Solar energy is widely regarded as an environmentally friendly and sustainable source of power. It reduces greenhouse gas emissions and dependency on fossil fuels, contributing to a cleaner environment. It also provides cost savings and enhances energy security. However, technical challenges persist. Poor installation, inadequate maintenance, and aging can degrade photovoltaic (PV) systems, leading to failures or faults. These issues increase the risk of power losses, electrical shocks, and fires. Direct Current (DC) arcs, in particular, pose a significant fire hazard in PV systems due to their unpredictability and high potential for damage. However, accurately defining parameters for real-world DC arc faults is difficult. Developing computational models of electric arcs is essential for simulating, analyzing, and detecting these faults. In that sense, this work provides a comprehensive overview of the prominent black-box arc models documented in the scientific literature, along with various methods for parameter identification, to facilitate the investigation of arc-related incidents within PV systems.

The present study aimed at investigating whether the hydroxychloroquine (HCQ) treatment would impact the neutralizing antibody production, viremia levels and the kinetics of serum soluble mediators upon planned 17DD-Yellow Fever (YF) primovaccination (Bio-Manguinhos-FIOCRUZ) of primary Sjögren’s syndrome (pSS). A total of 34 pSS patients and 23 healthy controls (HC) were enrolled. The pSS group was further categorized according to the use of HCQ (HCQ and Non-HCQ). The YF-plaque reduction neutralization test (PRNT ≥1:50), YF viremia (RNAnemia) and serum biomarkers analyses were performed at baseline and subsequent time-points (Day0/Day3–4/Day5–6/Day7/Day14-D28). The pSS group showed PRNT titers and seropositivity rates similar to those observed for HC (GeoMean = 238 vs 440, p = .11; 82% vs 96%, p = .13). However, the HCQ subgroup exhibited lower seroconversion rates as compared to HC (GeoMean = 161 vs 440, p = .04; 69% vs 96%, p = .02) and Non-HQC (GeoMean = 161 vs 337, p = .582; 69% vs 94%, p = .049). No differences in YF viremia were observed amongst subgroups. Serum biomarkers analyses demonstrated that HCQ subgroup exhibited increased levels of CCL2, CXL10, IL-6, IFN-γ, IL1-Ra, IL-9, IL-10, and IL-2 at baseline and displayed a consistent increase of several biomarkers along the kinetics timeline up to D14–28. These results indicated that HCQ subgroup exhibited a deficiency in assembling YF-specific immune response elicited by 17DD-YF primovaccination as compared to Non-HCQ subgroup. Our findings suggested that hydroxychloroquine is associated with a decrease in the humoral immune response after 17DD-YF primovaccination.

Abstract The re-emergence of yellow fever (YF) urged new mass vaccination campaigns and, in 2017, the World Health Organization approved the use of the fractional dose (FD) of the YF vaccine due to stock shortage. In an observational cross-sectional investigation, we have assessed viremia, antibodies, soluble mediators and effector and memory T and B-cells induced by primary vaccination of volunteers with FD and standard dose (SD). Similar viremia and levels of antibodies and soluble markers were induced early after immunization. However, a faster decrease in the latter was observed after SD. The FD led to a sustained expansion of helper T-cells and an increased expression of activation markers on T-cells early after vaccination. Although with different kinetics, expansion of plasma cells was induced upon SD and FD immunization. Integrative analysis reveals that FD induces a more complex network involving follicular helper T cells and B-cells than SD. Our findings substantiate that FD can replace SD inducing robust correlates of protective immune response against YF.

Succession following the abandonment of traditional management practices can pose severe consequences for the conservation of semi-natural dry grassland communities. In the present study, we investigated whether the quantity of floral resources changes during succession of semi-natural dry grasslands and how this is related to pollinator richness and the number of pollination interactions at the community level. We addressed this issue by quantifying floral resources (i.e., number of flowers, nectar volume and number of pollen grains) and monitoring pollination interactions in dry grassland communities at different stages of succession, defined as the total cover of plant species of forest edges. The relationship between the quantity of floral resources and cover of plant species of forest edges was significantly hump-shaped, i.e., regardless of the type of floral resource, all peaked at intermediate values of cover of plant species of forest edges. The richness of animal-pollinated plants in bloom also showed a hump-shaped relationship with the cover of plant species of forest edges, while the richness of pollinator species and the number of pollination contacts were indirectly related to the cover of plant species of forest edges, as they were significantly associated with the number of flowers and the richness of animal-pollinated plants in bloom. Results suggest that succession of dry grasslands after abandonment may affect a crucial function in terrestrial ecosystems, namely animal-mediated pollination. Nevertheless, the conditions of early succession, which could be achieved by the presence of scattered shrubs, could ultimately be favourable for the pollination function in dry grasslands.

