Abstract Background Cuproptosis has been studied in various aspects as a new form of cell death. Aims We hope to explore the molecular patterns and genes related to cuproptosis in evaluating and predicting the prognosis of hepatocellular carcinoma (HCC), as well as the impact of tumor immune microenvironment. Methods and Results Sixteen cuproptosis related gene (CRGs) and cuproptosis related molecular and gene characteristics were comprehensively analyzed from 492 HCC samples. Cuproptosis related molecular patterns were generated by consensus clustering algorithm, including cuproptosis clusters, cuproptosis gene clusters (CGC) and cuproptosis score (CS). The characteristics of tumor microenvironment (TME) and tumor immune cells were described by the ssGSEA and ESTIMATE algorithms. Cuproptosis score was established to assess the clinical characteristics, prognostic and immunotherapy. The role and mechanism of CRG (ATP7A) in HCC, as well as its relationship with TME and immune checkpoints, have been further explored. The results of somatic mutation, copy number variations (CNV), and CRGs expression in HCC suggested the CRGs might participate in the HCC oncogenesis. The cuproptosis clusters were closely related to the clinical pathological characteristics, biological processes, and prognosis of HCC. The three CGC was revealed to be consistent with the three immune infiltration characterizations, including immune‐high, immune‐mid, and immune‐low subtypes. BackgroundThe correlations between cuproptosis and long noncoding RNAs (lncRNAs) with the tumor microenvironment (TME), immunotherapy, and some other characteristics of hepatocellular carcinoma (HCC) remain unclear.MethodsSixteen cuproptosis regulators and 356 cuproptosis-related lncRNAs (CRLnc) were identified from 374 HCC profiles in The Cancer Genome Atlas (TCGA) database. Six differentially expressed CRLnc were selected, and a prognostic risk model based on the CRLnc signature (CRLncSig) was constructed. The prognostic power of the model was verified. Moreover, a cuproptosis-related gene cluster (CRGC) was generated based on six lncRNAs and differentially expressed genes. The relationship between immune cell infiltration in the TME, immunotherapy, CRLncSig, and CRGC was demonstrated through various algorithms, Tumor Immune Dysfunction and Exclusion (TIDE), tumor mutational burden (TMB), etc. Potential drugs and sensitivity to those agents were evaluated for the risk model. LncRNA AL158166.1 was selected and verified in HCC tissues and cell lines, the impact of its knockdown and overexpression in HCC cells was examined, and the copper (Cu) concentration and the cuproptosis-related gene expression were detected.ResultsA CRLncSig prognostic risk model with good predictive ability was constructed. The low-risk group had a longer overall survival (OS), lower tumor purity, more extensive immune cell infiltration, higher immune score, enrichment in immune-activated pathways, and more positive response to immunotherapy versus the high-risk group. CRGC-B exhibited the best OS and the lowest tumor stage; the immune cell infiltration analysis was similar to the low-risk group in CRLncSig. CRGC-B belonged to the “immune-high” group of the TME. The low-risk group had a higher TIDE score and susceptibility to antitumor drugs. The lncRNA AL158166.1 had the highest hazard ratio. The levels of AL158166.1 were higher in HCC tissues versus healthy tissues. Knockdown of AL158166.1 could lead to an increase in intracellular Cu concentration, induce DLAT low expression, and inhibit the proliferation and migration of HCC cells, whereas overexpression of AL158166.1 exerted the reverse effect.ConclusionOverall, a new CRLncSig prognostic risk model and a cuproptosis-related molecular signature were constructed and evaluated. The model and signature were associated with the prognosis, immune infiltration, and immunotherapy of HCC. Inhibiting the lncRNA AL158166.1 may induce cuproptosis and showed potential for the inhibition of tumors. Evaluation of the CRLnc, CRLncSig, and CRGC may enhance our understanding of the TME, determine the effectiveness of immunotherapy, and act as a marker for the prognosis of HCC.

