Abstract Tissue-derived extracellular vesicles (EVs) are emerging as pivotal players to maintain organ homeostasis, which show promise as a next-generation candidate for medical use with extensive source. However, the detailed function and therapeutic potential of tissue EVs remain insufficiently studied. Here, through bulk and single-cell RNA sequencing analyses combined with ultrastructural tissue examinations, we first reveal that in situ liver tissue EVs (LT-EVs) contribute to the intricate liver regenerative process after partial hepatectomy (PHx), and that hepatocytes are the primary source of tissue EVs in the regenerating liver. Nanoscale and proteomic profiling further identify that the hepatocyte-specific tissue EVs (Hep-EVs) are strengthened to release with carrying proliferative messages after PHx. Moreover, targeted inhibition of Hep-EV release via AAV-shRab27a in vivo confirms that Hep-EVs are required to orchestrate liver regeneration. Mechanistically, Hep-EVs from the regenerating liver reciprocally stimulate hepatocyte proliferation by promoting cell cycle progression through Cyclin-dependent kinase 1 (Cdk1) activity. Notably, supplementing with Hep-EVs from the regenerating liver demonstrates translational potential and ameliorates insufficient liver regeneration. This study provides a functional and mechanistic framework showing that the release of regenerative Hep-EVs governs rapid liver regeneration, thereby enriching our understanding of physiological and endogenous tissue EVs in organ regeneration and therapy.

Abstract Plastic bronchitis is a relatively uncommon illness that has been reported in all age groups. This case report describes a specific manifestation of plastic bronchitis in two pediatric brothers influenced by both smoke inhalation and influenza A virus infection. The therapeutic approach mainly involved symptomatic supportive care, antiviral therapy, repeated bronchoscopic alveolar lavage, and bronchial cast removal. Eventually, both patients went into remission. Bronchoscopy proved to be helpful in diagnosing and treating these cases.

The deep sand gravel type pore brine in Qaidam Basin belongs to the unsaturated chloride type brine with high calcium, high sodium and low potassium. 480 kg of gravel-type pore brine at 1000～2000 meters underground in Dalangtan mining area was selected for the test, and evaporated and crysta-llized in the natural state in summer. The distribution law of ions in the solid and liquid phases, the salt crystallization law and crystal morphology were analyzed, and the main precipitation characteristics of potassium were studied. The results show when evaporating in the outdoor natural state, the salt precipitation process is mainly divided into four stages: halite→carnallite→bischofite→tachyhydrite. The salt evolution law in the halite stage conforms to the phase diagram of Na+, K+, Mg2+//Cl-—H2O at 25 ℃, and that in carnallite and bischofite stages conforms to the phase diagram of K+, Ca2+, Mg2+//Cl-—H2O system at 25 ℃. There is a separate precipitation stage of potassium, at which potassium is mainly precipitated in the form of carnallite, and the recovery rate of potassium is 85.76%. The K+content in the potassium mixed salt can reach 9.27%, which is a high quality raw material for the production of potassium fertilizer. Calcium precipitates after the brucite stage. The experimental results can provide references for the development and utilization of this type of brine.

Abstract Background Metabolic reprograming and immune escape are two hallmarks of cancer. However, how metabolic disorders drive immune escape in head and neck squamous cell carcinoma (HNSCC) remains unclear. Therefore, the aim of the present study was to investigate the metabolic landscape of HNSCC and its mechanism of driving immune escape. Methods Analysis of paired tumor tissues and adjacent normal tissues from 69 HNSCC patients was performed using liquid/gas chromatography‐mass spectrometry and RNA‐sequencing. The tumor‐promoting function of kynurenine (Kyn) was explored in vitro and in vivo. The downstream target of Kyn was investigated in CD8+ T cells. The regulation of CD8+ T cells was investigated after Siglec‐15 overexpression in vivo. An engineering nanoparticle was established to deliver Siglec‐15 small interfering RNA (siS15), and its association with immunotherapy response were investigated. The association between Siglec‐15 and CD8+ programmed cell death 1 (PD‐1)+ T cells was analyzed in a HNSCC patient cohort. Results A total of 178 metabolites showed significant dysregulation in HNSCC, including carbohydrates, lipids and lipid‐like molecules, and amino acids. Among these, amino acid metabolism was the most significantly altered, especially Kyn, which promoted tumor proliferation and metastasis. In addition, most immune checkpoint molecules were upregulated in Kyn‐high patients based on RNA‐sequencing. Furthermore, tumor‐derived Kyn was transferred into CD8+ T cells and induced T cell functional exhaustion, and blocking Kyn transporters restored its killing activity. Accroding to the results, mechanistically, Kyn transcriptionally regulated the expression of Siglec‐15 via aryl hydrocarbon receptor (AhR), and overexpression of Siglec‐15 promoted immune escape by suppressing T cell infiltration and activation. Targeting AhR in vivo reduced Kyn‐mediated Siglec‐15 expression and promoted intratumoral CD8+ T cell infiltration and killing capacity. Finally, a NH2‐modified mesoporous silica nanoparticle was designed to deliver siS15, which restored CD8+ T cell function status and enhanced anti‐PD‐1 efficacy in tumor‐bearing immunocompetent mice. Clinically, Siglec‐15 was positively correlated with AhR expression and CD8+PD‐1+ T cell infiltration in HNSCC tissues. Conclusions The findings describe the metabolic landscape of HNSCC comprehensively and reveal that the Kyn/Siglec‐15 axis may be a novel potential immunometabolism mechanism, providing a promising therapeutic strategy for cancers.

