The temporal signals from a large gas detector may show dynamical scaling due to many correlated space points created by the charged particles while passing through the tracking medium. This has been demonstrated through simulation using realistic parameters of a Time Projection Chamber (TPC) being fabricated to be used in ALICE collider experiment at CERN. An interesting aspect of this dynamical behavior is the existence of an universal scaling which does not depend on the multiplicity of the collision. This aspect can be utilised further to study physics at the device level and also for the online monitoring of certain physical observables including electronics noise which are a few crucial parameters for the optimal TPC performance., Comment: 5 pages, 6 figures

This paper presents the simulation of the GLAST high energy gamma-ray telescope. The simulation package, written in C++, is based on the Geant4 toolkit, and it is integrated into a general framework used to process events. A detailed simulation of the electronic signals inside Silicon detectors has been provided and it is used for the particle tracking, which is handled by a dedicated software. The ALICE experiment at CERN will propose unprecedented requirements for event building and data recording. New technologies will be adopted as well as ad-hoc frameworks, from the acquisition of experimental data up to the transfer onto permanent media and its later access. These issues justify a careful, in-depth planning and preparation. The ALICE Data Challenge is a very important step of this development process where simulated detector data is moved from dummy data sources up to the recording media using processing elements and data-paths as realistic as possible. - Abstract
This document describes the guidelines developed by the Scientific Association Gruppo Tossicologi Forensi Italiani (GTFI) for the determination of psychoactive substances in biological samples for forensic toxicology and medico-legal purposes. The guidelines provide indications and recommendations for performing forensic toxicological and medico-legal analyses with a high level of quality, with particular reference to the use of appropriate analytical techniques and established analytical procedures and methods shared by the national scientific community, in order to ensure results of high reliability and reproducibility. The correct methods for the determination of alcohol and psychoactive drugs in biological samples may also be of interest to other scientific communities, given the wide range of applications of the various toxicological examinations required and carried out not only for forensic purposes, but also for clinical-diagnostic purposes, the results of which may constitute evidence in administrative or criminal contexts. In this sense, and in the framework of an already existing collaboration between the GTFI and the Italian Society of Clinical Pathology and Laboratory Medicine (SIPMeL), the publication of an extract from the GTFI guidelines may also be useful for professionals belonging to these scientific societies who perform laboratory tests for various toxicological purposes. This document represents an abbreviated version of the original guidelines (Revision No. 6 of 2022), which can be accessed in full at the link https://www.gtfi.it/wp-content/uploads/2023/02/LineeGuidaGTFI-MaterialeBiologico-rev06-08giu2022.pdf, in order to keep the length within appropriate limits for a scientific journal. - Abstract
The aim of the study was to validate the impact of the single-nucleotide polymorphism rs2413739 (T > C) in the PACSIN2 gene on thiopurines pharmacological parameters and clinical response in an Italian cohort of pediatric patients with acute lymphoblastic leukemia (ALL) and inflammatory bowel disease (IBD). In ALL, PACSIN2 rs2413739 T allele was associated with a significant reduction of TPMT activity in erythrocytes (p = 0.0094, linear mixed-effect model, multivariate analysis considering TPMT genotype) and increased severe gastrointestinal toxicity during consolidation therapy (p = 0.049). A similar trend was present also for severe hematological toxicity during maintenance. In IBD, no significant effect of rs2413739 could be found on TPMT activity, however azathioprine effectiveness was reduced in patients carrying the T allele (linear mixed effect, p = 0.0058). In PBMC from healthy donors, a positive correlation between PACSIN2 and TPMT protein concentration could be detected (linear mixed effect, p = 0.045). These results support the role of PACSIN2 polymorphism on TPMT activity and mercaptopurine adverse effects in patients with ALL. Further evidence on PBMC and pediatric patients with IBD supports an association between PACSIN2 variants, TPMT activity, and thiopurines effects, even if more studies are needed since some of these effects may be tissue specific.

- Abstract
Purpose: Idiopathic nephrotic syndrome (INS) is the most frequent form of childhood nephrotic syndrome. Steroids represent the best therapeutic option; however, inter-individual differences in their efficacy and side effects have been reported. To date, there is no way to predict patients’ resistance and/or dependence. Alterations in the cytokine profile of INS patients might contribute to proteinuria and glomerular damage and affect drug sensitivity. Methods: The cytokine plasma levels were measured in 21 INS children at diagnosis to investigate the association among cytokines pattern and clinical response. Patients were selected on the basis of their clinical response: 7 steroid sensitive (SS), 7 dependent (SD), and 7 resistant (SR). Significant results were then analyzed in 41 additional pediatric INS patients. Results: Within the 48 cytokines analyzed, macrophage migration inhibitory factor (MIF) was a good predictor of steroid response. Indeed, SR patients showed significantly higher MIF plasma levels compared with all others (p = 0.022; OR = 4.3, 95%CI = 1.2–25.4): a cutoff concentration of MIF > 501 pg/ml significantly discriminated SR patients (sensitivity = 85.7%, specificity = 71.4%). On the contrary, SD patients showed lower MIF plasma levels compared with others (p = 0.010; OR = 0.12, 95%CI = 9.2 × 10−3–6.7 × 10−1). Significant results were confirmed in the entire cohort. Conclusions: Our comprehensive cytokine analysis indicates that assessing MIF plasma levels at diagnosis could predict response to glucocorticoids in children with INS.