Abstract Background The aim of the study was to investigate the risk of human papillomavirus (HPV) genotyping particularly vaccine genotypes and multiple infections for cervical precancer and cancer, which might contribute to developing genotype-specific screening strategy and assessing potential effects of HPV vaccine. Methods The HPV genotypes were identified using the Seq HPV assay on self-collected samples. Hierarchical ranking of each genotype was performed according to positive predictive value (PPV) for cervical intraepithelial neoplasia 2/3 or worse (CIN2+/CIN3+). Multivariate logistic regression model was used to estimate the odds ratios (ORs) with 95% confidence interval (CI) of CIN2+ according to multiplicity of types and vaccine types. Results A total of 2811 HPV-positive women were analyzed. The five dominant HPV genotypes in high-grade lesions were 16/58/52/33/18. The overall ranking orders were HPV16/33/35/58/31/68/18/ 56/52/66/51/59/45/39 for CIN2+ and HPV16/33/31/58/45/66/52/18/35/56/51/68/59/39 for CIN3+. The risks of single infection versus co-infections with other types lower in the hierarchy having CIN2+ were not statistically significant for HPV16 (multiple infection vs. single infection: OR = 0.8, 95%CI = 0.6-1.1, P = 0.144) or other genotypes (P > 0.0036) after conservative Bonferroni correction. Whether HPV16 was present or not, the risks of single infection versus multiple infection with any number (2, ≥2, or ≥ 3) of types for CIN2+ were not significantly different. In addition, HPV31/33/45/52/58 covered by nonavalent vaccine added 27.5% of CIN2, 23.0% of CIN3, and 12.5% of cancer to the HPV16/18 genotyping. These genotype-groups were at significantly higher risks than genotypes not covered by nonavalent vaccine. Moreover, genotypes covered by nonavalent vaccine contributed to 85.2% of CIN2 lesions, 97.9% of CIN3 and 93.8% of cancers. Conclusions Partial extended genotyping such as HPV33/31/58 but not multiplicity of HPV infections could serve as a promising triage for HPV-positive self-samples. Moreover, incidence rates of cervical cancer and precancer were substantial attributable to HPV genotypes covered by current nonavalent vaccination.

BackgroundAs an “immune-privileged organ”, the liver has higher rates of both spontaneous tolerance and operational tolerance after being transplanted compared with other solid organs. Also, a large number of patients still need to take long-term immunosuppression regimens. Liver transplantation (LT) rejection involves varieties of pathophysiological processes and cell types, and a deeper understanding of LT immune response is urgently needed.MethodsHomogenic and allogeneic rat LT models were established, and recipient tissue was collected on postoperative day 7. The degree of LT rejection was evaluated by liver pathological changes and liver function. Differentially expressed genes (DEGs) were detected by transcriptome sequencing and confirmed by reverse transcription-polymerase chain reaction. The functional properties of DEGs were characterized by the Gene Ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG), and Reactome pathway analyses. The cells infiltrating the graft and recipient spleen and peripheral blood were evaluated by immunofluorescence and flow cytometry.ResultA total of 1,465 DEGs were screened, including 1,177 up-regulated genes and 288 down-regulated genes. GO enrichment and KEGG pathway analysis indicated that DEGs were involved in several immunobiological processes, including T cell activation, Th1, Th2 and Th17 cell differentiation, cytokine-cytokine receptor interaction and other immune processes. Reactome results showed that PD-1 signaling was enriched. Further research confirmed that mRNA expression of multiple immune cell markers increased and markers of T cell exhaustion significantly changed. Flow cytometry showed that the proportion of Treg decreased, and that of PD-1+CD4+ T cells and PD-1+CD8+ T cells increased in the allogeneic group.ConclusionUsing an omic approach, we revealed that the development of LT rejection involved multiple immune cells, activation of various immune pathways, and specific alterations of immune checkpoints, which would benefit risk assessment in the clinic and understanding of pathogenesis regarding LT tolerance. Further clinical validations are warranted for our findings.

The purpose is to explore new cultivation modes of college students’ national cognition and cultural acceptance. Deep learning (DL) technology and Educational Psychology theory are introduced, and the influence of art journal reading on college students’ national cognition and cultural acceptance is analyzed under Educational Psychology. Firstly, the background of Educational Psychology, national cognition and cultural acceptance, and learning system are discussed following a literature review. The DL technology is introduced to construct the journal reading guidance system. The system can provide users with art journals and record the user habits like reading duration and preferences. Secondly, hypotheses are proposed, and a questionnaire survey is designed, with 12 specific indicators to investigate and collect research data. Finally, the collected data are analyzed. The results show that women’s cognition of Chinese traditional culture, Chinese excellent revolutionary culture, and Chinese national identity is higher than that of men. By comparison, men’s cognition of Chinese advanced socialist culture is higher than women’s. After using the journal reading guidance system, the cognition of female college students on traditional Chinese culture is improved by 16.3%. Before and after reading art journals, the overall national cognition and cultural acceptance of Minority students are higher than that of Han students. The overall cognition of Literature and History students is higher than that of Science and Engineering students in traditional Chinese culture and China’s excellent revolutionary culture and lower in advanced Chinese socialist culture and Chinese national identity. The overall cognition of college students’ party members to the advanced socialist culture is higher than league members. As students read more art journals through the guidance system, their overall national cognition and cultural acceptance have increased. Therefore, reading art journals can promote college students’ national cognition and cultural acceptance. A national cognition and cultural acceptance promotion system that conforms to the current situation of college students is constructed. The finding provides a reference for developing complex emotion recognition technology in human-computer interaction.