Abstract Malocclusion, identified by the World Health Organization (WHO) as one of three major oral diseases, profoundly impacts the dental-maxillofacial functions, facial esthetics, and long-term development of ~260 million children in China. Beyond its physical manifestations, malocclusion also significantly influences the psycho-social well-being of these children. Timely intervention in malocclusion can foster an environment conducive to dental-maxillofacial development and substantially decrease the incidence of malocclusion or reduce the severity and complexity of malocclusion in the permanent dentition, by mitigating the negative impact of abnormal environmental influences on the growth. Early orthodontic treatment encompasses accurate identification and treatment of dental and maxillofacial morphological and functional abnormalities during various stages of dental-maxillofacial development, ranging from fetal stages to the early permanent dentition phase. From an economic and societal standpoint, the urgency for effective early orthodontic treatments for malocclusions in childhood cannot be overstated, underlining its profound practical and social importance. This consensus paper discusses the characteristics and the detrimental effects of malocclusion in children, emphasizing critical need for early treatment. It elaborates on corresponding core principles and fundamental approaches in early orthodontics, proposing comprehensive guidance for preventive and interceptive orthodontic treatment, serving as a reference for clinicians engaged in early orthodontic treatment.

Abstract While spin–orbit torque (SOT) devices are extensively investigated due to their potential for use in neural network computation, it remains challenging to explore the hardware for neural networks. In this paper, the field‐free memristive SOT switching of the CoPt single layer is used to propose a neuromorphic hardware circuit for detecting edges in images. Owing to its threefold symmetry of inversion, the polarity of SOT switching can be reversed by rotating the current by 60°. Moreover, the process of current‐induced SOT switching exhibits stable multi‐state magnetic switching behavior, and can be controllably tuned by using the pulse of the current. As the slope of the applied ramp pulse current increased, the wave of the anomalous Hall resistance changed from a curve with normally memristive property to trigonometric, and finally to cosine. The design of the hardware circuit for a single SOT device is subsequently formulated to detect the edges in images. The results of experiments verified the capability of this device to detect the edge lines in images with high accuracy, which confirms its potential for use in the hardware of neuromorphic computing platforms. The work here provides guidance for the application of SOT‐based devices to neuromorphic hardware.

Abstract Bacterial drug resistance monitoring in hospitals is a crucial aspect of healthcare management and a growing concern worldwide. In this study, we analysed the bacterial drug resistance surveillance in our hospital from 2022 Q1 to 2023 Q2. The main sampling sources were respiratory, blood, and urine-based, and the main clinical infections were respiratory and genitourinary in nature. Specimens were inoculated and cultured; bacterial strains were isolated using a VITEK® 2 Compact 60-card automatic microorganism identifier (bioMerieux, Paris, France) and their matching identification cards were identified, and manual tests were supplemented for strain identification. The most common Gram-positive bacteria detected were Staphylococcus aureus, followed by Enterococcus faecalis (E. faecalis), Staphylococcus epidermidis (S. epidermidis), and Staphylococcus haemolyticus (S. haemolyticus). The most common Gram-negative bacteria detected were Escherichia coli, Klebsiella pneumoniae, and Pseudomonas aeruginosa. The most prevalent multidrug-resistant bacteria were those producing extended-spectrum beta-lactamases, followed by methicillin-resistant Staphylococcus aureus, followed by carbapenem-resistant Enterobacterales. This study suggests that the prevention and control of infections in the respiratory and genitourinary systems should be the focus of anti-infective work and that the use of antimicrobials should be reduced and regulated to prevent the emergence and spread of resistant bacteria.

