Abstract Pygmy jerboas are one of the smallest taxa of rodents. They exhibit distinctly different morphological and biological characteristics from other subfamilies, such as more restricted distribution, species richness, reproductive ability, and population size. Agricultural expansion and the development of new energy projects in recent years lead to sharp decline of their natural populations. Here, we assembled and annotated the first reference genome for the subfamily Cardiocraniinae using Illunima and Nanopore sequencing from the thick-tailed pygmy jerboa, Salpingotus crassicauda. The final genome is 2.44 Gb in size, with a contig N50 length of 13.71 Mb and a BUSCO completeness of 96.35%. A total of 23,344 protein-coding genes were annotated in the final genome. We also determined the mitochondrial genome of this species and annotated 13 protein-coding genes, 22 tRNAs, and 2 rRNA. These genomic assemblies provide resources in studying phylogeny and adaptive evolution of Dipodidae, as well as implementing conservation management of jerboas.

Abstract Background Ferroptosis is a type of regulated necrosis and glutathione peroxidase 4 (GPX4) has been recognized as a key enzyme that protects against ferroptosis. However, the significance of GPX4 in polymorphonuclear neutrophils (PMNs) of systemic lupus erythematosus (SLE) has not been explored. So we examined GPX4 mRNA in PMNs and analyzed its association with serological and clinical features. Methods A single center research from the Department of Rheumatology and Immunology of the Third Affiliated Hospital, Southern Medical University was conducted between December 2020 and September 2022. Real‐time transcription‐polymerase chain reaction analysis was used to determine the expression of GPX4 mRNA in PMNs from patients suffering from different rheumatoid immune diseases and healthy controls. The associations of GPX4 RNA levels in SLE patients with serological and clinical indicators were assessed by Spearman's correlation analysis. Receiver operating characteristic curve analysis was performed to evaluate the diagnostic value of GXP4 mRNA for SLE. Results GPX4 mRNA levels were lower in SLE patients than those in healthy individuals, patients with rheumatoid arthritis and axial spondyloarthritis. GPX4 mRNA levels were negatively correlated with the number of positive autoantibodies (r = −0.3072, p = 0.0300), anti‐dsDNA antibody (r = −0.3654, p = 0.0336), antinucleosome antibody (r = −0.4052, p = 0.0263), erythrocyte sedimentation rate (r = −0.3773, p = 0.0069), C‐reactive protein (r = −0.4037, p = 0.0036), and SLE disease activity index (r = −0.3072, p = 0.0300). Abstract The two-spotted spider mite, Tetranychus urticae Koch (Acari: Tetranychidae), is a notorious pest in agriculture that has developed resistance to almost all chemical types used for its control. Here, we assembled a chromosome-level genome for the TSSM using Illumina, Nanopore, and Hi-C sequencing technologies. The assembled contigs had a total length of 103.94 Mb with an N50 of 3.46 Mb, with 87.7 Mb of 34 contigs anchored to three chromosomes. The chromosome-level genome assembly had a BUSCO completeness of 94.8%. We identified 15,604 protein-coding genes, with 11,435 genes that could be functionally annotated. The high-quality genome provides invaluable resources for the genetic and evolutionary study of TSSM.

Abstract The plum fruit moth Grapholita funebrana (Tortricidae, Lepidoptera) is an important pest of many wild and cultivated stone fruits and other plants in the family Rosaceae. Here, we assembled its nuclear and mitochondrial genomes using Illumina, Nanopore, and Hi-C sequencing technologies. The nuclear genome size is 570.9 Mb, with a repeat rate of 51.28%, and a BUCSO completeness of 97.7%. The karyotype for males is 2n = 56. We identified 17,979 protein-coding genes, 5,643 tRNAs, and 94 rRNAs. We also determined the mitochondrial genome of this species and annotated 13 protein-coding genes, 22 tRNAs, and 2 rRNA. These genomes provide resources to understand the genetics, ecology, and genome evolution of the tortricid moths.

Abstract Energy information is vulnerable to malicious denial of service (DoS) attacks due to the diversity and openness of the smart grid environment. In order to cope with the above challenges, this paper first proposes to adopt Markov hopping model to describe the random packet loss of measurement due to DoS attacks. Then, based on Holt two‐parameter exponential smoothing and untraced Kalman filtering techniques, a one‐step predictive value compensation method for measurement data loss is proposed, and an improved dynamic untraced particle filtering algorithm based on data fusion compensation strategy is designed. Finally, an IEEE‐30 bus system is used to simulate the proposed dynamic state estimation method, which proves that the proposed method can effectively resist DoS attack.