Abstract Background Self-sampling for human papillomavirus (HPV) testing is a feasible option to improve the cervical screening coverage. However, an ideal triage method for HPV-positive self-samples does not yet exist. The aim of this study was to explore the utility of HPV genotyping and p16INK4a immunostaining (p16) in triaging HPV-positive self-samples, focusing on HPV-positive, cytology-negative (HPCN) women. Methods A total of 73,699 women were screened in a cervical screening project in China via SeqHPV assay on self-samples. HPV-positive women were called-back and collected cervical sample for p16 immunostaining and liquid-based cytology, those who met any result of HPV16/18+ or visual inspection with acetic acid (VIA) + or p16+ were referred for colposcopy, and HPCN women with adequate data on p16 and pathology were analyzed. A triage strategy was considered acceptable if the negative predictive value (NPV) for cervical intraepithelial neoplasia 3 or worse (CIN3+) was 98% or more, combined with an improvement of sensitivity and specificity for CIN2+/CIN3+ in reference to the comparator, being HPV16/18 + . Results A total of 2731 HPCN women aged 30–64 years were enrolled, 136 (5.0%) CIN2+ and 53 (1.9%) CIN3+ were detected. Five triage strategies met the criteria: p16+; HPV16/33+; ‘HPV16+ or HPV33/58/31/35+&p16+’; ‘HPV16/33+ or HPV58/31/35+&p16+’; HPV16/18/31/33/45/52/58 + & p16+. These strategies required less or similar colposcopy referrals, and less colposcopies to detected one case of CIN2+/CIN3+, achieving favorable false positive (negative) rates to the comparator. Among them, p16 staining detected 83.1% (79.2%) of underlying CIN2 + (CIN3+) in HPCN women. Moreover, three triage strategies were favorable in sensitivity and/or specificity to the ‘HPV16/33+’ strategy: p16+; ‘HPV16+ or HPV33/58/31/35 + &p16+’; HPV16/18/31/33/45/52/58 + &p16 + . Conclusions Genotyping for HPV16/33 could be utilized to optimize the management of HPCN women. Moreover, p16 immunostaining, either alone or combined with extended genotypes, is more effective than HPV genotypes alone in the triage of HPCN women.

Background: For cervical cancer screening, routine cytology has a high specificity but a lower sensitivity. In older women, atrophy, which may mimic HSIL, presents a diagnostic challenge. p16 is a widely used biomarker for histological diagnosis of HSIL. Our objective was to evaluate PathCIN® p16INK4a immunocytology in identification of high grade dysplasia vs. benign atrophy. Methods: As part of a multi-center screening program, 3351 women were co-tested by p16 immunocytology. Among women referred for colposcopy on basis of cytology and high-risk HPV status, those with atrophy were older than the population screened (52 vs. 43 years). Cases from older women with atrophy (n ​= ​116) and controls without atrophy (n ​= ​47) were identified by re-examination of Pap smears. The detection of CIN2+ was compared for p16, cytology and HR-HPV results. Results: The sensitivity of routine cytology (≥LSIL) was much lower for cases with atrophy (17%) than non-atrophic cases (75%). The sensitivity of p16 immunocytology and of HR-HPV testing was high (88%–100%) both with and without atrophy. The specificity of routine cytology (≥LSIL) was higher for cases with atrophy (79%) than non-atrophic cases (38%). The specificity of p16 immunocytology was high (88–95%) and the specificity of HR-HPV testing was low (31%–33%) both with and without atrophy. Combining p16 with HPV testing and/or routine cytology had no benefit, as compared to p16 staining alone. Conclusions: p16 immunocytology compares favorably with routine cytology and HPV testing in the differential diagnosis of HSIL and benign atrophy. It is more sensitive than cytology for atrophic specimens, and is more specific than HPV testing. p16 immunocytology may decrease the need for colposcopy referrals and could be a useful tool for early detection of cervical cancer in peri- and post-menopausal women, who are more likely to have HSIL coexisting with atrophy.

