Research indicates that brain-region-specific synapse loss and dysfunction are early hallmarks and stronger neurobiological correlates of cognitive decline in Alzheimer's disease (AD) than amyloid plaque and neurofibrillary tangle counts or neuronal loss. Even though the precise mechanisms underlying increased synaptic pruning in AD are still unknown, it has been confirmed that dysregulation of the balance between complement activation and inhibition is a crucial driver of its pathology. The complement includes three distinct activation mechanisms, with the activation products C3a and C5a, potent inflammatory effectors, and a membrane attack complex (MAC) leading to cell lysis. Besides pro-inflammatory cytokines, the dysregulated complement proteins released by activated microglia bind to amyloid β at the synaptic regions and cause the microglia to engulf the synapses. Additionally, research indicating that microglia-removed synapses are not always degenerating and that suppression of synaptic engulfment can repair cognitive deficits points to an essential opportunity for intervention that can prevent the loss of intact synapses. In this study, we focus on the latest research on the role and mechanisms of complement-mediated microglial synaptic pruning at different stages of AD to find the right targets that could interfere with complement dysregulation and be relevant for therapeutic intervention at the early stages of the disease. Chat GPT is a highly advanced chat bot that stands out from other chat bots due to its remarkable ability to generate impressive prose in mere seconds. It has gained a lot of attention and sparked concerns about its potential impact on student assessments in higher education and various other fields. The goal of this study is to assess the potential benefits and drawbacks of using Chat GPT technology in educational settings, as well as any prospective impacts on student learning outcomes and educator pedagogy. The design of the study is a literature review. Our research has shown that there are both proponents and critics of using Chat GPT in education. Many proponents argue that Chat GPT has the potential to revolutionize the way we teach and learn. Its ability to generate human-like responses and adapt to the user's language and style of communication makes it an effective tool for personalized learning. On the other hand, there are also concerns about the use of Chat GPT in education. Critics contend that it might not be as successful at teaching critical thinking and problem-solving techniques and that it might result in a decline in human connection and individualized learning. According to our research, Chat GPT has certain drawbacks and difficulties even though it has the potential to be an effective teaching tool. Therefore, it is important that educators and policymakers carefully consider the potential advantages and disadvantages of using Chat GPT in their classrooms and develop appropriate strategies for responsible use. TikTok, a well-known application for creating and posting short videos on social media, is an untested educational tool. This study examines ways in which TikTok has impacted pharmacy education with a specific focus on learning concerning drugs. The study compared two groups to see if there are differences between the group using traditional learning strategies - such as textbooks and online resources - and the group using the TikTok platform. Multiple-choice tests were used to evaluate the performance of the two groups. Results show a statistically significant difference between traditional and TikTok-supported learning groups regarding academic performance, highlighting the potential of TikTok-supported learning for improving learning outcomes. These results make valuable contributions to the study of teaching methods and provide evidence of encouraging impacts for implementing advanced social media-based learning strategies in traditional learning environments. We evaluated the dependency of neutrophil [O.sup.-.sub.2] production on PTK-Lyn and MAPK-ERK1/2 in rats after thermal injury. Activation of PTK-Lyn was assessed by immunoprecipitation. Phosphorylation of ERK1/2 was assessed by Western blot analysis. [O.sup.-.sub.2] production was measured by isoluminol-enhanced luminometry. Imaging technique was employed to measure neutrophil [[[Ca.sup.2+]].sub.i] in individual cells. Thermal injury caused marked upregulation of Lyn and ERK1/2 accompanying enhanced neutrophil [O.sup.-.sub.2] production. Treatment of rats with PTK blocker (AG556) or MAPK blocker (AG1478) before burn injury caused complete inhibition of the respective kinase activation. Both AG556 and AG1478 produced an ~66% inhibition in [O.sup.-.sub.2] production. Treatment with diltiazem (DZ) produced an ~37% inhibition of [O.sup.-.sub.2] production without affecting Lyn or ERK1/2 activation with burn injury. [Ca.sup.2+] mobilization was upregulated with burn injury but not affected by treatment of burn rats with AG556. Unlike the partial inhibition of burn-induced [O.sup.-.sub.2] production by AG556, AG1478, or DZ, platelet-activating factor antagonist (PAFa) treatment of burn rats produced near complete inhibition of [O.sup.-.sub.2] production. PAFa treatment also blocked activation of Lyn. The findings suggest that the near complete inhibition of [O.sup.-.sub.2] production by PAFa was a result of blockade of PTK as well as [Ca.sup.2+] signaling. The mechanism of alcohol-mediated increased infection in burn patients remains unknown. With the use of a rat model of acute alcohol and burn injury, the present study ascertained whether acute alcohol exposure before thermal injury enhances gut bacterial translocation. On day 2 postinjury, we found a severalfold increase in gut bacterial translocation in rats receiving both alcohol and burn injury compared with the animals receiving either injury alone. Whereas there were no demonstrable changes in intestinal morphology in any group of animals, a significant increase in intestinal permeability was observed in ethanol- and burn-injured rats compared with the rats receiving either injury alone. We further examined the role of intestinal immune defense by determining the gut-associated lymphoid (Peyer's patches and mesenteric lymph nodes) T cell effector responses 2 days after alcohol and burn injury. Although there was a decrease in the proliferation and interferon-[gamma] by gut lymphoid T cells after burn injury alone; the suppression was maximum in the group of rats receiving both alcohol and burn injuries. Furthermore, the depletion of CD[3.sup.+] cells in healthy rats resulted in bacterial accumulation in mesenteric lymph nodes; such CD[3.sup.+] cell depletion in alcohol- and burn-injured rats furthered the spread of bacteria to spleen and circulation. In conclusion, our data suggest that the increased intestinal permeability and a suppression of intestinal immune defense in rats receiving alcohol and burn injury may cause an increase in bacterial translocation and their spread to extraintestinal sites.
