Abstract

BACKGROUND: The Chinese chestnut (Castanea mollissima) is widely cultivated in China for nut production. This plant also plays an important ecological role in afforestation and ecosystem services. To facilitate and expand the use of C. mollissima for breeding and its genetic improvement, we report here the whole-genome sequence of C. mollissima. FINDINGS: We produced a high-quality assembly of the C. mollissima genome using Pacific Biosciences single-molecule sequencing. The final draft genome is ∼785.53 Mb long, with a contig N50 size of 944 kb, and we further annotated 36,479 protein-coding genes in the genome. Phylogenetic analysis showed that C. mollissima diverged from Quercus robur, a member of the Fagaceae family, ∼13.62 million years ago. CONCLUSIONS: The high-quality whole-genome assembly of C. mollissima will be a valuable resource for further genetic improvement and breeding for disease resistance and nut quality.

Abstract

Chestnut plants (Castanea) are important nut fruit trees worldwide. However, little is known regarding the genetic relationship and evolutionary history of different species within the genus. How modern chestnut plants have developed local adaptation to various climates remains a mystery. The genomic data showed that Castanea henryifirst diverged in the Oligocene ~31.56 million years ago, followed by Castanea mollissima,and the divergence between Castanea seguiniiand Castanea crenataoccurred in the mid-Miocene. Over the last 5 million years, the population of chestnut plants has continued to decline. A combination of selective sweep and environmental association studies was applied to investigate the genomic basis of chestnut adaptation to different climates. Twenty-two candidate genes were associated with temperature and precipitation.We also revealed the molecular mechanism by which CmTOE1interacts with CmZFP8and CmGIS3to promote the formation of non-glandular trichomes for adaptation to low temperature and high altitudes. We found a significant expansion of CER1genes in Chinese chestnut (C. mollissima) and verified the CmERF48regulation of CmCER1.6adaptation to drought environments. These results shed light on the East Asian chestnut plants as a monophyletic group that had completed interspecific differentiation in the Miocene, and provided candidate genes for future studies on adaptation to climate change in nut trees.

Abstract

An optimized protocol was established to isolate large numbers of mature living generative cells from Nicotiana tabacum, Vicia faba L., and Iris tectorium Maxim. Fresh pollen grains on the day of anthesis were incubated in a solution containing 9% mannitol. The optimal pH and temperature for the reaction were 5.8 and 24°C, respectively. For a short time, large amount of generative cells were subsequently released into the solution and were immediately collected for preparation. The isolated pollen protoplasts were demonstrated to be living by microscopy in a fluorochromatic reaction with fluorescein diacetate (FDA). The isolation rate was above 95%.

Abstract

To investigate the relation between fertilization and lectin receptor distribution, fluorescein isothiocyanate (FITC) conjugated Canavalia ensiformis agglutinin (Con A) and Triticum vulgaris agglutinin (WGA) were applied as markers to localize Con A and WGA receptors on plasma membrane of female cells in tobacco before and after fertilization by use of single cell manipulation and cooled-CCD as well. Results illustrated that polarized distribution of Con A receptor before fertilization changed to uniform after fertilization on plasma membrane of central cell and egg cell. And WGA receptors were present on the surface membrane of unfertilized female cells, but they were absent on that of fertilized female cells. So fertilization induced changes in distribution patterns of lectin receptors and composition of glycoconjugates and might therefore induce molecular modifications of plasma membrane of female cells.

Abstract

The presence of mosaicism in the organization of concanavalin agglutinin (Con A) binding sites on murine egg cells was first reported 30 year ago. This discovery has triggered extensive studies into the roles of glycoproteins in gamete interactions in animals. This report comprises the first account of the existence of the mosaicism in higher plants. The distribution of Con A binding sites on both egg cells and central cells of tobacco (Nicotiana tabacum) was found to be polar and apparently determined by the location of the nucleus of the cell. On central cells, Con A binding sites were distributed on the section of the plasma membrane surface near the nucleus. By contrast, the binding sites on egg cells were concentrated away from the nucleus. Therefore, polarity of the plasma membrane component of female cells was confirmed for the first time. It is proposed that such polarized ConA binding sites could be involved in sperm recognition.

Abstract

Chestnuts are important ecological forest tree species. They can form mycorrhizal connections with a variety of ectotrophic mycorrhizal fungi. Mycorrhizal hyphae can expand the absorbing area to facilitate the intake of nutrients, thereby promoting plant growth and development. In this study, a chestnut-fungus symbiosis system was established in vitro , and typical mycorrhizal structures (root tips bifurcated and apical enlargement) were found after three-month inoculation. Phosphorus content and H + efflux rate of chestnut roots increased significantly after being inoculated with Scleroderma citrinum compared to the non-inoculated control. The increased H + efflux rate provides indirect evidence for the role of ecto-mycorrhizae in increased phosphate uptake. Finally, phylogenetic tree was clustered by MEGA5.0 based on amino acid sequences of five phosphate transporters of Castanea mollissima and known phosphate transporters of other plants. The results showed that CmPT4 was in a clade with known mycorrhizal phosphate transporters, and thus this gene was preliminarily a predicted mycorrhizal phosphate transporter. Further real-time quantitative PCR showed that five phosphate transporter genes ( CmPT1, CmPT2, CmPT3, CmPT4, and CmPT5 ) were upregulated under phosphorus deficiency treatment, and only CmPT4 was induced in the mycorrhizal roots of chestnut. Therefore, It can be concluded CmPT4 is a mycorrhizal phosphate transporter gene in Castanea mollissima . This study provides a foundation for further analysis of chestnut mycorrhizal formation and phosphate uptake improvement by phosphate transporters. [ABSTRACT FROM AUTHOR]