Diabetes, a prevalent chronic condition, significantly increases the risk of mortality from COVID-19, yet the underlying mechanisms remain elusive. Emerging evidence implicates Cathepsin L (CTSL) in diabetic complications, including nephropathy and retinopathy. Our previous research identified CTSL as a pivotal protease promoting SARS-CoV-2 infection. Here, we demonstrate elevated blood CTSL levels in individuals with diabetes, facilitating SARS-CoV-2 infection. Chronic hyperglycemia correlates positively with CTSL concentration and activity in diabetic patients, while acute hyperglycemia augments CTSL activity in healthy individuals. In vitro studies reveal high glucose, but not insulin, promotes SARS-CoV-2 infection in wild-type cells, with CTSL knockout cells displaying reduced susceptibility. Utilizing lung tissue samples from diabetic and non-diabetic patients, alongside Leprdb/dbmice and Leprdb/+mice, we illustrate increased CTSL activity in both humans and mice under diabetic conditions. Mechanistically, high glucose levels promote CTSL maturation and translocation from the endoplasmic reticulum (ER) to the lysosome via the ER-Golgi-lysosome axis. Our findings underscore the pivotal role of hyperglycemia-induced CTSL maturation in diabetic comorbidities and complications.

Abstract Tortricidae is one of the largest families in Lepidoptera, including subfamilies of Tortricinae, Olethreutinae, and Chlidanotinae. Here, we assembled the gap-free genome for the subfamily Chlidanotinae using Illumina, Nanopore, and Hi-C sequencing from Polylopha cassiicola, a pest of camphor trees in southern China. The nuclear genome is 302.03 Mb in size, with 36.82% of repeats and 98.4% of BUCSO completeness. The karyotype is 2n = 44 for males. We identified 15412 protein-coding genes, 1052 tRNAs, and 67 rRNAs. We also determined the mitochondrial genome of this species and annotated 13 protein-coding genes, 22 tRNAs, and one rRNA. These high-quality genomes provide valuable information for studying phylogeny, karyotypic evolution, and adaptive evolution of tortricid moths.

Immunotherapy shows substantial advancement in cancer and is becoming widely used in clinical practice. A variety of biomarkers have been proposed to predict the efficacy of immunotherapy, but most of them have low predictive ability. Tertiary lymphoid structures (TLSs), the aggregation of multiple lymphocytes, have been found to exist in various tumor tissues. TLSs have been shown to correlate with patient prognosis and immunotherapy response. This review summarizes the characteristics of TLSs and the inducing factors of TLS formation, presents available evidence on the role of TLSs in predicting immunotherapy response in different cancers, and lastly emphasizes their predictive potential for neoadjuvant immunotherapy efficacy.

Abstract Autoimmune congenital heart block (ACHB) is a passively acquired immune-mediated disease characterized by the presence of maternal antibodies against components of the Ro/SSA and La/SSB ribonucleoprotein complex that mainly affects the cardiac conducting system. ACHB occurs in 2% of women with positive anti-Ro/SSA and anti-La/SSB antibodies and causes a high risk of intrauterine fetal death, neonatal mortality, and long-term sequelae. In this review, we first describe a case of ACHB to provide preliminary knowledge. Then, we discuss the possible pathogenic mechanisms of ACHB; summarize the pregnancy management of patients with positive anti-Ro/SSA and anti-La/SSB antibodies and/or rheumatic diseases, the prevention of ACHB, and the treatment of ACHB fetuses; and propose routine screening of these antibodies for the general population. Careful follow-up, which consists of monitoring the fetal heart rate, is feasible and reassuring for pregnant women with positive anti-Ro/SSA and/or anti-La/SSB antibodies to lower the risk of ACHB in fetuses. Moreover, maternal administration of hydroxychloroquine may be useful in preventing ACHB in pregnant women with anti-Ro/SSA and/or anti-La/SSB antibodies.