BACKGROUND: The generation of induced pluripotent stem cells (iPSC) has been a game-changer in translational and regenerative medicine; however, their large-scale applicability is still hampered by the scarcity of accessible, safe, and reproducible protocols. The porcine model is a large biomedical model that enables translational applications, including gene editing, long term in vivo and offspring analysis; therefore, suitable for both medicine and animal production. AIM: To reprogramme in vitro into pluripotency, and herein urine-derived cells (UDCs) were isolated from porcine urine. METHODS: The UDCs were reprogrammed in vitro using human or murine octamer-binding transcription factor 4 (OCT4), SRY-box2 (SOX2), Kruppel-like factor 4 (KLF4), and C-MYC, and cultured with basic fibroblast growth factor (bFGF) supplementation. To characterize the putative porcine iPSCs three clonal lineages were submitted to immunocytochemistry for alkaline phosphatase (AP), OCT4, SOX2, NANOG, TRA1 81 and SSEA 1 detection. Endogenous transcripts related to the pluripotency (OCT4, SOX2 and NANOG) were analyzed via reverse transcription quantitative real-time polymerase chain reaction in different time points during the culture, and all three lineages formed embryoid bodies (EBs) when cultured in suspension without bFGF supplementation. RESULTS: The UDCs were isolated from swine urine samples and when at passage 2 submitted to in vitro reprogramming. Colonies of putative iPSCs were obtained only from UDCs transduced with the murine factors (mOSKM), but not from human factors (hOSKM). Three clonal lineages were isolated and further cultured for at least 28 passages, all the lineages were positive for AP detection, the OCT4, SOX2, NANOG markers, albeit the immunocytochemical analysis also revealed heterogeneous phenotypic profiles among lineages and passages for NANOG and SSEA1, similar results were observed in the abundance of the endogenous transcripts related to pluripotent state. All the clonal lineages when cultured in suspension without bFGF were able to form EBs expressing ectoderm and mesoderm layers transcripts. CONCLUSION: For the first time UDCs were isolated in the swine model and reprogrammed into a pluripotent-like state, enabling new numerous applications in both human or veterinary regenerative medicine.

Several opportunities for embryo development, stem cell maintenance, cell fate, and differentiation have emerged using induced pluripotent stem cells (iPSCs). However, the difficulty in comparing bovine iPSCs (biPSCs) with embryonic stem cells (ESCs) was a challenge for many years. Here, we reprogrammed fetal fibroblasts by transient expression of the four transcription factors (Oct4, Sox2, Klf4, and c-Myc, collectively termed "OSKM" factors) and cultured in iPSC medium, supplemented with bFGF, bFGF2i, leukemia inhibitory factor (LIF), or LIF2i, and then compared these biPSC lines with bESC to evaluate the pluripotent state. biPSC lines were generated in all experimental groups. Particularly, reprogrammed cells treated with bFGF were more efficient in promoting the acquisition of pluripotency. However, LIF2i treatment did not promote continuous self-renewal. biPSCs (line 2) labeled with GFP were injected into early embryos (day 4.5) to assess the potential to contribute to chimeric blastocysts. The biPSC lines show a pluripotency state and are differentiated into three embryonic layers. Moreover, biPSCs and bESCs labeled with GFP were able to contribute to chimeric blastocysts. Additionally, biPSCs have shown promising potential for contributing to chimeric blastocysts and for future studies.

Abstract Humans constantly shed deoxyribonucleic acid (DNA) into the surrounding environment. This DNA may either remain suspended in the air or it settles onto surfaces as indoor dust. In this study, we explored the potential use of human DNA recovered from air and dust to investigate crimes where there are no visible traces available—for example, from a recently vacated drugs factory where multiple workers had been present. Samples were collected from three indoor locations (offices, meeting rooms and laboratories) characterized by different occupancy types and cleaning regimes. The resultant DNA profiles were compared with the reference profiles of 55 occupants of the premises. Our findings showed that indoor dust samples are rich sources of DNA and provide an historical record of occupants within the specific locality of collection. Detectable levels of DNA were also observed in air and dust samples from ultra-clean forensic laboratories which can potentially contaminate casework samples. We provide a Bayesian statistical model to estimate the minimum number of dust samples needed to detect all inhabitants of a location. The results of this study suggest that air and dust could become novel sources of DNA evidence to identify current and past occupants of a crime scene.