Abstract Objective To investigate the effect of sequential distalization on increasing gaps in the maxillary anterior teeth, focusing on the control of torque and three-dimensional teeth movement during anterior retraction with clear aligners in extraction cases. Methods We recruited 24 patients who were undergoing extraction bilateral maxillary first premolars with clear aligners. According to a predetermined increment in the spaces between the maxillary anterior teeth, the patients were divided into three groups: those with no gap (9 cases), a 0.5 mm gap (6 cases) and a 1.0 mm gap (9 cases). In each group, a 2.0 mm en-mass retraction was applied on the anterior teeth. Plaster casts of the upper full dentition were obtained both before and after a 2 mm retraction. The palatal folds were used to overlap each pair of models. The three-dimensional movement of the teeth and the change of torque for the anterior teeth were subsequently analyzed using Geomagic Studio 2014 software. Results The change in torque in the groups with added gaps was significantly smaller than that in the group with no gaps (P 0.05). In the labial-lingual and vertical directions, the displacements of the central and lateral incisors were smaller in the groups with additional gaps compared to those in the groups without gaps (P 0.05). Then, a comparison was made between the displacement of the second premolar to the second molar in the mesial-distal direction across all groups. This paper considers Cyber-Physical Systems (CPSs) in the presence of Denial-of-Service (DoS) attacks, and investigates the networked control problem under data rate limitations. We develop a framework for CPSs in the presence of DoS attacks, and present a state-estimate control scheme for CPSs under DoS attacks. For the different communication status, the information on the plant state is transmitted to the controller by employing a different data rate in order to mitigate the influence of DoS attacks. Sufficient conditions on the data rate for mean square observability and stabilization of CPSs are derived by applying information theoretic tools of source coding. Our result is less conservative than some existing results. An illustrative example is given to demonstrate the effectiveness of the proposed quantization-coding scheme.

This paper investigates the state estimation problem for linear time-invariant systems where sensors and controllers are connected by a stationary memoryless digital communication channel. Time delay, packet dropout, and data rate limitation occur simultaneously in such a channel. We discuss observability of such systems under communication constraints. A real-time transport protocol (RTP) is used for data transmission. A quantization, coding, and control scheme on the basis of RTP is proposed in order to overcome such difficulties. Sufficient conditions for observability are provided in this case. Our result contains some existing results, and shows that there exists the inherent tradeoff between control and communication costs. Illustrative examples are given to demonstrate the effectiveness of the proposed quantization, coding, and control scheme.

Abstract Background During maxillary arch expansion with a clear aligner (CA), buccal tipping of the posterior teeth often occurs, resulting in an unsatisfactory arch expansion effect. The aim of this study was to analyze the appropriate maxillary arch expansion stride length and torque compensation angle for maxillary dentition to achieve an ideal moving state when a CA was used for upper arch expansion. Methods This study established a three-dimensional (3D) finite element model including a CA, maxilla, periodontal ligament (PDL), and maxillary dentition. The stress distribution, stress situation, expansion efficiency, and movement trends of the maxillary dentition during upper arch expansion of different stride (0.1 mm, 0.2 mm, and 0.3 mm) and torque compensation (0°, 0.5°, 1°, and 1.5°) were measured. Results Maxillary arch expansion lead to buccal tilt of the posterior teeth, lingual tilt of the anterior teeth, and extrusion of the incisors. As the angle of compensation increased, the degree of buccal tilt on the posterior teeth decreased, with this reducing the efficiency of upper arch expansion. When the stride length was 0.1 mm, the torque compensation was 1.2°, and when stride length was 0.2 mm and the torque compensation was approximately 2°, there was a tendency for the posterior teeth to move bodily. However, when the stride length was 0.3 mm, the increase in torque compensation could not significantly improve the buccal tilt phenomenon. In addition, the equivalent von-Mises stress values of the maxillary root, PDL, and alveolar bone were in the same order of magnitude. Conclusions This study indicated that the posterior teeth cause a degree of buccal tilt when maxillary arch expansion is ensured. The specific torque compensation angle should be determined based on the patient’s situation and the desired effect.