BackgroundCancer diagnosis based on machine learning has become a popular application direction. Support vector machine (SVM), as a classical machine learning algorithm, has been widely used in cancer diagnosis because of its advantages in high-dimensional and small sample data. However, due to the high-dimensional feature space and high feature redundancy of gene expression data, SVM faces the problem of poor classification effect when dealing with such data.MethodsBased on this, this paper proposes a hybrid feature selection algorithm combining information gain and grouping particle swarm optimization (IG-GPSO). The algorithm firstly calculates the information gain values of the features and ranks them in descending order according to the value. Then, ranked features are grouped according to the information index, so that the features in the group are close, and the features outside the group are sparse. Finally, grouped features are searched using grouping PSO and evaluated according to in-group and out-group.ResultsExperimental results show that the average accuracy (ACC) of the SVM on the feature subset selected by the IG-GPSO is 98.50%, which is significantly better than the traditional feature selection algorithm. Compared with KNN, the classification effect of the feature subset selected by the IG-GPSO is still optimal. In addition, the results of multiple comparison tests show that the feature selection effect of the IG-GPSO is significantly better than that of traditional feature selection algorithms.ConclusionThe feature subset selected by IG-GPSO not only has the best classification effect, but also has the least feature scale (FS). More importantly, the IG-GPSO significantly improves the ACC of SVM in cancer diagnostic.

(Aim) Gene expression data is typically high dimensional with a limited number of samples and contain many features that are unrelated to the disease of interest. Existing unsupervised feature selection algorithms primarily focus on the significance of features in maintaining the data structure while not taking into account the redundancy among features. Determining the appropriate number of significant features is another challenge. (Method) In this paper, we propose a clustering-guided unsupervised feature selection (CGUFS) algorithm for gene expression data that addresses these problems. Our proposed algorithm introduces three improvements over existing algorithms. For the problem that existing clustering algorithms require artificially specifying the number of clusters, we propose an adaptive k-value strategy to assign appropriate pseudo-labels to each sample by iteratively updating a change function. For the problem that existing algorithms fail to consider the redundancy among features, we propose a feature grouping strategy to group highly redundant features. For the problem that the existing algorithms cannot filter the redundant features, we propose an adaptive filtering strategy to determine the feature combinations to be retained by calculating the potentially effective features and potentially redundant features of each feature group. (Result) Experimental results show that the average accuracy (ACC) and matthews correlation coefficient (MCC) indexes of the C4.5 classifier on the optimal features selected by the CGUFS algorithm reach 74.37% and 63.84%, respectively, significantly superior to the existing algorithms. (Conclusion) Similarly, the average ACC and MCC indexes of the Adaboost classifier on the optimal features selected by the CGUFS algorithm are significantly superior to the existing algorithms. In addition, statistical experiment results show significant differences between the CGUFS algorithm and the existing algorithms.

Abstract Background Clinical diagnosis based on machine learning usually uses case samples as training samples, and uses machine learning to construct disease prediction models characterized by descriptive texts of clinical manifestations. However, the problem of sample imbalance often exists in the medical field, which leads to a decrease in classification performance of the machine learning. Methods To solve the problem of sample imbalance in medical dataset, we propose a hybrid sampling algorithm combining synthetic minority over-sampling technique (SMOTE) and edited nearest neighbor (ENN). Firstly, the SMOTE is used to over-sampling missed abortion and diabetes datasets, so that the number of samples of the two classes is balanced. Then, ENN is used to under-sampling the over-sampled dataset to delete the "noisy sample" in the majority. Finally, Random forest is used to model and predict the sampled missed abortion and diabetes datasets to achieve an accurate clinical diagnosis. Results Experimental results show that Random forest has the best classification performance on missed abortion and diabetes datasets after SMOTE-ENN sampled, and the MCC index is 95.6% and 90.0%, respectively. In addition, the results of pairwise comparison and multiple comparisons show that the SMOTE-ENN is significantly better than other sampling algorithms. Conclusion Random forest has significantly improved all indexes on the missed abortion dataset after SMOTE-ENN sampled.

