Abstract Background Osteosarcoma (OS) is the most common bone malignant tumor in children, and its prognosis is often poor. Anoikis is a unique mode of cell death.However, the effects of Anoikis in OS remain unexplored. Method Differential analysis of Anoikis-related genes was performed based on the metastatic and non-metastatic groups. Then LASSO logistic regression and SVM-RFE algorithms were applied to screen out the characteristic genes. Later, Univariate and multivariate Cox regression was conducted to identify prognostic genes and further develop the Anoikis-based risk score. In addition, correlation analysis was performed to analyze the relationship between tumor microenvironment, drug sensitivity, and prognostic models. Results We established novel Anoikis-related subgroups and developed a prognostic model based on three Anoikis-related genes (MAPK1, MYC, and EDIL3). The survival and ROC analysis results showed that the prognostic model was reliable. Besides, the results of single-cell sequencing analysis suggested that the three prognostic genes were closely related to immune cell infiltration. Subsequently, aberrant expression of two prognostic genes was identified in osteosarcoma cells. Nilotinib can promote the apoptosis of osteosarcoma cells and down-regulate the expression of MAPK1. Conclusions We developed a novel Anoikis-related risk score model, which can assist clinicians in evaluating the prognosis of osteosarcoma patients in clinical practice. Analysis of the tumor immune microenvironment and chemotherapeutic drug sensitivity can provide necessary insights into subsequent mechanisms. MAPK1 may be a valuable therapeutic target for neoadjuvant chemotherapy in osteosarcoma.

ObjectiveDistant metastasis occurs in some patients at the first diagnosis of nasopharyngeal carcinoma (NPC), the prognosis is poor, and there are significant individual differences. This study established a nomogram model of lung metastasis of NPC as a supplement to TNM staging.MethodsThe training cohort is used to build the nomogram model, and the validation cohort is used to evaluate the model. The training cohort of 177 patients is from the Surveillance, Epidemiology, and End Results (SEER) database. Factors affecting overall survival (OS) in patients with lung metastasis of NPC analysis by Cox regression analysis and then a nomogram were established. 122 patients from the Affiliated Tumor Hospital of Guangxi Medical University were selected as the external validation cohort. The concordance index (C-index), the area under the curve (AUC), and the calibration curve were used to assess the accuracy of the nomogram and used the decision curve analysis (DCA) curve to measure the clinical benefit capacity of the model. The patients were separated into two groups with different risks, and the “Kaplan-Meier (KM)” survival analysis was used to evaluate the differentiation ability of the model.ResultsAge, T-stage, radiation, chemotherapy, and brain metastases can affect the OS in NPC with lung metastasis. A nomogram was developed according to the above five factors. The C-index of the training cohort and the validation cohort were 0.726 (95% CI: 0.692-0.760) and 0.762 (95% CI: 0.733-0.791). The AUC of the nomogram was better than that of the TNM staging. In the training cohort, the nomogram predicted OS AUC values of 0.767, 0.746, and 0.750 at 1, 2, and 3 years, TNM stage of 0.574, 0.596, and 0.640. In the validation cohort, nomogram predictions of OS AUC values of 0.817, 0.857, and 0.791 for 1, 2, and 3 years, TNM stage of 0.575, 0.612, and 0.663. DCA curves suggest that nomogram have better clinical net benefits than TNM staging. The KM survival analysis shows that the nomogram has a reasonable risk stratification ability.ConclusionThis study successfully established a nomogram model of NPC lung metastasis, which can be used as a supplement to TNM staging and provide reference for clinicians.

Sea cucumber (Apostichopus japonicus) is one of the cultured species with the highest production value to a single species in China’s marine agricultural industry with high nutritional and medicinal value. Body color influences numerous biological functions in sea cucumbers, and it has become a key trait in selective breeding. It is found that a cultivation environment with strong light intensity leads to a lighter body color of A. japonicus compared to dark conditions. However, little research has been conducted on the molecular mechanisms regulating body color in different breeding environments. In this study, we used Illumina sequencing to examine gene expression patterns in green and purple A. japonicus exposed to different culture conditions. Overall, the genes in the body wall of purple individuals were more affected by the environment. We also identified pathways that were potentially influenced by the breeding environment, such as “Drug metabolism - cytochrome P450”, “Porphyrin and chlorophyll metabolism”, “Phosphatidylinositol signaling system”, “TGF-beta signaling”, and “mTOR signaling”. The aim of this study was to determine the light conditions and breeding environment that are more favorable to the requirements of body color in A. japonicus and apply them to the selection of high-quality breeds. The results obtained will support the breeding of A. japonicus with specific traits and the production of seedlings to promote the development of this maricultural industry.

Abstract Recent studies have shown great performance of Transformer-based models in long-term time series forecasting due to their ability in capturing long-term dependencies. However, Transformers have their limitations when training on small datasets because of their lack in necessary inductive bias for time series forecasting, and do not show significant benefits in short-time step forecasting as well as that in long-time step as the continuity of sequence is not focused on. In this paper, efficient designs in Transformers are reviewed and a design of decomposing residual convolution neural networks or DRCNN is proposed. The DRCNN method allows to utilize the continuity between data by decomposing data into residual and trend terms which are processed by a designed convolution block or DR-Block. DR-Block has its strength in extracting features by following the structural design of Transformers. In addition, by imitating the multi-head in Transformers, a Multi-head Sequence method is proposed such that the network is enabled to receive longer inputs and more accurate forecasts are obtained. The state-of-the-art performance of the presented model are demonstrated on several datasets.

Abstract Sea cucumber is a morphologically diverse and ecologically important clade of echinoderms. The sea cucumber Apostichopus japonicus is the most economically valuable species of sea cucumber. The initial assembly of the A. japonicus genome was released in 2017. However, this genome assembly is fragmented and lacks relative position information of genes on chromosomes. In this study, we produced a high-quality chromosome-level genome of A. japonicus using Pacbio HiFi long-reads and Hi-C sequencing data. The assembled A. japonicus genome spanned 671.60 Mb with a contig N50 size of 17.20 Mb and scaffold N50 size of 29.65 Mb. A total of 99.9% of the assembly was anchored to 23 chromosomes. In total, 19,828 genes were annotated, and 97.2% of BUSCO genes were fully represented. This high-quality genome of A. japonicus will not only aid in the development of sustainable aquaculture practices, but also lay a foundation for a deeper understanding of their genetic makeup, evolutionary history, and ecological adaptation.

Aim To emphasize the role of non-sulfonamides in the treatment of Nocardia infection and reduce the adverse reactions caused by sulfonamides. Methods We retrospectively analyzed a case of cutaneous nocardiosis in an immunocompetent individual. The colonies obtained by staining the pus in the lesion with antacid and culturing the agar plates were identified by flight mass spectrometry. The pathogenic identification showed Nocardia brasiliensis infection and the patient was treated with amoxicillin-clavulanic acid. Results After treatment with amoxicillin and clavulanic acid, the ulcer gradually peeled and crusted, leaving dark pigmentation. The patient has finally recovered. Conclusion: Sulfonamides are the first-line antibacterial agents for years in treatment of nocardiosis but are of great toxicity and side effects. This patient was successfully treated with amoxicillin-clavulanic acid and it provided a reference protocol for patients with sulfonamide-resistant Nocardia or sulfonamides intolerance.

