- Abstract
Conservation translocations are becoming common conservation practice, so there is an increasing need to understand the drivers of plant translocation performance through reviews of cases at global and regional levels. The establishment of the Italian Database of Plant Translocation (IDPlanT) provides the opportunity to review the techniques used in 186 plant translocation cases performed in the last 50 years in the heart of the Mediterranean Biodiversity Hotspot. We described techniques and information available in IDPlanT and used these data to identify drivers of translocation outcomes. We tested the effect of 15 variables on survival of translocated propagules as of the last monitoring date with binomial logistic mixed-effect models. Eleven variables significantly affected survival of transplants: life form, site protection, material source, number of source populations, propagation methods, propagule life stage, planting methods, habitat suitability assessment, site preparation, aftercare, and costs. The integration of vegetation studies in the selection of suitable planting sites significantly increased the success of translocation efforts. Although posttranslocation watering had a generally positive effect on translocation outcome, other aftercare techniques did not always increase transplant survival. Finally, we found that how funds were spent appeared to be more important than the actual amount spent. Plant translocations in Italy and in the Mediterranean area should account for the complexity of speciation, gene flow, and plant migrations that has led to local adaptations and has important implications for the choice and constitution of source material.

- Abstract
Recent literature on school counseling has focused on the need for new directions for school counseling and redefined roles for school counselors. However, since the 1984 supplement to the High School and Beyond (HS&B) survey, no national data have been collected to describe guidance counseling programs and activities. To help address this lack of current information, the National Center for Education Statistics (NCES) conducted a survey on high school guidance counseling in spring 2002 for the Office of Vocational and Adult Education, U.S. Department of Education. The survey, conducted through the NCES Fast Response Survey System (FRSS), provides a description of public high school guidance programs, activities, and staff in 2002. This E.D. Tab report summarizes findings for all public high schools in the 2002 FRSS survey and the 1984 supplement to HS&B. Findings for schools in the FRSS survey are also presented by the following school characteristics: enrollment size, locale, percentage of college-bound students, and number of vocational courses offered per 100 students. This executive summary presents highlights of findings for all public high schools and compares results from the FRSS survey and the supplement to HS&B concerning program goals, written plans, and selected guidance activities. Three appendixes contain the research methodology, the FRSS Survey Instrument, and selected items from the 1984 supplement to the HS&B survey. (Contains 11 references and 20 tables.) (GCP)

- Abstract
IDPlanT is the Italian Database of Plant Translocation, an initiative of the Nature Conservation Working Group of the Italian Botanical Society. IDPlanT currently includes 185 plant translocations. The establishment of a national database on plant translocation is a key step forward in data sharing and techniques improvement in this field of plant conservation. 5. Modified streptozotocin‐induced diabetic model in rodents

Abstract Streptozotocin (STZ)‐induced type I diabetes mellitus (DM) models have been pivotal in diabetes research due to their ability to mimic the insulin‐dependent hyperglycemia akin to human type I diabetes. However, these models often suffer from poor induction rates and low survival post‐STZ induction, especially in long‐term experiments, necessitating insulin supplementation, which introduces additional variables to experiments. To address this, we present a novel modification to the STZ‐induced DM model in C57BL/6J mice to improve survival rates without insulin supplementation. Our method involves non‐fasting, low‐dose STZ injections dissolved in pH‐neutral phosphate buffer saline instead of acidic sodium citrate buffer, administered over 5 days. We observed hyperglycemia induction in 94.28% of mice within a week post‐injection, with stable high blood glucose levels, stable body weight, and minimal mortality up to 21 weeks. Notably, omitting 10% sucrose in water and fasting did not affect hyperglycemia induction. Our findings suggest that the modified protocol not only decreases the experimental effort of the researchers, but reduces animal stress and mortality, thus enhancing experimental outcomes and animal welfare. By optimizing the STZ‐induced DM model in C57BL/6J mice, our study provides a valuable resource for researchers aiming to study diabetes and its complications while minimizing experimental variability and animal usage.

