush (13%), and then hole-type (pit) latrines (4%). Washable latrine floors had significantly higher E. coli densities (6.7 × 102 CFU/25 cm2) compared to non-washable floors (1.3 × 102 CFU/25 cm2) (p = 0.05), despite washable floors indicating improved latrines. The in-depth front-end analysis found that moist latrine surfaces had significantly elevated E. coli densities (1.2 × 103 CFU/25 cm2) compared to the dry ones (14.3 CFU/25 cm2) (p < 0.001), highlighting the importance of maintaining dry latrine surfaces. Latrine floors and mid-walls were the most frequently contaminated surfaces, emphasising the need to clean and disinfect these surfaces. Only 46% of the households reported always using soap for handwashing after defecation, exacerbating the risk of transmitting faecal pathogens. This study highlights that latrine cleanliness and hygiene are as crucial as latrine infrastructures for the effective disruption of faecal pathogens transmission during latrine use. Safe drinking water access is a human right, but data on safely managed drinking water services (SMDWS) is lacking for more than half of the global population. We estimate SMDWS use in 135 low- and middle-income countries (LMICs) at subnational levels with a geospatial modeling approach, combining existing household survey data with available global geospatial datasets. We estimate that only one in three people used SMDWS in LMICs in 2020 and identified fecal contamination as the primary limiting factor affecting almost half of the population of LMICs. Our results are relevant for raising awareness about the challenges and limitations of current global monitoring approaches and demonstrating how globally available geospatial data can be leveraged to fill data gaps and identify priority areas in LMICs.