The combined strain paths have significant effects on the formability on the sheet metal. In this work, aiming at a widely-used AA5052-O sheet metal, the uniaxial tension and the Nakazima tests were conducted on the large-scale specimens subjected to the pre-tension deformation. First, a novel gripping fixture was designed and manufactured to achieve the uniform pre-strains of the large specimen. Then, two types of strain paths of the tension - tension and tension - bulging were implemented to reveal the evolutions of the mechanical properties and forming limit curves (FLCs), respectively. Moreover, the forming limit stress curves (FLSCs), which are more insensitive to the changes of strain paths, are calculated from the experimental FLCs to improve the prediction accuracy of finite element (FE) simulations. Compared to the conventional necking criterion of path transition and the FLC criterion, the FLSC criterion can effectively enhance the simulation precision in the biaxial tension region.

Background: Antepartum depression, with an incidence of 20.7%, is a pressing global public health concern due to its detrimental effects on both the physical and mental health of pregnant women, as well as the potential risk it poses for depression in their offspring. Nevertheless, there is a lack of consensus among existing studies regarding this issue. Here, we systematically evaluated the relationship between maternal perinatal depression and offspring depression by meta-analysis. Methods: We conducted a comprehensive search for relevant studies in Pubmed, Embase, The Cochrane Library, CNKI, Wanfang, VIP, and Chinese Biomedical Literature Service System databases. The prospective cohort studies, which were published in English or Chinese, reported the occurrence of maternal prenatal and/or postnatal depression within one year postpartum and assessed the subsequent development of depression in their offspring, were included. Study quality was assessed with the Newcastle-Ottawa Scale. Review Manager 5.4 software was used for meta-analysis. Subgroup analysis was performed. Publication bias was evaluated with a funnel plot. Results: Totally, 12 studies were included. The meta-analysis found that maternal perinatal depression increased the risk of offspring depression by 1.64 (95% confidence interval (95% CI): 1.37, 1.96, p < 0.001). Subgroup analysis showed that the risk of offspring depression was significantly increased in the European population with maternal perinatal depression (odds ratio (OR) = 1.90, 95% CI (1.49, 2.42), p < 0.001), but not in the Australian and the American populations. The combined effect sizes of maternal antepartum and postpartum depression were (OR = 1.70, 95% CI (1.27, 2.27), p < 0.001) and (OR = 1.74, 95% CI (1.31, 2.32), p < 0.001), respectively. The combined effect size of the relationship of maternal perinatal depression with offspring depression in childhood and adulthood was (OR = 1.70, 95% CI (1.28, 2.25), p < 0.001) and (OR = 1.60, 95% CI (1.27, 2.02), p < 0.001), respectively. The adjusted and unadjusted combined effect sizes were (OR = 1.44, 95% CI (1.14, 1.82), p < 0.001) and (OR = 1.97, 95% CI (1.49, 2.60), p < 0.001), respectively. There may be some publication bias in the included studies. Conclusions: Maternal perinatal depression is associated with an increased risk of depression in offspring. Effective prevention and management of depression in perinatal women is necessary to mitigate the risk of depression in offspring.

In an increasingly competitive market environment, as an important decision for strategic change, CEO succession is a key step to driving sustainable development. The formulation of succession plans and how to improve the effectiveness of internal promotion mechanisms have been researched topics in executive team management. To explore the relationship between corporate executive succession planning and corporate performance, using 3955 CEO succession samples from 2599 companies during 2001-2015 in China, the Probit model, and the two-stage least squares method, the relationship between corporate CEO succession planning and corporate performance under the director-cum-CEO succession model was analysed. The results reveal that the formulation or not of succession plans in firms, particularly in listed firms, directly affects firm performance. Director-CEO internal succession decisions reduce firm performance relative to other CEO succession models. Conclusions obtained from this study indicate the relationship between succession planning and organisational performance at the micro level and provide a theoretical reference to further promote research related to succession decisions in listed companies and the longterm development interests of firms.

Background: Gastric cancer (GC) is the fifth leading cancer in the world, and there is a high mortality rate in China. Exploring the relationship between the prognosis of GC and the expression of related genes is helpful to further understand the common characteristics of the occurrence and development of GC and provide a new method for the identification of early GC, so as to provide the best therapeutic targets. Methods: Vascular endothelial growth factor (VEGF) and markers of epithelial-mesenchymal transition (EMT) were investigated immunohistochemically using tumor samples obtained from 196 GC tissues and adjacent tumor tissues. Abstract Background The stromal antigen 3 (STAG3) gene encodes an adhesion complex subunit that can regulate sister chromatid cohesion during cell division. Chromosome instability caused by STAG3 gene mutation may potentially promote tumor progression, but the effect of STAG3 on hepatocellular carcinoma (HCC) and the related molecular mechanism are not reported in the literature. The mechanism of the occurrence and development of HCC is not adequately understood. Therefore, the biological role of STAG3 in HCC remains to be studied, and whether STAG3 might be a sensitive therapeutic target in HCC remains to be determined. Methods The expression and clinical significance of STAG3 in HCC tissues and cell lines were determined by RT–qPCR and immunohistochemistry analyses. The biological functions of STAG3 in HCC were determined through in vitro and in vivo cell function tests. The molecular mechanism of STAG3 in HCC cells was then investigated by western blot assay. Results The mRNA expression of STAG3 was lower in most HCC cells than in normal cells. Subsequently, an immunohistochemical analysis of STAG3 was performed with 126 samples, and lower STAG3 expression was associated with worse overall survival in HCC patients. Moreover, cytofunctional tests revealed that the lentivirus-mediated overexpression of STAG3 in HCC cells inhibited cell proliferation, migration, and invasion; promoted apoptosis; induced G1/S phase arrest in vitro; and inhibited tumor growth in vivo. Furthermore, studies of the molecular mechanism suggested that the overexpression of STAG3 increased Smad3 expression and decreased CDK4, CDK6, cyclin D1, CXCR4 and RhoA expression. Conclusion STAG3 exhibits anticancer effects against HCC, and these effects involve the Smad3-CDK4/CDK6-cyclin D1 and CXCR4/RhoA pathways. STAG3 is a tumor-suppressor gene that may serve as a potential target for molecular therapy, which provides a new idea for the treatment of HCC.

Embryonic development, especially metamorphosis and settlement, has a major impact on the life history of marine invertebrates. Apostichopus japonicus is an economically important species of sea cucumber. In this study, we performed RNA sequencing on six key stages of A. japonicas development: fertilized eggs, blastula, gastrula, auricularia, doliolaria, and pentactula. A total of 32,353 genes were identified and annotated as a reference gene set for subsequent pairwise comparison analysis. After filtering out low-quality genes, the dynamic molecular responses to development were revealed by WGCNA. The results showed that of the 20 modules, genes in the blue, yellow, and darkslateblue modules were highly correlated with the gastrula, auricularia, and blastula stages, respectively. GO terms for “RNA” and “proteasome complex” were most significantly enriched in the blue module. In the darkslateblue and yellow module, receptors of signaling pathways and metabolic processes were significantly enriched, respectively. All DEGs were categorized into 34 terms, mainly associated with signal transduction and cellular immunity. The expression pattern of genes associated with adhesion, cell cycle, signal, transcription factor, extracellular matrix (ECM), and cytoskeleton was analyzed according to gene function. The results of this study facilitated a more comprehensive understanding of the molecular characteristics of sea cucumber embryonic development and will provide theoretical guidance for larva rearing in sea cucumber culture.