IntroductionThe potential neuroprotective and regenerative properties of electrical stimulation (ES) were studied in rhodopsin knockout mice (Rho−/−), a murine model of inherited retinal degeneration. The study focused on assessing the impact of varying ES frequencies on visual functions and photoreceptor cell survival in Rho−/− mice.MethodsTo elucidate the impact of electrical stimulation on cone survival, Rho−/− mice received either sham or transpalpebral ES using biphasic ramp or rectangular waveforms at 100 µA amplitude, starting at six weeks of age. The treatment duration spanned from one to three weeks. The optimal treatment frequency of ES sessions was determined by applying ES every one, two, or three days in three separate groups of Rho−/− mice. The sham group received daily treatments without the application of ES.ResultsOur study revealed significant improvement of visual function in Rho−/− mice following daily or every-other-day noninvasive transpalpebral ES, as evidenced by electroretinogram and optomotor response-based visual behavior assays. Concurrently, assessment of outer nuclear thickness and immunohistochemistry for the cone photoreceptor cell marker PNA demonstrated pronounced increases in the survival of rods and cones and improvement in the morphology of the inner and outer segments.DiscussionThis study underscores the protective effect of non-invasive ES in rhodopsin knockout-induced retinal degenerative disorders, providing a foundation for developing targeted therapeutic interventions for retinitis pigmentosa.

Assessing genetic diversity of narrow endemic plants is essential for their conservation. Gymnospermium scipetarum subsp. eddae (Berberidaceae) is an Italian endemic limited to a narrow forest area in the southern Apennines. Here we used random amplified polymorphic DNA (RAPD) markers to investigate population genetic differentiation and diversity in relation to distribution and microhabitat conditions. In all five extant populations and 134 individuals analysed we found a relatively low level of population genetic diversity (average Shannon-Weaver diversity index = 0.280; mean percentage of polymorphic bands = 57.45%; mean Nei's gene diversity H
e = 0.187). Percentage of polymorphic bands (PPB) at species level was higher than at the population level (83.33%), while total species diversity (He = 0.210) was comparable to that found in the two southernmost subpopulations. AMOVA revealed a weak differentiation among populations, which shared three genetic pools in their genetic structure. The estimated gene flow among populations was relatively high (mean Nm = 5.320), while the Mantel test revealed no significant correlation between geographical and genetic distances at the population level. We argue that information on genetic diversity of G. eddae have important implications for conservation programmes, particularly for the establishment of ex-situ collections and restocking conservation actions. Retinitis pigmentosa is a hereditary retinal disease that affects rod and cone photoreceptors, leading to progressive photoreceptor loss. Previous research supports the beneficial effect of electrical stimulation on photoreceptor survival. This study aims to identify the most effective electrical stimulation parameters and functional advantages of transcorneal electrical stimulation (tcES) in mice affected by inherited retinal degeneration. Additionally, the study seeked to analyze the electric field that reaches the retina in both eyes in mice and post-mortem humans. In this study, we recorded waveforms and voltages directed to the retina during transcorneal electrical stimulation in C57BL/6J mice using an intraocular needle probe with rectangular, sine, and ramp waveforms. To investigate the functional effects of electrical stimulation on photoreceptors, we used human retinal explant cultures and rhodopsin knockout (Rho–/–) mice, demonstrating progressive photoreceptor degeneration with age. Human retinal explants isolated from the donors’ eyes were then subjected to electrical stimulation and cultured for 48 hours to simulate the neurodegenerative environment in vitro. Photoreceptor density was evaluated by rhodopsin immunolabeling. In vivo Rho–/– mice were subjected to two 5-day series of daily transcorneal electrical stimulation using rectangular and ramp waveforms. Retinal function and visual perception of mice were evaluated by electroretinography and optomotor response (OMR), respectively. Immunolabeling was used to assess the morphological and biochemical changes of the photoreceptor and bipolar cells in mouse retinas. Oscilloscope recordings indicated effective delivery of rectangular, sine, and ramp waveforms to the retina by transcorneal electrical stimulation, of which the ramp waveform required the lowest voltage. Evaluation of the total conductive resistance of the post-mortem human compared to the mouse eyes indicated higher cornea-to-retina resistance in human eyes. The temperature recordings during and after electrical stimulation indicated no significant temperature change in vivo and only a subtle temperature increase in vitro (~0.5–1.5°C). Electrical stimulation increased photoreceptor survival in human retinal explant cultures, particularly at the ramp waveform. Transcorneal electrical stimulation (rectangular + ramp) waveforms significantly improved the survival and function of S and M-cones and enhanced visual acuity based on the optomotor response results. Histology and immunolabeling demonstrated increased photoreceptor survival, improved outer nuclear layer thickness, and increased bipolar cell sprouting in Rho–/– mice. These results indicate that transcorneal electrical stimulation effectively delivers the electrical field to the retina, improves photoreceptor survival in both human and mouse retinas, and increases visual function in Rho–/– mice. Combined rectangular and ramp waveform stimulation can promote photoreceptor survival in a minimally invasive fashion.