Semi-continuous anaerobic digestion (AD) of food waste (FW) was conducted by solar intelligent control system to explore the impact of temperature fluctuation on microorganisms and its engineering applicability. According to analysis of the AD system at different organic loading rate (OLR), introduction of the solar intelligent control system into AD could save electric energy. Temperature fluctuation caused by climate and other factors reduced methane production by 31.79%. Before reaching the optimum OLR, the methane yield increased with the promotion of OLR and was at the optimum when OLR = 4.5 kg ⋅ m−3 ⋅ d−1. Enzyme activities changed with the change of pH. Lower pH limited the enzyme activities. At the initial stage of the reaction, Clostridium and Lactobacillus dominated, which made content of amylase and lipase reach 264.07 U/mL (solar group), 331.02 U/mL (electric group), and 276.44 U/mL (solar group), 306.75 U/mL (electric group), respectively. Temperature fluctuation brought in by the solar intelligent control system had a great effect on microbial activity. Microbial community structure was influenced by the OLR change. Methanobacterium became the main methanogenic bacterium at the middle and late stage. After analysis, it is feasible to introduce solar energy into AD.

Abstract Activation of multiple inflammasomes in monocytes/macrophages is associated with the pathogenesis of systemic lupus erythematosus (SLE). Gasdermin D (GSDMD)-mediated pyroptosis, a common consequence of multiple activated inflammasomes, is a programmed cell death with strong inflammatory responses. This suggested that targeting monocyte/macrophage pyroptosis might provide an opportunity to cure SLE. Here, we aimed to investigate the effect of disulfiram (DSF), a small molecule inhibitor of pyroptosis, and its potential therapeutic mechanism for SLE. The mRNA expression of GSDMD and IL-1β were significantly increased in peripheral blood mononuclear cells (PBMCs) from SLE patients. Importantly, we found serum from SLE patients rather than healthy controls induced GSDMD-mediated pyroptosis in THP-1 cells, as evidenced by enhanced LDH release, increased number of PI-positive cells, and high expression of full-length GSDMD and N-terminal GSDMD. Interestingly, treatment with DSF obviously inhibited pyroptosis of THP-1 cells induced by serum from SLE patients. Of note, DSF administration reduced proteinuria, serum anti-dsDNA level, and renal immune complex. It also attenuated renal damage in PIL mice. Further research found that the high level of serum IL-β and GSDMD-mediated pyroptosis of glomerular macrophages in PIL mice were rescued with DSF treatment. These data implied that GSDMD-mediated monocytes/macrophages pyroptosis played an important role in the pathogenesis of SLE and DSF might be a potential alternative therapeutic agent for SLE.

Abstract An increase in apoptosis and/or defects in the clearance of apoptotic cells resulting in massive secondary necrosis have been recognized as the main causes of systemic lupus erythematosus (SLE). Recent findings have revealed that gasdermin E (GSDME)-mediated pyroptosis is a mechanism associated with secondary necrosis. We aimed to investigate the effects of GSDME-mediated pyroptosis on disease activity in lupus mice. In vivo, high levels of GSDME expression were observed in the renal tubules of pristane-induced lupus (PIL) mice and SLE patients. In lupus mice, GSDME knockout or SP600125 administration effectively ameliorated lupus-like features by inhibiting GSDME-mediated renal tubular epithelial cell pyroptosis. In vitro, treatment with tumour necrosis factor-α (TNF-α) plus cycloheximide (CHX) or SLE sera induced HK2 cells to undergo pyroptosis in a caspase-3- and GSDME-dependent manner. Likewise, SP600125 significantly reduced GSDME expression and decreased pyroptosis in HK2 cells. GSDME-mediated pyroptosis may be associated with SLE pathogenesis, and targeting GSDME may be a potential strategy for treating SLE.

The removal of arsenic contamination from the aqueous environment is of great importance in the conservation of the Earth's water resources, and surfactants are a promising material used to modify magnetic nanoparticles to improve adsorption properties. Therefore, it is important to develop efficient and selective adsorbents for arsenic. Surface modification of Fe3O4 was carried out using anionic, cationic and zwitterionic surfactants to obtain composite Fe3O4@SDS, Fe3O4@CTAB, Fe3O4@SNC 16 and Fe3O4@NPC 16 (collectively referred to as Fe3O4@surfactants). The synthesized composite Fe3O4@surfactants magnetic nanoparticles were characterized by XRD, TEM and FTIR. The As(V) removal characteristics of the composite magnetic nanoparticles from the aqueous solution were evaluated by adsorption batch experiments which indicated the possibility of effective application of the surfactant-modified Fe3O4 magnetic nanoparticles for the removal of As(V) from aqueous solution. The adsorption equilibrium of the composites was reached in 30 min and the kinetic data followed the pseudo-second-order model. Langmuir equation could represent the adsorption isotherm data very well. Moreover, under the identical conditions, Fe3O4@CTAB showed maximum capacity of adsorption for As(V) (55.671 mg g−1), with its removal efficiency being much higher than that of the other composites. In addition, the Fe3O4@surfactants composite magnetic nanoparticles retained 93.5% of its initial arsenic removal efficiency even after re-using it five times. The mechanism of arsenic adsorption by Fe3O4@surfactants composite magnetic nanoparticles was proved to be complexation via electrostatic attraction, which was mainly innersphere in nature.