The internet facilitates the formation of the information society while also accelerating the viral spread of negative news and negative emotions, increasing public uncertainty and depression and impeding consensus, especially in the post-pandemic period. Mindfulness intervention, which has a positive effect on attention focus, self-regulation, and subjective wellbeing, is proven to mitigate negative emotional effects, and even alter mind patterns. The study aimed to give insight into the effect of mindfulness in the new media field, concerning trait mindfulness improvement, emotional arousal and regulation, and implicit attitudes from the perspective of intra-personal communication and positive communication. The study conducted a randomized pre-test–post-test control group design, with 3 (condition groups: mindfulness vs. placebo vs. control) × 2 (test times: pre vs. post). Participants who were exposed to negative news coverage with negative emotional arousal received 14 consecutive days of intervention. The results showed that mindfulness training can improve trait mindfulness effectively on the whole, especially in facets of describing, acting awareness, and non-judgment, and mitigate the negative effect from bad information coverage, while mindfulness intervention on mind patterns and expectations on controversial issues still awaited future empirical research. The present study intended to bridge the bonding between positive psychology and new media studies by focusing on individual attention improvement and negative emotion regulation, in the expectation that trait mindfulness can be beneficial in individual infodemic syndromes such as judgment bias and information exhaustion, and avoidance.

Sterile alpha and HEAT/Armadillo motif-containing protein (SARM) is conserved in evolution and negatively regulates TRIF-dependent Toll signaling in mammals. The SARM protein from Litopenaeus vannamei and its Drosophila orthologue Ectoderm-expressed (Ect4) are also involved in immune defense against pathogen infection. However, the functional mechanism of the protective effect remains unclear. In this study, we show that Ect4 is essential for the viral load in flies after a Drosophila C virus (DCV) infection. Viral load is increased in Ect4 mutants resulting in higher mortality rates than wild-type. Overexpression of Ect4 leads to a suppression of virus replication and thus improves the survival rate of the animals. Ect4 is required for the viral induction of STAT-responsive genes, TotA and TotM. Furthermore, Ect4 interacts with Stat92E, affecting the tyrosine phosphorylation and nuclear translocation of Stat92E in S2 cells. Altogether, our study identifies the adaptor protein Ect4 of the Toll pathway contributes to resistance to viral infection and regulates JAK/STAT signaling pathway.

IntroductionMicroplastics (MPs) and cadmium (Cd) are persistent pollutants in aquatic environments. Sea cucumbers are susceptible to MPs and Cd due to their feeding behavior.MethodsThis study, based on Illumina sequencing, compared the transcriptomes of A. japonicus before and after Cd and/or MPs exposure. Additionally, we detected the changes of catalase (CAT), and superoxide dismutase (SOD) activity, glutathione (GSH), and malondialdehyde (MDA) content in sea cucumbers.Results and DiscussionHigh concentration of MPs caused the increase of SOD activity. High concentration combined treatment resulted in significant up regulation of these four indicators in A. japonicus and had the largest number of differential expression genes (DEGs) reaching 1,618 DEGs, consisting of 789 up regulated along with 829 down regulated DEGs. Transcriptome results showed that Cd induced up regulation of intestinal FAS associated death domain protein (FADD) expression, which may cause apoptosis and inflammation. The increase of intestinal putative heparan sulfate 2-O-sulfotransferase in cadmium treatment groups provided a mechanism for host defense. The imbalance of expression of the NOD-like receptor (NLR) family inflammatory bodies and caspase 6 in the microplastic treatment group also led to the inflammatory reaction in the intestine of sea cucumber. Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway analysis showed that in the process of fatty acid metabolism, MPs and Cd showed antagonistic effects, mainly in the inconsistent expression of Stearoyl CoA Ddesaturase (SCD1) protein. The significant changes of Toll interacting protein (TOLLIP) and E-selectin (SELE) in all Cd and MPs treatment groups may indicate the key immune response genes of sea cucumber to Cd exposure and MPs exposure. These genes were involved in the immune defense of sea cucumber exposed to different levels of Cd and MPs. This study provided insights into the mechanism of dietary MPs and Cd intake in an economically and ecologically important invertebrate species.

Exosomes are small extracellular vesicles that contain nucleic acids such as microRNAs and may participate in important biological processes. We made the initial report of exosomes from sea cucumber Apostichopus japonicus, that were classically cup-shaped and had an average size of 74.65 nm, and identified specific exosome biomarkers (HSP70, TSG101, and CD9). We explored changes in the global expression of microRNAs in exosomes from the commercially important A. japonicus under normal conditions and heat-stressed conditions for 3 and 7 d. We found that heat stress increased exosome production and modified the expression profiles of the microRNAs that they contained. Novel_mir31, novel_mir132, novel_mir26, miR-92_1, and novel_mir27 were commonly found to be differentially expressed in three comparison groups, indicating their importance in the heat stress response. The microRNA expression levels were validated by qPCR. Function analysis of the target genes of these microRNAs indicated they were involved mainly in replication and repair in the initial response of A. japonicus to heat stress exposure. Conversely, during acclimation to the high temperature conditions, the target genes of the differentially expressed microRNAs were primarily involved in metabolism adjustments. Our results will contribute to a better understanding of the regulatory roles of exosomes in sea cucumber, and provide insights into the functions of sea cucumber exosome-shuttled microRNAs against environmental stresses exacerbated by global warming.

Abstract Background IgA nephropathy (IgAN) is the most common primary glomerulonephritis globally. Increasing evidence suggests the importance of host immunity in the development of IgAN, but its dynamics during the early stage of IgAN are still largely unclear. Results Here we successfully resolved the early transcriptomic changes in immune cells of IgAN by conducting single-cell RNA-sequencing (scRNA-seq) with peripheral blood mononuclear cells. The differentially expressed genes (DEGs) between control and IgAN were predominantly enriched in NK cell-mediated cytotoxicity and cell killing pathways. Interestingly, we discovered that the number and cytotoxicity of NK cells are significantly reduced in IgAN patients, where both the number and marker genes of NK cells were negatively associated with the clinical parameters, including the levels of urine protein creatinine ratio (UPCR), serum galactose-deficient IgA1 and IgA. A distinctive B cell subset, which had suppressed NFκB signaling was predominantly in IgAN and positively associated with disease progression. Moreover, the DEGs of B cells were enriched in different viral infection pathways. Classical monocytes also significantly changed in IgAN and a monocyte subset expressing interferon-induced genes was positively associated with the clinical severity of IgAN. Finally, we identified vast dynamics in intercellular communications in IgAN. Conclusions We dissected the immune landscape of IgAN at the single-cell resolution, which provides new insights in developing novel biomarkers and immunotherapy against glomerulonephritis.

