6. The tea plant CsLHT1 and CsLHT6 transporters take up amino acids, as a nitrogen source, from the soil of organic tea plantations

Abstract

Organic tea is more popular than conventional tea that originates from fertilized plants. Amino acids inorganic soils constitute a substantial pool nitrogen (N) available for plants. However, the amino-acid contents in soils of tea plantations and how tea plants take up these amino acids remain largely unknown. In this study, we show that the amino-acid content in the soil of an organic tea plantation is significantly higher than that of a conventional tea plantation. Glutamate, alanine, valine, and leucine were the most abundant amino acids in the soil of this tea plantation. When 15N-glutamate was fed to tea plants, it was efficiently absorbed and significantly increased the contents of other amino acids in the roots. We cloned seven CsLHT genes encoding amino-acid transporters and found that the expression of CsLHT1, CsLHT2, and CsLHT6 in the roots significantly increased upon glutamate feeding. Moreover, the expression of CsLHT1 or CsLHT6 in a yeast amino-acid uptake-defective mutant, 22∆10α, enabled growth on media with amino acids constituting the sole N source. Amino-acid uptake assays indicated that CsLHT1 and CsLHT6 are H+-dependent high- and low-affinity amino-acid transporters, respectively. We further demonstrated that CsLHT1 and CsLHT6 are highly expressed in the roots and are localized to the plasma membrane. Moreover, overexpression of CsLHT1 and CsLHT6 in Arabidopsis significantly improved the uptake of exogenously supplied 15N-glutamate and 15N-glutamine. Taken together, our findings are consistent with the involvement of CsLHT1 and CsLHT6 in amino-acid uptake from the soil, which is particularly important for tea plants grown inorganic tea plantations.

24. The effect of intercropping leguminous green manure on theanine accumulation in the tea plant: A metagenomic analysis.

Abstract

Intercropping is a widely recognised technique that contributes to agricultural sustainability. While intercropping leguminous green manure offers advantages for soil health and tea plants growth, the impact on the accumulation of theanine and soil nitrogen cycle are largely unknown. The levels of theanine, epigallocatechin gallate and soluble sugar in tea leaves increased by 52.87% and 40.98%, 22.80% and 6.17%, 22.22% and 29.04% in intercropping with soybean–Chinese milk vetch rotation and soybean alone, respectively. Additionally, intercropping significantly increased soil amino acidnitrogen content, enhanced extracellular enzyme activities, particularly β‐glucosidase and N‐acetyl‐glucosaminidase, as well as soil multifunctionality. Metagenomics analysis revealed that intercropping positively influenced the relative abundances of several potentially beneficial microorganisms, including Burkholderia, Mycolicibacterium and Paraburkholderia. Intercropping resulted in lower expression levels of nitrification genes, reducing soil mineral nitrogen loss and N2O emissions. The expression of nrfA/H significantly increased in intercropping with soybean–Chinese milk vetch rotation. Structural equation model analysis demonstrated that the accumulation of theanine in tea leaves was directly influenced by the number of intercropping leguminous green manure species, soil ammonium nitrogen and amino acid nitrogen. In summary, the intercropping strategy, particularly intercropping with soybean–Chinese milk vetch rotation, could be a novel way for theanine accumulation. Summary statement: The theanine content of tea leaves increased by 52.87% and 40.98% when intercropped with soybean–Chinese milk vetch rotation and soybean alone, respectively. The accumulation of theanine in tea leaves was directly influenced by the intercropping green manure species and their quantity, as well as the levels of soil ammonium nitrogen and amino acid nitrogen. Abstract

Background Tea, one of the most important commercial crops on earth, is strongly affected by weeds on productivity and quality. Bioherbicides are shedding new light on weed control in tea gardens in an economical and safe manner. Results A pathogenic strain SYNJC-2-2 was isolated from diseased leaves of a noxious weed, goosegrass (Eleusine indica) from a tea garden in Zhejiang Province, China. It was identified as the fungal species Bipolaris bicolor based on the morphological characteristics and phylogenetic analysis. The potential of the B. bicolor strain SYNJC-2-2 as a bioherbicide was assessed by determining its efficacy to control weeds and selectivity to crops, its infection process and the influence of environmental conditions on conidial production and germination. The ED90 (effective dose of conidia resulting in 90 disease index) of SYNJC-2-2 on goosegrass was 2 × 104 conidia mL-1 . Additionally, three Poaceae weeds, Setaria viridis, Microstegium vimineum and Pennisetum alopecuroides, were also extremely susceptible to SYNJC-2-2. SYNJC-2-2 was safe to 14 out of 17 crop species in 9 families, especially tea plants. Conidial germination, hyphal growth and appressorial formation occurred within 3 to 6 h on goosegrass leaves. Hyphae invaded leaf tissues mainly through epidermal cell junctions and cracks, causing cell death and necrotic lesions within 2 days on inoculated leaves and killing goosegrass plants within 7 days. Furthermore, SYNJC-2-2 has a strong adaptability to environmental variables and high conidial production capacity on goosegrass juice agar media. Conclusion B. bicolor strain SYNJC-2-2 has the potential to be developed as a bioherbicide for controlling goosegrass, especially in tea gardens. This article is protected by copyright.