Abstract The population genetic structure of invasive species can be strongly affected by environmental and landscape barriers to dispersal. Disentangling the relative contributions of these factors to genetic divergence among invading populations is a fundamental goal of landscape genetics with important implications for invasion management. Here, we relate patterns of genetic divergence in a global invasive agricultural pest, Colorado potato beetle (CPB; Leptinotarsa decemlineata), to environmental and landscape factors along an invasion front in Northwestern China. We first used microsatellite markers and spatial‐temporal samples to assess broad patterns of genetic diversity as well as fine‐scale changes in patterns of genetic divergence. We then distinguished the relative contributions of five factors to genetic divergence among front populations: geographic distance (isolation by distance), climate dissimilarity (isolation by environment), and least‐cost distances (isolation by resistance) modeled with three factors: climate suitability, cropland cover, and road networks. Genetic diversity broadly decreased from West to East, with the exception being Eastern China. Low levels of genetic diversity and varying degrees of divergence were observed in Northwestern China, reflecting the potential effect of landscape heterogeneity. Least‐cost distance across cropland cover was most positively correlated with genetic divergence, suggesting a role of croplands in facilitating gene flow. The contribution of climate to genetic divergence was secondary, whether modeled in terms of local adaptability or connectivity of the climatic landscape, suggesting that constraints to CPB gene flow imposed by a harsh climate may be ameliorated in agricultural landscapes. No evidence was found for an obvious effect of road networks on genetic divergence and population structuring. Our study provides an example of how agricultural landscape connectivity can facilitate the spread of invasive pests, even across a broad climatic gradient. More broadly, our findings can guide decisions about future land management for mitigating further spread.

Population genetic structure is strongly affected by dispersal events, especially for migratory species. The investigation of population structure is therefore conducive to increasing our understanding of species dispersal. Spodoptera litura (Fabricius) (Lepidoptera: Noctuidae) is an important tobacco pest in China causing serious damage to multiple crops. In this study, we explore its dispersal dynamics by clarifying the fine-scale population genetics using 545 S. litura samples collected from tobacco plantations at 24 locations (mainly in Baise, Hechi, and Hezhou, Southern China). We analyzed the genetic diversity, genetic structure, and gene flow of these populations using seven microsatellite loci. Our results revealed high genetic diversity and low population genetic structure among S. litura. The genetic distance was uncorrelated with geographical distance, indicating the complete randomness of dispersal among the local populations. Our results suggest that the movement scope of contemporary S. litura might be much higher than the local-level spatial scale, which will provide a theoretical basis for pest management.

Humans and ecosystems are severely damaged by the existence of As(III/V) in the aquatic environment. Herein, an advanced Fe3O4@SFBC (Fe3O4-sunflower straw biochar) adsorbent was fabricated by co-precipitation method with sunflower straw biochar (SFBC) prepared at different calcination temperatures and different SFBC/Fe mass ratios as templates. The optimal pH for As(III/V) removal was investigated, and Fe3O4@SFBC shows removal efficiency of 86.43% and 95.94% for As(III) and As(V), respectively, at pH 6 and 4. The adsorption effect of calcining and casting the biochar-bound Fe3O4 obtained at different temperatures and different SFBC/Fe mass ratios were analyzed by batch experiments. The results show that when the SFBC biochar is calcined at 450 °C with an SFBC/Fe mass ratio of 1:5, the adsorption of As(III) and As(V) reaches the maximum, which are 121.347 and 188.753 mg/g, respectively. Fe3O4@SFBC morphology, structure, surface functional groups, magnetic moment, and internal morphology were observed by XRD, FTIR, SEM, TEM, and VSM under optimal working conditions. The material shows a small particle size in the range of 12–14 nm with better magnetic properties (54.52 emu/g), which is suitable for arsenic removal. The adsorption mechanism of As(III/V) by Fe3O4@SFBC indicates the presence of chemisorption, electrostatic, and complexation. Finally, the material was used for five consecutive cycles of adsorption–desorption experiments, and no significant decrease in removal efficiency was observed. Therefore, the new adsorbent Fe3O4@SFBC can be efficiently used for arsenic removal in the aqueous system.