IntroductionThe virtuality, concealment, uncertainty and complexity of online trading make the online food trading market have security risks, while the lack of information, information asymmetry and imperfect market system make the “lemon problem” in the market increasingly obvious.MethodsIn order to clearly understand and manage the “lemon problem” in the online food trading market, we built an evolutionary game model involving the seller, buyers and online food trading platform, deeply analyzed the formation process of the “lemon problem” in the online food trading market, and revealed the influencing factors and effects of each subject’s strategy choice from the perspectives of subsidy, punishment, cost, and benefit.ResultsFindings of this study reveal that: (1) In the online food trading market, the strategy of the seller, buyer and platform will be stable in six situations, and the “lemon problem” will emerge with the development and evolution of the online food trading market. (2) The strategy of each subject in the online food trading market will be affected by variables like cost difference between positive performance and negative performance of the seller, punishment from the buyer with positive participation to the seller with negative performance, subsidy from the platform with positive regulation to the seller with positive performance, etc., and different factors have different influence directions and degrees on the subject strategy. (3) In the online food trading market, cost, punishment, subsidy and benefit have different effects on the subject’s strategy. Among them, cost and cost difference have the most significant impact on the subject’s strategy, followed by punishment and benefit difference, and subsidy and additional benefit have less impact on the subject’s strategy.DiscussionBased on our study findings, it is proposed that by constructing a complete and standardized system of online food trading market from the aspects of examination and verification institution, reward and punishment institution, and supervision institution, it will be able to provide reference for managing the “lemon problem” in the online food trading market, promoting the sustainable development of the market, and ensuring the safety of online food.

Sustainable livelihoods are those that are able to cope with and recover from stress and shocks, and that maintain or strengthen livelihood capacity and livelihood capital, without damaging the foundations of the natural environment. In this paper, the IPHACT framework was constructed by improving the classic IPAT equation, and the key factors affecting production output and livelihood sustainability, as well as the factor differences among different livelihood strategy groups, were analyzed. We took 24 counties and districts of Ankang, Shangluo and Hanzhong in the Qin-Ba Mountain area of southern Shaanxi Province as examples, using a survey of farmers’ livelihood status and livelihood capital accounting. The results show that the amplification effect of population size on the environmental impact of livelihood output is widespread and generally significant. Both livelihood sustainability and livelihood benefit passed the significance test in the multi-model analysis, and the negative effect of livelihood sustainability proved the negative correlation between the environmental impact of livelihood output and livelihood sustainability, that is, the higher the livelihood output dependent on natural capital, the greater the environmental impact. The livelihood transformation of Hanzhong City is developing in the direction of reducing the environmental impact of livelihood output, and farmers have successfully practiced green livelihood transformation by changing their livelihood strategies. On the road to common prosperity, livelihood demand will inevitably increase. Reducing the dependence on natural capital is the key to effectively enhancing the sustainability of livelihoods.

Background: Chondroitin sulphate synthase 3 (CHSY3) is an important enzyme that regulates glycosylation, but it has not been reported in tumours. This study explored for the first time the oncological features of CHSY3 in stomach adenocarcinoma (STAD).Methods: We analysed CHSY3 expression in STAD through the Cancer Genome Atlas (TCGA) database and verified our findings by immunohistochemical staining and Western blot experiments. The prognostic value of CHSY3 in STAD was analysed through the biological aspects of CHSY3 in STAD, such as communal clinical follow-up survival data, methylation sites, tumour immune microenvironment (TIME) and immune cell surface checkpoints. Finally, the immune-evasion potential of CHSY3 in STAD was assessed on the Tumor Immune Dysfunction and Exclusion (TIDE) website and immunohistochemical staining experiment.Results:CHSY3 overexpression in STAD was associated with a poor prognosis based on immunohistochemical staining and Western blot experiments. Multivariate Cox analysis suggested that CHSY3 could be an independent prognostic risk factor. Pathway enrichment and TIME analysis demonstrated that CHSY3 up-regulated mesenchymal activation and immune activation signals in STAD, while TIDE assessment revealed that the risk of immune evasion was significantly higher in the high CHSY3 expression group than in the low CHSY3 expression group. Risk model scores based on CHSY3-associated immune cell surface checkpoints also presented poor prognosis, and immune evasion was significantly higher in the high-risk group than in the low-risk group.Conclusions: This study analysed CHSY3 from multiple biological perspectives and revealed that CHSY3 can be a biomarker of poor prognosis and mediates the TIME immune-evasion status in STAD.

We demonstrate a hybrid solution for simultaneous transfer of ultra-stable optical frequency, RF frequency and time signal on a 100 km fiber spool with the same wavelength based on coherent demodulation and optical phase-lock-loop (OPLL). The one pulse per second (1PPS) is encoded by phase and RF frequency is encoded with an intensity modulator. The noise caused by fiber link is compensated by the round-trip method. And the bad effect on optical frequency caused by optical phase modulation (PM) can be cleaned using OPLL. When the fiber link is stabilized, the transferred time deviation (TDEV) can be 2.4E-11@1s and 1.9E-12@104s, RF frequency stability can achieve 8.0E-14@1s and 2.5E-17@104s, the modified Allan deviation (MDEV) of cleaned optical frequency can be 1.4E-16@1s and 1.6E-19@104s.

The core enterprise oriented entrepreneurship ecosystem plays an important role in promoting the sustainable development of innovation and entrepreneurship and realizing the multi-agent win-win symbiosis. Based on the structure of the core enterprise oriented entrepreneurship ecosystem, this paper constructs an evolutionary game model with the participation of core enterprises, start-up enterprises and the government, analyzes the equilibrium point and corresponding stability conditions for the evolutionary game model, and carries out a simulation analysis on different stability conditions from the aspects of the initial willingness of the participants, the intensity of the subsidy and punishment, the cost and benefit of the strategy selection. The results show that: (1) In order to achieve different equilibrium states, different stability conditions should be satisfied, and there may be multiple stable points under each stability condition. (2) The initial intention will positively affect the subject’s positive strategy selection, and negatively affect the subject’s negative strategy selection. (3) The strategy selections of core enterprises, start-up enterprises and the government will be affected by many factors, and different factors have different influences on the directions and degrees of the main strategy selections. Based on these conclusions, we can build a unified information platform, set up a moderately high, flexible reward and punishment mechanism, and build a multi-party collaborative governance system. In addition, we can also use the stability conditions to judge the development status of the ecosystem to promote the formation of the ideal core enterprise oriented entrepreneurship ecosystem.

Triggered by COVID-19, one of the most dramatic crashes in the stock market in history occurred in March 2020. The sharp reductions in NASDAQ insurance stock indexes were observed after the occurrence of COVID-19 and in March 2020. In this study, the NASDAQ insurance stock markets (including NASDAQ Insurance Index, Developed Markets Insurance Index, and Emerging Markets Insurance Index) and NASDAQ Composite Index are utilized. The “scissors difference” between the NASDAQ Insurance Index and NASDAQ Composite Index is observed. The dynamic effects of the COVID-19 epidemic and the March 2020 crash on the multifractality of four series are explored. Firstly, the apparent and intrinsic multifractality, the components of multifractality, and the dynamic effects of the COVID-19 epidemic on these indexes are analyzed. Secondly, the multifractal cross-correlation between the NASDAQ Insurance Index and NASDAQ Composite Index is investigated. The dynamic influence of the COVID-19 epidemic on the cross-correlation is examined. The multifractal analysis results reveal that four series both before and after the occurrence of COVID-19 have multifractal characteristics. The stronger multifractal characteristics and the greater multifractal degree are obtained after the occurrence of COVID-19. The intrinsic multifractality of the three indexes ascends largely after the occurrence of COVID-19. The multifractal cross-correlation analysis illustrates that the cross-correlation between two indexes before and after the occurrence of COVID-19 is multifractal. The stronger multifractal cross-correlations and greater multifractal degrees are shown. The contribution of the intrinsic multifractal cross-correlation increased after the occurrence of COVID-19.