Plant traits—the morphological, anatomical, physiological, biochemical and phenological characteristics of plants—determine how plants respond to environmental factors, affect other trophic levels, and influence ecosystem properties and their benefits and detriments to people. Plant trait data thus represent the basis for a vast area of research spanning from evolutionary biology, community and functional ecology, to biodiversity conservation, ecosystem and landscape management, restoration, biogeography and earth system modelling. Since its foundation in 2007, the TRY database of plant traits has grown continuously. It now provides unprecedented data coverage under an open access data policy and is the main plant trait database used by the research community worldwide. Increasingly, the TRY database also supports new frontiers of trait-based plant research, including the identification of data gaps and the subsequent mobilization or measurement of new data. To support this development, in this article we evaluate the extent of the trait data compiled in TRY and analyse emerging patterns of data coverage and representativeness. Best species coverage is achieved for categorical traits—almost complete coverage for ‘plant growth form’. However, most traits relevant for ecology and vegetation modelling are characterized by continuous intraspecific variation and trait–environmental relationships. These traits have to be measured on individual plants in their respective environment. Despite unprecedented data coverage, we observe a humbling lack of completeness and representativeness of these continuous traits in many aspects. We, therefore, conclude that reducing data gaps and biases in the TRY database remains a key challenge and requires a coordinated approach to data mobilization and trait measurements. Summary: Microglia shift toward an inflammatory phenotype during aging that is thought to exacerbate age-related neurodegeneration. The molecular and cellular signals that resolve neuroinflammation post-injury are largely undefined. Here, we exploit systems genetics methods based on the extended BXD murine reference family and identify IGFBPL1 as an upstream cis-regulator of microglia-specific genes to switch off inflammation. IGFBPL1 is expressed by mouse and human microglia, and higher levels of its expression resolve lipopolysaccharide-induced neuroinflammation by resetting the transcriptome signature back to a homeostatic state via IGF1R signaling. Conversely, IGFBPL1 deficiency or selective deletion of IGF1R in microglia shifts these cells to an inflammatory landscape and induces early manifestation of brain tauopathy and retinal neurodegeneration. Therapeutic administration of IGFBPL1 drives pro-homeostatic microglia and prevents glaucomatous neurodegeneration and vision loss in mice. These results identify IGFBPL1 as a master driver of the counter-inflammatory microglial modulator that presents an endogenous resolution of neuroinflammation to prevent neurodegeneration in eye and brain.