Methods: Included in this analysis were women in a cervical cancer screening project conducted in central China who tested positive for high-risk human papillomavirus (hrHPV) and returned back for triage with complete data of primary HPV testing, liquidbased cytology (LBC) analysis, P16 immuno-stained cytology interpretation, and pathology diagnosis. The included patients were grouped by age: ≥50 (1,127 cases) and <50 years (1,430 cases). The accuracy of LBC and P16 cytology in the detection of pathology ≥HSIL was compared between the two groups, and the role of P16 immunostain in differentiating benign cervical lesions from cytology ≥HSIL was further analyzed. Results: One hundred sixty-seven women (14.8%; 167/1,127) in the ≥50 group and 255 (17.8%, 255/1,430) in the <50 group were pathologically diagnosed as HSIL (Path-HSIL). LBC [≥Atypical Squamous Cell Of Undetermined Significance (ASCUS)] and P16 cytology (positive) respectively detected 63.9% (163/255) and 90.2% (230/255) of the Path-≥HSIL cases in the <50 group and 74.3% (124/167) and 93.4% (124/167) of the Path-≥HSIL cases in the ≥50 group. LBC matched with pathology in 105 (41.2%) of the 255 Path-≥HSIL cases in the <50 group and 93 (55.7%) of the 167 Path-≥HSIL cases in the ≥50 group. There were five in the <50 group and 14 in the ≥50 group that were Path-≤LSIL cases, which were interpreted by LBC as HSIL, but negative in P16 cytology. Conclusion: P16 cytology facilitates differentiation of Path-≤LSIL from LBC-≥HSIL for women 50 years of age and above. It can be used in the lower-resource areas, where qualified cytologists are insufficient, as the secondary screening test for women aged ≥50 to avoid unnecessary biopsies and misinterpretation of LBC primary or secondary screening [ABSTRACT FROM AUTHOR]

Background: This study aimed to evaluate three different patterns of cervical cancer screening strategies for detection of cervical diseases. Methods: In total, 10,186 women aged 21–70 years attending cervical screening program were recruited and cotested by human papillomavirus (HPV) assays and cytology. Three-year histological follow-up data was recorded on women with abnormal screening results, and six clinically common screening algorithms were evaluated. Results: Significantly better protection against cervical intraepithelial neoplasia 2 or worse (CIN2+) at three-year follow-up was associated with a negative HPV result than by normal cytology at baseline. HPV screening was more sensitive and less specific than cytology screening. Moreover, HPV screening with HPV16/18 and reflex cytology (atypical squamous cells of undetermined significance [ASCUS] threshold) showed a similar sensitivity (94.6% vs. 98.2%, p = 0.125) and a superior specificity as compared to cotesting reflex HPV16/18 and cytology (ASCUS threshold) for CIN2+ (95.8% vs. 95.1%, p < 0.0001), achieving a colposcopy referral rate of 5.4%, and consuming 4.8 colposcopies and 4.4 cytology tests to find one CIN2+. Conclusions: HPV screening with triage of HPV-positive women by HPV16/18 genotyping and cytology provided a good equilibrium between screening effectiveness, the number of cytology tests required, and referral rates; HPV testing was similar in sensitivity to cotesting and safer than cytology, thus especially suitable for large population-based screening programs.

The related literature is studied to explore the psychological characteristics of juvenile delinquency groups and implement their psychological characteristics model for the intervention of health behavior. Drawing on the results of current literature research, the Youth Psychological Characteristics Crime Prevention Questionnaire (YPPQ) was compiled, which can be simply referred to as the Crime Prevention Questionnaire. The whole psychological characteristics of juvenile delinquency are analyzed by means of a questionnaire. Firstly, the YPPQ scores of different groups were compared, and a structured interview was conducted on the reasons for the crime of the problem youth group. Secondly, data analysis was carried out on the results of questionnaires and interviews, and the psychological characteristics of juvenile delinquency were summarized. A “mixed hierarchical intervention model” was proposed to intervene in the mental health behavior of juvenile delinquency groups, and corresponding intervention strategies were also proposed. The results reveal that through the questionnaire survey, the educational background of juvenile subjects was generally distributed in middle school, the number of juveniles with primary school education was less than 30% of the juvenile delinquency groups, the middle school education accounted for more than 60% of the juvenile delinquency groups, and the approximate age was about 18 years old. The largest number in each group were adolescents with secondary school education, indicating the importance of psychological education on crime prevention for adolescents in secondary school. By comparing the YPPQ test scores of different groups, the adolescent group has higher test scores than the juvenile delinquency groups in five of the dimensions. Through the comparative analysis of the YPPQ test results of the juvenile delinquency groups, the problem youth group, and the normal youth group, it is found that the YPPQ has high reliability and validity, so its detection and evaluation are highly feasible. By comparing the odds ratio (OR) of each question in the YPPQ test between the experimental group and the control group, it is found that the psychological characteristics of the experimental group are significantly affected by family, school, and even society. Finally, it proposes a “mixed hierarchical intervention model” for juvenile delinquency to intervene in health behaviors. The purpose is to provide some research ideas for the study of the psychological characteristics of juvenile delinquency groups and to put forward some suggestions for the prevention of juvenile delinquency and the intervention of health behavior.