Gonads play a critical role in the reproduction of sea cucumbers, and their development is regulated by a large number of genes. To date, the ideal reference genes (RGs) for studying sea cucumber gonadal development have not been reported. In this study, we selected ten RGs and conducted quantitative real-time PCR analysis to identify the most stable among Apostichopus japonicus gonads at four different stages of gonadal development. We used ∆Cq, GeNorm, NormFinder, BestKeeper, and RefFinder to evaluate the stability of candidate genes. EF1β and Grb2 together were the preferred combination of stable RGs among all of the gonad samples and for the ovaries alone. UBQ combined with RPS4 was the best combination of stable RGs in the testes. This study aimed to provide a basis for analysis of differences in gene expression related to gonadal development in sea cucumbers, and the results provide valuable information for further studies of the molecular mechanisms involved in sea cucumber gonadal development.

Mental health is a major public health issue that affects social development. This study aims to explore the psychological state of residents of Hainan Island and its influencing factors during the COVID-19 pandemic and to provide a scientific basis for the formulation of psychological counseling measures to be used after the pandemic. We used the nonprobability snowball sampling method to conduct an online survey from February 21 to February 28, 2020, and collected a sample of 533 respondents. Using a binary logistic regression model and network analysis, the psychological state of residents and the main factors were analyzed during the concentrated outbreak of COVID-19 (from January 20 to February 16, 2020). The study found that during the pandemic, 92.7% of the respondents were in a poor state of mind, and 54.2% experienced severe psychological stress. The mental state is spatially expressed as a pattern of "high in the middle and low in the surroundings." Second, within the four-week sample, the overall psychological tension showed an inverted U-shaped trend. Respondents feeling stressed were most common in the second week, when they accounted for 87.99% of the total. Third, individual characteristics such as gender, age, fixed expenditure, and family size significantly affect the mental state. Women, the elderly, residents with fixed expenditures, and residents with large families are at greater risk of psychological stress. Finally, external factors such as the distance between residents and the location of cases and the node degree of the pandemic transmission network have a significant impact on the psychological state. However, residents in the least developed areas of Hainan Island, which are far away from active cases and have a low node degree, are more prone to psychological pressure. The government needs to pay special attention to these groups when constructing a long-term mechanism of psychological crisis intervention and increase public health resource investment in underdeveloped areas.

The ability of the food industry to practice a high degree of corporate social responsibility is related to whether national food safety can be effectively guaranteed. By taking the food enterprises in China’s A-share listed companies from 2009–2018, this paper probes into the influence of China’s macroeconomic policy uncertainty on corporate social responsibility, and depicts the differencent impact of enterprise’s practice of social responsibility under government association and market association, respectively. The results firstly show that, the uncertainty of economic policy has a negative effect on the social responsibility of food enterprises, with a regression coefficient of -0.013. Secondly, nonstate-owned enterprises are more vulnerable to macroeconomic fluctuations in the practice of social responsibility. Thirdly, in the case of greater economic policy uncertainty, enterprises with stronger market connections in the food industry tend to be more conservative in their CSR. The above research results not only verify the path of transmission of economic policy uncertainty to corporate social responsibility, but also provide important ideas and references for improving the level of corporate social responsibility in the food industry and ensuring food safety.

Abstract Background Pyroptosis belongs to a novel inflammatory programmed cell death pathway, with the possible prognosis of endometrial cancer related to the terminal protein GSDMD. Hydrogen exerts a biphasic effect on cancer by promoting tumor cell death and protecting normal cells, which might initiate GSDMD pathway-mediated pyroptosis. Methods We performed immunohistochemical staining and western immunoblotting analysis to observe expression of NLRP3, caspase-1, and GSDMD in human and xenograft mice endometrial cancer tissue and cell lines. We investigated treatment with hydrogen could boost ROS accumulation in endometrial cancer cells by intracellular and mitochondrial sources. GSDMD shRNA lentivirus was used to transfect endometrial cancer cells to investigate the function of GSDMD protein in pyroptosis. Propidium iodide (PI) staining, TUNEL assay, measurement of lactate dehydrogenase (LDH) release and IL-1β ELISA were used to analysis pyroptosis between hydrogen-supplemented or normal culture medium. We conducted in vivo human endometrial tumor xenograft mice model to observe anti-tumor effect in hydrogen supplementation. Results We observed overexpression of NLRP3, caspase-1, and GSDMD in human endometrial cancer and cell lines by IHC and western immunoblotting. Hydrogen pretreatment upregulated ROS and the expression of pyroptosis-related proteins, and increased the number of PI- and TUNEL-positive cells, as well as the release of LDH and IL-1β, however, GSDMD depletion reduced their release. We further demonstrated that hydrogen supplementation in mice was sufficient for the anti-tumor effect to inhibit xenograft volume and weight of endometrial tumors, as mice subjected to hydrogen-rich water displayed decreased radiance. Tumor tissue sections in the HRW groups presented moderate-to-strong positive expression of NLRP3, caspase-1 and GSDMD. Hydrogen attenuated tumor volume and weight in a xenograft mouse model though the pyroptotic pathway. Conclusions This study extended our original analysis of the ability of hydrogen to stimulate NLRP3 inflammasome/GSDMD activation in pyroptosis and revealed possible mechanism (s) for improvement of anti-tumor effects in the clinical management of endometrial cancer.

A constrained optimization model and an iterative optimization algorithm based on PSO are designed for rural leisure tourism passenger flow scheduling. Compared with the traditional tourist dispatching scheme, this model maximizes the overall tourist experience and operation profit of the whole region on the base of protection of tourists' travel experience and the interests of operators in the dispatching spots. Simulations and comparisons are taken to evaluate the feasibility and effectiveness of the model and the optimization. The simulation results show that compared with the shortest-distance-based traffic scheduling scheme and the gravity-model-based scheme, the new model and optimization could meet the requirements of the rural leisure tourists dispatching and bring better tourist experience and tourism profit.

Abstract Background Gastric cancer is a common malignant tumor with high morbidity and mortality worldwide, which seriously affects human health. Gramicidin is a short peptide antibiotic which could be used for treating infection induced by bacteria or fungi. However, the anti-cancer effect of gramicidin on gastric cancer cells and its underlying mechanism remains largely unknown. Results Gastric cancer cells SGC-7901, BGC-823 and normal gastric mucosal cells GES-1 were treated with different concentrations of gramicidin respectively. The results of CCK-8 experiment revealed cellular toxicity of gramicidin to cancer cells while cell colony formation assay showed that gramicidin significantly inhibited the proliferation of gastric cancer cells, but had little effect on normal gastric mucosal cells. In addition, the wound healing assay showed that gramicidin inhibited the migration of SGC-7901 cell. Meanwhile, apoptosis and cell cycle analysis revealed that gramicidin induced cell apoptosis with G2/M cell cycle inhibition. Furthermore, western blot analysis demonstrated that gramicidin down-regulated the expression of cyclinD1 and Bcl-2 as well as the FoxO1 phosphorylation. Conclusions The current study illustrated the anti-tumor activity of gramicidin on gastric cancer cells, providing a possibility for gramicidin to be applied in clinical practice for the treatment of gastric cancer.

