The concentration and composition of phytoplankton populations are measured by an optical method which can be used either in situ or remotely. This method is based upon the in vivo light absorption characteristics of phytoplankton. To provide a data base for testing assumptions relative to the proposed method, visible absorbance spectra of pure cultures of 20 marine phytoplankton were obtained under laboratory conditions. Descriptive and analytical statistics were computed for the absorbance spectra and were used to make comparisons between members of major taxonomic groups and between groups. Spectral variation between the members of the major taxonomic groups was observed to be considerably less than the spectral variation between these groups. In several cases the differences between the mean absorbance spectra of major taxonomic groups are significant enough to be detected with passive remote sensing techniques.

The theoretical concepts underlying remote sensing of estuarine parameters using laser excitation are examined. The concepts are extended to include Mie scattering as a measure of the total suspended solids and to develop the water Raman signal as an internal standard. Experimental validation of the theory was performed using backscattered laser light from a laboratory tank to simulate a remote-sensing geometry. Artificially prepared sediments and biological cultures were employed to check specific aspects of the theory under controlled conditions. Natural samples gathered from a variety of water types were also analyzed in the tank to further enhance the simulation. The results indicate that it should be possible to remotely quantify total suspended solids, dissolved organics, attenuation coefficient, chlorophyll a, and phycoerythrin in estuarine water using laser excitation.

The calculation of indices of phytoplankton population composition from chlorophyll a fluorescence at 685 nm excited by narrow band light at 454 and 539 nm is discussed. The ratio of the fluorescence excited by light of these two wavelengths is a function of the distribution of the phytoplankton between two color groups, designated the golden-brown and the green. The golden-brown group consists of those species which have the highly photosynthetically active carotenoid-chlorophyll-a-protein complexes, i.e. members of the classes Bacillariophyceae, diatoms Dinophyceae, dinoflagellates, and some members of the class Prymnesiophyceae. The green color group consists those species of phytoplankton which apparently lack those complexes, i.e. members of the classes Chlorophyceae, Euglenophyceae, Prasinophyceae, Eustigmatophyceae, Xanthophyceae, and a few members of the Prymnesiophyceae. A few species of phytoplankton appear to have intermediate characteristics, and would apparently belong to neither group. Most of these species are members of the class Cryptophyceae. The composition index for this class is examined in detail.

Phytoplankton were grown in the laboratory for the purpose of testing a remote fluorosensor. The fluorosensor uses a unique four-wavelength dye laser system to excite phytoplankton bearing chlorophyll and to measure the chlorophyll fluorescence generated by this excitation. Six different species were tested, one at a time, and each was grown two to four times. Fluorescence measured by the fluorosensor provides good quantitative measurement of chlorophyll concentrations for all species tested while the cultures were in log phase growth. Fluorescene cross section ratios obtained in the single species tank tests support the hypothesis that the shape of the fluorescence cross section curve remains constant with the species (differences in fluorescence cross section ratios are a basis for determining composition of phytoplankton according to color group when a multiwavelength source of excitation is used. Linear relationships exist between extracted chlorophyll concentration and fluorescence measured by the remote fluorosensor during the log phase growth of phytoplankton cultures tested.

In an effort to improve understanding of the effects of variations in background water on reflectance spectra, laboratory tests were conducted with various concentrations of sewage sludge diluted with several types of background water. The results from these tests indicate that reflectance spectra for sewage-sludge mixtures are dependent upon the reflectance of the background water. Both the ratio of sewage-sludge reflectance to background-water reflectance and the ratio of the difference in reflectance to background-water reflectance show spectral variations for different turbid background waters. The difference in reflectance is the only parameter considered.

An aircraft-borne remote system is presented that utilizes narrow-band light from multiple dye lasers to excite selected algae photopigments and then measures the resultant flourescence emitted from chlorophyll a at 685 nm. Tests were conducted with both pure and mixed cultures of marine algae from a series of field tests taken from piers and bridges of Narragansett Bay, and a prototype remote fluorosensor was flown over the Bay during the 1978 winter-spring diatom bloom. Remote fluorescence obtained at hover points over sea-truth stations showed correlations with in situ fluorescence, total chlorophyll a, and cell count. It was concluded that the ratio of remote fluorescence to direct chlorophyll a concentration was less variable than expected, and the distribution of total chlorophyll a between two major photoplankton color groups showed three distinct areas, within the Bay, of green and golden-brown species.

A lidar system is developed that uses four selected excitation wavelengths to induce chlorophyll 'a' fluorescence which is indicative of both the concentration and diversity of phytoplankton. The operating principles of the system and the results of measurements of phytoplankton fluorescence in a controlled laboratory environment are presented. A comparative study of results from lidar fluorosensor laboratory tank tests using representative species of phytoplankton in single and multispecies cultures from each of four color groups reveals that (1) there is good correlation between the fluorescence of chlorophyll 'a' remotely simulated and detected by the lidar system and in-situ measurements using four similar excitation wavelengths in a flow-through fluorometer; (2) good correlation exists between the total chlorophyll 'a' calculated from lidar-fluorosensor data and measurements obtained by the Strickland-Parsons method; and (3) the lidar fluorosensor can provide an index of population diversity.

Development and characteristics of vacuum probe surface sampler for obtaining particulate contamination on surfaces

A description is presented of a unique system for the sterilization and sterile repair of spacecraft and the results of a test program designed to assess the biological integrity and engineering reliability of the system. This trailer-mounted system, designated the model assembly sterilizer for testing (MAST), is capable of the dry-heat sterilization of spacecraft and/or components less than 2.3 meters in diameter at temperatures up to 433 K and the steam sterilization of components less than 0.724 meter in diameter. Sterile access to spacecraft is provided by two tunnel suits, called the bioisolator suit systems (BISS), which are contiguous with the walls of the sterilization chambers. The test program was designed primarily to verify the biological and engineering reliability of the MAST system by processing simulated space hardware. Each test cycle simulated the initial sterilization of a spacecraft, sterile repair of a failed component, removal of the spacecraft from the MAST for mating with the bus, and a sterile recycle repair.