Abstract Background Neutrophil extracellular trap (NET) formation has been described to be closely involved in the pathogenesis of systemic lupus erythematosus (SLE). In this study, we aimed to investigate the effect of polydatin (PD) on NET formation and its effects on disease activity in lupus-prone mouse models. Methods In vitro, neutrophils from SLE patients and healthy people stimulated with phorbol 12-myristate 13-acetate (PMA) or phosphate-buffered saline (PBS) were treated with PD, and reactive oxygen species (ROS) production and NET formation examined. In vivo, pristane-induced lupus (PIL) mice were treated with vehicle, PD, mycophenolate mofetil (MMF) or cyclophosphamide (CYC) while MRL/lpr mice were treated with vehicle or PD. Proteinuria, serum autoantibodies, ROS production, NET formation and kidney histopathology were tested. Results Consistent with previous findings, blood neutrophils from SLE patients showed increased spontaneous NET formation. Both in vivo and in vitro, PD treatment significantly inhibited ROS production and NET release by neutrophils. In MRL/lpr mouse model, PD administration reduced the proteinuria, circulating autoantibody levels, and deposition of NETs and immune complex in the kidneys. In addition, PD treatment ameliorated lupus-like features in PIL mice as MMF or CYC did. Conclusions PD treatment inhibited ROS-mediated NET formation and ameliorated lupus manifestations in both PIL mice and MRL/lpr mice. These results highlight the involvement of NETosis in SLE pathogenesis and reveal that PD might be a potential therapeutic agent for SLE or other autoimmune diseases.

Hydroxychloroquine (HCQ) is one of the most commonly prescribed immune-suppressants in treating rheumatoid arthritis (RA). Our previous research showed that HCQ suppressed RA development by inhibiting T follicular helper (Tfh) cells directly. Dendritic cells (DCs) serve as the link between innate and acquired immunity. Whether HCQ suppressed Tfh cell through DCs was not clear. In current study, we found that HCQ efficiently inhibited CD86, chemokine (C-X-C motif) receptor 4 (CXCR4) expression and interferon-α (IFN-α) secretion of healthy donor derived purified DCs stimulated by RA patient serum. To mimic RA, collagen-induced arthritis (CIA) mouse model was used and treated with HCQ daily for fifty-four days prior to sacrifice. We found HCQ inhibited DC maturation and migration to lymph nodes (LNs), manifested as down-regulated expression of CD40, CD80, CD86, MHCII (I-Aq) on LN DCs. In addition, HCQ reduced the level of chemokine receptor 7 (CCR7) and L-selectin on peripheral blood DCs and diminished percentage of LN DCs. Of note, HCQ only inhibited CpG ODN 1826-induced IL-12 secretion by bone marrow DCs (BMDCs) stimulated by various toll like receptor (TLR) agonists. Mechanistically, HCQ down-regulated the expression of TLR9 not only in healthy donor PBMC-derived DCs stimulated by RA patient serum, but also in LN DCs of CIA mice and CpG-activated BMDCs. Furthermore, arthritis scores in TLR9−/− mice were much lower than that in wild type mice with impaired maturity and migration capability of DCs. Collectively, activation of DCs contributes to the pathogenesis of RA and HCQ shows protective effects on RA by inhibition of DC activation via blocking TLR9.