ObjectiveThe present study aimed to evaluate the role of integrating the pretreatment neutrophil-to-lymphocyte ratio (NLR) into the eighth edition of the AJCC staging system for nasopharynx cancer in an endemic region.MethodsBetween May 2007 and December 2012, a total of 713 cases with NPC were retrospectively analyzed. The separation ability in terms of overall survival (OS), local failure-free survival (LFFS), distant metastasis-free survival (DMFS), and failure-free survival (FFS) was evaluated. The discriminatory ability was assessed using Harrell’s concordance index (c-index). Recursive partitioning analysis (RPA) was conducted and incorporated with pretreatment NLR.ResultsWhen integrated with NLR, the separate and discriminatory abilities for N classifications were improved in terms of OS and DMFS, but not for T categories. By using Recursive partitioning analysis, five subgroups were generated. Compared with the overall stage, the integration of NLR could not enhance the separate and discriminatory abilities. However, patients in the RPA 4 group gained significant benefits in terms of OS (HR 0.390 (95%CI 0.212-0.716), P = 0.002) and FFS (HR 0.548 (95%CI 0.314-0.958), P = 0.032) from the additional adjuvant chemotherapy after concurrent chemoradiotherapy.ConclusionThe integration of NLR into the 8th edition of the AJCC staging system could enhance the separation and discriminatory abilities for N classifications, but not for T categories. In addition, patients in the RPA 4 group could benefit from the addition of adjuvant chemotherapy to concurrent chemoradiotherapy.

Ischemia-reperfusion (I/R) injury contributes to the morbidity and mortality of ischemic strokes. As an in vitro model, oxygen-glucose deprivation and reperfusion (OGD/R) exposure induces neuronal injury. Low-dose ethanol preconditioning (EtOH-PC) was reported to alleviate neuronal apoptosis during OGD/R. However, whether the mitochondrial BKCa (mitoBKCa) channel is involved in the neuroprotective effect of EtOH-PC during OGD/R is not clearly defined. This study attempts to explore the mediation of the mitoBKCa channel in the neuroprotective effect of EtOH-PC on OGD/R-induced neuronal apoptosis and the underlying mechanisms. OGD/R model was established using primary cortical neurons that were preincubated with ethanol. Subsequently, the cell viability was measured by CCK-8 assay, and the apoptotic cells were determined by TUNEL assay. Annexin V/7-AAD staining and mitochondrial membrane potential using JC-10 were detected by flow cytometry. Western blot analysis was performed to check the apoptosis-related proteins. In the mixed primary culture, 95% neurofilament-positive cells were cortical neurons. Low-dose EtOH-PC (10 mmol/L) for 24 h significantly attenuated the OGD2h/R24h-induced neuronal apoptosis through activating the BKCa channel. Further investigations suggested that ethanol pretreatment increased the mitochondrial membrane potential (MMP) and downregulated the production of cleaved caspase 3 in OGD/R-injured neurons by activating the mitoBKCa channel. Low-dose ethanol pretreatment significantly attenuated the OGD/R-induced neuronal apoptosis mediated by the mitoBKCa channel which modulated the mitochondrial function by impeding the uncontrolled opening of mitochondrial permeability transition pore (MPTP).

The interaction process between farmers’ livelihood and land use is a core link between the interaction and the coupling of the human–land system. It is a complex evolutionary process that involves several issues such as resource-intensive utilization and economic development. This study first constructs a dynamic model of the coupling system of farmers’ livelihood efficiency and land use and designs two types of 31 scenarios based on the farmer’s livelihood mode and land use. This study further simulates the coupling coordination relationship in different scenarios and then proposes suggestions for sustainable development. The findings of our study show that with the development of time, the livelihood capital, livelihood output, and land use level first showed a decline and then an increase. The results also reveal that livelihood efficiency and coordination degree are related to the livelihood mode and land use mode of farmers, while the land use level is not significantly associated with the livelihood mode of farmers. Pure-agriculture farmers have the lowest livelihood efficiency and coordination degree when they have no planting and breeding poultry, while part-time farmers have the highest land use level and coordination degree when they plant cash crops and breeding livestock. Besides, non-agriculture farmers have the highest livelihood efficiency and the lowest land use level when they neither plant crops nor breed livestock. To improve the level of coupling coordination, it is necessary not only to flexibly adjust the farmer’s livelihood and land use mode but also to optimize the allocation of various resources to promote the coordinated and sustainable development of farmers.

Although the outbreak of the COVID-19 pandemic caused serious restrictions on human activities in and around Shanghai, China, the period can be viewed as a helpful experiment to investigate the correlation between PM2.5 and O3 concentrations. In this study, the hourly PM2.5 and O3 series in four cities (i.e., Shanghai, Jiaxing, Nantong and Suzhou) from 27 November 2019 to 23 March 2020 are used. The “seesaw effect” is observed in the study data. The dynamic impacts of the COVID-19 pandemic on the multifractal cross-correlations and the coordinated control degree of PM2.5-O3 are examined in these cities. First of all, the multifractal cross-correlations, multifractality components and dynamic influences of the COVID-19 pandemic on cross-correlations between PM2.5 and O3 in four cities are illustrated. Furthermore, a new quantification index, ζ, evaluating the coordinated control degree of PM2.5-O3 is developed, validated and compared. The multifractal cross-correlation analysis results reveal that the cross-correlations between PM2.5 and O3 in and around Shanghai both before and during the COVID-19 partial lockdown have multifractal characteristics. Moreover, there are weaker multifractal cross-correlation degrees of PM2.5-O3 in four cities during the COVID-19 partial lockdown. The multifractal cause analysis based on stochastic simulation illustrates that the impacts of multifractality due to the nonlinear correlation part are greater than the linear correlation part and the fat-tailed probability distribution part in and around Shanghai. The intrinsic multifractal cross-correlations decreased in all cities during the COVID-19 lockdown. However, the effects of the COVID-19 lockdown on the multifractal cross-correlations are limited from the perspective of intrinsic multifractality. The mean values of ζ in and around Shanghai all increase during the COVID-19 partial lockdown, which indicates that the PM2.5-O3 coordinated control degrees in all four cities become weaker.

Apostichopus japonicus achieves intestinal regeneration in a short period after evisceration, and multiple genes are involved in this process. The transcriptome of A. japonicus was screened for regeneration-associated protein (Aj-Orpin), a gene that is specifically upregulated during intestinal regeneration. The expression and function of Aj-Orpin were identified and investigated in this study. The 5′ and 3′ RACE polymerase chain reaction (PCR) was used to clone the full-length cDNA of Aj-Orpin. The open reading frame codes for a 164 amino-acid protein with an EF-hand_7 domain and overlapping signal peptides and transmembrane regions. Moreover, Aj-Orpin mRNA and protein expression during intestinal regeneration was investigated using real-time quantitative PCR and Western blot. The expression pattern of Aj-Orpin in the regenerating intestine was investigated using immunohistochemistry. The results showed that Aj-Orpin is an exocrine protein with two EF-hand-like calcium-binding domains. Expression levels were higher in the regenerating intestine than in the normal intestine, but protein expression changes lagged behind mRNA expression changes. Aj-Orpin was found to play a role in the formation of blastema and lumen. It was primarily expressed in the serosal layer and submucosa, suggesting that it might be involved in proliferation. These observations lay the foundation for understanding the role of Orpin-like in echinoderm intestinal regeneration.