Unit charts are a common type of chart for visualizing scientific data. A unit chart is a chart used to communicate quantities of things by making the number of symbols on the chart proportional to the number of items represented. An accurate perception of the order of magnitude is essential to evaluating whether a unit chart can effectively convey information. Previous studies have primarily focused on perceptual properties at small order-of-magnitude scales or the efficacy of pictographs in unit charts. However, few researchers have explored the perceptual effectiveness of unit charts when representing large orders of magnitude. In this study, we performed a series of sampling measurements to investigate the visual–perceptual characteristics of unit charts when representing asymmetric interactions such as large-scale numbers. The results showed that under the restriction of the current conventional display medium, unit charts still offer a significant advantage over bar charts in a single-scale visual overview. However, this comes at the cost of a longer response time. Although this study constitutes basic research, accumulating evidence about how people reason about magnitudes beyond human perception is critical to the field of information science. This study may contribute to understanding how viewers perceive unit charts and the factors that influence graphical perception. This article provides some specific guidelines for designing unit charts that may be useful to visualization designers.

In order to optimize the resource allocation of the traditional publishing industry in the new media era, it is proposed to integrate the traditional publishing and digital publishing industries to solve the problem of unbalanced resource distribution under dual-track conditions. Professional talents with innovative entrepreneurial ability and psychology in colleges and universities are cultivated to promote the integration and reform process of the publishing industry under the background of new media art. First, the study analyzes the digital reform issues facing the development of the publishing industry in the new media era. Second, in view of the development situation of the publishing industry in the Yangtze River Delta, it is proposed to establish a development model of integrated publishing in the Yangtze River Delta through resource allocation. Then, under the new media art form, the teaching mode of creative and entrepreneurial talents training in art colleges and universities is optimized to cultivate students’ innovative ability and entrepreneurial positive psychology. The research results show that the number of books printed in Shanghai in the Yangtze River Delta is 13,000 types per year, and the number is still rising; however, periodicals and newspapers are affected by the new media industry, and the number of publications is declining. The printing volume has dropped by 50% in 9years; the questionnaire survey results show that 68% of the students are very interested in entrepreneurial activities, but 53% of the students have not carried out entrepreneurial activities at all, indicating that the students’ entrepreneurial ability is insufficient. The results provide a reference for studying the reform direction of the publishing industry and cultivating entrepreneurial talents in the context of new media.

Background/objectiveHuman papillomavirus (HPV) genotyping and cytology have been recommended for colposcopy triage, but it is unclear which combinations of high-risk HPV (hrHPV) types and cytology with various thresholds provide clinically useful information for the triage after primary HPV screening on self-collected samples.MethodChinese Multi-site Screening Trial (CHIMUST) database focused on self-collected samples was reviewed using the results of Cobas4800 HPV assay. Absolute risks of each genotype for cervical intraepithelial neoplasia 2 or worse/ 3 or worse (CIN2+/CIN3+) were calculated. Triage of atypical squamous cells of undetermined significance (ASCUS) or worse cytology was used as the comparator, and diagnostic accuracy for paired comparisons between algorithms was obtained using McNemar's test.ResultsA total of 10, 498 women were included, the overall prevalence of hrHPV, HPV16, HPV18, and Other hrHPV genotypes were 13.7%, 2.4%, 0.8%, and 10.5%, respectively. HPV16-positive women had the highest absolute risk among various genotypes for CIN2+/CIN3+ whether in normal or abnormal cytology (ASCUS or worse) and among all age groups. When compared with the comparator, combining HPV16 positivity and/or high-grade squamous intraepithelial lesion (HSIL) or worse yielded higher specificity (97.7% vs. 97.0%, pConclusionsTriage of HPV-positive women on self-collected samples by combining HPV16 or HPV16/18 genotyping with different thresholds of cytology could provide tradeoffs in sensitivity for detecting cervical lesions and colposcopy referral rates, and tailor management in various circumstances of clinical practice.