Airway epithelial injury in response to allergens such as toluene diisocyanate (TDI) leads to persistent airway inflammation. Pyroptosis is recognized as a strong proinflammatory cell death process. However, the role of pyroptosis in bronchial epithelial injury and airway inflammation in TDI-induced asthma remains unknown.In this study, cytotoxic effect of TDI on 16HBE cells (a human bronchial epithelial cell line) was detected. Then a TDI-induced experimental asthma mouse model was established for in vivo study.Here we found that TDI induced pyroptosis in 16HBE cells, as evidenced by enhanced expressions of caspase-1 and elevated levels of LDH, IL-1β and HMGB1. As expected, TDI-induced inflammatory cell death was significantly blocked by a specific NLRP3 inflammasome inhibitor. Intriguingly, in asthmatic mice, the increased cleavages of caspase-1 and pyroptotic executioner gasdermin D (GSDMD) in bronchial epithelial cells were decreased by NLRP3 inflammasome inhibitor. Furthermore, inhibition of NLRP3 inflammasome attenuated airway hyper-responsiveness and airway inflammation, accompanied by lower levels of IL-1β, IgE and Th2-related cytokines.Our data suggest that bronchial epithelial pyroptosis exacerbates airway inflammation and hyper-responsiveness in TDI-induced asthma via NLRP3 inflammasome activation and GSDND cleavage. Therefore, NLRP3 inflammasome-mediated pyroptosis may be a potential treatment target for TDI-induced asthma.

Systemic lupus erythematosus (SLE) is a heterogeneous autoimmune disease characterized by excessive inflammatory and immune responses and tissue damage. Increasing evidence has demonstrated the important role of programmed cell death in SLE pathogenesis. When apoptosis encounters with defective clearance, accumulated apoptotic cells lead to secondary necrosis. Different forms of lytic cell death, including secondary necrosis after apoptosis, NETosis, necroptosis, and pyroptosis, contribute to the release of damage-associated molecular patterns (DAMPs) and autoantigens, resulting in triggering immunity and tissue damage in SLE. However, the role of autophagy in SLE pathogenesis is in dispute. This review briefly discusses different forms of programmed cell death pathways and lay particular emphasis on inflammatory cell death pathways such as NETosis, pyroptosis, and necroptosis and their roles in the inflammatory and immune responses in SLE.

Rice water weevil (RWW) is divided into two types of population, triploid parthenogenesis and diploid bisexual reproduction. In this study, we explored the meiosis of triploid parthenogenesis RWW (Shangzhuang Town, Haidian District, Beijing, China) by marking the chromosomes and microtubules of parthenogenetic RWW oocytes via immunostaining. The immunostaining results show that there is a canonical meiotic spindle formed in the triploid parthenogenetic RWW oocytes, but chromosomes segregate at only one pole, which means that there is a chromosomal unipolar division during the oogenesis of the parthenogenetic RWW. Furthermore, we cloned the conserved sequences of parthenogenetic RWW REC8 and Tws, and designed primers based on the parthenogenetic RWW sequence to detect expression patterns by quantitative PCR (Q-PCR). Q-PCR results indicate that the expression of REC8 and Tws in ovarian tissue of bisexual Drosophila melanogaster is 0.98 and 10,000.00 times parthenogenetic RWW, respectively (p < 0.01). The results show that Tws had low expression in parthenogenetic RWW ovarian tissue, and REC8 was expressed normally. Our study suggests that the chromosomal unipolar division and deletion of Tws may cause parthenogenesis in RWW.

Thrips pests are causing increasing damage to many crops, including vegetables, fruits, and ornamental plants. RNA interference (RNAi) has been recognized as a potential approach for thrips control. However, the RNAi of thrips is often restricted by the lack of efficient dsRNA delivery method. Here, we developed a nanocarrier-mediated dsRNA oral delivery system for efficient RNAi in the melon thrips, Thrips palmi. We first determined RPL17 and EF1α as the optimal reference genes among different developmental stages, tissues, adult ages, and starvation treatments. Two aminopeptidase N genes, TpAPN2 and TpAPN3, expressed at a relatively high level in the midgut, were selected as target genes for RNAi. In our delivery system, dsRNA solutions were absorbed by cucumber leaves and delivered to thrips by feeding. Naked dsRNAs exhibited no RNAi effect, whereas the nanocarrier SPc (star polycation)-loaded dsRNAs induced a 43.7% and 48.9% decrease in the expression levels of TpAPN2 and TpAPN3, respectively. Inhibited functions of TpAPN2 and TpAPN3 resulted in 36.0% and 53.9% reduction in the feeding amount and 28.5% and 35.5% decrease in egg production, respectively. Co-silencing of TpAPN2 and TpAPN3 led to a mortality of 48.3%, higher than the single-RNAi of TpAPN2 (23.3%) and TpAPN3 (30%). The newly developed dsRNA delivery system is easy to manipulate with improved RNAi efficacy, providing a powerful tool for gene functional study of thrips. [ABSTRACT FROM AUTHOR]