- Abstract
ALICE is a general-purpose heavy-ion experiment designed to study the physics of strongly interacting matter and the quark-gluon plasma in nucleus-nucleus collisions at the LHC. It currently includes more than 900 physicists and senior engineers, from both nuclear and high-energy physics, from about 80 institutions in 28 countries. The experimentwas approved in February 1997. The detailed design of the different detector systems has been laid down in a number of Technical Design Reports issued between mid-1998 and the end of 2001 and construction has started for most detectors. Since the last comprehensive information on detector and physics performance was published in the ALICE Technical Proposal in 1996, the detector as well as simulation, reconstruction and analysis software have undergone significant development. The Physics Performance Report (PPR) will give an updated and comprehensive summary of the current status and performance of the various ALICE subsystems, including updates to the Technical Design Reports, where appropriate, as well as a description of systems which have not been published in a Technical Design Report. The PPR will be published in two volumes. The currentVolume I contains: 1. a short theoretical overview and an extensive reference list concerning the physics topics of interest to ALICE, 2. relevant experimental conditions at the LHC, 3. a short summary and update of the subsystem designs, and 4. a description of the offline framework and Monte Carlo generators. Volume II, which will be published separately, will contain detailed simulations of combined detector performance, event reconstruction, and analysis of a representative sample of relevant physics observables from global event characteristics to hard processes. © 2004 IOP Publishing Ltd.

ALICE; ALICE is a general-purpose heavy-ion experiment designed to study the physics of strongly interacting matter and the quark-gluon plasma in nucleus-nucleus collisions at the LHC. It currently includes more than 900 physicists and senior engineers, from both nuclear and high-energy physics, from about 80 institutions in 28 countries. The experiment was approved in February 1997. The detailed design of the different detector systems has been laid down in a number of Technical Design Reports issued between mid-1998 and the end of 2001 and construction has started for most detectors. Since the last comprehensive information on detector and physics performance was published in the ALICE Technical Proposal in 1996, the detector as well as simulation, reconstruction and analysis software have undergone significant development. The Physics Performance Report (PPR) will give an updated and comprehensive summary of the current status and performance of the various ALICE subsystems, including updates to the Technical Design Reports, where appropriate, as well as a description of systems which have not been published in a Technical Design Report.

The ALICE experiment at CERN will propose unprecedented requirements for event building and data recording. New technologies will be adopted as well as ad-hoc frameworks, from the acquisition of experimental data up to the transfer onto permanent media and its later access. These issues justify a careful, in-depth planning and preparation. The ALICE Data Challenge is a very important step of this development process where simulated detector data is moved from dummy data sources up to the recording media using processing elements and data-paths as realistic as possible. We will review herein the current status of past, present and future ALICE Data Challenges, with particular reference to the sessions held in 2002 when - for the first time - streams worth one week of ALICE data were recorded onto tape media at sustained rates exceeding 300 MB/s.

(3-Glycidoxypropyl)trimethoxysilane (GPTMS) derived hybrid organic-inorganic materials have been obtained by a new multistep nonhydrolytic/hydrolytic synthesis. The new synthesis has been achieved using titanium(IV) chloride instead of titanium alkoxides to catalyze the epoxy ring opening in the organically modified alkoxide. The precursor sols were studied by multinuclear solution-state magnetic resonance (NMR) techniques, gas chromatography coupled with mass spectrometry (GC-MS), and electrospray ionization (ESI) experiments. The derived powdered gels were characterized by multinuclear solid-state magic angle spinning (MAS) NMR, Fourier transform infrared (FT-IR), and matrix-assisted laser desorption/ionization (MALDI) experiments. All these results show that epoxy ring opening is achieved at low temperature and within short reaction times. Interestingly, several reactions occur simultaneously and are involved in the early polymerization process, leading to fast catalyst deactivation in solution, chlorination of GPTMS organic groups, and titanium dispersion inside precursor sols. Despite a rather complex reaction pathway, this method can be used to prepare transparent hybrid organic-inorganic materials, whose properties can satisfy the requirements for photonic applications.

The aim of this study was to evaluate the in vitro steroid sensitivity as a predictor of clinical response to glucocorticoids in childhood idiopathic nephrotic syndrome (INS). Seventy-four patients (median age 4.33, interquartile range [IQR] 2.82-7.23; 63.5% male) were enrolled in a prospective multicenter study: in vitro steroid inhibition of patients' peripheral blood mononuclear cell proliferation was evaluated by [methyl-
3 H] thymidine incorporation assay at disease onset (T0) and after 4 weeks (T4) of treatment. Steroid dependence was associated with increased in vitro sensitivity at T4 assessed both as drug concentration inducing 50% of inhibition (IC50 ; odds ratio [OR] = 0.48, 95% confidence interval [CI] = 0.24-0.85; P = 0.0094) and maximum inhibition at the highest drug concentration (Imax ; OR = 1.13, 95% CI = 1.02-1.31; P = 0.017). IC50 > 4.4 nM and Imax < 92% at T4 were good predictors for optimal clinical response.