Abstract: All known populations of the Sardinian endemic Centaurea filiformis Viv. (Asteraceae) were studied in order to understand the impact of both geographic and ecological factors on the genetic structuring of this species. Fourteen populations and 234 individuals were sampled. The demographic structure of the populations and the reproductive ecology were estimated in 28 plots. Population genetic analyses were based on SSR markers. Genetic structure was investigated by spatial Bayesian methods. Average densities of 0.51 individuals m−2 were detected, with a prevalence of adults. Ten species of pollinators were identified; C. filiformis ability to self‐pollinate and myrmecochory were demonstrated experimentally. The populations displayed an average heterozygosity value of He = 0.576 and high genetic differentiation (overall FST = 0.218). Bayesian analysis suggests that five is the most probable number of gene pools of origin. A strong correlation between geographic distances and genetic distances among populations was highlighted. The demographic population structure of C. filiformis is dominated by adults, suggesting that it is a stable‐regressive or senile species, investing more in local persistence than colonisation ability. Despite the scattered distribution, the populations studied do not present evidence of genetic erosion. The analysis of genetic differentiation reveals very high differentiation levels among populations, thus indicating that effective barriers exist against gene flow. A general conclusion is that population distribution results in a clear genetic structure for the populations studied, and that geography and not ecology is shaping the present distribution of this species. Contribution to the knowledge of the Sardinian Laurus nobilis L. stands, a priority habitat of the 92/43/EEC Directive. The results of the phytosociological study of the Sardinian Laurus nobilis L. stands are here presented. The statistical analysis of the surveys, carried out on the best known populations, allowed us to recognize four new plant communities, referred to one association and three subassociations. In Sardinia the Laurus nobilis L. stands are present on effusive and metamorphic substrata, on typic xerorthent or lithic xerorthent soils, in the oceanic pluviseasonal Mediterranean bioclimate. The investigated stands show a good conservation status and high recruitment levels, in relation to the high naturalness of the ravines and valleys in which they occur.

Hybridisation plays a prominent role in plant evolution due to its influence on genetic diversity, fitness and adaptive potential. We identify a case of on-going hybrid evolution of floral phenotypes in disjunct populations of Cyclamen balearicum and C. repandum subsp. repandum on Corsica and Sardinia., Hybrid populations on the two islands contain similar patterns of variation in flower colour and size but are probably at different stages in the evolutionary process of hybridisation, and differences in the frequency of floral types and flower size suggest hybrid vigour that may contribute to the dynamics and maintenance of hybrid forms., In a review of cases of hybridisation in Mediterranean plants we found an equivalent number of cases for the contemporary occurrence of mixed hybrid populations, as there are cases of homoploid hybrid species differentiation., We argue for the development of a conservation strategy for Mediterranean plants that integrates the need to protect not just pure endemic species (some of hybrid origin) but also mixed populations where adaptive variation and new species are evolving due to contemporary hybridisation. Authors report some results about a phytosociological study concerning the wetlands to the south of Olbia (Gallura, north-eastern Sardinia). Vegetation analysis allowed to detect 21 associations, among them two are new and denominated Halimionio portulacoidis-Limoniastretum monopetali ass. nova and Ephedro fragilis-Pistacietum lentisci ass. nova. Among them, the association Halimiono portulacoidis-Limoniastretum monopetali shows a particular phytogeographical interest, because it sets the Limoniastrum monopetalum vegetation in the only Sardinian station of this shrubby Plumbaginacea. From a syntaxonomic point of view, described syntaxa are included in 9 vegetation classes. Plant landscape of the area was reconstructed by the study of chain contacts among different communities, placed in space according to humidity and salinity gradients, determined by substratum texture and micromorphology. On the basis of phytocoenotic diversity stressed by this research, this study area is believed to be deserving of protection aiming at conserving and managing its populations and plant communities.