The growing phenomenon of population aging is redefining demographic dynamics, intensifying age-related conditions, especially dementia, projected to triple by 2050 with an enormous global economic burden. This study investigates visual arts-mediated Cognitive Activation Therapy (CAT) as a non-pharmacological CAT intervention targets both biological aging [leukocyte telomere length (LTL), DNA methylation age (DNAmAge)] and cognitive functionality. Aligning with a broader trend of integrating non-pharmacological approaches into dementia care. The longitudinal study involved 20 patients with mild to moderate neurocognitive disorders. Cognitive and functional assessments, and biological aging markers -i.e., LTL and DNAmAge- were analyzed before and after CAT intervention. Change in LTL was positively correlated with days of treatment (p =0.0518). LTL significantly elongated after intervention (p =0.0269), especially in men (p =0.0142), correlating with younger age (p =0.0357), and higher education (p =0.0008). DNAmAge remained instead stable post-treatment. Cognitive and functional improvements were observed for Copy of complex geometric figure, Progressive Silhouettes, Position Discrimination, Communication Activities of Daily Living—Second edition, Direct Functional Status (p < 0.0001) and Object decision (p =0.0594), but no correlations were found between LTL and cognitive gains. Visual arts-mediated CAT effectively mitigates cellular aging, especially in men, by elongating LTL. These findings underscore the potential of non-pharmacological interventions in enhancing cognitive and functional status and general well-being in dementia care. Further research with larger and longer-term studies is essential for validation.

Campanhas publicitárias são meticulosamente elaboradas para alcançar o maior número possível de pessoas, utilizando uma variedade de estratégias adaptáveis para conquistar públicos-alvo. À medida que surgem questões sociais somadas a esse contexto, há uma crescente intenção de representar uma ampla gama de diversidade. Esta pesquisa visa analisar a recepção do público em relação a uma campanha publicitária pró-LGBT lançada pelo Burger King, veiculada no YouTube, compreendendo alguns aspectos abordados nas reações à campanha. Para isso, filiamo-nos à Análise Crítica do Discurso (Fairclough, 2003), que concebe discurso como uma parte essencial da prática social, influenciadora e mantenedora de estruturas de poder. Nosso corpus consistiu em 20 comentários retirados da seção de discussão da campanha publicitária no YouTube. Utilizamos o sistema de Avaliatividade (Martin; White, 2005) como ferramenta analítica da semântica do discurso, mais especificamente o subsistema de Engajamento, para verificar como esses 20 comentaristas se posicionaram e interagiram com outras vozes em seus textos. Os comentários foram classificados conforme as subcategorias de Engajamento e, discutidos a partir de uma perspectiva linguístico-social. Os resultados revelaram que a maioria dos discursos é marcada por comentários LGBTfóbicos, que contestam a legitimidade da campanha e apresentam argumentos para negar a importância de abordar o tema na mídia. Esses discursos utilizam estratégias que dificultam o diálogo e excluem a comunidade LGBT+ dos espaços de consumo.

Independent vector analysis (IVA) can be viewed as an extension of independent component analysis (ICA) to multiple datasets. It exploits the statistical dependency between different datasets through mutual information. In the context of motor imagery classification based on electroencephalogram (EEG) signals for the brain–computer interface (BCI), several methods have been proposed to extract features efficiently, mainly based on common spatial patterns, filter banks, and deep learning. However, most methods use only one dataset at a time, which may not be sufficient for dealing with a multi-source retrieving problem in certain scenarios. From this perspective, this paper proposes an original approach for feature extraction through multiple datasets based on IVA to improve the classification of EEG-based motor imagery movements. The IVA components were used as features to classify imagined movements using consolidated classifiers (support vector machines and K-nearest neighbors) and deep classifiers (EEGNet and EEGInception). The results show an interesting performance concerning the clustering of MI-based BCI patients, and the proposed method reached an average accuracy of 86.7%.