Most species of the class Holothuroidea are able to regenerate most of their internal organs following a typical evisceration process, which is a unique mechanism that allows sea cucumbers to survive in adverse environments. In this review, we compare autotomy among different type of sea cucumber and summarize the histocytological events that occur during the five stages of intestinal regeneration. Multiple cytological activities, such as apoptosis and dedifferentiation, take place during wound healing and anlage formation. Many studies have focused on the molecular regulation mechanisms that underlie regeneration, and herein we describe the techniques that have been used as well as the development-related signaling pathways and key genes that are significantly expressed during intestinal regeneration. Future analyses of the underlying mechanisms responsible for intestinal regeneration should include mapping at the single-cell level. Studies of visceral regeneration in echinoderms provide a unique perspective for understanding whole-body regeneration or appendage regeneration.

This study aims to find out the strategic priorities of the technical factors to have sustainable low carbon industry. For this purpose, a hybrid multi-criteria decision-making (MCDM) is proposed that contains three different stages. First, the economic criteria required for sustainable development and the technical needs of the low carbon industry are defined by making a comprehensive literature review. After that, economic criteria are weighted by using the fuzzy decision-making trial and evaluation laboratory (DEMATEL) methodology. Finally, technical factors are ranked with the help of the fuzzy technique for order preference by similarity to ideal solution (TOPSIS) approach. Moreover, another analysis is also performed by considering fuzzy Vise Kriterijumska Optimizacija I Kompromisno Resenje (VIKOR) to evaluate the consistency of the analysis results. The main motivation of this study is to define the primary technical factors to minimize carbon emission problem by proposing a hybrid MCDM model. The findings indicate that research and development for renewable sources has the greatest importance for low-carbon industry. In addition, the analysis results of fuzzy TOPSIS and fuzzy VIKOR are quite similar. This situation demonstrates the consistency and coherency of the ranking results. Hence, it is recommended that countries should mainly give importance to the research and development investments so that the costs of renewable energy problems can be minimized. This situation can attract the attentions of the companies to invest in these projects. In this way, it will be possible to use a cleaner energy in industrial production.

Abstract Background The goals of this study were to explore the expression profiles and functional networks of long non-coding RNAs (lncRNAs) and messenger RNAs (mRNAs) in mesenchymal stromal cells (MSCs) involved in regulating the function of monocytes and to clarify the mechanisms by which MSCs exert immunoregulatory effects on monocytes. Methods MSCs and CD14+ monocytes were separately isolated. The immunoregulatory effects of MSCs on monocytes were determined by flow cytometry. lncRNAs and mRNAs that were differentially expressed (DE) between the control group (MSCs only) and co-culture group (MSCs co-cultured with monocytes) were identified through high-throughput sequencing and bioinformatic analyses and were confirmed by qRT-PCR. Bioinformatic analyses were performed to identify the critical biological functions and signalling pathways involved in MSC-mediated monocyte regulation and to identify the functional networks formed between DE mRNAs and lncRNAs. Results MSCs showed a strong ability to induce monocyte migration but inhibited monocyte differentiation into M1 macrophages. A total of 145 DE lncRNAs and 768 DE mRNAs were identified between the control and co-culture groups. Significant fold changes in lncRNAs and mRNAs were confirmed by qRT-PCR. GO analysis demonstrated that DE mRNAs and lncRNAs were highly associated with terms related to binding and biological regulation. KEGG analysis revealed 122 significantly regulated pathways, including the cytokine-cytokine receptor pathway and chemokine signalling pathway. Interaction and co-expression networks were constructed for DE mRNAs and lncRNAs, and several key microRNAs were identified in the competitive endogenous RNA (ceRNA) network. Target genes of the DE lncRNAs were analysed to predict critical mRNA-lncRNA axes involved in the immunoregulatory function of MSCs. Conclusions Our research describes the lncRNA and mRNA expression profiles and functional networks involved in MSC-mediated regulation of monocytes. These results provide possible molecular mechanisms for the immunoregulatory function of MSCs and may help to elucidate possible molecular therapeutic targets in MSCs for the treatment of autoimmune diseases.

Ecological compensation affects farmers’ livelihoods, as well as sustainability and social equity. Yulin City is the core area of the energy “Golden Triangle” in China. The farmers livelihood strategy model was used to analyse data from a household survey and interviews with selected farmers. The paper analyses the current livelihood strategies of farmers and impact of ecological compensation from the perspective of participation and income. The results showed that: (1) the significant difference between the coal resource development and oil and gas resource development was agricultural planting. Migrant work and non-agricultural self-employment in the coal resource development region were higher than that of the oil and gas resource development region. (2) The income from agricultural planting in the coal resource development region was significantly lower than that of the oil and gas resource development region. The labour, non-agricultural self-employment, and transfer incomes were higher in the coal resource development region than that in the oil and gas resource development region. (3) Ecological compensation has a significant negative effect on participation and income from agricultural planting, as well as a positive effect on participation and income from non-agricultural self-employment. We have proposed measures to strengthen ecological compensation mechanisms and improve the farmers’ livelihoods, in order to inform decision-making and high-quality sustainable development in energy development region.

Circular economy is a critical approach to realize the coordinated development of society, economy, and ecological environment. Given the fact that urban is a complex system in which human beings integrate material, energy, information, and natural environment and interact and influence each other, reviewing the urban circular economy research and development could benefit for having a better and comprehensive understanding on urban complexity. Mainly based on the Chinese literature studies from 1999 to 2020, this study aims to present an in-depth analysis of the research themes, policy systems, and index system of Chinaʼs urban-scale circular economy, discuss the changes and evolution trends of themes, levels, and perspectives in time series, sort out the policy systems at both the national and local levels, and analyze the design principles and application fields of indicators. Finally, we propose that future development of an urban circular economy should be built based on modern techniques, technologies, and models. The construction of development mechanism on the circular economy should be framed as “government-led, market-driven, legal norms, policy support, technological support, and public participation” and inject concepts such as “Internet +,” “sharing economy,” “Internet of Things,” and “artificial intelligence.”

Introduction: Environmental protection is an essential issue for sustainable development, and its execution power mainly comes from individual environmental protection behavior. This study investigates the resident environmental protection behaviors and influencing factors in the ecologically fragile areas of western China based on a total of 1062 households by adopting the participatory assessment method. Then, this study used the structural equation model to empirically analyze the impact of social capital, environmental risk perception, and environmental awareness on residents’ environmental protection behavior.Outcomes: The results showed that environmental awareness is the basis for practicing environmental protection behavior, consistent with the existing research; social capital has a significant impact on residents’ environmental protection behavior, with an impact coefficient of 0.347. Hence, environmental risk perception has an indirect influence on environmental protection behavior through environmental awareness. Besides, compared with urban residents, rural residents’ environmental awareness and social capital have a stronger role in environmental protection behavior.Conclusions: The study clarified the influence path of residents’ environmental protection behavior, thus effectively providing a decision-making basis for the government to guide residents in the environmental governance system.