We present here the results from a phytosociological and synchorological study of the deciduous oak woods of Sardinia. This vegetation analysis has allowed the individuation of three new associations: Lonicero implexae-Quercetum virgilianae, Ornithogalo pyrenaici-Quercetum ichnusae and Glechomo sardoae-Quercetum congestae. The Lonicero implexae-Quercetum virgilianae association includes the woods on calcareous substrata spread throughout Northern Sardinia, which can occasionally be found in southern areas; the first of these are rich in mesophilous species and are attributed to the subassociation cyclaminetosum repandi, while the others are referred to quercetosum virgilianae. The Ornithogalo pyrenaici-Quercetum ichnusae association, more widely distributed in Central-Northern Sardinia, includes the climatophilous deciduous woods that are found on lithological substrata of a non-carbonatic nature, and in particular, on andesites, trachytes and metarenites. The typical aspect is referred to the subassociation cytisetosum villosi, while the subassociation ilicetosum aquifolii includes the more mesophilous woods on basaltic substrata. The Glechomo sardoae- Quercetum congestae association always includes the woods found on the non-carbonatic substrata in the higher altitude areas with an oceanic pluviseasonal temperate bioclimate. Of this, the subassociation quercetosum congestae, on the granite areas, and oenanthetosum pimpinelloidis, on those metamorphic and of basalt, are proposed. At the higher hierarchical levels, the association Lonicero implexae-Quercetum virgilianae is referred to the endemic Sardinian-Corsican suballiance Clematido cirrhosae-Quercenion ilicis, of the alliance Fraxino orni-Quercion ilicis, while for the other associations the new suballiance Paeonio morisii-Quercenion ichnusae, (holotypus: Glechoma sardoae-Quercetum congestae ass. nova) particular to the Sardinian biogeographic subprovince, is proposed, of the alliance Pino calabricae-Quercion congestae, the order Quercetalia pubescentipetraeae, and the class Querco roboris-Fagetea sylvatica.

A syntaxonomic revision of the holm-oak and cork-oak woods of Sardinia is here presented. The geological and phytoclimatic heterogeneity of the island and the wide ecological amplitude of the holm-oak, result in a large phytocoenotic diversity of the Sardinian holm-oak woods. These are referred to five associations: Pyro amygdaliformis-Quercetum ilicis, Prasio majoris-Quercetum ilicis ass. nova, Galio scabri-Quercetum ilicis, Saniculo europaeae-Quercetum ilicis ass. nova and Aceri monspessulani-Quercetum ilicis. From the phytogeographic point of view, in the subhumid-humid, meso-supramediterranean bioclimatic belts of Sardinia the presence of the association Galio scabri-Quercetum ilicis is found to be remarkable. As far as the cork-oak woods are concerned, the associations Galio scabri-Quercetum suberis and Violo dehnhardtii-Quercetum suberis ass. nova are here reported. The Sardinian-Corsican holm-oak and cork-oak woods define the western limit of the European central-Mediterranean alliance Fraxino orni-Quercion ilicis of which in Sardinia there are many characteristic and differential entities. In the Sardinian-Corsican holm-oak woods, however, there are endemic entities (Arum pictum ssp. pictum, Helleborus lividus ssp. corsicus, Digitalis purpurea var. gyspergerae, Quercus ichnusae and Paeonia morisii) and other western-centred entities with little or no spread on the Italian peninsula (Galium scabrum, Clematis cirrhosa and Teucrium scorodonia). These entities allow the proposal that within the alliance Fraxino orni-Quercion ilicis can be recognised the new Sardinian-Corsican suballiance Clematido cirrhosae-Quercenion ilicis (typus: Prasio majoris-Quercetum ilicis ass. nova), which is endemic to the Sardinian and Corsican sectors of the Italo-Tyrrhenian Province. Consequently, the suballiance Fraxino orni-Quercenion ilicis, which is the typical of the alliance Fraxino orni-Quercion ilicis, has also been proposed.

Mediterranean agro-silvo-pastoral systems play a key role in view of the positive contribution that they could offer to a sustainable development of European agriculture. The knowledge of the vegetation dynamics and of the processes and land uses favoring different vegetation types related to the same actual potential natural vegetation (PNV) could represent a sound reference framework for monitoring and managing plant biodiversity in these systems. The aim of the research was to evaluate plant diversity along a gradient of use intensity comparing the actual vegetation versus the PNV. The results of our research showed that in the studied Mediterranean agro-silvo-pastoral system, included in the same environmental unit, human activities enhanced plant biodiversity. Moreover, the case study presented here confirmed the effectiveness of those landscape approaches comparing actual vegetation versus the PNV for plant biodiversity monitoring and reinforced previous studies showing the effect of human activities on plant community diversity at the environmental unit scale in different biogeographical contexts.