Public environmental cognition is an important basis for optimizing environmental management and reducing tensions between humans and land. Although the level of environmental cognition is a gradual process under normal conditions, it often changes qualitatively because of major public emergencies. During the 2019 new coronavirus epidemic (COVID-19), the most significant public health event in recent years, 24,188 national samples were obtained based on a network survey. The comprehensive evaluation method was used to assess the impact of major public events on public environmental cognition and the characteristics of spatial and temporal distribution. The findings are as follows. (1) During the epidemic period, sudden public health emergencies effectively promoted the national residents’ environmental awareness, whether urban residents or rural; most respondents generally agreed with the concept of “respect nature and cherish life”. (2) The environmental cognition of national residents was higher in the northwest and lower in the northeast of China, which is suitable for economic and social development and humanistic tradition. (3) There was a clear positive correlation between environmental awareness and education level. (4) During the epidemic, nervousness of respondents had a negative effect on environmental cognition. This study provides scientific support and a basis for decision making for the government to carry out environmental management optimization and improve the ecological and environmental cognition of the public, as well as devise effective intervention mechanisms with different time and space dimensions for similar future public health emergencies.

Abstract Objective This study aimed to evaluate the safety and efficacy of three‐dimensional conformal radiotherapy (3D‐CRT) and hepatic resection for patients with hepatocellular carcinoma (HCC) involving portal vein tumor thrombus (PVTT). Methods We retrospectively analyzed 323 HCC patients involving PVTT. Among them, 134 patients underwent 3D‐CRT, while 189 controls treated with hepatic resection (HR). Survival rate and prognostic analysis were performed using Kaplan‐Meier method and Cox regression analyses. Results The 1‐, 2‐, and 3‐year overall survival (OS) of RT group and HR group was 54% vs 62%, 33% vs 47%, and 18% vs 43%, respectively (P = 0.003). In the subgroup of PVTT type analysis, the 1‐, 2‐, and 3‐year OS in RT group was 65%, 39%, and 19%, respectively, while that in HR group was 83%, 53%, and 42%, respectively, in type I PVTT (P

Background: Astaxanthin from natural sources is typically esterified with fatty acids; hence, it must be hydrolyzed to remove esters before identification and quantification by conventional HPLC. Alkaline-catalyzed saponification and enzyme-catalyzed enzymolysis are the most commonly used de-esterification methods. However, information on the efficiency and isomerization during de-esterification of natural astaxanthin esters by these two methods remains scarce. Therefore, we conducted two HPLC-based experiments to determine which method is better for hydrolyzing astaxanthin esters. Results: To assess the effect of enzymolysis (0.67 U/mL cholesterol esterase, at 37°C) and saponification (0.021 M NaOH, at 5°C) conditions on free astaxanthin recovery and destruction or structural transformation of astaxanthin, we varied the total treatment time across a range of 195 min. The results showed that enzymolysis and saponification were complete in 60 min and 90 min, respectively. After complete hydrolysis, the maximum free astaxanthin recovery obtained by enzymolysis was 42.6% more than that obtained by saponification. The identification of by-products, semi-astacene and astacene, during the process of saponification also indicated that a more severe degradation of astaxanthin occurred during saponification. Moreover, the composition of astaxanthin isomers during saponification was similar to that of the isomers during enzymolysis between 30 min and 75 min (all-trans:9-cis:13-cis = 21:3:1, approximately) but dramatically changed after 90 min, whereas the composition in the enzymolysis treatment remained relatively stable throughout. Conclusion: Compared with saponification, enzymolysis with cholesterol esterase was recommended as a more accurate method for de-esterification of natural astaxanthin esters for further qualitative and quantitative HPLC analysis.How to cite: Su F, Xu H, Yang N, et al. Abstract Background PHD-finger domain protein 5A (PHF5A) is a highly conserved small transcriptional regulator also involved in pre-mRNA splicing; however, its biological functions and molecular mechanisms in non-small cell lung cancer (NSCLC) have not yet been investigated. The purpose of this study was to determine the functional relevance and therapeutic potential of PHF5A in lung adenocarcinoma (LAC). Methods The expression of PHF5A in LAC tissues and adjacent non-tumor (ANT) tissues was investigated using immunohistochemistry of a tissue microarray, qRT-PCR, western blot and bioinformatics. The function of PHF5A was determined using several in vitro assays and also in vivo assay by lentiviral vector-mediated PHF5A depletion in LAC cell lines. Results PHF5A was highly upregulated in LAC tissues compared with the ANT counterparts, and closely associated with tumor progression and poor patient prognosis. These results were further confirmed by findings of the TCGA database. Moreover, functional studies demonstrated that PHF5A knockdown not only resulted in reduced cell proliferation, increased cell apoptosis, and cell cycle arrest, but also suppressed migration and invasion in LAC cells. PHF5A silencing was also found to inhibit LAC tumor growth in nude mice. Microarray and bioinformatics analyses revealed that PHF5A depletion led to dysregulation of multiple tumor signaling pathways; selected factors in key signaling pathways were verified in vitro. Conclusions The data suggest for the first time that PHF5A is an oncoprotein that contributes to LAC progression by regulating multiple signaling pathways, and may constitute a prognostic factor and potential new therapeutic target in NSCLC.

This paper aims to identify effective mechanisms for government poverty alleviation measures based on the livelihood sustainability of farm households in Southern Shaanxi province, China. The paper utilizes data from 414 farm households, collected through field observations and in-depth interviews in 24 rural communes in Qinba Mountain Area of Shaanxi province, China. Using theoretical research methods and employing the sustainable livelihood approach (SLA) framework, this paper analyzes poverty alleviation measures as well as the impact of varied capital availability on sustainable livelihood. The study shows that developing local industries and governmental financial support improve the sustainable livelihood of farmers and eradicate absolute poverty. The findings of this study further indicate that there is a positive correlation between poverty alleviation measures and natural and social capital for sustainable livelihood. The paper provides empirical and quantitative evidence on alleviation of poverty, and the findings will help improve the sustainability of livelihood capability of farming households. This study suggests impactful approaches to stabilizing mechanisms for poverty alleviation in rural areas over the longer term.

Genetic linkage maps have become an indispensable tool for genetics and genomics research. Sea cucumber (Apostichopus japonicus), which is an economically important mariculture species in Asia, is an edible echinoderm with medicinal properties. In this study, the first SNP-based high-density genetic linkage map was constructed by sequencing 132 A. japonicus individuals (2 parents and 130 offspring) according to a genotyping-by-sequencing (GBS) method. The consensus map was 3181.54 cM long, with an average genetic distance of 0.52 cM. A total of 6144 SNPs were assigned to 22 linkage groups (LGs). A Pearson analysis and QTL mapping revealed the correlations among body weight, body length, and papillae number. An important growth-related candidate gene, protein still life, isoforms C/SIF type 2 (sif), was identified in LG18. The gene was significantly highly expressed during the larval developmental stages. Its encoded protein reportedly functions as a guanine nucleotide exchange factor. These results would facilitate the genetic analysis of growth traits and provide valuable genomic resources for the selection and breeding of new varieties of sea cucumbers with excellent production traits.
